Microwave-assisted extraction (MAE) was investigated for extraction of phenolic compounds from tomato with maximised antioxidant activities using response surface methodology (RSM) coupled with a central composite design, and in vitro antioxidant assays (FRAP and ORAC). MAE was more efficient for greater antioxidant activities and higher total phenolic contents than solvent extraction. The optimal MAE processing parameters were 96.5 °C, 2.06 min, 66.2% ethanol for FRAP, and 96.5 °C, 1.66 min, 61.1% ethanol for ORAC. The models were successfully applied to 20 tomato cultivars, whose total phenolic contents (TPC) and indexes (TPI) were 489.30–997.45 mg gallic acid equivalent/100 g dry weight (DW) and 281.34–468.52 mg/100 g DW, respectively. Eight phenolic compounds were identified. Individual phenolics were 6.10–42.73 mg/100 g DW. The FRAP, but not the ORAC value showed good correlation with the TPC or TPI. The methodologies developed and the knowledge acquired in this study will provide useful information to tomato breeders and food processors.  相似文献

The antioxidant capacities and phenolic composition in six native, commercially grown, Australian herbs and spices were investigated. Tasmannia pepper leaf, followed by anise myrtle and lemon myrtle contained the highest levels of total phenolics (TP; 102.1; 55.9 and 31.4 mg gallic acid equivalents (GAE)/g dry weight (DW), respectively). Tasmannia pepper leaf exhibited the highest oxygen radical absorbance capacity (ORAC assay) followed by lemon myrtle and anise myrtle. Anise myrtle exhibited the highest total reducing capacity [TRC; Ferric Reducing Antioxidant Power (FRAP) assay], followed by Tasmannia pepper leaf and lemon myrtle. Australian bush tomato, with TP content of 12.4 ± 0.9 mg GAE/gDW and TRC of 206.2 μMol Fe+2/gDW, resembled the Chinese Barbary Wolfberry fruit. The TP content of Tasmannia pepper berry (16.86 mg GAE/gDW) was similar to that of black pepper, but it’s TRC was 25% lower. Cinnamic acids and flavonoids, tentatively identified by mass spectrometry, were identified as the main sources of antioxidant activities.  相似文献

Honey possesses antioxidant and antibacterial activities; however the compounds that are responsible for these activities are still under investigation. To determine whether there are common features/compounds that underlie the antioxidant and antibacterial activities, 20 Canadian honeys were analysed using the oxygen radical absorbance capacity (ORAC) and the broth micro-dilutions assay, respectively. Principal component analysis revealed the highest correlation between ORAC and Maillard reaction-like products (MRLP) (R = 0.938, p < 0.0001). In addition, the extremely significant correlations between ORAC, MRLPs, phenolic content and honey colour may suggest that these compounds represent the same chemical entity and exert their antioxidant activity while being part of a higher molecular weight structure. Moreover, the antioxidant activity and MRLPs were also the dominant variables contributing to the antibacterial activity against Escherichia coli (R = 0.737, p < 0.0001 and R = 0.65, p < 0.002, respectively) however no such correlation against Bacillus subtilis. In conclusion, MRLPs in honey appear to underlie the antioxidant and partially the antibacterial activity against E. coli.  相似文献

The effect of roasting on oxidative stability, antioxidant capacity and the content of antioxidant phytochemicals in Pistacia terebinthus oil was investigated. Oils were extracted from P. terebinthus fruits roasted at 180 °C for 0–40 min. Roasting was found to cause an increase in the passage of phenolic compounds to the oil whereas the level of tocopherols, lutein and β-carotene was decreased. Antioxidant capacity and oxidative stability of P. terebinthus oil increased with roasting. As an indicator of the presence of Maillard reaction products in oil, hydroxymethylfurfural (HMF) level and colour intensity was measured and found to increase with increasing roasting time. Fatty acid composition was not affected significantly by roasting.  相似文献

