Poly[(diethylaminoethyl methacrylate)‐graft‐(ethylene glycol)] hydrogels were prepared with a molar ratio of 10:1 of diethylaminoethyl methacrylate to poly(ethylene glycol) of number‐average molecular weights (Mn) 200, 400 and 1000 g mol?1 using tetra(ethylene glycol) dimethacrylate to give a crosslinking ratio between 0.5 and 4.0 %. Glucose oxidase and catalase were immobilized in the matrix during polymerization. The maximum enzyme loading used was 6.6 × 10?4 g of glucose oxidase per g of polymer. The equilibrium and dynamic swelling properties of these hydrogels were investigated. The pH‐dependent equilibrium swelling characteristics showed a sharp transition between the swollen and the collapsed state at pH 7.0. The dynamic response of the hydrogel discs to pH was analyzed in pulsatile pH conditions. The effects of particle size, crosslinking and molecular weight of poly(ethylene glycol) (PEG) on the dynamic swelling response were investigated. Novel amphiphilic star‐shaped terpolymers comprised of hydrophobic poly(?‐caprolactone), pH‐sensitive polyaminoester block and hydrophilic poly(ethylene glycol) (Mn = 1100, 2000 g mol?1) were synthesized using symmetric pentaerythritol as the core initiator for ring‐opening polymerization (ROP) reaction of ?‐caprolactone functionalized with amino ester dendrimer structure at all chain ends. Subsequently, a second ROP reaction was performed by means of four‐arm star‐shaped poly(?‐caprolactone) macromer with eight ‐OH end groups as the macro‐initiator followed by the attachment of a poly(ethylene glycol) block at the end of each chain via a macromolecular coupling reaction. The molecular structures were verified using Fourier transform infrared and 1H NMR spectroscopies and gel permeation chromatography. The terpolymers easily formed core–shell structural nanoparticles as micelles in aqueous solution which enhanced drug solubility. The hydrodynamic diameter of these agglomerates was found to be 91–104 nm, as measured using dynamic light scattering. The hydrophobic anticancer drug curcumin was loaded effectively into the polymeric micelles. The drug‐loaded nanoparticles were characterized for drug loading content, encapsulation efficiency, drug–polymer interaction and in vitro drug release profiles. Drug release studies showed an initial burst followed by a sustained release of the entrapped drug over a period of 7days at pH = 7.4 and 5.5. The release behaviours from the obtained drug‐loaded nanoparticles indicated that the rate of drug release could be effectively controlled by pH value. Altogether, these results demonstrate that the designed nanoparticles have great potential as hydrophobic drug delivery carriers for cancer therapy. © 2015 Society of Chemical Industry  相似文献

Novel pH‐responsive PEGylated hollow nanocapsules (HNCaps) were fabricated through a combination of distillation–precipitation copolymerization and surface thiol–ene ‘click’ grafting reaction. For this purpose, SiO2 nanoparticles were synthesized using the Stöber approach, and then modified using 3‐(trimethoxysilyl)propyl methacrylate (MPS). Afterward, a mixture of triethyleneglycol dimethacrylate (as crosslinker), acrylic acid (AA; as pH‐responsive monomer) and MPS‐modified SiO2 nanoparticles (as sacrificial template) was copolymerized using the distillation–precipitation approach to afford SiO2@PAA core–shell nanoparticles. The SiO2 core was etched from SiO2@PAA using HF solution, and the obtained PAA HNCaps were grafted with a thiol‐end‐capped poly(ethylene glycol) (PEG) through a thiol–ene ‘click’ reaction to produce PAA‐g‐PEG HNCaps. The fabricated HNCaps were loaded with doxorubicin hydrochloride (DOX) as a model anticancer drug, and their drug loading and encapsulation efficiencies as well as pH‐dependent drug release behavior were investigated. The anticancer activity of the drug‐loaded HNCaps was extensively evaluated using MTT assay against human breast cancer cells (MCF7). The cytotoxicity assay results as well as superior physicochemical and biological features of the fabricated HNCaps mean that the developed DOX‐loaded HNCaps have excellent potential for cancer chemotherapy. © 2020 Society of Chemical Industry  相似文献

