Despite the great variety in chicken photoreceptors, existing morphogenetic studies only deal with two types: rods and cones. We have therefore examined by scanning electron microscopy the first appearance and maturation of different retinal photoreceptors in 36 chicken embryos (Gallus domesticus), aged 5-19 days prehatching. On day 5 of incubation, chicken retinae were only composed of proliferating ventricular cells devoid of photoreceptors. On day 8, outer mitotic cells were separated from inner differentiating photoreceptors, by the transient layer of Chievitz. Ball-like protrusions appeared at the ventricular surface, representing the first signs of photoreceptor inner segment formation. From day 10 onward, double cones, single cones, and rods could be clearly distinguished, and occasional cilia were detected at their tip. On day 12, inner segments had increased in length and diameter, and frequently carried a cilium representing the beginning of outer segment formation. On day 14, most photoreceptors displayed a distinct outer segment. On day 19, photoreceptors had essentially assumed adult morphology. Based on the shape of their outer segments, two subtypes of cones and three subtypes of double cones could be distinguished. Throughout development, we observed microvilli close to maturing photoreceptors, either originating from their lateral sides, from their tip, or from Müller cells. Microvillus density peaked between day 12 and 14, indicating an important role in photoreceptor morphogenesis. Unilateral occlusion of the eyes of posthatching chicken reduced the proportion of double cones to single cones in the retina, indicating dependence of retinal morphogenesis upon functional activity of visual cells.  相似文献

The photoreceptor population in Xenopus consists of a green-sensitive rod (lambda(max) = 523 nm), a blue-sensitive rod (lambda(max) = 445 nm) and three classes of cone. The largest cone is red-sensitive (lambda(max) = 611 nm). The intermediate cone is presumed to be blue-sensitive based on physiological criteria, whereas the miniature cone may be UV-sensitive. Horizontal cells (HC) are of two sorts: axon-bearing and axonless. The axon-bearing HC is of the luminosity type and probably contacts all types of photoreceptor. The axonless HC is of the chromaticity type and contacts only intermediate (blue) cones and at least one type of rod. During development dendrites of HCs and bipolar neurons penetrate photoreceptor bases. A progressive maturation of HC and bipolar synapses with rods and cones occurs between tadpoles stages 37/8 and 46. Neighboring rods and cones are joined by gap junctions. During this same period, the outer segments are laid down and photopigments synthesized. A linear relation was found between the quantum capturing ability of the rod and its absolute threshold. Mature rods of the Xenopus retina release glutamate in a calcium-dependent manner. Glutamate release was found to be a linear function of calcium influx through L-type calcium channels. Both types of HC possess ionotropic glutamate receptors of the AMPA subtype.  相似文献

This review summarizes our present state of knowledge about spectrally different photoreceptor cell types in the Xenopus retina. The classification of the photoreceptors was based on morphology, combined with immunolabelling with various anti-visual pigment antibodies and other molecular probes on semithin sections and retinal wholemounts. The majority of photoreceptors is represented by rods. Altogether 97-98% of the total rod population consists of the principal ("red") rods that are selectively labeled by N-terminal specific anti-bovine rhodopsin monoclonal antibodies (mAbs) and are maximally sensitive to green light. The other, rare, blue-sensitive rod type ("green rod") is thinner, not stained by these antibodies but binds C-terminal specific anti-rhodopsin mAbs. The major representatives of the cones are red-sensitive and consist of a morphologically heterogeneous group comprising both (principal and accessory) members of double cones, as well as large single cones. Outer segments in this group are selectively labeled by mAb COS-1, specific to the L/M group of cone visual pigments. Another, relatively rare cone type is similar in size, but slightly smaller than the large single cone and is not labeled by mAb COS-1. This cone type is assumed to have a blue-sensitive cone visual pigment. The third, least abundant, and immunocytochemically distinct cone type is a small single (miniature) cone, which binds mAb OS-2 relatively strongly, and anti-rhodopsin mAbs 4B4 and 1D4 weakly. By exclusion, this small single cone may be identical with the UV-sensitive cone. Further studies are needed, however, to identify the color sensitivity of the latter two cone types.  相似文献

Transmission electron microscopy (TEM) was performed on the near-surface material (depth <500 nm) of tapered roller bearing inner rings (cones) that were tested at two levels of boundary-lubricated conditions in mineral oil with no additives. Site-specific thinning of cross section cone surface sections for TEM analyses was conducted using the focused ion beam (FIB) milling technique. High-resolution structural and compositional characterization of near-surface material and surface layers was performed on an untested cone as well as cones tested at Λ~1.1 and 0.3. This approach revealed near-surface microstructural distortion and grain size gradients that were attributed to surface finishing operations during manufacture. The characteristics of oxide surface layers and micro-cracks on the tested bearing surfaces were evaluated and found to depend on lubrication conditions.  相似文献

