Response surface methodology (RSM) was applied to optimize the oven drying conditions for lycopene content (Y1) and lipophilic antioxidant capacity (Y2) in decanter, a by-product of the pink guava puree industry. Two-factor central composite design was employed to determine the effects of two independent variables, namely temperature (X1: 50-80 °C) and drying time (X2: 4-6 h). Lycopene content and lipophilic antioxidant capacity were measured using high-performance liquid chromatography (HPLC) and the ABTS radicals scavenging assay, respectively. A β-carotene bleaching assay was also applied to measure the antioxidant activity. Response surface plots showed that an increase in temperature and time significantly reduced the response variables. The optimum oven conditions for drying of decanter with minimum lycopene degradation were 43.8 °C for 6.4 h, with a predicted lycopene content of 14 mg/100 g and antioxidant capacity of 21 μmol LE/100 g. To validate the optimized model, the experimental values were compared with the predicted values to check the adequacy of the model. The experimental values were found to be in agreement with those predicted, indicating the suitability of the model for optimizing the oven drying conditions for decanter.  相似文献

Psidium cattleianum (strawberry guava) is one of many underutilised edible fruits that grow wild in Jamaica, and could potentially be commercially exploited to yield health and economic benefits. In this study, the total phenolics, proximate contents, and antioxidant, anti-inflammatory, and antimicrobial activities of P. cattleianum and P. guajava (common guava), a well-known species, were compared. Strawberry guavas were found to be superior to common guavas in antioxidant and antimicrobial activities, total phenolics and vitamin C content. They also possessed relatively high fibre content (24.9%). The hexane and ethyl acetate extracts of strawberry guavas showed cyclooxygenase-2 enzyme inhibitory activities of 18.3% and 26.5%, respectively (250 μg/mL), indicating anti-inflammatory activity. The EtOAc and MeOH extracts of P. guajava showed 56.4% (COX-2) and 44.1% (COX-1) inhibitory activity, respectively. Additionally, nine compounds were isolated from strawberry guava fruits, some of which demonstrated anti-inflammatory activity. These results indicate that strawberry guavas are beneficial for health.  相似文献

The ability of brown seaweed extracts, Ascophyllum nodosum, Laminaria hyperborea, Pelvetia canaliculata, Fucus vesiculosus and Fucus serratus to protect against tert-butyl hydroperoxide (tert-BOOH) induced stress in Caco-2 cells was investigated. Oxidative stress was determined by measuring alteration in the enzymatic activity of catalase (CAT) and superoxide dismutases (SOD) and cellular levels of glutathione (GSH). L. hyperborea, P. canaliculata and F. serratus significantly protected against tert-BOOH induced SOD reduction but did not protect against the reduction in CAT activity or the increased cellular levels of GSH. The ability of F. serratus and F. vesiculosus to protect against H2O2 and tert-BOOH induced DNA damage was also assessed. The DNA protective effects of the two seaweed extracts was compared to those of three metal chelators; deferoxamine mesylate (DFO), 1,10-phenanthroline (o-phen) and 1,2-Bis(2-aminophenoxy)ethane-N,N,N′,N′-tetraacetic acid tetrakis (BAPTA-AM). F. serratus and F. vesiculosus significantly protected (P < 0.05) against H2O2 (50 μM) induced DNA damage but not tert-BOOH induced damage.  相似文献