Novel pH‐ and temperature‐responsive chitosan‐graft‐poly[2‐(N,N‐dimethylamino)ethyl methacrylate] (chitosan‐g‐PDMAEMA) copolymers were successfully synthesized by homogeneous atom transfer radical polymerization (ATRP) under mild conditions. Chitosan macroinitiator was prepared by phthaloylation of amino groups of chitosan and subsequent acylation of hydroxyl groups of chitosan with 2‐bromoisobutyryl bromide. The copolymers were obtained by ATRP of 2‐(N,N‐dimethylamino)ethyl methacrylate and they can self‐assemble into stable micelles in water. Hybrid micelles with a PDMAEMA corona incorporating gold nanoparticles (Au NPs) were prepared in situ via the reduction of HAuCl4 with NaBH4. The pH and temperature responses of the copolymer micelles and hybrid micelles were characterized using UV‐visible spectroscopy and dynamic laser light scattering. The morphology of the micelles was observed using transmission electron microscopy and atomic force microscopy. The PDMAEMA corona of the micelles acts as the ‘nanoreactor’ and the ‘anchor’ for the in situ formation and stabilization of Au NPs. Therefore, the spatial distribution of Au NPs within the micelles can be tuned by varying the temperature and pH value. Copyright © 2010 Society of Chemical Industry  相似文献

A convenient approach has been developed for the preparation of microsize hydrogels composed of crosslinked poly(acrylic acid) (PAA) and poly(N‐isopropylacrylamide) (PNIPAm). First, semi‐interpenetration polymer networks of hydropropylcellulose (HPC) and PNIPAm‐co‐PAA copolymer are formed through the copolymerization and crosslinking of monomer acrylic acid and N‐isopropylacrylamide in HPC aqueous solution. After the selective removal of HPC from networks due to ionization of PAA units and disruption of hydrogen bonding with increasing pH, PNIPAm‐co‐PAA microgels are obtained, whose volume is confirmed to be responsive to both temperature and pH. Doxorubicin hydrochloride (Dox) can be encapsulated in PNIPAm‐co‐PAA microgels with high drug loading driven by the electrostatic interaction, and a sustained‐release characteristic of Dox from the microgels is observed under physiological pH value and temperature. In vitro cell experiments, the drug‐loaded microgels can be taken up by LoVo cells and release their payload in cell cytoplasm without loss of drug efficacy. This indicates that PNIPAm‐co‐PAA microgels might be a potential drug delivery carriers especially for water‐soluble or polypeptide drugs. © 2011 Wiley Periodicals, Inc. Multifunctional hybrid nanoparticles, Fe3O4@poly[(2‐dimethylamino)ethyl methacrylate]‐block‐poly(2‐hydroxyethyl methacrylate)‐graft‐carbazole, with pH‐responsivity, superparamagnetism and fluorescence for targeted drug delivery and release have been synthesized. The nanoparticles have a core‐shell structure as determined from transmission electron microscopy, pH‐responsivity as determined from hydrodynamic radius analysis, superparamagnetism as determined from vibrating sample magnetometry and fluorescence as determined from fluorescence spectroscopy and fluorescence microscopy. The release behavior of model drug progesterone indicates that the release rate can be effectively controlled by altering the pH of the environment. The multifunctional nanoparticles could be applied extensively in targeted drug delivery and release, and with fluorescence they can serve as efficient tracers to