Four underutilized Georgia-grown fruit crops, namely loquat (Eriobotrya japonica), mayhaw (Crataegus sp.), fig (Ficus carica), and pawpaw (Asimina triloba), and their leaves were analysed for total polyphenols by Folin–Ciocalteau method, and antioxidant capacity by ferric-reducing antioxidant power (FRAP) and Trolox-equivalent antioxidant capacity (TEAC) assays. Organic acids and phenolic compounds were identified by RP-HPLC. For lipid profile, fruits were separated into two fractions – seed and fruit (i.e., without seed); lipid was extracted using the Folch method and analysed for fatty acids, phytosterols, tocopherols, and phospholipids. The major organic acid identified in all samples was malic acid (177–1918 mg/100 g FW). The predominant phenolic acids in all the fruits were gallic (1.5–6.4 mg/100 g FW) and ellagic (0.2–33.8 mg/100 g FW), and the most abundant flavonoid was catechin (12.2–37.8 mg/100 g FW). Total lipid content varied from 0.1% in mayhaw fruit to 21.5% in pawpaw seed. Linoleic acid was the predominant fatty acid in all of the samples (28.2–55.7%).  相似文献

The phenolic compounds in extracts from pressed olive cake were investigated. Free phenolic compounds were extracted from olive cake using methanol. To liberate bound phenolic compounds, the olive cake was subjected to basic and acidic hydrolysis followed by methanol extraction. The individual phenolic compounds and antioxidant activity of the extracts were determined. The highest total phenolic content and antioxidant activity were obtained using methanol extraction for 12 h at 70 °C. The RP-HPLC profiles for full-fat and defatted olive cake showed that protocatechuic acid, hydroxybenzoic acid, sinapic acid, p-coumaric acid, rutin and hesperidin were the predominant free phenolic compounds. Meanwhile, syringic acid, sinapic acid, caffeic acid and protocatechuic acid were the predominant bound phenolic acids. A positive correlation was observed between total phenolic content and antioxidant activity. The results indicated that most of the phenolic compounds in olive products were present in their free forms (75–90% of total phenolic content), while bound phenolic compounds were only a small proportion (10–25%) of total phenolic content.  相似文献

The effects of high intensity pulsed electric fields (HIPEF) (35 kV/cm with 4 μs bipolar pulses at 200 Hz for 800 or 1400 μs) or thermal (90 °C, 60 s) treatments over phenolic and carotenoid compounds of a fruit juice–soymilk (FJ–SM) beverage stored at 4 °C were evaluated and compared, having the untreated beverage as a reference. Coumaric acid, narirutin and hesperidin were the most abundant phenolic compounds in the FJ–SM beverage, while the main carotenoids were lutein, zeaxanthin and β-carotene. Immediately after HIPEF or heat processing, hesperidin content of the beverage showed a huge rise, resulting in a significant increase on the total phenolic concentration. Regarding carotenoid concentration, HIPEF or thermal treatment lead to a significant decrease; lutein, zeaxanthin and β-cryptoxanthin being the most affected compounds. In contrast, the content of some individual phenolics and carotenoids increased with time, while others tended to decrease or remained with no significant changes with regards to their initial values. Total phenolic concentration seemed to be highly stable during storage; while, total carotenoid content gradually diminished, irrespectively of the treatment applied. Overall, the changes observed in HIPEF treated FJ–SM beverage were less than those in the heat processed one. Hence, HIPEF is a feasible technology to obtain FJ–SM beverages with extended shelf-life and a similar profile of antioxidant compounds to freshly made beverages.  相似文献

Fatty acids (FA), phytosterols, tocopherols, phenolic compounds, total carotenoids and hydrophilic and lipophilic ORAC antioxidant capacities were evaluated in 16 cultivars of Sacha inchi (SI) seeds with the aim to valorise them and offer more information on the functional properties of SI seeds. A high α linolenic (α-Ln) fatty acid content was found in all cultivars (ω3, 12.8–16.0 g/100 g seed), followed by linoleic (L) fatty acid (ω6, 12.4–14.1 g/100 g seed). The ratio ω6/ω3 was within the 0.83–1.09 range. γ- and δ-tocopherols were the most important tocopherols, whereas the most representative phytosterols were β-sitosterol and stigmasterol. Contents of total phenolics, total carotenoids and hydrophilic and lipophilic antioxidant capacities ranged from 64.6 to 80 mg of gallic acid equivalent/100 g seed; from 0.07 to 0.09 mg of β-carotene equivalent/100 g of seed; from 4.3 to 7.3 and, from 1.0 to 2.8 μmol of Trolox equivalent/g of seed, respectively, among the evaluated SI cultivars. Results showed significant differences (p < 0.05) among the evaluated SI cultivars in the contents of ω3, ω6, antioxidant capacities and other evaluated phytochemicals. SI seeds should be considered as an important dietary source of health promoting phytochemicals.  相似文献

