Stingless bee honey samples from west Amazonian Ecuador were studied for their physiochemical, chemical and functional properties. Reducing sugars (44.9 ± 5.72 g/100 g), water (34.1 ± 4.34 g/100 g), free acidity (31.8 ± 4.05 meq/100 g), diastase activity (1.60 ± 0.20 u.d./g), hydroxymethylfurfural (15.0 ± 1.91 mg/kg), electrical conductivity (0.48 ± 0.06 mS/cm), ash (0.28 ± 0.04 g/100 g), colour (150 mm Pfund) were determined as physicochemical parameters. Melissopalynological analyses were processed evidencing pollen belonging to 14 plant families. Glucose (25.5 ± 3.41 g/100 g), fructose (25.2 ± 3.37 g/100 g) and sucrose (3.72 ± 0.49 g/100 g) contents were determined by HPLC, evidencing equal concentrations between fructose and glucose. Coumarins and flavonoids were determined by densitometric HPTLC: fraxin and bergamotin (0.065 ± 0.009; 0.035 ± 0.005 μg/g) among coumarins; luteolin (0.045 ± 0.006 μg/g), quercitrin (0.020 ± 0.003 μg/g), isoramnetin (0.015 ± 0.002 μg/g) among flavonoids. Among the vitamin E isomers, evaluated by HPLC, the occurrence of the only β-tocopherol (1.12 ± 0.15 μg/g) was noted. All these results were compared with those acquired for two multifloral Apis mellifera honeys. DPPH and β-carotene bleaching tests were performed, showing interesting values for Ecuadorian honey samples, higher than those shown by multifloral A. mellifera honeys (88.1 ± 11.1 DPPH inhibition%; 70.8 ± 8.90 β-carotene inhibition%). Antibacterial activity, against both Gram positive and Gram negative bacteria, revealed MIC values (10–50 μg/ml) always lower than those of A. mellifera honeys. Ecuadorian Meliponinae honey samples also showed anti-mutagenic activity assayed with Saccharomyces cerevisiae D7 strain, inhibiting back mutation over the entire range of concentrations.  相似文献

This paper presents the centesimal and mineral composition, fatty acid profile of the lipidic fraction, phenolic and anthocyanin contents, and antioxidant activity of Gaylussacia brasiliensis fruit. The results indicated the following composition: moisture (81.30%), lipids (0.62%), proteins (0.56%), carbohydrates (10.74%), dietary fiber (6.53%), and ash (0.25%). The main elements comprising the mineral composition were K, Mg, Ca, and Fe. The fatty acid composition was characterized by a high content of polyunsaturated fatty acids (62.2%) and a high PUFA/SFA ratio (2.92). The G. brasiliensis fruit contained considerable amounts of phenolics (492.87 mgAG/100 g) and anthocyanins (240.43 mg/100 g), which contribute to its high antioxidant activity. This study highlights the potential of this fruit as an important source of both nutritional and bioactive compounds available in the native Brazilian flora.  相似文献

Salvia sclareoides is an aromatic herb native to Portugal, of which phenolic content (Folin–Ciocalteau method), chemical profile (HPLC/DAD), antioxidant activity (DPPH, β-carotene/linoleic acid assays), acute toxicity (MTT method, adapted for non-adherent cells), genotoxicity (short-term chromosomal aberration assay) and prion binding properties were evaluated in the acetone, water, ethanol, methanol and n-butanol extracts. The latter presented the highest phenolic content and antioxidant activity (DPPH assay), and was the single one with the flavonoids (+)-catechin, kaempferol O-glucoside and quercetin. Vanillic acid was the major component of all extracts but gallic, gentisic, caffeic, syringic, coumaric and ferulic acids were also found in some extracts. Only the n-butanol extract had components binding to the cellular form of human prion protein detected by NMR which showed specificity for two regions of the folded domain and for the unstructured N-terminal region. Extracts were not cytotoxic nor genotoxic, reinforcing the potential of S. sclareoides for nutraceutical purposes.  相似文献

Leaves and products made from them are the major focus of research on bioactive compounds of mate (Ilex paraguariensis). However, studies on other parts of the plant, are scarce. The aim of this study was to determine the methylxanthines, phenolic compounds, and antioxidant activity of mate bark (residual biomass) and compare with those of mate leaves. The high antioxidant activity of mate bark and its high concentration of total polyphenols were apparent in both the aqueous and the methanolic extracts, the values of which were greater than those detected in the leaves. Of the phenolic acids identified, the levels of chlorogenic acid and 4,5-dicaffeoylquinic acid in the samples were significantly higher in the methanolic bark extract. With regard to methylxanthines, considerable concentrations were detected in the samples. The results obtained contribute to the viability of exploiting this residue, broadening the use of the mate plant.  相似文献

