Five extracts from five plants, of which four are endemic to Turkish flora, were screened for their possible in vitro antioxidant activities by two complementary test systems, namely DPPH free radical-scavenging and β-carotene/linoleic acid. In the first case, Pelargonium endlicherianum extract exerted greater antioxidant activity than those of other plant extracts studied with an IC50 value of 7.43 ± 0.47 μg/ml, followed by Hieracium cappadocicum (30.0 ± 0.14 μg/ml). When compared to the synthetic antioxidant BHT (18.0 ± 0.40 μg/ml), the methanolic extract of P. endlicherianum exhibited more than two fold greater antioxidant activity. In the β-carotene/linoleic acid test system, the most active plant was P. endlicherianum with 72.6% ± 2.96 inhibition rate, followed by H. cappadocicum (55.1% ± 2.33) and Verbascum wiedemannianum (52.5% ± 3.11). Antioxidant activities of curcumin and ascorbic acid were also determined as positive controls in parallel experiments.  相似文献

Various analytical methods based on the HPLC-DAD technique were used to determine 38 phenolic compounds in red wines. While anthocyanins and hydroxycinnamic acids were determined by direct injection of wine samples, hydroxybenzoic acids, catechins, procyanidins and flavonols required an analytical pretreatment involving liquid–liquid partitioning with ethyl ether followed by solid-phase extraction on C18 mini-columns. The proposed analytical methods were used to establish the phenol composition of Mencía and Brancellao, two varietal young red wines, and its influence on colour stability during storage in bottles for one year. At the end of malolactic fermentation, Mencía wine was found to contain much greater amounts of anthocyan pigments than was Brancellao wine. This resulted in a higher colour density but a weaker hue in Mencía wine than in Brancellao wine. Phenolic compounds evolved similarly in both wines during storage; changes in such compounds involved a decrease in the levels of monomeric anthocyanins, phenolic acids, epicatechin and flavonols, and an increase in those of procyanidins. The absence of a relationship between the changes in colour density and monomeric anthocyanins in both wines suggests that copigmentation and polymerization with other phenolic compounds (viz. phenolic acids, catechins and/or flavonols) prevail over degradation of the pigments.  相似文献

Aqueous extract, proanthocyanidin rich extract, and organic extracts of Cymbopogon schoenanthus L. Spreng (lemon grass) shoots from three different locations in South Tunisia were screened for their antioxidant, acetylcholinesterase and antimicrobial activities. In addition to the evaluation of these activities, the contents of flavonoids and total phenolic compounds were determined.Antioxidant activity measured by DPPH assay showed that the proanthocyanidin extract exhibited higher antioxidant activity than the aqueous extract. Extract concentration providing 50% inhibition (IC50) ranged from 16.4 ± 6.8 μg/mL to 26.4 ± 6.8 μg/mL. The antioxidant activity was also determined using the β-carotene/linoleic acid bleaching test. The best results (IC50 = 0.11 ± 0.10 mg/mL) were obtained with the proanthocyanidin extract of the plants collected from the desert region (Dhibat).The greatest acetylcholinesterase inhibitory activity (IC50 = 0.23 ± 0.04 mg/mL) was exhibited by the ethyl acetate and methanol extracts of the plants collected from the mountainous region. It seems that extracts obtained with more polar solvents gave better results.The proanthocyanidin extracts showed a good antimicrobial activity against Streptococcus sobrinus at low concentration (MIC = 4 mg/mL). Therefore, these extracts could be used to prevent carious lesions by inhibiting S. sobrinus growth.  相似文献

