We isolated four compounds, fustin, gallic acid, 3′,4′,7-trihydroxyflavone, and fisetin from Rhus verniciflua. These compounds showed electron donation ability (87–94%) that was stronger than butylated hydroxyanisole (52%). Gallic acid (OD700 = 1.98) showed the highest reducing power, and the other isolated compounds (OD700 = 0.66–1.31) showed stronger activity than α-tocopherol (OD700 = 0.21). The ethyl-acetate fraction had the highest phenolic content (723 mg GAE/g), followed by the 80% ethanolic extract (597 mg GAE/g). For Gram-negative bacteria, fisetin had the most potent anti-bacterial activity (MIC = 8 μg/ml) against Escherichia coli. 3′,4′,7-Trihydroxyflavone (106%), the 80% ethanolic extract (101%), and ethyl-acetate (98%) had the most powerful α-glucosidase inhibitory effect at 50 μg/ml. R. verniciflua extracts have potential as functional food additives.  相似文献

The present study investigates the antimicrobial activity of various solvent extracts of Eruca sativa (aerial and root) and seed oil against-antibiotic resistant Gram-negative (Escherichia coli, Pseudomoms aeruginosa and Shigella flexneri) and Gram-positive (Staphylococcus aureus and Bacillus subtilis) bacteria. Among the various preparations, seed oil was the most active, exhibiting a maximum zone inhibition of 97% for Gram-positive bacteria and of 74–97% for Gram-negative bacteria. The MIC of the seed oil was found to be 65–75 and 60–70 μg/ml for Gram-negative and Gram-positive bacteria, respectively. Analytical investigation on main volatile and non-volatile components was performed on seed oil. Among the formers allyl isothiocyanate (40 μg/g), 3-butenyl isothiocyanate (260 μg/g), 4-methylsulfinybutyl isothiocyanate (sulforaphane 743 μg/g), 2-phenylethyl isothiocyanate (159 μg/g) and bis(isothiocyanatobutyl)disulphide (∼5000 μg/g) were determined by head space/SPME/GC–MS analysis. Free fatty acids were 1.6% w/w of the oil and overall 25 fatty acids were identified. Erucic and oleic acids were the main fatty acids both in the free (7.8 and 2.1 mg/ml) and esterified forms (50.6% w/w and 14.9% w/w of total fatty acids). The essential oil from underground parts of Ferula heuffelii from N.E. Serbia, was analysed using GC and GC–MS. The main compounds of the essential oil were elemicin (35.4%) and myristicin (20.6%). The essential oil exhibited the best antimicrobial activity against two strains of Candida albicans (MIC = 7.0 and 13.7 μg/ml), as well as against Micrococcus luteus (MIC = 13.7 μg/ml), Staphylococcus epidermidis (MIC = 17.6 μg/ml), Bacillus subtilis (MIC = 21.1 μg/ml) and Micrococcus flavus (MIC = 28.2 μg/ml). In the DPPH radical scavenging assay, essential oil showed substantial activity with SC50 = 22.43 μl/ml. The essential oil was also tested for antispasmodic activity. It inhibited spontaneous contraction of isolated rat ileum dose-dependently, and at the concentration of 86.64 μg/ml exhibited 50% of the maximum effect of atropine. After incubation with 75.00 μg/ml of essential oil, acetylcholine did not induce contractions of ileum, and at 250.00 μg/ml, the essential oil almost completely abolished the spasmodic effect of potassium chloride (80 mM).  相似文献

