The antioxidant activities of different polarity solvents extracts from barley were evaluated by various antioxidant assays, including reducing power, free radical scavenging and lipid oxidation inhibition. Those various antioxidant activities were compared to standard antioxidants, butylated hydroxyluene (BHT) and ascorbic acid (Vit. C). The properties of the extracting solvents significantly affected the total phenolics, proanthocyanidins and antioxidant activities of barley extract. The highest contents of total phenolics and proanthocyanidins were obtained from extraction with 70% acetone. For three different solvent extracts, the antioxidant activities were in this order: 70% acetone extract > 70% ethanol extract ? 70% methanol extract. Reducing powers of three extracts and their scavenging effects on 2, 2-diphenyl-1-picrylhydrazyl radicals were effective at amount of 200 μg. The 70% acetone extract of barley exhibited high antioxidant activity in linoleic acid system, which was not significantly (P < 0.05) different from BHT during the incubation time.  相似文献

The aim of this work is to examine the chemical composition and antioxidant activity of separated essential oils and different solvent extracts of Thymus praecox subsp. skorpilii var. skorpilii (TPS). The ethanol, acetone, methanol, hexane, aqueous extracts and separated essential oils of TPS were assessed for their antioxidant activities. Antioxidant activities were evaluated by reduction of Mo(VI) to Mo(V), reducing power, superoxide scavenging activity, free radical-scavenging activity, metal chelating activity, linoleic acid peroxidation, hydrogen peroxide scavenging activity and peroxide scavenging activity. Essential oils were characterized in total to be 41 components, whereas 9 components were isolated by column chromatography for antioxidant activity. TPS essential oil was found to contain thymol (40.31%) and o-cymene (13.66%) as the major components. The ethanol, methanol and water extracts exerted significant free radical-scavenging activity. The methanol and water extracts displayed highest superoxide scavenging activity. The water extract has the highest total phenolics (6.211 mg gallic acid (GAE)/g DW) and flavonoids (0.809 mg quercetin/g DW).  相似文献

Cultivated and wild ginseng leaves were examined for their various antioxidant activities. Both ginseng leaves were extracted with methanol and sequentially partitioned with solvents in an order of increasing polarity. Among various solvent extracts in cultivated and wild ginseng leaves, EtOAC extracts in both ginseng leaves showed the most powerful scavenging activities against DPPH radicals. Data on other antioxidant activities, measured by inhibition rates against lipid peroxidation and linoleate oxidation, revealed similar results, showing the highest activities in EtOAC extracts, followed by butanol, water, chloroform and hexane extracts, in both cultivated and wild ginseng leaves. EtOAC extracts of wild ginseng leaves contained more phenolics (9.71 g:4.87 g/100 g, dry basis) and flavonoids (3.03 g:2.34 g/100 g, dry basis) than cultivated ginseng leaves. When EtOAC extracts were acid-hydrolyzed, two aglycones of flavonoids, quercetin (0.43 and 0.66 g/100 g, dry basis) and kaempferol (1.23 and 1.50 g/100 g, dry basis) were detected in cultivated and wild ginseng leaves. These differences in concentrations of key antioxidants between two different ginseng leaves seemed to be responsible for their differences in antioxidant activities.  相似文献

The physical and antioxidant properties of alginate biodegradable film incorporating white, red and extruded white ginseng extracts were investigated. No differences in moisture contents of all alginate film samples were noticeable (p > 0.05) except those film samples incorporated without ginseng extract, which had the highest moisture content. The addition of ginseng extract to alginate film decreased tensile strength and elastic modulus (p < 0.05) but increased the percent elongation at break. The presence of extracts did not significantly affect the water vapor permeability of the film samples. Alginate film containing extruded white ginseng extract at a barrel temperature of 130 °C presented the highest (61.12%) free-radical scavenging activity against the 2,2-diphenyl-1-picryhydrazyl radical (DPPH) in methanol, followed by film containing white ginseng extruded at a barrel temperature of 115 °C. Following these samples in terms of free-radical scavenging activity were film samples containing red and white ginseng extracts, respectively. These results revealed that extruded white ginseng extract has good potential to be incorporated into alginate to make antioxidant biodegradable film or coating for various food applications.  相似文献

