A survey was conducted to determine the occurrence of fumonisin B1, B2 and B3 during 2007 in 186 samples of organic and conventional locally available corn products. Samples included baby food (n = 62), corn flour (11), cornflakes (23), pasta (14), cookies (17) and other corn products (59) were obtained from popular markets of Valencia (Spain) and Perugia (Italy). The analytical method used pressurized liquid extraction and liquid chromatography/electrospray ionization tandem mass spectrometry with a triple quadrupole (QqQ) analyser. Of the 104 Spanish samples, 22% contained levels in the range of 2–449 µg kg?1, 2–229 µg kg?1 and 6–105 µg kg?1 for FB1, FB2 and FB3, respectively, while 19 (23%) of the 82 Italian samples were positive with quantifiable levels between 2–235 µg kg?1, 3–187 µg kg?1, and 4–40 µg kg?1 for fumonisins B1, B2 and B3, respectively. Overall, none of the Italian samples and only one organic baby food sample from a Spanish market was above the maximum permitted levels established by European legislation. Fumonisins were found mostly in corn flour followed by cookies and cornflakes. Eleven samples from Spain and nine samples from Italy were organic products, being contaminated the 72% and 77% of the samples, respectively. Analysis of the results showed that levels of fumonisins in corn products were similar in Italy and Spain. The safety of fumonisin intake through corn products was demonstrated by the calculation of the estimated daily intake of both populations considering organic and conventional products separately, which ranged from 1.7 × 10?3 to 0.72 µg kg?1 bw day?1 and comparing them with the provisional maximum total daily intake (PMTDI) of 2 µg kg?1 bw day?1 established by the European Union.  相似文献

The occurrence of deoxynivalenol (DON) was investigated in 514 cereal-based products (corn-based, n = 125; barley-based, n = 96; wheat-based, n = 94; rice-based, n = 199) marketed in Korea during 2007?2008, and estimates of DON intake were determined. Samples were analysed by high-performance liquid chromatography (HPLC) with ultraviolet light (UV) detection after immunoaffinity clean-up. The limits of detection (LOD) and limits of quantification (LOQ) were 2.2 and 5.6 µg kg–1, respectively. Recoveries and repeatability expressed as coefficients of variation (CV) were 82.3–100% and 2.4–15.3% in beer, bread and dried corn. The incidences and mean levels of DON were 56% and 68.9 µg kg?1 for corn-based products, 49% and 24.1 µg kg?1 for wheat-based products, 43% and 7.5 µg kg?1 for barley-based products, and 16% and 3.4 µg kg?1 for rice-based products, respectively. The estimated daily intake of DON from the consumption of rice-based, wheat-based, barley-based and corn-based products were 0.0038 µg kg?1 bw day?1, 0.0032 µg kg?1 bw day?1, 0.0015 µg kg?1 bw day?1 and 0.0002 µg kg?1 bw day?1, respectively. These values represent 0.38%, 0.32%, 0.25% and 0.01% of the provisional maximum tolerable daily intake (PMTDI) of 1 µg kg?1 bw day?1. These results indicate that rice-based products are major contributors to DON exposure in Korea, even though the current exposure level is unlikely to cause adverse health effects.  相似文献

