Representative cottonseed salad oils, corn oils, lards and hydrogenated vegetable shortenings, and portions of the same fats heated at 182 C for 120 hr were fed as 20% of nutritionally adequate diets to weanling albino rats in longevity studies. Differences in the responses of rats fed diets containing the unheated and heated fats were generally small with respect to rates of gain, 12 th week and adult weights, efficiencies of utilization of absorbed energy, incidences of grossly detectable diseases and longevities. There were no indications that the feeding of the heated fats had shortened survival times in comparison with the comparable unheated fat. Animals fed hydrogenated vegetable shortening, heated or unheated, survived the longest. However, gains were slightly slower with the heated cotton-seed oil diets, and food efficiencies were slightly lower with the heated cottonseed oil and heated lard diets because of decreased digestibilities of these fats. The usual disabilities of old age such as nephritis, respiratory disease and periarteritis were present in all groups. The incidence of mammary tumors was high but did not differ significantly with the kind of fat, heaated or unheated. Tumor incidence other than mammary was similar in both sexes and there was no significant difference between fresh and heated fats. Absence of adverse effects attributable to the heated fats during the life span of the rats in further evidence of the safety of these fats of the quality customarily consumed by the human population.  相似文献

Since it was first suggested that high dietary fat is a risk factor in colon cancer, there have been several studies to test this hypothesis. Epidemiologic studies suggested a positive association between dietary fat and colon cancer. Laboratory animal model studies demonstrated that not only the amount of rat, but also types of fat differing in fatty acid composition are important determining factors in colon tumor development. Chemically-induced colon tumor incidence was increased in rats fed the semipurified diets containing 23% corn oil, safflower oil, lard or beef tallow (high-fat) as compared to those fed 5% corn oil, safflower oil, lard or beef tallow diets (low-fat). Diets containing 23% conconut oil, olive oil or fish oil, or high-fat diets containing varying levels oftrans fat, had no colon tumor-enhancing effect compared to their respective low fat diets. The stage at which the effect of dietary fat is exerted appears to be mostly during the post-initiation phase of colon carcinogenesis. Lack of a colon tumor enhancing effect of dietary fish oil is observed both during the initiation and postinitiation phases. The mechanisms by which various dietary fats increase colon carcinogenesis are not fully understood. In most instances, however, the high-fat diet appears to enhance tumorigenesis through elevation of agents, such as secondary bile acids, that act as promoters of tumor development. Lack of colon tumor promotion by dietary fish oil andtrans fat appears to be mediated through their effect on mucosal ornithine decarboxylase activity, colonic secondary bile acids and/or prostaglandin synthesis. Our objective was to determine the relative rates ofin vivo triglyceride (TG) secretion and the composition of very low density lipoproteins (VLDL) in rats fed different dietary saturated fats. Male Sprague-Dawley rats (150–200 g) were fed diets containing 16% corn oil, or 14% butterfat, 14% beef tallow, 14% olive oil, or 14% coconut oil plus 2% corn oil for 5 wk. Changes in plasma TG specific radioactivity were determined in individual, unanesthetized fasted rats after injection of 100 μCi [2-3H]glycerol. Nonlinear regression analysis using a 2-compartment model was used to determine the fractional rate constant for TG turnover in plasma. The plasma TG pool was 33–40% larger with beef tallow than with corn, olive or coconut oil feeding (p<0.05), and 20% larger with beef tallow than with butterfat feeding. The rate of TG secretion into plasma (mg/min/100 g body weight) was 60% higher in animals fed beef tallow than corn or coconut oil (p<0.05) and 26–33% higher in animals fed beef tallow than olive oil or butterfat. Differences in VLDL composition (% wt) were also noted. Our data suggest that greater TG secretion is the primary factor contributing to the larger TG pool with ingestion of beef tallow relative to butterfat, corn or coconut oil. These results suggest that different dietary saturated fats have unique effects on TG metabolism in rats. Male Sprague-Dawley rats were fed diets with 10%, 30%, or 50% of energy derived from fat for two weeks. The fats used were beef tallow, olive oil, peanut oil and butter. Aortic prostacyclin (PGI2) production, platelet aggregation and thromboxane A2 (TXA2) production and plasma and aortic phospholipid (PL) content were measured. Butter- and beef tallow-feeding reduced aortic PGI2 production and collagen-induced TXA2 production in a dosedependent manner as the level of fat in the diet increased. Neither olive oil nor peanut oil had any effect on aortic PGI2 production or collagen-induced TXA2 production. Butter-feeding also resulted in a decrease in collageninduced platelet aggregation; however, none of the other fats had any effect on collagen-induced platelet aggregation. The observed decreases in aortic PGI2 and collagen-induced TXA2 production were paralleled by similar decreases in aortic and plasma PL arachidonic acid content and an increase in eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA). Only the most saturated fats, butter and beef tallow, had significant inhibitory effects on prostanoid production and platelet aggregation.  相似文献