This study assessed a new experimental model to analyze the effects of sodium hypochlorite solution 2.5% for 10 min and chlorhexidine 2% for 15 s on cones surface alterations. Twenty five gutta-percha and 25 thermoplastic synthetic polymer-based cones had their last 1 mm from the tapered end examined by scanning electron microscopy (SEM) before and after being exposed to the disinfectant solutions. To allow the surfaces to be compared, the samples were not metalized, a voltage of 1 kV was used and the images were captured within standardized angles. The samples were immersed on the test solutions. The comparison of the alterations of the cones was performed using the software program Adobe Photoshop CS. A measurement was taken of the standard deviations of the pixel intensity in this area, higher standard deviations represent more irregular areas. Differences in standard deviation before and after disinfection for each cone were compared between groups by means of analysis of variance supplemented by the Tukey test. Gutta-percha cones showed higher standard deviations than Resilon cones (P < 0.001). In the gutta-percha cones, sodium hypochlorite generated more irregular areas than chlorhexidine and the control group (P < 0.001). The present study has demonstrated the feasibility of using a modified SEM method. This opens opportunities to the use of new and precise tools for evaluating a sample in different moments.  相似文献

The fabrication of silicon cantilever‐based scanning near‐field optical microscope probes with fully aluminium‐coated quartz tips was optimized to increase production yield. Different cantilever designs for dynamic‐ and contact‐mode force feedback were implemented. Light transmission through the tips was investigated experimentally in terms of the metal coating and the tip cone‐angle. We found that transmittance varies with the skin depth of the metal coating and is inverse to the cone angle, meaning that slender tips showed higher transmission. Near‐field optical images of individual fluorescing molecules showed a resolution < 100 nm. Scanning electron microscopy images of tips before and after scanning near‐field optical microscope imaging, and transmission electron microscopy analysis of tips before and after illumination, together with measurements performed with a miniaturized thermocouple showed no evidence of mechanical defect or orifice formation by thermal effects.  相似文献

A consortium of microorganisms with the capacity to degrade crude oil has been characterized by means of confocal laser scanning microscopy (CLSM), transmission electron microscopy (TEM), and scanning electron microscopy (SEM). The analysis using CLSM shows that Microcoleus chthonoplastes is the dominant organism in the consortium. This cyanobacterium forms long filaments that group together in bundles inside a mucopolysaccharide sheath. Scanning electron microscopy and transmission electron microscopy have allowed us to demonstrate that this cyanobacterium forms a consortium primarily with three morphotypes of the heterotrophic microorganisms found in the Microcoleus chthonoplastes sheath. The optimal growth of Microcoleus consortium was obtained in presence of light and crude oil, and under anaerobic conditions. When grown in agar plate, only one type of colony (green and filamentous) was observed.  相似文献

The principles of image formation in natural color scanning electron microscopy (NC-SEM) are discussed in detail. This method is based on the frequency characteristic of the human visual system. It is shown that the Mach effect and masking effect are important in the characteristics. The former, which can enhance structural details, is visually similar to the edge effect in secondary electron (SE) images, and the latter is required for proper representation of very degraded color information obtained from a light microscope. When using these effects suitably, an NC-SEM image with the resolution equivalent to that of an SEM image can be acquired, though it is composed of an SEM image and a special video microscopy (VM) image with a resolution much lower than the SEM image of the identical view. The NC-SEM is more effective than the SEM in observation. interpretation, and analysis of various samples with important color information.  相似文献

Using transmission electron microscopy (TEM) and scanning force microscopy (SFM) together, it was possible to verify important structural features of a nanostructured bulk material such as the kp‐morphology in an ABC triblock copolymer. By applying suitable imaging techniques during the SFM measurements it was possible to determine the morphology without additional manipulation steps in between. In comparison, TEM investigations on this type of material usually require selective staining procedures prior to the measurement. Also electron beam damage is often encountered during TEM measurements especially if components such as poly(methacrylates) are present. In contrast, SFM measurements can be assumed not to significantly change the phase dimensions of the components.  相似文献

