In an effort to find a new whitening agent, we have found that the methanol extract of the dried skin of Allium cepa showed inhibition of melanin formation. Bioassay-guided fractionation led to the isolation of quercetin (1) and quercetin 4’-O-β-glucoside (3) from A. cepa as the inhibitors of melanin formation in B16 melanoma cells with IC50 values of 26.5 and 131 μM, respectively. In addition, we evaluated the effect of some quercetin derivatives, such as isoquercitrin (2), quercetin 3,4’-O-diglucoside (4), rutin (5) and hyperin (6) on B16 melanoma cells. These quercetin derivatives did not show any inhibition of melanin formation. Furthermore, the ORAC values of compounds 16 were 7.64, 8.65, 4.82, 4.32, 8.17 and 9.34 μmol trolox equivalents/μmol, respectively. Dried skin of red onion showed inhibitory activity against melanin formation in B16 melanoma cells, as well as antioxidant properties.  相似文献

There is increasing evidence that flavonols demonstrate beneficial properties for human health. Quercetin is the major flavonol present in onion (Allium cepa cv) and is present predominantly as quercetin 3,4′-diglucoside and quercetin 4′-monoglucoside. These compounds are known to be potent free radical scavengers and antioxidants, and are considered to be protective against cardiovascular disease. Analysis for the presence of these compounds has therefore become more important. Robust capillary zone electrophoresis and high performance liquid chromatography procedures were developed for profiling and quantifying the levels of quercetin 3,4′-diglucoside and quercetin 4′-monoglucoside in 70% methanol/water extracts of six different onion varieties available in Victoria, Australia. Quercetin 3,4′-diglucoside, which is not commercially available as a reference standard, was isolated from freeze-dried onion powder by preparative high performance liquid chromatography and used to quantify the levels in the onion extracts. Significant differences in the levels and ratios of the two compounds were seen between red, brown and white onion varieties (e.g. ‘Redwing’; quercetin 3,4′-diglucoside 191 mg/100 g DW, quercetin 4′-monoglucoside 85 mg/100 g DW; ‘Cream Gold’, quercetin 3,4′-diglucoside 153 mg/100 g DW, quercetin 4′-monoglucoside 58 mg/100 g DW, ‘Spanish white’; quercetin 3,4′-diglucoside <1 mg/100 g DW, quercetin 4′-monoglucoside <1 mg/100 g DW).  相似文献

Antimicrobial activity of different concentrations (50, 100, 200, 300 and 500 ml/l) of essential oil extracts of three type of onions (green, yellow and red) and garlic against two bacteria, Staphylococcus aureus, Salmomella Enteritidis, and three fungi, Aspergillus niger, Penicillium cyclopium and Fusarium oxysporum, was investigated. The essential oil (EO) extracts of these Allium plants (garlic and onions) exhibited marked antibacterial activity, with garlic showing the highest inhibition and green onion the lowest. Comparatively, 50 and 100 ml/l concentrations of onions extracts were less inhibitory than 200, 300 and 500 ml/l concentrations. However, with garlic extract, high inhibitory activity was observed for all tested concentrations. S. aureus showed less sensitivity towards EO extracts inhibition, however S. Enteritidis was strongly inhibited by red onion and garlic extracts. The fungus F. oxysporum showed the lowest sensitivity towards EO extracts, whereas A. niger and P. cyclopium were significantly inhibited particularly at low concentrations. Conclusively, where seasoning is desired, essential oil extracts of onions and garlic can be used as natural antimicrobial additives for incorporating in various food products.  相似文献

