The total content of free, esterified and bound phenolics of the peel and flesh of four potato varieties (Purple, Innovator, Russet and Yellow) were determined using the Folin–Ciocalteu method. The respective antioxidant activities of these potatoes and/or their skins were evaluated using several in vitro assays. The phenolic profiles of potato peel and flesh samples were determined using high-performance liquid chromatography with photodiode array detection (HPLC-DAD). Bound and esterified phenolics contributed as much or even more than the free phenolics to the antioxidant activity of the peels; extracts from Purple variety showing the highest activity. The peels of all varieties showed significantly (p < 0.05) higher phenolic content and antioxidant activities than their respective flesh. Chromatographic data showed differences in the amounts, but not in types, of phenolic compounds in the potato peel samples. Thus, potato processing discard may be used in food formulations and their extracts could potentially be employed as an effective source of antioxidants in food systems.  相似文献

Phenolic compounds, antioxidant and antibacterial activities of defatted Moringa oleifera seed flour (DMF) were investigated. The total phenolic and flavonoid contents were measured using colorimetric methods. Free phenolic acid and flavonoid profiles were analyzed by high performance liquid chromatography, while antioxidant capacities were evaluated using scavenging assays of 1,1-diphenyl-2-picrylhydrazyl radical (DPPH), ferric reducing antioxidant power and total antioxidant capacity. The results showed that extractability of phenolic compounds was significantly higher (p < 0.05) in bound phenolic extract (4173.00 ± 32.22 mg gallic acid equivalents (GAE)/100 g) than in free phenolic extract (780.00 ± 14.2 mg GAE/100 g) and it showed higher antioxidant and antimicrobial activities. The IC50 value for DPPH radical scavenging activity was 0.9 ± 0.05 and 14.9 ± 0.07 mg/mL for bound phenolic and free phenolic extracts, respectively. Bound phenolic extract was more effective (minimum inhibitory concentration (MIC), 0.06–0.157%) than free phenolic extract (MIC, 0.117–0.191%) against tested bacteria. Ten phenolic compounds (gallic acid, p-coumaric acid, ferulic acid, caffeic acid, protocatechuic acid, cinnamic acid, catechin, epicatechin, vanillin and quercetin) were identified and quantified in both extracts. These natural plant phenolics from Moringa seeds could be a good source of antioxidants and antibacterials for food and pharmaceutical industries.  相似文献

Five commonly consumed vegetables in Malaysia namely, four-angled bean (Psophocarpus tetragonolobus D.C.), French bean (Phaseolus vulgaris L.), long bean (Vigna sesquipedalis L.), snow pea (Pisum sativum var. macrocarpon L.) and snap pea (Pisum sativum) were blanched in boiling water for 10 min. The contents of total phenolics, ascorbic acid and β-carotene, and the antioxidant capacity as typified by β-carotene and free radical scavenging activity (DPPH) assays were determined for the raw and blanched vegetables. The study revealed that blanching caused a significant (p < 0.05) increase in β-carotene content [fresh (389–539 µg/100 g), blanched (510–818 µg/100 g)], except in snow pea. Conversely, there was a significant (p < 0.05) decrease in ascorbic acid content [fresh (1.2–7.8 mg/100 g), blanched (0.67–3.8 mg/100 g)]. After blanching, the total phenolic content and antioxidant activity either decreased or increased depending on the type of vegetables. The total phenolic content was positively correlated with the antioxidant activity of the studied vegetables to some extent, but not with ascorbic acid or β-carotene.Industrial relevanceBeans (four-angled bean, French bean and long bean), and peas (snow and snap pea) are commonly consumed vegetables in Malaysia. However, the scientific information regarding blanching and its effect on the total phenolic content and antioxidant activities is very limited. This work showed that blanching caused an increase in the carotene, total phenolic content and antioxidant activities in some of the selected vegetables. This work is important since, consumption of raw vegetables does not have high nutritional qualities and antioxidant activities as shown in this work.  相似文献