Olive trees play an important role in the Moroccan agro-economy, providing both employment and export revenue. However, the olive oil industry generates large amounts of wastes and wastewaters. The disposal of these polluting by-products is a significant environmental problem that needs an adequate solution. On one hand, the phytotoxic and antimicrobial effects of olive mill wastewaters are mainly due to their phenolic content. The hydrophilic character of the polyphenols results in the major proportion of natural phenols being separated into the water phase during the olive processing. On other hand, the health benefits arising from a diet containing olive oil have been attributed to its richness in phenolic compounds that act as natural antioxidants and are thought to contribute to the prevention of heart diseases and cancers. Olive mill wastewater (OMW) samples have been analysed in terms of their phenolic constituents and antioxidant activities. The total phenolic content, flavonoids, flavanols, and proanthocyanidins were determined. The antioxidant and radical scavenging activity of phenolic extracts and microfiltred samples was evaluated using different tests (iron(II) chelating activity, total antioxidant capacity, DPPH assays and lipid peroxidation test). The obtained results reveal the considerable antioxidant capacity of the OMW, that can be considered as an inexpensive potential source of high added value powerful natural antioxidants comparable to some synthetic antioxidants commonly used in the food industry.  相似文献

Phenolic compounds in oil palm fruit (E. guineensis) were extracted in soluble free (SFP), insoluble-bound (ISBP) and esterified (EFP) forms. The total phenolic content (TPC) of the oil palm fruit extracts was determined using the Folin–Ciocalteu method and found to range from 5.03 to 9.04 g/L per g of dried weight (DW). The antioxidant activities of oil palm phenolic extracts were analysed using free radical scavenging assays and results showed that oil palm phenolic extracts contained antioxidant activities in the order of ISBP > EFP > SFP. Eight different phenolic acids were identified and quantified using a simple reversed-phase high performance liquid chromatography (HPLC) with a diode array detector (DAD) and liquid chromatography/tandem mass spectrometry (LC/MS/MS). Ferulic, p-hydroxybenzoic and p-coumaric acid were the dominant phenolic acids found in oil palm fruit extracts and ranged from 55 to 376 μg/g of DW.  相似文献

Using the bleaching of the free radical scavenger 1,1-diphenyl-2-picrylhydrazyl (DPPH) as the guiding assay, a novel compound, 4-(3-(3,4-dihydroxyphenyl)acryloyl)-6-hydroxy-1-methoxy-1,2,3,4-tetrahydronaphthalene-2-carboxylic acid, was isolated from water-soluble extract of Canna edulis Ker with ten known compounds: rosmarinic acid, salvianolic acid B, ferulic acid, caffeic acid, 1-caffeoylquinic acid, 3-caffeoylquinic acid, 4-caffeoylquinic acid, 5-caffeoylquinic acid, salicylic acid and gallic acid. The structures were characterized by spectroscopic methods including extensive 2D-NMR techniques. Moreover, antioxidant activities of total extract and the new compound were determined through six different kinds of modes. In view of antioxidant activity confirmed, C. edulis extract and the new compound can be developed as natural food additives. In addition, the correlations between concentration and antioxidant activity and among the modes were analyzed. The results of DPPH and ABTS radical scavenging and FRAP assay exhibited high consistency. However, those for other modes showed difference to a certain extent.  相似文献

Phenolic compounds were separated from the leaves of Cyclocarya paliurus (Batal.) Ijinskaja and their bioactivities were evaluated through an in vitro PTP1B inhibitory assay. Bioassay-guided fractionation of the ethanol extract has resulted in the isolation of a naphthoquinone derivative, (1R, 2R, 4R)-1,2,4-trihydroxy-1,2,3,4-tetrahydro-naphthalene-1-O-β-d-glucopyranoside, named cyclonoside A, and a lactone, (4R, 5S, 6R)-8,9,10-trihydroxy-4-[3′,4′-dihydroxyphenyl]-1,6-dioxaspiro[4,5]decan-2-one, named cyclospirolide, along with 10 known phenolic compounds: quercetin-3-O-α-d-glucuronide, quercetin-3-O-β-d-glucuronide, myricetin-3-O-β-d-glucuronide, 1-caffeoylquinic acid, 3-caffeoylquinic acid, 4-caffeoylquinic acid, 5-caffeoylquinic acid, caffeic acid, 5-hydroxynaphthalene-1, 4-di-O-β-d-glucopyranoside and piceid. The structures of these compounds were established by means of spectroscopic methods including extensive 2D NMR techniques and chemical evidence. Among all the compounds, 1, 2, 4, 5, 10 and 11 showed strong inhibition against PTP1B, with IC50 values ranging from 1.922 ± 0.480 to 10.50 ± 2.67 μg/mL. The results suggested that the extract from this plant could be used as a potential source for functional food ingredient with anti-obesity.  相似文献

