The chloroform and aqueous fractions of Carissa opaca fruit, a traditional medicinal fruit in Pakistan possessed a high amount of total phenolic and flavonoid contents as compare to other solvent fractions with potent antioxidant activities in scavenging DPPH, superoxides, hydroxyl, hydrogen peroxide, ABTS radicals, and had strong iron chelating activity. On the other hand, the ethyl acetate fraction showed the highest inhibition of β-carotene linoleic acid peroxidation and phosphomolybdate assay. A high correlation coefficient existed between EC50 values of DPPH, superoxides, hydroxyl, hydrogen peroxide, ABTS radicals, total phenolics and flavonoids, but a non significant correlation was found in the case of iron chelaters, β-carotene and phosphomolybdate assay. This study verified that the chloroform and aqueous fractions have strong antioxidant activities which were correlated with its high level of phenolics and flavonoids. These fractions can be used as a source of potential antioxidant or functional food material.  相似文献

洋甘菊各萃取相抗氧化活性及其有效成分分析   总被引:1,自引:0,他引:1  
目的:筛选洋甘菊醇提物抗氧化活性最强的萃取组分,并对该组分抗氧化活性的有效成分进行研究。方法:采用DPPH·、ABTS+·、O2-·自由基清除能力和FRAP抗氧化功能测试方法,对比研究了洋甘菊醇提物不同极性萃取组分的抗氧化活性,并运用HPLC等方法对抗氧化能力最强萃取组分进行有效成分分析。结果:洋甘菊乙酸乙酯萃取组分的抗氧化能力最强,对DPPH·、ABTS+·和O2-·的IC50分别为25.6、30.6和83.3 mg/mL,总抗氧化能力(以trolox计)为16.7 mg/mL。成分分析表明,乙酸乙酯萃取相中总酚和总黄酮含量分别为25.84%和14.93%,在检测到的10种多酚或黄酮类化合物中,木犀草素、蒙花苷和木犀草素-7-葡萄糖含量较丰富。结论:洋甘菊醇提物的乙酸乙酯萃取组分具有良好的抗氧化能力可能与其富含的多酚和黄酮成分有关。  相似文献

Three fractions (peel, pulp and aril) of gac fruit (Momordica cochinchinensis Spreng) were investigated for their phytochemicals (lycopene, beta-carotene, lutein and phenolic compounds) and their antioxidant activity. The results showed that the aril had the highest contents for both lycopene and beta-carotene, whilst peel (yellow) contained the highest amount of lutein. Two major phenolic acid groups: hydroxybenzoic acids and hydroxycinnamic were identified and quantified. Gallic acid and p-hydroxybenzoic acid were found in all fractions. Ferulic acid and p-hydroxybenzoic acid were most evident in pulp. Myricetin was the only flavonoid found in all fractions. Apigenin was the most predominant flavonoid in pulp (red), whereas rutin and luteolin gave the highest content in aril. The extracts of different fractions exhibited different levels of antioxidant activity in the systems tested. The aril extract showed the highest FRAP value. The greatest antioxidant activities of peel and pulp extracts were at immature stage, whereas those in the seed extracts increased from mature stage to ripe stage. The contents of total phenolic and total flavonoid in peel and pulp decreased during the fruit development stage (immature > ripe fruit) and subsequently displayed lower antioxidant capacity, except for the seed.  相似文献

Antioxidant activities of ethanolic extract from dill flower and its various fractions were evaluated with 2,2-diphenyl-1-picrylhydrazyl radical scavenging, Trolox equivalent antioxidant capacity, reducing power, chelating power, and β-carotene bleaching assays. The flower extract was successively separated into n-hexane, ethyl acetate and ethanol soluble fractions by liquid–liquid partition. Dill leaf and seed extracts were used for comparison. In all assays, the flower extract showed higher antioxidant activity than the leaf and seed extracts. With regard to various fractions of the flower extract, the sequence for antioxidant activity was ethyl acetate fraction > ethanol fraction > original flower extract > n-hexane fraction. Phenols including flavonoids and proanthocyanidins should be responsible for antioxidant abilities of the flower extract. Chlorogenic acid, myricetin, and 3,3’,4′,5,7-pentahydoxyflavan (4 → 8)-3,3′,4′,5,7-pentahydoxyflavan were the major phenolic acid, flavonoid, and proanthocyanidin, respectively, in the dill flower extract.  相似文献

