建立了环带斑褶菇菌丝体粗多糖的甲醇一步提取法.采用正交设计,优化了提取工艺,并采用传统的水提醇沉法验证了新工艺的应用效果.结果表明,甲醇一步提取法可以有效地应用于环带斑褶菇菌丝体粗多糖的制备.采用甲醇一步提取法提取环带斑褶菇菌丝体粗多糖的最佳工艺为:料液比1:40,提取温度55℃,提取时间6h.甲醇一步提取法提取的粗多糖为白色半透明结晶,蛋白含量0.02%,得率23.46%;而水提醇沉法提取的粗多糖为浅褐色粉末,蛋白含量0.36%,得率22.04%.甲醇一步提取法有待于在其他生物粗多糖提取中得到更大范围的应用. 相似文献

从板蓝根中提取多糖,并用苯酚-硫酸测定其含量。方法:运用正交设计选取微波法和超声法的最佳工艺条件,用均匀设计选取渗漉法的最佳工艺条件,比较三种方法在最佳工艺条件下的得率。结果:超声法、微波法、渗漉法提取板蓝根多糖的得率分别为5.72%、6.24%、4、42%。微波法和超声法适合于板蓝根多糖的提取。 相似文献

The gelling behaviour of linseed polysaccharide was measured in the presence of sucrose as a function of both polysaccharide and sucrose concentrations. Yield stress, consistency index, and flow behaviour index were determined using the Herschel-Bulkley model. Transition concentrations of 0.7% for the polysaccharide and 55% for sucrose were found. Yield stress and consistency index increased with both polysaccharide and sucrose concentrations while flow behaviour index decreased. A polynomial model of the third degree on centred data after natural log transformation was found to give a satisfactory fit for the variations in rheological constants. The models for both yield stress and consistency index were a complex power law, and for flow behaviour index a complex semi-log relationship, on sucrose and polysaccharide concentrations. 相似文献

A husk polysaccharide component from a malt causing premature yeast flocculation was isolated and characterized, and its effect on the fermentation pattern of Saccharomyces cerevisiae 2036 established. A particular malt husk extract, obtained using a mild aqueous extraction procedure, induced premature flocculation when added to fermentations in normal wort. In sodium dodecyl sulphate-polyacrylamide gel electrophoresis (SDS-PAGE) of the malt husk extract, 4 protein bands (42,600; 17,500; 15,100 and 13,100 daltons) and a high molecular weight polysaccharide were identified. These components were separated by gel chromatography and their effect on the fermentation pattern established. Only the polysaccharide fraction induced premature flocculation. Sugar analysis of this fraction indicated the presence of arabinose, galactose, glucose, mannose, xylose, rhamnose and an acidic sugar component. The polysaccharide fraction showed haemoagglutination activity, which was not inhibited by any of 11 simple sugars tested. 相似文献

采用超声波+复合酶法预处理对Phellinus linteus子实体水溶多糖进行提取,并对提取多糖成分差异进行初步分析。采用超声波技术预处理桑黄子实体,利用正交实验研究了料液比、时间、功率各因素对桑黄多糖得率的影响;在超声波优化结果基础上,进行纤维素酶+木瓜蛋白酶法处理,研究了纤维素酶用量、处理时间、木瓜蛋白酶用量、处理时间各因素对多糖得率的影响。研究表明:在料液比1:20、超声波(400W、20min)和纤维素酶(0.8%、pH5.5、45℃、60min)、木瓜蛋白酶(2%、pH5.5、50℃、120min)双重预处理条件下,再进行多糖提取,多糖得率最高。研究表明采用超声波+复合酶法预处理方法提取多糖,与传统的热水提取法相比,极大提高了提取效率,为Phellinus linteus多糖的产业化打下了基础。 相似文献
