Agaricus bisporus, Lentinula edodes, Pleurotus ostreatus, Pleurotus eryngii and Grifola frondosa were analyzed for antioxidant capacity, as measured by oxygen radical absorbance capacity (lipophilic and hydrophilic) (ORACtotal), hydroxyl radical averting capacity (HORAC), peroxynitrite radical averting capacity (NORAC), superoxide radical averting capacity (SORAC) assays, Folin-Ciocalteu reagent and ergothioneine (ERG) content. ORACtotalvalues ranged from 39 to 138 μmol of Trolox equivalents (TE)/g dry weight (dw). HORAC values ranged from 3.0 to 13.6 μmol of caffeic acid equivalents/g dw. NORAC values ranged from 2.0 to 9.0 μmol TE/g dw. SORAC values ranged from 0.37 to 2.6 kunit superoxide dismutate equivalents/g dw. Polyphenols ranged from 4.2 to 10.6 mg gallic acid equivalents/g dw. A. bisporus mushrooms, especially portabellas, had higher antioxidant capacity relative to the specialty mushrooms tested. ERG ranged from 0.21–2.6 mg/g dw with L. edodes, P. ostreatus, G. frondosa containing a statistically significant greater amount compared to A. bisporus. A good correlation was found between ORACtotal and polyphenols (R2 = 0.86).  相似文献

A microwave-assisted extraction system was used to extract phenolic antioxidants from peanut skins. The effects of microwave power (10%, 50%, 90% nominal), irradiation time (30, 90, 150 s) and sample mass (1.5, 2.5, 3.5 g) on total phenolic content (TPC), ORAC (oxygen radical absorbance capacity) level and resveratrol content of peanut skin extracts (PSE) were investigated. Peanut skins were extracted with 37.5 ml of 30% ethanol (EtOH) in water. A response surface method was used to estimate optimum extraction conditions, based on TPC, ORAC level and resveratrol content. The maximum predicted TPC, under the optimised conditions (90% microwave power, 30 s irradiation time and 1.5 g skins), was 143.6 mg gallic acid equivalent (GAE)/g skins. The highest ORAC value was 2789 μmol trolox equivalents (TE)/g skins, which occurred at 90% power, 150 s and 1.5 g of skins. Resveratrol was identified in PSE by LC–MS–MS analysis.  相似文献

Three wine grapes, Norton (Vitis aestivalis), Cabernet Franc clone1, and Cabernet Franc clone313 (Vitis vinifera), collected from a Virginia vineyard were evaluated and compared for their antioxidant properties and phenolic profile. All grape extracts exerted remarkable antioxidant activities. Their oxygen radical absorbance capacity (ORAC) values were not significantly different from one another, ranging from 22.9 to 26.7 μmol TE/g of fresh weight. The Cabernet Franc clone1 had the strongest 1,1-diphenyl-2-picrylhydrazyl (DPPH) radicals scavenging activity (8.8 μmol TE/g) compared to the Norton or Cabernet Franc clone313 grape extracts (7.9 μmol TE/g and 5.4 μmol TE/g, respectively). The Norton grape contained significantly higher total phenolic, anthocyanin, and flavonoid content than the Cabernet Franc grapes (p < 0.05). The hydroxybenzoic acids, in particular gallic acid, were the major phenolic acids in all the grape extracts. The Norton grape variety was found to be rich in malvidin-diglucoside and malvidin-glucoside, while the malvidin-diglucoside was negligible in the Cabernet Franc grapes. The results suggest a remarkable impact of grape genotype on its antioxidant properties and phenolic composition in Virginia-grown wine grapes.  相似文献

The antioxidant activity (oxygen radical absorbance capacity, ORAC) and total phenols and flavonoids were determined in extracts from digestive tract, gonads, muscles and respiratory apparatus of sea cucumber, Cucumaria frondosa. Total phenols content varied from 22.5 to 236.0 mg of gallic acid equivalents/100 g dw, and flavonoids from 2.9 to 59.8 mg of rutin equivalents/100 g. ORAC values ranged from 140 to 800 μmol of Trolox equivalents/g dw. Among all extracts, best antioxidant potencies were observed in ethyl acetate extracts from digestive tract, and in acetonitrile-rich fractions obtained from mixed extracts using acetonitrile/TFA (trifluoroacetic acid) acidified water on muscles, gonads and respiratory apparatus. The weakest potencies were observed with water extracts from digestive tract and respiratory apparatus, and with water-rich fractions obtained from mixed extraction of gonads and muscles. A significant correlation was observed between ORAC values and total phenol content in extracts and fractions of gonads and muscles, but ORAC and phenols were not correlated in digestive tract and respiratory apparatus extracts. ORAC values were significantly correlated (p < 0.05) with total flavonoids in all extracts. Successive eluates obtained from solid-phase extraction of water-rich fractions using C18 cartridge showed ORAC values (105–500 μmol of TE/g) reaching up to 2.3 times the potency of their parent fractions. Flavonoids are suggested to be mainly responsible for observed activities. Our results provide a first quantitative evaluation of C. frondosa tissues as useful sources of antioxidants for human consumption.  相似文献