The aim of this work was to examine the chemical constituents and antioxidant potential of water-soluble fractions from the commonly consumed vegetable, Allium vineale. The water-soluble fraction, containing phenolic compounds, was extracted with ethyl acetate to obtain flavonoids which were separated and purified by repeated column chromatography over Sephadex LH-20, RP C18 and silica gel. The isolated compounds were identified according to their physicochemical properties and spectral data (UV, HPLC–TOF/MS, 1H NMR, 13C NMR and 2D NMR). Three flavonoids were isolated and identified as chrysoeriol-7-O-[2″-O-E-feruloyl]-β-d-glucoside (1), chrysoeriol (2), and isorhamnetin-3-β-d-glucoside (3). Antioxidant studies of the aqueous extract and three isolated compounds, 1, 2, 3, were undertaken and they were found to have significant antioxidant activity. Antioxidant activities were evaluated for total antioxidant activity by the ferric thiocyanate method, ferric ion (Fe3+) reducing antioxidant power assay (FRAP), ferrous ion (Fe2+) metal chelating activity, and DPPH free radical-scavenging activity. The water-soluble ethyl acetate and methanol extraction methods were also compared using HPLC–TOF/MS.  相似文献

青钱柳叶活性成分的抗氧化活性及UPLC-QTOF-MS/MS分析   总被引:2,自引:0,他引:2  
利用96孔板法测定青钱柳叶不同溶剂(水、70%乙醇、乙酸乙酯和正己烷)提取物中总酚、总黄酮含量及抗氧化活性(DPPH自由基清除能力、还原能力、总抗氧化能力),考察酚类物质含量与抗氧化活性的相关性,并采用超高效液相色谱-四极杆飞行时间串联质谱(ultra performance liquid chromatography coupled with quadrupole time-of-flight tandem mass spetrometry,UPLC-QTOF-MS/MS)分析提取物中主要活性成分。结果表明:不同提取物中总酚、总黄酮含量及抗氧化活性存在显著性差异且70%乙醇溶液提取物表现出最高的总酚(219.01 mg GAE/g)、总黄酮含量(7.23 mg CE/g)及最强的DPPH自由基清除能力(35.46 mg TE/g)和还原能力(1.89 mmol Fe SO_4/g);总酚、总黄酮含量与DPPH自由基清除能力、还原能力之间呈正相关,与总抗氧化能力呈显著负相关,表明多酚类物质是青钱柳中主要的抗氧化剂。UPLC-QTOF-MS/MS分析70%乙醇溶液提取物并初步鉴定出22种化合物,包括2种有机酸、4种酚酸、5种黄酮、8种三萜皂苷类和3种酯类,其中酚酸和黄酮类化合物是主要的抗氧化活性成分,有机酸、三萜皂苷及酯类化合物可能是潜在的抗氧化活性成分。  相似文献

目的:研究恶味乳杆菌B2(Lacbacillus perolens B2)生物转化对银杏花粉黄酮组分及其抗氧化活性的影响。方法:分析银杏花粉及其生物转化产物的提取物和各自石油醚相、乙酸乙酯相和正丁醇相萃取物的得率和高效液相色谱图,通过2,2’-连氮基-双-(3-乙基苯并二氢噻唑啉-6-磺酸)、1,1-二苯基-2-三硝基苯肼自由基清除能力评价其体外抗氧化活性,通过对H2O2损伤小鼠巨噬细胞RAW264.7的保护作用评价其细胞抗氧化活性。结果:通过L. perolens B2的生物转化,提取物及各相萃取物得率均有所提高,银杏花粉主要黄酮苷被转化为以山柰酚为代表的黄酮苷元,并富集于乙酸乙酯萃取相。体外抗氧化活性评价显示,通过L. perolens B2的生物转化,提取物和各相萃取物的体外抗氧化能力均得到提高,其中生物转化产物的乙酸乙酯相萃取物的体外抗氧化活性最高。细胞实验显示,供试的银杏花粉及其生物转化产物的提取物和各自的乙酸乙酯相萃取物均可减缓由H2O2损伤导致的RAW264.7细胞凋亡,并且存在剂量依赖关系。生物转化产物的提取物及乙酸乙酯相萃取物对H2O2损伤RAW264.7细胞的保护能力均高于对应的银杏花粉样品,其中生物转化产物的乙酸乙酯相萃取物保护能力最强,当其质量浓度为15 μg/mL时,几乎可以完全保护RAW264.7细胞不受H2O2损伤。结论:通过L. perolens B2的生物转化,银杏花粉主要黄酮苷被转化为以山柰酚为代表的黄酮苷元,提取物及各相萃取物的得率、体外抗氧化活性及对H2O2诱导损伤的RAW264.7细胞的保护作用均显著提高(P<0.05),其中生物转化产物的乙酸乙酯相萃取物抗氧化活性最高。  相似文献

