The stabilities of the bacterial community structures supported by seven full-scale biological reactors treating pharmaceutical wastewater were investigated by denaturing gradient gel electrophoresis (DGGE) of polymerase chain reaction (PCR) amplified 16S rRNA gene fragments. Effluent quality from this treatment process was consistently high with respect to BOD5 (<30 mgl(-1)), soluble COD (<500 mgl(-1)), and total ammonia (< 5 mgl(-1) as N) concentrations. Long-term community structure stability was studied by comparing the similarity of PCR-DGGE fingerprints from samples collected 87 days apart between which the influent wastewater characteristics were relatively stable. The Dice index (Cs) of similarity was moderately high for the first four reactors (Cs = 0.61-0.77) and very high for the last three reactors (Cs = 0.89-0.91). Short-term community structure stability was studied by comparing PCR-DGGE fingerprints from samples collected 15 days apart between which the influent wastewater characteristics changed significantly, while the effluent quality remained consistently high. The bacterial community composition of each of the seven bioreactors showed a moderate community shift (Cs = 0.70-0.76). Short-term variability in influent wastewater composition, therefore, affected a greater community shift than did long-term operation treating a wastewater of relatively consistent composition. These results indicate that functionally stable wastewater treatment bioreactors have stable microbial community structures under normal operating conditions but are able to adapt in response to perturbations to sustain high effluent quality.

Changes in microbial community structure, associated with changes in process performance, were investigated with respect to the sludge retention time (SRT) in bioreactors treating thiocyanate. Among the seven reactors operated at 0.8-3.0 d SRTs, respectively, the reactor at 2.0 d SRT displayed the maximal thiocyanate removal rate of 240.2mg/L/d. However, the thiocyanate removal efficiency suddenly decreased from 96.1% to 43.1% when the SRT was reduced from 2.0 to 1.8d, corresponding to a 50.1% drop in the removal rate. Microbial communities in the reactors operated at short SRTs, near washout, were analyzed by denaturing gradient gel electrophoresis (DGGE) based on bacterial 16S rRNA genes. All band sequences recovered were assigned to two phyla, Proteobacteria and Bacteriodetes. A Thiobacillus-like microorganism was commonly detected in all the reactors and is suggested to be the main organism responsible for thiocyanate decomposition. Several DGGE band sequences were closely related to the environmental clones detected in environments rich in sulfur and/or nitrogen compounds. Statistical analysis of the DGGE profiles demonstrated that the structure of thiocyanate-degrading communities, as well as the process performance, changed with change in SRT. The microbial community profiles were not always more closely related to those at similar SRT than those at less similar SRT on the non-metric multidimensional scaling (NMDS) map. This was also supported by clustering analysis. These results were contrary to the general notion that the community structures in continuous systems will be controlled by the washout of microbial populations. Our experimental results suggest that the structure of a microbial thiocyanate-degrading community at a given SRT would not be determined only by the washout effect.  相似文献

The total, ammonia-oxidizing, and denitrifying Bacteria in a full-scale membrane bioreactor (MBR) were evaluated monthly for over one year. Microbial communities were analyzed by denaturing gradient gel electrophoresis (DGGE) and clone library analysis of the 16S rRNA and ammonia monooxygenase (amoA) and nitrous oxide reductase (nosZ) genes. The community fingerprints obtained were compared to those from a conventional activated sludge (CAS) process running in parallel treating the same domestic wastewater. Distinct DGGE profiles for all three molecular markers were observed between the two treatment systems, indicating the selection of specific bacterial populations by the contrasting environmental and operational conditions. Comparative 16S rRNA sequencing indicated a diverse bacterial community in the MBR, with phylotypes from the α- and β-Proteobacteria and Bacteroidetes dominating the gene library. The vast majority of sequences retrieved were not closely related to classified organisms or displayed relatively low levels of similarity with any known 16S rRNA gene sequences and thus represent organisms that constitute new taxa. Similarly, the majority of the recovered nosZ sequences were novel and only moderately related to known denitrifiers from the α- and β-Proteobacteria. In contrast, analysis of the amoA gene showed a remarkably simple ammonia-oxidizing community with the detected members almost exclusively affiliated with the Nitrosomonas oligotropha lineage. Major shifts in total bacteria and denitrifying community were detected and these were associated with change in the external carbon added for denitrification enhancement. In spite of this, the MBR was able to maintain a stable process performance during that period. These results significantly expand our knowledge of the biodiversity and population dynamics of microorganisms in MBRs for wastewater treatment.  相似文献

