The agouti (Dasyprocta prymnolopha, Wagler 1831) is a wild rodent of great zootechnical potential, a fact that enables anatomical and morphological studies to support management actions with this animal. In this perspective, this study aimed to describe the anatomy and histology of the agouti stifle joint. Four adult agoutis were used, two females and two males. The animals were submitted to dissection and identification of the structures of the stifle joint. For light microscopy study, samples of the patellar ligament, cranial and caudal cruciate ligaments, medial and lateral collateral ligaments were used. Agouti has a highly congruent patellofemoral joint; elongated patella; medial and lateral fabellae at the proximal insertion of the gastrocnemius muscle; medial and lateral meniscus with lunula; in addition to the presence of the following ligament structures: patellar ligament, cranial and caudal cruciate ligaments, medial and lateral collateral ligaments, meniscofemoral ligament, caudal meniscal ligament of the medial meniscus, and medial and lateral cranial ligaments. The patellar ligament presents bundles of parallel collagen fibers with a straight path and coated fibroblasts; collateral and cruciate ligaments had loose and dense connective tissue, coated fibroblasts and collagen bundle undulations, the latter most expressive in the caudal cruciate ligament. Thus, except for the shape and angulation of the stifle, which allows specific movements, the agouti stifle has structures analogous to that of other rodents and domestic animals.  相似文献

Stem cells are present in the adult tissues of most diverse species. Bone marrow is recognized to be the most exploited site to obtain stem cells and cell progenitors. The objective of the present study was to characterize hematopoietic progenitor (HP) morphology and analyze the performance of adherent cell progenitors (ACPs) cultivated in vitro from black‐rumped agouti bone marrow (Dasyprocta prymnolopha). Bone marrow aspirates were obtained from tibia crest and used to prepare histological slides and identify cell morphology. Cells were also scattered on culture plates for later isolation, expansion, and quantification. Smears obtained from bone marrow demonstrated HPs at different stages of maturity. In culture, these cells showed fibroblastoid morphology and a strong tendency to form colonies, demonstrated by the presence of cell aggregates, cytoplasmic elongations lying side by side. An 80% cell confluence was observed at 18 days in culture and progressive reduction in the percentage of nonadherent mononuclear cells. After eight passes, a mean cell viability of 96.07% was observed, from a pool of 1.6 × 107 cells (ACP). Thirteen 25‐cm2 culture bottles were trypsinized, resuspended in freezing medium, stored in 14 criotubes at a concentration of 1 × 106 cells per milliliter, and placed in liquid nitrogen at ?196°C. Agouti bone marrow demonstrated high plasticity, moreover different HP lines, and a population of adherent cells demonstrated morphology similar to mesenchymal stem cells in culture. Microsc. Res. Thirty adult agoutis (Dasyprocta primnolopha) from the Nucleus of Study and Preservation of Wild Animals at the Federal University of Piauí were used. Blood scrubs of these animals were colored by the Leishman method and analyzed in light microscopy. The cells had been measured using programs that analyze images (Leica QWin - Image Processing and Analysis Software). Mature erythrocytes, basophil reticulocytes, lymphocytes, eosinophils, neutrophils, monocytes, and thrombocytes were identified. Agoutis' erythrocytes presented elliptical form, without nucleus with an average diameter of 5.64 micromeres ± 0.38. The lymphocytes are spherical cells with scarce cytoplasm, dense and with a very centralized rounded nucleus measuring an average diameter of 13.20 micromeres ± 0.35. The monocytes are slightly basophilic, with a spherical nucleus, central constriction, and an average diameter of 20.59 micromeres ± 0.32. The neutrophils are spherical, with a polymorphic lobulated nucleus, with an average diameter of 11.2 micromeres ± 0.20. The eosinophils are spherical with lobulated nucleus and with an average diameter of 14.25 micromeres ± 0.36. Only five basophils were observed, with abundance of cytoplasmic granules with 9.8 micrometers of diameter ± 0.30. Thrombocytopenic pleomorphism was frequent. There were similarities in the cellular constituents in peripheral blood of agoutis and of other rodents and humans. The cellular types from the peripheral blood, the morphology, and morphometry of the blood's cells did not vary according to sex.  相似文献

The zebrafish pineal gland plays a fundamental role in the regulation of the circadian rhythm through the melatonin secretion. The pinealocytes, also called photoreceptive cells, are considered the morphofunctional unit of pineal gland. In literature, the anatomical features, the cellular characteristics, and the pinealocytes morphology of zebrafish pineal gland have not been previously described in detail. Therefore, this study was undertaken to analyze the structure and ultrastructure, as well as the immunohistochemical profile of the zebrafish pineal gland with particular reference to the pinealocytes. Here, we demonstrated, using RT-PCR, immunohistochemistry and transmission electron microscopy, the expression of the mRNA for rhodopsin in the pineal gland of zebrafish, as well as its cellular localization exclusively in the pinealocytes of adult zebrafish. Moreover, the ultrastructural observations demonstrated that the pinealocytes were constituted by an outer segment with numerous lamellar membranes, an inner segment with many mitochondria, and a basal pole with the synapses. Our results taken together demonstrated a central role of zebrafish pinealocytes in the control of pineal gland functions.  相似文献