目的:对木贼的乙醇提取物不同萃取部位进行总酚酸、黄酮含量测定及抗氧化活性比较,以探知木贼抗氧化活性物质基础。方法:木贼经70%乙醇超声提取浓缩后,采用有机溶剂萃取法依次得到石油醚、乙酸乙酯、正丁醇、水相4个部位萃取物;利用紫外-分光光度法比较各萃取部位总黄酮、酚酸含量差异;以抗坏血酸为阳性对照,比较不同萃取部位还原能力、羟自由基(·OH)、1,1-二苯基-2-三硝基苯肼(DPPH·)、2,2'-联氮-双(3-乙基苯并噻唑-6-磺酸)二铵盐(ABTS+·)清除率的差异,评价各部位抗氧化能力强弱。结果:木贼各萃取物总黄酮含量在5.70~61.55 mg/g 范围内,总酚酸含量在4.07~19.10 mg/g 范围内,乙酸乙酯萃取物总黄酮与酚酸含量明显高于其他萃取物,其还原能力、羟自由基、DPPH·、ABTS+·清除率也明显高于正丁醇、石油醚和水相萃取物,乙酸乙酯萃取物对羟自由基、ABTS+·、DPPH·清除能力IC50值分别为:(0.321±0.0026)、(0.213±0.0010)和(0.169±0.0014) mg/mL。结论:经各相溶剂萃取后木贼有效成分得到初步分离,乙酸乙酯相萃取部位总黄酮和酚酸相对含量及抗氧化活性明显强于总提物和其他萃取部位,且其抗氧化能力呈量效关系。实验发现木贼中黄酮和酚酸类成分与其抗氧化能力密切相关。  相似文献

Smilax excelsa L. leaves are used widely in the Black Sea region of Turkey for consumption in the daily diet and in folk medicine for their medicinal properties. In the present study, different antioxidant tests were employed in order to evaluate the antioxidant activities of water, infusion, ethanol and ethyl acetate extracts of S. excelsa leaves. In addition, the results were compared with natural and synthetic antioxidants. The levels of total phenolics, total flavonoids and anthocyanins of the extracts were also determined. The extracts were found to have different levels of antioxidant properties in the test models used. All extracts had good total phenolic and flavonoid contents, inhibited lipid peroxidation, showed radical scavenging and iron-chelating activities. Therefore, the leaves of the plant could be considered as a significant natural antioxidant source.  相似文献

Hovenia acerba Lindl. is a perennial tree of Rhamnaceae family. In the present study, four solvents (80% methanol, ethyl acetate, n-hexane, and water) were used to extract the compounds of Hovenia acerba Lindl. peduncles (HAP). The total contents of phenolics and tannins (TPC and TTC), as well as their antioxidant activities (DPPH, ABTS, and FRAP) of these four different extracts including 80% methanol extract (ME), ethyl acetate extract (EE), n-hexane extract (HE) and water extract (WE) were compared. The antioxidant activities of HAP showed positive correlations with TPC and TTC, indicating that phenolics maybe a major contributor to the antioxidant activities of HAP. The significant difference between antioxidant activities and TPC/TTC implied the respective contribution extent of phenolics for the antioxidant activities of four solvent extracts. Besides, a total of 117 compounds, including 7 oligosaccharides, 11 organic acids, 41 phenolics, 1 steroids, 25 terpenes, 11 fatty acids, 9 amino acids, and 12 other compounds were identified or tentatively characterized in HAP for the first time. Among them, 23 bioactive constituents, including 4 organic acids, 5 phenolic acids, and 14 flavonoids, were quantified. The comparison of characterization and quantitation of 4 extracts demonstrated that 80% methanol showed the widest range of selectivity for both HAP flavonoid aglycones and glycosides, while ethyl acetate preferred to extract the flavonoids aglycone from HAP. Secondly, water and 80% methanol were excellent solvents for the extraction of phenolic acids. And both ethyl acetate and 80% methanol perform well at the extraction of terpenes and steroids.  相似文献

