Holstein cows (n = 72, 28 primiparous) were fed for ad libitum intake a forage mixture (corn and haycrop silages, 63:37 DM) and a 71:29 ratio of two grain mixtures (13.8 and 36.2% CP, DM basis) at 1 kg/2.5 kg of the previous day's milk during 1 to 28 DIM (covariate period). Thereafter, rations were balanced and adjusted for energy and CP weekly based on monthly DHI data, forage analyses, and NRC requirements. Cows were blocked by parity (1 vs. greater than 1) and assigned randomly to receive one grain (blend of 13.8 and 36.2% CP grains, herd mean) or two grains according to individual needs. Daily yields of milk, 4% FCM, SCM, and resulting intakes of grain were higher for cows fed two grains, but the increase in daily DMI and the decreases in daily forage intake and SCM:grain ratio were not significant. Fewer pluriparous cows (3 vs. 8 out of 19) on the two-grain treatment conceived after first service, but overall services per conception (1.97) were equal. Percentage of cows with SCC below linear score 4 (91%) was equal between treatments. Means adjusted by covariate 4% FCM resulted in more modest treatment effects. Advantage of two-grain versus one-grain treatment for milk per day was 7.7% for parity 1 and only 1.6% for parities greater than 1; corresponding responses (nonsignificant) for 4% FCM were 2.8 and 4.3, respectively. Forage DMI depressions on two-grain treatment were not significant. When covariate-adjusted means were used, the advantage in income over feed cost for individualized feeding of CP was $106 and $36/yr per cow in parities 1 and greater than 1 when cows produced 8209 kg (313 DIM) and 8527 kg (320 DIM) of milk, respectively. 相似文献

ABSTRACTThe effect of ozone on deoxynivalenol (DON) detoxification was investigated. Ozone treatment could significantly reduce the levels of DON in wheat; 53% of DON in wheat was decomposed with 90 mg l–1 of ozone at a flow rate of 15 l min–1 for 4 h. The safety of DON-contaminated wheats (DCWs) untreated/treated by ozone was also evaluated. Institute of Cancer Research (ICR) mice were divided into a standard diet group and five experimental diet groups for a 51-day orally administration experiment. In the experiment, no remarkable changes in the general appearance of the mice were observed, and all the mice survived until the scheduled necropsy. The results of sub-chronic toxicity indicated that mice fed on DCWs alone had significantly decreased in body weight gain, thymus and spleen weights, ratios of liver, thymus and spleen to body weight, blood indices (red blood cell, haemoglobin, white blood cell), and pro-inflammatory cytokines (interleukin-6 (IL-6), tumour necrosis factor alpha (TNF-α)), while showing a significant increase in alanine aminotransferase, aspartate aminotransferase, blood creatinine and blood urea nitrogen levels. Histopathological examination indicate that DON elicited some degree of toxicity on the liver, kidney and thymus tissue. Mice fed on DCWs treated by ozone mitigated the adverse effects compared with mice fed on DCWs. All the results suggested that the deleterious effects of DON could be highly reduced by ozone, and ozone itself shows minor toxic effects on animals in this process. 相似文献

The influence of stage of lactation and corresponding diets on rates of protein degradation (kd) is largely unstudied. Study objectives were to measure and compare in situ ruminal kd of crude protein (CP) and estimate rumen CP escape (rumen-undegradable protein; RUP) of selected feeds by cows at 3 stages of lactation fed corresponding diets, and to determine the incubation times needed in an enzymatic in vitro procedure, using 0.2 units of Streptomyces griseus protease per percent of true CP, that predicted in situ RUP. Residue CP was measured after in situ fermentation for 4, 8, 12, 24, 36, 48, and 72 h of 5 protein sources and 3 total mixed rations, which were fed to the in situ cows. Two nonlactating (dry) cows and 2 cows each at 190 (mid) and 90 (peak) days of lactation were used. Each pair of cows was offered free-choice diets that differed in composition to meet their corresponding nutrient requirements. Diets had decreasing proportions of forages and contained (dry matter basis) 11.9, 15.1 and 16.4% CP and 54.3, 40.3 and 35.3% neutral detergent fiber, for dry, mid, and peak TMR (TMR1, TMR2, and TMR3), respectively. Intakes were 10.3, 21.4, and 23.8 kg of dry matter/d, respectively. Kinetic CP fractions (extractable, potentially degradable, undegradable, or slowly degradable) were unaffected by treatment. Lag time and kd varied among feeds. The kd was faster for all feeds (0.136/h) when incubated in dry-TMR1 cows compared with mid-TMR2 (0.097/h) or peak-TMR3 (0.098/h) cows, and no differences in lag time were detected. Calculated RUP, using estimated passage rates for each cow based on intake, differed between dry-TMR1 (0.382) and mid-TMR2 (0.559) or peak-TMR3 (0.626) cows, with a tendency for mid-TMR2 to be different from peak-TMR3. Using the average kd and lag time obtained from dry-TMR1 to calculate RUP for mid-TMR2 and peak-TMR3 cows using their passage rates reduced RUP values by 6.3 and 9.5 percentage units, respectively. Except for that of herring meal, in vitro residue CP at 6, 12, and 48 h of enzymatic hydrolysis was correlated (r = 0.90) with in situ RUP of peak-TMR3, mid-TMR2, and dry-TMR1, respectively. Although confounded within treatments, stage of lactation, diet, and intake appeared to affect CP degradation parameters and RUP. Using kd from nonlactating cows, or the RUP calculated from them, may bias diet evaluation or ration formulation for lactating cows. In addition, enzymatic in vitro predictions of RUP should be measured using incubation times that are appropriate for lactating cows. 相似文献

