We report on fluorescence enhancement in near field optical spectroscopy by apertureless microscopy. Our apertureless microscope is designed around a confocal fluorescence microscope associated with an AFM head. First, we show that the confocal microscope alone allows single molecule imaging and single molecule fluorescence analysis. When associated with the AFM head, we demonstrate, for the first time to our knowledge, that single molecule fluorescence is enhanced under the silicon tip. We tentatively attribute this effect to field enhancement under the tip.  相似文献

A quadrature optical detection technique, based on polarized balanced-homodyne interferometry, has been developed for specific application to apertureless near-field scanning optical microscopy (ANSOM). With such technique, multiplicative background interference, inficiating quantitative optical imaging in standard homodyne-based ANSOM, can be suppressed. Periodic modulation of interferometric optical phase, typically employed in heterodyne-based ANSOMs even to such purpose, is not needed in the present configuration. Homodyne detection also facilitates detection of harmonic components of the ANSOM optical signal at the probe/sample distance modulation frequency, necessary for near-field discrimination and suppression of artifacts. Furthermore, since amplitude signal is not affected by phase fluctuations generated in the optical path of the interferometer, an optical fiber could be included in one interferometer arm, to couple the ANSOM head to the detection system, obtaining improved versatility of the instrument. A demonstration of the interferometer performance is given by a test confocal optical scan of a mirror surface. This technique, as applied to near-field microscopy, is anticipated to provide absolute values of optical contrast not depending on background interference and topography artifacts.  相似文献

We report on the fabrication, characterization and application of a probe consisting of a single gold nanoparticle for apertureless scanning near-field optical microscopy. Particles with diameters of 100 nm have been successfully and reproducibly mounted at the end of sharp glass fibre tips. We present the first optical images taken with such a probe. We have also recorded plasmon resonances of gold particles and discuss schemes for exploiting the wavelength dependence of their scattering cross-section for a novel form of apertureless scanning near-field optical microscopy.  相似文献

We present an analytical model able to explain the optical signal recorded during our experimental approach curves in the infrared at a wavelength lambda=10.6 microm, with a home-made apertureless near-field scanning optical microscope ANSOM. This model uses classical electrodynamics to calculate the scattering cross section of the oscillating tip, considered as a dipole, and its dielectric image in the sample as a function of the tip-sample separation from the near-field to the far-field regime. The dipoles are placed in a non-uniform electric field because of the standing wave arising from the interference between the incident and the specular laser beams. We also added a background field coming from a scatterer on the surface in order to account for zeroing of the optical signal for particular tip-sample separation and interference patterns.  相似文献

This report presents the Apertureless Scanning Optical Near-Field Microscope as a powerful tool for the characterization of modern optoelectronic and photonic components with sub-wavelength resolution. We present an overview of the results we obtained in our laboratory over the past few years. By significant examples, it is shown that this specific probe microscopy allows for in situ local quantitative study of semiconductor lasers in operation, integrated optical waveguides produced by ion exchange (single channel or Y junction), and photonic structures.  相似文献

We present advances in experimental techniques of apertureless scanning near-field optical microscopy (aSNOM). The rational alignment procedure we outline is based upon a phase singularity that occurs while scanning polarizers around the nominal cross-polarized configuration of s-polarized excitation and p-polarized detection. We discuss the theoretical origin of this topological feature of the setup, which is robust against small deviations, such as minor tip misalignment or shape variations. Setting the polarizers to this singular configuration point eliminates all background signal, allowing for reproducible plasmonic eigenmode mapping with optimal signal-to-noise ratio.  相似文献

We present the first studies of dyed human hair fibres performed with an apertureless scanning near‐field optical microscope. Samples consisted of 5‐µm‐thick cross‐sections, the hair fibres being bleached and then dyed before being cut. Hair dyed with two molecular probes diffusing deep inside the fibre or mainly spreading at its periphery were investigated at a wavelength of 655 nm. An optical resolution of about 50 nm was achieved, well below the diffraction limit; the images exhibited different optical contrasts in the cuticle region, depending on the nature of the dye. Our results suggest that the dye that remains confined at the hair periphery is mainly located at its surface and in the endocuticle.  相似文献

