The influence of lactate/phosphate enhancement on meat color and lipid oxidation stability, tenderness, protein degradation, and protein aggregation of early postmortem beef muscles packaged in a high oxygen modified atmosphere packaging (HiOx-MAP; 80% O2, 20% CO2) were studied. At 24 hr postmortem, three bovine muscles (longissimus, semimembranosus, and adductor; n = 10, respectively) were enhanced (10% injection rate) with either lactate (2.5%)/phosphate (0.3%) solution or water, packaged in HiOx-MAP, stored 9 days at 1 °C, and then displayed for 7 days at 1 °C. The lactate/phosphate injection significantly improved color stability (higher a* values) of all three bovine muscles throughout display period. Accumulation of lipid oxidation determined by 2-thiobarbituric acid-reactive substances values was also decreased (P < 0.05) in the lactate/phosphate injection compared to the water treatment during storage and display periods. The objective tenderness values of longissimus and semimembranosus were also improved (P < 0.05) by the lactate/phosphate enhancement treatment compared to the water treatment based on star probe measurement. There were no significant differences found in desmin and troponin-T degradation, or oxidative cross-linking of myosin between treatments. The results suggest that lactate/phosphate enhancement has beneficial effects on color and lipid oxidation stability, and tenderness development of beef cuts under HiOx-MAP conditions.  相似文献

Pelvic and Achilles suspension methods for beef carcasses were compared for four gender-age groups (24 month bulls, 34 month bulls, heifers, and cows) and five muscles [M. longissimus dorsi (LD), M. semimembranosus (SM), M. adductor (AD), M. psoas major (PM), and M. gluteus medius (GM)]. Pelvic suspension increased muscle and sarcomere lengths in the SM, LD, GM, and AD muscles. The following effects were significant (p < 0.05). Peak force was reduced by pelvic suspension in the LD and GM of bulls-24 and bulls-34, but not heifers and cows. Furthermore, peak forces decreased for the SM after pelvic suspension in bulls-24, bulls-34, and heifers. For the AD, the only decrease in peak force was for bulls-34. Water-holding capacity increased and purge in vacuum bags decreased for pelvic suspension of all muscles except the PM. Although the effects of pelvic suspension varied somewhat between gender-age groups and muscles, this method of hanging carcasses merits industrial consideration because it improves muscle yields, tenderness, and reduces variation within muscles.  相似文献

The objective of this study was to identify proteins in bovine longissimus dorsi muscle that are related to tenderness. Two dimensional difference in gel electrophoresis (2D-DIGE) was used to compare the sarcoplasmic fractions from steaks that differed in star probe values at 14 days postmortem. The intensity of myosin light chain 1 (MLC1) was greater in the sarcoplasmic fraction prepared from steaks that had lower star probe values. It was hypothesized that μ-calpain catalyzes the release MLC1 into the sarcoplasmic fraction. Myofibrils from beef longissimus dorsi were purified and incubated with μ-calpain and the appropriate buffer controls. μ-Calpain was added at 1.23 μg (0.0875 U) of pure μ-calpain/mg myofibrillar protein. Incubations of one and 120 min had a greater abundance of MLC1 in the supernatants than the control incubations. As a consequence of μ-calpain proteolysis, MLC1 is rapidly released from the myofibril and is a potential indicator of proteolysis and improvement in beef tenderness.  相似文献

This study was carried out to compare the physicochemical and microbiological characteristics of beef and pork jerky, prepared from whole muscle of beef semimembranosus (BSM), pork semimembranosus (PSM), pork longissimus dorsi (PLD), and pork psoas major (PPM). The BSM and PSM jerky had higher moisture content, and PPM jerky had lower water activity than other jerky samples during 30 days of storage at 25 °C (P < 0.05). Pork jerky samples had higher lightness value than beef jerky, while PSM jerky had higher pH value than other jerky samples (P < 0.05). The shear force and TBARS values of PPM jerky were higher than those of other jerky samples (P < 0.05). Saturated fatty acid (SFA, %) was significantly higher in the BSM jerky than others, while unsaturated fatty acid (UFA, %) was significantly higher in the PSM and PLD than BSM and PPM jerky samples (P < 0.05). The PPM jerky showed a significant increase in UFA (%) during storage, and a significantly decrease in microbial count after storage of 30 days (P < 0.05).  相似文献

