Fourteen red-fleshed plum (Prunus salicina Erhr. and hybrids) and eight peach [Prunus persica (Batsch) L.] genotypes were characterized for their total phenolic and anthocyanin contents. Selected rich phenolic genotypes showed high antioxidant activity, stable colour properties and good antimicrobial activity. Results indicated positive correlations between phenolic compounds (r2 = 0.83) and antioxidant activity for both types of fruit. Colorants prepared from an anthocyanin rich plum genotype showed similar hue to that of synthetic colorant FD&C red 3 and higher stability than a commercial red grape colorant with respect to time, temperature and pH. Additionally, a selected rich phenolic plum genotype exhibited strong antimicrobial activity against Salmonella Enteritidis and Escherichia coli O157:H7. This study proposes that selection of crops high in phenolic compounds can be related to enhanced functional properties and opens the possibility of breeding fruits with targeted functional properties for the fresh produce and processing market.  相似文献

Using “natural green” plant extracts or their derived products in various food and beverage applications is an increasing trend in the food industry. Selection of these plant extracts and their application depends on their functional properties, availability, cost effectiveness, consumer awareness and their effect on the sensory attributes of the final product. Green tea extract (GTE) and grape seed extract (GSE) are two popular plant extracts that have been widely used in various food and beverage applications. Green tea is a widely consumed beverage that has attracted more attention in the recent years due to its health benefits like antioxidant, antimicrobial, anticarcinogenic and anti-inflammatory properties. Grape seed extract is derived from the grape seeds that is extracted, dried and purified to produce polyphenolic compounds-rich extract that also has well documented antioxidant, antimicrobial and anti-inflammatory properties. These two plant extracts (polyphenolic and proanthocyanidin rich compounds) have potential antioxidant properties by inhibiting the lipid oxidation and warmed over flavors and antimicrobial activities against major food borne pathogens like Listeria monocytogenes, Salmonella Typhimurium, Escherichia coli O157:H7, and Campylobacter jejuni in preventing pathogen contamination. Furthermore, they have demonstrated synergism in antimicrobial activity when used in combination with organic acids (malic, tartaric acid, benzoic acids etc.), bacteriocins like nisin or chelating agents like EDTA in various model systems including fresh produce (fruits and vegetables), raw and ready-to-eat meat and poultry products. Apart from beneficial effects of grape seed extract in food safety and quality, concerns regarding the side effects of GSE are also addressed. Nevertheless, persistent recalls of the food products involving foodborne pathogens despite various control measures calls for efficient bacteriostatic and bactericidal agents and technologies to deliver the active components for an effective inhibition of pathogens. Therefore, further research involving electrostatic spray and nanoscale delivery of the active components present in these natural, green, plant extracts and using them as a component in multiple hurdle approach would enhance the food safety and quality in addition to providing alternative “green” solutions to the food processors.  相似文献

Free radical scavenging and antibacterial activities were determined for several red wines from different grape varieties from the southern Serbia wine region. The amount of polyphenols and anthocyanins in selected wines was investigated and the potential antibacterial activity of the wines against pathogenic strains of Staphylococcus aureus and Escherichia coli was also determined. The free radical scavenging and antibacterial activity of the wines was correlated with polyphenolic and anthocyanin content. The significant differences in the antioxidant and antibacterial activities between the wines from grape cultivars (Cabernet Sauvingon and Pinot Noir) and the indigenous varieties (Vranac and Prokupac) were not confirmed. The hypothesis of a protective effect of red wine is supported by our results that show that the red wines with higher amounts of polyphenols and anthocyanins had higher antioxidant and antibacterial properties.  相似文献

The phenolic composition, antioxidant activity, and antimicrobial activity of extracts from vine leaves of six grape varieties collected in May, August, and September was studied. The phenolic potential of the extracts was dependent on variety and picking-time. Extracts of leaves collected in September were the richest in total phenols, flavonoids, flavonols, and stilbenes. The antioxidant properties determined by ferric reducing antioxidant power and 2,2-diphenyl-1-picrylhydrazyl assays, and antimicrobial activity against Stapylococcus aureus, Bacillus cereus, Campylobacter jejuni, Escherichia coli, and Salmonella Infantis were good and in correlation with the chemical composition changes of the leaf extracts. The results indicated that leaves remaining on the vine in September after the grape harvest could be especially promising as an inexpensive source of effective antioxidant/antimicrobial agents.  相似文献