Antioxidant activities (μmol Trolox equivalent (TE)/g fresh weight) of 19 sweet potato genotypes with distinctive flesh colour (white, cream, yellow, orange and purple) were measured by oxygen radical absorbance capacity (ORAC), 2,2-diphenyl-1-picrylhydrazyl (DPPH), and 2,2′-azinobis(3-ethyl-benzothiazoline-6-sulfonic acid) (ABTS). Total phenolics were measured using the Folin–Ciocalteau method, total anthocyanins by the pH-differential method, and β-carotene by HPLC. The total antioxidant activity (hydrophilic + lipophilic ORAC) was highest (27.2 μmol TE/g fresh weight (fw)) for NC415 (purple-fleshed) and lowest (2.72 μmol TE/g fw) for Xushu 18 (white-fleshed). The hydrophilic-ORAC values were significantly correlated with the DPPH (R2 = 0.859) and ABTS (R2 = 0.761) values. However, the lipophilic-ORAC values were poorly correlated with the β-carotene contents (R2 = 0.480). The total phenolic contents (0.011–0.949 mg chlorogenic acid equivalent/g fw) were highly correlated with the hydrophilic-ORAC (R2 = 0.937) and DPPH (R2 = 0.820) values. Therefore, the total phenolic content can serve as a useful indicator for the antioxidant activities of sweet potatoes.  相似文献

The polyphenolic content and antioxidant capacity in the seeds of 11 red grape varieties (five international and six native) widely cultivated in Turkey were investigated. Total phenolic, total flavanol and total polymeric procyanidin content ranged from 79.2 to 154.6, 89.2 to 179.4, and 27.0 to 43.3 mg/g seed, respectively. While (+)-catechin (4.71–23.8 mg/g seed) was found as main flavanol, galloylated catechin monomer and dimeric procyanidin amounts varied between 2.89–17.2 and 0.97–2.97 mg/g seed, respectively. All seed extracts showed remarkable DPPH radical scavenging activity (EC50) and oxygen radical scavenging capacity (ORAC) ranging from 2.71 to 4.62 μg/mL and 1425.9 to 3009.2 μmol Trolox equivalent/g seed, respectively. With high amount of total phenolic content and antioxidant activity, seeds of Okuzgozu, Papaz Karasi, Ada Karasi and Kalecik Karasi varieties could be evaluated as dietary supplement.  相似文献

Polyphenolic antioxidant profiles of yellow camellia   总被引:1,自引:0,他引:1  
Yellow camellia (section Nitidissima Chang) is a subgroup of tea plant with golden yellow flowers. It is a very rare species with potential as antioxidant rich plant as it is closely related to tea. We found, for the first time, that the antioxidant capacity and polyphenolic constituents of six yellow camellia species varied widely. Among them, the dry leaf of Camellia impressinervis had the highest level of oxygen radical absorbance capacity (ORAC, 2270.9 μmol TE/g), total phenolic content (TPC, 475.6 μmol GAE/g), and proanthocyanidin content (PAC, 66.1 μmol CE/g). The ORAC level correlated well with TPC (R2 = 0.9402), and PAC (R2 = 0.9954). The compounds responsible for antioxidant activities were further profiled by HPLC and LC–MS analysis. Compared to the commonly consumed tea leaves, yellow camellia leaves contain more diverse phenolic compounds, including ellagitannins, proanthocyanidins, taxifolin deoxyhexose, apigenin derivatives, kaempferol derivative, quercetin derivatives, glucosyl isorhamnetin, (epi)catechin-(epi)afezelechin polymers and platphyllosides. Unlike tea leaves, no caffeine is detected in the yellow camellia leaves. The antioxidant activity of the polyphenolic compounds were further verified by precolumn treatment of the extracts with a stable radical 2,2-diphenyl-1-picrylhydrazyl (DPPH).  相似文献

