Two series of poly(ε‐caprolactone)‐b‐poly(ethylene glycol)‐b‐poly(ε‐caprolactone) triblock copolymers were prepared by the ring opening polymerization of ε‐caprolactone in the presence of poly(ethylene glycol) and dibutylmagnesium in 1,4‐dioxane solution at 70°C. The triblock structure and molecular weight of the copolymers were analyzed and confirmed by 1H NMR, 13C NMR, FTIR, and gel permeation chromatography. The crystallization and thermal properties of the copolymers were investigated by wide‐angle X‐ray diffraction (WAXD) and differential scanning calorimetry (DSC). The results illustrated that the crystallization and melting behaviors of the copolymers were depended on the copolymer composition and the relative length of each block in copolymers. Crystallization exothermal peaks (Tc) and melting endothermic peaks (Tm) of PEG block were significantly influenced by the relative length of PCL blocks, due to the hindrance of the lateral PCL blocks. With increasing of the length of PCL blocks, the diffraction and the melting peak of PEG block disappeared gradually in the WAXD patterns and DSC curves, respectively. Three types of pegylated amphiphilic copolymers of poly(δ‐valerolactone) (PVL) were copolymerized with methoxy poly(ethylene glycol) (MePEG) and poly(ethylene glycol) (PEG4000 and PEG10,000), respectively. Pegylation of PVL allowed copolymers possessing amphiphilic property and efficiently self‐assembled to form micelles with a low critical micelle concentration (CMC) in the range of 10?7–10?8M. The average molecular weight of copolymers was in the range of 10,000–20,000 Da, and the polydispersity of copolymers was about 1.7–1.8. Higher mobility of low molecular weight PEG (i.e., MePEG and PEG4000) than high molecular weight PEG10,000 allowed valerolactone ring opening more efficient in terms of PVL/MePEG and PVL/PEG4000 copolymers possessing longer chain length in hydrophobic domain. Pegylated PVL with low CMC and triblock structure was preferred to encapsulate drug during micelle formation. Although all of these amphiphilic copolymers exhibited controlled release character, the micelles formed by triblock copolymer possessed a more stable core‐shell conformation than that by diblock copolymer, and resulted in the release of drug from triblock micelles slower than that from diblock micelles. © 2006 Wiley Periodicals, Inc. Novel amphiphilic star‐shaped terpolymers comprised of hydrophobic poly(?‐caprolactone), pH‐sensitive polyaminoester block and hydrophilic poly(ethylene glycol) (Mn = 1100, 2000 g mol?1) were synthesized using symmetric pentaerythritol as the core initiator for ring‐opening polymerization (ROP) reaction of ?‐caprolactone functionalized with amino ester dendrimer structure at all chain ends. Subsequently, a second ROP reaction was performed by means of four‐arm star‐shaped poly(?‐caprolactone) macromer with eight ‐OH end groups as the macro‐initiator followed by the attachment of a poly(ethylene glycol) block at the end of each chain via a macromolecular coupling reaction. The molecular structures were verified using Fourier transform infrared and 1H NMR spectroscopies and gel permeation chromatography. The terpolymers easily formed core–shell structural nanoparticles as micelles in aqueous solution which enhanced drug solubility. The hydrodynamic diameter of these agglomerates was found to be 91–104 nm, as measured using dynamic light scattering. The hydrophobic anticancer drug curcumin was loaded effectively into the polymeric micelles. The drug‐loaded nanoparticles were characterized for drug loading content, encapsulation efficiency, drug–polymer interaction and in vitro drug release profiles. Drug release studies showed an initial burst followed by a sustained release of the entrapped drug over a period of 7days at pH = 7.4 and 5.5. The release behaviours from the obtained drug‐loaded nanoparticles indicated that the rate of drug release could be effectively controlled by pH value. Altogether, these results demonstrate that the designed nanoparticles have great potential as hydrophobic drug delivery carriers for cancer therapy. © 2015 Society of Chemical Industry  相似文献