A survey of 11 mycotoxins in 348 wheat flour samples marketed in Hebei province of China were analysed by liquid chromatography-tandem mass spectrometry, was carried out. The selected mycotoxins consisted of four aflatoxins (AFs: AFB1, AFB2, AFG1 and AFG2) and seven Fusarium toxins, i.e. deoxynivalenol (DON), nivalenol, 3-acetyldeoxynivalenol and 15-acetyldeoxynivalenol, zearalenone, Fusarenon-X and deoxynivalenol-3-glucoside. Results indicated that most of the wheat samples analysed were contaminated with mycotoxins. Wheat was most susceptible to DON (91.4% contamination), with a mean level of 240 μg kg?1. On average the probable daily intake (PDI, expressed as µg kg?1 body weight day?1) of mycotoxins was within the provisional maximum tolerable daily intake (PMTDI, 2.0 µg kg?1 of body weight day?1) as set by the Joint FAO/WHO Expert Committee on Food Additives. Nevertheless, exposure assessment revealed that the maximum PDI of mycotoxins was 4.06 µg kg?1 body weight day?1, which was twice the PMTDI value. Fumonisins B1, B2 and B3 (FB1, FB2 and FB3) and aflatoxins B1 (AFB1), B2 (AFB2), G1 (AFG1) and G2 (AFG2) are both major mycotoxins of food concern, because of their wide range of concentration and possible co-occurrence. Therefore, a contamination survey in corn and wheat flour by liquid chromatography–tandem mass spectrometry was carried out. Quantification of fumonisins and aflatoxins was based on internal calibration (by the use of 13C34-fumonisin) and external calibration, respectively. Fumonisins were detected in 95% of corn samples and in 7% of wheat flour samples, with the mean level (FB1?+?FB2?+?FB3) of 441?µg?kg?1 and 0.09?µg?kg?1, respectively. Low levels of aflatoxins were detected in 37% of the samples with a mean level (B1?+?B2?+?G1?+?G2) of 0.12?µg?kg?1. Fumonisins and aflatoxins were not detected in 29% of the samples analysed. Simultaneous occurrence of fumonisins and aflatoxins was observed in 12% of samples.  相似文献

The main objective of this research was to perform an exposure assessment of aflatoxins intake through consumption of maize products in Serbia, Croatia, and Greece. Food consumption survey of maize-based products has been performed during 2017 in the three countries with at least 1,000 interviewees per country covering their dietary habits and body weight. Values for the concentration of aflatoxins were extracted from available research published in the last ten years. Finally, a Monte Carlo analysis of 100,000 iterations was performed to estimate the intake of aflatoxins from consumption of maize-based products. Results revealed that the estimated average exposure of adults to aflatoxins, from maize consumption, in each of the three countries was between 0.44 ng kg?1 bw day?1 and 5.59 ng kg?1 bw day?1. Margin of exposure values for the mean exposure levels, in all three countries, were between 30 and 389. Estimations for hepatocellular carcinoma cases/year/105 individuals, depending on the HBsAg+ prevalence, were 0.075–0.098, 0.006–0.008 and 0.020–0.026 for Serbia, Croatia and Greece, respectively.  相似文献

Aflatoxins were surveyed in 256 rice samples taken from retail markets in different provinces of Iran during October 2007 and July 2008. A methanol/water (80?:?20,?v/v) mixture and an aflatoxin immunoaffinity column (IAC) were used for extraction and clean-up. Mycotoxins were determined using HPLC with fluorescence detection and post-column derivatization using a photo-ionization cell. Levels of contamination ranged 0.0–5.8?ng?g?1 (mean, 1.4?ng?g?1) and 0.1–6.3?ng?g?1 (mean, 1.6?ng?g?1) for AFB1 and total aflatoxins, respectively. AFB1 was detected in almost all samples. Results showed that 55 samples (21.5%) were contaminated with more than 2?µg?kg?1 of AFB1, while seven samples (2.7 %) contained more than 4?µg?kg?1 total aflatoxins. The calculated probable daily intake of AFB1 from rice for Iranians ranged 1.4–5.8?ng AFB1 per kg body weight per day for average consumers and, hence. exceeding the estimated provisional maximum tolerable daily intake.  相似文献

The objective of this study was to determine the distribution of an economically important class of mycotoxins, the aflatoxins, in rice milling fractions. Rice plants grown under field production conditions are frequently infected with types of pathogenic fungi that produce toxic metabolites (mycotoxins). Paddy (seeds) rice from healthy plants in the field was collected and stored on a farm under humid, poorly ventilated conditions. Samples were milled into four fractions (hulls, brown rice, bran and white rice) and analysed for aflatoxins (B1, B2, G1 and G2) using a validated method. Rice fractions from healthy plants, which contained low levels of aflatoxins (less than 1?µg?kg?1), were used to determine the efficiency of the extraction method. Seeds stored under poor conditions were found to be contaminated with aflatoxins B1 and B2 as were the fractions. The sums of AFB1 and AFB2 in stored paddy rice, hulls, brown rice, bran and white rice were 141, 39, 158, 367 and 56?µg?kg?1, respectively. The ratio of aflatoxin B1 and B2 was about 10?:?1. AFG1 and AFG2 were less than 1?µg?kg?1. Thus, brown rice contained 92.9% of the aflatoxins in paddy rice, whereas white rice contained only 27.9%.  相似文献