The potential antioxidant activities of three phlorotannins (phloroglucinol, eckol and dieckol) purified from Ecklonia cava collected in Jeju Island were investigated to evaluate their potential value as the natural products for foods or cosmetic application. In this study, antioxidant activities were measured by electron spin resonance spectrometry (ESR) technique for scavenging effects of free radicals such as 1,1-diphenyl-2-picrylhydrazyl (DPPH), alkyl, hydroxyl (HO) and superoxide anion radical (O2 •−) and by comet assay for protecting effects against H2O2-mediated DNA damage. The results show that all the phlorotannins have the potential DPPH, alkyl, hydroxyl and superoxide radical scavenging activities. Especially, eckol samples scavenged around 93% of DPPH at 0.25, 0.5, 1 mg/mL of concentrations and were higher than the other phlorotannins, such as phloroglucinol and dieckol samples. Also, protecting effects of the phlorotannins against H2O2-mediated DNA damage increased with increased concentrations of the samples in the L5178 mouse T-cell lymphoma cell lines (L5178Y-R). In conclusion, these results suggest that the three phlorotannins purified from E. cava have the potential inhibitory effect on H2O2-mediated DNA damage and harmful free radicals and can be used as antioxidants in cosmetic, foods and drug industry.  相似文献

Sugarcane (Saccharum officinarum L.) juice is widely consumed by people of the tropics and subtropics. It has been used to cure jaundice and liver-related disorders in Indian systems of medicine. Its possible mechanism of action was examined in terms of antioxidant availability. The assays involved different levels of antioxidant action such as oxygen radical absorbance capacity (ORAC), radical scavenging abilities using 1,1-diphenyl-2-picryl hydrazyl (DPPH); 2,2′-azobis-3-ethyl benzthiazoline-6-sulfonic acid (ABTS); ferric reducing antioxidant power (FRAP); and protection of membranes examined by inhibition of lipid peroxidation. In addition, the content of phenols and total flavonoids were measured. The aqueous extracts of three varieties of sugarcane were studied. These varieties showed good antioxidant properties and were also able to protect against radiation induced DNA damage in pBR322 plasmid DNA and Escherishia coli cultures. In conclusion, the study reveals that the ability of sugarcane juice to scavenge free radicals, reduce iron complex and inhibit lipid peroxidation, may explain possible mechanisms by which sugarcane juice exhibits its beneficial effects in relation to its reported health benefits.  相似文献

The aim of the study was to investigate phenolic composition, antioxidative, protective and cytotoxic effects of Pleurotus eryngii and Auricularia auricula-judae. Analysis of phenolic compounds in these edible mushrooms species has been carried out by high-performance liquid chromatography (HPLC). Protective effect of these mushrooms on H2O2 induced oxidative cell damage was determined by using MTT (3-(4,5-Dimethylthiazole-2-yl)-2,5-diphenyltetrazolium bromide, a yellow tetrazole) assay. Antioxidant activities of the mushrooms extracts were evaluated by using complementary in vitro assays. In addition, the measurement of total antioxidant compounds in the extracts was carried out. All the extracts exhibited protective effect against H2O2 induced oxidative cell damage but the highest activity was observed for A. auricula-judae aqueous extract (89.5 ± 1.8% cell viability at 0.1 mg/ml). P. eryngii methanolic extract showed the highest ferrous iron chelating ability (IC50 = 0.42 ± 0.03 mg/ml). A. auricula-judae extracts (at concentration of 0.025–0.100 mg/ml) were not toxic to baby hamster kidney fibroblast cell line (BHK 21). These results suggest that these mushrooms may be used as a potential source of natural antioxidants for food supplementation or in the development of nutraceuticals.  相似文献

About 90–98% of native lycopene exists in the all-E form, but 79–88% of the lycopene found in the human body are Z-isomeric forms. Thermally-induced geometrical isomerisation of lycopene occurred within 24 h of refluxing in ethyl acetate and the proportion of Z-isomers increased from 5.8% to 49.9%. Accordingly, the concentration of lycopene required to double quinone reductase (QR) activity in Hepa 1c1c7 cells decreased from >100 to ∼22 μg/mL following thermo-isomerisation, while cell viability was retained at >87% at levels up to 50 μg/mL. The inhibition of nitric oxide (NO) production in activated RAW 264.7 macrophages was 50% at ∼100 μg/mL thermo-isomerised lycopene and increased to >80% when the concentration in the medium was increased to 500 μg/mL. No significant inhibition of NO evolution by macrophages occurred with native (∼94% all-E) lycopene. Both QR induction and NO inhibition bioassays revealed that the structural changes evoked by thermo-isomerisation were accompanied by enhanced biological functionality.  相似文献