The model of apertureless near-field optical microscope is developed taking into account the multipole moment of probe. In the case of samples with small dielectric absorption, the multipole moments are shown to be responsible for the appearance of additional resonances in the spectrum of scattered signal. The influence of multipole moments is especially pronounced in the near-field microscopy with modulation of tip-sample distance. A good agreement of the theoretical results with experimental data in the case of resonant interaction of the probe and sample is demonstrated.  相似文献

The cells and tissues of many marine invertebrates and their associated flora contain fluorescent pigments and proteins, many of which have been utilized commercially and provide marker molecules in other systems for fluorescence imaging technology. However, in the study of marine invertebrates and their symbioses these naturally occurring molecules have been seen to limit or confound fluorescence microscopy analyses. Here we demonstrate the endogenous fluorescence associated with two marine invertebrates (coral and foraminifera) and describe how these qualities can be utilized in fluorescence microanalyses. Understanding and imaging the diversity of fluorescent molecules provide insight into how fluorescence microscopy techniques can now be applied to these complex systems.  相似文献

Quasi-two-colour femtosecond pump and probe spectroscopy and near-field scanning optical microscopy are combined to study the carrier dynamics in single semiconductor nanostructures. In temporally, spectrally and spatially resolved measurements with a time resolution of 200 fs and a spatial resolution of 200 nm, the non-linear change in reflectivity of a single quantum wire is mapped in real space and time. The experiments show that carrier relaxation into a single quantum wire occurs on a 100 fs time scale at room temperature. Evidence is given for a transient unipolar electron transport along the wire axis on a picosecond time and 100 nm length scale.  相似文献

A new design of a tunneling near-field optical microscope (TNOM) combined with an atomic force microscope (AFM) is presented. This design can be used to generate three different images of the sample's surface: a non-contact (tapping mode) AFM image, a conventional TNOM and an image of a modulation signal of the conventional TNOM, which we call AC-TNOM. The images are obtained simultaneously, using a single light source. It is shown that the AC-TNOM has better resolution ( approximately 200A) and contrast compared to conventional TNOM ( approximately 400A).  相似文献

Spin-sensitive optical near-field microscopy and spectroscopy are proposed based on the study on the conserved quantities in optical near-field interactions of atoms with dielectric surfaces. A two-step photoionization spectra of Cs atoms resolving hyperfine structures are demonstrated near a planar dielectric surface by using evanescent waves. These techniques of state/spin-selective excitation and highly sensitive detection, combined with the techniques of optical pumping, will open up possibilities of space- and polarization-sensitive detection of optical near‐fields using atomic probes. This novel method provides us with a useful technique for the observation of polarization nature of the optical near-field and controlling the spin states of mesoscopic electronic systems.  相似文献

Near-field optical structure of the centromere region of undyed polytene chromosomes has been observed using an apertureless near-field optical microscope that detects the intensity of light scattered from an atomic force microscope tip under laser illumination. The centromere is of primary importance to the functioning of the chromosome in the cell during cell division. It is also particularly interesting for structural/optical studies since its DNA repeat sequences are highly conserved among organisms and it is possible that they play a part in the centromere self assembly (Clark and Wall 1996).  相似文献

We study the influence of metal roughness on the near‐field distribution generated by an aperture or an apertureless (scattering) probe. Different experimental parameters are investigated: roughness magnitude, aperture form, distribution of the roughness. Our results show that aluminium roughness has a dramatic impact on the emission characteristics of a near‐field probe and in particular on its polarization sensitivity. Apertureless or scattering probes appear to be less sensitive to roughness and to provide a well confined field even with a somewhat rough probe.  相似文献

We solve numerically the three-dimensional vector form of Maxwell's equation for the situation of near-field excitation and collection of luminescence from a single quantum dot, using a scanning near-field optical fibre probe with subwavelength resolution. We highlight the importance of polarization-dependent effects in both the near-field excitation and collection processes. Applying a finite-difference time domain method, we calculate the complete vector fields emerging from a realistic probe structure which is in close proximity to a semiconductor surface. We model the photoluminescence from the quantum dot in terms of electric dipoles of different polarization directions, and determine the near-field luminescence images of the dot captured by the same probe. We show that a collimating effect in the high index semiconductor significantly improves the spatial resolution in the excitation-collection mode. We find that the spatial resolution, image shape and collection efficiency of near-field luminescence imaging strongly depend on the polarization direction as represented by the orientation of the radiating electric dipoles inside the quantum dot.  相似文献