Chromatographic purification of the dichloromethane-soluble fraction of alga, on neutral alumina, using increasing concentrations of ethylacetate/n-hexane as eluents, yielded seven labdane diterpenoids (1–7) as major constituents of green alga Ulva fasciata. Structures of these diterpenoids were established using extensive spectroscopic techniques. Antimicrobial assay showed that the compounds labda-14-ene-3α,8α-diol (2) and labda-14-ene-8α-hydroxy-3-one (4) were inhibitory to the growth of Vibrio parahaemolyticus and Vibrio alginolyticus with minimum inhibitory concentrations of 30 μg/ml by 2, and 40 μg/ml by 4, respectively against the former and 30 μg/ml by 2, and 80 μg/ml by 4, respectively, against the latter. Structure–activity relationship analyses revealed that the compounds with electronegative hydroxyl or carbonyl group(s) exhibit greater activities, apparently by proton exchange reaction with the basic aminoacyl residue at the macromolecular receptor site of virulent enzymes of pathogenic bacteria. These might provide promising therapeutic agents against infections with multi-resistant Gram-negative fish pathogenic bacteria.  相似文献

Essential oil from the aerial parts of Artemisia indica was analysed by GC-FID and GC–MS. A total of 43 compounds representing 96.8% of the oil were identified and the major components were found to be artemisia ketone (42.1%), germacrene B (8.6%), borneol (6.1%) and cis-chrysanthenyl acetate (4.8%). Antimicrobial activity of the oil was evaluated against seven clinically significant bacterial and two fungal strains. The essential oil and its major constituents exhibited moderate to potent, broad-spectrum antibacterial and antifungal activities targeting both Gram-positive and Gram-negative bacteria. In vitro cytotoxicity evaluation against four human cancer cell lines THP-1 (leukemia), A-549 (lung), HEP-2 (liver) and Caco-2 (colon) showed that the essential oil exhibited concentration dependant growth inhibition in the 10–100 μg/ml dilution range, with IC50 values of 10 μg/ml (THP-1), 25 μg/ml (A-549), 15.5 μg/ml (HEP-2) and 19.5 μg/ml (Caco-2). It was interesting to note that the essential oil also exhibited potent antioxidant activity.  相似文献

Plectranthus barbatus, known as “falso boldo” in Brazil, is used in herbal tea or cooked as a vegetable. Infusions and decoctions of leaves from P. barbatus were analysed for their inhibition of acetylcholinesterase and their antioxidant activity. The decoction showed high inhibition activity (31% inhibition with 0.5 mg of extract/ml) and also high antioxidant activity (IC50 = 45.8 ± 0.5 μg of dry extract/ml in the DPPH test; IC50 = 69.8 ± 3.1 μg of dry extract/ml in the β-carotene–linoleic acid test). Rosmarinic acid, scutellarein 4′-methyl ether 7-O-glucuronide and (16S)-coleon E were the main constituents identified. These compounds have antiacetylcholinesterase activity. Rosmarinic acid and the scutellarein derivative have IC50 = 440 μg/ml and 1 mg/ml, respectively. One milligram per millilitre of (16S)-coleon E showed 61% inhibition of the enzyme. Other Plectranthus species, P. ecklonii, P. fructicosus, P. lanuginosus and P. verticillatus, were also analysed and the results obtained correlated with the content in rosmarinic acid.  相似文献

This study provides the first chemical investigation of wild-harvested fruits of Vanilla pompona ssp. grandiflora (Lindl.) Soto-Arenas developed in their natural habitat in the Peruvian Amazon. Flowers were hand-pollinated and the resulting fruits were analysed at different developmental stages using an HPLC-DAD method validated for the quantification of glucovanillin and seven other compounds. The method showed satisfactory linearity (r2 > 0.9969), precision (coefficient of variation <2%), recoveries (70–100%), limit of detection (0.008–0.212 μg/ml), and limit of quantification (0.027–0.707 μg/ml). The evaluation of crude and enzyme-hydrolyzed Soxhlet-extracted samples confirmed the leading role of glucosides in fruit development. LC–ESI-MS studies corroborated the identities of four glucosides and seven aglycones, among them vanillin (5.7/100 g), 4-hydroxybenzyl alcohol (3.6/100 g), and anisyl alcohol (7.1/100 g) were found in high concentrations. The attractive flavor/aroma profile exhibited by wild V. pompona fruits supports studies focused on the development of this species as a specialty crop.  相似文献