The fluorescent amphiphilic block copolymer methoxy poly(ethylene glycol) (mPEG)‐b‐poly(ethyl cyanoacrylate) (PECA)–2‐(N‐carbazolyl) ethyl methacrylate (CzEMA) was synthesized via living oxyanion‐initiated polymerization. mPEG‐b‐PECA–CzEMA was characterized by gel permeation chromatography, 1H‐NMR, and Fourier transform infrared spectroscopy. The results indicate that the polymerization was well controlled with a narrow molecular weight distribution. The mPEG‐b‐PECA–CzEMA nanoparticles prepared by nanoprecipitation techniques showed a narrow size distribution with an average diameter of less than 100 nm. The mPEG‐b‐PECA–CzEMA exhibited a strong carbazole fluorescence. Furthermore, it was found that the fluorescence intensity of mPEG‐b‐PECA–CzEMA was sensitive to a change in solvent. The results indicate that a subtle change in the state of the polymer micellar association may have altered the state of carbazole groups, which was responsible for the fluorescence emission. © 2011 Wiley Periodicals, Inc. Methoxy poly(ethylene glycol)‐b‐poly(L ‐lactic acid) (MPELLA) was prepared by the melt polycondensation of methoxy poly(ethylene glycol) and L ‐lactic acid. The structure and properties of MPELLA were characterized by IR, 1H‐NMR, differential scanning calorimetry, and wide‐angle X‐ray diffraction. To estimate its feasibility as a vehicle for paclitaxel, MPELLA nanoparticles were prepared by a self‐emulsification/solvent evaporation method. The paclitaxel‐loaded nanoparticles (PMTs) showed a spherical morphology with an inner core and an outer shell. The size, size distribution, and loading capacity of PMTs were also measured. The release kinetics of paclitaxel from PMTs in vitro was studied. The results show that paclitaxel can be effectively incorporated into MPELLA nanoparticles, which provide a delivery system for paclitaxel and other hydrophobic or toxic compounds. © 2005 Wiley Periodicals, Inc. A series of poly(?‐caprolactone)‐b‐poly(ethylene glycol) (PCL‐b‐PEG) block copolymers with different molecular weights were synthesized with a salicylaldimine‐aluminum complex in the presence of monomethoxy poly(ethylene glycol). The block copolymers were characterized by 1H NMR, GPC, WAXD, and DSC. The 1H NMR and GPC results verify the block structure and narrow molecular weight distribution of the block copolymers. WAXD and DSC results show that crystallization behavior of the block copolymers varies with the composition. When the PCL block is extremely short, only the PEG block is crystallizable. With further increase in the length of the PCL block, both blocks can crystallize. The PCL crystallizes prior to the PEG block and has a stronger suppression effect on crystallization of the PEG block, while the PEG block only exerts a relatively weak adverse effect on crystallization of the PCL block. © 2007 Wiley Periodicals, Inc. This work evaluates the transdermal drug delivery properties of amphiphilic copolymer self‐assembled nanoparticles by skin penetration experiments in vitro. Paclitaxel‐loaded methoxy poly(ethylene glycol)‐block‐poly(D ,L ‐lactic acid) diblock copolymer nanoparticles (PNPs) were prepared by a solid dispersion technique and were applied to the surface of excised full‐thickness rat skin in Franz diffusion cells. HPLC, transmission electron microscopy, Fourier transform infrared spectroscopy and 1H NMR were used to assay the receptor fluid. The results show that the amphiphilic copolymer nanoparticles with the entrapped paclitaxel are able to penetrate rat skin. Ethanol can improve the delivery of PNPs and increase the cumulative amount of paclitaxel in the receptor fluid by 3 times. Fluorescence microscopy measurements indicate that the PNPs can penetrate the skin not only via appendage routes including sweat ducts and hair follicles but also via epidermal routes. Poly(L ‐lactic acid) (PLLA), poly(ε‐caprolactone) (PCL), and their films without or blended with 50 wt% poly(ethylene glycol) (PEG) were prepared by solution casting. Porous films were obtained by water‐extraction of PEG from solution‐cast phase‐separated PLLA‐blend‐PCL‐blend‐PEG films. The effects of PLLA/PCL ratio on the morphology of the porous films and the effects of PLLA/PCL ratio and pores on the physical properties and biodegradability of the films were investigated. The pore size of the blend films decreased with increasing PLLA/PCL ratio. Polymer blending and pore formation gave biodegradable PLLA‐blend‐PCL materials with a wide variety of tensile properties with Young's modulus in the range of 0.07–1.4 GPa and elongation at break in the range 3–380%. Pore formation markedly increased the PLLA crystallinity of porous films, except for low PLLA/PCL ratio. Polymer blending as well as pore formation enhanced the enzymatic degradation of biodegradable polyester blends. Copyright © 2006 Society of Chemical Industry  相似文献