The objective of this study was to assess antioxidant and antiproliferative activities of four different Turkish pomegranate varieties (Hatay, Hicaz, Adana and Antalya) using an in vitro HepG2 cancer cell model. All the pomegranate extracts employed in this study significantly diminish the proliferation of HepG2 cells in a dose‐dependent manner. The total phenolic acid, anthocyanin and flavonoid contents for each of the four varieties were determined. The Hatay pomegranate variety had the highest total phenolic acid (337.4 ± 2.34 mg/100 g) and flavonoid (58.42 ± 2.25 mg/100 g) contents of the pomegranates examined. Antioxidant activities of the pomegranates were determined using DPPH and ABTS radical scavenging assays. The lack of correlation between colour index value and antioxidant–antiproliferative activities suggested that phenolic acids and flavonoids are predominant compounds influencing pomegranate's bioactivity rather than anthocyanins. Individual phenolic acids found in Hatay pomegranates were determined, using an HPLC system, as gallic acid being the most predominant phenolic compound.  相似文献

以矮丛红豆越橘为研究对象,用体积分数95%乙醇提取后,分别用石油醚、乙酸乙酯、正丁醇进行萃取,评价乙醇粗提取物和不同溶剂萃取物中总酚、总黄酮、总花色苷、齐墩果酸含量及体外抗氧化能力(1,1-二苯基-2-三硝基苯肼(1,1-diphenyl-2-picrylhydrazyl,DPPH)自由基、2,2’-联氮双(3-乙基苯并噻唑啉-6-磺酸)二铵盐自由基(2,2’-azinobis-(3-ethylbenzthiazoline-6-sulphonate),ABTS~+·)清除能力)。红豆越橘粗提取物及不同溶剂萃取物均具有一定的抗氧化活性,其中乙酸乙酯、正丁醇萃取物的抗氧化活性较强,乙酸乙酯萃取物具有最好的DPPH自由基清除能力(半抑制浓度(50%inhibitory concentration,IC_(50))为0.27 mg/m L)与ABTS+·清除能力(IC_(50)为0.05 mg/m L)。不同溶剂萃取物的抗氧化活性与总黄酮和总酚的含量具有较强的相关性。  相似文献

Chinese herbs employed in traditional Chinese medicine (TCM) have been used for centuries in the practice of medicated diet and dietetic therapy. The seed of Cuscuta chinensis Lam. (Convolvulaceae), a commonly used traditional Chinese herb, is frequently added in Chinese cooking and preparation of refreshments, including porridge and alcoholic beverages, to nourish the human body. In the present study, we compared the antioxidant activities of water and ethanol extracts from the seeds C. chinensis and also of its different organic fractions, including n-hexane, ethyl acetate, n-butanol and organic water, by assessing their DPPH (1,1-diphenyl-2-picryl hydrazine) free radical-scavenging, superoxide anion scavenging, anti-superoxide anion formation and anti-lipid peroxidation abilities. The flavonol contents of all test samples were analyzed by high-performance liquid chromatography with an ultraviolet (HPLC–UV) detector. The results showed that there is a direct correlation of the flavonol content with the antioxidant activities from the extracts and fractions of C. chinensis. Moreover, the ethyl acetate fraction demonstrated significantly better and higher antioxidant effects, and also had a higher flavonol content than had the remaining samples (P < 0.05). The water fractions, however, exhibited the weakest antioxidant activity, and had low concentrations of flavonols. Thus, we suggest that the ethanol extract of C. chinensis, but not its water extract, could be used as a dietary nutritional supplement to promote human health and prevent oxidation-related diseases, due to its antioxidant properties.  相似文献

Nyctanthes arbor-tristis (Harsingar) leaf extracts are extensively used in Indian traditional medicine. The acetone-soluble fraction of its ethyl acetate extract showed impressive antioxidant activity as revealed by several in vitro experiments, e.g., DPPH, hydroxyl and superoxide radicals, as well as H2O2 scavenging assays. Moreover, its preventive capacity against Fe(II)-induced lipid peroxidation of liposomes and γ-ray-induced DNA damage also confirmed this. The strong reducing power and high phenolics and flavonoids contents could be responsible for the antioxidant activity.  相似文献

