Fruit from 42 blueberry (Vaccinium spp.) cultivars, including 36 rabbiteye cultivars (Vaccinium ashei Reade), three V. ashei hybrid derivatives, and three northern highbush (Vaccinium corymbosum L.) standards were evaluated for antioxidant capacity, individual flavonoid content, and the contribution of each identified phenolic compound to total antioxidant activity. Considerable variation was found in flavonoid content, antioxidant activity, and their contribution to total antioxidant activity among cultivars. Among 42 blueberry cultivars, the rabbiteye ‘Early May’ contained the highest amount of chlorogenic acid, myricetin 3-arabinoside, quercetin derivatives, and delphinidin-, cyanidin-, petunidin-, and malvidin-basis anthocyanins. ‘Early May’ cultivar also had the highest antioxidant activity (88.2 μmol TE/g fw). ‘Owen’, ‘Bluegem’, ‘Clara’, Climax’, and ‘Centurion’ were among the other rabbiteye cultivars that also had high levels of flavonoids and antioxidant activities. In contrast, the pink-fruited V. ashei hybrid, ‘Pink Lemonade’, had the lowest content of flavonoids and lowest antioxidant activity. The mean flavonoid content and antioxidant activity of rabbiteye cultivars was higher than those among northern highbush and V. ashei hybrids. The antioxidant activity of V. ashei hybrid derivatives was derived mainly from chlorogenic acid, myricetin, and quercetin, which contributed 62.5% of total antioxidant activity, whereas anthocyanins (malvidin, petunidin, delphinidin, and cyanidin) were the main contributors to the antioxidant activity of rabbiteye cultivars (76.2%) and northern highbush standards (76.8%). Blueberry cultivars identified to have high phenolic content and high antioxidant activity could be used as parents for future blueberry breeding programmes to develop new blueberry cultivars with higher antioxidant activity and further improve human health.  相似文献

The aroma impact components of the rabbiteye blueberry vinegars made from wine fermented without skin-contact (JV), wine fermented with skin-contact (WV), or blueberries (BV) were evaluated. Headspace volatiles were isolated by solid-phase microextraction (SPME). Headspace volatiles were analysed by gas chromatography–mass spectrometry (GC–MS) and gas chromatography–olfactometry (GCO). The potent odourants were evaluated by the ‘Osme’ method. Acetic acid, 2/3-methyl-butanoic acid, phenethyl acetate, 2-phenylethanol, octanoic acid, eugenol, and phenylacetic acid were the most important aroma-active compounds identified in all three treatments. The contributions of these compounds to each sample were different. There were more aroma-active compounds identified from the BV than from the other two treatments. The type of fermentation impacts the aroma components of rabbiteye blueberry vinegars.  相似文献

Fruit from 42 blueberry (Vaccinium species) cultivars, including 36 rabbiteye cultivars (Vaccinium ashei Reade), 3 V. ashei hybrid derivatives and 3 northern highbush (V. corymbosum L.) standards, were evaluated for antioxidant activities (AA), levels of antioxidant enzymes [catalase (CAT), ascorbate peroxidase (AsA-POD), glutathione reductase (GR), glutathione peroxidase (GSH-POD), superoxide dismutase (SOD), dehydroascorbate reductase (DHAR), monodehydroascorbate reductase (MDAR) and guaiacol peroxidase (G-POD)], and antioxidant nonenzyme components [ascorbic acid (ASA) and glutathione (GSH)]. The results of this study showed that cultivars had varying levels of AA, a wide range of antioxidant enzyme activities and various amounts of nonenzyme components. The correlations between AA and all of the enzyme activities or nonenzyme components were positive and high in the 42 cultivars tested in this study. The correlation (R2) values between AA and CAT, SOD, DHAR, MDAR, GSH-POD, GR, G-POD and AsA-POD were 0.91, 0.91, 0.91, 0.91, 0.90, 0.90, 0.90, and 0.90, respectively. The correlations between AA and ASA and GSH were 0.94 and 0.92, respectively. Among all 42 cultivars, ‘Early May’ had the highest activities of the AA, AsA-POD, G-POD, DHAR and MDAR and the highest amounts of ASA and GSH, whereas ‘Elliott’ had the highest levels of GR, GSH and SOD.  相似文献