Male Sprague Dawley rats were fed semipurified diets containing 20% fat for 15 weeks. The dietary fats were corn oil, soybean oil, palm oil, palm olein and palm stearin. No differences in the body and organ weights of rats fed the various diets were evident. Plasma cholesterol levels of rats fed soybean oil were significantly lower than those of rats fed corn oil, palm oil, palm olein or palm stearin. Significant differences between the plasma cholesterol content of rats fed corn oil and rats fed the three palm oils were not evident. HDL cholesterol was raised in rats fed the three palm oil diets compared to the rats fed either corn oil or soybean oil. The cholesterol-phospholipid molar ratio of rat platelets was not influenced by the dietary fat type. The formation of 6-keto-PGF was significantly enhanced in palm oil-fed rats compared to all other dietary treatments. Fatty acid compositional changes in the plasma cholesterol esters and plasma triglycerides were diet regulated with significant differences between rats fed the polyunsaturated corn and soybean oil compared to the three palm oils.  相似文献

A semisynthetic diet containing 15% by weight of dietary fat was fed to six groups of male Wistar rats for 28 days. Two groups received the distilable fraction of fresh corn oil (DCO) or fresh olive oil (DOO), two groups the distillable fraction of the thermally oxidized fats (OCO,OOO), and two groups received the respective fresh fats as controls (FCO, FOO). Substantial changes in the fatty acid composition occurred in the fats upon thermal oxidation. Only the rats that received OOO showed overt symptoms of heated fat toxicity. This was reflected in the histological scores of these animals, with the liver sustaining the most numerous and severe lesions. Tissue fatty acid changes of any significance were confined largely to the polar liver lipids of the rats that were fed OCO or OOO. The results of this study would suggest that the relatively greater toxicity of OOO, compared to OCO, may in part be due to the high oleic:linoleic acid ratio of the fresh olive oil and in part to a higher tocopherol content of the corn oil.  相似文献

Summary The Korpaczy modification of the Stamm reaction for detecting rancidity in fats has been applied to a variety of fats and oils, including hog and beet fat, cottonseed oil, a number of other vegetable and seed oils, and a group of marine oils. By means of cellulose acetate electrophoresis and measurement of radioactivity, lipoproteins (d≤1,006) of lymph collected from rats fed a diet containing fats added to labeled fatty acids for 20–24 hr, have been separated into two kinds differing in their size. Fats with different saturated and unsaturated fatty acid content were tested: corn oil, lard, mutton tallow, tripalmitin, corn oil-lard mixture, and randomized palmitic-linoleic triglycerides. Factors studied were: (a) increased amounts of these fats given alone or included in a test meal; (b) added amounts of nutrients (other than fats) in the test meal; (c) adaptation to a diet containing 20% fat for 10 days. The major part of the labeled lymph lipid was always transported by the smaller particles. Size distribution of lymph fat particles was influenced by some of the factors studied. Generally, unsaturated fats produced higher amounts of larger particles.  相似文献

Summary There is increasing evidence that the abnormal nutritional properties of highly autoxidized fats are related to the polymers which develop during autoxidation. Lard and cottonseed oil were aerated at 95°C. for 200 hrs. and molecularly distilled; and the residue fractions, non-volatile at 275 to 300°C., were studied. Diets containing 20% of autoxidatively produced polymeric residue, fed to albino rats, led to diarrhea and rapid death, but when this residue was reduced to 10%, most of the animals were gradually able to tolerate it. At the 4 or 7% level it was well tolerated, but growth was reduced. There were no distinctive histological lesions, and withdrawal of the polymer permitted immediate realimentation without evidence of subsequent injuries. The polymeric residue from autoxidized cottonseed oil exerted a greater growth-depressant effect than that from lard, and the latter, more than that from a hydrogenated vegetable oil used for deep-fat frying for 80 hrs. at 190°C. Addition of fresh fat to the polymeric residues decreased their growth-depressant effect. When rats were fed a measured amount of diet sufficient to maintain their weight, the caloric requirement necessary for weight maintenance gradually decreased. When the dietary fat source consisted of polymeric residue to the extent of 4 to 10%, the caloric requirement for weight maintenance decreased relatively little, if at all. The polymeric residue from autoxidized lard was, in this respect, as effective as that from autoxidized cottonseed oil. We investigated modes whereby stearic acid (18∶0) exerts a neutral or cholesterol-lowering effect using dietary fats which provided graded levels of 18∶0 and distinct triacylglycerol (TAG) profiles. Male Sprague-Dawley rats (150–175 g) were fed diets containing 0.2% cholesterol and 16% fat from corn oil, or from 1% corn oil plus 15% lard (13.2% 18∶0), beef tallow (19.2% 18∶0) or cocoa butter (34.7% 18∶0) for 3 wk, and then killed in a fasted or fed state. Chylomicron (CM) fatty acid profiles suggested reduced absorption of 18∶0 with greater 18∶0 intake. CM TAG profiles indicated a reduction or loss of two TAG species compared to the TAG profiles of the stearate-rich diets: 1-palmitoyl-2-oleoyl-3-stearoyl glycerol (POS) and 1,3-distearoyl-2-oleoyl glycerol (SOS). Hepatic total cholesterol concentrations were 54–77% lower (P<0.01) in the cocoa butter-fed than the lard- and beef tallow-fed groups. The cocoa butter group showed a significantly lower ratio of high-density lipoprotein esterified/free cholesterol than all other groups. Hepatic stearoyl-CoA and oleoyl-CoA concentrations, the substrate and product for hepatic δ9 desaturase, were not significantly different for corn oil-fed and cocoa butter-fed groups in spite of a large difference in 18∶0 intake. These data suggest that the neutral or cholesterol-lowering effect of 18∶0 is not due to hepatic conversion of stearic to oleic acid, and that POS and SOS are poorly absorbed from stearate-rich dietary fats.  相似文献

