Four diets prepared, respectively, with 0%, 2%, 4% and 8% of a high-linoleic added fat were administered (76 days of treatment) to a sample of 112 pigs of four breeds (Landrace, Large White, Duroc and a crossbreed Landrace × Duroc). The effects of diet and breed on the evolution of the fatty acid composition of backfat were examined by taking biopsies. Over time, a continuous increase in stearic, palmitic and oleic acids throughout the pig’s life was observed. Oleic acid showed the smallest differences among the four diets at the end of the experiment, while stearic and palmitic acid showed higher differences according to the increase in the percentage of dietary fat. Stearic acid showed the highest rate of increase over time, according to the increasing intake of linoleic acid (diets 1–4). These increases were compensated by a decrease in linoleic acid, although this decrease tended to stabilize according to a higher percentage of added fat and also, for diet 4 (8% fat), an increase in linoleic acid was observed at the end of the experiment. Among the minor fatty acids, arachidonic acid showed a clear decrease over time, although higher levels at the end of the experiment were observed for diets including 4% and 8% of added fat, compared to the other two diets including lower amounts of linoleic acid. Moreover, a significant effect was observed for the factor breed. So, Duroc pigs showed the highest rate of deposit of linoleic acid and the lowest of stearic acid, while the other three breeds showed similar rates.  相似文献

The triacylglycerol (TAG) composition of edible Mediterranean molluscs (Eledone moschata, Sepia officinalis, Todarodes sagittatus) and crustacean (Penaeus kerathurus) was studied using a combination of preparative RP–HPLC and GC/MS. In S. officinalis and T. sagittatus mantle TAG, the main fatty acids were C16:0 and C18:0 while in E. moschata they were C18:1ω-9, C16:0, C20:5ω-3 and C22:6ω-3. In P. kerathurus muscle and cephalothorax TAG, the main fatty acids were C16:0, C18:0, C18:1ω-9, C20:4ω-6, C20:5ω-3, C22:6ω-3 and C16:0, C18:1ω-9, C20:4ω-6, C20:5ω-3, C22:6ω-3, respectively.  相似文献

Volatile organic acids in freeze-dried Cheonggukjang were quantified using a stable-isotope dilution assay (SIDA) according to the fermentation period. Five organic acids, acetic acid, propanoic acid, 2-methylpropanoic acid, butanoic acid, and 3-methylbutanoic acid, were identified using solid-phase microextraction (SPME) in conjunction with gas chromatography–mass spectrometry (GC–MS). The contents of volatile organic acids in Cheonggukjang were highly dependent on the fermentation period and they increased during fermentation. Moreover, the branched-chained organic acids (namely 2-methylpropanoic acid and 3-methylbutanoic acid) were formed earlier and were present at much higher contents than the corresponding straight-chained organic acids during Cheonggukjang fermentation.  相似文献

Farmed New Zealand King Salmon (Oncorhynchus tshawytscha) was prepared according to common consumer techniques; raw, poached, steamed, microwaved, pan fried (no added oil), oven baked (no added oil) and deep fried (in sunflower oil). The fatty acid profile was investigated to determine the optimal preparation techniques to achieve both optimal sensory and nutritional qualities, in particular the levels of long chain polyunsaturated omega-3 fatty acids. The modified Bligh and Dyer method was used for lipid extraction and the Hartman and Lago method for FAMES preparation. Fatty acid composition was determined by gas chromatography. There were moisture and lipid losses during cooking amongst the different methods. The fatty acid profile showed only minor differences between the methods apart from an increase in PUFA in the deep fried salmon due to linoleic acid uptake from the frying oil. In all the cooking methods the omega-3 fatty acids were well preserved. However, deep fried showed the lowest amounts of omega-3 fatty acids. As the results showed good preservation of omega-3 fatty acids regardless of cooking method, there may be possible “internal protection” of omega-3 fatty acids in King Salmon that warrants future research.  相似文献