The intake of total aflatoxins (AFT) and aflatoxin B1 (AFB1) from food in Japan was estimated from AFT and AFB1 concentration and frequency data in 24 foods (884 samples) from a 3-year retail market survey from the summer of 2004 to the winter of 2006, and by food consumption data from the National Health and Nutrition Survey performed in 2005. The AFT and AFB1 survey revealed that peanut, peanut products, cocoa, chocolate, pistachio, white pepper, red pepper, almond, job's tears, buckwheat and corn grits are considered to be contributors of AFT (or AFB1) intake in Japan (maximum AFB1 (AFT) levels ranged from 0.21 to 28.0 µg kg?1 (from 0.21 to 9.0 µg kg?1)) in AFT-contaminated food. A probabilistic approach using the Monte Carlo method was carried out to simulate an estimate of the AFT (or AFB1) intake distributions in each age group in Japan. In this study, AFB1 intake ranged from 0.003 to 0.004 ng kg?1 body weight day?1 (from lower to upper limits), and the potential risk for cancer using a formula devised by the Joint Food and Agricultural Organization/World Health Organization (FAO/WHO) Expert Committee on Food Additives (JECFA) was estimated at 0.00004–0.00005 person/year/100,000 persons, even though this was in the higher levels (95.0th percentile) of the consumer population. The aim of this study was to assess the dietary exposure of adults in Hong Kong to nitrate and nitrite from vegetables. If all vegetables consumed were raw, the dietary exposure to nitrate for average consumers was estimated to be 4.4?mg?kg?1 body weight (bw)?day?1 and, for high consumers, was estimated to be 13?mg?kg?1?bw?day?1, which is about 120 and 350% of acceptable daily intake (ADI), respectively. If all vegetables consumed were cooked, the dietary exposure to nitrate from vegetables for the average adult consumer was estimated to be 3.5?mg?kg?1?bw?day?1 and, for high consumer, was estimated to be 10?mg?kg?1?bw?day?1, which is about 95 and 270% of ADI, respectively. On the other hand, the dietary exposure to nitrite from vegetables for average and high consumers were well below the ADI.  相似文献

In view of the frequent occurrence of mycotoxins in cereals, a study was initiated to assess the exposure of the Hungarian adult population. Consumption data for 1360 individuals, based on a 3-day questionnaire, indicated that white bread accounted for the major intake of cereal-based products. Various cereal products were analysed for 16 mycotoxins by a LC/MS/MS multi-toxin method with LOD of 16?µg?kg?1 and LOQ of 50?µg?kg?1. Deoxynivalenol (DON) was most frequently detected, but no acetyl-deoxynivalenol was present in detectable concentrations. Consumer exposure was calculated with standard Monte Carlo probabilistic modelling and point estimates, taking into account bread consumption and DON contamination in independently taken wheat flour and wheat grain samples. Over 55% of cases the DON intake were below 15% of the provisional maximum tolerable daily intake (PMTDI) of 1?µg/(kg?bw)/day. However, in 5–15% of cases, the intake from bread consumption alone exceeded the PMTDI. Wheat grain data led to the higher percentage. Intakes estimated from both data sets were at or below the acute reference dose (ARfD) of 8?µg/(kg?bw)/day in 99.94–99.97% of cases.  相似文献