Purified flavonoid extract from illuminated parsley (Petroselinum crispum (Mill.) Nym.) cell culture was administered by gavage to Wistar rats. The dose corresponded to 6.9 mg flavonoids on aglycone base/kg body mass. Segments of the gastrointestinal wall from stomach to colon, their luminal contents, and liver and kidneys were collected at time intervals between 1 and 12 h and investigated by HPLC of the respective extracts for flavonoids. The spreading of the flavonoids was accompanied by partial deglycosylation that began already in the stomach where at first quercetin and later apigenin, chrysoeriol and isorhamnetin aglycones were detected. We got evidence of flavonoid absorption by the stomach that does not require the liberation of aglycones. Due to obvious differences in metabolization and absorption rates the composition and the content of flavonoids changes in the gastrointestinal segments and their contents with time. Flavonoids could be detected neither within the gastrointestinal lumen after 12 h nor in the kidneys at any time. But traces of flavonoids were found in the livers at 1.5 and 12 h.  相似文献

以小浆果果树红树毒叶片为实验材料,利用分光光度法测定其总黄酮含量,并采用烘箱贮存法研究了不同添加量红树毒叶片提取物粗黄酮在猪油中的抗氧化性。结果表明:鲜样中红树毒叶片总黄酮含量是278.73mg/100g,干样中是533.71mg/100g;红树毒叶片提取物粗黄酮对猪油有一定的抗氧化作用,并随添加量的增加而增强;增效剂以(柠檬酸)对红树毒叶片提取物粗黄酮无增效作用,而Vc具有一定的增效作用。抗氧化活性顺序为:0.02%槲皮素>0.02%PG>0.02%BHT>0.4%粗黄酮>0.05%粗黄酮 0.05%Vc>0.2%粗黄酮>0.1%粗黄酮>0.05%粗黄酮>0.02%粗黄酮>0.05%粗黄酮 0.05%CA>对照。  相似文献

Frequent consumption of nuts has been linked to a lowered risk of cardiovascular disease. Phytochemicals, especially phenolics, in nuts may be considered to be the major bioactive compounds for health benefits. Nine types of tree nuts and peanuts commonly available in the United States were evaluated for total phenolic and flavonoid contents, antioxidant, and antiproliferative activities. The profiles of total phenolics and flavonoids, including both soluble free and bound forms, were investigated by utilizing solvent extraction, base digestion, and solid-phase extraction methods. Walnuts had the highest total phenolic and flavonoid contents (1580.5 ± 58.0 mg/100 g, 744.8 ± 93.3 mg/100 g, respectively). Walnuts also possessed the highest total antioxidant activity (458.1 ± 14.0 μmol of vitamin C equiv/g of nut). Both soluble phenolic and flavonoid contents were positively correlated with total antioxidant activity (R2 = 0.9901, p < 0.05; and R2 = 0.9749, p < 0.05, respectively). The proliferation of HepG2 and Caco-2 cells was significantly inhibited in a dose-dependent pattern after exposure to the extracts of nuts, with walnuts and pecans exhibiting the highest antiproliferative activity. These results provide new knowledge about health functions of nuts and may influence consumers toward purchasing nuts exhibiting greater potential health benefits.  相似文献

以覆盆子醇提物及其不同极性部位的总酚、黄酮含量和抗氧化活性为研究目的,采用还原力,1,1-二苯基-2-苦基肼(DPPH)和OH自由基清除能力抗氧化体系来评价它们的抗氧化活性,研究结果表明:乙酸乙酯相的总酚和黄酮含量最高,分别为(328.33±13.75)mg/g和(298.25±25.64)mg/g(p<0.05);该相在不同抗氧化体系中的抗氧化活性也最强,清除DPPH和OH的能力显著强于BHT(p<0.05)。覆盆子醇提物及其不同极性部位中的总酚含量与还原力,DPPH和OH清除能力呈正相关,相关系数分别为0.9197、0.9321和0.9808,这表明总酚是覆盆子提取物的主要抗氧化活性成分。  相似文献

This work reports the nutritional potential and the antiradical activity of the cladodes of the Brazilian Opuntia monacantha Haw. (Cactaceae), as well the isolation of kaempferol and isorhamnetin through activity-guided fractionation from its active MeOH extract. TLC, HPLC-DAD and NMR techniques were used to detect and identify the compounds. The water (91.1%), ash, protein, fibre and lipid contents (15.0, 5.4, 18.5 and 1.4 g/100 g, respectively) were shown to be quite similar to the mean values of other Opuntia spp., some widely used as food and forage. The antioxidant activity of their MeOH extract and of its n-hexane, EtOAc and n-BuOH fractions was assessed by measuring the ability of the fractions to scavenge DPPH radical, showing an activity enhancement for the fractions in comparison to the crude MeOH extract. The well-known free-radical scavenging activity of the isolated flavonoids reinforces the contribution of these compounds to the presented activity of the O. monacantha cladodes.  相似文献