Microbial community structure of a lab scale thermophilic aerobic wastewater treatment reactor was analyzed by a combination of culture-independent methods. Quinone profile method provides for chemical analysis of respiratory quinone molecular species, which corresponds to bacterial groups. Denaturing gradient gel electrophoresis (DGGE) of PCR-amplified 16S rDNA partial sequences (PCR-DGGE) clarifies community changes at species level, as DGGE can separate DNA fragments of different sequences. Certain phvlogenetic groups of bacterial cells can be labeled by fluorescence in situ hybridization (FISH). Quinone profile showed a predominant presence of MK-7. PCR-DGGE revealed that constituents of the community were unchanged during the stable phase. FISH demonstrated the existence of the relatives of Bacillus lentus and B. thermocloacae in considerable proportions. The community was mainly composed of Bacillaceae, and obligate thermophilic and mesophilic Bacillus appeared in spite of the temperature fluctuation from 35 degrees C to 60 degrees C. The combination of these culture-independent methods revealed the community precisely enough to evaluate the reactor performance.  相似文献

Three identical sequential batch reactors (SBRs) were each inoculated with sludge from a full-scale wastewater treatment plant (WWTP) treating a waste stream of different origin, i.e. a hospital, a meat processing company, and a municipal WWTP. The SBRs were run in parallel for 84 consecutive days to investigate whether the reactors would become more phylogenetically similar or stay separated concerning their functionality and microbial communities. Overall, the nitrification functionality was high throughout the experiment, and the size and structure of the sludge flocs were very similar. The total bacterial and ammonia-oxidizing bacterial (AOB) communities were analyzed by PCR-DGGE. Cluster analysis demonstrated very distinct bacterial communities in the three SBRs, not showing any trend becoming more similar. The carrying capacity, dynamics and functional organization of the communities were assessed by DGGE analysis and based on these patterns the range-weighted richness, moving window analysis, and constructing Pareto-Lorenz evenness distribution curves were calculated. Between the SBRs, highly comparable internal structure and dynamics of the AOB communities were observed, although they had only one AOB DGGE band in common. These observations indicate that community characteristics such as the extent of biodiversity and dynamics are more important indicators of good microbial functionality than the presence of certain specific species.  相似文献

Quantitative changes in methanogenic community structures, associated with performance data, were investigated in three anaerobic batch digesters treating synthetic glucose medium, whey permeate, and liquefied sewage sludge. All digesters were initially seeded with anaerobic sludge obtained from a local municipal wastewater treatment plant. Dynamics of methanogenic populations were monitored, at order and family levels, using real-time PCR based on the 16S rRNA gene. The molecular monitoring revealed that, in each digester, the quantitative structure of methanogenic community varied continuously over treatment time and the variation corresponded well to the changes in chemical profiles. Biphasic production of methane, associated with successive increases in aceticlastic (mainly Methanosarcinaceae) and hydrogenotrophic (mainly Methanomicrobiales) methanogenic groups, was observed in each digester. This corresponded to the diauxic utilization of acetate and longer-chain volatile fatty acids (C3-C6), mainly propionate. Additionally, the non-metric multidimensional scaling (NMDS) analysis of the quantification results demonstrated that the community shift patterns in three digesters were totally different from each other. Considering that the operating conditions in all trials were identical except substrates, the differences in quantitative shift profiles were suggested to be due to the different substrate compositions. This implied that the composition of wastewater could affect the evolution of quantitative methanogenic community structure in an anaerobic process. Overall, our results suggested that more attention to quantitative as well as qualitative approaches on microbial communities is needed for fundamental understanding of anaerobic processes, particularly under dynamic or transitional conditions.  相似文献