A milk production and digestibility trial with 32 cows (2 X 2 factorial with 8 cows per treatment) was conducted to test animal response to varying rumen degradation of dietary starch and protein. Four rations (barley-cottonseed, barley-brewers dried grains, milo-cottonseed, and milobrewers dried grains) were compared in early lactation cows fed 65% concentrate and 35% forage for 60 d. Digestibilities of starch were higher for barley than milo diets; whereas, those for organic matter were higher for cottonseed meal than brewers dried grains. Milk production was highest on barley-cottonseed with mean adjusted values of (kg/d): 37.4, 34.9, 34.2, and 34.6 for respective treatments. Dry matter intakes were not different among rations. Milk fat was higher on milo than barley with respective diets averaging (%): 3.1, 2.9, 3.4, and 3.6, and resulting in lowest FCM for the barley-brewers dried grains diet. Milk protein was not altered by treatment. These data suggest that responses to varying protein degradability can be altered by rate of starch breakdown in the rumen. 相似文献

The digestive and metabolic utilization of protein (50% lactoalbumin + 50% casein) and fat (43.0% butter, 29.5% olive oil, 14.7% soy oil, 9.8% MCT and 3.0% lecithin) provided by two commercial diets used in clinical enteral nutrition (normoproteic, 16.1% protein and 20.8% fat, and hyperproteic, 23.1% protein and 14.9% fat), was studied in adult rats (mean body weight 180 g). The diet containing the greater amount of protein improved the digestive utilization of nitrogen, and although nitrogen retention was optimal, it failed to rise further when the dietary protein supply was increased. A study was conducted to determine the rumen degradation and intestinal digestibility of crude protein (CP) and AA, and AA composition of the rumen-undegradable protein (RUP) from 3 sources of blood meal (BM1, BM2, and BM3), canola meal (CM), low-fat distillers dried grains with solubles (LFDG), soybean meal (SBM), and expeller soybean meal (ESBM). Two Holstein cows fitted with ruminal and proximal duodenal cannulas were used for in situ incubation of 16 h and for the mobile bag technique. To correct for bacterial contamination of the RUP, 2 methods were used: purines and DNA as bacterial markers. Ruminal degradations of CP were 85.3, 29.8, 40.7, 75.7, 76.9, 68.8, and 37.0 ± 3.93% for BM1, BM2, BM3, CM, LFDG, SBM, and ESBM, respectively. Ruminal degradation of both total essential AA and nonessential AA followed a similar pattern to that of CP across feedstuffs. Based on the ratio of AA concentration in the RUP to AA concentration in the original feedstuff, ruminal incubation decreased (ratio <1) the concentrations of His, Lys, and Trp, and increased (ratio >1) the concentrations of Ile and Met across feedstuffs. Compared with purines, the use of DNA as bacterial marker resulted in a higher estimate of bacterial CP contamination for CM and lower estimates for LFDG and ESBM. Intestinal digestibility of RUP could not be estimated for BM1, BM3, and SBM due to insufficient recovery of residue. For the remaining feedstuffs, intestinal digestibility of RUP was highest for ESBM, followed by BM2 and LFDG, and lowest for CM: 98.8, 87.9, 89.7, and 72.4 ± 1.40%, respectively. Intestinal absorbable dietary protein was higher for BM2 compared with CM and LFDG, at 61.7, 17.9, and 20.7 ± 2.73% CP, respectively. As prices fluctuate, intestinal absorbable protein or AA may be used as a tool to aid in the selection among feedstuffs with different protein quality. 相似文献