In the present work the influence of temperature, xanthan gum and fructose addition on rheological behavior in steady state of blueberry puree was evaluated. The blueberry pulp was formulated with addition of xanthan gum (1.6, 2.0, 2.5, 3.0 and 3.3%) and fructose (6.6, 10.0, 15.0, 20.0 and 23.4%). Rheological data were obtained by shear rate scan tests in a rotational rheometer, with geometry of concentric cylinders in temperatures of 27, 40, 60, 80 and 93 °C. The thixotropy analysis was made through the calculation of the difference between the areas under up and down cycles’ flow curves. The analysis of the formulation influence on puree viscosity was carried out in two stages: i) selection of an appropriate rheological model for representation of data obtained in experiments and ii) statistical analysis of formulation and temperature influence on rheological parameters of selected model. Three 2-parameter models (Bingham, Ostwald–de Waele and Casson) and three 3-parameter models (Herschel–Bulkley, Mizrahi–Berk and Sisko) were tested. Based on the values of correlation coefficients and variance of the estimated parameters, Casson model was chosen for fitting of experimental data. The content of xanthan gum appears as a determinant variable of the rheological behavior of the puree, both at short and long times. The correlation among the variables studied was represented through statistical models which could be used for the development of formulations with specified viscosity in the range of additive concentrations studied.  相似文献

Basic blueberry processing includes juice processing or winemaking. By-products obtained from the juice and wine industry can be a source of new value-added products such as phenolic antioxidant supplements or ingredients for food processing. The phenolic compositions of products and by-products (pomaces) depend mainly on processing techniques such as duration of skin contact, crushing, pressing, and others. The present study was to evaluate the effects of fermentation type on retention of total anthocyanins, total phenolics, and antioxidant activity of blueberry by-products. Total phenolics (TPH), total anthocyanins (ACY), antioxidant activities (β-carotene bleaching assay and ferric thiocyanate assay), and antiradical activity (DPPH radical-scavenging assay) of rabbiteye blueberry (Vaccinium ashei) by-products (juice, wine, and vinegar pomaces) were determined. The wine pomace (WP) had higher TPH, antioxidant activities and antiradical activity. Vinegar pomace (VP) had the lowest ACY, TPH, antiradical activity, and antioxidant activities. The results indicate that the antioxidant and antiradical activities of blueberry by-products were not significantly affected by the wine making process. Acetification significantly decreased TPH, ACY, antioxidant activities, and antiradical activity. However, VP still maintained an important phenolics concentration and antioxidant activity.  相似文献

Frozen rabbiteye blueberries (Vaccinium ashei) were processed into juice (BJ), wines made without (BW1) or with (BW2) skin contact fermentation, and vinegars made from BW1 (JV), BW2 (WV) or blueberries (BV). Total phenolics, total anthocyanins, antioxidant activities (beta-carotene bleaching assay and ferric thiocyanate assay), and antiradical activity (DPPH radical-scavenging method) of these fluid products were determined. The differences in total anthocyanin contents of all blueberry products were significant. The BW2 had the highest content of anthocyanins and polyphenols and the highest beta-carotene bleaching activity and antiradical activity. Acetification decreased total anthocyanin content, total polyphenols and antioxidant activities. Correlations indicate that anthocyanins made significant contributions than did phenolics to antioxidant activities of products. The abilities of BJ, BW1 (wine from blueberry juice), and BW2 to inhibit linoleic acid peroxidation were high (∼95%). The abilities of vinegar products to inhibit linoleic acid peroxidation were low. The results indicate that skin-contact fermentation is a better method for obtaining higher antioxidant activity of blueberry products. Also, acetification significantly decreased anthocyanins and antioxidant activities.  相似文献