The ultrasound-assisted enzyme hydrolysis (UAEH) procedure for extraction of phenolics from olive waste (OW) was established. The optimized conditions obtained with response surface methodology (RSM) were pH 5.75, treatment 40 min and temperature 55 °C, the yield obtained reached to 4.0%. The extraction kinetics model was fitted well to the ln (Ct/C0) = kt, and the rate constants (k) increased with the increased in the extraction temperature. Moreover, the values of k were higher and the activation energy was lower than ultrasound treatment alone. The thermodynamic parameters indicated the extraction process was easier under the UAEH. Compared with the petroleum ether and n-butyl alcohol, ethyl acetate extract fraction had high phenolic content (64.4 mg/g), especially hydroxytyrosol and tyrosol (17.2 mg/g and 14.1 mg/g), and exhibited excellent antioxidant activities in DPPH·, ABTS+·and FRAP assay. These finding revealed that OW could be used for a enrich source of natural phenolic compounds for development of food industry.Industrial relevanceOlive waste (OW) has been widely paid attention as sources of high-added value compounds, such as phenolic which can be obtained by the several methods. And phenolic extract as a food additive can not only improve the sensory quality of food, but also can enhance antioxidant capacity in fatty food matrices. Moreover, the use of this extract has better economic benefits than synthetic additives usually used in food industry. In this study, a novel green, simple and efficient technology was employed to extract the phenolic compounds, which was suitable for large-scale industrial use. The results indicated that ultrasound-assisted enzyme hydrolysis (UAEH) has significant potential in the extraction process of phenolics from OW exhibiting higher phenolic yield in shorter extraction times.  相似文献

The methanolic extract of Canavalia ensiformis (L.) DC (Jack bean) seed, an underutilized food legume collected from India was analyzed for antioxidant and health relevant functional properties. The raw seeds contained total free phenolic content of 12.98 ± 1.63 g catechin equivalent/100 g extract DM. The raw seed extract exhibited ferric reducing/antioxidant power (FRAP, 1218 mmol Fe[II]/mg extract), inhibition of β-carotene degradation (49.81%), radical scavenging activity against DPPH (56.78%) and superoxide (35.89%). In addition, 77.56% of α-amylase and 75.45% of α-glucosidase enzyme inhibition characteristics were found under in vitro starch digestion bioassay. Sprouting + oil-frying caused an apparent increase on the total free phenolic content with significant improvement on antioxidant and free-radical scavenging capacity, while soaking + cooking as well as open-pan roasting treatments showed diminishing effects. Inhibition of α-amylase and α-glucosidase enzyme activities were declined to 22.69 and 42.69%, respectively during sprouting + oil-frying treatment is more desirable for the dietary management of type II diabetic patients.  相似文献

Mushrooms have become attractive as functional foods and as a source of physiologically beneficial bioactive compounds. Herein, we describe and compare the chemical constituents (phenolic compounds, macronutrients, sugars, fatty acids, tocopherols and ascorbic acid) of four wild edible mushrooms widely appreciated in gastronomy: Armillaria mellea (Vahl) P. Kumm., Calocybe gambosa (Fr.) Donk, Clitocybe odora (Fr.) P. Kumm., Coprinus comatus (O.F. Müll.) Pers. Furthermore, the antioxidant activity of their water soluble polysaccharidic and ethanolic fractions was studied by three different in vitro assays. C. comatus revealed the highest concentrations of sugars (43.23/100 g dry weight), PUFA (77.46%), phenolic compounds (45.02 mg/kg), tocopherols (301.03 μg/100 g) and, among all of the fractions tested, its ethanolic fraction showed the highest antioxidant activity (EC50 < 2.6 mg/ml). C. odora revealed one of the highest ascorbic acid (172.65 mg/100 g) contents and its water soluble polysaccharidic fraction showed the best antioxidant properties (EC50 < 3.6 mg/ml) among the polysaccharidic fractions. The studied mushrooms species could potentially be used in well-balanced diets and as a source of bioactive compounds.  相似文献