Corn is an important food and feedstuff in China and worldwide. The problems caused by aflatoxin B1-contaminated corn (ACC) are of great concern. Our previous studies have demonstrated that ozone can effectively degrade AFB1 in corn, prompting us to investigate the in vivo toxicity of treated ACC. In this study, 35 Kunming mice were used to assess the in vivo toxicity of ozone treated ACC. Results indicated that compared to mice fed with basal feedstuff (provided by the Shanghai SLAC Laboratory), those fed with ACC have significantly decreased mean weight as well as total protein (TP), albumin (ALB), and globulin (GLB) contents (p < 0.05). On the other hand, the liver and kidney/body weight ratio as well as the serum alanine transaminase (ALT), aspartate aminotransferase (AST), and alkaline phosphatase (ALP) levels significantly increased (p < 0.05). Obvious histopathological changes were found in the liver and kidney. When mice were fed with the ozone-treated ACC, no significant differences were observed in the mean weight, the liver and kidney/body weight ratio and in the major serum indexes ALT, TP, ALB, and GLB (p > 0.05). However, AST and ALP significantly increased (p < 0.05), and slight histopathological changes were found in liver tissues. This study indicated that ACC may lead to significant changes in various physiological characteristics and biochemical indexes in liver and kidney tissues, but ozone treatment of ACC could significantly reduce these changes. 相似文献

We compared ruminal dry matter (DM) and crude protein (CP) degradation kinetics of fresh forages of alfalfa (vegetative, early bud, early flowering, and late flowering stages) and bromegrass with endophyte-free and endophyte-infected tall fescue (tillering, stem elongation, heading, and flowering stages) by using nonlinear models. Duplicate Dacron bags were incubated for 0, 3, 6, 9, 12, 16, 24, 36, 48, and 60 h in 2 Simmental x Angus steers fitted with ruminal cannulas. The effects of animal, forage species, and maturity within forage species were evaluated. A first-order kinetics model was determined to be suitable for estimation of degradation profiles relative to models with variable rate of degradations. Alfalfa had higher soluble DM (36.6%), lower insoluble potentially digestible DM (43.0%), higher rate (13.8%/h), and higher extent of DM degradation (66.3%) than grasses (27.2, 53.5, 6.7%/h, and 54.6% for soluble DM, insoluble potentially digestible DM, rate, and extent of DM degradation, respectively). The extent of CP degradation was similar among forages (74.7%), but alfalfa had a higher CP degradation rate than grasses (16.1 vs. 12.5%/h). Extents of degradation of DM and CP decreased with maturity in alfalfa and in grasses. Ruminally undegradable CP (RUP) was higher in alfalfa (40.0 g/kg of DM) than in grasses (34.9 g/kg of DM), and decreased with maturity in grasses (40.4 to 28.3 g/kg of DM) but not in alfalfa. The amount of RUP that was potentially degradable in the rumen was not different among forage species (22.1 g/kg). As forage CP concentration decreased, the RUP (as a percentage of CP) increased but, as a percentage of forage DM, decreased. Species of forage had important effects on ruminal DM and CP degradation when incubated in fresh form. 相似文献