record magnetic targeting routes. BACKGROUND: Much interest has recently been shown in fundamental studies and technological developments of polymeric nanoparticles. Novel dual temperature‐ and pH‐sensitive stabilized nanoparticles composed of maleilated dextran (Dex‐MA) and N‐isopropylacrylamide (NIPAAm) were prepared and investigated to be used for the controlled release of 10‐hydroxycamptothecin. RESULTS: The resultant nanoparticles showed spherical geometry, with diameters ranging from 60 to 300 nm, the variation achieved by adjusting the composition ratio or the amounts of surfactant. The size of the nanoparticles increased and the zeta potential reversed with an increase of pH. Also, the lower critical solution temperature (LCST) decreased at pH = 3.0, but increased at pH = 6.0, which suggests that the LCST is dependent on pH. Drug release results obtained showed encapsulation and loading efficiencies were 62.9% and 0.82%, respectively. Cytotoxicity assay of this copolymer indicated no significant toxicity. CONCLUSION: Novel dual temperature‐ and pH‐sensitive Dex‐MA‐graft‐PNIPAAm nanoparticles may provide some advantages for anti‐cancer drug delivery due to a tunable LCST from 35 to 45 °C and controllable dimensions. Surfactant‐free nanoparticles of poly(DL ‐lactide‐co‐glycolide) (PLGA) nanoparticles were prepared with or without poly(L ‐lactide)‐poly(ethylene oxide) (LE) diblock copolymer (abbreviated as PLGA/LE and PLGA nanoparticles) by dialysis method. LE diblock copolymer was used to make PLGA nanoparticles to alternate conventional surfactant. The size of PLGA and PLGA/LE nanoparticles was 295.3 ± 171.3 and 307.6 ± 27.2 nm, respectively, suggesting LE diblock copolymer might be coated onto the surface of nanoparticles. Observation of scanning electron microscope (SEM) showed that PLGA/LE nanoparticles have spherical shapes ranging ~ 200–500 nm. In 1H‐NMR study, characteristic peaks of the methyl protons of PLGA disappeared in D2O, whereas characteristic peaks of the methyl proton of both PEG and PLGA were shown in both CDCl3 and D2O, indicating that LE diblock copolymer coated on the surface of the PLGA nanoparticles. The higher the initial content of drug, the higher the drug contents and the lower the loading efficiency. PLGA/LE nanoparticles at higher drug contents resulted in slower adriamycin·HCl (ADR) release rate than that of lower drug contents. Also, slower release rate of ADR was achieved by entrapped into the PLGA/LE nanoparticles, whereas LE polymeric micelles showed rapid ADR release. © 2003 Wiley Periodicals, Inc. Amphiphilic copolymers with cationic hydrophilic moieties and different ratios of hydrophobic portion to hydrophilic portion were designed and synthesized via the combination of hydrosilylation reactions and quaternization reactions. The structures were characterized through Fourier transform infrared spectroscopy, 1H NMR , 13C NMR and gel permeation chromatography. The measurements of critical micelle concentrations, electrical conductivities and zeta potentials indicated that the copolymers could self‐assemble into nanoparticles with charges around the surface in aqueous solution. The sizes of the micelles were between 67 nm and 104 nm detected by dynamic light scattering. The self‐assembled micelles were used as drug carriers to encapsulate a model drug (tocopherol), and their drug‐loading content (DLC ) and efficiency (DLE ) were determined by UV ?visible spectra, resulting in considerable drug‐loading capacity to a tocopherol maximum up to 17.2% (DLC ) and 80.3% (DLE ) with a size of 90 nm. The blank micelles and drug‐loaded micelles displayed a spherical shape detected by transmission electron microscopy, which demonstrated not only the self‐assembly behaviors but also the drug‐loading performances of the cationic amphiphilic copolymers. All the results demonstrated that the cationic amphiphilic copolymers could be used as potential electric‐responsive drug carriers. © 2017 Society of Chemical Industry  相似文献