This study was aimed to investigate the influence of solvent (water and ethanol in concentration: 50%, 70% and 96%) and extraction temperature (25–80) °C on polyphenols extraction of grape seed (Vitis vinifera L. cv. “Frankovka”) cultivated in Eastern Croatia. The best results were reached using 50% ethanol at 80 °C. The most abundant individual polyphenolic compound was catechin which makes the average of 45.11% of the total phenolic content followed by epicatechin (34.45%), procyanidin B2 (12.90%), gallic acid (5.34%), gallocatechin (1.58%), epicatechin gallate (1.01%). The antioxidant activity of extracts was determined by DPPH‐method and it was highly dependent on extraction solvent and temperature. The total extractable proanthocyanidins content of extract had the highest positive correlation (r = 0.994) with antioxidant activity, which indicated that these compounds might be the most important antioxidant in examined grape seed extracts. Relationship between phenolic content and colour of extracts was observed.  相似文献

The extracts of crude polyphenols (seeds, pulp + skin, whole) from four different cultivars of Indian grapes were used in this study. The total polyphenolic contents of grape polyphenolic extracts (GPEs) were determined and their in vitro antioxidant and anti-initiating activities evaluated. The total polyphenolic contents, expressed in terms of gallic acid/catechin/procyanidin B3 equivalents, were found to vary significantly. Antioxidant activity of GPEs, particularly the seedless variety, was evident from significant dose-dependent inhibition of lipid peroxidation and DPPH activity. GPEs and catechin inhibited the microsome-catalysed activity of cytochrome P450 isozymes (1A1, 1A2, 2B1) in a dose-dependent manner, by the decreased formation of resorufin. The inhibitory activity of GPEs on nitrite-mediated N-nitrosation of dimethylamine and N-methylaniline appears to correlate significantly with the total polyphenolic contents. Furthermore, six individual polyphenols present in GPEs were quantitated by HPLC, wherein procyanidin B3 was a major constituent.  相似文献

Antimutagenic and antimicrobial activities of pu-erh tea   总被引:2,自引:0,他引:2  
The biological action of water extract of pu-erh tea (WEPT) was evaluated by Salmonella mutagenesis assay and bacteria test. Like green tea, oolong tea and black tea, WEPT showed neither cytotoxicity and nor mutagenicity toward Salmonella typhimurium TA98 and TA100 with or without S9 mix (an external metabolic activation system). WEPT at 0.5-5 mg/plate expressed a dose-dependent inhibitory effect against both the mutagenicity of aflatoxin B1 (AFB1), an indirect mutagen which requires metabolic activation, and 4-nitroquinoline-N-oxide (NQNO), a direct mutagen, in Salmonella typhimurium TA98 and TA100. In general, the antimutagenic activity of WEPT against AFB1 and NQNO was weaker than other tea extract because of the least amount of total catechin in WEPT. For antimicrobial action, WEPT, green tea, oolong tea and black tea at 2.0 mg/ml showed inhibitory effect on growth of Staphylococcus aureus and Bacillus subtilis, but no effect on Escherchia coli. Obviously, WEPT has potential antimicrobial effect on gram-positive Staphylcoccus aureus and B. subtilis than that of gram-negative E. coli. In addition, caffeine and epicatechin (EC), main polyphenolic compounds in WEPT, showed both antimutagenic and antimicrobial effect against strains mentioned above, which may partially account for the antimutagenic and antimicrobial action of pu-erh tea.  相似文献

The polyphenolic content and antioxidant capacity in the seeds of 11 red grape varieties (five international and six native) widely cultivated in Turkey were investigated. Total phenolic, total flavanol and total polymeric procyanidin content ranged from 79.2 to 154.6, 89.2 to 179.4, and 27.0 to 43.3 mg/g seed, respectively. While (+)-catechin (4.71–23.8 mg/g seed) was found as main flavanol, galloylated catechin monomer and dimeric procyanidin amounts varied between 2.89–17.2 and 0.97–2.97 mg/g seed, respectively. All seed extracts showed remarkable DPPH radical scavenging activity (EC50) and oxygen radical scavenging capacity (ORAC) ranging from 2.71 to 4.62 μg/mL and 1425.9 to 3009.2 μmol Trolox equivalent/g seed, respectively. With high amount of total phenolic content and antioxidant activity, seeds of Okuzgozu, Papaz Karasi, Ada Karasi and Kalecik Karasi varieties could be evaluated as dietary supplement.  相似文献