Yanang (Tiliacora triandra) is a vegetable used in many cuisines of the northeast of Thailand and Lao People’s Democratic Republic. This paper reports the optimised extraction and some physicochemical characteristics of polysaccharide gum from Yanang leaves. The optimised extraction condition was at the leaf:water ratio of 1:6.6, 85 °C, and 100 min extraction time. The major monosaccharide constituent of Yanang gum was xylose, together with substantial amounts of other neutral sugars. The FT-IR spectra of Yanang gum were similar to that of xylan. Yanang gum exhibited shear-thinning flow behaviour and the extent of shear-thinning was concentration dependent. The mechanical spectra of Yanang gum at low concentration (0.5%) were typical of semi-diluted to concentrated solutions. However, with increasing concentration, the solution behaved like a weak gel.  相似文献

This study was carried out to analyze the nutritional composition, mineral content and antioxidant activity of Kalopanax pictus leaves. K. pictus leaves are high in carbohydrates (60.37%) and crude protein content (27.05%) on a dry weight basis with low content of fatty acids (1.45%). Hyperoside (1), 3,5-di-O-caffeoyl quinic acid (2), methyl 3,5-dicaffeoyl quinate (3), and 3-O-feruloylquinic acid (4) were isolated and purified from this plant as well. In all the chemical assays used, compound 3 exerted better 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging activity, reducing power ability, and was a potent inhibitor of hydrogen peroxide (H2O2)-induced human embryonic kidney 293 (HEK 293) cell death and lipid peroxidation, whereas compound 4 presented as a powerful radical scavenger against superoxide radicals. Our results suggest that the chemical components of K. pictus leaves possess good antioxidant capacities and this species might have potential applications in the food and medical industries.  相似文献

Analysis by gas chromatography-mass spectrometry led to the identification of 20 volatiles from the steam distilled oil of the leaves from Chamaecyparis obtusa Siebold et Zuccarni. Seven constituents of the oil were tested for contact and fumigant activity against adults of Callosobruchus chinensis (L.) and Sitophilus oryzae (L.). Responses varied with compound and dose rather than increasing exposure time beyond 1 or 2 days. In the impregnated-paper test with the constituents [bornyl acetate, (+)-limonene, myrcene, α-phellandrene, α-pinene, sabinene and terpinolene], at 0.1 mg/cm2, bornyl acetate, α-phellandrene and terpinolene caused 97%, 97% and 87% mortality, respectively, against C. chinensis adults 1 day after treatment. At 0.05 mg/cm2, moderate activity was achieved with bornyl acetate (56% mortality), α-phellandrene (75%) and terpinolene (55%). Against S. oryzae adults, at 0.26 mg/cm2, terpinolene caused 93% mortality 2 days after treatment, whereas 80% mortality 4 days after treatment was observed with bornyl acetate and α-phellandrene. The toxicity of these compounds was significantly decreased at 0.18 mg/cm2. In a fumigation test with S. oryzae adults, bornyl acetate and terpinolene were much more effective in sealed containers than in open ones, indicating that the insecticidal activity of these compounds was largely attributable to fumigant action. These naturally occurring materials derived from leaves of Chamaecyparis could be useful for managing populations of C. chinensis and S. oryzae.  相似文献

A polyphenol oxidase was purified and characterised from leaves of the common spiderflower. Purification sequentially with ammonium sulphate, dialysis, DEAE-Sepharose ion-exchange chromatography and Sephadex G-75 gel filtration chromatography resulted in 37.8-fold enrichment in the specific activity and 44.3% recovery of the total activity. Purified PPO is a monomeric protein of 52.6 kDa revealed by Coomassie and active staining and Western blot. It was optimally active at pH 8.0 and 60 °C, and stable from pH 3.0 to 9.0 and below 60 °C. It displayed enzymatic activity towards monophenols, diphenols and triphenols, especially towards diphenols, and substrate specificity towards methylated and methoxylated substrates. Its activity was slightly increased by 0.1% SDS, heavily inhibited by Hg2+ and Pb2+, and completely inhibited by 1.0 mM of ascorbic acid, l-cysteine, β-mercaptoethanol, sodium diethyldithiocarbamate and thiourea, and by 10 mM of dithioerythritol, sodium metabisulphite and sodium sulphite.  相似文献