This study was designed to examine the in vitro antioxidant activities and rosmarinic acid levels of the methanol extracts of Salvia verticillata subsp. verticillata and S. verticillata subsp. amasiaca. The extracts were screened for their possible antioxidant activity by two complementary test systems, namely DPPH free radical-scavenging and β-carotene/linoleic acid systems. In the first case, S. verticillata subsp. verticillata was superior to the subsp. amasiaca with an IC50 value of 14.5 ± 1.21 μg mg−1. In the β-carotene/linoleic acid test system, inhibition capacity of S. verticillata subsp. verticillata was 74.4 ± 1.29%. Antioxidant activities of BHT, ascorbic acid, curcumin and α-tocopherol were determined in parallel experiments. Activity of rosmarinic acid was also screened for better establishing the relationship between rosmarinic acid level and antioxidant activity for the plant extracts. S. verticillata subsp. verticillata had the highest rosmarinic acid level with a value of 28.7 ± 0.89 μg mg−1. There is a strong correlation between the rosmarinic acid level and antioxidant activity potential. Our results showed that rosmarinic acid and its derivatives are more likely to be responsible for most of the observed antioxidant activities of Salvia species.  相似文献

Galicia (the north-western corner of Spain) is a Spanish region with several old-traditional winegrowing areas. Vitis vinifera L. cv. Mencía is one of the most often used to produce quality red wines but the tendency of the world wine-making market is to reward the production of wines that have particular and differentiated characteristics. In Galicia, there are other red cultivars such as Gran Negro which were not exploited for their potential to produce quality red wines. Dynamics of anthocyanin and flavonol accumulation in Gran Negro berries were studied separately in two different positions within the cluster (tips and shoulders) during 30 days before harvest to assess the existence of different polyphenol quality in both positions and as a consequence, red wines with different qualities. Dynamics of anthocyanins over ripening confirmed that anthocyanins presented upward trends (from 1510 to 1727 mg/kg in tips; and from 1532 to 1728 mg/kg in shoulders) but dynamics of flavonols confirmed that maximum values were reached and stabilised thirty days before harvest (from 45 to 39 mg/kg in tips; and from 49 to 45 mg/kg in shoulders). No differences were observed in anthocyanin and flavonol contents collected from tips and shoulders which indicates that is not necessary to harvest them separately. The results from Gran Negro were compared with those of Brancellao and Mouratón.  相似文献

Mencía is one of the most often used to produce quality red wines in the five Galicia Denomination of Origin. However, there are traditional cultivars such as Brancellao, which were not exploited for their potential to produce quality red wines. Dynamics of anthocyanin and flavonol accumulation were studied separately in two different positions within the cluster (tips and shoulders), during 30 days before harvest. The objective of separating berries is to assess the existence of different polyphenol quality in both positions; as a consequence, the selection and harvest of those berries with a higher content of anthocyanins and flavonols could produce red wines with different qualities. Derivatives of five anthocyanins (malvidin, peonidin, petunidin, delphinidin and cyanidin) were detected in skins at both positions within the cluster. Anthocyanin contents stabilized in the 30 days prior to harvest in the berries from the shoulders whereas they continue to increase in those from the tips. Derivatives of six flavonols (quercetin, myricetin, kaempherol, laricitrin, isorhamnetin and syringetin) were detected in skin and flesh at both positions within the cluster. Dynamics of anthocyanins (from 400 to 515 mg/kg in tips; and from 598 to 574 mg/kg in shoulders) and flavonols (from 19 to 29.3 mg/kg in tips; and from 22.7 to 29.4 mg/kg in shoulders) over ripening confirmed that these polyphenols presented upward trends. Therefore, it is not necessary to harvest Brancellao berries separately and a high quality red wine will be obtained with berries from entire clusters regarding these compounds.  相似文献