Size-exclusion chromatography (SEC) and activity-guided fractionation of honeys allowed the isolation of high molecular weight brown compounds, ranging in size from 66 to 235 kDa that exhibited peroxyl radical-scavenging activity. Their concentrations, antioxidant activity and degree of browning increased after heat-treatment of honeys, suggesting that they represent melanoidins. Chemical analysis of melanoidins demonstrated the presence of proteins, polyphenols and oligosaccharides. Heat-treatment caused an increased incorporation of phenolics into high molecular weight melanoidins and drastically decreased the protein content in these fractions with a concomitant appearance of high molecular weight protein–polyphenol complexes of reduced solubility. LC–ESI–MS demonstrated the presence of oligosaccharide moieties, supporting the postulated origin of melanoidins. The changes in the phenolic content of melanoidins from heated honeys were strongly correlated with their oxygen radical absorbance capacity (ORAC) values (R = 0.75, p < 0.0001), indicating that polyphenols contribute to the antioxidant activity of melanoidins. In summary, honey melanoidins are multi-component polymers consisting of protein–polyphenol–oligosaccharide complexes. A direct interaction between polyphenols and melanoidins resulted in a loss or gain of function for melanoidin antioxidant activity.  相似文献

Mango (Mangifera indica L.) is an economically important fruit throughout the world. ‘Ataulfo’ mango, a leading cultivar in Mexico, has the highest content of phenolic compounds among several commercial varieties of mango. However, the individual identification and antioxidant contribution of these phenols during ripening of mango fruit is unknown. Qualitative and quantitative analysis of the major phenolic compounds found in ‘Ataulfo’ mango fruit pulp was conducted in four stages of ripeness, using high-performance liquid chromatography coupled to mass spectrometry. The antioxidant contribution of each of the major phenolic compounds was calculated. The major compounds identified were chlorogenic acid (28–301 mg/100 g DW), gallic acid (94.6–98.7 mg/100 g DW), vanillic acid (16.9–24.4 mg/100 g DW), and protocatechuic acid (0.48–1.1 mg/100 g DW). The antioxidant contribution of the four phenolic acids increased during ripening. Gallic acid accounted for the highest contribution (39% maximum value), followed by chlorogenic acid (21% maximum value). This could indicate that these phenolic compounds may have an important role in the antioxidant metabolism in ‘Ataulfo’ mango fruit during ripening, and promoting health benefits to consumers.  相似文献

In this study, determination of phenolic acids as well as investigation of antioxidant activity of methanolic extracts from the fruits and leaves of Chenopodium album is described. Extracts were subjected to acidic hydrolysis in order to obtain total free phenolic acids. However, some of phenolic acids were identified and quantified by HPLC-DAD. The results were confirmed by LC-MS equipped with MS-ESI. In addition, Folin–Ciocalteu method was applied to determine the total phenolic contents. The antioxidant activity of C. album extracts was examined by using DPPH and hydroxyl radical-scavenging activity assays. Results revealed that the leaves extract exhibits better performance in antioxidant assays and in the higher total phenolic contents (3066 mg of GAE/100 g) when compared to fruits extract (1385 mg of GAE/100 g). From these results it has been revealed that the methanolic extracts of C. album from fruits and leaves have great potential as a source for natural health products.  相似文献

Many natural antioxidant extracts were reported to potentially reduce the acrylamide formation during Maillard reaction. In this work, the effect of pure antioxidant molecules on acrylamide net amount was investigated in an equimolar asparagine–glucose system (10 mM) during heating (200 °C). All experimentations were performed in the newly designed test cell. Cinnamic acid and six phenolic compounds (gallic, ferulic, coumaric, caffeic acids, catechin, and epicatechin), displaying different antioxidant capacity, were added at 5 mM to the model system. The results indicated no reduction effect of the tested compounds on acrylamide net amount. Moreover, in the case of ferulic acid a slight but significant promoting effect was observed. An increase of antioxidant activity was also noticed during heating of model solution spiked with caffeic acid but the acrylamide level was not affected.  相似文献