Four (red, violet, white and green) varieties of Allium cepa were studied for their total phenolic contents (TPC), antioxidant (AOA) and free radical scavenging activities (FRSA). The TPC varied from 4.6 to 74.1 mg/g GAE, AOA varied from 13.6% to 84.1% and FRSA showed wide range in terms of IC50 (inhibitory concentration) from 0.1 to 15.2 mg/ml, EC50 (efficient concentration) from 4.3 to 660.8 mg/mg and ARP (antiradical power) from 0.15 to 23.2. The outer dry layers of red and violet varieties showed better inhibition of lipid peroxidation assayed by ammonium thiocyanate than α-tocopherol. The non-site-specific inhibition of hydroxyl radical induced deoxyribose degradation was also higher in the outer dry layers of red and violet varieties than in their middle and inner layers. The outer layers were also potential inhibitors of nitroblue tetrazolium chloride (NBT) reduction caused by superoxide anions. On the other hand the ferrous ion chelating capacity of the red and violet varieties was highest in the inner layers. Specific phenolic composition performed through HPLC and LC–MS/MS showed the presence of gallic acid, ferulic acid, protocatechuic acid, quercetin, and kaempferol. The unutilised outer layers of the red variety were a rich source of quercetin (5110 μg/g) with high AOA, FRSA and also showed significant protection of DNA damage caused by free radicals.  相似文献

A fructan that acts as an anti-influenza A virus substance was isolated from hot water extract of the green leafy part of a Welsh onion (Allium fistulosum L.). The structure of the fructan was characterised and elucidated by chemical and spectroscopic analyses. The fructan was composed of terminal (21.0%) and 2,1-linked β-d-Fruf residues (65.3%) with 1,6-linked β-d-Glcp residues (13.7%). The molecular weight of the polysaccharide and polydispersity was estimated to be 1.5 × 103 and 1.18, respectively. Although the fructan did not show anti-influenza A virus activity in vitro, it demonstrated an inhibitory effect on virus replication in vivo when it was orally administered to mice. In addition, the polysaccharide enhanced the production of neutralising antibodies against influenza A virus. Therefore, the antiviral mechanism of the polysaccharide seemed to be dependent on the host immune system, i.e., enhancement of the host immune function was achieved by the administration of the polysaccharide. From our observations, the fructan from Welsh onions is suggested to be one of the active principles which exert an anti-influenza virus effect.  相似文献

This work reports preliminary results on the potential of a metal oxide sensor (MOS)-based electronic nose, as a non-destructive method to discriminate three “Tropea Red Onion” PGI ecotypes (TrT, TrMC and TrA) from each other and the common red onion (RO), which is usually used to counterfeit. The signals from the sensor array were processed using a canonical discriminant function analysis (DFA) pattern recognition technique. The DFA on onion samples showed a clear separation among the four onion groups with an overall correct classification rate (CR) of 97.5%.  相似文献

The polysaccharide fraction extracted with simulated gastric juice from onion bulbs contained a mixture of galactan with short-length sugar chains, pectic polysaccharides and evident content of proteinaceous material. Galacturonan and rhamnogalacturonan were the main constituents of the linear regions of the sugar chains of the pectic polysaccharides. The ramified regions included rhamnogalacturonan-I. NMR data revealed that the side chains of the ramified region contained mainly 1,4-linked β-d-galactopyranose residues and lesser content of 1,3-linked β-d-galactopyranose and 1,5-linked α-l-arabinofuranose residues. Furthermore, the proteinaceous material was determined to be partly linked to the sugar chains.  相似文献

A separation/enrichment procedure has been developed for the determination of selenium in garlic and onion samples by electrothermal atomic absorption spectrometry (ET-AAS) as a slurry sampling after preconcentration with 3,3-diaminobenzidine (DAB) reagent on the activated carbon. The influences of pH, time, amount of carbon and complexing reagent were outlined. The effect of acids used in the digestion of samples was also studied and compared. Selenium level was found to be 0.024 μg g−1 for onion (n = 5; LOD – 0.5 μg L−1; LOQ – 1.7 μg L−1) and 0.015 μg g−1 for garlic (n = 5; LOD – 1.3 μg L−1; LOQ – 3.3 μg L−1). Three different samples of garlic were analyzed by k0-instrumental neutron activation analysis (k0-INAA) at the Jozef Stefan Institute (JSI). The data obtained by k0-INAA show that the content of selenium overlapped the results obtained by ET-AAS.  相似文献