The ethanol extracts from 24 samples plant species commonly found in Thailand were investigated and compared on their antioxidant activity by ABTS assay. The ethanol extract from the leaves of guava (Psidium guajava) showed the highest antioxidant capacity with the TEAC value of 4.908 ± 0.050 mM/mg, followed by the fruit peels of rambutan (Nephelium lappaceum) and mangosteen (Garcinia mangostana) with the TEAC values of 3.074 ± 0.003 and 3.001 ± 0.016 mM/mg, respectively. The further investigation of guava leaf extracts from different solvents; n-hexane, ethyl acetate, n-butanol, and methanol, was examined using ABTS and FRAP assays. The total phenolic content was done by Folin–Ciocalteu reaction. The results indicated that the methanol fraction possessed the highest antioxidant activity, followed by the butanol and ethyl acetate fractions, respectively. The hexane fraction showed the lowest antioxidant activity. The results demonstrated that the mechanism of antioxidant action of guava leaf extracts was free radical scavenging and reducing of oxidized intermediates. The phenolic content in guava leaf fraction played a significant role on the antioxidant activity via reducing mechanisms.  相似文献

Lactic-fermented cabbage, similar to Kimchi in Korea, is a very popular fermented vegetable product in Taiwan and China. In this study, fermented cabbage prepared by a dry-salt method was first extracted with water and methanol. Antioxidant activity such as DPPH radical scavenging effects, reducing power and Fe2+-chelating ability of the solvent extracts of fermented cabbage was determined and the effect of fermentation on the change of antioxidant activity, total phenolic and total flavonoid content was also investigated. Results revealed that antioxidant activity observed on the Chinese cabbage mixture may vary with extraction solvents and fermentation. Generally, the methanol extract of the cabbage mixture showed a higher DPPH radical scavenging activity and reducing activity than the water extract. Although, fermentation did not alter the Fe2+-chelating ability and reducing activity of the methanol extract of the cabbage mixture, it reduced these same antioxidant activities in the water extract. Amongst the various extracts examined, the methanol extract of fermented cabbage showed the highest DPPH radical scavenging effect. On the other hand, the highest Fe2+-ion chelating and reducing activities were exerted by the methanol extracts of both the cabbage mixture and the fermented cabbage, which showed no significant difference (p < 0.05). Additionally, the type of solvent and fermentation were also found to affect the total phenolic and flavonoid content of the extracts. Fermentation increased the total phenolic content of the methanol extract, whilst reducing the total flavonoid content of the water extract. Furthermore, changes in the antioxidant activity observed on the extracts of the cabbage mixture and fermented cabbage did not coincide exactly with the total phenolic and total flavonoid content.  相似文献

Pineapple has several beneficial properties including antioxidant activity. The fruit of pineapple was extracted with ethyl acetate, methanol and water. The phenolic content of the extracts was determined by Folin-Ciocalteu method and antioxidant activity was assayed through some in vitro models such as antioxidant capacity by phosphomolybdenum, ??-carotene-linoleate, and radical scavenging activity using ??,??-diphenyl-??-picrylhydrazyl (DPPH) method. The phenolic contents of the extracts as caffeic acid equivalents were found to be highest in methanol (51.1%) followed by ethyl acetate (13.8%) and water extract (2.6%). Antioxidant capacity of the extracts as equivalent to ascorbic acid (??mol/g of the extract) was in the order of methanol extract > ethyl acetate extract > water extract. In comparison with butylated hydroxyanisole (BHA), at 100 ppm of concentration, the antioxidant and free radical scavenging activities of the extracts assayed through ??-carotene-linoleate and DPPH method were also found to be highest with methanol extract followed by ethyl acetate and water extracts. The results indicated that the extent of antioxidant activity of the extract is in accordance with the amount of phenolics present in that extract and the pineapple fruit being rich in phenolics may provide a good source of antioxidant.  相似文献