Nine phenolic acids were identified and quantified by HPLC–MS in leaves and 10 in seeds of kale (black cabbage). The free, ester (methanol-soluble), glycoside and ester-bound (methanol-insoluble) phenolic acid contents of the leaves were 487, 532, 4989 and 6402 ng/g fresh weight, respectively. Ferulic and caffeic acids (total contents; 4269 and 4887 ng/g, respectively) were the most abundant. The seed contents of these fractions were 1993, 1477, 1231 and 4909 ng/g dry weight (DW), respectively, and sinapic acid was the most abundant (5037 ng/g DW). The fractions’ total phenolic contents, determined colorimetrically, were highly correlated with their DPPH scavenging capacity, and in antimicrobial activity assays, with nine test organisms representing a wide array of taxa, all of the fractions were effective against Staphylococcus aureus, Enterococcus faecalis, Bacillus subtilis and (most strongly) Moraxella catarrhalis. Antimicrobial and antioxidant activities of kale phenolics in free and conjugated forms are discussed.  相似文献

The oxygen radical absorbance capacity (ORAC), antiproliferative activities and mycosporine-like amino acid (MAA) profiles of methanol extracts from two grades of dulse harvested from locations varying in UV-exposure (west vs east coasts of Grand Manan Island, NB) were determined in the present study. MAAs confirmed by LC/MS in both grades 1 (low-UV) and 2 (high-UV) dulse were palythine, shinorine, asterina-330, palythinol and porphyra-334; usujirene was present only in grade 2 dulse. ORAC values of grade 1 and 2 dulse extracts were 36.42 and 38.78 μmol Trolox/g extract. B16-F1 murine skin melanoma cell proliferation was inhibited (p < .05) by 68.5% and 91.9% by grade 1 and 2 dulse extracts at 6.0 mg/mL. The antiproliferative efficacy of grade 2 dulse was greater (p < .05) than grade 1 from 0.375 to 6.0 mg/mL. MAA differences between the grade 1 and 2 dulse extracts likely influenced the antiproliferative efficacies, despite the similar ORAC values.  相似文献

Comparative analysis of FA, NL and sterol profiles in the Nephropsnorvegicus (langoustine), Palinurusvulgaris (lobster) and Penaeuskerathurus (shrimp) muscle and cephalothorax showed that C16:0, C16:1ω-7, C18:0, C18:1ω-9, C20:4ω-6, EPA and DHA were found to be their major FA. Highest EPA occurred in langoustine muscle TL, DHA in both shrimp tissues TL while C20:4ω-6 in lobster muscle and cephalothorax TL. Muscle and cephalothorax NL consisted mainly of sterols (42.5–54.4% and 13.7–43.1%) and triacylglycerols (35.4–45.8% and 44.6–59.4%). Cholesterol was the major sterol (70.90–98.58% and 97.10–98.31% of total sterols in muscle and cephalothorax respectively) followed by avenasterol (0.18–20.32% and 0.07–0.70%) and β-sitosterol (0.29–7.30% and 0.23–0.75%). Lower concentrations of brassicasterol, stigmasterol, Δ7-stigmasterol, campesterol and campestanol were also found. The examined crustaceans muscle (edible part) was found to be a good ω-3 PUFA source for the consumers, while the cephalothorax (which is usually discarded) could be used effectively as a source for ω-3 PUFA production.  相似文献

To investigate the nutritional value of the diatom Cyclotella cryptica (Reimann, Lewin, and Guillard) as an alternative feed for use in the aquaculture industry, the heterotrophic growth characteristics and resultant fatty acid profile of the microalga were studied when cultivated under a variety of controlled salinity and temperature conditions. In addition, the effects of pH on the growth characteristics were also studied. The maximum specific growth rate was affected by initial pH and cultivation temperature, but not by salinity. The optimal pH and temperature ranges for growth were 7.2 to 8.1 and 22.5 to 25.0 °C, respectively. Lipid accumulation and the fatty acid composition were also affected by cultivation temperature and salinity. The optimal temperature range and salinity level for lipid accumulation were 18.0 to 25.0 °C and 11.2 psu, respectively. In all cases the fatty acid distribution was similar, with the most abundant fatty acids being palmitic acid (16:0), palmitoleic acid (16:1 n-7), stearidonic acid (18:4 n-3, SDA), eicosapentaenoic acid (20:5 n-3, EPA), and decosahexaenoic acid (22:6 n-3, DHA).  相似文献