The aim of this study was to determine the phenolic profile of seven different varieties of ripe date palm fruit (Phoenix dactylifera) from Algeria by LC–DAD–MS (ESI+), to investigate their respective antioxidant activities by the DPPH· method and to estimate their phenolic content using the Folin–Ciocalteu method. The total phenolic content was in the range of 2.49 ± 0.01 to 8.36 ± 0.60 mg gallic acid equivalents (GAE) per 100 g fresh fruit. This fruit was shown to possess an antioxidant activity, giving values of antiradical efficient (AE) from 0.08 ± 0.00 to 0.22 ± 0.00. The phenolic contents and the antiradical efficiencies of the different varieties were highly correlated (R2=0.975). All the varieties were found to contain mainly p-coumaric, ferulic and sinapic acids and some cinnamic acid derivatives. Three different isomers of 5-o-caffeoylshikimic acid were detected. Different types of flavonoids were identified, mainly flavones, flavanones and flavonol glycosides.  相似文献

Copao (Eulychnia acida Phil., Cactaceae) is an endemic species occurring in arid areas of northern Chile. The fruits are commercialized by peasants within the Elqui and Limari valleys and are appreciated for its acidic and refreshing taste. We now report the total phenolic (TP) and total flavonoid (TF) content, antioxidant activity, phenolic composition and main phenolic distribution in pulp and epicarp of copao fruits from different harvesting places from both valleys. The ascorbic acid content was determined in fresh fruit pulp, epicarp and juice. The phenolic-enriched extract was analyzed for antioxidant effect and composition. Ferulic acid, 9,10-dihydroxy-4,7-megastigmadien-3-one hexoside, isorhamnetin and quercetin glycosides were identified by HPLC–DAD–MS/MS analysis. The main compounds were isolated and fully characterized by NMR techniques. The main phenolic in the samples was isorhamnetin-3-O-[α-rhamnopyranosyl-(1  6)-β-glucopyranoside]. The HPLC pattern of the phenolic-enriched extracts of the fruits allows a differentiation of samples from the Elqui and Limari valleys. All fruit extracts and Amberlite-retained fraction from the methanolic extract were devoid of toxicity against human gastric AGS cells and human lung fibroblasts, with IC50 values > 400 μg/mL for AGS and 344 to > 400 μg/mL for fibroblasts, respectively. The compound identification, associated with the antioxidant activity and insignificant cell toxicity, adds relevant information for the possible development of this native fruit into a new crop.  相似文献

Ready-to-eat shredded orange and purple carrots, packed in air (control), or in modified atmosphere packaging [MAP; (90%N2 + 5%O2 + 5%CO2 and 95%O2 + 5%CO2)], and stored chilled for up to 13 days, were examined for their antioxidant activity and contents of anthocyanins, carotenoids and phenolics, as well as sensory quality. Total antioxidant activity, carotenoids and phenolics of purple carrots were initially 2.8-, 2.3- and 2.9-fold higher than orange carrots, respectively. Total antioxidant activity remained relatively constant in orange carrots during storage under all treatment conditions, whereas a highly significant decrease (P<0.01) in (95%O2 + 5%CO2)-treated purple carrots was observed. The content of anthocyanin, only found in purple carrots, was decreased slightly during the storage period, and this was particularly significant in the 95%O2 + 5%CO2 treatment. In both orange and purple carrots, loss of total carotenoids occurred in the 95%O2 + 5%CO2 treatment. Total phenolic content of purple carrots increased at a much higher rate during storage than orange carrots. The MAP treatment (90%N2 + 5%O2 + 5%CO2) gave better sensory quality and extended shelf-life for purple carrots (∼2–3 days longer shelf-life than other treatments,) but, no difference was observed for orange carrots. Shredded purple carrot can be stored under 90%N2 + 5%CO2 + 5%O2 treatment for up to 10 days and high nitrogen treatment may be used in maintaining the storage quality of shredded purple carrots. Thus, purple carrots may be used in place of orange carrots to take advantage of their nutraceutical components.  相似文献