Dorystoechas hastata (D. hastata) is a monotypic plant endemic to Antalya province of Turkey. D. hastata leaves are used to make a tea locally called “çalba tea”. Diethyl ether (E), ethanol (A), and water (W) were used for the sequential preparation of extracts from dried D. hastata leaves. A hot water extract (S) was also prepared by directly boiling the powdered plant in water. The antioxidant activities of the extracts were tested by ferric thiocyanate (FTC) and 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging methods. E extract exhibited the greatest antioxidant activity with FTC method, whereas S extract exhibited the lowest IC50 value (6.17 ± 0.53 μg/ml) for DPPH radical scavenging activity. Total phenolic contents of the extracts were estimated by Folin–Ciocalteu method and S extract was found to contain the highest amount (554.17 ± 20.83 mg GAE/g extract) of phenolics. Extract A contained highest flavonoid content and there was a inverse linear correlation (R2 = 0.926) between IC50 values for DPPH radical scavenging activity and flavonoid contents of all extracts. Reducing power of extracts increased in a concentration-dependent manner. S extract was found to possess higher reducing power than equivalent amount of ascorbic acid at 20 and 25 μg/ml concentrations. Linear correlation between reducing power and concentration of E, A, and W extracts (R2 > 0.95) was observed. A, W, and S extracts contained relatively high levels of proline. The results presented suggest that D. hastata may provide a natural source of antioxidants and proline.  相似文献

Defatted wheat germ (DWG) is the main by-product of the wheat germ oil extraction process. Its nutritional value has well been accepted. In this study, the antioxidant properties of 30% ethanol, 50% ethanol, 70% ethanol, 100% ethanol, and aqueous extracts of DWG were measured using various in vitro assays. Among the DWGEs (DWG extracts) tested, the 70% ethanol extract showed the best DPPH radical scavenging power while the 100% ethanol extract showed the highest ABTS radical scavenging activity and reducing power. In addition, both the 70% ethanol extract and the 50% ethanol extract exhibited relatively higher antioxidant activity in linoleic acid system. The extracts in question exhibited total phenolic contents ranging from 13.98 to 16.75 mg GAE/g. DWG, as a source of natural antioxidants, can be used to formulate nutraceuticals with potential applications to reducing the level of oxidative stress. The antioxidant potency of the DWG extracts could be the basis for its health promoting potential.  相似文献

以黄色洋葱皮为原料,分别利用70%甲醇、70%乙醇和70%丙酮作为提取溶剂进行多酚类物质提取。测定提取物中的总酚含量、总黄酮含量并研究其抗氧化活性(DPPH自由基清除能力、ABTS自由基清除能力以及铁还原能力)。研究结果显示:利用甲醇作为提取剂所得的多酚类物质提取率最高(12.35%),并且提取物中总酚和总黄酮含量最高,分别为83.17、34.31 mg/g。同时,甲醇提取物对DPPH自由基和ABTS自由基的半抑制浓度IC50分别为0.059和0.031 mg/mL,低于抗坏血酸而高于乙醇提取物和丙酮提取物。  相似文献

The antioxidant properties and total phenolic contents of methanol, acetone and water extracts of mulberry (Morus indica L.) leaves were examined. Various experimental models including iron (III) reducing capacity, total antioxidant capacity, DPPH radical scavenging activity and in vitro inhibition of ferrous sulphate-induced oxidation of lipid system were used for characterization of antioxidant activity of extracts. The three extracts showed varying degrees of efficacy in each assay in a dose-dependent manner. Methanolic extract with the highest amount of total phenolics, was the most potent antioxidant in all the assays used. In addition, the effect of temperature (50 °C and 100 °C), pH (3, 5, 7, 9 and 11) and storage (5 °C) on the antioxidant activity of methanolic extract was investigated. The antioxidant activity of the extract remained unchanged at 50 °C and was maximum at neutral pH. The extract stored at 5 °C in the dark was stable for 30 days after which the antioxidant activity decreased (p ? 0.05) gradually. On the basis of the results obtained, mulberry leaves were found to serve as a potential source of natural antioxidants due to their marked antioxidant activity.  相似文献

The antioxidant and quinone reductase (QR) inducing activities of American, Asian, and Siberian ginseng have been reported using various plant materials, solvents, and assays. To directly establish their comparative bioactivity, the effects of extracts obtained from acidified methanol (MeOH), a gastrointestinal mimic (GI), and hexane (Hex) on free radical scavenging and QR induction were tested. Siberian ginseng-MeOH had the highest total phenolic content at 52.6 μmol gallic acid equivalents/g. GI liberated ?50% more phenolics than MeOH and Hex from American and Asian ginseng. Siberian ginseng-Hex was most effective at inducing QR activity in Hepa1c1c7 cells. GI and MeOH extracts of American and Asian ginseng exhibited comparable HOCl scavenging activity, but were ?4.6-fold more potent than Siberian ginseng. Siberian ginseng was the most effective scavenger of ONOO. Siberian-MeOH had the highest ferric reducing antioxidant power (FRAP), and American ginseng-GI had the highest oxygen radical absorbance capacity (ORAC). Thus, the components in ginseng have antioxidant and QR-inducing activity which are dependent upon the dose, method of extraction, radical species, and plant species.  相似文献