Diblock copolymers with different poly(ε‐caprolactone) (PCL) block lengths were synthesized by ring‐opening polymerization of ε‐caprolactone in the presence of monomethoxy poly(ethylene glycol) (mPEG‐OH, MW 2000) as initiator. The self‐aggregation behaviors and microscopic characteristics of the diblock copolymer self‐aggregates, prepared by the diafiltration method, were investigated by using 1H NMR, dynamic light scattering (DLS), and fluorescence spectroscopy. The PEG–PCL block copolymers formed the self‐aggregate in an aqueous environment by intra‐ and/or intermolecular association between hydrophobic PCL chains. The critical aggregation concentrations of the block copolymer self‐aggregate became lower with increasing hydrophobic PCL block length. On the other hand, reverse trends of mean hydrodynamic diameters were measured by DLS owing to the increasing bulkiness of the hydrophobic chains and hydrophobic interaction between the PCL microdomains. The partition equilibrium constants (Kv) of pyrene, measured by fluorescence spectroscopy, revealed that the inner core hydrophobicity of the nanoparticles increased with increasing PCL chain length. The aggregation number of PCL chain per one hydrophobic microdomain, investigated by the fluorescence quenching method using cetylpyridinium chloride as a quencher, revealed that 4–20 block copolymer chains were needed to form a hydrophobic microdomain, depending on PCL block length. © 2006 Wiley Periodicals, Inc. Poly(ethylene glycol)‐poly(L ‐lactide) diblock and triblock copolymers were prepared by ring‐opening polymerization of L ‐lactide with poly(ethylene glycol) methyl ether or with poly(ethylene glycol) in the presence of stannous octoate. Molecular weight, thermal properties, and crystalline structure of block copolymers were analyzed by 1H‐NMR, FTIR, GPC, DSC, and wide‐angle X‐ray diffraction (WAXD). The composition of the block copolymer was found to be comparable to those of the reactants. Each block of the PEG–PLLA copolymer was phase separated at room temperature, as determined by DSC and WAXD. For the asymmetric block copolymers, the crystallization of one block influenced much the crystalline structure of the other block that was chemically connected to it. Time‐resolved WAXD analyses also showed the crystallization of the PLLA block became retarded due to the presence of the PEG block. According to the biodegradability test using the activated sludge, PEG–PLLA block copolymer degraded much faster than PLLA homopolymers of the same molecular weight. © 1999 John Wiley amp; Sons, Inc. Atactic poly(3‐hydroxybutyrate) (a‐PHB) and block copolymers of poly(ethylene glycol) (PEG) with poly(ε‐caprolactone) (PCL‐b‐PEG) were synthesized through anionic polymerization and coordination polymerization, respectively. As demonstrated by differential scanning calorimetry (DSC) and dynamic mechanical thermal analysis (DMTA) measurements, both chemosynthesized a‐PHB and biosynthesized isotactic PHB (i‐PHB) are miscible with the PEG segment phase of PCL‐b‐PEGs. However, there is no evidence showing miscibility between both PHBs and the PCL segment phase of the copolymer even though PCL has been block‐copolymerized with PEG. Based on these results, PCL‐b‐PEG was added, as a compatibilizer, to both the PCL/a‐PHB blends and the PCL i‐PHB blends. The blend films were obtained through the evaporation of chloroform solutions of mixed components. Excitingly, the improvement in mechanical properties of PCL/PHB blends was achieved as anticipated initially upon the addition of PCL‐b‐PEG. © 2001 John Wiley & Sons, Inc. Grafting of poly(ε‐caprolactone) (PCL) and poly(lactide) (PLA) chains on poly(vinyl alcohol) backbone (PVA degree of hydrolysis 99%) was investigated using MgH2 environmental catalyst and melt‐grown ring‐opening polymerization (ROP) of ε‐caprolactone (CL) and L ‐lactide (LA), that avoiding undesirable toxic catalyst and solvent. The ability of MgH2 as catalyst as well as yield of reaction were discussed according to various PVA/CL/MgH2 and PVA/LA/MgH2 ratio. PVA‐g‐PCL and PVA‐g‐PLA were characterized by 1H‐ and 13C‐NMR, DSC, SEC, IR. For graft copolymers easily soluble in tetrahydrofuran (THF) or chloroform, wettability and surface energy of cast film varied