Endogenous tocopherols in extracted lipids from Jack beans (Canavalia gladiata DC.) were determined by high-performance liquid chromatography (HPLC), and were investigated in relation to the fatty acids (FA) distribution of triacylgycerols (TAG) and phospholipids (PL). The dominant tocopherols were (δ)-tocopherol (78.9–96.5 mg%) and (γ)-tocopherol (42.1–56.1 mg%) with much smaller amounts of (α)-tocopherol (1.1–1.3 mg%). The lipids of Jack beans comprised mainly TAG (34.6–38.6 wt.%) and PL (54.8–57.4 wt.%), and other components were also detected in minor proportions (0.3–3.8 wt.%). The PL components included phosphatidyl choline (46.2–48.7 wt.%), phosphatidyl inositol (23.4–29.6 wt.%) and phosphatidyl ethanolamine (18.5–21.2 wt.%). Comparison of these different beans showed, with a few exceptions, no significant differences (P > 0.05) in FA distribution. The FA distribution of TAG among the five beans was evident in the Jack beans: unsaturated FA (93.3–95.3 wt.%) were predominantly concentrated at the sn-2 position and saturated FA (33.6–34.4wt.%) primarily occupying the sn-1 position or sn-3 position. The results obtained from this work would provide useful information to both producers and consumers for manufacturing functional foods or beverages in Japan and elsewhere.  相似文献

To determine the impacts of finishing diet and tissue type and location on fatty acid composition and palatability of Jersey beef, twenty steers were assigned to a factorial treatment design with initial weight (Light vs. Heavy) and finishing diet (70 vs. 85% concentrate) as treatments. Ribeye steaks were collected for sensory evaluation. Muscle, seam and subcutaneous (s.c.) fat from steaks, kidney fat (KF) and omental fat (OMF) were collected for fatty acid analysis. Initial weight and finishing diet had little impact on beef palatability. The 85% concentrate decreased polyunsaturated fatty acids (PUFA) in muscle and increased trans fatty acids in all tissues (P < 0.05). The monounsaturated:saturated fatty acid ratio (MUFA:SFA) was highest in s.c. fat, intermediate in muscle and seam fat, and lowest in KF and OMF. The PUFA:SFA was highest in muscle, intermediate in s.c. and seam fat, and lowest in KF and OMF. Fatty acid composition differed greatly among tissues and the lower concentrate diet increased omega-3 and PUFA percentages in muscle.  相似文献

Aroma is an essential factor affecting the quality of tea (Camellia sinensis) products. While changes of volatile compounds during tea manufacturing have been intensively studied, the effect of environmental factors on volatile contents of fresh tea leaves has received less attention. We found that C. sinensis var. Yabukita kept in darkness by shading treatment for 3 weeks developed etiolated leaves with significantly increased levels of volatiles, especially volatile phenylpropanoids/benzenoids (VPBs). Upstream metabolites of VPBs, in particular shikimic acid, prephenic acid, and phenylpyruvic acid, showed lower levels in dark treated than in control leaves, whereas the contents of most amino acids including l-phenylalanine, a key precursor of VPBs, were significantly enhanced. In addition, analysis by ultra performance liquid chromatography-time of flight mass spectrometry, capillary electrophoresis–time of flight mass spectrometry, high performance liquid chromatography, and gas chromatography–mass spectrometry indicated that volatile and non-volatile metabolite profiles differed significantly between dark treated and untreated leaves.  相似文献