Salvia officinalis (SO) and Thymus vulgaris (TV) are medicinal plants well known for their curative powers. However, the molecular mechanisms responsible for these abilities of sage and thyme have not been fully understood yet. In this study we investigated the composition and the quantitative estimation of plant extracts, the protective effects of plant extracts against hydrogen peroxide- and 2,3-dimethoxy-1,4-naphthoquinone-induced DNA damage, and levels of enzymatic and non-enzymatic antioxidants (superoxide dismutase, glutathione peroxidase, glutathione) in human HepG2 cells. To measure antioxidative activity of plant extracts we used three assays: 1,1-diphenyl-2-picrylhydrazyl (DPPH), ferric reducing antioxidant power (FRAP) and 2,2′-azinobis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS). The results showed that the oxidant-induced DNA lesions were significantly reduced in cells pre-treated with the plant extracts studied. The observed DNA-protective activity could be explained by both elevation of GPx activity in cells pre-treated with SO and TV and antioxidant activity of SO and TV.  相似文献

The total phenolic content (TPC), total antioxidant status (TAS), free radical scavenging capacity, inhibition of low density lipoprotein (LDL) cholesterol and metal chelating capacity of extracts of whole black and whole white sesame seeds and their hull fractions in 80% aqueous ethanol were investigated. The TPC of whole black sesame seeds and hull extracts were 29.9 ± 0.6 and 146.6 ± 0.6 mg catechin equivalents/g crude ethanolic extract, respectively. The corresponding values for white sesame were 10.6 ± 1.6 and 29.7 ± 0.9 mg catechin equivalents/g crude ethanolic extract. The TAS as determined by Trolox equivalent antioxidant capacity assay and expressed as Trolox equivalents was highest for black sesame hulls (65.9 ± 1.7) while white seeds showed the lowest (4.4 ± 0.6). Free radical scavenging capacity of sesame extracts (5–40 μg/mL) was measured using 2,2-diphenyl-1-picrylhydrazyl radical. The highest scavenging capacity was obtained at 40 μg/mL and was 94.9 ± 0.8, 25.1 ± 0.4, 14.4 ± 0.9 and 2.5 ± 0.4 for black sesame hulls, black sesame seeds, white sesame hulls and white sesame seeds, respectively. Inhibition of LDL oxidation at 100 ppm level was highest for black sesame hulls (96.7%) followed by those for white sesame hulls (84.6%), black sesame (78.4%) and white sesame seeds (57.3%). Sesame products displayed good ferrous ion chelating capacities, which ranged from 12% to 46% and 17% to 62% at 50 and 100 ppm levels, respectively. Results demonstrated considerable antioxidant activity of sesame products tested especially black sesame hulls.  相似文献

Cowpeas contain phenolic compounds with potential health benefits. The effect of simulated gastrointestinal digestion on phenolic composition of cooked cowpeas and the ability of the digests to inhibit radical-induced DNA damage was determined. A red and a cream-coloured cowpea type were used. The phenolic composition of acetone extracts and enzyme digests of cooked cowpeas was determined using UPLC-MS. Compounds such as p-hydroxybenzoic acid, p-coumaric acid, coumaroylaldaric acid and feruloylaldaric acid were present in the acetone extracts of the cooked cowpeas but were not detected in the enzyme digests. Glycosides of quercetin and myricetin decreased upon in vitro gastrointestinal digestion of cooked cowpeas whereas flavan-3-ols were hardly present except catechin glucoside. The enzyme digest of the red cowpea type was about thrice as effective as that of the cream cowpea type in protecting DNA from oxidative damage. The observation that enzyme digests of cooked cowpeas inhibited radical-induced DNA damage suggests that cowpea phenolics retain some radical scavenging activity after gastrointestinal digestion.  相似文献