The fruits of Pilosocereus arrabidae (Cactaceae) is found in the Grumari shoal, located in the Rio de Janeiro state, Brazil and, several studies have already highlighted the importance of consuming its fruits. This work aims to investigate the physical, mineral and physicochemical properties of its fruits as well as to establish the knowledge about their chemical constituents and antioxidant properties. The peel (Pe) and pulp (Pu) extracts were obtained by maceration with the following solvents: hexane (HX), dichloromethane (DCL), ethyl acetate (EAC) and ethanol 70% (ET). The extracts were analyzed by Gas Chromatography coupled to Mass Spectrometry (GC–MS) and, by High Performance Liquid Chromatography (HPLC–DAD and HPLC–MS) for its chemical investigation. For the antioxidant activity investigations the ORAC, DPPH and ferrous ion chelating capacity (FICC) tests were performed. As results, we found higher yields for peels (72%) compared to pulps (28%). By the physical–chemical analyses we point out the fruits as a good source of fiber (pulp: 6.01 mg/100 g; peel: 8.02 mg/100 g). The minerals were analyzed by the method of issuing flames and indicated high levels of selenium (DRI for pulp and peel: 147%) and manganese (DRI pulp: 97.69% and DRI peel: 269.56%). The total flavonoid contents of the fruits performed by HPLC–DAD presented 0.45 μg equiv. in quercetin/mL of peel EAC extract and 0.25 μg equiv. in rutin/mL of pulp EAC extract. The antioxidant activities by the ORAC, FICC and DPPH methods indicated that the ET extracts showed antioxidant activities above the standards adopted for the tests. Among these, we highlight the ET extract of the pulp with EC50 of 17.57 ± 0.27 μg/mL, lower than Ginkgo biloba EGB761® (23.40 ± 0.04 mg/mL). By the FICC test the EAC extract of the peel and pulp showed 70.0% and 53.4% activity, respectively, at 500 mg/mL, higher than the standard quercetin (50.0%). By the HPLC–DAD and HPLC–MS methods there were detected, for the first time on this species, the presence of the following flavonoids on the EAC extracts: quercetin, rutin, catechin, dihydrokaempferol, quercetin 3 or 4′-O-glucoside, kaempferol and isorhamnetin. By the GC–MS analysis there were detected on the DCL extracts saturated fatty acids (palmitic and stearic) and, on the HX extracts, methylated sugars (peel) and menthol (pulp). To sum up, the fruits of P. arrabidae display antioxidant potential correlated to flavonoid presence, and, high levels of selenium, manganese and fibers, characteristics that can promote beneficial effects on human health.  相似文献

Citharexylum solanaceum is a native fruit from Brazil, which both bioactive compounds and antioxidant potential were not yet investigated. Thus, the freeze-dried extracts of seed and pulp + skin of C. solanaceum fruits were obtained after solid-liquid extraction with ethanol and their bioactive compounds composition, namely phenolic compounds and carotenoids, were determined. The antioxidant capacity of both extracts against physiologically relevant reactive oxygen species (ROS) and reactive nitrogen species (RNS) was further investigated. Both C. solanaceum extracts showed high contents of phenolic compounds; however, pulp + skin extract presented 2.4-times more phenolic compounds (33.54 mg/g) than the seed extract (14.09 mg/g). Verbascoside (phenylpropanoid) was the major compound identified in both extracts (11–25 mg/g). Regarding the carotenoid composition, all-trans-lutein (14–42 μg/g) and all-trans-β-carotene (13–44 μg/g) were the major compounds in both extracts. The high content of phenolic compounds and carotenoids in pulp + skin extract might explain its higher scavenging capacity against all the ROS/RNS as compared to seed extract. In general, both extracts showed better scavenging capacity for the RNS than for the ROS. Our results indicate that C. solanaceum fruits can be explored as an important natural source of antioxidant compounds.  相似文献

