In this paper the overall antioxidant power, expressed as Briggs–Rauscher antioxidant index, of decoction or cold infusions of dried Hibiscus sabdariffa flowers was determined at 25 and 37 °C, to compare the scavenger ability of the beverages at either room or physiological temperature. Total polyphenol contents and the absorbance of anthocyanin pigments were also determined, and the trend with the overall antioxidant capability is considered. Combined photometric and pH-metric titrations were acquired to obtain information on the colour-total acidity relationship of the product. The results show that the decoction preparation protocol provides karkadè with the highest nutritional value and that the polyphenol content can account for the antioxidant capability of H. sabdariffa-based beverages. Moreover, a quantitative relationship between acid–base and redox chemistry was found. The H. sabdariffa-based drinks can be considered as protective beverages and a regular consumption of karkadè might be proposed to ensure protection against free radicals.  相似文献

Response surface methodology was used to investigate the effect and interactions of processing variables such as roselle extract (0.1–1.3%), soybean oil (5–20%) on physicochemical, textural and sensory properties of cooked pork patties. It was found that reduction in thickness, pH, L* and b* values decreased; however, water-holding capacity, reduction in diameter and a* values increased, respectively, as the amount of roselle increased. Soybean oil addition increased water-holding capacity, reduction in thickness, b* values of the patties. The hardness depended on the roselle and soybean oil added, as its linear effect was negative at p < 0.01. The preference of color, tenderness, juiciness, and overall quality depend on the addition of roselle and soybean oil. The maximum overall quality score (5.42) was observed when 12.5 g of soybean oil and 0.7 g of roselle extract was added. The results of this optimization study would be useful for meat industry that tends to increase the product yield for patties using the optimum levels of ingredients by RSM.  相似文献

Gundelia tournefortii L. is an important food source and a well-known medicinal plant in Eastern Anatolia. Therapeutic effects of medicinal plants are known to be closely related to their antioxidant capacities. Antioxidant activities of G. tournefortii, both for the aerial parts and seeds, were investigated by using both 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging and lipid peroxidation inhibition methods. The seeds were found to have higher antioxidant potential than the aerial, with IC50 values of 0.073 mg/mL for DPPH scavenging and 0.146 mg/mL for lipid peroxidation inhibition capacities. In addition, total phenolic contents of the Gundelia tournefortii L. extracts, especially the seed extracts correlates to its high antioxidant activity with 105.1 ± 8.7 μg gallic acid equivalents (GAEs) per mg of seed extract. Plant extracts with high phenolics content are known to have important effects on various enzymes, as well as glutathione-S-transferases, which are important detoxification enzymes in phase II systems with an important role in developing multi-drug resistance to chemotherapy in tumour cells. Consequently, the effects of G. tournefortii extracts on crude cytosolic glutathione-S-transferase was also studied and the seed extracts have shown effective inhibition of cytosolic GST activity, with an IC50 of 97.51 μg/mL.  相似文献

In the present study the quantification of the polyphenolic fraction, anthocyanins and other polar compounds, the antioxidant capacity and the anti-hyperlipemic action of the aqueous extract of Hibiscus sabdariffa has been achieved. Seventeen compounds were successfully quantified either by HPLC-DAD or HPLC-ESI-TOF-MS. Six of them were directly quantified by their corresponding standards, whereas the rest were indirectly quantified as equivalents using standards of similar compounds. The antioxidant capacity have also been estimated by comparing different assays, i,e, Trolox equivalent antioxidant capacity (TEAC), ferric reducing antioxidant power (FRAP), oxygen radical absorbance capacity (ORAC), and measurement of thiobarbituric acid reacting substances (TBARS). H. sabdariffa showed high reducing capacity in FRAP assay and significant capability to scavenge peroxyl radicals in the ORAC assay. Nevertheless, the extract exhibited poor efficacy to inhibit peroxyl radicals in lipid systems. The plant extract also exhibited the capacity to decrease serum triglyceride concentration on hyperlipemic mouse model.  相似文献