Crude polyphenols were extracted from tobacco leaf by 80% ethanol solution with ultrasonic treatment and then purified by a macroporous resin. The polyphenols from tobacco leaf (PTL) were subjected to analyses by reverse-phase high-performance liquid chromatography (RP-HPLC) and electrospray ionization mass spectrometry (ESI-MS). The dominant polyphenols in tobacco leaf were identified as chlorogenic acid and rutin. Furthermore, the antioxidant activities of PTL were investigated, including scavenging activities of 1,1-diphenyl-2-picrylhydrazyl (DPPH) radicals (5.02 μg/ml IC50 value), hydroxyl radicals (49.6 μg/ml IC50 value) and superoxide anion radicals (44.0 μg/ml IC50 value), inhibition activity of lipid peroxidation (132 μg/ml IC50 value) and reducing power. The proliferation inhibition activities on Escherichia coli, Staphylococcus aureus and Bacillus subtilis were also measured for evaluating the antimicrobial activity of PTL. The diameters of inhibition zones were 20.23 ± 0.42, 17.66 ± 0.86 and 12.89 ± 0.29 mm, respectively. The results showed that PTL had great potential as antioxidant and antimicrobial agent.  相似文献

Composition and antioxidant and antimicrobial activities of essential oil and methanol extract polar and nonpolar subfractions of Stachys inflata were determined. GC and GC/MS analyse of the essential oil showed 45 constituents representing 95.46% of the oil, the major components linalool (28.55%), α-terpineol (9.45%), spathulenol (8.37%) and (2E)-hexenal (4.62%) constituted 50.99% of it. Essential oil and extracts were also tested for their antioxidant activities using 2,2-diphenyl-1-picrylhydrazyl (DPPH) and β-carotene/linoleic acid assays. In the DPPH test, IC50 value for the polar subfraction was 89.50 μg/ml, indicating an antioxidant potency of about 22% of that of butylated hydroxytoluene (IC50 = 19.72 μg/ml) for this extract. In β-carotene/linoleic acid assay, the best inhibition belonged to the nonpolar subfraction (77.08%). Total phenolic content of the polar and nonpolar extract subfractions was 5.4 and 2.8% (w/w), respectively. The plant also showed a week antimicrobial activity against three strains of tested microorganisms. Linalool and α-terpineol were also tested as major components of the oil and showed no antioxidant but considerable antimicrobial activities.  相似文献

Phenolic compounds, related to antioxidative and antifungal properties of ethanolic extracts from five commercial grape cultivars (three red and two white) grown in Turkey were determined. A reversed-phase high performance liquid chromatography (RP-HPLC) procedure was developed, and a total 18 different phenolic compounds were identified. Total phenolic contents of the extracts were determined using Folin–Ciocalteau method. Antioxidant activities of the extracts were evaluated by using DPPH radical scavenging and phosphomolybdenum methods. All extracts exhibited strong antioxidant and antiradical activity. Phenolic compounds and antioxidant activities of the extracts were variety dependent. Antifungal activities of the pomaces and extracts were screened by both in vitro agar-well diffusion assay and antifungal activity in apple and orange juices in situ using Zygosaccharomyces rouxii and Z. bailii. Antifungal activities revealed that the pomaces and extracts of Gamay and Kalecik karasi could be more effective antifungal agents than those of Emir, Narince and Okuzgozu grape cultivars.  相似文献

The aim of this study is to investigate in vitro antioxidant, antimicrobial, and anticancer activities of the acetone extracts of the lichens Cladonia furcata, Cladonia pyxidata and Cladonia rangiferina and their atranorin and fumarprotocetraric acid constituents. Antioxidant activity was evaluated by free radical scavenging, superoxide anion radical scavenging, reducing power, and determination of total phenolic and flavonoid compounds. As a result of the study atranorin had largest free radical scavenging activity with IC50 values 131.48 μg/mL. Moreover, the tested samples had effective reducing power and superoxide anion radical scavenging. Total content of phenol and flavonoid in extracts was determined as pyrocatechol equivalent, and as rutin equivalent, respectively. The strong relationships between total phenolic and flavonoid contents and the antioxidant effect of tested extracts were observed. The antimicrobial activity was estimated by determination of the minimal inhibitory concentration by the broth microdilution method. The most active was fumarprotocetraric acid with minimum inhibitory concentration values ranging from 0.031 to 0.125 mg/mL. Anticancer activity was tested against FemX (human melanoma) and LS174 (human colon carcinoma) cell lines using MTT method. All samples were found to be strong anticancer activity toward both cell lines with IC50 values ranging from 10.97 to 41.23 μg/mL.  相似文献

