Noradrenergic projections from the locus coeruleus (LC) project to the olfactory bulb (OB), a cortical structure implicated in odor learning and perceptual differentiation among similar odorants. The authors tested the role of OB noradrenaline (NA) in short-term olfactory memory using an animal model of LC degeneration coupled with intrabulbar infusions of NA. Specifically, the authors lesioned cortical noradrenergic fibers in mice with the noradrenergic neurotoxin N-Ethyl-N-(2-chloroethyl)-2-bromobenzylamine hydrochloride (DSP4) and measured the effects on an olfactory habituation/spontaneous discrimination task. DSP4-treated mice failed to habituate to repeated odor presentations, indicating that they could not remember odors over the 5-min intertrial interval. The authors then infused NA bilaterally into the OBs of both DSP4-treated and nonlesioned control animals at two concentrations (10-3M and 10-5M, 2 μl/side). In DSP4-treated animals, NA administration at either concentration restored normal habituation and spontaneous discrimination performance, indicating that noradrenergic neuromodulation mediates these aspects of perceptual learning and that its efficacy does not require activity-dependent local regulation of NA release. Functional OB learning mechanisms may be necessary for normal odor recognition and differentiation among physically similar odorants. The present study investigated age-related differences in discrimination and reversal learning for olfactory and visual stimuli in 6-month and 24-month-old rats. Rats were trained to discriminate between two pseudo-randomly selected odors or objects. Once each animal reached a criterion on discrimination trials, the reward contingencies were reversed. Young and aged rats acquired the olfactory and visual discrimination tasks at similar rates. However, on reversal trials, aged rats required significantly more trials to reach the learning criterion on both the olfactory and visual reversal tasks than young rats. The deficit in reversal learning was comparable for odors and objects. Furthermore, the results showed that rats acquired the olfactory task more readily than the visual task. The present study represents the first examination of age-related differences in reversal learning using the same paradigm for odors and objects to facilitate cross-modal comparisons. The authors tested how prior odor enrichment affects the spontaneous discrimination of both preexposed and novel odors. Experimental rats were exposed to single odors or to pairs of similar or dissimilar odors for 1-hr periods twice daily over 20 days. Spontaneous discriminations between pairs of similar odors were tested before and after the odor exposure period using an olfactory habituation task. The authors found that (a) experimental rats did not spontaneously discriminate similar odor pairs before the exposure period, whereas they spontaneously discriminated them after the enrichment period, and (b) the improvement of performance was not selective for the odors used during enrichment. These results show that odor experience changes perception in the manner predicted based on other groups' electrophysiological experiments. Adult C57BL/6J mice were trained in a fully automated operant task to discriminate between 2 simultaneously presented odors for water reward. Each mouse was trained on 16 different discrimination problems to a criterion of 90% correct in a block of 20 trials for each problem. Each of the mice tested reached criterion on all discriminations; the number of errors made before reaching criterion was greatest for the first few problems and decreased substantially thereafter. Acquisition performance on early trials of novel discriminations suggested that mice form learning sets for olfactory cues. The mice were trained on 4 additional problems and tested for memory with probe trials at retention intervals of 1, 2, 4, and 8 weeks. Good retention of olfactory cues was seen even at the longest memory delay. Automated testing of olfactory discriminations should be useful for assessing learning and memory abilities in genetically modified mice. Carbon chain length in several classes of straight-chain aliphatic odorants has been proposed as a model axis of similarity for olfactory research, on the basis of successes of studies in insect and vertebrate species. To assess the influence of task on measured perceptual similarities among odorants and to demonstrate that the systematic similarities observed within homologous odorant series are not task specific, the authors compare 3 different behavioral paradigms for rats (olfactory habituation, generalization, and discrimination). Although overall patterns of odorant similarity are consistent across all 3 of these paradigms, both quantitative measurements of perceptual similarity and comparability with 2-deoxyglucose imaging data from the olfactory bulb are dependent on the specific behavioral tasks used. The present study assessed whether benzodiazepines impair the acquisition, performance, and reversal of olfactory and auditory go/no-go discriminations in rats. Experiment 1 showed that midazolam (0.5-2 mg/kg sc) did not affect the performance of a well-learned two-odor olfactory discrimination and moderately facilitated performance of a go/no-go auditory discrimination. Experiment 2 found that midazolam (1 mg/kg) increased the number of errors made in the acquisition of a novel go/no-go olfactory discrimination task and in the reversal of a previously well-learned olfactory discrimination. However, midazolam did not affect the acquisition and reversal of an equivalent auditory discrimination task. Experiment 3 showed that diazepam (1 mg/kg) also impaired the acquisition and reversal of a novel olfactory discrimination task. Taken together, these results indicate that benzodiazepines cause a selective impairment of olfactory discrimination learning. This may reflect an effect of benzodiazepines in the glomerular circuitry of the olfactory bulb and at downstream olfactory processing sites such as the piriform cortex and orbitofrontal cortex. The associative structure mediating goal-directed action was investigated using