The chemical profiles of bioactive essential oil and extracts obtained by hydrodistillation (HD) and supercritical fluid extraction (SFE), respectively, from Lavandula viridis were compared. The SFE was performed at 40 °C and at extraction pressures of 12 or 18 MPa in two different separators. Evaluation of the essential oil and SFE extracts by GC–FID and GC–IT–MS revealed that oxygen-containing monoterpenes were the major constituents in both cases, but there were important differences between the chemical profiles produced by the different extraction techniques. More compounds were isolated by HD but higher yields were achieved by SFE. Camphor was the main component identified in the essential oil (31.59 ± 1.32%), and in extracts from the first (1.61 ± 0.34%) and second SFE separators (22.48 ± 1.49%) at 12 MPa. In contrast, the first separator SFE extract at 18 MPa (heavy compounds) was dominated by myrtenol (5.38 ± 2.04%) and camphor (4.81 ± 1.93%), whereas the second separator SFE extract (volatiles) was dominated by verbenone (13.97 ± 5.27%). The essential oil and heavy compound extracts from the first separator possessed antioxidant and anti-cholinesterase activities. Our data show that phytochemicals from the aerial parts of L. viridis could be developed as natural antioxidant and anti-cholinesterase drugs, with particular applications in the symptomatic treatment of Alzheimer’s disease.  相似文献

The antioxidant activity of chloroform and methanol extract of roots and stems of Rhubarb (Rheum ribes L.), which are used for medicinal purposes and also its fresh stems and petioles are consumed as vegetable, was studied. The antioxidant potential of both extracts of roots and stems were evaluated using different antioxidant tests, namely total antioxidant (lipid peroxidation inhibition activity), DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging, superoxide anion radical scavenging, ferric reducing power, and cupric reducing power (CUPRAC), and metal chelating activities. Total antioxidant activity was also measured according to the β-carotene bleaching method, and all four extracts exhibited stronger activity than known standards, namely butylated hydroxytoluene (BHT) and α-tocopherol. Particularly, higher activity was exhibited by roots with 93.1% and 84.1% inhibitions of chloroform and methanol extracts, while 82.2% and 82.0% inhibitions by stem extracts, respectively. However, both methanol extracts exhibited higher DPPH radical scavenging activity than the corresponding chloroform extracts, moreover, methanol extract of the stems showed better activity than BHT. In addition, both root extracts showed more potent superoxide anion radical scavenging activity than BHT, and comparable with well known radical scavenger l-ascorbic acid. Except chloroform extract of the roots, the other three extracts exhibited better metal chelating activity than quercetin. Also, total phenolic and flavonoid contents in both extracts of the roots and stems of R. ribes were determined as pyrocatechol and quercetin equivalents, respectively.  相似文献

The present study determined the chemical composition, fatty acid (FA) content and antioxidant capacity of meat from goats supplemented with Moringa oleifera leaves (MOL) or sunflower cake (SC) or grass hay (GH). The meat from goat supplemented with MOL had higher concentrations of total phenolic content (10.62 ± 0.27 mg tannic acid equivalent E/g). The MOL significantly scavenged 2,2′-azino-bis-3-ethylbenzothiazoline-6-sulfonic-acid (ABTS) radical to 93.51 ± 0.19% (93.51 ± 0.19%) and 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical to 58.95 ± 0.3% than other supplements. The antioxidative effect of MOL supplemented meat on catalase (CAT), reduced glutathione (GSH), superoxide dismutase (SOD) and lipid oxidation (LO) was significantly (P < 0.05) higher than other meat from goat feed on grass hay or those supplemented with sunflower seed cake. The present study indicated that the anti-oxidative potential of MOL may play a role in improving meat quality (chemical composition, colour and lipid stability).  相似文献

Nitraria tangutorun Bobr., a unique kind of fruit, widely spreads in the Qinghai-Tibet Plateau. In the present study, nine anthocyanins were identified in two variations (purple fruit and red fruit) of N. tangutorun by HPLC/DAD-ESI/MS. Cyanidin-3-O-(trans-p-coumaroyl)-diglucoside (215.76 ± 22.91 mg of Mv3G5G equivalent per 100 g of fresh weight) and pelargonidin-3-O-(p-coumaroyl)-diglucoside (5.13 ± 0.35 mg of Mv3G5G equivalent per 100 g of fresh weight) were the main anthocyanins in the purple and red fruits respectively. In addition, most of the anthocyanins were acylated by coumaric acid, and the rare anthocyanins that naturally presented a coumaric acid in both cis and trans configurations have been detected. Furthermore, the extract of the two variations showed significantly different antioxidant activity (p < 0.01) according to DPPH, ABTS and FRAP assay. Purple fruit possessed higher antioxidant activity than red fruit. There were significant correlations between antioxidant activity and both the total polyphenol content and anthocyanins content. This work is valuable for elucidation of anthocyanins composition in N. tangutorun and for further utilization as a functional food and medicine material.  相似文献