The antioxidant properties and total phenolic contents of methanol, acetone and water extracts of mulberry (Morus indica L.) leaves were examined. Various experimental models including iron (III) reducing capacity, total antioxidant capacity, DPPH radical scavenging activity and in vitro inhibition of ferrous sulphate-induced oxidation of lipid system were used for characterization of antioxidant activity of extracts. The three extracts showed varying degrees of efficacy in each assay in a dose-dependent manner. Methanolic extract with the highest amount of total phenolics, was the most potent antioxidant in all the assays used. In addition, the effect of temperature (50 °C and 100 °C), pH (3, 5, 7, 9 and 11) and storage (5 °C) on the antioxidant activity of methanolic extract was investigated. The antioxidant activity of the extract remained unchanged at 50 °C and was maximum at neutral pH. The extract stored at 5 °C in the dark was stable for 30 days after which the antioxidant activity decreased (p ? 0.05) gradually. On the basis of the results obtained, mulberry leaves were found to serve as a potential source of natural antioxidants due to their marked antioxidant activity.  相似文献

Antioxidant activity and phenolic compounds of sumac extracts were investigated. Sumac was extracted in methanol and subjected to solvent–solvent partitioning to yield two fractions as ethyl acetate and aqueous. Methanol extract was further fractioned over Sephadex LH-20 column. Antioxidant activity of extracts and fractions were screened using ferric thiocyanate and DPPH radical scavenging methods. Phenolic composition of active fraction(s) was determined by HPLC–MS systems. Those fractions which exhibited strong antioxidant activity were rich in anthocyanins and hydrolysable tannins. While gallic acid was the main phenolic acid in the extracts, anthocyanin fraction contained cyanidin, peonidin, pelargonidin, petunidin, and delphinidin glucosides and coumarates. Pentagalloyl glucose was abundant in the hydrolysable tannin fraction. Effective scavenging concentration (EC50) on DPPH radical was 0.70 μg/mL both in ethyl acetate and tannin fractions, and 5.33 μg/mL in anthocyanin rich fraction. Same extracts and fractions showed moderate lipid peroxidation inhibition effect compared with the synthetic antioxidants. The findings demonstrate that sumac can be used as a natural antioxidant.  相似文献

Ten nanofiltration flat-sheet membranes and eight tight ultrafiltration membranes with nominal MWCOs ranging from 0.2 to 150 kDa were tested to concentrate anthocyanin extract from roselle. A pilot system was used, which featured a membrane cell with an effective area of 0.0155 m2. Permeate fluxes were recorded for transmembrane pressures between 0.5 and 3.0 MPa, while keeping all other operating conditions constant (volumetric reduction ratio 1, 35 °C). Retention values of total soluble solids, acidity and anthocyanins increased with transmembrane pressure. With similar permeate fluxes at average transmembrane pressure, retention of anthocyanins is significantly higher for nanofiltration membranes than for ultrafiltration membranes. A membrane was then selected for an industrial trial on the basis of its flux, retention of compounds of interest and energy consumption per liter of permeate. The trial using a 2.5-m2 filtration surface, could concentrate roselle extract from 4 to 25 g total soluble solids per 100 g, with 100% retention of anthocyanins. No significant damages were observed when comparing concentrate quality with the initial roselle extract.  相似文献

Antioxidant activity and hepatoprotective potential of Phyllanthus niruri, a widely used medicinal plant, were investigated. Methanolic and aqueous extract of leaves and fruits of P. niruri showed inhibition of membrane lipid peroxidation (LPO), scavenging of 1,1-diphenyl-2picrylhydrazyl (DPPH) radical and inhibition of reactive oxygen species (ROS) in vitro. Antioxidant activity of the extracts were also demonstrable in vivo by the inhibition of the carbon tetrachloride (CCl4) – induced formation of lipid peroxides in the liver of rats by pretreatment with the extracts. CCl4 – induced hepatotoxicity in rats, as judged by the raised serum enzymes, glutamate oxaloacetate transaminase (GOT) and glutamate pyruvate transaminase (GPT), was prevented by pretreatment with the extracts, demonstrating the hepatoprotective action of P. niruri.  相似文献

Methanol extract from juemingzi (Cassia tora L.) was fractionated by liquid-liquid partition using ethyl acetate, n-butanol and water, respectively. The antioxidant activity of three different fractions was evaluated using different antioxidant tests, including inhibition of peroxidation of linoleic acid, reducing power, free-radical scavenging and ferrous ions chelating activity. In the above four assays, all the fractions showed antioxidant potential to varying degrees. Among these fractions, ethyl acetate fraction exhibited more antioxidant potency than other fractions. Furthermore, ethyl acetate fraction was found to be more effective in protecting LDL against oxidation in a concentration-dependent manner. The data suggest that juemingzi especially ethyl acetate-soluble fraction may have a preventive effect against atherosclerosis by inhibiting LDL oxidation.  相似文献