Armadillos, Xenarthras representatives, known for adaptability to different ecosystems, own specific morphophysiological characteristics that are not known and deserve to be studied. The aim of this study was to describe the morphology of cartilage of the larynx of the nine‐banded armadillo (Dasypus novemcinctus). Five dead armadillos were donated by the Chico Mendes Institute of Biodiversity (ICMBio‐PI) to the Federal University of Piauí. The animals were fixed and dissected for removal of the larynx. The cartilages were identified and described, photodocumented, and schematized. Fragments with about 0.5 cm of each cartilage were collected and submitted to classical histology for Hematoxylin–Eosin coloring. The slides were assembled in enterlan and analyzed under a light microscope. The larynx of the armadillo (D. novemcinctus) is located in the mentonian region, ventral to the esophagus, and due to the total positioning of the tongue in the oral cavity, there is also a cranial cervical position in this species. The larynx has five cartilages, they are: a cricoid, a thyroid, an epiglottis, and two arytenoids. The corniculate process is present; however, the cuneiform process is absent. The epiglottis has a discrete bifurcation at its apex. In all cartilages epithelial variations are observed. The tissues are varied from squamoso stratified to cylindrical pseudostratified, with propria lamina rich in mucoserosas glands. With the exception of epiglottic cartilage, predominantly elastic, the rest are hyaline. The larynx of D. novemcinctus, although the same number of cartilages, differs morphologically and microscopically from the larynx of other species.  相似文献

This research was aimed to present the histological and ultrastructure properties of the adrenal gland in the Persian squirrel. Two male and female animals were included in the study. The adrenal gland was bean-shaped and located on the cranial pole of kidney. The enveloping capsule was dense connective tissue that reacted positively with Periodic-Acid Schiff (PAS) and Masson trichrome stainings. The parenchyma of the gland consisted of two-part, namely cortex and medulla; the cortex had three layers: zona glomerulosa (ZG), zona fasciculata (ZF), and zona reticularis (ZR). The cells of the ZG were mainly spherical and ovoid with circular arrangement and few lipid droplets in TEM micrographs. The cells of the ZF were columnar and spherical that were arranged in cord-like rows. Transmission electron microscopy (TEM) indicated conspicuous lipid droplets and mitochondria in this zone. The cells of the ZR were arranged in a tangled networks and were almost similar to those in the ZF. TEM images showed fewer lipid vesicles in the ZR compared to the ZF and ZG. Chromaffin cells were located in the medulla of the adrenal gland in two layers. TEM images showed that some of them were smaller and contained fewer secretory granules; other cells were larger and contained more electron-dense secretory granules. Immunofluorescence staining showed that steroidogenic factor 1 (SF1) expressed from cortex to the corticomedullary junction (CMJ) and tyrosine hydroxylase (TH) expressed in the medulla. In conclusion, the results indicated both similarities and differences between the adrenal gland of the Persian squirrel and other animals such as mammals and rodents.  相似文献

All termite species (Isoptera) are eusocial. These insects live in societies where the division of labor is a predominant characteristic, conditioned by the presence of castes with different morphology, ontogeny, and development. The soldier caste is unique among social insects and it is responsible for colony defense. Soldiers belonging to the Nasutitermitinae subfamily are very peculiar, since they may be polymorphic and present a nasus in addition to either developed or vestigial mandibles. The defensive secretions of soldiers of the neotropical Nasutitermitinae have been the aim of several chemical studies, but few data exist concerning the anatomy and histology of the exocrine glands. This article presents a comparative study on the anatomy of the frontal gland of soldiers of several Nasutitermitinae species: Syntermes dirus (Burmeister), Syntermes nanus (Constantino), Constrictotermes cyphergaster (Silvestri), Nasutitermes corniger (Motschulsky) and Velocitermes heteropterus (Silvestri), with emphasis on the ultramorphology and ultrastructure of the frontal tube.  相似文献

A (001) surface of molybdenum, covered by one monolayer of sulphur was prepared in UHV and characterized by LEED, Auger and XPS. This surface was found to be stable in air for periods of several days. STM images of the surface, obtained in air in the topographic and local barrier height modes, revealed the atomic arrangement of sulphur atoms in domains with 1×2 and 2×1 periodicities. Boundaries between domains, adsorbate and substrate defect structures and crystallites formed during the initial oxidation of the Mo substrate were observed.  相似文献