Microgel particles were prepared, made of hydroxypropylcellulose‐graft‐(acrylic acid) (HPC‐g‐AA) and acrylic acid(AA). The particles undergo reversible volume phase transitions in response to both pH and temperature changes while keeping the inherent properties of PAA and HPC‐g‐AA. Dynamic light scattering measurements reveal that the average hydrodynamic radius and hydrodynamic radius distributions of the microgel particles depend on temperature and pH. The microgels exhibit excellent pH sensitivity and a higher swelling ratio at higher pH in aqueous solution. In vitro release study shows that the amount of insulin released from the microgels is less at pH = 1.2 than at pH = 6.8. The results indicate that the resultant microgels seem to be of great potential for intelligent oral drug delivery. Copyright © 2012 Society of Chemical Industry  相似文献

A series of pH‐sensitive composite hydrogel beads, carboxymethyl cellulose‐g‐poly(acrylic acid)/attapulgite/sodium alginate (CMC‐g‐PAA/APT/SA), were prepared by combining CMC‐g‐PAA/APT composite and SA, using Ca2+ as the ionic crosslinking agent and diclofenac sodium (DS) as the model drug. The effects of APT content and external pH on the swelling properties and release behaviors of DS from the composite hydrogel beads were investigated. The results showed that the composite hydrogel beads exhibited good pH‐sensitivity. Introducing 20% APT into CMC‐g‐PAA hydrogel could change the surface structure of the composite hydrogel beads, decrease the swelling ability, and relieve the burst release effect of DS. The drug cumulative release ratio of DS from the hydrogel beads in simulated gastric fluid was only 3.71% within 3 hour, but in simulated intestinal fluid about 50% for 3 hour, 85% for 12 hour, up to 90% after 24 hour. The obtained results indicated that the CMC‐g‐PAA/APT/SA hydrogel beads could be applied to the drug delivery system as drug carriers in the intestinal tract. © 2011 Wiley Periodicals, Inc. Amphiphilic thermally sensitive poly(N‐isopropylacrylamide)‐block‐poly(tetramethylene carbonate) block copolymers were synthesized by ring‐opening polymerization of tetramethylene carbonate with hydroxyl‐terminated poly(N‐isopropylacrylamide) (PNiPAAm) as macro‐initiator in the presence of stannous octoate as catalyst. The synthesis involved PNiPAAm bearing a single terminal hydroxyl group prepared by telomerization using 2‐hydroxyethanethiol as a chain‐transfer agent. The copolymers were characterized using 1H NMR and Fourier transform infrared spectroscopy and gel permeation chromatography. Their solutions show reversible changes in optical properties: transparent below the lower critical solution temperature (LCST) and opaque above the LCST. The LCST depends on the polymer composition and the media. Owing to their amphiphilic characteristics, the block copolymers form micelles in the aqueous phase with critical micelle concentrations (CMCs) in the range 1.11–22.9 mg L?1. Increasing the hydrophobic segment length or decreasing the hydrophilic segment length in the amphiphilic diblock copolymers produces lower CMCs. A core‐shell structure of the micelles is evident from 1H NMR analyses of the micelles in D2O. Transmission electron microscopic analyses of micelle morphology show a spherical structure of both blank and drug‐loaded micelles. The blank and drug‐loaded micelles have an average size of less than 130 nm. Observations show high drug‐entrapment efficiency and drug‐loading content for the drug‐loaded micelles. Poly(N‐isopropylacrylamide‐co‐hydroxyethyl methacrylate) [P(NIPAM‐co‐HEMA)] copolymer was