Sphallerocarpus gracilis roots harvested at two different times (T1: 12-10-2009 and T2: 15-04-2010) were analysed for their main nutrients, phenolics, antioxidant activity, DNA damage protective effect and microstructure. Results revealed that higher amounts of starch were detected in T1 S. gracilis root, whereas higher levels of reducing sugar, protein, amino acids and phenolic compounds were determined in T2 S. gracilis root. The antioxidant activity was assessed by DPPH, FRAP and TEAC assay and T2 S. gracilis root was found to possess higher antioxidant activity. Also, results of the correlation analysis indicated that caffeic, p-coumaric and gallic acids contributed substantially to the antioxidant property and there was significant correlation between DPPH radicals scavenging capacity and FRAP and TEAC. The highest and lowest DNA damage protective effects were found of free phenolic extracts from T2 and T1 S. gracilis roots, respectively. The microstructure of S. gracilis root varied greatly with the harvest time.  相似文献

The potential therapeutic effects of Costa Rican guava (Psidium friedrichsthalianum) extracts for chronic obstructive pulmonary disease were examined. The ethyl acetate fraction displayed the highest antioxidant activity, as compared to the hexane, chloroform, and n-butanol fractions, as well as the crude extract. This fraction was evaluated for its anti-inflammatory activity response relationship against interleukin-8 (IL-8) and inhibition of matrix metalloproteinase-1 (MMP-1) expression before and after treatment with cigarette smoke. The ethyl acetate fraction exhibited inhibitory activity against IL-8 production and MMP-1 expression, showing the most potent inhibitory activities in both assays at 100 μg/mL, and nine compounds (19) were found. Phenolic compounds 1-O-trans-cinnamoyl-β-d-glucopyranose (2), ellagic acid (3), myricetin (4), quercitrin (7), and quercetin (9) were identified using standard compounds or literature reports from related species. Compounds 1, 5, 6, and 8 were tentatively identified as 1,5-dimethyl citrate (1), sinapic aldehyde 4-O-β-d-glucopyranose (5), 3,3′,4-tri-O-methylellagic acid-4′-O-d-glucopyranoside (6), and 1,3-O-diferuloylglycerol (8), All nine compounds are reported for the first time in Costa Rican guava.  相似文献

Seven new (17) and three known (810) abietane diterpenoids were isolated from the methanolic extract of the aerial parts of Isodon lophanthoides var. graciliflorus (Lamiaceae), a folk Chinese medicine and an herb for functional beverages. They were identified as 16-acetoxylsugiol (1), graciliflorin E (2), graciliflorin F (3), 15-O-methylgraciliflorin F (4), 15-hydroxy-20-deoxocarnosol (5), 3β-hydroxysempervirol (6), 15-hydroxy-1-oxosalvibretol (7), abieta-8,11,13-triene-14,19-diol (8), 6,12,15-trihydroxy-5,8,11,13-abietatetraen-7-one (9), and 3α-hinokiol (10) based on the spectroscopic data including COSY (correlated spectroscopy), HMBC (heteronuclear multiple bond correlation), and HR-ESI-MS (high-resolution electrospray ionization mass spectrometry). All the compounds except 10 were obtained from I. lophanthoides for the first time. Compounds 1, 2, 5, 6, 8, and 9 exhibited in vitro cytotoxicity against A549 (human lung adenocarcinoma), MCF-7 (human breast adenocarcinoma), and HeLa (human cervical carcinoma) cell lines with the IC50 values of 1.79–52.67 μM.  相似文献