Ten hydroalcoholic extracts of edible plants from the Calabria region (Italy) were evaluated for their in vitro antioxidant and antiradical properties and in vivo topical anti-inflammatory activity. All the extracts had radical-scavenging and/or antioxidant properties, the most active plants being hawkweed oxtongue and viper’s bugloss. The best free radical (DPPH·)-scavenging activity was found in hawkweed oxtongue and chicory leaves extracts (IC50 = 25 and 26 μg/ml, respectively). Hawkweed oxtongue, poppy and viper’s bugloss extracts showed the greatest inhibition of linoleic acid oxidation (IC50 = 3 μg/ml). Viper’s bugloss and hawkweed oxtongue extracts had the greatest antioxidant effect on bovine brain peroxidation (IC50 = 11 and 22 μg/ml). All the extracts also showed an anti-inflammatory effect: 300 μg/cm2 provoked oedema reductions ranging from 18% to 43%. Cress was the most active plant. Chicory leaves contained the highest amount of phenolics (190 mg/g) whilst Rush crimps contained the highest amount of flavonoids (32.9 mg/g), followed by hawkweed oxtongue (15.8 mg/g). Cress contained the highest number of sterols. Among them, γ-sitosterol (12.2%) and ergost-5-en-3-ol (3β) (4.5%) were found to be the major constituents. Moreover, three of the identified molecules (stigmasta-5,23-dien-3β-ol, stigmasta-5,24(28)-dien-3-ol (3β,22E) and 9,19-cyclolanost-24-en-3-ol (3β)) were found in this plant only.  相似文献

Aqueous extract, proanthocyanidin rich extract, and organic extracts of Cymbopogon schoenanthus L. Spreng (lemon grass) shoots from three different locations in South Tunisia were screened for their antioxidant, acetylcholinesterase and antimicrobial activities. In addition to the evaluation of these activities, the contents of flavonoids and total phenolic compounds were determined.Antioxidant activity measured by DPPH assay showed that the proanthocyanidin extract exhibited higher antioxidant activity than the aqueous extract. Extract concentration providing 50% inhibition (IC50) ranged from 16.4 ± 6.8 μg/mL to 26.4 ± 6.8 μg/mL. The antioxidant activity was also determined using the β-carotene/linoleic acid bleaching test. The best results (IC50 = 0.11 ± 0.10 mg/mL) were obtained with the proanthocyanidin extract of the plants collected from the desert region (Dhibat).The greatest acetylcholinesterase inhibitory activity (IC50 = 0.23 ± 0.04 mg/mL) was exhibited by the ethyl acetate and methanol extracts of the plants collected from the mountainous region. It seems that extracts obtained with more polar solvents gave better results.The proanthocyanidin extracts showed a good antimicrobial activity against Streptococcus sobrinus at low concentration (MIC = 4 mg/mL). Therefore, these extracts could be used to prevent carious lesions by inhibiting S. sobrinus growth.  相似文献

Phenolic antioxidants seem to be partly responsible for the protective effects against cardiovascular diseases attributed to moderate wine consumption. Grape skins greatly contribute to the phenolic composition of red wine. In this paper, the in vitro antioxidant activity of red grape (Vitis vinifera) skins is determined. We show that the radical scavenging activity (C 50 values) against 2,2-diphenyl-1-picrylhydrazyl (DPPH·) of grape skin extracts is relatively high (3.2–11.1 mg dried skin/mg DPPH·) in relation to other foodstuffs and, as expected, is influenced by grape variety, stage of grape ripening and vintage. The antioxidant potential of grape skins seems to be transferred into wine since grape varieties with skins exhibiting high antioxidant potential also resulted in wines with high antioxidant activity. Statistically significant correlations were found between antioxidant activity and phenolic content (total polyphenols, proanthocyanins, catechins and anthocyanins) for both grape skins and wines.  相似文献

The present study investigated the antioxidant and cytoprotective effects of purified ethanolic extracts of Cytisus multiflorus, Lamium album L. and Thymus citriodorus plants. These extracts showed high antioxidant activity in DPPH and reducing power assays. Using a model of chemical stress induced by potassium dichromate (DK) in human hepatoblastoma HepG2 cells, 50 μg/mL of C. multiflorus, L. album and T. citriodorus extracts decreased the rate of reactive oxygen species (ROS) production by 35%, 26% and 20%, respectively, when exposed to 25 μM of DK. This effect was also observed for the treatment of cells with individual polyphenolic compounds determined in the extracts, or with mixtures prepared with individual polyphenolic compounds simulating the phenolic composition of the extracts. Additionally, the purified ethanolic extracts and the prepared polyphenolic mixtures showed a cytoprotective effect against DK-induced toxicity. The overall results emphasize the contribution of polyphenols in antioxidant and cytoprotective properties of the studied plants.  相似文献