Sugarcane molasses is a rich source of antioxidant materials with peroxyl radical scavenging effects. To explore the potent antioxidant activity of sugarcane molasses against 2,2′-azobis(2-amidinopropane) dihydrochloride (AAPH)-induced peroxyl radicals, 7 methanolic fractions of sugarcane molasses (F1–F7) were separated via bioassay-guided fractionation and evaluated by oxygen radical absorbance capacity (ORAC), cellular antioxidant activity (CAA), and oxidative DNA damage protective activity assays. The ORAC values of sugarcane molasses fractions ranged from 4399 to 6266 μmol TE/g, whilst the EC50 values for CAA ranged from 3.7 to 5.9 μg/ml. Moreover, it was found that sugarcane molasses fractions, particularly F6 and F7, could protect against oxidative DNA damage caused by peroxyl radicals at an effective concentration of 100 μg/ml. Ten phenolic constituents were identified in the fractions, including known antioxidative compounds, viz., schaftoside, isoschaftoside, ferulic acid, p-coumaric acid, and p-hydroxybenzaldehyde.  相似文献

The carotenoid compositions, antioxidant activities and the potential cardio-protective role of 13 tomato cultivars with distinct colour were studied. Colour coordinates were evaluated by colorimeter and the carotenoid compositions were analysed by UPLC. Red tomatoes had the highest total carotenoid contents (TCC) and antioxidant activities, followed by purple, orange, pink and yellow ones. The TCC were 120.5–278.0 μg/g DW, and the antioxidant activities were 21.32–40.07 μmol TE/g DW (PCL), 64.42–89.98% (DPPH) and 10.47–13.76 μmol TE/g DW (ORAC), respectively. The lipophilic extracts were also found to prevent cell death in a cell-based model system using cardiac H9c2 cells and H2O2, via attenuation of the caspase-3 and matrix metalloproteinase-2 activities. The extracts of different tomatoes showed strong but different antioxidant activities. Roles of total and individual carotenoids in the antioxidant activities were studied and lycopene showed the highest correlation. Results of this study can be used to guide the development of new tomato cultivars and functional foods, and benefit the consumers.  相似文献

Seven primocane fall-bearing raspberry (Rubus idaeus L.) cultivars, Nova (red), Dinkum (red), Heritage (red), Autumn Britten (red), Josephine, Anne (yellow), Fall Gold (yellow) were analysed for potential health promoting properties including their inhibitory effect on starch and fat digestive enzymes, antioxidant activities, and phenolic composition. The tested raspberry extracts showed no detectable inhibition of pancreatic α-amylase and lipase. However, all the extracts exhibited potent inhibition of α-glucosidase with IC50 from 16.8 to 34.2 μg/mL. Four phenolic compounds, ellagic acid, cyanidin-diglucoside, pelargonidin-3-rutinoside, and catechin were identified as the active α-glucosidase inhibitors. The raspberry extracts also possessed significant antioxidant activities with oxygen radical absorbance capacities (ORAC) ranging from 136.7 to 205.2 μmol Trolox equivalents (TE)/g dry weight fruit and DPPH radical scavenging activities from 305 to 351 μmol TE/g. The total phenolic content of raspberry cultivars varied significantly from 40.9 to 98.5 mg of gallic acid equivalents/g dry weight. The anthocyanin content varied widely from 0.1 to 9.5 mg cyanidin 3-glucoside equivalents/g. Nine phenolic acids were quantified in raspberries and their total amounts varied from 157.3 to 713.5 μg/g. The enzyme inhibition and antioxidant properties of raspberry cultivars were not correlated with their total phenolic, anthocyanin, and phenolic acid content. Overall, ‘Dinkum’ and ‘Josephine’ raspberry varieties possess higher total phenolic content, ORAC, DPPH radical scavenging activity, and α-glucosidase inhibitory activity than other five cultivars.  相似文献

The in vitro antioxidant properties of different extracts (water, alcohol, alcohol:water, hexane or chloroform extract) of curry leaves (Murraya koenigii L.) were evaluated using various assays. The alcohol:water (1:1) extract of curry leaves (AWEC) showed the highest antioxidant and free radical scavenging activity. It inhibited membrane lipid peroxidation by 76%, at 50 μg/ml, scavenged 93% of superoxides at 200 μg/3 ml and scavenged approximately 90% of hydroxyl and 1,1-diphenyl-2-picrylhydrazayl radicals at 4–5-fold lower concentrations compared to the other tested extracts. In addition, the alcohol:water extract reduced cytochrome c and ferric ion levels, chelated ferrous ions and inhibited ferrous sulfate:ascorbate-induced fragmentation and sugar oxidation of DNA. These results establish the antioxidant potential of AWEC, which could be used as natural antioxidant source.  相似文献