synthesized by controlled radical polymerization from respective N‐isopropylacrylamide (NIPAM) and hydroxyethyl methacrylate (HEMA) monomers with a predetermined ratio. To prepare the thermosensitive and biodegradable nanoparticles, new thermosensitive graft copolymer, poly(L ‐lactide)‐graft‐poly(N‐isoporylacrylamide‐co‐hydroxyethyl methacrylate) [PLLA‐g‐P(NIPAM‐co‐HEMA)], with the lower critical solution temperature (LCST) near the normal body temperature, was synthesized by ring opening polymerization of L ‐lactide in the presence of P(NIPAM‐co‐HEMA). The amphiphilic property of the graft copolymers was formed by the grafting of the PLLA hydrophobic chains onto the PNIPAM based hydrophilic backbone. Therefore, the graft copolymers can self‐assemble into uniformly spherical micelles ò about 150–240 nm in diameter as observed by the field emission scanning electron microscope and dynamic light scattering. Dexamethasone can be loaded into these nanostructures during dialysis with a relative high loading capacity and its in vitro release depends on temperature. Above the LCST, most of the drugs were released from the drug‐loaded micelles, whereas a large amount of drugs still remains in the micelles after 48 h below the LCST. © 2011 Wiley Periodicals, Inc. Network formation was monitored by shear storage modulus (G′) during free radical crosslinking polymerization to investigate the effects of pH and ethylenediaminetetraacetic acid (EDTA; a complex agent). Three types of acrylic monomers, acrylic acid (AAc), 2‐acrylamidoglycolic acid (AmGc), and 2‐acrylamido‐2‐methyl propanesulfonic acid (AmPS), were polymerized in the presence of a crosslinking agent. The ratio of crosslinking agent (methylene bis‐acrylamide; MBAAm) to monomer was varied as: 0.583 × 10?3, 1.169 × 10?3, 1.753 × 10?3, and 2.338 × 10?3. G′ of the hydrogel in crosslinking polymerizations of AAc and AmPS was effectively increased by addition of EDTA, which was not the case for the crosslinking polymerization of AmGc. The order of magnitude of G′ differed based on the acidity of monomer. The maximum values of G′ in crosslinking polymerizations of AAc, AmGc, and AmPS were ~20,000 Pa, 6000 Pa, and 400 Pa, respectively. G′ varied linearly with the molecular weight between crosslinks (Mwc). pH and EDTA‐complex affected the rate of intramolecular propagation during crosslinking polymerization. Our results indicated that G′ was primarily affected by the following factors in the order: (1) acidity of monomer, (2) Mwc, and (3) physical interactions induced by pH and EDTA. © 2014 Wiley Periodicals, Inc. Grafting of agar and κ‐carrageenan with polyvinylpyrrolidone (PVP, average molecular weight 10,000 D) in an aqueous medium at a pH of about 7 produced agar‐graft‐PVP and κ‐carrageenan‐graft‐PVP blends capable of forming hydrogels. The reaction was carried out with microwave irradiation in the presence of a water‐soluble initiator, potassium persulfate. Optimum microwave irradiation conditions for obtaining hydrogels of the grafted products were achieved. The structural characteristics and thermal stability of the grafted blends were studied by Fourier transform infrared, 13C‐NMR, and thermogravimetric analyses. Appearance of new IR bands at 1661, 1465, and 1426 cm?1 in the grafted products indicated the insertion of PVP into the polysaccharide structure. Powder X‐ray diffraction studies revealed the enhanced crystallinity in the products compared to in the control polysaccharides as well as PVP. Agar and κ‐carrageenan were grafted to a considerable degree, with 62.5 E % and 125 G % for agar‐graft‐PVP and 65.5 E % and 131 G % for κ‐carrageenan‐graft‐PVP. Optical micrographs of the grafted blends indicated considerable changes in the morphology of the agar and the κ‐carrageenan, substantiating the X‐ray