Thirty-eight midlactating Holstein cows averaging 597 kg of body weight (SD = 59) were used to determine the effects of dietary flaxseed on protein requirement and N excretion in urine and feces. Milk yield and composition, intake, and digestibility were also determined. Cows were allotted from wk 20 to 30 of lactation to 1 of 4 TMR containing 1) no flaxseed (control) and 16% protein (MPC), 2) whole flaxseed and 16% protein (MPF), 3) no flaxseed (control) and 18% protein (HPC), and 4) whole flaxseed and 18% protein (HPF). Cows fed high protein diets had greater feed intake than those fed medium protein diets (20.2 vs. 18.4 kg/d), and cows fed no flaxseed had greater dry matter intake than those fed flaxseed (20.1 vs. 18.5 kg/d). Milk yield was lower for cows fed MPF (20.3 kg/d) than for those fed HPC (24.4 kg/d), HPF (24.9 kg/d), or MPC (24.0 kg/d). Milk protein and lactose concentrations were similar for cows fed MPC and HPC, but flaxseed decreased milk protein concentration in cows fed MPF or HPF compared with cows fed the control diets. Milk fat concentration was similar in cows fed diets with or without flaxseed, but it was decreased by higher protein concentration. Digestibility was generally reduced when diets contained flaxseed and lower protein concentration. Dietary protein had no effect while dietary flaxseed increased fecal N excretion. Retention of N was lower in cows fed flaxseed compared with cows fed the control diets. Feeding flaxseed decreased milk concentrations of short- and medium-chain fatty acids and increased those of long-chain fatty acids. In vivo and in vitro gastric emptying of protein fractions of three milk replacers containing either milk protein (control), a mixture (50:50 on a CP basis) of milk protein and native whey protein concentrate, or a mixture (50:50 on a CP basis) of milk protein and heated whey protein concentrate was studied. In vivo gastric emptying was measured in three preruminant calves fitted with reentrant duodenal cannulas and used in a 3 x 3 Latin square design. In vitro gastric emptying was determined after enzymatic digestion in an artificial stomach. In vivo and in vitro flow rates of protein N (12% TCA-insoluble N) and total N were higher for milk replacers containing whey proteins than for control. Gastric emptying of NPN (12% TCA-soluble N) was slightly higher for diets containing whey proteins than for that containing milk proteins. Gastric emptying of all protein fractions was similar for the two milk replacers containing whey proteins. In vivo and in vitro results were significantly correlated, suggesting that the in vitro method reproduced conditions for proteolysis and could be used to predict gastric digestion of protein fractions. 相似文献

Three ruminally and duodenally cannulated cows were assigned to an incomplete 4 × 4 Latin square with four 14-d periods and were fed diets supplemented with urea, solvent soybean meal, xylose-treated soybean meal (XSBM), or corn gluten meal to study the effects of crude protein source on omasal canal flows of soluble AA. Soluble AA in omasal digesta were fractionated by ultrafiltration into soluble proteins greater than 10 kDa (10K), oligopeptides between 3 and 10 kDa (3-10K), peptides smaller than 3 kDa (small peptides), and free AA (FAA). Omasal flow of total soluble AA ranged from 254 to 377 g/d and accounted for 9.2 to 15.9% of total AA flow. Averaged across diets, flows of AA in 10K, 3-10K, small peptides, and FAA were 29, 217, 50, and 5 g/d, respectively, and accounted for 10.3, 71.0, 17.5, and 1.6% of the total soluble AA flow. Cows with diets supplemented with solvent soybean meal had higher flows of Met, Val, and total AA associated with small peptides than those whose diets were supplemented with XSBM, whereas supplementation with corn gluten meal resulted in higher total small peptide-AA flows than did XSBM. Averaged across diets, 27, 75, and 93% of soluble AA in 10K, 3-10K, and peptides plus FAA flowing out of the rumen were of dietary origin. On average, 10% of the total AA flow from the rumen was soluble AA from dietary origin, indicating a substantial escape of dietary soluble N from ruminal degradation. Omasal concentrations and flows of soluble small peptides isolated by ultrafiltration were substantially smaller than most published ruminal small peptide concentrations and outflows measured in acid-deproteinized supernatants of digesta. 相似文献

The colorimetric method for the determination of carbohydrate by the phenol-sulphuric acid reagent was modified for the estimation of macroquantities of total carbohydrate in formulated fishery products. Aqueous suspensions of the samples are reacted with the phenol-sulphuric acid reagent and the carbohydrate content is estimated by comparison to glucose standards. This technique is rapid and the carbohydrate contents obtained are in good agreement with values calculated by difference from the proximate composition. It is suggested that this technique could be used in determining the proximate composition of various food samples. The protein content could then be estimated by difference to within ±6 % (relative) of values obtained by direct protein determinations. 相似文献

Greasy hair is a common problem in Europe. The first step in developing anti-grease hair products must be to establish a sensitive protocol for measuring any changes in perceived hair greasiness.
Sensitive clinical trials and in vivo evaluation methods of determining hair greasiness have been developed that show significant differences in the perceived hair greasiness following the use of different shampoos.
Products tested in two clinical trials for efficacy as anti-grease shampoos were an anti-grease shampoo containing 2% 3,4-thiolanediol as the active ingredient, and a baby shampoo based on mild surfactants. The same placebo shampoo based on ether sulphates used in typical European shampoo formulations was used in both tests. A shampoo containing 2% zinc pyrithione was included in the trials as a control, since there were indications that this would increase the amount of hair greasiness.
Neither of the test products were shown to be effective anti-grease shampoos. The shampoo containing 2% zinc pyrithione was shown to produce significantly more greasiness than both the placebo and the test shampoos.
Since significant differences could be shown between the different shampoos, the protocol and in vivo evaluation techniques used in these clinical trials are considered to be validated.