This study was designed to examine the in vitro antioxidant activities of the methanol extracts of six Salvia species [Salvia caespitosa Montbret & Aucher ex Bentham (ENDEMIC), Salvia hypargeia Fisch. & Mey. (ENDEMIC), Salvia euphratica subsp. euphratica Montbret & Aucher ex Bentham (ENDEMIC), Salvia sclarea L., Salvia candidissima subsp. candidissima Montbret & Aucher ex Bentham and Salvia aethiopis L.] from Turkey. The extracts were screened for their possible antioxidant activities by two complementary test systems, namely DPPH free radical-scavenging and β-carotene/linoleic acid systems. Non-polar subfractions of the methanol extracts of Salvia species studied did not show any antioxidant activity in both test systems. In the first case, the most active plant was S. euphratica subsp. euphratica, an endemic species, with an IC50 value of 20.7 ± 1.22 μg/ml, followed by S. sclarea (IC50 = 23.4 ± 0.97 μg/ml) among the polar subfractions. In the β-carotene/linoleic acid test system, polar extract of S. hypargeia was superior to the polar extracts of other Salvia species studied (69.2% ± 1.90%). This activity was followed by S. sclarea with 63.5% ± 4.24% inhibition rate. The inhibition rate of the synthetic antioxidant, buthylated hydroxytoluene (BHT), was also determined to be 96%. Since the polar extracts of Salvia species dealt with here exhibited excellent antioxidant activities when compared to BHT, it seems possible to keep perishable fat-containing food longer by direct addition of an extract of sage.  相似文献

Different commercial edible oilseed samples were analyzed in order to evaluate the compounds involved in antioxidant activity. Fatty acids, triacylglycerols, tocopherols, chlorophylls, β-carotene, squalene, phenolic compounds and CIE Lab coordinates were assayed. Strong chemical variability depending on the oilseed variety was observed. Antioxidant activity was evaluated for the oils, both for their methanol-soluble phase and the fraction insoluble in methanol. DPPH radical scavenging activity expressed in TEAC ranged between 0.45 and 2.30 mmol/l in peanut and maize oils, respectively. Free radical scavenging activity was mainly influenced by tocopherols content (r = +0.70) in oils and polyunsaturated fatty acids (r = +0.61) in the fraction nonsoluble in methanol. Variability of the correlation between the antioxidant activity and the composition of the oilseeds could be attributed to the differences in the squalene, chlorophylls, carotenoids, and phenols contents of the oils and their mutual interactions.  相似文献

Antioxidant capacity and antimicrobial activities of Laetiporus sulphureus (Bull.) Murrill. extracts obtained with ethanol were investigated in this study. The study was aimed at determining the antioxidant activity (DPPH free radical-scavenging, β-carotene/linoleic acid systems), total phenolic content and total flavonoid concentration of L. sulphureus. Inhibition values both of L. sulphureus ethanol and the standards increased parallel with the elevation of concentration in the linoleic acid system. Inhibition values of L. sulphureus (LS) extract, BHA and α-tocopherol standards were found to be 82.2%, 96.4% and 98.6%, respectively, at a concentration of 160 μg/ml. DPPH free radical-scavenging activity was found to exhibit 14%, 26%, 55% and 86% inhibition, respectively, at concentrations of 100, 200, 400 and 800 μg/ml. Total flavanoids were 14.2 ± 0.12 μg mg−1 (quercetin equivalent) while the phenolics were 63.8 ± 0.25 μg mg−1 (pyrocatechol equivalent) in the extract. Positive correlations were found between total phenolic content in the mushroom extracts and their antioxidant activities. Edible mushrooms may have potential as natural antioxidants. L. sulphureus showed narrow antibacterial activity against Gram-negative bacteria and strongly inhibited the growth of the Gram-positive bacteria tested. The crude extract exhibited high anticandidal activity on Candida albicans. Therefore, the extracts could be suitable as antimicrobial and antioxidative agents in the food industry.  相似文献