The effect of additives and post-treatment incubation conditions on the recovery of high pressure and heat-injured (i.e., processed at 620 MPa and 95 and 100 °C for 5 min) spores of Clostridium botulinum strains, 62-A (proteolytic type A) and 17-B (nonproteolytic type B) was studied. High pressure and heat-injured spores were inoculated into TPGY (Trypticase–Peptone–Glucose–Yeast extract) anaerobic broth media containing additives (lysozyme, l-alanine, l-aspartic acid, dipicolonic acid, sodium bicarbonate, and sodium lactate) at various concentrations (0–10 μg/ml) individually or in combination. The spore counts of high pressure and heat-injured 62-A and 17-B recovered from TPGY broth containing lysozyme (10 μg/ml) incubated for 4 months versus that recovered from peptone–yeast extract–glucose–starch (PYGS) plating agar containing lysozyme (10 μg/ml) incubated under anaerobic conditions for 5 days were also compared. None of the additives either individually or in combination in TPGY broth improved recovery of injured spore enumeration compared to processed controls without additives. Addition of lysozyme at concentrations of 5 and 10 μg/ml in TPGY broth improved initial recovery of injured spores of 17-B during the first 4 days of incubation but did not result in additional recovery at the end of the 4 month incubation compared to the processed control without lysozyme. Adding lysozyme at a concentration of 10 μg/ml to PYGS plating agar resulted in no effect on the recovery of high pressure and heat-injured 62-A and 17-B spores. The recovery counts of high pressure and heat-injured spores of 62-A and 17-B were lower (i.e., <1.0 log units) with PYGS plating agar compared to the MPN method using TPGY broth as the growth medium.  相似文献

Zanthoxylum avicennae is commercially used as healthcare products for the improvement of chronic hepatitis and rheumatism. Its fruit is also used as a spice. Five new coumarins, 5′-methoxyauraptene (1), 6,5′-dimethoxyauraptene (2), 5′-methoxycollinin (3), 7-((2′E,5′E)-7′-methoxy-3′,7′-dimethylocta-2′,5′-dienyloxy)coumarin (4), and 6-methoxy-7-((2′E,5′E)-7′-methoxy-3′,7′-dimethylocta-2′,5′-dienyloxy)coumarin (5), have been isolated from the leaves of Z. avicennae, together with 15 known compounds (620). The structures of these new compounds were determined through spectroscopic and MS analyses. Compounds 6, 915, and 18 exhibited inhibition (IC50 ? 8.29 μg/ml) of superoxide anion generation by human neutrophils in response to formyl-l-methionyl-l-leucyl-l-phenylalanine/cytochalasin B (fMLP/CB). Compounds 6, 10, 12, 14, and 18 inhibited fMLP/CB-induced elastase release with IC50 values ?6.30 μg/ml.  相似文献

A GC–MS analysis of Erysimum diffusum Ehrh. (Brassicaceae) glucosinolate autolysis products led to the identification of a new mustard oil constituent – 4-isothiocyanatobutanoic acid. The identity of the new glucosinolate breakdown product was confirmed by means of co-injection of the synthetic compound in the GC with the root autolysate and derivatization reaction to the well known methyl ester. Methyl ester of this acid was long believed to be a genuine glucosinolate hydrolysis product but we now provide evidence that it could be an artifact of the isolation procedure. 4-Isothiocyanatobutanoic acid was assayed for antimicrobial activity and demonstrated significant inhibitory (10–220 μg/ml) and microbicidal (10–1175 μg/ml) activity against important human pathogens.  相似文献