采用rDNA ITS序列分析对一株野生桑黄菌(SH)进行鉴定,研究了其子实体不同溶剂(70%乙醇、甲醇、乙酸乙酯、氯仿)提取物中的总黄酮、总多酚与抗氧化活性的相关性以及4种提取物的抑菌活性。结果表明:该野生桑黄菌为粗毛纤孔菌(Inonotus hispidus),其子实体不同溶剂提取物的总黄酮、总多酚、抗氧化活性和抑菌活性存在显著差异,其中甲醇提取物中的总黄酮和总多酚含量最高,分别为(14.78±0.16)mg芦丁/g和(67.42±1.41)mg没食子酸/g;70%乙醇提取物的和DPPH·清除能力ABTS+·清除能力最强,分别为(43.83±0.03)和(116.08±0.24)mg Trolox/g,甲醇提取物的铁离子还原能力最强,为(126.24±3.02)mg Trolox/g;总多酚和总黄酮含量与FRAP法之间呈极显著正相关(p<0.01),总多酚与ABTS+·清除能力和DPPH·清除能力之间呈显著相关(p<0.05),而总黄酮与ABTS+·清除能力和DPPH·清除能力不相关;70%乙醇提取物和甲醇提取物的抑菌效果最好,其中对草莓灰葡萄孢霉的抑菌效果最好(抑菌圈直径>30 mm),其次是对金黄色葡萄球菌(抑菌圈直径>15 mm),对大肠杆菌的抑菌效果最差(抑菌圈直径>10 mm)。研究发现粗毛纤孔菌甲醇提取物的总多酚、总黄酮含量最高,抗氧化活性和抑菌活性最强,是天然抗氧化剂的良好来源。  相似文献

Water, water/methanol (1/1), methanol and ethanol crude extracts from a brown alga Stypocaulon scoparium were examined for total phenolic contents (TPC) using Folin–Ciocalteu method. DPPH scavenging assay was performed to measure the radical scavenging activities (RSA) of the extracts. Results showed a significant association between the antioxidant potency and the TPC. The aqueous extract showed both, the highest antioxidant activity and highest phenolic contents. The identification and quantification of phenolic antioxidants were carried out with a rapid and simple method of reverse phase high performance liquid chromatography (RP-HPLC). This method was developed for the simultaneous analysis of 14 polyphenols, namely gallic acid, catechin, epicatechin, rutin, p-coumaric acid, myricetin, quercetin and protocatechuic, vanillic, caffeic, ferulic, chlorogenic, syringic and gentisic acids. The chromatographic separation of 14 polyphenols was achieved in less than 40 min by RP-HPLC (Varian, Pursuit XRs C18 column, 5 μm, 250 mm × 4.6 mm) using linear gradient elution of methanol and water (0.1% formic acid) with a flow rate of 1 ml/min. The antioxidant and the tyrosinase inhibitory activities of 4 different solvents (acetone, ethanol, methanol, and water) for preparation of extracts from guava (branch, fruit, leaf, and seed) were evaluated by measuring total phenolic contents (TPC), DPPH radical scavenging activity, ABTS radical scavenging activity, reducing power (RP), and tyrosinase inhibitory activity. The extracts of branch and leaf showed relatively higher antioxidant properties than those of fruit and seed. The highest TPC (141.28 mg/g gallic acid equivalents), DPPH radical scavenging activity (IC50=34.01 μg/mL), ABTS radical scavenging activity (IC50=3.23 μg/mL), and RP (IC50= 75.63 μg/mL) were found in acetone extract of leaf, while water extract of seed had the lowest antioxidant activity. The tyrosinase inhibitory activity of ethanol extract from guava leaf was 69.56%, which was the highest activity among the extracts. These results indicate that useful bioactive substances exist in the guava branch as well as leaf extracts.  相似文献