Chinese chive seeds (Allium tuberosum Rottl.) (grown in China) were investigated. Density, thousand-grain weight, and hectolitre weight of seeds were 1.27 g/cm3, 4.9 g, and 71 kg/100 l, respectively. The results showed that Chinese chive seeds contained high amounts of oil (15.8%), dietary fibre (18.2%) and crude protein (12.3%). Oil of seeds was composed of 10.1% saturated and 90.0% unsaturated fatty acids. Linoleic(69.1%) and palmitic (7.0%) were the most abundant unsaturated and saturated fatty acids, respectively. Chinese chive seeds contained 4.5 mg/kg of thiamin, 2.8 mg/kg of riboflavin and 55.1 mg/kg of niacin. The mineral contents of the seed of A. tuberosum, for iron, calcium and zinc, were 580 mg/kg, 1328 and 80.8 mg/kg, respectively. Analysis of the amino acid content of Chinese chive seed revealed that it was a rich source of the essential amino acids, isoleucine, tryptophan and lysine. The study revealed that Chinese chive seeds had high levels of nutritionally important components, such as oil, minerals and essential amino acids.  相似文献

Onion (Allium cepa L.) is an important vegetable crop consumed primarily for its ability to enhance the flavor of other foods. The quality of onion depends on its pungency. While highly pungent onions are popular in India, less pungent ones are preferred in other countries. However, the variability in pungency in bulbs of different cultivars however has not been investigated. The present investigation was therefore undertaken to determine the pungency in three popular Indian onion cultivars viz. N-2-4-1, B-780 and Phule Safed. Randomly selected bulb samples of the cultivars were analyzed for the content of pyruvic acid, total soluble solids (TSS) and reducing, non-reducing and total sugars. The red variety N-2-4-1 showed higher level of pungency, while other two varieties were comparatively less pungent.  相似文献

The aim of this work was to examine the chemical constituents and antioxidant potential of water-soluble fractions from the commonly consumed vegetable, Allium vineale. The water-soluble fraction, containing phenolic compounds, was extracted with ethyl acetate to obtain flavonoids which were separated and purified by repeated column chromatography over Sephadex LH-20, RP C18 and silica gel. The isolated compounds were identified according to their physicochemical properties and spectral data (UV, HPLC–TOF/MS, 1H NMR, 13C NMR and 2D NMR). Three flavonoids were isolated and identified as chrysoeriol-7-O-[2″-O-E-feruloyl]-β-d-glucoside (1), chrysoeriol (2), and isorhamnetin-3-β-d-glucoside (3). Antioxidant studies of the aqueous extract and three isolated compounds, 1, 2, 3, were undertaken and they were found to have significant antioxidant activity. Antioxidant activities were evaluated for total antioxidant activity by the ferric thiocyanate method, ferric ion (Fe3+) reducing antioxidant power assay (FRAP), ferrous ion (Fe2+) metal chelating activity, and DPPH free radical-scavenging activity. The water-soluble ethyl acetate and methanol extraction methods were also compared using HPLC–TOF/MS.  相似文献

A phytochemical investigation of the fresh bulbs of Allium nigrum L. led to the isolation of new spirostane-type glycosides as two inseparable isomer mixtures, nigrosides A1/A2 (1a/1b) and nigrosides B1/B2 (2a/2b), two new cholestane-type glycosides, nigrosides C and D (3 and 4), together with the known compounds, 25(R,S)-5α-spirostan-2α,3β,6β-trio1-3-O-β-d-glucopyranosyl-(1 → 2)-O-[β-d-xylopyranosyl-(1 → 3)]-O-β-d-glucopyranosyl-(1 → 4)-β-d-galactopyranoside (5a/5b) and 25(R,S)-5α-spirostan-2α,3β,6β-trio1 3-O-β-d-glucopyranosyl-(1 → 2)-O-[4-O-(3S)-3-hydroxy-3-methylglutaryl-β-d-xylopyranosyl-(1 → 3)]-O-β-d-glucopyranosyl-(1 → 4)-β-d-galactopyranoside (6a/6b), isolated from this plant for the first time. All structures were elucidated mainly by spectroscopic analysis (1D and 2D NMR experiments, FABMS, HRESIMS) and by comparison with literature data. Cytotoxicity of the isolated compounds was assessed against human colon carcinoma (HT-29 and HCT 116) cell lines. Compounds 5a/5b and 6a/6b were found to be the most active with IC50 values 1.09 and 2.82 μM against HT-29 and 1.59 and 3.45 μM against HCT 116, respectively.  相似文献