A total of 51 Brazilian teas from the species Camellia sinensis, Peumus boldus, Matricaria recutita, Baccharis trimera, Cymbopogon citratus, Pimpinella anisum, Mentha piperita, and Ilex paraguariensis were analyzed in terms of phenolic compounds, color and in vitro antioxidant capacity using ferric reducing antioxidant power (FRAP) and 1,1-diphenyl-2-picrylhydrazyl (DPPH) assays. Data were processed using principal component analysis (PCA), hierarchical cluster analysis (HCA) and linear discriminant analysis (LDA). Significant (P < 0.01) correlations between antioxidant activity measured by DPPH and FRAP assays with the total phenolic compounds (r = 0.87; r = 0.90, respectively) and flavonoids (r = 0.79; r = 0.77, respectively) were attained. The compounds that displayed significant (P < 0.05) correlations with the antioxidant activity were gallic acid, catechin, epicatechin, procyanidin B2, quercetrin, and caffeine. PCA was a suitable approach to check for similarities among tea samples, explaining up to 50% of data variability. Four groups were suggested using HCA, in which cluster 3 showed the highest content of total phenolic compounds, flavonoids, antioxidant activity, gallic acid, and caffeine. All samples included in this group were from C. sinensis. The overall classification capacity obtained by LDA was 82.00%, in which 100% of samples from I. paraguariensis, C. citratus, M. recutita, and P. boldus were adroitly classified, while 60% of teas from P. anisum, 80% of M. piperita teas, and 88.24% of C. sinensis teas were correctly classified.  相似文献

Banana peel is an underused by-product that can be processed to obtain flour that is more easily stored for further uses. The extracts of banana peel flour exhibited a high total phenolic content (around 29 mg/g, as GAE) due to the occurrence of important amounts of flavonoid phenolics: highly polymerized prodelphinidins (around 3952 mg/kg), followed by decreasing lower contents of flavonol glycosides (mainly 3-rutinosides and predominantly quercetin-based structures, accounting for around 129 mg/kg), B-type procyanidin dimers and monomeric flavan-3-ols (jointly around 126 mg/kg). The high total phenolic content of extracts of banana peel flour is likely responsible for the very high antioxidant activity (μM/g, as Trolox equivalents) measured by three different methods: FRAP, around 14 μM/g; ABTS, around 242 μM/g; and ORAC, around 436 μM/g. All these results suggest the interest in going in depth of the good use of banana peel as a profitable source of bioactive phenolic compounds.  相似文献

The hepatoprotective effect of water extracts of sweet orange (Citrus sinensis) peel (WESP) and its biological compound, hesperidin (HD), on oxidative stress in vivo, were investigated. HD was the major compounds among the ten compounds identified using HPLC-DAD and HPLC-MS/MS analysis. Oral administration of WESP to rats at 10 and 100 mg/kg bw for 28 consecutive days before a single dose of CCl4 (2 ml/kg bw) demonstrates a significant protective effect by lowering the levels of aspartate aminotransferase (AST) and alanine aminotransferase (ALT), and by improving the histological architecture of the rat liver. WESP attenuated oxidative stress by increasing the content of hepatic glutathione (GSH), and by a dramatic increase in the activities of superoxide dismutase (SOD) and glutathione peroxidase (GPx). WESP induced a significant CYP2E1 activity, which suggests that WESP may be a substrate of CYP2E1. WESP at a dose of 1.0 mg/kg bw and HD at 0.1 mg/kg bw did not sustain the protective effect against oxidative stress, in vivo. This study demonstrated that citrus peel protects rat liver from CCl4-induced injury by attenuating hepatic oxidative stress, which suggests that WESP can be used as a therapeutic antihepatotoxic agent for the treatment of hepatic injury.  相似文献