The contents of Pb and Cd in two species of Iranian tuna fish (yellowfin and skipjack), which were caught from the Persian Gulf and Oman Sea, and the effects of canning processing steps on their contents were assessed by electrothermal atomic absorption spectrometry. The results revealed that the levels of lead and cadmium throughout the processing steps in yellowfin were in range of 0.154 ± 0.019–0.441 ± 0.025 μg/g and 0.029 ± 0.002–0.084 ± 0.0005 μg/g, respectively. Pb and Cd concentrations from received fish to final product in skipjack were found to be in range of 0.072 ± 0.031–0.218 ± 0.031 μg/g and 0.016 ± 0.001–0.062 ± 0.002 μg/g, respectively. The limit of detection for lead and cadmium were 0.058 μg/g (11.6022 μg/l) and 0.0007 μg/g (0.1485 μg/l), respectively. Results from paired sample t-test analysis showed that defrosting, cooking, and sterilisation by autoclave would reduce the contents of lead and cadmium, considerably.  相似文献

The present study analysed the antioxidant activity of the technical Cashew Nut Shell Liquid (tCNSL) using the 2,2-diphenyl-1-picrylhydrazyl (DPPH) scavenging assay and the xanthine oxidase assay, as well as in vivo evaluation by Saccharomycescerevisiae assay. Also, the chemical composition of tCNSL was determined by gas chromatography–mass spectrometry (GC–MS), revealing the presence of cardanols (40.26%), cardols (29.95%), phytosterol (10.68%), triacontanes (4.66%) and anacardic acid (1.79%). The DPPH-based assay results showed that tCNSL 1000 μg/ml reduced the radical level by 88.9%, and the scavenging of hydroxyl radicals in the xanthine oxidase assay indicated significant antioxidant activity (IC50 = 702 μg/ml). In addition, tCNSL exerts an important protective effect against oxidative stress in yeast when used in 100–500 μg/ml concentration range, when exposed to paraquat or H2O2, indicating an in vivo antioxidant effect.  相似文献

Phytochemical compositions of five varieties of black soybeans (Glycine max) and their stabilities at room temperature, 4 and −80 °C over 14 months were determined by HPLC systems with electrochemical (ECD) and UV detectors. Polyphenol profiling was carried out by a liquid chromatography–electrospray ionisation-mass spectrometry (LC–ESI-MS) with orbitrap as a mass analyser in both positive and negative ion modes, and polyphenols in aglycone forms were quantified by HPLC–ECD. Five different varieties of black soybeans (G. max) contained 249–405 μg/g dry wt of γ-tocopherol and 6.76–14.98 μg/g dry wt of lutein. Major polyphenols in black soybeans (G. max) were daidzein (193–288 μg/g dry wt) and genistein (145–223 μg/g dry wt), mainly present as glucosides and acetyl glucosides. No significant decrease was found in total phenols of black soybeans (G. max) stored at room temperature, 4 or −80 °C for 14 months. On the other hand, lutein and γ-tocopherol degraded significantly within a month of storage at room temperature (p < 0.01), whereas they remained stable up to 6 months at 4 °C and up to 14 months at −80 °C. The current study indicates that black soybeans (G. max) are rich source of γ-tocopherol and phenols (isoflavones, flavonols, flavan-3-ols, proanthocyanidins and anthocyanin) and that the levels vary depending upon varieties. In addition, storage at low temperature is recommended to reduce the loss of fat-soluble phytochemicals in black soybeans (G. max) over an extended period of time.  相似文献

This study was designed to examine the chemical composition and in vitro antioxidant activity of the essential oil of Clinopodium vulgare. GC–MS analysis of the oil resulted in the identification of 40 compounds, representing 99.4% of the oil; thymol (38.9%), γ-terpinene (29.6%) and p-cymene (9.1%) were the main components. The samples were subjected to a screening for their possible antioxidant activity by using 2,2-diphenyl-1-picrylhydrazyl (DPPH) and β-carotene-linoleic acid assays. In the first case, IC50 value of the C. vulgare essential oil was determined as 63.0 ± 2.71 μg/ml. IC50 value of thymol and γ-terpinene, the major compounds of the oil, was determined as 161 ± 1.3 μg/ml and 122 ± 2.5 μg/ml, respectively, whereas p-cymene did not show antioxidant activity. In β-carotene-linoleic acid system, C. vulgare essential oil exhibited 52.3 ± 1.19% inhibition against linoleic acid oxidation. In both systems, antioxidant capacities of BHT, curcumine and ascorbic acid were also determined in parallel experiments.  相似文献