in relation with the length and number of hydrophobic chains. Aqueous solution of micelle‐like particles was realized by dissolution in THF then addition of water. In this article, a new kind of biodegradable poly(ε‐caprolactone)‐poly(ethylene glycol)‐poly(ε‐caprolactone)‐based polyurethane (PCEC‐U) copolymers were successfully synthesized by melt‐polycondensation method from ε‐caprolactone (ε‐CL), poly(ethylene glycol) (PEG), 1,4‐butanediol (BD), and isophorone diisocyanate (IPDI). The obtained copolymers were characterized by 1H‐nuclear magnetic resonance (1H‐NMR), FTIR, and gel permeation chromatography (GPC). Thermal properties of PCEC‐U copolymers were studied by DSC and TGA/DTG under nitrogen atmosphere. Water absorption and hydrolytic degradation behavior of these copolymers were also investigated. Hydrolytic degradation behavior was studied by weight loss method. 1H‐NMR and GPC were also used to characterize the hydrolytic degradation behavior of PCEC‐U copolymers. The molecular weight of PCL block and PEG block in soft segment and the content of hard segment strongly affected the water absorption and hydrolytic degradation behavior of PCEC‐U copolymers. © 2009 Wiley Periodicals, Inc. Core–shell type nanoparticles of poly(L ‐lactide)/poly(ethylene glycol) (LE) diblock copolymer were prepared by a dialysis technique. Their size was confirmed as 40–70 nm using photon correlation spectroscopy. The 1H‐NMR analysis confirmed the formation of core–shell type nanoparticles and drug loading. The particle size, drug loading, and drug release rate of the LE nanoparticles were slightly changed by the initial solvents that were used. The drug release behavior of LE core–shell type nanoparticles showed an initial burst during the first 12 h and then a sustained release until 100 h. The degradation behavior of LE block copolymer nanoparticles was divided into three phases: the initial rapid degradation phase, the stationary phase, and the rapid degradation phase until complete degradation. It was suggested that lidocaine release kinetics were predominantly governed by the diffusion mechanism in the initial burst phase and after that by both of the diffusion and degradation mechanisms. © 2002 Wiley Periodicals, Inc. Poly(p‐dioxanone)–poly(ethylene glycol)–poly(p‐dioxanone) ABA triblock copolymers (PEDO) were synthesized by ring‐opening polymerization from p‐dioxanone using poly(ethylene glycol) (PEG) with different molecular weights as macroinitiators in N2 atmosphere. The copolymer was characterized by 1H NMR spectroscope. The thermal behavior, crystallization, and thermal stability of these copolymers were investigated by differential scanning calorimetry and thermogravimetric measurements. The water absorption of these copolymers was also measured. The results indicated that the content and length of PEG chain have a greater effect on the properties of copolymers. Ethylene‐vinyl acetate copolymer (EVA)/poly(?‐caprolactone) (PCL) blend (50/50 w/w) with co‐continuous morphology was prepared via melt mixing for fabricating microporous EVA membrane materials through selective solvent extraction. Shear flow and quiescent annealing techniques were employed to control co‐continuous phase size in the EVA/PCL blend, and the time‐ and temperature‐dependent relations of phase size were then evaluated theoretically. Using these techniques, microporous EVA membrane materials with various pore sizes ranging from 2 µm to more than 200 µm were obtained. In contrast to the porous EVA membrane prepared by the traditional way of solvent casting/particulate leaching, the as‐obtained microporous membrane shows a higher level of interconnectivity and much narrower pore size distribution with uniform pore structure. © 2013 Society of Chemical Industry  相似文献

Summary: Poly(ε‐caprolactone)‐polyglycolide‐poly(ethylene glycol) monomethyl ether random copolymers were synthesized from ε‐caprolactone (ε‐CL), glycolide (GA) and poly(ethylene glycol) monomethyl ether (MPEG) using stannous octoate as catalyst at 160 °C by bulk polymerization. The copolymers with different composition were synthesized by adjusting the weight ration of reaction mixture. The resultant copolymer with a weight ratio (10:15:75) of MPEG2000, GA, and CL was characterized by IR, 1H NMR, GPC and DSC. The new biodegradable copolymer has potential for medical applications since it is combined with properties of PCL, PGA and MPEG.