Ethanol extracts of Chardonnay grape and black raspberry seed flours were evaluated for their capacity to suppress lipid oxidation, preserve important fatty acids, and inhibit microbial growth. They were also tested for radical scavenging activity against DPPH and peroxyl radicals as reflected in oxygen radical absorbance capacity (ORAC), and total phenolic content (TPC). Both tested seed flour extracts suppressed lipid oxidation and rancidity development in fish oil. Black raspberry seed flour extract significantly reduced the degradation of biologically important n − 3 PUFA under accelerated oxidative conditions. Black raspberry and Chardonnay seed flour extracts at 165 and 160 μg seed flour equivalents/mL, respectively exhibited bacteriocidal activity against Escherichia coli and growth inhibition of Listeria monocytogenes under experimental conditions. Both seed flour extracts exhibited DPPH radical quenching activity and Chardonnay had the stronger ORAC of 663 μmol Trolox equivalents per gram seed flour and the higher TPC of 99 mg gallic acid equivalents/g flour. The data from this study suggest the potential for developing natural food preservatives from these seed flours for improving food stability, quality, safety, and consumer acceptance.  相似文献

Antioxidant capacity (AC), total phenolic content (TPC) and total carotenoid content (TCC) in palm oils at various stages of the refining process from two technological modes were determined. The obtained mean FRAP and DPPH values for the methanolic extracts of palm oils from mode 1 (19.5–102.8 μmol TE/100 g and 18.8–103.0 μmol TE/100 g) were lower than for oils from mode 2 (25.6–134.8 μmol TE/100 g and 25.4–135.4 μmol TE/100 g). The total phenolics (4.1–12.4 mg GA/100 g) and carotenoids (0.18–45.8 mg/100 g) in the studied oils were correlated with their antioxidant capacities determined by FRAP and DPPH methods (r 0.6623–0.9878). During the refining process, for both technological modes resulted in a loss of AC by 80%, TPC by 26–55% and TCC by 99%. The bleaching step caused the highest losses of AC as determined by FRAP 41% and 46%, DPPH by 43% and 48%, while TPC loss was 45% and 23% and loss of carotenoids was 49% and 56%, in mode 1 and mode 2, respectively.  相似文献

The effects of varying doses of UV and ultrasound (US), and incubation times at 25 °C on total phenolics (TP), antioxidant capacity by two methods, trolox equivalent antioxidant capacity (TEAC) and oxygen radical absorbance capacity (ORAC) and sensory overall acceptance (OA) of treated peanuts were investigated. Optimum process parameters for UV and US were determined using response surface methodology. UV and US increased TP, TEAC and ORAC total antioxidant capacity (TAC) of most treated peanuts, whereas OA decreased in all treatments compared to controls. US was more effective than UV in increasing total antioxidants. Optimum US process parameters at 36 h incubation time will result in products with TAC ≥ 68 μM TE/g, TP ≥ 1.51 mg GAE/g, TEAC ≥ 2.76 μM TE/g, and OA ≥ 5 or neither like nor dislike.  相似文献

The total phenols content (Folin–Ciocalteau assay) and antioxidant capacity (ferric reducing/antioxidant power – FRAP) of 41 plants from Córdoba (Argentina) were analyzed. Phenol content ranged from 8.2 to 100.2 mg GAE/g. FRAP ranged from 85.2 to 1862.0 μmol of Fe(II)/g. Capparis atamisguea had the lowest values of total phenols content and antioxidant capacity (8.2 mg GAE/g and 85.2 μmol of Fe(II)/g, respectively), while Ligaria cuneifolia exhibited the highest values (100.2 mg GAE/g and 1862.0 μmol of Fe(II)/g, respectively). A significant linear correlation (p < 0.05) was found (0.9125) between phenols content and antioxidant capacity. Results support the idea that these plants may be a good source of natural antioxidants for food applications. Plants from the Asteraceae family (the most representative of the Córdoba flora) were further tested for their DPPH radical scavenging activity. Some plant extracts were tested in a simple food system to investigate to their potential use in foods.  相似文献