Three ruminally and duodenally cannulated, lactating Holstein cows were used in a 3 x 3 Latin square experiment to study the effects of differing levels of nonstructural carbohydrate and degradable intake protein on ruminal digestibility and microbial protein production. Three diets were formulated to contain 1) 38 and 13.2%, 2) 31 and 11.8%, and 3) 24 and 9% nonstructural carbohydrate and degradable intake protein as percentages of the DM, respectively. Dry matter intakes were similar for all diets (21.9, 21.1, and 18.3 kg/d for diets 1, 2, and 3, respectively). Likewise, microbial efficiency, as estimated from purine analysis, was unaffected by diet and averaged 24 g of microbial N/kg of OM digested for all treatments. Ruminal digestion of OM averaged 66.6, 65.1, and 55.7% for diets 1, 2, and 3, respectively, resulting in lower microbial N flow per day for diet 3 (317, 333, and 202 g, respectively). Digestion of nonstructural carbohydrate and CP followed similar trends as did OM digestion, whereas NDF digestion remained similar across all diets. These results indicate that nonstructural carbohydrate greater than 24% and ruminally degradable protein greater than 9% of DM will enhance microbial protein flow from the rumen. 相似文献

The proximate mineral composition and indispensable amino acid pattern were determined for samples of Snail (Vivapara quadrata), periwinkle (Littorina littorea), crayfish (Palamonetes varians) and some locally smoked fresh water fish (Pisces spp.).The crude protein contents of smoked fish, crayfish, snail and periwinkle were 75·31, 74·84, 65·29 and 60·93% dry matter, respectively. These values were 56·9, 54·9, 35·2 and 26·1%, respectively higher than that of whole hen's egg. All the samples, except snail, also had higher ash values. Whole hen's egg, however, contained more crude fat (40·23%) and gross energy (34·74 Kj/g) than the test samples.The minerals calcium and phosphorus were higher in the test samples than in whole hen's egg. Samples analysed contained higher amounts of tryptophan than whole hen's egg and, with the exception of snail, the samples also contained more lysine. The test samples have high chemical scores ( ratios). Comparison has also been made of the indispensable amino acid patterns of these samples with those of cow's milk, human milk and the FAO (1957) provisional pattern. 相似文献

The study of colonic flora composition and metabolism presents considerable methodological problems. Attempts to circumvent these problems have led to the development of numerous in vitro and in vivo models to simulate the human colon and its microbial population. In terms of in vivo models, conventional laboratory animals have many limitations. Data of greater relevance to man can be obtained by using germ-free rodents associated with human colonic bacteria. The applications of such animals to studies of toxicity of chemicals and gastrointestinal infections are discussed. The advantages and disadvantages of the various in vitro systems for studying gut microflora and its metabolic activity (from simple static cultures to the more sophisticated continuous and semicontinuous flow models) are reviewed. The apparatus involved is described together with practical information on media, running conditions, and sampling. The bacteriological and metabolic criteria for establishing the similarity of the models to the in situ colonic flora are also discussed. The final sections of the review are devoted to the major applications (current and future) of the models, including fermentation studies on dietary fiber, metabolism of nutrients and foreign compounds (including carcinogens) in food, and the investigation of colonization resistance. 相似文献

The accuracy with which the Streptomyces griseus, ficin, and neutral protease with amylase in vitro methods could be used to predict in situ protein degradation of concentrate feeds was evaluated. Four types of soy products and two types of distillers grains were incubated using each of the four techniques for .5, 1, 2, 4, 8, 12, 24, and 48 h. The percentages of initial CP that remained undegraded over time were determined. Comparison of the degradation curves and contrast analysis of the data indicated that the in vitro methods generated degradability estimates in conflict with those obtained by the in situ method. However, the neutral protease with amylase method ranked the test feeds according to the extent of CP degradation at 24 h most similar to that in situ. Regression equations developed with the neutral protease with amylase degradability estimates explained 78, 76, and 74% of the variation in the in situ estimates obtained after 12, 18, and 24 h of incubation, respectively. At least 69% of the variation in the 18-h in situ estimates could be explained by the neutral protease with amylase estimates obtained after 1, 2, 4, 8, and 12 h of incubation. Little relationship was found between the ficin and S. griseus versus in situ results. Although none of the in vitro methods resulted in degradation curves consistently related to those generated by the in situ technique, relationships were found between protein degradability estimates obtained by the neutral protease with amylase method at specific time points and those obtained by the in situ technique. 相似文献