Membrane fouling remains a major obstacle for wider application of membrane bioreactors (MBRs) to wastewater treatment. Polysaccharides in mixed liquor suspensions in the reactors are thought to be mainly responsible for the evolution of membrane fouling in MBRs. However, details of polysaccharides causing membrane fouling in MBRs are still unknown. In this study, polysaccharides in a mixed liquor suspension of a pilot-scale MBR treating municipal wastewater were fractionated by using lectins, special proteins that bind to specific polysaccharides depending on their properties. Fouling potentials of the fractionated polysaccharides were assessed by bench-scale dead-end filtration tests. It was clearly shown that the degrees of fouling caused by fractionated polysaccharides were significantly different. The amounts of polysaccharides in each fraction could not explain the variations in the fouling, indicating the presence of polysaccharides with high specific fouling potentials. To investigate structures and origins of the polysaccharides with high fouling potentials, matrix-assisted laser desorption/ionization (MALDI)-time of flight (TOF)/mass spectrometry (MS) analysis was applied to the fractionated polysaccharides after partial hydrolysis. Several mass peaks obtained could be assigned to fragments of structures of polysaccharides (i.e., oligosaccharides) reported in a database/literature. This is the first report showing the plausible structures of polysaccharides in MBRs based on MS. A deeper understanding and effective control of membrane fouling in MBRs could be achieved with information obtained by the approach used in this study.  相似文献

A pilot scale submerged ultra-filtration membrane bioreactor (MBR) was used for the aerobic treatment of domestic wastewater over 9 months of year 2006 (28th March to 21st December). The MBR was installed at a municipal wastewater facility (EMASAGRA, Granada, Spain) and was fed with real wastewater. The experimental work was divided in 4 stages run under different sets of operation conditions. Operation parameters (total and volatile suspended solids, dissolved oxygen concentration) and environmental variables (temperature, pH, COD and BOD5 of influent water) were daily monitored. In all the experiments conducted, the MBR generated an effluent of optimal quality complying with the requirements of the European Law (91/271/CEE 1991). A cultivation-independent approach (polymerase chain reaction-temperature gradient gel electrophoresis, PCR-TGGE) was used to analyze changes in the structure of the bacterial communities in the sludge. Cluster analysis of TGGE profiles demonstrated significant differences in community structure related to variations of the operation parameters and environmental factors. Canonical correspondence analysis (CCA) suggested that temperature, hydraulic retention time and concentration of volatile suspended solids were the factors mostly influencing community structure. 23 prominent TGGE bands were successfully reamplified and sequenced, allowing gaining insight into the identities of predominantly present bacterial populations in the sludge. Retrieved partial 16S-rRNA gene sequences were mostly related to the α-Proteobacteria, β-Proteobacteria and γ-Proteobacteria classes. The community established in the MBR in each of the four stages of operation significantly differed in species composition and the sludge generated displayed dissimilar rates of mineralization, but these differences did not influence the performance of the bioreactor (quality of the permeate). These data indicate that the flexibility of the bacterial community in the sludge and its ability to get adapted to environmental changes play an important role for the stable performance of MBRs.  相似文献

The effect of temperature was studied on the efficiency of soluble COD removal and bacterial community development during the aerobic biological treatment of a pharmaceutical wastewater. Using wastewater and bacterial inoculum obtained from the full-scale facility treating this wastewater, batch laboratory cultures were operated at 5°C intervals from 30°C to 70°C. Following four culture transfers to allow for bacterial acclimation, residual soluble COD levels were measured and bacterial community fingerprints were obtained by denaturing gradient gel electrophoresis (DGGE) of polymerase chain reaction (PCR)-amplified 16S rRNA gene fragments. Soluble COD removal efficiency declined as temperature increased from 30°C (62%) to 60°C (38%). Biological treatment of this wastewater failed to occur at temperatures higher than 60°C. Gradual shifts in bacterial community structure were detected as temperature increased, including a concomitant reduction in the number of different bacterial populations. The impact of temperature on a two-stage biological treatment process was also compared. Better soluble COD removal was achieved when both reactors were operated at 30°C compared to a system where the two stages were consecutively operated at 55°C and 30°C. These results indicate that operation of aerobic biological wastewater treatment reactors at elevated temperatures can have adverse effects on process performance.  相似文献