To evaluate the potential public health risk of ethyl carbamate (EC), EC exposure from fermented foods and beverages for Hong Kong population was estimated. In 276 samples analysed, EC was detected (limit of detection (LOD) at 0.4?µg?kg?1) in 202 samples (73%), with higher levels in fermented red bean curd (150–650?µg?kg?1) and yellow wine (140–390?µg?kg?1), while low or non-detected (ND) in preserved vegetables (ND–10?µg?kg?1) and fermented tea (ND–15?µg?kg?1). The estimated dietary exposure from all fermented foods and beverages was 8.27?ng?kg?1?bw?day?1, while exposure excluding alcoholic beverages was 5.42?ng?kg?1?bw?day?1, with calculated margins of exposure (MOEs) at 3.6?×?104 and 5.5?×?104 respectively. The risk of adverse health effects was low for the average population but higher (MOE?of?103) for high consumers of alcoholic beverages especially habitual drinkers of alcoholic types with high EC contents.  相似文献

The aim of this work was to determine deoxynivalenol (DON) contamination in wheat-based food products in Argentina and to estimate DON exposure. The numbers of samples were determined according to a developed sampling plan. A total of 156 samples of different wheat products were randomly collected from food markets in Luján, Argentina, and analyzed for DON by gas chromatography. DON contamination ranged 7–271?ng?g?1 for French bread, 5–149?ng?g?1 for Vienna bread, 11–85?ng?g?1 for crackers, 8–85?ng?g?1 for pizza, and was 79?ng?g?1 for noodles. The maximum contribution to DON intake was 7% of the PMTDI for French bread; the minimum was less than 1% for noodles. Assuming all groups had eaten all sampled foods and summing all groups’ intake contribution, the highest estimate DON exposure would only be <14% (for the 18–24-year old men group) of the DON daily dietary intake.  相似文献

This study has examined the occurrence of aflatoxins in 168 samples of different fractions obtained during the processing of cocoa in manufacturing plants (shell, nibs, mass, butter, cake and powder) using an optimised methodology for cocoa by-products. The method validation was based on selectivity, linearity, limit of detection and recovery. The method was shown to be adequate for use in quantifying the contamination of cocoa by aflatoxins B1, B2, G1 and G2. Furthermore, the method was easier to use than other methods available in the literature. For aflatoxin extraction from cocoa samples, a methanol–water solution was used, and then immunoaffinity columns were employed for clean-up before the determination by high-performance liquid chromatography. A survey demonstrated a widespread occurrence of aflatoxins in cocoa by-products, although in general the levels of aflatoxins present in the fractions from industrial processing of cocoa were low. A maximum aflatoxin contamination of 13.3?ng?g?1 was found in a nib sample. The lowest contamination levels were found in cocoa butter. Continued monitoring of aflatoxins in cocoa by-products is nevertheless necessary because these toxins have a high toxicity to humans and cocoa is widely consumed by children through cocoa-containing products, like candies.  相似文献

This paper reported on selective Raman spectroscopic and mass spectrometric determination of functionalized aflatoxins. Performance parameter methods encompassed six component mixtures of AFs, B1, G1, M1 and B2A, G2A, and M2A at concentration range of ∈ 0.096–21.581?ng g?1. Limits of detection (LODs) and limits of quantitation (LOQ) are 0.135 and 0.45 ng g?1, respectively. The matrix effect of AFB2A, AFG2A, and AFM2A in multicomponent mixtures is evaluated. The LODs of 0.11–0.09 ng g?1 is achieved. Raman spectroscopic method performance parameters at concentration range of ∈ 0.10–5,000 μg kg?1 for six component mixtures (r 2?=?0.97651) are LOD?=?10.13 μg kg?1 and LOQ?=?33.76?μg kg?1. The designed experimental schedule and sampling model is tested on contaminated grain samples. As far as the biochemistry and genetic engineering of aflatoxins and their major metabolites involved the high regioselectivity and binding affinity to N7 guanine residue at DNA, causing carcinogenic and mutagenic effects, the data reported are of emergency on the frontier fields of analytical chemistry, environmental bioengineering, monitoring, and assessment of human health risk from aflatoxins contamination. Aflatoxins are considered as biochemical weapons. Therefore, the essence of the study is on the instrumental methodology, which would be of interest and encompassed public security research and related scientific elaborations of more effective methods for (1) prevention, (2) reducing of the damages, and/or (3) acquisition of the outgrowths from biological (chemical) terrorist attacks.  相似文献

