Box-Behnken响应曲面法优化药对生-炒酸枣仁中总黄酮及总皂苷的提取工艺 总被引:1,自引:0,他引:1
目的:通过响应曲面法优化药对生-炒酸枣仁中总黄酮及总皂苷的提取工艺。方法:采用乙醇回流法,以总黄酮及总皂苷得率为考察指标,在单因素实验基础上,利用Box-Behnken响应曲面法优化药对生-炒酸枣仁中总黄酮及总皂苷的最佳提取工艺。结果:优化后该药对总黄酮及总皂苷的最佳提取工艺为乙醇体积分数70%,液固比20mL·g~(-1),提取温度80℃,提取时间60min,总皂苷得率为(6.29±0.06)%,总黄酮得率为(2.48±0.03)%。结论:该提取工艺稳定可靠,可为药对生-炒酸枣仁的临床应用及开发利用提供参考。 相似文献

目的确定柴胡多糖超声提取的最佳条件。方法采用苯酚-硫酸法显色,紫外可见分光光度法(UV)在488 nm波长处测定柴胡多糖的吸光度,以多糖得率、多糖含量为指标,采用单因素和Box-Behnken响应面法优化实验条件。结果超声提取柴胡多糖的最佳工艺参数为:超声时间26min,料液比1∶21(g∶m L),超声次数为2次。结论最佳提取条件下的多糖含量为67.13%,综合评分98.28%,与预测值99.47%相近,说明该工艺优化结果合理可行,可用于柴胡多糖的提取。 相似文献

超声辅助提取工艺当今在食品工业中扮演着越来越重要的作用,它有着便捷高效、提取工艺成熟等优点。研究了从荔枝核中提取皂苷的超声波辅助提取优化工艺流程。根据单因素试验法对荔枝核皂苷的相关参数进行了实验优化,分别为:提取时间、固液比、乙醇浓度、提取温度和超声波功率。实验结果显示,当固液比、提取时间、超声功率、提取温度、乙醇浓度分别设定为1:60g/mL,30分钟,420W,45℃以及70%时可以得到最好的提取效果。在最佳工艺条件下,皂苷的最大产率为41.80mg/g,比传统溶剂提取产率36.80mg/g高13.59%。可以看到超声辅助提取可以很好地提高产率,缩短提取时间,为将来在工业上进一步利用超声提取技术提取荔枝核皂苷提供了很好的参考依据。 相似文献

目的:应用响应曲面法优化酸性染料比色法测定板蓝根总生物碱的测定方法。方法:在单因素考察实验的结果上,选取缓冲液pH值、缓冲溶液用量和酸性染料用量为3个显著因素进行Box-Benhnken中心组合设计,以板蓝根总生物碱提取率为衡量标准,通过响应曲面法对测定条件进行优化。结果:板蓝根-乙醇-乙酸乙酯(酸碱处理)条件下得出的总生物碱吸光度、含量以及固形物转移率最高,醋酸-醋酸钠缓冲液溶液pH=4.8,缓冲液用量5 mL,溴甲酚绿用量5 mL,三氯甲烷10 mL,萃取1次为板蓝根总生物碱含量测定的最优条件。结论:综合比较三种不同工艺对板蓝根总生物碱的提取效果,板蓝根-乙醇-乙酸乙酯(酸碱处理)效果最好。缓冲溶液的pH值和酸性染料的用量对测定板蓝根总生物碱的含量有显著的影响。 相似文献

以硫磺为原料,采用低温熔融法制备了具有较好高温稳定性的聚合硫粉体。通过单因素实验研究了稳定剂类型、反应温度、反应时间和稳定剂用量对聚合硫高温稳定性和单程转化率的影响。在单因素实验基础上采用Box-Behnken Design(BBD)实验设计方法,利用Design-Expert软件建立反应温度、反应时间、稳定剂用量3个因素的影响规律,通过方差分析回归建立数学模型,预测和验证最优生产工艺。结果表明,在反应温度为246 ℃、反应时间为48.23 min、稳定剂用量为0.28%条件下,制备的聚合硫显示出较好的高温稳定性。 相似文献