Computer tomography has been used frequently for the 3‐D visualization of plant anatomical traits but sample preparation has been widely neglected. Without any preparation smaller (i.e., up to 1 × 1 cm2) turgescent or semi‐dry plant samples (especially leaf samples) diminish the image quality of a scan due to gradual water loss and therefore constant movement. A suitable preparation for scans of turgescent and semi‐dry plant samples with a high resolution μCT (<1–5 μm) has to be very thin, heat‐resistant (up to 35°C), have a low attenuation coefficient, and should not alter the water content and structure of the sample. Several agents have been tested, but only a coating with vaseline conserved the water content of a plant sample efficiently. However, water molecules and vaseline both attenuate the X‐ray beam, which decreases the image quality of scans of turgescent or semi‐dry plant samples. Therefore, trade‐offs between the spatial resolution, sample water content, sample size, and image quality have to be considered: larger samples have to be placed further away from the X‐ray tube, which leads to a lower spatial resolution; water and preparation agents attenuate the X‐ray beam, causing low‐quality images which may be accompanied by motion artifacts compared to a scan of a dry sample, where no preparation is necessary. The detailed understanding of the physical and reaction chemistry of engineering lubricants is key to new developments in the future. Here, we draw together the main chemical/engineering literature in the first systematic review of the standard anti‐wear additive used as an aviation lubricant, tricresyl phosphate (TCP), focusing on understanding the links between the surface chemistry, tribology and decomposition of TCP. While there is still debate concerning the mechanism of TCP, it is clear that it is activated by moisture, oxygen or oxidised metal surfaces. Its anti‐wear properties arise from the resulting formation of iron phosphate or polyphosphate on the contact surfaces. Evidence stresses the importance of chemistry at the boundary layer and a mechanism involving pre‐coordination of TCP at the surface, resulting in activation at the P‐centre and subsequent attack by residual H2O or surface O2−. This perspective provides a potential baseline for the development of future phosphorus‐based high‐performance additives. Copyright © 2015 John Wiley & Sons, Ltd.  相似文献

This article presents the results of analyses carried out on a lot of 11 fragments of Bronze Age ceramics discovered in the site from B?ile Figa (Beclean town, Transylvania, Romania), where salt exploitation occurred. The samples were analyzed by means of optical microscopy, on the basis of which the morphology and distribution of the inclusions in the ceramic paste was established. Likewise, the samples were analyzed by scanning electron microscopy with energy‐dispersive X‐ray spectrometry, producing microphotographs for each sample, and on the basis of the X‐ray spectra, the elemental composition in gravimetric percentages was established. Through the micro‐Fourier‐transform infrared spectroscopy analysis, on the basis of characteristic group vibrations, it was possible to ascertain the nature of the compounds from the ceramic samples. These analyses will contribute to the reconstruction of the prehistoric technologies for salt exploitation.  相似文献

Thirty white‐spotted geckos, Tarentola annularis, from the South Sinai desert in Egypt, were examined for helminth parasites. Spauligodon aspiculus was observed to infect 19 geckos with 63.33% as a prevalence of parasitic infection. The present nematode species is separated from congeners by morphological and metrical characteristics such as lateral alae, aspinose filamentous tail, and no spicule, and three pairs of caudal papillae with posterior pair excluded from envelopment by the caudal alae in the male worms, and knobbed eggs, and postbulbar vulva in females. It compared morphometrically with other Spauligodon species described previously and showed few differences in measurements. Molecular characterization based on the partial 28S rRNA nuclear ribosomal gene sequence showed that there was a close identity, up to 72%, with other sequences retrieved from GenBank. Phylogenetic analysis showed that the parasite sequence in conjunction with existing data facilitates the investigation of the placement of this pharyngodonid species within Oxyuridae. The present species is deeply embedded in the genus Spauligodon with close relationships to previously described Spauligodon nicolauensis (gb| JN619349.1, and JF829243.1) as more related sister taxa. This study highlights the importance of combining genetic and morphological data with taxonomy in pharyngodonid species.  相似文献

Ticks of Rhipicephalus sanguineus species have great medical and veterinary importance for being a vector of various diseases. In an attempt to minimize their action on the host, people have resorted to chemical control by using various acaricides, such as selamectin. Although previous studies have demonstrated its toxic action in domestic animals, no studies focused on the detection of cell death when exposed to selamectin. For this reason, the technique for detecting autophagic cell death was used in order to demonstrate the responses of rabbits' skin tissues pre‐infested with R. sanguineus and exposed to different concentrations of selamectin. The obtained results when exposed to 100 and 80% concentrations of selamectin showed a strong mark of acid phosphatase on the cells of the connective tissue of the dermis and hair follicles, whereas the ones exposed to the 50% concentration had a weak mark on the cells of the connective tissue of the dermis and moderate staining in hair follicles. It became clear that, when used at high concentrations (100 and 80%), selamectin is capable to induce a large scale occurrence of the autophagic cell death process. On the other hand, the concentration of 50% causes minor morphophysiological changes in the skin of rabbit hosts when evaluated the cell death process. Therefore, the data confirms that selamectin is a powerful dose‐dependent toxic agent causes increased activity of the enzyme acid phosphatase. Microsc. Res.