The effect of processing on functional compounds in buckwheat was investigated. Extractions of buckwheat flour were carried out before and after roasting or extrusion. Folin–Ciocalteu assays indicated that processing did not cause any change in total phenolic content in buckwheat flour. Roasted (200 °C, 10 min) dark buckwheat flour exhibited an increase in non-polar compounds as well as polar compounds whereas extrusion exhibited increase only in polar compounds. Antioxidant activity test (DPPH) showed that roasting at 200 °C for 10 min decreased the antioxidant activity slightly whereas extrusion (170 °C) did not cause any change. The results suggest that processing conditions can be optimized to retain the health promoting compounds in buckwheat products.  相似文献

Obtaining an extract with high antioxidant activity using environmentally friendly technologies and low-cost raw materials is of great interest. In the present work, a combined extraction process developed by our research group involving ultrasound treatment and agitated solvent extraction was evaluated. This method was compared in terms of yield, composition, and economical feasibility to traditional extraction methods, including ultrasound assisted, agitated bed and soxhlet extraction with ethanol (acidified or not). The proposed method maximizes the extraction of phenolic compounds with acceptable degradation of anthocyanin pigments from an unusual source: Brazilian jabuticaba (Myrciaria cauliflora) skins. The use of ultrasonic irradiation continuously supporting a main extraction process has demonstrated increased performance but implies in high consumption of energy and consequently, money. However, the procedure described in this paper appears to be a viable option because it uses shorter ultrasonic irradiation and results in high antioxidant activity extracts, and the anthocyanin profile corroborates literature data (cyanidin-3-glucoside and delphinidin-3-glucoside).  相似文献

Portulaca oleracea fractions obtained from the crude extract of the plant by reversed-phase separation were investigated for their antioxidant activities and phenolic compounds content. Five fractions were classified based on optical absorption between the wavelength range of 200–400 nm. Fraction 3 displayed higher absorption than the other fractions. The total amount of quantified phenolics in this fraction was found to be quite high compared to that of crude extract. Chlorogenic, caffeic, p-coumaric, ferulic and rosmarinic acids were the free phenolic acids, and quercetin and kaempferol were the free flavonoids detected in Fraction 3. IC50 value of crude extract was found to be 511.8 μg/ml whereas Fraction 3 exhibited an IC50 value of 154.1 μg/ml. TEAC of Fraction 3 was found to be almost four times higher than that of the crude extract. Fraction 3 was also found to show the highest lipid peroxidation inhibiting capacity, determined by TBARS assay.  相似文献

This paper focused on the assessment of antioxidant property and fatty acid composition of four Centaurea species. The antioxidant activity of its methanol extract was evaluated by several in vitro experiments including phosphomolybedum assay, DPPH assay, β-carotene/linoleic acid, ferric and cupric reducing power. Total phenolic and flavonoid contents were also evaluated. The methanol extract of Centaurea pulcherrima var. pulcherrima showed the superior free radical scavenging activity, linoleic acid inhibition capacity, reducing power and also had the highest total phenolic content. A significant relationship between antioxidant capacity and total phenolic components was found. The oils of Centaurea taxa were also analysed for fatty acid concentration by gas chromatography. The principal fatty acids in the species were palmitic acid (23.38–30.49%) and linoleic acid (20.19–29.93%). These findings suggest that the Centaurea species could be used as a potential source of new natural antioxidants and unsaturated fatty acids in food industry, cosmetics and pharmaceutical preparations.  相似文献