Berries of Vaccinium meridionale Swartz native to Colombia were analysed for chemical composition, total phenolic content, anthocyanin content, and antioxidant activity. In addition, high-performance liquid chromatography with photodiode array detection (HPLC–DAD) and HPLC-electrospray ionisation tandem mass spectrometry (ESI–MS/MS) were used to determine anthocyanin and phenolic composition. Anthocyanin content was 329.0 ± 28.0 mg cyanidin 3-glucoside equivalents/100 g (fresh weight) FW and total phenolic content was 758.6 ± 62.3 mg gallic acid equivalent/100 g FW. Cyanidin 3-galactoside was the major anthocyanin while the most abundant non-anthocyanin phenolic was chlorogenic acid.  相似文献

3-O-caffeoylquinic acid, 5-O-caffeoylquinic acid (chlorogenic acid), and 5-O-caffeoylquinic acid methyl ester, as elucidated by 1H and 13C NMR, were isolated from leaves of Etlingera elatior. This is the first report of caffeoylquinic acids (CQA) including chlorogenic acid (CGA) in Zingiberaceae. Leaves of Etlingera species were rich in total phenols and CQA, and non-cytotoxic to normal human liver and African green monkey kidney cells. Content of CQA of E. elatior, Etlingera fulgens, and Etlingera rubrostriata leaves was significantly higher than leaves of Ipomoea batatas, and comparable to flowers of Lonicera japonica. CGA found only in leaves of E. elatior and E. fulgens was significantly higher in content than flowers of L. japonica, the commercial source.  相似文献

Antibacterial effects of American cranberry (Vaccinium macrocarpon) concentrate on foodborne pathogens, Escherichia coli O157:H7, Listeria monocytogenes, Salmonella Typhimurium, and Staphylococcus aureus in vitro were investigated. Cranberry concentrate at various concentrations was prepared in distilled water (DW) or Brain Heart Infusion (BHI) broth. Pathogens were inoculated in each sample and incubated at 21 and 4 °C for 0, 1, 5, 7, and 24 h (DW samples) and 0, 1, 3, and 5 days (BHI samples). Transmission electron microscopy (TEM) was used to study the effects of cranberry concentrate on cellular structure of pathogens. DW results showed that S. Typhimurium and L. monocytogenes were reduced to non-detectable levels at 5 h in 100 μl/ml treatment at 21 and 4 °C. At 24 h, no target pathogens were detected from the 100 μl/ml treatment. BHI data indicated that the 100 μl/ml treatment reduced the four pathogens by 3-8 log CFU/ml compared with the control on Day 5 at 21 and 4 °C. TEM revealed damage to the bacterial cell walls and membranes. Cranberry concentrate has antibacterial effects on the four foodborne pathogens. Based on potential health benefits and proven antimicrobial effects, American cranberry concentrate may have dual applications as a food preservative.  相似文献

Mugua, fruit of the genus Chaenomeles, is a valuable source of health food and Chinese medicine. To elucidate the bioactive compounds of five wild Chaenomeles species, extracts from fresh fruits were investigated by HPLC-DAD/ESI-MS/MS. Among the 24 polyphenol compounds obtained, 20 were flavan-3-ols (including catechin, epicatechin and procyanidin oligomers). The mean polymerisation degree (mDP) of procyanidins was examined by two acid-catalysed cleavage reactions; the mDP value was the highest in Chaenomeles sinensis and the lowest in Chaenomeles japonica. Total polyphenol content (TPC) reached 46.92 and 46.28 mg/g gallic acid equivalents (GAE) in Chaenomeles speciosa and Chaenomeles thibetica, respectively, although their main bioactive compounds were different (being epicatechin and procyanidin B2 in the former, and catechin and procyanidin B1 in the latter). These two species also exhibited equally strong free radical scavenging activities. Our results further showed that the antioxidant ability of Chaenomeles fruits was significantly correlated to their total polyphenol contents. Two triterpenes (oleanolic acid and ursolic acid) were the highest quantity in Chaenomeles cathayensis and C. thibetica, respectively.  相似文献

Anthocyanins are polyphenols that are widely distributed in plants, and contribute to the brilliant blue, red or purple colour in leaves, flowers or fruits. Their attractive colours and their abundance in the north-east of China make the berries of Vaccinium uliginosum a new and excellent source of natural pigments. The pigments were extracted from the V. uliginosum berry using 0.5% TFA in ethanol solution, purified by an X-5 resin column and characterised by HPLC–DAD analysis. All of the peak assignments were confirmed by low-resolution electrospray mass spectrometry. The results showed that 11 anthocyanins and 2 flavonols were present in the V. uliginosum berry.  相似文献