The chemical composition of the hydrodistillate of aerial parts of Cymbopogon jawarancusa, a natural grass considered as major forage for animal nutrition, used in food because of the presence of sufficient concentration of minerals like calcium and potassium was analysed by capillary GC–FID, GC–MS and 13C NMR. Seventeen constituents representing 97.8% of the total oil with piperitone (58.6%) and elemol (18.6%) as major constituents were identified. In vitro cytotoxicity of the oil and its constituents on human cancer cell lines THP-1 (leukemia), A-549 (lung), HEP-2 (liver) and IGR-OV-1 (ovary) was evaluated by Sulphorhodamine-B assay. The oil was found to be more potent than its components against cancer cell lines tested with IC50 of 6.5 μg/ml (THP-1), 6.3 μg/ml (A-549), 7.2 μg/ml (HEP-2) and 34.4 μg/ml (IGR-OV-1). Antioxidant activity of oil and its constituents was evaluated by DPPH assay. In conclusion, the results demonstrate potent cytotoxic and antioxidant effects of oil, and its components like piperitone, α-pinene, β-caryophyllene and β-elemene.  相似文献

Satureja cuneifolia Ten. is a well-known aromatic plant which is frequently used as a spice and herbal tea in Anatolia. S. cuneifolia oil was analyzed by gas chromatography/mass spectrometry (GC/MS). The major components of S. cuneifolia oil were carvacrol (44.99%) and p-cymene (21.61%). The essential oil of S. cuneifolia exhibited antimicrobial activity against all of the tested foodborne and spoilage bacteria. The minimum inhibitory concentration (MIC) values for test bacteria which were sensitive to the essential oil of S. cuneifolia were in the range of 600–1400 μg/ml. Antioxidant activities of the essential oil and the methanolic extract from S. cuneifolia were evaluated by using DPPH radical scavenging, β-carotene–linoleic acid bleaching and metal chelating activity assays. In addition, the amounts of total phenol components in the plant methanolic extract (222.5 ± 0.5 μg/mg) and the oil (185.5 ± 0.5 μg/mg) were determined.  相似文献

The essential oils of four Teucrium species were studied and 150 components, in all, were identified. All oils were rich in sesquiterpenes (50.1–55.8%). Spathulenol and δ-cadinene were the main compounds of Teucrium brevifolium oil; caryophyllene and 4-vinyl guaiacol predominated in Teucrium flavum. Carvacrol and caryophyllene oxide predominated in Teucrium montbretii ssp. heliotropiifolium, while carvacrol and caryophyllene were the most abundant components in Teucrium polium ssp. capitatum. The oil which most effectively inhibited LPS-induced NO production in macrophage cell line RAW 264.7 was that from T. brevifolium (IC50 = 7.1 μg/ml), followed by T. montbretii ssp. heliotropiifolium and T. polium ssp. capitatum (IC50 = 16.5 and 29.4 μg/ml, respectively). The in vitro cytotoxic assay on three human cancer cell lines showed that the most antiproliferative oils were those from T. polium ssp. capitatum and T. montbretii ssp. heliotropiifolium on CACO-2 cell lines (IC50 = 52.7 and 92.2 μg/ml, respectively). The T. brevifolium oil showed a selective cytotoxicity on COR-L23 while significant activity was exerted by T. polium oil on C32.  相似文献