Therapeutic effects of several medicinal plants and vegetables, which are commonly used as food and in folk medicine against many disease, are well known. Antioxidant capacities of Heracleum persicum Desf., Prangos ferulacea (L.) Lindl., Chaerophyllum macropodum Boiss. species from Apiaceae family were evaluated by determining their effects on DPPH radical scavenging, and lipid peroxidation inhibition, as well as their total phenolic contents. Potential natural glutathione-S-transferase inhibitors have gained great importance in the last decade especially because of the role of glutathione-S-transferases in developing resistance to chemotherapy. The effect of dense phase carbon dioxide (DPCD) processing (34.5 MPa, 8% CO2, 6.5 min, and 40 °C) on phytochemical, sensory and aroma compounds of hibiscus beverage was compared to a conventional thermal process (HTST) (75 °C for 15 s) and a control (untreated beverage) during refrigerated storage (4 °C). The overall likeability of the hibiscus beverage for all treatments was not affected by storage up to week 5. DPCD process retained more aroma volatiles as compared to HTST. Aroma profiles in the beverages were mainly composed of alcohols and aldehydes with 1-octen-3-ol, decanal, octanal, 1-hexanol, and nonanal as the compounds with the highest relative percentage peak areas. A loss of only 9% anthocyanins was observed for the DPCD processed hibiscus beverage. Phytochemical profiles in the hibiscus beverage included caffeoylquinic acids, anthocyanins, and flavonols. No major changes in total phenolics and antioxidant capacity occurred during the 14 weeks of storage.  相似文献

Hibiscus sabdariffa leaf, the edible part of H. sabdariffa Linne, is usually ignored and discarded. The object of the study was to examine the anticancer properties of H.sabdariffa L. leaf extract (HLE). First, HLE was demonstrated to be rich in polyphenols, including catechin and ellagic acid (EA). 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) data showed that among three kinds of human prostate cancer (CaP) cells, androgen-dependent LNCaP cells were the most susceptible to HLE. HLE and its purified compound EA were evaluated for apoptotic activities. Molecular data showed the effect of HLE in LNCaP cells might be mediated via both intrinsic and extrinsic apoptotic pathways. Finally, HLE inhibited the growth of LNCaP cells in xenograft tumour studies. As a result, our data presented the first evidence of HLE as an apoptosis inducer in LNCaP cells, and these findings may open interesting perspectives to the strategy in human CaP treatment.  相似文献

This study was designed to examine the in vitro antioxidant activities and rosmarinic acid levels of the methanol extracts of Salvia verticillata subsp. verticillata and S. verticillata subsp. amasiaca. The extracts were screened for their possible antioxidant activity by two complementary test systems, namely DPPH free radical-scavenging and β-carotene/linoleic acid systems. In the first case, S. verticillata subsp. verticillata was superior to the subsp. amasiaca with an IC50 value of 14.5 ± 1.21 μg mg−1. In the β-carotene/linoleic acid test system, inhibition capacity of S. verticillata subsp. verticillata was 74.4 ± 1.29%. Antioxidant activities of BHT, ascorbic acid, curcumin and α-tocopherol were determined in parallel experiments. Activity of rosmarinic acid was also screened for better establishing the relationship between rosmarinic acid level and antioxidant activity for the plant extracts. S. verticillata subsp. verticillata had the highest rosmarinic acid level with a value of 28.7 ± 0.89 μg mg−1. There is a strong correlation between the rosmarinic acid level and antioxidant activity potential. Our results showed that rosmarinic acid and its derivatives are more likely to be responsible for most of the observed antioxidant activities of Salvia species.  相似文献

In this study, roselle extract was cold sterilized by crossflow microfiltration. At laboratory scale the optimal operating conditions at volumetric reduction ratio (VRR) 1 cannot be generalized when VRR is greater than 1. Indeed, at VRR 1 the optimal transmembrane pressure was 2.5 bar with a permeate flux of 185 L h−1 m−2, while at VRR 20 optimal pressure was 3.7 bar with a permeate flux of 95 L h−1 m−2. A simple model that allowed to represent the effect of VRR and transmembrane pressure on the permeate flux was adjusted with the experiments done at laboratory scale. This model used to predict the permeate flux using a semi-industrial pilot scale equipment gave a good match between the experimental and calculated performances. Permeate was collected aseptically and stored at 4, 20 and 37 °C for 90 days. A roselle extract storage at 4 and 20 °C was appropriate for consumption after a 3 month shelf life.  相似文献