diffraction data. A plausible mechanism for the crosslinking of PVP to agar and κ‐carrageenan is proposed. These hydrogels exhibited enhanced water‐holding capacity despite weaker gel strength than that in the respective control polysaccharides. © 2006 Wiley Periodicals, Inc. Chitosan‐graft‐β‐cyclodextrin (CS‐g‐β‐CD) copolymer was synthesized by conjugating β‐cyclodextrins to chitosan molecules through click chemistry. The copolymer structure was characterized by Fourier transform infrared spectroscopy (FTIR) and nuclear magnetic resonance (NMR). CS‐g‐β‐CD/CMC nanoparticles were prepared by a polyelectrolyte complexation process in aqueous solution between CS‐g‐β‐CD copolymer and carboxymethyl chitosan (CMC), which was used to load anticancer drug (Doxorubicin hydrochloride, DOX·HCl) with hydrophobic group. The particle size, surface charge, zeta potential, and morphology of the nanoparticles were characterized with dynamic light scattering. The drug loading efficiency and in vitro release of DOX·HCl of the nanoparticles were measured by ultraviolet spectrophotometer. The results demonstrated that the size, surface charge and drug loading efficiency of the nanoparticles could be modulated by the fabrication conditions. The drug loading efficiency of CS‐g‐β‐CD/CMC nanoparticles was improved from 52.7% to 88.1% because of the presence of β‐CD moieties with hydrophobic cavities, which can form inclusion complexes with the drug molecules. The in vitro release results showed that the CS‐g‐β‐CD/CMC nanoparticles released DOX·HCl in a controlled manner, importantly overcoming the initial burst effect. Methoxy poly (ethylene glycol)‐graft‐α, β‐poly (aspartic acid) derivatives (mPEG‐g‐PAA‐N3) were synthesized by sequential ring‐opening reaction of polysuccinimide (PSI) with mPEG‐NH2 (MW: 2000 Da), and 1‐azido‐3‐aminopropane, respectively. Then N2‐(hex‐5‐yne)‐diethylenetriamine‐tetra‐t‐butylacetate (DTTA‐der) was conjugated to mPEG‐g‐PAA‐N3 by click cycloaddition. After deprotection of carboxylic groups, mPEG‐g‐PAA‐DTTA macromolecular ligands were obtained. MPEG‐g‐PAA‐(DTTA‐Gd) complex nanomicelles were fabricated from mPEG‐g‐PAA‐DTTA and Gadolinium chloride. The formation of nanomicelles was confirmed by fluorescence spectrophotoscopy and particle size measurements. It was found that all the nanomicelles showed spherical shapes with core‐shell structures and narrow size distributions. Their sizes ranged from 50 to 80 nm, suggesting their passive targeting potential to tumor tissue. With the increase of graft degree (GD) of mPEG, the sizes of mPEG‐g‐PAA‐(DTTA‐Gd) nanomicelles showed a tendency to decrease. Compared with gadopentetate dimeglumine (Gd‐DTPA), mPEG‐g‐PAA‐(DTTA‐Gd) nanomicelles showed essential decreased cytotoxicity to KB cell line and enhanced T1‐weighted signal intensity, especially at low concentration of gadolinium (III), suggesting their great potentials as magnetic resonance imaging contrast agents. © 2010 Wiley Periodicals, Inc. Reduction‐responsive drug delivery systems have recently gained intense attention in intracellular delivery of anticancer drugs. In this study, we developed a PEGylated polypeptide, poly(ethylene glycol)‐block‐poly(?‐propargyloxycarbonyl‐l ‐lysine) (PEG113b‐PPAL), as a novel clickable substrate for conjugation of reduction‐responsive side chains for antineoplastic drug delivery. PEG113b‐PPAL was synthesized through ring‐opening polymerization of alkyne‐containing N‐carboxyanhydride monomers. A designed disulfide‐containing side chain was introduced onto the PEGylated polypeptide by click reaction. The obtained copolymer PEG113b‐P(Lys‐DSA) formed micelles by self‐assembly, which exhibited reduction‐responsive behavior under the stimulus of 10 mmol L–1 glutathione (GSH) in water. A small molecule intermediate, compound 2 , was