Beef steaks from longissimus lumborum, semimembranosus, and adductor muscles (n = 10; respectively) were cut at 24 h postmortem, randomly assigned to either high-oxygen modified atmosphere packaging (HiOx-MAP; 80% O2, 20% CO2) or vacuum (VAC), and displayed for 9 days at 1 °C. HiOx-MAP packaged beef steaks had a rapid increase in lipid oxidation and a decrease in color stability during display. The steaks in HiOx-MAP had significantly lower tenderness and juiciness scores, and higher off-flavor scores compared to steaks in VAC. HiOx-MAP condition did not affect the postmortem degradation of troponin-T or desmin. Furthermore, autolysis of μ-calpain was not influenced by packaging. SDS–PAGE, immunoblotting, and diagonal-PAGE revealed oxidative cross-linking of myosin heavy chain in meat packaged in HiOx-MAP. These results suggest that the HiOx-MAP system may negatively affect meat quality characteristics by inducing lipid and myoglobin oxidation and cross-linking/aggregation of myosin by protein oxidation.  相似文献

The effects of: a) applications of oil drip (from aged salted pork fat) onto dry-cured ham surface and b) application of a temperature of 35 °C for 4 days after 234 days of processing (HTST treatment) were evaluated. The oil application reduced moisture, proteolysis and white film in semimembranosus, microbial counts in adductor and the intensity of hollow extent, toasted flavour, adhesiveness, pastiness (in semimembranosus) and chewiness (in semimembranosus and biceps femoris) and increased the intensity of nutty flavour (in both muscles), aged flavour, hardness, fibrousness and overall liking (in semimembranosus). The objective of the present study was to evaluate the relationship between small heat shock proteins (sHSP) degradation and tenderness development of beef loins at different ultimate pH (pHu). A total of twelve loins (M. longissimus dorsi) from steers were obtained at 1 day post mortem. Shear force and proteolysis of each loin were analyzed at 1 and 28 days post mortem. The loins at intermediate pHu (5.8 to 6.0) showed more variation in tenderness compared to the loins at low pHu (< 5.8), where few samples were still tough (> 10 kgF) at 28 days. The intact sHSP20 was more pronounced (P < 0.05) in the intermediate pHu loin compared to the low pHu counterpart. Further, high correlations between the degradation of both sHSP and myofibrillar proteins were observed (e.g. r = 0.94; degraded sHSP27 and degraded desmin). The result of this study suggests that the extent of sHSP degradation could be an indicator of myofibrillar protein degradation and tenderness.  相似文献

Muscles (n = 18) were dissected from each side of 20 lamb carcasses to characterize physical, chemical, and histological traits. Muscles from the right sides were used to measure color values, expressible moisture, pH, total collagen content, and sarcomere length. Muscles from the left sides were aged for 7 d and used to determine Warner–Bratzler shear (WBS) force values. The M. adductor and M. semimembranosus had the darkest colored lean (lowest L values), whereas the M. latissimus dorsi and M. tensor fasciae latae had the lightest colored lean (highest L values). The M. triceps brachii, M. pectoralis profundus, and M. latissimus dorsi had among the lowest numerical expressible moistures whereas the M. adductor and the M. longissimus lumborum had among the highest. The M. infraspinatus was found to have the highest collagen content (9.0 mg/g) and the M. psoas major had the longest sarcomere length (3.1 μm). Of the muscles sampled, the M. serratus ventralis had one the lowest WBS values (21.8 N) and the M. semimembranosus the highest (42.6 N). On average, the largest muscles identified were the M. gluteobiceps, M. gluteus medius, M. longissimus lumborum, M. longissimus thoracis, and M. semimembranosus, which would likely be suitable in terms of physical dimensions for used in a variety of individual muscle applications. Other muscles, however, such as the M. teres major and M. tensor fasciae latae, may be too small in size and dimension to be used for individual application.  相似文献

A total of 180 pigs was slaughtered in the same slaughterhouse, but divided in six different trials distributed over Winter (December–March) and Summer (April–September). Meat quality measurements (pH, electrical conductivity, color and/or water-holding capacity) were carried out 30 min, 24 and/or 35 h after slaughter in three different muscles: M. gracilis, M. semimembranosus and M. longissimus dorsi. A tendency towards a higher proportion of PSE meat during Summer was found in the examined muscles. Moreover a higher protein, higher dry matter content, a lower water/protein ratio and a lower slicing yield were found for the cooked hams suggesting a higher PSE prevalence in the Summer. A lairage time between 2 and 4 h during Summer and less than 2 h during Winter was related to a lower proportion of PSE meat. The correlation coefficients between the individual meat quality variables were moderate, but showed the predictive power of the pH measured 24 h post-mortem in the M. gracilis for meat quality.  相似文献

The objective of this study was to develop a non-destructive method for classifying cooked-beef tenderness using hyperspectral imaging of optical scattering on fresh beef muscle tissue. A hyperspectral imaging system (λ = 922–1739 nm) was used to collect hyperspectral scattering images of the longissimus dorsi muscle (n = 472). A modified Lorentzian function was used to fit optical scattering profiles at each wavelength. After removing highly correlated parameters extracted from the Lorentzian function, principal component analysis was performed. Four principal component scores were used in a linear discriminant model to classify beef tenderness. In a validation data set (n = 118 samples), the model was able to successfully classify tough and tender samples with 83.3% and 75.0% accuracies, respectively. Presence of fat flecks did not have a significant effect on beef tenderness classification accuracy. The results demonstrate that hyperspectral imaging of optical scattering is a viable technology for beef tenderness classification.  相似文献