Syzygium aqueum is a medicinal plant which is grown in tropical regions. In this study, the ethanolic extracts of S. aqueum leaf were investigated for its antihyperglycaemic activity. Our investigation revealed its effectiveness in inhibiting the carbohydrate hydrolysing enzymes, α-glucosidase (EC50 = 11 μg/ml) and α-amylase (EC50 = 8 μg/ml), at significant level than the commercial drug acarbose (EC50 = 28 μg/ml, α-glucosidase; EC50 = 12 μg/ml, α-amylase). In addition, the ethanolic leaf extracts were able to inhibit the key enzyme in the polyol pathway, aldose reductase (EC50 = 0.03 μg/ml) and prevent the AGEs formation by 89%. Six flavonoid compounds, 4-hydroxybenzaldehyde (1), myricetin-3-O-rhamnoside (2), europetin-3-O-rhamnoside (3), phloretin (4), myrigalone-G (5) and myrigalone-B (6), were isolated from the ethanolic leaf extracts. Compounds (2) and (3) showed high inhibitory activities, with EC50 values of 1.1 μM and 1.9 μM against α-glucosidase and EC50 values of 1.9 μM and 2.3 μM against α-amylase, respectively. These findings provide a strong rationale to establish S. aqueum’s capability as an antihyperglycaemic agent.  相似文献

The essential oils of Origanum ehrenbergii and O. syriacum collected in Lebanon were analysed by GC and GC–MS and evaluated for their anticholinesterase, NO production inhibitory activities, and antioxidant properties. O.ehrenbergi essential oil was characterised by the presence of 37 components, representing 94.9% of the total oil of which thymol (19%) and p-cymene (16.1%) were the main abundant compounds. Thirty-six compounds characterised the O.syriacum essential oil, representing 90.6% of the total oil. The most abundant components were thymol (24.7%) and carvacrol (17.6%). O. ehrenbergii demonstrated interesting scavenging effects on DPPH with an IC50 value of 0.99 μg/ml. In addition, both O. ehrenbergii and O. syriacum oils inhibited oxidation of linoleic acid after 30 min of incubation, as well as after 60 min of incubation with IC50 values of 42.1 and 33.6 μg/ml, and 46.9 and 58.9 μg/ml, respectively. Interestingly, O. ehrenbergii oil inhibited NO production in the murine monocytic macrophage cell line RAW 264.7 with an IC50 value of 66.4 μg/ml. Acetylcholinesterase (AChE) and butyrylcholinesterase (BChE) inhibition was assessed by modifications of the Ellman’s method. O. ehrenbergii exhibited a strong activity against both cholinesterases with IC50 values of 0.3 μg/ml. The data suggest that O. ehrenbergii and O. syriacum oils could be used as a valuable new flavour with functional properties for food or nutriceutical products with particular relevance to supplements for the elderly.  相似文献

Siberian pine (Pinus sibirica Du Tour) seeds, commonly known as cedar nuts, are ascribed a number of medicinal properties. In this study, we report the qualitative–quantitative composition, antioxidant activity and cell viability-related properties of a defatted aqueous-acetone-soluble P. sibirica seed extract. The total phenolic and total tannin contents were estimated at 266 ± 3.9 mg gallic acid/g and 115 ± 7.8 mg tannic acid/g, respectively. Reverse-phase chromatographic analysis of the crude extract indicated the presence of a chromatographic hump indicative of the presence of proanthocyanidins. After acid hydrolysis, the presence of hydroxylated benzoic and cinnamic acids, flavanones and flavan-3-ols was confirmed. After thiolysis, (+)-catechin was identified as more abundant than (−)-epicatechin, suggesting that this molecule was the main terminal unit of the proanthocyanidins within this extract. The extract demonstrated iron(III)-reductive (AscAE = 650 ± 5.10 μmol ascorbic acid/g) and iron(II) chelating (EC50 = 20.1 ± 2.1) activities and the ability to scavenge 1,1-diphenyl-2-picrylhydrazyl (IC50 = 257 ± 2.36 μg/ml) and hydroxyl (IC50 = 338 ± 6.49 μg/ml) free radicals. When the effects of P. sibirica extract were assessed in a tumourigenic SH-SY5Y neuroblastoma cell line, it was found that the cell viability was diminished in the presence of P. sibirica extract (0.2–1.0 mg/ml), as indicated by decreased membrane integrity (LDH assay) and mitochondrial metabolic activity (MTT assay), but the level of p53 protein was not changed (Western blot).  相似文献