Arge pullata Zadd is an important phytophagous pest that damages red birch Betula albo‐sinensis in Hubei Province, South China. Massive ecological and economic losses have been caused by this species, which threatens the ecological security of the Shennongjia Nature Reserve. To investigate the mechanoreception, chemoreception, and oviposition processes of A. pullata, scanning electron microscopy and optical confocal microscopy were used to reveal the typology, morphology, and distribution of ovipositor and antennal sensilla. The results show that A. pullata has clavate antennae and eight types of sensilla in total, including sensilla chaetica, sensilla trichodea (types 1–3), sensilla basiconica, sensilla coeloconica (types 1 and 2), and Böhm's bristles. Sensilla trichodea type 1 distributed only on male antennae; the densities of sensilla trichodea type 2 and sensilla basiconica differed between the sexes. The binding pattern of ovipositor valvulae was discovered, and one type of sensilla chaetica, two types of sensory pits, and tooth‐like cones as well as two types of microtrichia were found in the ovipositor. Based on morphological evidence and research on Hymenoptera, putative functions are suggested to increase our understanding of the mechanisms by which this species finds hosts and mates, and how oviposition takes place. The present study explores a novel design of cone flowmeter for bi-directional flow metering application. Two identical cone shapes are machined with their base circle surfaces joined together with a small step in between them and differential pressure measurement is done across the apex of the cones. The bi-directional cone flowmeter is tested under fully developed flow conditions and its performance under double 90° bend (out-of-plane) is also evaluated. The bi-directional cone flowmeter is tested in a circular pipe (inside diameter of 101 mm) with water as the working medium for the flow Reynolds number ranging from 1.18×105 to 5.48×105. Influence of the half cone angle (α) and the location of static pressure taps on the coefficient of discharge (Cd) of a cone flowmeter are studied. Two cones with half cone angles α=30° and α=45° with a constant constriction ratio (β) of 0.75 are studied. Static pressure taps are located on both sides of the bi-directional cone. Two sets of locations of static pressure taps are studied. First set includes two static pressure taps on the pipe wall in the planes of apexes of the bi-directional cone—called apex taps. Second set includes pressure taps on the pipe wall in the planes at a distance D/4 away from the apexes of the bi-directional cone—called D/4 taps. Double 90° bend (out-of-plane) is placed at 1.5D, 5.5D, 9.5D and 13.5D upstream to the bi-directional cone flowmeter. It is observed that the apex static pressure taps located in the plane of apexes of the bi-directional cone result in statistically consistent coefficient of discharge for all Reynolds numbers covered in this study. The results suggest that the bi-directional cone flowmeter is insensitive to the swirl created by double 90° bend (out-of-plane) placed at the upstream of cone flowmeter, if placed at a distance of 9.5D or more.  相似文献

设计了一种新型结构高速刀柄,刀柄的结构特征为实心短锥双面定位,并在锥面上开环形沟槽形成分锥体。由分锥体组合而成的大柔度锥面允许刀柄与主轴锥孔以较大过盈量配合联接,同时刀柄分锥体高速旋转时能够产生较大的离心膨胀与主轴锥孔膨胀相匹配,提高了刀柄与主轴接口的极限转速和联接刚度。对刀柄结构进行了设计,利用有限元分析对刀柄的主要几何参数、过盈量以及极限转速进行了确定。研究结果证明,与常用的HSK刀柄相比,所设计的新型高速刀柄能够显著提高刀柄的极限转速。  相似文献

The aim of this study was to investigate the damage of cutting tools coated by physical vapor deposition (PVD) during the continuous turning of a titanium alloy. The investigation utilized scanning electron microscopy (SEM), electron probe micro-analysis (EPMA), and transmission electron microscopy (TEM). It was found that a TiN coating on the tool wore faster than an uncoated cemented carbide tool. The damage mode of the coating on the rake face was fracture without plastic deformation. Additionally, there was a pattern to the crystal orientation relationship at some of the interfaces between the adhered workpiece material and the TiN coating. The crystal orientation relationship presumably produced a strong bond between the adhered material and the coating. The coating damage was thus caused by the force exerted by the adhered materials on the grain boundary on the damaged coating surface. A comparison of the tool damages during the machining of Ti-6Al-4V alloy and alloy 718 suggested that the damage of the coating depended on the interfacial strength between the adhered material and the coating, as well as the strength of the adhered material at a high temperature. Hence, to prevent the damage of the tool during the machining of a titanium alloy, it is preferable to use a ductile material (e.g., cemented carbide) rather than a brittle material (e.g., ceramic).  相似文献

分析了直母线锥盘金属带式无级变速器带的轴向偏移。阐述了传统控制方法的弊端:传统曲母线锥盘与金属片产生角点接触,在传动比i=1附近传动比变化不连续;在传动比i=1时,预置带的轴向偏移量只能使带的最大偏移量减小到未调整时的50%~60%。提出复合母线锥盘,锥盘母线与金属片侧边共轭,既完全消除了金属带的轴向偏移,又与直母线锥盘具有相同的接触强度,提高了无级变速器的传动能力,扩大了无级变速器的应用范围。  相似文献