A human interferon beta expressed from a synthetic gene was produced in high cell density culture by recombinant Escherichia coli using an optimized linear feeding strategy. The optimal induction conditions to be determined consisted of inducer concentration and dry cell weight at the time of induction. For this purpose, the response surface methodology was applied. Under optimal conditions, the maximum interferon beta concentration and overall productivity of 2.2 g/l and 0.151 g/l h were obtained, respectively, as the highest amounts ever reported for this protein. Two optimal ranges of dry cell weight and IPTG concentration consisting of 50 g/l and 2.54 mM, and 70 g/l and 1.29 mM were predicted, respectively, at which maximum productivity was achieved. By using a novel feeding strategy with linear variation of specific growth rate during high cell density fermentation, the maximum biomass productivity of 5.037 g/l h was obtained in a defined medium during 16 h. Then, by applying the optimum induction conditions, we accomplished an increase in overall productivity by more than three-fold over the central point. This is the first report showing the high production of human interferon beta by a synthetic gene in a simple fed-batch high cell density culture of recombinant E. coli in a defined medium. 相似文献

通过响应面法对皂角苷浸提飞灰中Cu、Zn、Pb和Cd 4种重金属的实验条件进行优化,选取pH、皂角苷浓度、离子强度、温度、时间和固液比6个因素进行中心组合设计,使用design-expert8.0软件进行数据拟合,建立了Cu、Zn、Pb和Cd去除总量的工艺数学模型,通过方差分析得到6个因素对浸提液中4种重金属去除总量的主效应关系为: pH>固液比>离子强度>皂角苷浓度>温度>时间。对重金属去除总量数据进行design-expert8.0软件优化,得到皂角苷浓度为41.2g·L-1、时间为13.54 h、离子强度为0.64 mol·L-1、pH为2、固液比为1%和温度为23.4℃时,重金属总去除量达最大值,Cu、Zn、Pb和Cd的去除率分别达到55.12%、6.20%、17.80%和78.11%。 相似文献

探讨了超声波辅助技术提取蜂胶中的活性黄酮类化合物的工艺。实验以芦丁为标样,在单因素实验基础上,采用Box-Behnken实验设计方法,以提取温度、固液比、提取时间以及乙醇浓度为自变量,黄酮提取率为响应值,研究各自变量及其交互作用对黄酮提取率的影响。利用Design expert软件,建立了提取率与超声波处理各因素的二次多项式数学模型,获得了蜂胶中黄酮的最佳提取条件为:提取温度51.32℃,固液比1∶10,提取时间12.88 min,乙醇浓度88.86%,最佳黄酮提取率为52.39%。经过实验验证,黄酮提取率可达51.98%。 相似文献

Optimization of oil extraction from locust beans (Parkia biglobosa) using central composite rotatable design of RSM was carried out. Independent variables were roasting temperature (90, 100, 110, 120, 130°C) and roasting duration (5, 10 15, 20, 25 min), while the responses were oil yield (OY), free fatty acid (FFA), color (CO), stability, anisidine value (AV), and specific gravity (SG). Data obtained from this study were analyzed using ANOVA and regression analysis. OY, FFA, CO, oil stability, AV, and SG of the oil ranged between (19.0–22.5%), (0.65–3.1%), (37.9–199.9 mg/L), (3.6–6.4%), (11.4–59.9 mg/L), (0.7–0.9), respectively. Roasting conditions significantly influenced all dependent variables at p<0.05. Coefficients of determination (R2) of the generated models ranged from 0.49 to 0.98. Optimum roasting condition was 108°C for 10 min. This gave 22.3% OY, 1.7% FFA, 54.9 mg/L CO, 4.1% stability, 16.5 mg/L AV, and 0.9 SG. Desirability of 0.8 was obtained for optimum conditions. Practical applications: Generally, African locust bean is fermented to give condiment. It has not been used for commercial oil production despite its richness in oil, but soybean with a similar oil content of about 18–20% has been used both for condiment and commercial oil production. Research into the potential of locust bean as a source of edible oil is desirable. Roasting is among pre‐treatment methods employed in vegetable oil industry. Information on degree of influence of these parameters on quantity and quality of oil recovered oil from African locust benefits both entrepreneur and researchers. Also, the developed mathematical expression for OY, FFA, CO, oil stability, AV, and SG, as influenced by roasting temperature and duration is an effective tool in predicting these parameters.