Fresh and oxidized cotton seed oil (CO) olive oil (OO), chicken fat (CF) and beef fat (BF) were fed to male weanling rats for 33 to 108 weeks. Groups fed oxidized fats except OO showed a higher death rate than those fed the corresponding fresh fats. Groups fed oxidized CO and BF had the highest death rate. Histological studies of animals dying from natural causes showed more pronounced cardiac lesions in the animals fed oxidized CO. Serum, liver and brain cholesterol levels were not influenced by oxidized fats. Fatty acid composition of depot fats and of heart and liver lipids did not show significant differences between groups fed fresh and the corresponding oxidized fats. On the basis of variation ranges of fatty acids (83 milk fats) and triglycerides (763 milk fats) the limits of detection of foreign fats (13 different types) in milk fat are determined. In this respect, on the basis of certain ratios of triglycerides only for soybean oil, sunflower oil, and olive oil and also fo ratios of fatty acides only for soybean oil, sunflower oil, coconut oil and palm kernel oil low limits of detection of <5% were established. On the other hand triglyceride formulae with up to 8 individual triglycerides permit a sensitive qualitative (2–5%) and quantitative detection of all vegetable and also animal non-milk fats in milk fat, even with special feeding conditions of the cows (e.g. fats, Ca-soaps).  相似文献

A 91-day feeding study evaluated soybean oil, rapeseed oil, fully hydrogenated soybean oil, fully hydrogenated rapeseed oil, fully hydrogenated superglycerinated soybean oil and fully hydrogenated superglycerinated rapeseed oil at 7.5% of the diet in rats; a 16-wk feeding study evaluated soybean oil and the three rapeseed oils or fats at 15% of the diet. Each fat was fed to 40 rats as a mixture with soybean oil making up 20% of a semi-synthetic diet. No significant differences in body weight gains or diet-related pathology were seen in the 91-day study although the rats fed liquid rapeseed oil had slightly heavier hearts, kidneys and testes than the others. The rats fed the four fully hydrogenated fats ate more feed and had lower feed efficiencies than those fed oils but no differences were seen among the four hydrogenated fats. In the 16-wk feeding study, no pronounced pathology related to the diet was seen although the rats fed liquid rapeseed oil had a slightly higher incidence of histiocytic infiltration of cardiac muscle than the rats in the other groups. The female rats fed the three rapeseed oil fats gained significantly less weight and the females fed liquid rapeseed oil had enlarged hearts compared to the other groups. The absorbabilities of the six fats were measured in the 91-day study when fed as a mixture with soybean oil and as the sole source of dietary fat in a separate 15-day balance study. The four fully hydrogenated fats were poorly absorbed and the absorption of behenic acid from the two hydrogenated rapeseed oils was found to be 12% and 17% in the balance study and 8-40% in the feeding study. The adverse biological effects of unhydrogenated rapeseed oil containing erucic acid as reported in the literature do not occur with fully hydrogenated rapeseed oil. In addition, the low absorbability of the fully hydrogenated rapeseed oil is an added factor in its biological inertness.  相似文献