Aqueous and hydroalcoholic extracts from Verbena officinalis L. were obtained and characterised. The analysis by HPLC-DAD and LC-MS allowed the detection and identification of three iridoids, fifteen flavonoids and four phenolic acid derivatives. Four flavonoids, scutellarein 7-diglucuronide (9), scutellarein 7-glucuronide (13), pedalitin 6-galactoside (15) and scutellarein 7-glucoside (19) are reported for the first time from this plant. In addition, three new flavonoids have been isolated: scutellarein 7-O-(2-O-feruloyl)-diglucuronide (5), pedalitin 6-O-diglucuronide (6) and pedalitin 6-O-(2-O-feruloyl)-diglucuronide (13). To our knowledge, these flavonoids have not been reported as natural products. Both extracts showed significant antioxidant activity using three in vitro model systems and the results have been correlated with total phenolic and total flavonoid contents. The results have allowed establishing an important relation structure-activity and significant correlations have also been found between the mineral content and the flavonoids present in both extracts.  相似文献

This study was conducted in order to assess the effect of the inoculation of Glycine max (soybean) with Bradyrhizobium japonicum on the metabolite profile and antioxidant potential of its aerial parts. Extracts containing phenolic compounds and organic acids were analysed by HPLC–DAD and HPLC–UV, respectively. Volatiles profile was determined by HS-SPME/GC–IT-MS. The antioxidant activity of acidic and methanolic extracts was assessed against DPPH. Nodulation caused by B. japonicum led to an increase in the contents of phenolic compounds and organic acids. The same was observed for volatile compounds, with compounds like linalyl acetate, menthyl acetate and α-farnesene being detected only in inoculated plants. The phenolic extracts showed stronger antioxidant capacity than the organic acid extracts. In addition, extracts from plants nodulated with B. japonicum exhibited significantly higher antioxidant activity than control samples. These findings suggest that the inoculation with nodulating B. japonicum strains may be employed to manipulate the content of interesting metabolites in G. max aerial parts.  相似文献

To elucidate reasons for the observed variability in the antibacterial activity of honeys, we analysed a causal relationship between (a) honey floral sources and the activity and (b) the effect of honey storage on stability of compounds conferring this activity. Honeys from diverse floral sources were screened against Escherichia coli (ATCC 14948) and Bacillus subtilis (ATCC 6633) using the broth microdilution method. Among “active” honeys, 37% originated from buckwheat, 18% from clover and 12% from blueberry, indicating that these floral sources produced phytochemical(s) that inhibited bacterial growth. The stability of the putative phytochemical(s) was analysed in “active” honeys (MIC90 6.25% v/v) by measuring the activity every 3–6 months for a period of 1–3 years. A sharp decline in activity against both bacteria was observed in the first 3–6 months of storage. The decline coincided with major changes in chemical composition of honeys which included a significant change in colour (p < 0.0025), extremely significant change in concentration of UV-absorbing compounds (p < 0. 0001) and appearance of melanoidins. While these changes reduced E. coli sensitivity to honey, it rendered B. subtilis completely insensitive. Thus, the data indicates that the presence of phytochemical(s) conferring the antibacterial activity is sensitive to storage. The de-regulation of the antibacterial activity with the concomitant appearance of melanoidins suggests that the active phytochemical components might be sequestered into melanoidin aggregates, losing their function.  相似文献

Propolis extracts are currently used for the treatment of oronasal infections and as antioxidant agents. Ethanolic commercial Brazilian propolis extracts were assayed for their ability to inhibit the growth of Staphylococcus aureus and also for their ability to scavenge DPPH radicals. These activities were correlated with their total phenolic and flavonoid levels. In one group of extracts there was a strong linear relationship between total phenol contents and the measured activities, while in the other group this relationship was weaker. It was also found that flavonoid levels had a greater influence on the antioxidant activity of these extracts than on their antimicrobial profiles.  相似文献

The antimicrobial and antioxidant activities of crude ethanolic extract from Hippophaë rhamnoides L. (Elaeagnaceae) leaf, stem, root and seed, and their respective fractions, obtained by liquid-liquid extraction (LLE) using hexane (HF), ethyl acetate (EAF) and water (WF), were investigated. The crude extract was obtained by Pressurised Liquid Extraction (PLE), using ethanol at 100 bar and 60 °C. Antimicrobial activity was tested against food-borne and clinical microorganisms. Antioxidant activity was measured using the DPPH-radical scavenging and the ferric reducing antioxidant power (FRAP) assays. The phytochemical contents were examined by colorimetric methods. The results showed that crude extracts were active against Gram − and + strains, and that seed and root extracts were better radical scavengers than leaf and stem extracts. For all organs, the two activities tested were found to be higher in WF. These activities were correlated with the presence of phenolic compounds in active fractions. High Performance Thin Layer Chromatography (HPTLC) fingerprints confirmed presence of phenolic compounds in active extracts and fractions.  相似文献