Grape seed extracts (GSEs) were investigated in yeast cells harbouring defects in their antioxidant system (regarding the cellular growth and growth recovery from H2O2 insult). GSEs antioxidant activity was detected in wild-type and mutant strains Δcta1, Δgsh1 and Δoye2glr1, while pro-oxidant activity in Δsod1 cells was seen. Assessment of proliferation of prostate cancer PC3 and HBV-replicating HepG2 2.2.15 cells treated with GSEs has shown higher cytotoxicity of red grape seed extract (RW) than white grape seed extract (WW) subjective to dose and period of administration. No antiviral effect was detected by measuring the secreted virion particles in HepG2 2.2.15 cells treated with GSEs. The GSEs play a dual antioxidant/pro-oxidant role in vivo according with the cellular antioxidant system deficiencies and exhibit cytotoxic properties in PC3 and HepG2 2.2.15 cell lines, but no antiviral action against HBV.  相似文献

ABSTRACT: Antidesma thwaitesianum Müll. Arg. or mao is widely used as commercial products of juice and wine in Thailand. As a result, waste products from the mao plant, such as mao seeds (MS) and mao marcs (MM), are plentiful. We aimed to purify and analyze polyphenolic content in both MS and MM and to investigate the radical scavenging activities of these polyphenolics against 1,1‐diphenyl‐2‐picryl‐hydrazyl (DPPH) and 2,2′‐Azinobis (3‐ethylbenzothiazoline 6‐sulphonate) (ABTS) radicals and thiobarbituric acid reactive products (TBARP). The results showed MS and MM to be an abundant source of polyphenols (97.32 to 130 mg gallic acid equivalents [GAE]/g) and proanthocyanidins. The radical scavenging activities of MS/MM against DPPH and ABTS radicals (IC50 of 0.85 to 1.21 μg/mL) were significantly higher (P < 0.05) than that of standard trolox (IC50 of 5.05 μg/mL). Activity of MS/MM extracts were 3.74 and 3.80 μg/mL trolox eq/g f.w. for the DPPH and ABTS assays, respectively. The oxidation of erythrocyte membranes using 2‐thiobarbituric acid demonstrated that the protective effect of MS/MM on lipid peroxidation is as strong as grape seed proanthocyanidin extract. These findings suggest that polyphenolic compounds and proanthocyanidins isolated from these mao extracts had much higher antioxidant activities than those of standard trolox and exhibited similar antioxidant potential to grape seed proanthocyanidin extract. These findings may also increase value of mao waste products and allow development of commercial health products.  相似文献

The methanolic extracts of 16 Helichrysum species were investigated for their in vitro antioxidant, radical scavenging and antimicrobial activities. All the extracts showed strong antioxidant and radical scavenging activity. The highest total antioxidant capacity as ascorbic acid equivalents (AAE) of 194.64 mg/g dry extract was obtained for Helichrysum noeanum in the phosphomolybdenum assay. The highest IC50 value (7.95 μg/ml) was observed for the extract of Helichrysum stoechas subsp. barellieri in the DPPH assay. The total phenolic contents of the extracts ranged from 66.74 to 160.63 mg gallic acid equivalents (GAE)/g dry extract. The major component present in the extracts was identified as chlorogenic acid followed by apigenin-7-glucoside and apigenin by HPLC analysis. All the extracts showed significant antimicrobial activity against microorganisms containing 13 bacteria and two yeasts in the agar diffusion method.  相似文献

The influence of two extracts of grape skin and seeds from Vitis vinífera L. cv. País (Chilean black grapes), rich in proanthocyanidins (PAs), was evaluated on the inhibition of the angiotensin I-converting enzyme (ACE), and the inhibition was related to the type and number of subunits of the polymeric PAs chain. Size exclusion chromatography was used to purify the extract and its characterization was made by acid catalysis depolymerization followed by HPLC. ACE activity was measured by quantitative HPLC, measuring the hyppuric acid (HA) produced from the hydrolysis of hippuryl-l-histidyl-l-leucine (HHL) by ACE. Structural compositions differed significantly between both extracts: the skin extracts do not exhibit epicatechin (EC) and epicatechin gallate (ECG), and the seed extracts did not present epigallocatechin (EGC). Skin extracts have a higher mean degree of polymerization (mDP) than seed extracts and a higher inhibition power (IC50 = 0.14 ± 0.03 μM and IC50 = 0.480 ± 0.03 μmol/L), respectively. The catechin (IC50 = 1495 ± 90 μmol/L) and epicatechin (IC50 = 1772 ± 121 μmol/L) monomers exerted lower inhibition than the either grape extract. The structural differences between grape skin and seed PAs could influence the ACE inhibition capacity. The larger inhibitory power of skin extract was associated to greater OH availability, higher mDP and the presence of EGC.  相似文献