Phenolic constituents (total phenols, flavonoids, tannins, and anthocyanins), comparative antiradical potency and cytotoxicity of processed mango (Mangifera indica) kernel (PMK), processed bush mango (Irvingia gabonensis) kernel (PBMK) and their mixture (MKK) at 50:50 were evaluated. Antiradical assay of the samples was conducted using three different methodologies (lipid peroxidation, nitric oxide and ferric reducing anti-oxidant power (FRAP)). The three samples contained negligible amounts of anthocyanins (< 0.67 ng/g) compared with the other constituents (1.4 to 2.2 mg/g), largely of gallotannins and flavonoids. However, PBMK had the least flavonoid content, averaging 43%. Due to relative sensitivity of assay techniques to phenolic composition of sample extracts, results from FRAP assay favourably complemented those of another technique to give better reflection of sample’s radical scavenging capacity. Mean inhibitory concentration (IC50) values obtained, showed that the three samples had higher antioxidant capacity than reference quercetin. Similarly, the mean lethal concentration (LC50) values obtained from cytotoxicity assay indicated that the phenolic acid and flavonoid content of the three processed kernel samples were more tolerable physiologically compared with reference dichromate. Both observations were statistically significant (p < 0.05). Processed kernels of mango (Mangifera indica) (PMK) and bush mango (Irvingiagabonensis) (PBMK), therefore, could find application as neutraceuticals and antimicrobials.  相似文献

A study was undertaken with the objective of evaluating the storage stability of the polyphenols and the antioxidant activity in the peel from two varieties of potatoes, Penta and Marcy, grown in Ontario, Canada. Samples were stored at ?20, 4 and 25 °C for 0, 2, 4 and 8 weeks. Peel from Penta variety contained higher levels of total polyphenolic compounds, 2.4 mg/g of peel d.w. compare to Marcy, 1.14 mg/g of peel d.w. Chlorogenic and caffeic acids were the major polyphenolic compounds present in both verities. Levels of polyphenolic compounds in the peel were influenced by storage temperature with maximum loss observed at 25 °C. Storage time caused a decline in the levels of polyphenolic compounds up to 4 weeks at all temperatures followed by a significant increase at the end of week 8. There was a parallel increase in the antioxidant activity of the peel samples during storage and until the end of 8 weeks of storage. At all temperatures of storage, there was a significant relationship (r = 0.43, P  0.05) between the amount of polyphenolic compound in the peel samples and their antioxidant capacity. Results suggest potato peel as a valuable source of polyphenolic compounds requiring proper storage condition to maintain their antioxidant properties.  相似文献

Supercritical fluid extraction (SFE) was applied for the extraction of valuable compounds from grape (Vitis labrusca B.) peel. Extraction was carried out according to an orthogonal array design (OAD) and independent variables selected were temperature, pressure and modifier concentration. SFE process was optimized by using response surface methodology (RSM) for the extract yield, total phenols, antioxidants and total anthocyanins from grape peel. Effects of extraction temperature and pressure were found to be significant on all responses. Optimal SFE conditions were identified as 45–46 °C temperature, 160–165 kg cm? 2 pressure and 6–7% ethanol as modifier for maximum extract yield (12.31%), total phenols (2.156 mg GAE/100 mL), antioxidants (1.628 mg/mL) and total anthocyanins (1.176 mg/mL). Experimental values for response variables at these optimal conditions match well with the predicted values. Grape peel extracts obtained by SFE showed more than 93% DPPH radical scavenging activities.Industrial relevanceThis study describes the response surface optimization of supercritical fluid extraction (SFE) process for the enhanced recovery of total phenols, antioxidant and anthocyanins from grape peel. SFE uses CO2 as supercritical fluid which is environment friendly solvent; allows extraction at lower temperature and the extracts obtained possess higher quality and safety. Industrially, it may be used as a promising technique for the extraction of bioactive compounds from plant materials.  相似文献