The aim of the study was to investigate the mechanical properties and biodegradability of poly(trimethylenecarbonate‐ε‐caprolactone)‐block‐poly(p‐dioxanone) [P(TMC‐ε‐CL)‐block‐PDO] in comparison with poly(p‐dioxanone) and poly(glycolide‐ε‐caprolactone) (Monocryl®) monofilaments in vivo and in vitro. P(TMC‐ε‐CL)‐block‐PDO copolymer and poly(p‐dioxanone) were prepared by using ring‐opening polymerization reaction. The monofilament fibers were obtained using conventional melt spun methods. The physicochemical and mechanical properties, such as viscosity, molecular weight, crystallinity, and knot security, were studied. Tensile strength, breaking strength retention, and surface morphology of P(TMC‐ε‐CL)‐block‐PDO, poly(p‐dioxanone), and Monocryl monofilament fibers were studied by immersion in phosphate‐buffered distilled water (pH 7.2) at 37°C and in vivo. The implantation studies of absorbable suture strands were performed in gluteal muscle of rats. The polymers, P(TMC‐ε‐CL)‐block‐PDO, poly(p‐dioxanone), and Monocryl, were semicrystalline and showed 27, 32, and 34% crystallinity, respectively. Those mechanical properties of P(TMC‐ε‐CL)‐block‐PDO were comparatively lower than other polymers. A block copolymer based on poly(γ‐benzyl‐L ‐glutamate) (PBLG) as the hydrophobic part and poly(ethylene oxide) (PEO) as the hydrophilic part was synthesized and characterized. PBLG/PEO/PBLG (GEG) block copolymer nanoparticles were prepared using the dialysis technique. Fluorescence spectroscopy measurement suggested that GEG block copolymers were associated in water to form polymeric micelles and the critical micelle concentration (CMC) value of the GEG‐50 block copolymer was 0.0084 g/L. Particle‐size distribution of the GEG‐50 block copolymer based on the number average was 34.9 ± 17.6 nm. Also, the particle size and drug‐loading contents of GEG nanoparticles were significantly changed with the initial solvent used. From transmission electron microscope (TEM) observations, the GEG polymeric micelle was a nice spherical shape and the sizes ranged from approximately 20–60 nm in diameter. Results from assessing the drug‐loading contents against the initial solvent showed that the use of tetrahydrofuran (THF) or 1,4‐dioxane as the initial solvent resulted in higher drug‐loading contents than those of other solvents. In the drug‐release studies, the higher the molecular weight of the polymer and drug‐loading contents, the slower the drug release. Also, the initial solvent used was significantly affected not only in the drug‐loading contents but also in the drug‐release kinetics. © 2000 John Wiley & Sons, Inc. The objective of this study was to prepare high molecular weight poly(ester‐anhydride)s by melt polycondensation. The polymerization procedure consisted of the preparation of carboxylic acid terminated poly(?‐caprolactone) prepolymers that were melt polymerized to poly(?‐caprolactone)s containing anhydride functions along the polymer backbone. Poly(?‐caprolactone) prepolymers were prepared using either 1,4‐butanediol or 4‐(hydroxymethyl)benzoic acid as initiators, yielding hydroxyl‐terminated intermediates that were then converted to carboxylic acid‐terminated prepolymers by reaction with succinic anhydride. Prepolymers were then allowed to react with an excess of acetic anhydride, followed by subsequent polycondensation to resulting high molecular weight poly(ester‐anhydride)s. Upon coupling of prepolymers, size exclusion chromatography analyses showed an increase from 3600 to 70,000 g/mol in number‐average molecular weight (Mn) for the 1,4‐butanediol initiated polymer, and an increase from 7200 to 68,000 g/mol for the 4‐(hydroxymethyl)benzoic acid‐initiated polymer. 4‐Hydroxybenzoic acid and adipic acid were also used as initiators in the preparation of poly(?‐caprolactone) prepolymers.