ABSTRACT

The aim of the study was to assess the dietary intake of caramel colours and their by-products 4-methylimidazole (4-MEI) and 2-acetyl-4-tetrahydroxybutylimidazole (THI) for the Chinese population. Based on the typical and maximum reported use levels of caramel colours in 15 food categories, the dietary intakes of combined and single-class caramel colours of Classes I, III and IV were estimated with the food consumption data from the China National Nutrient and Health Survey. Using the mean values of 4-MEI and THI contents in Class III and Class IV Caramel colour samples, the exposures to 4-MEI and THI from dietary caramel colours were derived. The results showed that the combined and individual average dietary caramel colour intakes for the Chinese population of different age groups were estimated to be 232–60.3 mg kg?1 bw day?1 for combined caramels, 5.9–29.2 mg kg?1 bw day?1 for Class I, 7.7–29.6 mg kg?1 bw day?1 for Class III, 21.2–54.3 mg kg?1 bw day?1 for Class IV, which were far below the group acceptable daily intake (ADI) and respective ADIs. The combined intake of 4-MEI from Class III and IV caramel colours was estimated to be 3.8–5.2 μg kg?1 bw day?1 on average, and 12.9–27.1 μg kg?1 bw day?1 at 95th-97.5th percentile for the general population. The anticipated exposure to THI from Class III caramel colours was estimated to be 0.1–0.3 μg kg?1 bw day?1 on average and 0.5–1.7 μg kg?1 bw day?1 at 95th–97.5th percentile for the general population. The dietary caramel colours intakes and the exposures to 4-MEI and THI from dietary caramel colour for the Chinese population were considered to be of low health concern based on the present toxicological data. Soy sauce, vinegar and compound seasonings were found to be the main contributors to the dietary intake of caramel colours.  相似文献

The occurrence of aflatoxins (AF) B1, B2, G1, G2 and cyclopiazonic acid (CPA) in feeds, and AFM1 and CPA in milk was determined in dairy farms located in the northeastern region of São Paulo state, Brazil, between October 2005 and February 2006. AF and CPA determinations were performed by HPLC. AFB1 was found in 42% of feed at levels of 1.0–26.4 µg kg?1 (mean: 7.1 ± 7.2 µg kg?1). The concentrations of AFM1 in raw milk varied between 0.010 and 0.645 µg l?1 (mean: 0.104 ± 0.138 µg l?1). Only one sample was above the tolerance limit adopted in Brazil (0.50 µg l?1) for AFM1 in milk. Regarding CPA in feed, six (12%) samples showed concentrations of 12.5–153.3 µg kg?1 (mean: 57.6 ± 48.7 µg kg?1). CPA was detected in only three milk samples (6%) at levels of 6.4, 8.8 and 9.1 µg l?1. Concentrations of aflatoxins and CPA in feed and milk were relatively low, although the high frequency of both mycotoxins indicates the necessity to continuously monitor dairy farms to prevent contamination of feed ingredients.  相似文献

A survey of moulds and mycotoxins was performed on 99 rice samples taken from the Swedish retail market. The main objective was to study the mould and mycotoxin content in basmati rice and rice with a high content of fibre. Samples of jasmine rice as well as long-grain rice were also included. The samples were analysed for their content of ochratoxin A (high-performance liquid chromatography (HPLC)), aflatoxin B1, B2, G1, and G2 (HPLC, RIDA®QUICK), and mould (traditional cultivation methods in combination with morphological analysis). The majority of samples were sampled according to European Commission Regulation 401/2006. Subsamples were pooled and mixed before milling and both mould and mycotoxin analyses were performed on milled rice. The results showed that the majority of basmati rice (71%) and many jasmine rice samples (20%) contained detectable levels of aflatoxin B1 (level of quantification = 0.1 µg aflatoxin kg?1 rice). Two samples of jasmine rice and ten basmati rice samples contained levels over the regulated European maximum limits of 2 µg kg?1 for aflatoxin B1 or 4 µg kg?1 for total aflatoxins. Aspergillus was the most common mould genus isolated, but also Penicillium, Eurotium, Wallemia, Cladosporium, Epicoccum, Alternaria, and Trichotecium were found. The presence of Aspergillus flavus in 21% of the samples indicates that incorrect management of rice during production and storage implies a risk of mould growth and subsequent production of aflatoxin. Rough estimates showed that high rice consumers may have an intake of 2–3 ng aflatoxin kg?1 bodyweight and day?1 from rice alone. This survey shows that aflatoxin is a common contaminant in rice imported to Europe.  相似文献