In the present work, we report the results of a study aimed at evaluating the antiradical activity, the antioxidant activity and the acetylcholinesterase (E.C. 3.1.1.7.) inhibitory capacity of essential oils, ethanol and boiling water extracts from five aromatic herbs growing wild in Portugal and used in traditional food preparations: fennel (Foeniculum vulgare), mint (Mentha spicata), pennyroyal (Mentha pulegium), rosemary (Rosmarinus officinalis) and wild thyme (Thymus serpyllum). The water extracts of M. spicata and M. pulegium showed the highest radical-scavenging activity (DPPH test) values (IC50 = 5.7 ± 0.4 and 8.9 ± 0.2 μg ml−1 respectively). This activity was higher than that found with the standard antioxidant BHT. The ethanol extracts of M. spicata, T. serpyllum and F. vulgare showed the highest antioxidant activities measured by the β-carotene/linoleic acid assay, IC50 = 36.9 ± 0.1, 41.2 ± 0.1 and 68.7 ± 0.1 μg ml−1, respectively. The inhibition of AChE was higher in the essential oil fraction. The highest activity was found for R. officinalis with an IC50 = 69.8 ± 0.1 μg ml−1.  相似文献

The polyphenolic compound content has been determined in 15 Amazonian plant species (leaves, bark, stems, fruits, and seeds) used in folk medicine, using two complementary spectrophotometric methods. In addition, the antioxidant activity of the corresponding plant extracts has been determined by TEAC (trolox equivalent antioxidant capacity) and ORACFluorescein (oxygen radical antioxidant capacity using fluorescein as fluorescent probe) assays to identify naturally-rich sources of antioxidants. The plants under investigation showed a great range of TEAC (1.0 up to 347.1 μmol of trolox equiv./g) and ORAC (6.7 up to 1396.4 μmol of trolox equiv./g) values. These values were highly correlated to the concentration in total phenolics obtained by the Folin–Ciocalteau procedure (TEAC: r2 = 0.88, n = 65; ORAC: r2 = 0.70, n = 62), and in total flavanoids, quantified using the chromogen reagent p-dimethylaminocinnamaldehyde (TEAC: r2 = 0.75, n = 54; ORAC: r2 = 0.74, n = 51). The high antioxidant capacities found in leaves and bark of Byrsonima crassifolia, Inga edulis, Davilla kunthii and Cecropia palmata and also their great biomasses in the forest should stimulate further studies regarding the characterization, purification and concentration of their phenolic compounds.  相似文献

The study established baseline data on the total phenolic content and antioxidant activities of five sweet potato (Ipomoea batatas) varieties grown in the Philippines including Dakol, Emelda, Haponita, PSBSP and Violet. Phenolic content ranged from 192.7 to 1159.0 mg gallic acid equivalent (GAE) /100 g dry sample. Antioxidant activities were highest for Dakol, with an EC50 value of 0.7 ± 0.2 mg/mL for DPPH radical scavenging activity, 2.5 ± 0.5 mg/mL for reducing power, and 2.4 ± 0.3 mg/mL for iron-chelating ability, on a dry basis. However, Haponita had the best inhibitory action on linoleic acid oxidation at 99.4 ± 0.9%. Methanolic sweet potato extracts had higher radical scavenging activity, reducing power and oxidation inhibition than α-tocopherol and higher iron-chelating capacity than ethylenediamine tetraacetic acid (EDTA). Significant (∗P < 0.05) negative correlation was observed between total phenolic content and the EC50 for DPPH radical scavenging activity (R = −0.826), reducing power (R = −0.876) and iron-chelating capacity (R = -0.800).  相似文献

This study was designed to examine the in vitro antioxidant activities of Rosmarinus officinalis L. essential oil compared to three of its main components (1,8-cineole, α-pinene, β-pinene). GC–MS analysis of the essential oil resulted in the identification of 19 compounds, representing 97.97% of the oil, the major constituents of the oil were described as 1,8-cineole (27.23%), α-pinene (19.43%), camphor (14.26%), camphene (11.52%) and β-pinene (6.71%). The oil and the components were subjected to screening for their possible antioxidant activity by means of 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay and β-carotene bleaching test. In the DPPH test system, free radical-scavenging activity of R. officinalis L. essential oil, 1,8-cineole, α-pinene and β-pinene were determined to be 62.45% ± 3.42%, 42.7% ± 2.5%, 45.61% ± 4.23% and 46.21% ± 2.24% (v/v), respectively. In the β-carotene bleaching test system, we tested series concentration of samples to show the antioxidant activities of the oil and its main components, whereas the concentrations providing 50% inhibition (IC50) values of R. officinalis L. essential oil, 1,8-cineole, α-pinene and β-pinene were 2.04% ± 0.42%, 4.05% ± 0.65%, 2.28% ± 0.23% and 2.56% ± 0.16% (v/v), respectively. In general, R. officinalis L. essential oil showed greater activity than its components in both systems, and the antioxidant activities of all the tested samples were mostly related to their concentrations. Antioxidant activities of the synthetic antioxidant, ascorbic acid and BHT, were also determined in parallel experiments as positive control.  相似文献