Within the last 15–20 years, major advances have been publicized regarding the mechanism and general understanding of the partial hydrogenation of triglycerides, including soybean oil, cottonseed oil, corn oil and various animal fats (primarily hog and beef fats). Although edible shortenings, oleomargarine stocks and soap stocks are produced in large quantities, there is considerable doubt that the theory and fundamental information relative to hydrogenation is always applied to the fullest extent in designing and operating commercial reactors. This paper will review past accomplishments, the current state of the art and probable directions to be taken to obtain even further reactor improvements. Summary Soybean oil, cottonseed oil, and lard were heatedin vacuo for either 70 and/or 100 min. at 610°F. and were fed to male and female rats at a level of 15% in the diet over a complete life span. Nutritional indices such as growth, including food efficiency and digestibility of the fats, reproduction and lactation, longevity, and tissue cholesterol and total lipid were measured. No evidence of impaired nutrition or harmfulness was observed in any group except in those fed the more highly polymerized soybean oil, where a decrease in iodine value of 10% and a 100% increase in viscosity was associated with a slight depression in growth and an interference with reproductive performance in the female animal. This interference with the reproductive process was alleviated by supplementation with α-tocopherol. There was no evidence of increased tumor incidence in the groups ingesting the heated oils. Summary 1. Dilatometric curves between 0°C. and their melting points have been obtained for the following fats: lard, butterfat, cottonseed oil, peanut oil, a commercial margarine oil, a commercial all-hydrogenated vegetable shortening, three samples of soybean oil hydrogenated to different degrees, a hard butter fractionally crystallized from hydrogenated peanut oil, a mixture of tristearin and soybean oil, and a synthetic fat containing equal molar proportions of stearic and oleic acids. 2. The dilatometric curves, of volume change in the fat against temperature, were in every case composed of a series of straight lines, separated by sharp breaks or transition points. 3. The number of linear sections in the dilatometric curves corresponded in a general way with the known degree of complexity in the glycerides of the fats, and varied from two in the case of the relatively simple stearic-oleic glyceride mixture, to at least seven in the case of the all-hydrogenated shortening. Since each break in the curve must correspond to the disappearance of a distinct class of solid glycerides or glyceride complexes, the application of dilatometry to the qualitative and quantitative determination of glyceride composition in fats is suggested. 4. Only two of the fats examined, the mixture of tristearin and soybean oil, and the synthetic stearicoleic glyceride mixture, exhibited polymorphism, even after rapid solidification in ice water. The effect of saturated and unsaturated fats on in vitro formation and release of lipids and cholesterol from14C acetate by rat intestinal tissue was investigated. The rats were fed a basal diet enriched with either 25% corn oil or lard and then sacrificed after a 10- or 25-day feeding period. It was observed that a similar14C lipid content but a greater14C cholesterol content was found in the intestinal tissue of rats fed corn oil than in rats fed lard for 10 days. After a longer period of feeding of 25 days, the intestinal tissue14C cholesterol level was decreased in the corn oil fed rats without any significant effect on other lipids. These data suggest that corn oil in some way influences cholesterol biosynthesis depending upon its degree of unsaturation and the period of time for which it is fed. The decrease at the later time might involve some mechanism which aids in getting rid of accumulated tissue cholesterol. Less14C lipid and14C cholesterol were released by the intestinal tissue of rats fed the unsaturated fat as compared with those fed the saturated fat, suggesting a possible role in vivo in reducing blood lipids and blood cholesterol levels. A method is described for the quantitative determination of the fully saturated triglyceride composition of fats. The method involves a quantitative oxidation of the unsaturated triglycerides in the fat to a mixture of α ketol and dihydroxy compounds, using aqueous alkaline potassium permanganate and a phase transfer catalyst in a two phase reaction. The fully saturated triglycerides are fractionated from the oxidation products by column chromatography on silicic acid and their composition is quantified by gas liquid chromatography (GLC) using a flame ionization detector. Analyses of samples of lard, cocoa butter, olive oil, soybean oil, crude palm oil (ex Malaysia) and the stearin fraction from palm oil are given as examples of the method. Carcass fats were obtained from weanling rats fed a complete diet for 8 weeks, which consisted of 2% cottonseed oil and 10% of the following fats: (1) corn oil; (2) the fatty acids of corn oil; (3) triricinolein; (4) ricinoleic acid; (5) the hydrogenated fatty acids of castor oil ; and (6) commercial hydrogenated shortening. The fats were subjected to both pancreatic lipase and nonspecific hydrolysis ; the resulting acids converted into methyl esters by conventional methods, and subjected to gas Chromatographie analysis. From these data, the positional distri-bution of the component fatty acids, glyceride types, and isomeric forms were calculated. The results indicated a preferential placement of un-saturated acids in the 2- position of the carcass triglycerides and that the carcass fat composition in terms of unsaturated (U) and saturated (S) fatty acid composition is not greatly influenced by the S and U compositions of the dietary fat. It was found that hydroxy acids or their tri-esters are metabolized much the same as are normal triglycerides and exert no particular in-fluence upon the fat structure of the rat. Some type of relationship between the dietary U and the U3 in the carcass fat appears to be present. The glycerides of the carcass fats examined here are essentially a random mixture of the major glyceride types, but the isomeric forms (SUS, S SU, USU and UUS) are a definite non-random mixture.