The effects of toxins at ambient concentrations on microbial activity and community dynamics are poorly understood. We operated 4 membrane bioreactors (MBRs) in parallel; two reactors were continuously exposed to the toxin 3-chloroaniline (3-CA) at environmentally relevant levels, representing 25% of the total chemical oxygen demand (COD; Total COD = 400 mg l−1 d−1), and two reactors received no 3-CA. During the 70 d exposure to 3-CA the microbial communities never adapted as evidenced by a 48% and 14% reduction in COD and ammonia removal, respectively, compared to over 92% reduction for both measurements in the controls. The bacterial 16S rRNA gene was monitored using terminal restriction fragment length polymorphism (T-RFLP) analysis (n = 15 temporal grab samples per reactor) over the 70 d period. T-RFLP spectra analysis compared the rapid species turnover rate (STR) approach with the more computationally intensive non-metric multi-dimensional scaling (NMS) complemented with multi-response permutation procedure (MRPP). The methods revealed comparable findings and the presence of 3-CA selected for a more convergent community with less bacterial turnover. In contrast, the control MBRs were more divergent as evidenced by greater bacterial turnover variability. The importance of studying replicate reactors is highlighted by the fact that one of the two controls was significantly different from the treatment MBRs (p-value = 0.01, α = 0.05) whereas the other one was not (p-value = 0.24, α = 0.05). The study suggests that analysis of community dynamics with the rapid STR approach and with NMS/MRPP can lead to comparable results when targeting the 16S rRNA gene. The use of replicate bioreactors is essential for meaningful interpretation of microbial community patterns.  相似文献

Biomass characteristics and microbial community diversity between a submerged membrane bioreactor with mixed liquor recirculation (MLE/MBR) and a membrane bioreactor with the addition of integrated fixed biofilm medium (IFMBR) were compared for organic carbon and nitrogen removal from wastewater. The two bench-scale MBRs were continuously operated in parallel at a hydraulic retention time (HRT) of 24 h and solids retention time (SRT) of 20 d. Both MBRs demonstrated good COD removal efficiencies (>97.7%) at incremental inflow organic loading rates. The total nitrogen removal efficiencies were 67% for MLE/MBR and 41% for IFMBR. The recirculation of mixed liquor from aerobic zone to anoxic zone in the MLE/MBR resulted in higher microbial activities of heterotrophic (46.96 mg O2/gVSS h) and autotrophic bacteria (30.37 mg O2/gVSS h) in the MLE/MBR compared to those from IFMBR. Terminal Restriction Fragment Length Polymorphism analysis indicated that the higher nitrifying activities were correlated with more diversity of nitrifying bacterial populations in the MLE/MBR. Membrane fouling due to bacterial growth was evident in both the reactors. Even though the trans-membrane pressure and flux profiles of MLE/MBR and IFMBR were different, the patterns of total membrane resistance changes had no considerable difference under the same operating conditions. The results suggest that metabolic selection via alternating anoxic/aerobic processes has the potential of having higher bacterial activities and improved nutrient removal in MBR systems.  相似文献

The structure and microbial communities of biofilms developing on cross-flow nanofiltration (NF) membranes at different temperatures (20, 25 or 34 degrees C) and operation lengths (8h-24days) were studied. Feedwater comprised tertiary quality wastewater effluent or synthetic media mimicking effluents of intermediate quality. After each run, the membranes were autopsied for bacterial enumeration, bacterial community composition and microscopy visualization (SEM, CLSM and AFM/NSOM). Community composition was analyzed by polymerase chain reaction-denaturing gradient gel electrophoresis (PCR-DGGE) coupled with sequence analysis of 16S rRNA gene fragments from dominant bands. Deposition of polysaccharides and initial bacterial colonization were observed within 8h, whereas developed biofilms markedly affecting membrane permeability were evident from days 2-3 onwards. Regardless of applied conditions, the heterotrophic plate counts in the biofilm were 3-4x10(6)CFU/cm(2) and the thickness of the biofouling layer was 20-30microm. From a total of 22 sequences obtained from 14 independent experiments, most species identified were Gram negative (19 of 22 sequences). Proteobacteria were found to be a prevalent group in all cases (16 of 22 sequences) and among it, the beta-subclass was the most predominant (8 sequences), followed by the gamma-subclass (5 sequences). Pseudomonas/Burkholderia, Ralstonia, Bacteroidetes and Sphingomonas were the dominant groups found in most cases. Even though the microbial population might be important with respect to biofouling patterns, membrane permeability decline seems to be more substantially influenced by the formation and accumulation of exopolymeric substances (EPS).  相似文献