Transmission electron microscopy (TEM) was performed on cross-sectional samples of tapered roller bearing cone surfaces that were tested at two levels of local boundary lubrication severity, Λ ~ 1.1 and 0.3. Unlike our previously reported work in which a base mineral oil was used, the bearing tests were conducted in mineral oil with sulfur- and phosphorus-containing gear oil additives. Structural and compositional characterization of undetached antiwear surface layers on the base steel (cone raceway) revealed that the films contained crystalline and amorphous regions. A sharp interface (<~10 nm) that separated the surface layer and base steel was imaged. The surface layer for the cone tested at Λ ~ 1.1 consisted of Fe, O, and P, whereas that for the cone tested at Λ ~ 0.3 consisted of Fe, O, P, C, Ca, and S. Various TEM analytical techniques were used to study the segregation of these elements throughout the antiwear surface layer volume.  相似文献

Cellular composition of the adult zebrafish (Danio rerio) optic tectal cortex was examined in this study. Morphological techniques such as 1 μm thick serial plastic sections stained with osmium tetroxide and toluidine blue, modified rapid Golgi silver impregnation, GFAP immunohistochemistry, confocal microscopy, as well as scanning and transmission electron microscopy were used. Neuronal and glial components are described and the layers of the cortex are revisited. Specific neuronal arrangements as well as unique glial/ependymal cells are described. A three dimensional rendering of the astrocytic fiber arrangement in the marginal zone is presented and a composite drawing summarizes the cellular composition of the optic tectum.  相似文献

The objective of this paper is to present a method to describe the three-dimensional variations of the geometry of the three portions forming the distal part of the human femur: the medial and lateral femoral condyles and the intercondylar fossa. The contours of equally spaced sagittal slices were digitized on the distal femur to determine its surface topography. Data collection was performed using a digitizer system which utilizes low-frequency, magnetic field technology to determine the position and orientation of a magnetic field sensor in relation to a specified reference frame. The generalized reduced gradient optimization method was used to reconstruct the profile of each slice utilizing two primitives: straight-line segments and circular arcs. The profile of each slice within the medial femoral condyle was reconstructed using two circular arcs: posterior and distal. The profile of each slice within the lateral femoral condyle was reconstructed using three circular arcs: posterior, distal and anterior. Finally, the profile of each slice within the intercondylar fossa was reconstructed using two circular arcs: proximal-posterior and anterior, and a distal-posterior straight-line segment tangent to the proximal-posterior circular arc. Combining the data describing the profiles of the different slices forming the distal femur, the posterior portions of each of the medial and lateral femoral condyles were modelled using parts of spheres having an average radius of 20 mm. The anterior portion of the lateral condyle was approximated to a right cylinder having its circular base parallel to the sagittal plane with an average radius of 26 mm. The anterior portion of the intercondylar fossa was modelled using an oblique cylinder having its circular base parallel to the sagittal plane with an average radius of 22 mm. Furthermore, it is suggested that the distal portion of the lateral femoral condyle could be modelled using parts of two oblique cones while the distal portion of the medial femoral condyle could be modelled using a part of a single oblique cone, all cones having their circular bases parallel to the sagittal plane. It is also suggested that the posterior portion of the intercondylar fossa could be modelled using two oblique cones: a proximal cone having its base parallel to the sagittal plane and a distal cone having its base parallel to the frontal plane.  相似文献

Three dimensional data collection in electron microscopy is normally limited to a range of tilt angles that is less than +/- 90 degrees. Thus, even under the best conditions, experimental values of the structure factors will be missing within a solid cone in reciprocal space. Previous work has already shown that the missing cone of data can produce serious artifacts in three-dimensional density maps at modest resolution, for example approximately 15A. We now report, however, that a missing cone as large as +/- 30 degrees appears to have no serious effect on the three-dimensional density map of a protein at 3.6 A resolution, and we attribute this result to the fact that the electron density features are quite well separated from one another at that resolution. The map calculated with a +/- 30 degree missing cone is, furthermore, no more sensitive to noise (error) than is the full (isotropic) Fourier map. This result does not seem to be unreasonable in view of the fact that less than 14% of the data (i.e., signal) is lost due to the missing cone. Our numerical simulations therefore indicate that there should be no difficulty in interpreting high-resolution Fourier maps that can be produced with data that fall within realistic estimates of achievable resolution, tilt angles, and experimental error.  相似文献