The effect of village processing techniques on the aflatoxin content of corn and peanut products was investigated. In 30 trials, corn kernels were dehulled (bran removal), soaked for 24 h, washed and dried before grinding into flour and boiling in water to a thick consistency (Nshima). Shelled peanuts were either dry-roasted as whole kernels or ground into peanut meal and cooked. Dehulling, following by 24-h soaking (steeping) and subsequent washing significantly (P<0·05) reduced the aflatoxin B1 content of corn flour from 900 to 150 μg kg−1, and similarly that of aflatoxin G1 from 929 to 114 μg kg−1. Preparation of Nshima did not result into a substantial reduction in aflatoxin content, neither did extension of the cooking duration of 2 h afford any further decontamination. Whereas boiling peanut meal yielded a moderate reduction in the content of aflatoxins B1 and G1, roasting whole peanut kernels greatly reduced (P<0·001) the concentrations of the toxins from that in raw kernels (AFB1= 8600 μg kg−1 and AFG1=6200 μg kg−1) to 1300 and 1200 μg kg−1, respectively. These results indicate that specific processing techniques carried out in Zambian villages are effective in reducing aflatoxin carry-over into edible fractions, while others are not. © 1998 SCI.  相似文献

The aim of this study is to obtain data on the exposure of non-breast-fed infants to polychlorinated dibenzo-p-dioxins and dibenzofurans (PCDD/F), polychlorinated biphenyls (PCB), organochlorine pesticides (OCP), and bisphenol A (BPA) and its chlorinated derivatives through consumption of commercial infant foods with largest shares of the market in 22 European Union countries. The E-Screen bioassay was employed to assess the oestrogenicity of the baby foods and the ethoxyresorufin-O-deethylase (EROD) induction was measured to determine the levels of PCDD/F and PCB. Consequently, the highest total effective xenoestrogen burden (TEXB) of 73.60?pM?Eeq?g?1 was found in the soy-based formula and the EROD bioassay was always below the limit of quantification (LOQ) (3.5?pg?g?1). Overall, the estimated dietary exposure to BPA via commercial baby foods was lower than the tolerable daily intake (TDI) of 50?µg?kg?1 body weight (bw). Furthermore, the findings indicated that the dietary exposure of 0–9-month-old infants through the products investigated here does not exceed the maximum TDI of 4?pg WHO-TEQ (toxic equivalents)?kg?1 bw. However, exposure to more than 2?pg WHO-TEQ?kg?1?bw?day?1 might occur for 0–4-month-old infants consuming ‘starting’ hypoallergenic formula. Moreover, analysis of OCP indicated that the dietary exposure of non-breast-fed infants was not harmful. Considering the importance of early development and the vulnerability of infants and children, it is essential to determine their dietary exposure to contaminants in order to decide which efforts of risk reduction should receive highest priority.  相似文献

The aflatoxins content of 140 cotton seed samples were determined using high-performance liquid chromatography. Samples were obtained from wholesalers in Iran between May 2010 and June 2011. Aflatoxin B1 gave the highest incidence of contamination and was found in 129 of the 139 samples. The highest concentration of aflatoxin was 14.4?ng?g?1. Thirteen cotton seed samples (9.35%) were above one of the regulatory limits of the European Union (5?ng?g?1), but no sample was above the highest EU limit and the safety limit recommended by the FDA (20?ng?g?1) and regulatory limits of Iran (50?ng?g?1) for total aflatoxin.  相似文献