The effect of ambient and refrigerated storage temperature on post-harvest phytochemical qualities of broccoli florets was investigated during storage. Fresh broccoli florets were packed in polypropylene (PP) micro-perforated film bags and stored, under open ambient storage conditions (15 ± 1 °C, 55 ± 2% RH), and laboratory refrigerated storage (4 ± 0.5 °C, 50 ± 2% RH) for a total period of 144 h. Quality of broccoli florets was evaluated in terms of physiological weight loss (PLW), ascorbic acid content, chlorophyll content, β-carotene and total antioxidant activity. Samples packed in PP micro-perforated film showed significantly (P < 0.05) lower losses of PLW, ascorbic acid, chlorophyll, β-carotene and total antioxidant activity (5.51%, 4.53%, 18.9%, 4.04% and 16.4%, respectively), during storage for up to 144 h under refrigerated conditions. For better phytochemical retention, the broccoli florets should be packed in PP micro-perforated film bags and stored under refrigerated conditions.  相似文献

This study was designed to examine the in vitro antioxidant activities of the essential oil and methanol extracts of Satureja spicigera and S. cuneifolia from Turkish flora. GC and GC/MS analysis of the essential oils resulted in the identification of 40 and 29 compounds, representing the 99.4% and 99.5% of the oils, respectively. Major constituents of the oils were carvacrol (42.5% and 67.1%), γ-terpinene (21.5% and 15.2%) and p-cymene (20.9% and 6.7%), respectively. Methanol extracts were also obtained from the aerial parts of the plants. The samples were subjected to a screening for their possible antioxidant activities by using 2,2-diphenyl-1-picrylhydrazyl (DPPH) and β-carotene–linoleic acid assays. In general, samples obtained from S. cuneifolia exerted greater antioxidant activities than did those obtained from S. spicigera. In the DPPH test system, free radical-scavenging activity of S. spicigera oil was determined to be 127 ± 1.63 μg/ml, whereas IC50 value of S. cuneifolia was 89.1 ± 2.29 μg/ml. In the β-carotene–linoleic acid test system, antioxidant activities of the oil were 81.7 ± 1.14% and 93.7 ± 1.83%, respectively. Antioxidant activities of the synthetic antioxidant, BHT, ascorbic acid, curcumin and α-tocopherol were also determined in parallel experiments.  相似文献

Fish protein hydrolysates (FPH) with antioxidative properties were prepared using Pacific hake fish with high endogenous proteolytic activity from Kudoa paniformis parasitic infection. Infection level of ∼107K. paniformis spores/g fish mince or higher yielded FPH with high antioxidant potential by autolysis and/or Validase® BNP or Flavourzyme® 500L. Autolyzing fish mince containing 30 × 106 spores/g for 1 h at 52 °C and pH 5.50 produced FPH (named E-1h) with Trolox equivalent antioxidant capacity (TEAC) in the 2,2′-azinobis-(3-ethylbenzothiazoline-6-sulphonic acid) radical assay of 262 ± 2 μmol/g and oxygen radical absorbing capacity (ORAC) of 225 ± 17 μmol Trolox equivalents/g freeze-dried sample. E-1h FPH also exhibited a marked concentration-dependent scavenging activity on 1,1-diphenyl-2-picrylhydrazyl radical. Antioxidant activity of E-1h FPH was higher (p < 0.05) than BHA and α-tocopherol in a linoleic acid peroxidation system over prolonged storage (∼162 h). Antioxidative FPH from Pacific hake may be useful ingredients in food and nutraceutical applications.  相似文献