研究不同极性溶剂对红薯叶中酚类化合物的提取以及提取物抗氧化性的影响,并鉴定提取物中的主要抗氧化成分组成。分别采用极性不同的7 种溶剂(蒸馏水、甲醇、无水乙醇、丙酮、正丁醇、乙酸乙酯和氯仿)从红薯叶中提取多酚,并评价提取物中总酚、总黄酮和花青素的含量,以及对1,1-二苯基-2-三硝基苯肼(1,1-diphenyl-2-picrylhydrazyl,DPPH)自由基清除能力和还原能力,最后运用高效液相色谱-串联质谱(high performance liquidchromatography tandem mass spectroscopy,HPLC-MS/MS)技术分析抗氧化活性最好的提取物中多酚的主要组成成分。结果表明:提取溶剂的极性对红薯叶中多酚类化合物的提取效率和提取物抗氧化活性有很大的影响,水提物具有最高的粗提物得率((37.13±1.60)%),而甲醇提取物中总酚含量(13.80 mg GAE/g)和总黄酮含量((5.68±0.35)mg QE/g)最高,且具有最好的DPPH自由基清除能力(IC50为0.32 mg/mL)与还原能力(ρ0.5为0.95 mg/mL)。采用HPLC-MS/MS从红薯叶甲醇提取物中鉴定9 种、初步鉴定3 种酚类化合物,鉴定的化合物为咖啡酸、对羟基苯甲酸、1-咖啡酸奎宁酸、3-咖啡酸奎宁酸、异绿原酸A、异绿原酸B、异绿原酸C、3,4,5-三咖啡酰奎尼酸和金丝桃苷。  相似文献

Significant levels of antioxidant activities and phenolic components have been detected in wheat and wheat-based food products, indicating that wheat may serve as an excellent dietary source of natural antioxidants for disease prevention and health promotion. Several solvent systems have been used to prepare antioxidant extracts from wheat and wheat-based food products. This makes it difficult to compare and understand the antioxidant activities of wheat reported from different research groups. The purpose of this study was to evaluate the effects of four selected solvent systems at ambient temperature for 15 h and Soxhlet extraction with absolute ethanol on antioxidant activity measurements. The four solvent systems included 50% acetone (v/v), 70% methanol (v/v), 70% ethanol (v/v), and ethanol, and antioxidant activities were tested using ORAC, radical scavenging activities against stable DPPH and cation ABTS•+, and total phenolic content. The results showed that the extracting solvent significantly altered the antioxidant property estimations of wheat bran, and 50% acetone is a recommended solvent for extracting phenolic antioxidants from wheat bran for analytical purpose.  相似文献

以蜂胶为原料,分别以甲醇、乙醇、氯仿、蒸馏水为溶剂进行萃取,测定各萃取物总酚与黄酮类物质的含量,通过测定还原能力,清除DPPH 自由基能力的测定来比较4 种不同萃取物的抗氧化性。结果表明:甲醇萃取物具有较强的抗氧化性。甲醇萃取物浓度为0.25mg/ml 时还原力高达1.162,且浓度为0.05mg/ml 时对DPPH 自由基清除率为64.676%。乙醇萃取物通过石油醚、氯仿、乙酸乙酯、丙酮依次分级萃取后所得到4 种萃取物各项指标测定结果表明,氯仿萃取了蜂胶乙醇萃取物中主要成分,但乙酸乙酯和丙酮萃取物的还原能力及清除DPPH 自由基清除能力较佳,在浓度为0.05mg/ml 时还原力分别为2.087 和1.798,DPPH自由基清除率达87.724% 和88.013%。  相似文献

Experiments were conducted to determine if the contents of phenolics and browning reaction products and antioxidant activity of raisin extracts were closely dependent on the extraction solvent. Enhanced extraction yields were obtained from solvent containing higher water concentrations. However, total phenolic content (TPC) was highest for extracts obtained from solvent to water ratios of 60:40 (v/v), whereby the extract obtained from ethanol:water (60:40, v/v) had the highest TPC of 375 mg gallic acid equivalents (GAE)/100 g extract. HMF content was highest in extracts obtained from 60% solvent, regardless of solvent type. The extract obtained from 60% methanol had the highest HMF content at 199 μg/g extract. Although the 60% solvents provided extract with high antioxidant components, the antioxidant activity of raisin extracts obtained from 80:20 (v/v) solvent/water was significantly higher than other raisin extracts obtained from different solvent concentrations. Phenolic acids, HMF, and low-molecular-weight flavonoids were responsible for the antioxidant activity, but not the high-molecular-weight flavonoids.  相似文献