Seven flavonoid glycosides, kaempferol 3-O-α-L-rhamnopyranosyl (1→2)-[3-O-acetyl]-β-D-glucopyranoside (1), kaempferol 3-O-α-L-rhamnopyronosyl (1→2)-[6-O-acetyl]-β-D-glucopyranoside (2), kaempferol 3-O-α-L-rhamnopyranosyl-(1→2)-β-D-glucopyranoside (3), kaempferol 3-O-β-D-glucopyranoside (4), kaempferol 3,7-di-O-β-D-glucopyranoside (5), 7-O-β-D-glucopyranosyl kaempferol 3-O-α-L-rhamnopyranosyl-(1→2)-β-D-glucopyranoside (6), kaempferol 3-O-α-L-rhamnopyranosyl (1→2)-β-D-glucopyranoside-7-O-[2-O-(trans-p-coumaroyl)]-β-D-glucopyranoside (7) were isolated from the n-butanol fraction of Allium ursinum L. and the structures of these compounds were elucidated on the basis of mass spectrometry, 1H NMR, 13C NMR, HMQC and HMBC data. Among them, 1 and 2 are novel compounds and compounds 4 and 5 were isolated from this plant species for the first time.  相似文献

A steroidal saponin, named tuberoside A, together with six known compounds, were isolated from the seeds of Allium tuberosum Rottl. ex Spreng. On the basis of acid hydrolysis, comprehensive spectroscopic analyses and comparison with spectral data of known compounds, its structure was established as (24S, 25S)-5β-spirostan-2β,3β,24-triol 3-O-α-l-rhamnopyranoyl-(1→2)-O-[α-l-rhamnopyranosyl-(1→4)]-β-d-glucopyranoside. The six known compounds were thymidine, adenosine, 2-hydroxy purine, adenine, uracil, and thymine. 2-hydroxy purine, adenine, uracil and thymine are isolated from the seeds of A. tuberosum for the first time. This paper deals with the isolation and structural elucidation of the new saponin.  相似文献

The objective of this work was to analyse phenolic compounds and antiradical capacity of different parts of walnut fruit among six genotypes of Juglans regia L. Therefore, total phenolic and flavonoid content were determined and methanolic extracts of walnut genotypes were considered by the reducing power, DPPH (2,2-diphenyl-1-picrylhydrazyl), superoxide anion and nitric oxide radical scavenging. Significant differences were found in phenolic content and radical scavenging capacity of different parts of fruits and among various genotypes. High correlation coefficient (R2 = 0.81) was observed between phenol content and radical scavenging activity, but this was not always true (R2 = 0.01). These results demonstrated that walnut genotypes have different phenolic compounds and phenolic compounds have different radical scavenging power. The differences of phenolic compounds were confirmed by using high performance liquid chromatography (HPLC).  相似文献

St. John’s wort (Hypericum perforatum L.) herb is used as a herb-tea or as a food supplement because of its antidepressive properties. St. John’s wort, grown under different levels of UV-B radiation, was analysed for its concentrations of flavonoids, tannins and hypericin. A high level of UV-B radiation increased leaf concentrations of flavonoids from 6.31 to 9.00/100 g in dry matter (DM) and of tannins from 26.6 to 31.4/100 g in DM. The concentration of hypericin in leaves exposed to enhanced UV-B radiation (0.08/100 g in DM) was lower than that of leaves exposed to reduced UV-B radiation (0.09/100 g in DM). We conclude that the concentrations of bioactive substances in St. John’s wort herb can be altered by regulating the environment during plant growth.  相似文献