used as a model to investigate the thiol reduction mechanism of PEG113b‐P(Lys‐DSA) copolymers. The anticancer drug doxorubicin (DOX) was then loaded into the micelles with a drug loading content of 6.73 wt% and a loading efficiency of 40.3%. Both the blank and the drug‐loaded micelles (DOX‐loaded polylysine derived polymeric micelles (LMs/DOX)) adopted a spherical morphology, with average diameters of 48.0 ± 13.1 and 63.8 ± 20.0 nm, respectively. The in vitro drug release results indicated that DOX could be released faster from the micelles by the trigger of GSH in phosphate buffered saline. Confocal laser scanning microscopy and flow cytometer analysis further proved the intracellular delivery of DOX by LMs/DOX and their GSH‐sensitive release behavior. A 3‐(4,5‐dimethyl‐thiazol‐2‐yl)‐2,5‐diphenyl tetrazolium bromide assay showed that the polymers exhibited negligible cytotoxicity towards normal L929 cells or cancer MCF‐7 cells with a treated concentration up to 1.0 mg mL–1. This study synthesizes thermally sensitive block copolymers poly(N‐isopropylacrylamide)‐b‐poly(4‐methyl‐ε‐caprolactone) (PNIPA‐b‐PMCL) and poly(N‐isopropylacrylamide)‐b‐poly(4‐phenyl‐ε‐caprolactone) (PNIPA‐b‐PBCL) by ring‐opening polymerization of 4‐methyl‐ε‐caprolactone (MCL) or 4‐phenyl‐ε‐caprolactone (BCL) initiated from hydroxy‐terminated poly(N‐isopropylacrylamide) (PNIPA) as the macroinitiator in the presence of SnOct2 as the catalyst. This research prepares a PNIPA bearing a single terminal hydroxyl group by telomerization using 2‐hydroxyethanethiol (ME) as a chain‐transfer agent. These copolymers are characterized by differential scanning calorimetry (DSC), 1H‐NMR, FTIR, and gel permeation chromatography (GPC). The thermal properties (Tg) of diblock copolymers depend on polymer compositions. Incorporating larger amount of MCL or BCL into the macromolecular backbone decreases Tg. Their solutions show transparent below a lower critical solution temperature (LCST) and opaque above the LCST. LCST values for the PNIPA‐b‐PMCL aqueous solution were observed to shift to lower temperature than that for PNIPA homopolymers. This work investigates their micellar characteristics in the aqueous phase by fluorescence spectroscopy, transmission electron microscopy (TEM), and dynamic light scattering (DLS). The block copolymers formed micelles in the aqueous phase with critical micelle concentrations (CMCs) in the range of 0.29–2.74 mg L?1, depending on polymer compositions, which dramatically affect micelle shape. The experiment and dissipative particle dynamics simulation were carried out on four polymers with different block ratios for the investigation of the structure–property relationship of (poly(ε‐caprolactone)2‐[poly(2‐(diethylamino)ethyl methacrylate)‐b‐poly(poly(ethylene glycol) methyl ether methacrylate)]2 [(PCL)2(PDEA‐b‐PPEGMA)2] micelles. The miktoarm star polymers assembled into spherical micelles composed of PCL core, pH‐sensitive PDEA mesosphere and poly (ethylene glycol) methyl ether methacrylate (PPEGMA) shell. When decreasing pH from 7.4 to 5.0, the hydrodynamic diameter and transmittance of (PCL)2(PDEA‐b‐PPEGMA)2 micelles increased along with globule‐uneven‐extended conformational transitions, owing to the protonation of tertiary amine groups of DEA at lower pH conditions. Doxorubicin (DOX) was mainly loaded in the pH‐sensitive layer, and more DOX were loaded in the core when increasing drug concentrations. The in vitro DOX release from the micelles was significantly accelerated by decreasing pH from 7.4 to 5.0. The results demonstrated that the pH‐sensitive micelles could be used as an efficient carrier for hydrophobic anticancer drugs, achieving controlled and sustained drug release. © 2014 American Institute of Chemical Engineers AIChE J, 60: 3634–3646, 2014  相似文献