The effects of farming system, geographical origin, and grape variety on the in vitro antioxidant capacity, some physicochemical properties and chemical composition were investigated. Major and minor phenolic compounds, reducing and antioxidant assays using chemical and biological systems were determined in n = 62 100% purple grape Brazilian juices (Vitis labrusca L.) from organic and conventional systems from the three largest producing regions. Results were subjected to ANOVA, MANOVA, and principal component analysis. No distinction was observed between organic and conventional grape juices regarding the in vitro antioxidant/reducing capacities, but only through the analysis of individual phenolic acids. The geographical origin and grape variety played a more important role in distinguishing Brazilian grape juices according to the antioxidant activity and some chemical markers. MANOVA was applied and the combined effect of farming system and geographical origin was only significant for tonality (p = 0.044) and protocatechuic acid content (p = 0.030). Correlation analysis was performed and results showed that total phenolic content, total flavonoids, flavonols, anthocyanins, and condensed tannins were statistically associated (p < 0.05) with FRAP, ABTS, and reducing potential of the hydrophilic phenolic compounds (RPHPC). Ascorbic acid did not seem to be involved in the antioxidant potential of the selected purple grape juices. Protocatechuic acid was statistically correlated to FRAP (r = 0.4636, p < 0.001), ABTS (r = 0.3298, p = 0.010), and RPHPC (r = 0.3623, p = 0.004), while ferulic acid correlated with FRAP (r = 0.3410, p = 0.008) and RPHPC (r = 0.2662, p = 0.040). The inhibition of lipid peroxidation conducted at 37 °C in a biological system buffered at pH 7.4 was significantly (p < 0.05) correlated to RPHPC (r = 0.533, p = 0.013) and to the scavenging activity toward ABTS radical (r = 0.505, p = 0.020). In addition, principal component analysis revealed that the inhibition of lipid peroxidation was associated with higher contents of total phenolic content, flavonoids, anthocyanins, flavonols, condensed tannins, and protocatechuic acid. Theobroma cacao L. (Sterculiaceae) and cocoa-derived products are phenolics-rich food; these products are largely studied because of the antioxidant and antiradical in vitro properties of phenolic constituents. Cocoa hulls are the principal by-product of cocoa, separated from the cotyledons during the pre-roasting process or after the roasting process of T. cacao beans (de-hulling/de-husking step). This by-product is a matrix rich in fiber (namely insoluble, but also represented by pectins) and phenolics. Supercritical CO2 is a powerful mild technology able to extract and fractionate from plant or animal foods without the use of organic solvent. This approach was used to extract some phenolics fractions from cocoa hulls. Only two recovered fractions, (150 bar, 50 °C, re-dissolved in acetone; 200 bar, 50 °C, re-dissolved in acetone), apparently free from (-)epicatechin, catechin and phenolic acids, showed protective action in an in vitro test (SH-SY5Y cells, differentiated to a neuronal phenotype using retinoic acid and then exposed to ischemic damage), similar to the action of cabergoline and vitamin E. We suggest the use of supercritical CO2 for the isolation of bioactive fractions from cocoa hulls and an in vitro model as a useful model to study the antioxidant/antiradical properties of isolated phenolic pigments.  相似文献

In this work, the effect of the methods used for the freezing and drying of olive leaves on the polyphenol content and antioxidant capacity of the extracts was addressed. Thus, different methods were used to dry olive leaves (fresh or frozen by conventional (? 28 °C) or N2 freezing): hot air drying at 70 or 120 °C and freeze drying. The extracts were characterized by determining the phenolic content, antioxidant capacity and HPLC-DAD/MS–MS profile.Drying had a significant (p < 0.05) influence on the antioxidant potential of olive leaf extracts. Both the drying and freezing methods significantly (p < 0.05) influenced the concentration of the main polyphenols identified. Hot air drying provided a higher phenolic content, especially in oleuropein, than freeze drying. Thus, drying at 120 °C was the best processing condition. Freezing reduced the antioxidant potential as compared to fresh leaves, probably due to oxidase activation, although its influence was not dependent on the freezing method.  相似文献