The composition of the essential oil from Dictamnus dasycarpus was analysed by GC–MS; 47 constituents (88.9% of the total oil) were identified. The main compounds were syn-7-hydroxy-7-anisylnorbornene (29.4%), pregeijerene (15.5%) and geijerene (11.4%). The antimicrobial activity of the essential oil was evaluated against nine microorganisms using disc diffusion and broth microdilution methods. The oil showed the strongest bactericidal activity against Staphylococcus aureus ATCC 25923 and methicillin-resistant S. aureus. The in vitro cytotoxicity of the oil on six human cancer cell lines was also examined. The cytotoxicity of the oil on three human breast cancer cell lines was significantly stronger than on other cell lines.  相似文献

Portulaca oleracea fractions obtained from the crude extract of the plant by reversed-phase separation were investigated for their antioxidant activities and phenolic compounds content. Five fractions were classified based on optical absorption between the wavelength range of 200–400 nm. Fraction 3 displayed higher absorption than the other fractions. The total amount of quantified phenolics in this fraction was found to be quite high compared to that of crude extract. Chlorogenic, caffeic, p-coumaric, ferulic and rosmarinic acids were the free phenolic acids, and quercetin and kaempferol were the free flavonoids detected in Fraction 3. IC50 value of crude extract was found to be 511.8 μg/ml whereas Fraction 3 exhibited an IC50 value of 154.1 μg/ml. TEAC of Fraction 3 was found to be almost four times higher than that of the crude extract. Fraction 3 was also found to show the highest lipid peroxidation inhibiting capacity, determined by TBARS assay.  相似文献

Two new sesquiterpene glucosides, litchiosides A (1) and B (2), were obtained along with pumilaside A (3) and funingensin A (4) from lychee (Litchi chinensis Sonn.) seed. Their structures were elucidated on the basis of spectroscopic and chemical evidence. The in vitro antitumour activity of compounds 14 against A549, LAC, Hela, and Hep-G2 cell lines was determined using the MTT colourimetric assay. Pumilaside A (3) exhibited significant activity against all the tested cell lines with IC50 values of 0.012–6.29 μM, and funingensin A (4) showed moderate activity against Hep-G2 cells (IC50 = 39.27 μM), while litchiosides A (1) and B (2) were found to be inactive.  相似文献

The thermal stability and catalytic activity of endoglucanase (EngD) from mesophilic Clostridium cellulovorans were improved by evolutionary molecular engineering. Thermostable mutants were isolated after staggered extension process (StEP) with celE from thermophilic Clostridium thermocellum performed to conduct family shuffling and overlay screening of the resultant mutant library. The relative activity of the best-evolved clone has been improved of about 2 times higher at 50 °C and showed a higher kcat/Km value than its engD parental clone. We determined that these variants had two amino acid substitutions (L157N, Q158E) and confirmed their effects by substituting these amino acids in the parental gene by site-directed mutagenesis. These substitutions resulted in an increase in hydrophilic or charged residues. Our results demonstrate that in vitro recombination is an effective approach to improve the thermostability and enzymatic activity of a mesophilic enzyme.  相似文献

The peduncles of Hovenia dulcis, containing abundant nutrients and having a taste like a combination of raisin, clove, cinnamon and sugar, have been consumed as fruits and used as traditional herbal medicine for a long time in China. Up to date, little information is available about the polysaccharides from peduncles of H. dulcis (HDPS) and their potential bioactivity. In this study, three purified fractions were prepared by sequential purification of crude HDPS through ion-exchange chromatography and gel-filtration chromatography. The three fractions of HDPS-1, HDPS-2 and HDPS-3 were found to be homogeneous heteropolysaccharides mainly composed of rhamnose, arabinose, galactose and galacturonic acid with an average molecular weight of 235, 70 and 53 kDa, respectively. HDPS-3 was quite different from HDPS-1 and HDPS-2, as it contained much higher content of galacturonic acid (40.5%). In vitro immunostimulatory activity evaluation revealed that all the three fractions could significantly stimulate the proliferation of splenocytes and enhance phagocytosis, nitric oxide production and acid phosphatase activity of peritoneal macrophages, which suggested that HDPS had a potent immunostimulatory activity and could be explored as a potential natural immunomodulatory agent.  相似文献