The aerial parts of two endemic Pimpinella [Pimpinella anisetum Boiss. & Ball. and Pimpinella flabellifolia (Boiss.) Benth. ex Drude] were hydro-distilled to produce oils in the yields of 2.07% (v/w) and 2.61% (v/w), respectively. The oils were analysed by GC and GC/MS. Twenty-one and nineteen components were identified, representing 99.5% and 99.7% of the oils, respectively. The main compounds of P. anisetum were (E)-anethole (82.8%) and methyl chavicol (14.5%), whereas limonene (47.0%), (E)-anethole (37.9%) and α-pinene (6.0%) were the major constituents of P. flabellifolia. The oils were screened for their possible antioxidant activities by two complementary test systems, namely DPPH free radical-scavenging and β-carotene/linoleic acid systems. In the first case, P. anisetum oil exerted greater antioxidant activity than that of P. flabellifolia oil with an IC50 value of 5.62 ± 1.34 μg/ml. In the β-carotene/linoleic acid test system, the oil of P. anisetum was superior to P. flabellifolia with 70.5% ± 2.86 inhibition rate. Essential oils of the plants studied here were also screened for their antimicrobial activities against six bacteria and two fungi. The oils showed moderate antimicrobial activity against all microorganisms tested.  相似文献

Arils from six pomegranate (Punica granatum L.) cultivars obtained from various sites from the Mediterranean region of Turkey were evaluated for their chemical and antioxidant properties. These properties included total phenolics (TP), total monomeric anthocyanins (TMA), soluble solids (TSS), titratable acidity (TA), individual sugars and organic acids. Antioxidant capacities of arils were determined by both the ferric reducing antioxidant power (FRAP) and trolox equivalent antioxidant capacity (TEAC) assays. The antioxidant capacities averaged 5.60 and 7.35 mmol TE/l by the TEAC and FRAP methods. Variability among cultivars was greatest for TMA content (CV 132%); individuals ranged from 6.1 to 219 mg cy3-Gluc l−1. TP means averaged 1507 mg GAE/l. Levels of FRAP, TEAC, TP, and TMA were strongly correlated (r = 0.82–0.96). The major sugars were fructose (6.4 g/100 ml) and glucose (6.8 g/100 ml), the major acids were citric (1.78 g/100 ml) and malic (0.12 g/100 ml).  相似文献

The aim of this work was to use different assays to evaluate the antioxidant and vasodilatory properties of three typical food products from the Mediterranean area and to correlate these activities with their phenolic content. For this purpose, red wines Cannonau, liqueurs obtained by cold maceration of myrtle (Myrtus communis L.) berries and bitter honeys obtained from strawberry-tree flowers (Arbutus unedo L.) were analysed. The total phenolic (TP) content was measured spectrophotometrically with a modified Folin–Ciocalteau method and phenolic compounds were identified and dosed by HPLC-DAD and LC-MS/MS. Antioxidant activities were evaluated with DPPH, FRAP and ABTS assays and the in vitro vasodilatory effects were assessed using norepinephrine precontracted rat aortic rings. Cannonau wines and myrtle liqueurs showed high levels of TP (1978 ± 279 and 1741 ± 150 mg GAE/L, respectively), linearly correlated to the results of FRAP, ABTS, and DPPH assays. Their maximal vasodilatory activity was 61.7 ± 4.1% and 53.0 ± 3.0%, respectively. Although strawberry-tree honey contained relatively high levels of phenolic compounds (922 ± 38 mg GAE/kg), it did not induce vasodilation, even at the highest dose tested (0.206 g/L). These results indicate that foods with high levels of phenolic compounds should be studied using several different biological assays before being recommended to the general public as functional foods.  相似文献