The oxidative modification of low-density lipoprotein (LDL) is involved in the pathogenesis of atherosclerotic lesions through the formation of macrophage-derived foam cells. In the present study, we aimed to investigate the anti-atherosclerotic effect of Hibiscus sabdariffa leaf polyphenolic extract (HLP), which is rich in flavonoid. The inhibitory effect of HLP on oxidation and lipid peroxidation of LDL was defined in vitro. HLP showed potential in reducing foam cell formation and intracellular lipid accumulation in oxidised-LDL (ox-LDL)-induced macrophage J774A.1 cells under non-cytotoxic concentrations. Molecular data showed these influences of HLP might be mediated via liver-X receptor α (LXRα)/ATP-binding cassette transporter A1 (ABCA1) pathway, as demonstrated by the transfection of LXRα siRNA. Our data implied that HLP up-regulated the LXRα/ABCA1 pathway, which in turn led to stimulation of cholesterol removal from macrophages and delay atherosclerosis. These results suggested that HLP potentially could be developed as an anti-atherosclerotic agent.  相似文献

Radical scavenging and antibacterial properties of large thyme extracts isolated from five chemotypes of Thymus pulegioides L. growing wild in Lithuania were studied. The chemotypes were defined according to the main essential oil components: linalool (L), geranial/geraniol/neral (G/G/N), thymol (T), carvacrol/γ-terpinene/p-cymene (C/γT/pC) and thymol/carvacrol/γ-terpinene/p-cymene (T/C/γT/pC). The contents of phenolic compounds, flavonoids and flavonols were determined. It was found that the extracts of phenolic chemotypes containing remarkable concentrations of thymol and/or carvacrol were stronger DPPH and ABTS free radical scavengers in the model systems. The antibacterial activity of the extracts depended on the plant chemotype, extract preparation, solvent used and finally the sensitivity of bacteria. Bacillus cereus, Micrococcus luteus, Staphylococcus epidermidis and Staphylococcus aureus were the most sensitive to the all extracts applied, whereas Escherichia coli, Salmonella typhimurium and Enterobacter aerogenes remained resistant.  相似文献

Changes in total phenolics, antioxidant activity (AOX), carotenoids, capsaicin and ascorbic acid were monitored during three maturity stages in 10 genotypes of sweet pepper. In an attempt to explain the variations during maturity stages (green, intermediate and red/yellow), the data was expressed both on fresh and dry weight basis. All the antioxidant constituents (phenolics, ascorbic acid and carotenoids) and AOX, when expressed on fresh weight basis in general, showed an overall increasing trend during maturity in all the genotypes studied. On dry weight basis, phenolic content declined in majority of the genotypes during maturity to red stage. This decline was significant (P<0.05) in Parker, Torkel, HA-1038 and Flamingo. Genotype Flamingo and Golden Summer had the highest phenolic content of 852.0 mg 100 g−1 and 720.5 mg 100 g−1, at their final red and yellow maturity stages, respectively. With maturation, most of the cultivars showed a declining trend with regard to capsaicin content while total carotenoids and β-carotene content increased significantly. Anupam was a promising genotype in terms of both total carotenoids and β-carotene content. Ascorbic acid content declined progressively with advancing maturity. Genotype HA-1038 had the maximum content (3030 mg 100 g−1 dwb) at the green stage. AOX in general, increased with maturity and registered a 1.30-1.95fold increase from green to red stage.The study proposes the nutritional significance of consuming sweet peppers at the red maturity stage because of enhanced functional properties. Overall genotype Flamingo and Anupam represent superior genotypes for both nutrition and germplasm improvement.  相似文献

Tuberous roots of Decalepis hamiltonii (Dh) are consumed in southern India as pickles and beverage for their health benefits. The antioxidant potential of the roots was measured using various in vitro assays. Among the extracts, the methanolic and aqueous extracts showed high antioxidant activity measured as scavenging of DPPH, superoxide and hydroxyl radicals. Both the aqueous and methanolic extracts inhibited microsomal lipid peroxidation and exhibited strong reducing power and metal chelating activity. The antioxidant activity did not correlate with the phenolic content of the extracts. These results demonstrate the antioxidant potency of the root extracts which could be the basis for its alleged health promoting potential of Dh. The roots of Dh could serve as a new source of natural antioxidants or nutraceuticals with potential applications to reducing the level of oxidative stress and related health benefits.  相似文献