We investigated the metabolic profile and biological activities of the essential oil and polar extracts of Lavandula pedunculata subsp. lusitanica (Chaytor) Franco collected in south Portugal. Gas chromatography–mass spectrometry (GC–MS) analysis revealed that oxygen-containing monoterpenes was the principal group of compounds identified in the essential oil. Camphor (40.6%) and fenchone (38.0%) were found as the major constituents. High-performance liquid chromatography with diode array detection (HPLC–DAD) analysis allowed the identification of hydroxycinnamic acids (3-O-caffeoylquinic, 4-O-caffeoylquinic, 5-O-caffeoylquinic and rosmarinic acids) and flavones (luteolin and apigenin) in the polar extracts, with rosmarinic acid being the main compound in most of them. The bioactive compounds from L. pedunculata polar extracts were the most efficient free-radical scavengers, Fe2+ chelators and inhibitors of malondialdehyde production, while the essential oil was the most active against acetylcholinesterase. Our results reveal that the subspecies of L. pedunculata studied is a potential source of active metabolites with a positive effect on human health.  相似文献

Albumin, globulin and glutelin fractions were prepared from chickpea and oat seeds using sequential extractions. Molecular characteristics of individual protein fractions were investigated using sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) in combination with proteomic techniques. SDS-PAGE results revealed that chickpea albumin and globulin fractions (C-Ab and C-Gb) showed protein bands with molecular weights (MWs) related to subunits of legumin (11S globulin) and pea vicilin (7S globulin); oat protein fractions (O-Ab, O-Gb and O-Gt) showed most protein bands with MWs related to subunits of oat 12S globulin (avenalin). With proteomic analysis, eighteen tryptic peptides from chickpea globulin fraction showed sequence homology that corresponded to chickpea legumin α- and β-subunit (NCBI accession number: gi|6273402; theoretical mass 56,216 Da) while sixteen tryptic peptides from chickpea albumin fraction (C-Ab) were identified as chickpea provicilin precursor (NCBI accession number: gi|82173888; theoretical mass 51,390 Da); fifteen tryptic peptides from oat protein fractions were identified with origin from oat 12S seed storage globulin 1 (NCBI accession number: gi|134918; theoretical mass 58,508 Da). The identified tryptic peptide, ALIVPQNFAIAAK, was commonly found in chickpea glutelin fraction (C-Gt), rice glutelin fraction (R-Gt), and oat albumin, globulin and glutelin fractions (O-Ab, O-Gb and O-Gt).  相似文献

3,5-Dihydroxy-4-methoxybenzyl alcohol (DHMBA), an antioxidant isolated from the Pacific oyster (Crassostrea gigas), was studied in a cell-based fluorometric antioxidant assay using human hepatocyte-derived cells (C3A) and diphenyl-1-pyrenylphosphine (DPPP) as a fluorescent probe. In comparison with two hydrophilic antioxidants, DHMBA showed the stronger inhibition of DPPP-mediated fluorescence than chlorogenic acid and l-ascorbic acid: at a concentration of 320 μM of DPPP, the inhibition was 26.4 ± 2.6%, 11.1 ± 1.2%, and 0 ± 2.0% for DHMBA, chlorogenic acid, and l-ascorbic acid, respectively (mean ± SD, n = 4). Their relative oxygen radical absorbance capacities (ORAC) were dissociated with their cell-based antioxidant activities: 1.47 ± 0.40, 4.57 ± 0.30, and 0.53 ± 0.13 μmol TE/μmol for DHMBA, chlorogenic acid, and l-ascorbic acid, respectively (mean ± SD, n = 4). The amphiphilicity of DHMBA was better than chlorogenic acid and l-ascorbic acid might underlie this dissociation. Since the C3A cells are human hepatoma-derived cells, DHMBA might be useful in the prevention and treatment of liver diseases by involving an oxidation process.  相似文献