Activated sludge is one of the most abundant and effective wastewater treatment process used to treat wastewater, and has been used in developed countries for nearly a century. In all that time, several hundreds of studies have explored the bacterial communities responsible for treatment, but most studies were based on a handful of samples and did not consider temporal dynamics. In this study, we used the DNA fingerprinting technique called automated ribosomal intergenic spacer region analysis (ARISA) to study bacterial community dynamics over a two-year period in two different treatment trains. We also used quantitative PCR to measure the variation of five phylogenetically-defined clades within the Accumulibacter lineage, which is a model polyphosphate accumulating organism. The total bacterial community exhibited seasonal patterns of change reminiscent of those observed in lakes and oceans. Surprisingly, all five Accumulibacter clades were present throughout the study, and the total Accumulibacter community was relatively stable. However, the abundance of each clade did fluctuate through time. Clade IIA dynamics correlated positively with temperature (ρ = 0.65, p < 0.05) while Clade IA dynamics correlated negatively with temperature (ρ = −0.35, p < 0.05). This relationship with temperature hints at the mechanisms that may be driving the seasonal patterns in overall bacterial community dynamics and provides further evidence for ecological differentiation among clades within the Accumulibacter lineage. This work provides a valuable baseline for activated sludge bacterial community variation.  相似文献

The goal of this study was to investigate the bacterial community in activated sludge with an anaerobic side-stream reactor (ASSR), a process permitting significant decrease in sludge production during wastewater treatment. The study operated five activated sludge systems with different sludge treatment schemes serving as various controls for the activated sludge with ASSR. Bacterial communities were analyzed by denaturing gradient gel electrophoresis (DGGE), sequencing and construction of phylogenetic relationships of the identified bacteria. The DGGE data showed that activated sludge incorporating ASSR contained higher diversity of bacteria, resulting from long solids retention time and recirculation of sludge under aerobic and anaerobic conditions. The similarity of DGGE profiles between ASSR and separate anaerobic digester (control) was high indicating that ASSR is primarily related to conventional anaerobic digesters. Nevertheless, there was also unique bacteria community appearing in ASSR. Interestingly, sludge in the main system and in ASSR showed considerably different bacterial composition indicating that ASSR allowed enriching its own bacterial community different than that from the aeration basin, although two reactors were connected via sludge recirculation. In activated sludge with ASSR, sequences represented by predominant DGGE bands were affiliated with Proteobacteria. The remaining groups were composed of Spirochaetes, Clostridiales, Chloroflexi, and Actinobacteria. Their putative role in the activated sludge with ASSR is also discussed in this study.  相似文献

A membrane bioreactor (MBR) is a promising wastewater treatment technology, but there is a need for efficient control of membrane fouling, which increases operational and maintenance costs. Soluble microbial products (SMP) have been reported to act as major foulants in the operation of MBRs used for wastewater treatment. In this study, SMP in MBRs operated with different sludge retention times (SRTs) were investigated by means of various analytical techniques and their relations to the evolution of membrane fouling were considered. Bench-scale filtration experiments were carried out in a laboratory with synthetic wastewater to eliminate fluctuations that would occur with the use of real wastewater and that would lead to fluctuations in compositions of SMP. Three identical submerged MBRs were operated for about 50 days under the same conditions except for SRT (17, 51 and 102 days). Accumulation of SMP in the MBRs estimated by conventional analytical methods (i.e., the phenol-sulfuric acid method and the Lowry method) was significant in the cases of short SRTs. However, the degrees of membrane fouling in the MBRs were not directly related to the concentrations of SMP in the reactors estimated by the conventional analytical methods. Non-conventional analytical methods such as excitation-emission matrix (EEM) fluorescence spectroscopy revealed that characteristics of SMP in the three reactors considerably differed depending on SRT. Foulants were extracted from the fouled membranes at the end of the operation and were compared with SMP in each MBR. It was clearly shown that characteristics of the foulants were different depending on SRT, and similarities between SMP and the extracted foulants were recognized in each MBR on the basis of results of EEM measurements. However, such similarities were not found on the basis of results obtained by using the conventional methods for analysis of SMP. The results of this study suggest that the use of conventional methods for analysis of SMP is not appropriate for investigation of membrane fouling in MBRs.  相似文献