Suaeda fruticosa is an edible and medicinal halophyte known for its hypoglycaemic and hypolipidaemic activities. In this study, novel biological activities of the shoot extracts related to their phenolics were investigated. Results showed an appreciable total phenolic (31.8 mg GAE/g DW) in shoot extracts. The estimation of antioxidant capacities using oxygen radical absorbance capacity (ORAC method) and a cell based-assay (WS1) of four extracts (hexane, dichloromethane, methanol and water) showed that shoot methanol extract exhibit the highest antioxidant activities. The same extract displayed the utmost anti-inflammatory activity, inhibiting nitric oxide (NO) release, by 66.4% at 160 μg/ml in lipopolysaccharide (LPS)-stimulated RAW 264.7 macrophages. Besides, the dichloromethane extract showed the highest anticancer activity against human lung carcinoma (A-549) and colon adenocarcinoma cell lines (DLD-1, Caco-2 and HT-29) with specificity against DLD-1 (IC50 = 10 ± 1 μg/ml). These findings demonstrate the remarkable potentiality of this edible halophyte as valuable source of antioxidants which exhibit original and interesting anti-inflammatory and anticancer capacities.  相似文献

Antioxidant activity and phenolic compounds of sumac extracts were investigated. Sumac was extracted in methanol and subjected to solvent–solvent partitioning to yield two fractions as ethyl acetate and aqueous. Methanol extract was further fractioned over Sephadex LH-20 column. Antioxidant activity of extracts and fractions were screened using ferric thiocyanate and DPPH radical scavenging methods. Phenolic composition of active fraction(s) was determined by HPLC–MS systems. Those fractions which exhibited strong antioxidant activity were rich in anthocyanins and hydrolysable tannins. While gallic acid was the main phenolic acid in the extracts, anthocyanin fraction contained cyanidin, peonidin, pelargonidin, petunidin, and delphinidin glucosides and coumarates. Pentagalloyl glucose was abundant in the hydrolysable tannin fraction. Effective scavenging concentration (EC50) on DPPH radical was 0.70 μg/mL both in ethyl acetate and tannin fractions, and 5.33 μg/mL in anthocyanin rich fraction. Same extracts and fractions showed moderate lipid peroxidation inhibition effect compared with the synthetic antioxidants. The findings demonstrate that sumac can be used as a natural antioxidant.  相似文献

Most research on extraction of phenol compounds from wine by-products and commercial exploitation of extracts use grape seeds and/or skins as raw materials. Looking for alternative antioxidants sources, obtaining antioxidant extracts from wine lees (also known as dregs), a sub-exploited by-product of winemaking process, is here presented. Microwave-assisted extraction (MAE) of phenolic compounds from wine lees has been optimized using the total phenols index, the ORAC values and yield of the extraction as response variables. Under the optimal working conditions, the proposed MAE method provides better extraction efficiency in a much shorter time (17 min) than the conventional extraction method for phenolic compounds (24 h). The liquid extract obtained by MAE was spray-dried. The type and amount of excipients used, as well as the spray-drying temperature, were optimized in order to minimize the oxidation of phenolic compounds and maximize the yield of the spray-drying process. The total phenols index in the dried extract thus obtained was 36.8% (expressed as gallic acid), showing an ORAC value of 3930 μmol TE/g. Additionally, Mv3G, Cm-Mv3G, myricetin, quercetin, quercetin-3-β-glucoside, caffeic acid and p-coumaric acid were quantified in the dry extract by HPLC–DAD. The results indicate that wine lees antioxidant extracts can be a suitable and cheap alternative to those obtained from grape seeds or skins.  相似文献

The methanol extract from mature fruits of Aristotelia chilensis (Mol) Stuntz (Elaeocarpaceae) showed antioxidant activities and cardioprotective effects on acute ischemia/reperfusion performed in rat heart in vivo. This extract protected animals from heart damage by the incidence of reperfusion dysrythmias, and the no-recovery of sinus rhythm. On the other hand, the MeOH extract of the fruit was able to prevent these harmful events in the animal’s heart by diminishing lipid oxidation and reducing the concentration of thiobarbituric acid reactive substances (TBARS), a lipid peroxidation index. In addition, MeOH extract of A. chilensis was evaluated for DPPH (2,2-diphenyl-1-picrylhydrazyl) radical scavenging, crocin radical scavenging, oxygen radical absorption capacity (ORAC), ferric reducing antioxidant power (FRAP), an estimation of lipid peroxidation in liposomes through the inhibition of formation of TBARS. MeOH extract was found to have IC50 of 1.62 ppm against DPPH and 2.51 ppm against TBARS, compared with the juice, whose IC50 was 12.1 ppm and 9.58 ppm against DPPH and TBARS formation, respectively. Antioxidant activities of MeOH extract were strongly correlated with total polyphenol content. Consistent with this finding, MeOH had the greatest ORAC and FRAP values as percentage of activity. These results show that these fruits could be useful as antioxidant, cardioprotective and nutraceutical sources.  相似文献