congruent and incongruent conditional discriminations. The stimulus was the same as the outcome in each component of the congruent discriminations, whereas the stimulus of one component of the incongruent discriminations was the same as the outcome of the other component. Humans, but not rats, learned the congruent discrimination more rapidly than the incongruent discrimination, a difference that the authors attribute to the fact that outcome-response associations caused response conflict in the incongruent discrimination. Moreover, responding was resistant to outcome devaluation following incongruent, but not congruent, training, suggesting that both humans and rats adopted a stimulus-response strategy to resolve the incongruent discrimination. Dopaminergic modulation affects odor detection thresholds and olfactory discrimination capabilities in rats. The authors show that dopamine D? receptor modulation affects odor discrimination capabilities in a manner similar to the modulation of stimulus intensity. Performance in a simultaneous odor discrimination task was systematically altered by manipulations of both odorant concentration and D? receptor activation (agonist quinpirole, 0.025-0.5 mg/kg; antagonist spiperone, 0.5 mg/kg). Rats' discrimination performance systematically improved at higher odor concentrations. Blockade of D? receptors improved performance equivalent to increasing odor concentration by 2 log units, whereas activation of D? receptors reduced odor discrimination performance in a dose-dependent manner. Bulbar dopamine release may serve a gain control function in the olfactory system, optimizing its sensitivity to changes in the chemosensory environment. Experiments 1 and 2 address the controversy regarding the reliability of methamphetamine effects on interval timing. A temporal discrimination procedure was used, in which the rats were reinforced for pressing the left or the right levers after short and long signals, respectively. Methamphetamine (0.5 mg/kg sc) severely disrupted operant performance at 20-100 min after injection, which disabled the measurement of drug effects on temporal perception (Experiment 1). The same dose of methamphetamine shifted the psychometric function to the left at 100-180 min after injection, indicating an increase in subjective durations (Experiment 2). Although these results confirm the role of dopamine in interval timing, that a change in the speed of a neural clock mediates the methamphetamine-induced change in temporal perception is still a working hypothesis. Dopaminergic modulation of cortical activity has been implicated in the formation of reward associations. There is abundant evidence for dopaminergic effects on olfactory processing. Using an olfactory discrimination task, the authors show that D? and D? dopamine receptors can regulate rats' olfactory discrimination capacities and that the effects of receptor activation functionally oppose one another. Injection of either the D? agonist SKF 38393 (10 mg/kg) or the D? antagonist spiperone (0.62 mg/kg) facilitated the discrimination of similar odorants but had no effect on the discrimination of dissimilar odorants, whereas both the D, antagonist SCH 23390 (0.025 mg/kg) and the D? agonist quinpirole (0.2 mg/kg) significantly impaired rats' ability to discriminate similar and dissimilar odorants. Odor naming and recognition memory are poorer in children than in adults. This study explored whether such differences might result from poorer discriminative ability. Experiment 1 used an oddity test of discrimination with familiar odors on 6-year-olds, 11-year-olds, and adults. Six-year-olds were significantly poorer at discrimination relative to 11-year-olds and adults, who did not differ. Experiment 2 used the same procedure but with hard-to-name visual stimuli and compared only 6-year-olds with adults (as with the remaining experiments in this study). There was no difference in performance between these groups. Experiment 3 used the same procedure as Experiment 1 but with less familiar odors. Six-year-olds were significantly poorer at discrimination than adults. In Experiment 4 the researchers controlled for verbal labeling by using an articulatory suppression task, with the same basic procedure as in Experiment 1. Six-year-old performance was the same as for Experiment 1 and significantly poorer than that of adults. Impoverished olfactory discrimination may underpin performance deficits previously observed in children. These all may result from their lesser experience with odors, relative to adults. The role of the basolateral amygdala (BLA) in the consolidation of an association between an olfactory stimulus and footshock was investigated with a reversible lesion technique of posttraining infra-BLA infusions of tetrodotoxin. Rats receiving tetrodotoxin infusions following paired odor-shock presentations spent more time near the odor, and reacted differently on contact with the odor when tested 24 hr after training, than did rats receiving paired presentations and saline infusions, but they did not differ from rats receiving unpaired presentations and saline infusions. The results indicate that the BLA plays a similar role in influencing consolidation of olfactory-based memory as it does for memory based on other modalities. Thus, these findings strengthen the view that the BLA plays a general role in modulation of memory storage for emotionally arousing events. Some controversy still exists as to how binary odorant mixtures are behaviorally perceived, despite many studies aimed at understanding this phenomenon. Binary mixture perception by rodents is a first step in elucidating how more complex odor blends may be perceived. Research thus far has examined how the degree of component similarity, olfactory receptor overlap, relative concentration of components, and even olfactory enrichment affect the behavioral perception of binary mixtures. These studies have aimed to categorize binary mixtures into 1 of 3 rigid categories, but often the results conflict as to which category a particular mixture belongs. In the present article, the authors used a habituation/discrimination paradigm to determine whether