This study examined the relationship between collagen content as determined by hydroxyproline assay with other measures of connective tissue, shear force and tenderness in lamb muscle. Samples were taken from both m. longissimus lumborum (LL, loin) and the m. semimembranosus (SM, topside) of 99 lambs. Sensory tenderness and compression of the LL were not correlated to any measure of collagen or connective tissue, with one exception where compression was correlated (r = − 0.28; P < 0.05) to the percentage of connective tissue determined by imaging. Intramuscular fat (IMF) was linearly correlated (P < 0.05) to sensory tenderness and compression, such that a 1% increase in IMF increased the tenderness score by 2.3 ± 0.83 units and reduced compression by 0.73 ± 0.21 N. There was no correlation between SM shear force and collagen concentration. The data suggest that measurement of collagen concentration did not explain the variation in shear force and sensory tenderness observed in the meat from lambs.  相似文献

The objective was to determine the effect of muscle location on protein solubility and protein degradation in deep (DSM) and superficial (SSM) portion of beef semimembranosus. At 24 h postmortem, the semimembranosus was removed from beef carcasses (n = 10), packaged in high-oxygen modified atmosphere (80% O2 + 20% CO2), and displayed for 7 d at 1 °C. DSM had higher (P < 0.05) L*, a*, b*, and hue values than SSM throughout display. DSM had significantly higher protein denaturation and less protein concentration than SSM. Western blotting for μ-calpain autolysis revealed that DSM maintained more (P < 0.05) unautolyzed μ-calpain than SSM. This result coincided with less desmin and troponin-T degradation in samples from the DSM. These results confirm the hypothesis that increased protein denaturation in DSM results in minimal proteolysis by negatively affecting μ-calpain activation. This demonstrates a potential to alter progression of proteolysis and improvement in tenderness associated with postmortem storage.  相似文献

The effects of chilling method and moisture enhancement were examined for improving eating quality of semimembranosus (SM) and longissimus lumborum (LL) from 62 cull beef cows. Chilling method included hot boning muscles after 45 to 60 min postmortem or conventional chilling for 24 h. Moisture enhancement included 1) a non-injected control (CONT) or injection processing (10% of product weight) using 2) Sodium Tripolyphosphate/salt (Na/STP), 3) Sodium Citrate (NaCIT), 4) Calcium Ascorbate (CaASC), or 5) Citrus Juices (CITRUS). Chilling method by moisture enhancement treatment interactions (P < 0.09) were due to decreased hue, chroma and sarcomere length values in hot boned vs. conventionally chilled product (SM and LL) for CaASC vs. other moisture enhancement treatments. Chilling method by moisture enhancement treatment interactions (P < 0.05) were due to decreased shear force and increased tenderness in conventionally chilled vs. hot boned LL using CaASC vs. Na/STP. Moisture enhancement can improve tenderness of cull cow beef depending on combinations of chilling method and moisture enhancement treatments used.  相似文献

The objective of this study was to investigate the potential of a novel optical reflectance imaging method to predict beef tenderness. Two-dimensional (2D) optical reflectance in beef muscles induced by a point incident light was acquired. A set of five parameters were extracted from each reflectance image to describe quantitatively the reflectance profiles. Two parameters, q and B, were derived by numerically fitting the equi-intensity contours of the reflectance pattern. Two spatial gradients were calculated along the directions parallel and perpendicular to muscle fibers and total scattering intensity was obtained by excluding the specular reflectance. This method was applied to analyze 2D images of optical diffuse reflectance in 336 beef samples obtained from 14 steers in which large variations in tenderness were generated by altering animal genetics, suspension method and aging time as well as utilizing muscles varying in their inherent tenderness. Tenderness was evaluated using Warner–Bratzler shear force (WBSF). The effects of animal breed, muscle, types of suspension, and aging were investigated and results indicate that the scattering intensity measured at 1-d was correlated (R2 = 0.50 at λ = 720 nm) with 10-d WBSF in M. longissimus dorsi muscles; and the q parameters measured at 1-d was correlated (R2 = 0.46 at λ = 720 nm) with 10-d WBSF in M. psoas major muscles. These results show analyzing 2D reflectance images of meat surfaces provides valuable information regarding the physical characteristics of meat that are responsible for beef tenderness.  相似文献