The behavioural responses of Sitophilus granarius, S. oryzae and S. zeamais to synthetic 4S,5R-sitophinone alone and in combination with volatiles from kibbled carob have been investigated with a view to the development of a single lure to attract all three species. Sitophilus zeamais and S. oryzae were shown to respond to 4S,5R-sitophinone at amounts as low as 0.025 ng. Sitophilus granarius gave a significant response to 50 ng 4S,5R-sitophinone. Volatiles from kibbled carob were also shown to attract all three species. This is the first time that attraction of S. zeamais and S. oryzae to carob volatiles has been demonstrated. Fresh lures containing 4S,5R-sitophinone and carob volatiles attracted significantly more insects in pitfall bioassays for all three species than when either component was used alone. However, after 6 weeks a significant increase in response compared to the control was found only for S. oryzae and S. zeamais. The effect of insect age on response to the pheromone and carob volatiles was also studied for all three species. The response to carob volatiles decreased with increasing insect age for all three species. A significant response to 4S,5R-sitophinone was found only for 1-2-day-old adults of S. oryzae and S. zeamais but for all ages tested of S. granarius. The effect of the combination of pheromone and carob volatiles also decreased with increasing insect age.  相似文献

Unknown green pigment, responsible for greening in crushed garlic cloves, was purified and characterized by using a series of column chromatography, liquid chromatography–electrospray ionization (LC–ESI), fast atom bombardment (FAB), matrix-assisted laser desorption/ionization time-of-flight (MALDI-TOF) mass spectrometry (MS), and nuclear magnetic resonance (NMR) spectroscopy. The purified green pigment was highly polar and slightly viscous, with a garlic flavor, and easily turned to a yellow or brown color with exposure to room temperature. The absorption spectrum in methanol showed a crude methanolic green pigment-like profile with two absorbance maxima at 440 and 590 nm.  相似文献

Analysis by gas chromatography-mass spectrometry led to the identification of 20 volatiles from the steam distilled oil of the leaves from Chamaecyparis obtusa Siebold et Zuccarni. Seven constituents of the oil were tested for contact and fumigant activity against adults of Callosobruchus chinensis (L.) and Sitophilus oryzae (L.). Responses varied with compound and dose rather than increasing exposure time beyond 1 or 2 days. In the impregnated-paper test with the constituents [bornyl acetate, (+)-limonene, myrcene, α-phellandrene, α-pinene, sabinene and terpinolene], at 0.1 mg/cm2, bornyl acetate, α-phellandrene and terpinolene caused 97%, 97% and 87% mortality, respectively, against C. chinensis adults 1 day after treatment. At 0.05 mg/cm2, moderate activity was achieved with bornyl acetate (56% mortality), α-phellandrene (75%) and terpinolene (55%). Against S. oryzae adults, at 0.26 mg/cm2, terpinolene caused 93% mortality 2 days after treatment, whereas 80% mortality 4 days after treatment was observed with bornyl acetate and α-phellandrene. The toxicity of these compounds was significantly decreased at 0.18 mg/cm2. In a fumigation test with S. oryzae adults, bornyl acetate and terpinolene were much more effective in sealed containers than in open ones, indicating that the insecticidal activity of these compounds was largely attributable to fumigant action. These naturally occurring materials derived from leaves of Chamaecyparis could be useful for managing populations of C. chinensis and S. oryzae.  相似文献

This study determined the starvation tolerance of Tribolium castaneum (Herbst), Rhyzopertha dominica (F.) and Sitophilus oryzae (L.) in terms of both adult survival and reproduction, the impact of starvation on reproduction not having been studied before. Experiments were conducted at 30 °C and 55% or 70% r.h. using a laboratory strain and a field strain of each species. The number of progeny was a better indicator of the impact of starvation on a species than adult survival. Tribolium castaneum was the most tolerant species, requiring up to 35 d starvation before no progeny were produced. Rhyzopertha dominica and S. oryzae required up to 8 d starvation before no progeny were produced. The results suggest that hygiene will have a greater impact on populations of S. oryzae and R. dominica than T. castaneum.  相似文献