rats' perception of one component of a binary mixture of either perceptually similar or dissimilar components changed when the concentration of both components was varied together. The authors found that perception of a binary mixture changed with changing component concentration, such that one binary mixture could be categorized differently depending on component intensity. In each of 4 experiments animals were given a structural discrimination task that involved visual patterns composed of identical features, but the spatial relations among the features were different for reinforced and nonreinforced trials. In Experiment 1 the stimuli were pairs of colored circles, and pigeons were required to discriminate between patterns that were the mirror image of each other. A related task was given to rats in Experiment 2. Subjects solved these discriminations. For Experiment 3, some pigeons were given a discrimination similar to that used in Experiment 1, which they solved, whereas others received a comparable task but with 3 colored circles present on every trial, which they failed to solve. The findings from Experiment 3 were replicated in Experiment 4 using different patterns. The results are difficult to explain by certain connectionist theories of discrimination learning, unless they are modified to take account of the way in which compound stimuli are structured. 2 classes of experiment on the role of attention in discrimination learning are reviewed: (a) Investigations of the effect of attention on the amount learned about different cues have been interpreted as disproving noncontinuity theory (according to which animals attend to only 1 cue at a time). The fact that animals learn something about a 2nd cue, however, does not prove that attention has no effect on learning, and more recent evidence shows that it does. (b) If animals do not automatically attend to all cues, part of what they must learn in order to solve a discrimination problem is to attend to the relevant cue. Experiments on the acquired distinctiveness of cues, transfer along a continuum, and reversal learning provide evidence for the importance of such classificatory learning. (155 ref.) Cells in the orbitofrontal cortex (OF) respond to odors and their associated rewards. To determine how these responses are acquired and maintained, the authors recorded single OF units in rats performing an odor discrimination task. Approximately 64% of all cells differentiated between rewarded and nonrewarded odors. These odor valence responses changed during learning in 26% of all cells, and these changes were positively correlated with improving performance, supporting the idea that the information provided by these cells is used in learning the task. However, changes in odor valence responses were also observed after learning, and included not only increases in odor discrimination, but also decreases or mixed increases and decreases. Thus, only some of the changes in firing reflected acquisition of the task. The results suggest that learning triggers a continuing reorganization of OF neural ensembles representing odors and their rewards. The role of the hippocampus and perirhinal-entorhinal cortex was examined in an olfactory discrimination paradigm. Small neurotoxic lesions of the hippocampus (21% tissue damage) yielded relatively unimpaired olfactory retention across brief (30 s), intermediate (approximately 5 min), and 24-hr delays, whereas impairments were noted at 5-day retention intervals. Larger hippocampal lesions (63% tissue damage) spared memory at intermediate delays, with no impact at 8-day retention intervals. Aspiration lesions directed at the perirhinal-entorhinal cortex produced a variable performance pattern, with impairments noted at intermediate, 24-hr, and 5-day delays. Results suggest the hippocampus is not specifically involved in retaining olfactory information, with additional consideration given to the relationship between lesion size and memory impairment. Rats were given intermixed preexposure to the compound flavors AX and BX and to the compound CX in a separate block of trials (4 presentations of each compound). In Experiment 1, rats showed less generalization of conditioned aversion from AX to BX than from CX to BX, a perceptual learning effect. Experiment 2 showed that the formation of an excitatory association proceeded more readily between A and B than between C and B, suggesting that intermixed preexposure maintains the effective salience of A and B and does not establish inhibition between them, a process that would require prolonged preexposure. According to this analysis, salience modulation and associative inhibition may contribute to perceptual learning at different stages of preexposure. Two male quokkas (Setonix brachyurus: a herbivorous macropod marsupial) were trained to discriminate pairs of stimuli in the laboratory. Quokkas indicated their choice by pulling on 1 of 2 simultaneously presented cords. The quokkas' discrimination abilities were tested on 6 tactile and 6 visual discrimination tasks. Correct responses were rewarded with food. For both quokkas, all tactile tasks were learned to a criterion of 75% correct in up to 4 20-trial sessions. No visual task maintained criterion performance in 4 sessions. One tactile discrimination was reversed 10 times. Participants initially completed a discrimination task (D1) involving categorization of patterns with multiple common features, each feature being partly predictive of the correct response. In a subsequent target discrimination task (D2), these features were redistributed across new discriminative stimuli. The relative predictiveness of the features in D1 was either maintained in D2 (i.e., features were equally informative in D1 and D2) or switched (i.e., more informative features in D1 were made less informative in D2, and vice versa). Differential performance on D2 suggested that features most predictive of the correct D1 responses became more highly associable than features that were less predictive in D1. This finding suggests that the associability of individual stimulus elements changes as a consequence of their role in discrimination learning.