This study evaluated the interaction between medium voltage electrical stimulation, SmartStretch™ stretching and ageing treatments on key meat quality traits of hot boned sheep m. semimembranosus. Medium voltage stimulation reduced initial pH (P < 0.001), but did not impact on other meat quality traits. There was a significant interaction between stretch treatment and ageing (P < 0.001) for shear force such that samples which were both stretched and aged were the most tender. Sarcomere length was significantly (P < 0.001) increased by SmartStretch™ treatment. Control (no stretching) resulted in greater (P < 0.05) cooking loss, but there was significantly less purge loss (P < 0.05). The ratio 630/580 nm and a* colour values at 0 and 5 days decreased at a significantly (P < 0.05) slower rate when SmartStretch™ was applied. Overall medium voltage stimulation did not inhibit the effectiveness of the SmartStretch™ treatment. The SmartStretch™ treatment provided significant improvement in tenderness and the potential to increase meat display time.  相似文献

The objective of this study was to use proteomics to identify alterations of proteins that are related to tenderness. The longissimus dorsi (LD) were removed from ten beef carcasses at 24 h postmortem, and the two with the highest (HSP; average kg of force = 6.57) and lowest star probe values (LSP; average kg of force = 3.75) at 14 days postmortem were identified. Two-dimensional PAGE was used to compare the sarcoplasmic fraction of the LD from HSP and LSP steaks. A series of spots identified as phosphoglucomutase 1 (PGM1) were identified. Only the most alkaline isoform was identified as being unphosphorylated. The least phosphorylated isoform (isoform 5) had a greater density of the total protein (P < 0.05) and phosphorylated protein (P < 0.05) in the samples from HSP steaks compared to the samples from LSP steaks. This study illustrates the importance of identifying posttranslational modifications of proteins in the search for biomarkers.  相似文献

Manipulation of growth rate and/or diet has been shown to affect protein turnover and may be used to improve beef quality. This trial was conducted to evaluate the effects of average daily gain (ADG) and diet on animal performance, collagen characteristics and beef quality of two different muscles; longissimus dorsi (LD) and semitendinosus (ST). Seventy six Hereford and Angus steers were assigned to three dietary management regimens for finishing: high grain diet based on corn (n = 28), alfalfa pasture (n = 22) and grass pasture (n = 26). Average daily gains were greater (P < 0.001) in Herefords vs. Angus and for corn- vs. pasture finished cattle. Overall, total collagen content was greater (P < 0.001) and the percentage of total collagen that was heat soluble was lower (P < 0.0001) for ST than for LD muscle. The lowest (P < 0.05) values for both total and heat soluble collagens were found in animals finished on corn. WBSF values for LD were greater (P < 0.01) in grass-fed vs. alfalfa- and grain-finished cattle while there was no difference in WBSF values for ST between grass- and corn-fed animals. No correlation between ADG and WBSF was observed for any muscle. ADG was not correlated with collagen solubility in ST, but was correlated (P < 0.05) with collagen solubility in LD. A key finding is that growth rate affected heat soluble collagen in the two muscles to a different extent. In conclusion, this study shows that different feeding strategies may not influence the tenderness of all muscles in a similar way.  相似文献

This study evaluates the effect of replacing fat with surimi-like material (SLM) made from the porcine longissimus dorsi muscle on the physicochemical and sensory characteristics of pork patties. Pork patties were produced with different levels of fat and SLM using a commercial method. Pork patties produced with 20% SLM had the lowest fat content (1.76%, P < 0.001), and had a higher cooking yield (87.41 vs. 78.57%, P < 0.05) and less shrinkage (4.01 vs. 9.02%, P < 0.001) than patties produced with 20% back-fat (control). Moreover, patties produced with SLM exhibited more acceptable organoleptic characteristics, including tenderness (P < 0.01) and overall acceptability (P < 0.001), than full-fat control patties. These results indicated that SLM cannot only be used as a fat replacer, but that it also improves the physicochemical and sensory characteristics of low-fat pork patties.  相似文献

This study investigated the effect of ultimate pH (pHu) in beef on the degradation of large structural proteins during refrigerated storage using SDS-PAGE. M. longissimus dorsi from bull carcasses were selected and classified into three groups: low pHu (≤ 5.79), intermediate pHu (5.80–6.19) and high pHu (≥ 6.2) muscles. Samples were then stored at − 1.5 °C for 1, 2, 7, 14, 21 and 28 days. Meat tenderness was measured at each aging time. Depending on meat pHu, different protein patterns and degradation rates of structural proteins were found. Rapid changes of large structural proteins took place within 48 h post mortem. Besides titin and nebulin, degradation of filamin was clearly revealed. Two more large protein bands corresponding to myosin family members also exhibited fast decline with storage time. It suggested that the fast degradation of these proteins is a key factor in the improvement of meat tenderness.  相似文献