The behaviour of microbial populations responsible for degradation of the aromatic compounds, phenol, benzoate, and salicylate, and changes of microbial community structures in seawater microcosms were analysed quantitatively and qualitatively using MPN–PCR and PCR–DGGE. The purpose of the study was to investigate the ecology of the entire microbial community during bioremediation. Bacterial populations possessing catechol 1,2-dioxygenase (C12O) DNA were evidently the primary degraders of phenol and benzoate, but others possessing catechol 2,3-dioxygenase (C23O) DNA increased to enhance substrate degradation under high-load conditions when the substrates were present for long periods. However, salicylate degradation was evidently facilitated by specific bacterial populations possessing C23O DNA. PCR–DGGE analyses suggested that bacterial populations already relatively dominant in the original microcosm contributed to phenol degradation. Bacteria composing a minor fraction of the original population apparently increased and contributed to benzoate degradation. Bacterial populations possessing C23O DNA were responsible for salicylate degradation, however, and different degrading bacteria were evidently selected for, depending on the initial salicylate concentration. Microbial community structure tended to be simplified by aromatic compound degradation. Thus, microbial monitoring can elucidate the behaviour of bacterial populations responsible for aromatic compound degradation and be used to assess the effects of bioremediation on intact microbial ecosystems.  相似文献

Submerged membrane bioreactors (MBRs) are now widely used for various types of wastewater treatment. One drawback of submerged MBRs is the difficulty in removing nitrogen because intensive aeration is usually carried out in the tank and the MBRs must therefore be operated under aerobic conditions. In this study, the feasibility of treating municipal wastewater by a baffled membrane bioreactor (BMBR), particularly in terms of nitrogen removal, was examined. Simultaneous nitrification/denitrification in a single and small reaction tank was possible by inserting baffles into a normal submerged MBR as long as wastewater was fed in the appropriate way. To examine the applicability of the BMBR, pilot-scale experiments were carried out using real municipal wastewater. Although neither external carbon addition nor mixed liquor circulation was carried out in the operation of the BMBR, average removal rates of total organic carbon (TOC), total phosphorus (T-P) and total nitrogen (T-N) reached 85%, 97% and 77%, respectively, with the hydraulic retention time (HRT) of 4.7h. Permeability of the membrane could be maintained at a high level throughout the operation. It was found that denitrification was the limiting step in removal of nitrogen in the BMBR in this study. Various types of monitoring carried out in the BMBR also demonstrated the possibility of further improvements in its performance.  相似文献

A novel Bio-Entrapped Membrane Reactor (BEMR) packed with bio-ball carriers was constructed and investigated for organics removal and membrane fouling by soluble microbial products (SMP). An objective was to evaluate the stability of the filtration process in membrane bioreactors through backwashing and chemical cleaning. The novel BEMR was compared to a conventional membrane bioreactor (CMBR) on performance, with both treating identical wastewater from a food and beverage processing plant. The new reactor has a longer sludge retention time (SRT) and lower mixed liquor suspended solids (MLSS) content than does the conventional. Three different hydraulic retention times (HRTs) of 6, 9, and 12 h were studied. The results show faster rise of the transmembrane pressure (TMP) with decreasing hydraulic retention time (HRT) in both reactors, where most significant membrane fouling was associated with high SMP (consisting of carbohydrate and protein) contents that were prevalent at the shortest HRT of 6 h. Membrane fouling was improved in the new reactor, which led to a longer membrane service period with the new reactor. Rapid membrane fouling was attributed to increased production of biomass and SMP, as in the conventional reactor. SMP of 10-100 kDa from both MBRs were predominant with more than 70% of the SMP <100 kDa. Protein was the major component of SMP rather than carbohydrate in both reactors. The new reactor sustained operation at constant permeate flux that required seven times less frequent chemical cleaning than did the conventional reactor. The new BEMR offers effective organics removal while reducing membrane fouling.  相似文献