The anthocyanin composition of tamarillo (Solanum betaceum Cav., red variety) and Andes berry (Rubus glaucus Benth.) was determined by HPLC–PDA and HPLC–ESIMS. From the anthocyanin-rich extracts (AREs), pure compounds (17) were obtained by MLCCC (multilayer countercurrent chromatography) and further preparative HPLC, and their unequivocal structures were obtained by 1D and 2D NMR analyses. The new anthocyanin delphinidin 3-O-α-l-rhamnopyranosyl-(1 → 6)-β-d-glucopyranoside-3′-O-β-d-glucopyranoside, as well as the known cyanidin-3-O-rutinoside, pelargonidin-3-O-rutinoside, and delphinidin-3-O-rutinoside were identified as constituents of tamarillo fruit. Although the anthocyanin composition of Andes berry had been reported before in the literature, the unequivocal structure elucidation of the major compound, cyanidin-3-O-α-l-rhamnopyranosyl-(1 → 6)-O-β-d-xylopyranosyl-(1 → 2)-β-d-glucopyranoside, was achieved for the first time.  相似文献

The TaqMan real-time PCR has the highest potential for automation, therefore representing the currently most suitable method for screening, allowing the detection of fraudulent or unintentional mislabeling of species. This work describes the development of a real-time polymerase chain reaction (RT-PCR) system for the detection and identification of common octopus (Octopus vulgaris) and main substitute species (Eledone cirrhosa and Dosidicus gigas). This technique is notable for the combination of simplicity, speed, sensitivity and specificity in an homogeneous assay.  相似文献

Fruit stones and pericarps of Litchi chinensis, waste products of the food, were studied as a source of polymeric proanthocyanidins. The structures of the polymeric proanthocyanidins isolated from Litchi chinensis were characterized by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) coupled with high performance liquid chromatography electrospray ionization mass spectrometry (HPLC-ESI-MS) analysis. The spectra obtained through MALDI-TOF MS analysis revealed that the dominant A-type procyanidin polymers occurred in each polymer length with one or more A-type linkages. The polymeric proanthocyanidins of litchi fruit stones exhibited longer polymer length than those of fruit pericarps, with polymerization degrees up to 20 and 11 for fruit stones and pericarps, respectively. After depolymerization with toluene-??-thiol, HPLC-ESI-MS analysis showed that epicatechin and A-type dimer were the major constituent units, and the mean polymerization degrees were 15.4 and 5.8 for polymeric proanthocyanidins of fruit stones and pericarps, respectively. The antioxidant properties investigated using DPPH, ABTS and FRAP methods showed that the higher polymerization degree of polymeric proanthocyanidins from litchi fruit stones exhibited higher antioxidant activities than those from litchi pericarps.  相似文献

The chemical composition of the hydrodistillate of aerial parts of Cymbopogon jawarancusa, a natural grass considered as major forage for animal nutrition, used in food because of the presence of sufficient concentration of minerals like calcium and potassium was analysed by capillary GC–FID, GC–MS and 13C NMR. Seventeen constituents representing 97.8% of the total oil with piperitone (58.6%) and elemol (18.6%) as major constituents were identified. In vitro cytotoxicity of the oil and its constituents on human cancer cell lines THP-1 (leukemia), A-549 (lung), HEP-2 (liver) and IGR-OV-1 (ovary) was evaluated by Sulphorhodamine-B assay. The oil was found to be more potent than its components against cancer cell lines tested with IC50 of 6.5 μg/ml (THP-1), 6.3 μg/ml (A-549), 7.2 μg/ml (HEP-2) and 34.4 μg/ml (IGR-OV-1). Antioxidant activity of oil and its constituents was evaluated by DPPH assay. In conclusion, the results demonstrate potent cytotoxic and antioxidant effects of oil, and its components like piperitone, α-pinene, β-caryophyllene and β-elemene.  相似文献