The effect of processing on functional compounds in buckwheat was investigated. Extractions of buckwheat flour were carried out before and after roasting or extrusion. Folin–Ciocalteu assays indicated that processing did not cause any change in total phenolic content in buckwheat flour. Roasted (200 °C, 10 min) dark buckwheat flour exhibited an increase in non-polar compounds as well as polar compounds whereas extrusion exhibited increase only in polar compounds. Antioxidant activity test (DPPH) showed that roasting at 200 °C for 10 min decreased the antioxidant activity slightly whereas extrusion (170 °C) did not cause any change. The results suggest that processing conditions can be optimized to retain the health promoting compounds in buckwheat products.  相似文献

The fruit quality characteristics, phenolic compounds and antioxidant capacities of 24 sweet cherry (Prunus avium L.) cultivars grown on the mountainsides of the Etna volcano (Sicily, Italy) were evaluated. High-performance liquid chromatographic methods were used to identify and quantify sugars, organic acids and phenolics. A total of seven phenolic compounds were characterised as hydroxycinnamic acid derivatives (neochlorogenic acid, p-coumaroylquinic acid and chlorogenic acid) and anthocyanins (cyanidin 3-glucoside, cyanidin 3-rutinoside, pelargonidin 3-rutinoside and peonidin 3-rutinoside). The total anthocyanin content ranged from 6.21 to 94.20 mg cyanidin 3-glucoside equivalents/100 g fresh weight (FW), while the total phenol content ranged from 84.96 to 162.21 mg gallic acid equivalents/100 g FW. The oxygen radical absorbance capacity (ORAC) assay indicated that fruit of all genotypes possessed considerable antioxidant activity. The high level of phenolic compounds and antioxidant capacity of some sweet cherry fruits implied that they might be sources of bioactive compounds that are relevant to human health.  相似文献

Maillard reaction products (MRPs) were prepared from casein–glucose by refluxing for 130 min at 102 °C and initial pH 12.0 without pH control to investigate the characteristics of casein–glucose Maillard reaction and the antioxidant activity difference among different fractions of MRPs. Browning and intermediate products increased, however, the pH of the system decreased with increase in the heating time. Free amino group content decreased 78% during first 10 min and did not change nearly thereafter. Amino acid analysis indicated that lysine and arginine decreased significantly, and casein was partially hydrolysed to peptides or free amino acid. High molecular weight compounds were dominant in the MRPs, determined by high performance gel-filtration chromatography. After ultrafiltration, antioxidant activity of each MRPs fraction was investigated by DPPH radical-scavenging activity, reducing power, Fe2+ chelating activity and lecithin oxidation assay. MRPs of different molecular weight exhibited distinctly different antioxidant activities.  相似文献

Juices from fifteen citrus varieties (seven mandarins, four sweet oranges, one lemon, one grapefruit, and two pummeloes) of China were investigated mainly on quality parameters, total carotenoid, phenolic compounds (total phenolics, flavanone glycosides (FGs), and phenolic acids), and antioxidant capacity (ferric reducing antioxidant power (FRAP) assay and 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay). Among the fifteen varieties, Bendizao had the highest content of total carotenoid (10.02 mg/L), Satsuma had the highest content of narirutin (288.12 mg/L), Yinzaocheng had the highest content of hesperidin (533.64 mg/L), and Huyou had the highest content of naringin (348.53 mg/L), neohesperidin (265.25 mg/L) and total FGs (746.08 mg/L). As for total phenolic acids, Liubencheng had the highest content (72.61 mg/L). Hybrid 439 achieved the highest AA content (631.25 mg/L), and the highest total phenolics (1555.49 mg/L) and the greatest inhibition of DPPH radical (61.62%). Hamlin had the highest ascorbic acid equivalent antioxidant capacity (AEAC: 899.31 mg/L) determined by FRAP assay. Correlation coefficients of AA, total phenolics (gallic acid equivalent), FRAP (AEAC), DPPH (I%), total FGs and total phenolic acids indicated that AA played a major role for the antioxidant capacity of citrus juices, and phenolics also played an important role, which may be mainly ascribed to FGs, whilst phenolic acids seemed to play a minimal role. Furthermore, Huyou and Hybrid 439 were considered two valuable varieties from the view of antioxidant capacity and nutrition.  相似文献

Myrtle (Myrtus communis L.) berries extracts were prepared with solvents at different polarity (water, ethanol, and ethyl acetate) and analysed using different in vitro tests in order to evaluate their antioxidant properties. Antiradical and total antioxidant activities were measured with DPPH and FRAP tests, respectively. Their ability to protect biological molecules was assessed using the cholesterol and LDL oxidation assays. In addition, phenolic compounds and unsaturated fatty acids composition was analysed by HPLC–DAD and HPLC–MS/MS. Ethanol and water extracts showed the highest amount of extracted compounds, but the highest antiradical and antioxidant activities were found in ethanol and ethyl acetate extracts. These extracts were also the ones with the highest content of phenolic compounds. In addition, our results showed a highly significant correlation between the amount of total phenols and antiradical (R2 = 0.9993) or antioxidant activities (R2 = 0.9985) in these extracts. HPLC–DAD and HPLC–MS analyses showed significant quantitative and qualitative differences among these three extracts. The ethyl acetate extract had the highest protective effect in assays of thermal (140 °C) cholesterol degradation and Cu2+-mediated LDL oxidation, inhibiting the reduction of polyunsaturated fatty acids and cholesterol, and the increase of their oxidative products. These results suggest that because of these properties, myrtle berries could be used in dietary supplements preparations or as food additives.  相似文献

Ultra-high pressure homogenisation (UHPH) is a recently developed technology and is still under study to evaluate its effect on different aspects of its application to food products. The aim of this research work was to evaluate the effect of UHPH treatments on quality characteristics of apple juice such as antioxidant capacity, polyphenol composition, vitamin C and provitamin A contents, in comparison with raw (R) and pasteurised (PA) apple juice. Several UHPH treatments that include combinations of pressure (100, 200 and 300 MPa) and inlet temperatures (4 and 20 °C) were assayed. Apple juice was pasteurised at 90 °C for 4 min. Antioxidant capacity was analysed using the oxygen radical antioxidant capacity (ORAC), 2,2-diphenyl-1-picrylhydrazyl (DPPH), trolox equivalent antioxidant capacity (TEAC), ferric reducing antioxidant power (FRAP) assay while total phenolic content was determined by the Folin–Ciocalteau assay. According to the FRAP and DPPH assays, UHPH processing did not change apple juice antioxidant capacity. However, significant differences were detected between samples analysed by TEAC and ORAC assays. In spite of these differences, high correlation values were found between the four antioxidant capacity assays, and also with total polyphenol content. The analysis and quantification of individual phenols by HPLC/DAD analytical technique reflects that UHPH-treatment prevented degradation of these compounds. Vitamin C concentrations did not change in UHPH treated samples, retaining the same value as in raw juice. However, significant losses were observed for provitamin A content, but lower than in PA samples.  相似文献

Free and total fluorescence compounds and color formation were measured in three different milk–cereal based infant foods stored at 25, 30 and 37 °C for 9 months to evaluate the advanced and final stages of the Maillard reaction. Milk–cereal infant foods containing honey (B) or fruits (C) had fluorescent values higher than sample (A) without them. This difference could be ascribed to the higher monosaccharide (fructose and/or glucose) content of (B) and (C), which could increase susceptibility to the Maillard reaction. However, for color increase (ΔE), no significant differences (p < 0.05) among the three types of samples were found. During the storage period, a gradual increase in fluorescence and color was observed, and statistically significant differences among the three temperatures studied were detected, the values being greater at 37 °C than at 30 °C and 25 °C.  相似文献

Laying hens are known to be able to “bio-convert” health-promoting components from the diet into eggs. Wheat and corn are important sources of antioxidant phenolics in the human diet. Given the fact that wheat and corn are major feed ingredients of laying hens in Canada, the objectives of this study were to characterise the presence of novel phenolic compounds in egg yolk and to determine the effect of cooking methods on their content and activity. The total phenolic content of yolk extracts was 3.83 and 3.49 μmol gallic acid equivalents/g dry yolk for wheat-based and corn-based yolks, respectively. Antioxidant properties of yolk extracts were measured by the ORAC, DPPH and ABTS assays. ORAC values were 151.1 ± 26.1 and 155.9 ± 9.5 μmol TE/g for wheat-based and corn-based yolks, respectively. All cooking methods significantly reduced (p < 0.05) the antioxidant values. Ferulic acid was detected in trace amounts and no other phenolic compounds were found. However, tryptophan and tyrosine were found to be two main contributors to the antioxidant property of egg yolk. The contents of total free amino acids were 10081.0 and 10009.5 μg/g in wheat-based and corn-based yolks, respectively; cooking methods were found to reduce significantly the amount of free amino acids.  相似文献
