Both soft (SD, Bam) and dry (DD, Kharak) date varieties were stored at 4 °C for six months followed by an additional one week storage at 18 °C. Antioxidant compounds (total phenolic content (TPC) and total flavonoid content (TFC)) of the dates increased following storage. Cluster analysis applied on TPC and TFC data before and after storage obtained two statistically significant clusters of SD and DD indicating that TPC and TFC had different behaviors according to the types of dates. As antioxidant concentration in dates was dependent on type of dates, selection of variety with high antioxidant compounds may be recommended. Further cold storage of up to six months followed by one week storage at 18 °C may further improve the level of antioxidant compounds.  相似文献

Microwave-assisted extraction (MAE) was investigated for extraction of phenolic compounds from tomato with maximised antioxidant activities using response surface methodology (RSM) coupled with a central composite design, and in vitro antioxidant assays (FRAP and ORAC). MAE was more efficient for greater antioxidant activities and higher total phenolic contents than solvent extraction. The optimal MAE processing parameters were 96.5 °C, 2.06 min, 66.2% ethanol for FRAP, and 96.5 °C, 1.66 min, 61.1% ethanol for ORAC. The models were successfully applied to 20 tomato cultivars, whose total phenolic contents (TPC) and indexes (TPI) were 489.30–997.45 mg gallic acid equivalent/100 g dry weight (DW) and 281.34–468.52 mg/100 g DW, respectively. Eight phenolic compounds were identified. Individual phenolics were 6.10–42.73 mg/100 g DW. The FRAP, but not the ORAC value showed good correlation with the TPC or TPI. The methodologies developed and the knowledge acquired in this study will provide useful information to tomato breeders and food processors.  相似文献

Total phenols (TPC) and antioxidant properties were determined in chick-pea, green and red lentils and sweet chestnut flours, in both aqueous-organic extracts and their residues, by the Folin Ciocalteau method and by the FRAP assay, respectively. Plant lignans were quantified in flours by means of HPLC. In addition, the FRAP of plant lignans (secoisolariciresinol, lariciresinol, isolariciresinol, pinoresinol, matairesinol), their mixture and enterolignans (enterodiol and enterolactone) were determined. In all flours, the highest TPC values were found in the residue. Specific and varietal significant differences were observed in all parameters. The highest TPC (737.32 and 1492.93 mg/100 g d.w.) and FRAP (140.32 and 101.25 μmol/g d.w.) values were reached by green lentils in both aqueous-organic extract and residue, respectively. Sweet chestnuts had the highest total lignans (980.03 μg/100 g d.w.). It was also found that the plant lignans standards have a higher antioxidant activity than enterolignans standards and that matairesinol has the highest activity.  相似文献

Concentrated muscadine pomace extract was chromatographically analyzed for its individual phenolic, flavonoid, and anthocyanidin compounds. This extract was also characterized regarding its total phenolic content (TPC), total flavonoid content (TFC), antioxidative activities in terms of scavenging DPPH free radicals, reducing ferric, and chelating Fe2+. The TPC of this product was 34.1 ± 1.8 mg of gallic acid equivalents(GAE)/g of extract, and TFC was 3.0 ± 0.3 mg of quercetin equivalents/g of extract. Some phenolic compounds including ellagic acid, gallic acid, (−)-epicatechin, (−)-epigallocatechin, catechin, myricetin, quercetin, and kaempferol were identified. Some anthocyanidins including delphinidin, cyanidin, petunidin, peonidin, and malvidin were also identified in the extract by using a combination of retention and spectral properties on a reverse-phase HPLC–PDA. In addition, 3,3′,4,4′5,5′-hexahydroxystilbene-a resveratrol analogue present in the extract was identified for the first time by LC–MS. The results from this study demonstrate that the muscadine pomace extract is rich of natural antioxidants such as phenolics, flavonoids and anthocyanins, and possesses strong antioxidant properties. Besides, the developed methods can be used for routine quality control of the muscadine products for manufacturing efficiency and consistency.  相似文献

Total phenolic content (TPC) and total antioxidant capacity (TAC) of four onion varieties (red, white, yellow and sweet) and shallot from selected locations (Washington, Idaho, Oregon, Texas and Georgia) were determined using Fourier transform infrared (FT-IR) spectroscopy (4000–400 cm−1). The Folin–Ciocalteu (F–C) assay was used to quantify TPC and three assays were used to determine TAC, including 2,2-diphenyl-picrylhydrazyl (DPPH) assay, Trolox equivalent antioxidant capacity (TEAC) assay and ferric reducing antioxidant power (FRAP) assay. Partial least squares regression (PLSR) with cross-validation (leave-one-out) was conducted on onion and shallot extracts (n = 200) and their corresponding F–C, DPPH, TEAC and FRAP values were employed to obtain four independent calibration models for predicting TPC and TAC for the extracts. Spectra from an extra 19 independent extracts were used as an external validation set for prediction. A correlation of r > 0.95 was obtained between FT-IR predicted and reference values (by F–C, DPPH, TEAC and FRAP assay) with standard errors of calibration (SEC) and standard errors of cross-validation (SECV) less than 2.85, 0.35 and 0.45 μmol Trolox/g FW of extracts for TEAC, FRAP and DPPH assay, respectively; and 0.36 mg gallic acid/g FW of extracts for the F–C assay. In addition, cluster analysis (principal component analysis (PCA)) and discriminant function analysis (DFA) could differentiate varieties of onions and shallot based upon infrared spectral features. Loading plots for the various chemometrics models indicated that hydroxyl and phenolic functional groups were most closely correlated with antioxidant capacity. The use of mid-infrared spectroscopy to predict the total antioxidant capacity of vegetables provides a rapid and precise alternative to traditional wet chemistry analysis.  相似文献

Fruit weight, antioxidant capacity, total anthocyanins, total phenolics, ascorbic acid, soluble solid content (SSC), reducing sugar and acidity of a number of selected cornelian cherry (Cornusmas L.) genotypes of varied pigmentation were investigated. Two methods, namely β-carotene bleaching and ferric reducing antioxidant power (FRAP) were used to determine total antioxidant capacity, while Folin–Ciocalteu reagent was used to determine total phenols. Fruit weight, SSC and ascorbic acid content of genotypes were 2.09–9.17; 12.53–21.17% and 29–112 mg/100 g, respectively. Antioxidant activity and total phenolic content varied among genotypes and 44-18 genotype had the highest antioxidant capacity using both methods. This genotype also had the highest total phenolic (74.8 mg GAE/g DW) and total anthocyanin (115 mg cyanidin-3-glucoside equivalents /100 g FW) content. There are linear relationships between antioxidant capacities and total phenols. The present study demonstrates the potential of certain cornelian cherry genotypes, notably 44-18, for improvement of nutritional value through germplasm enhancement programmes.  相似文献

Total phenolic content (TPC) and antioxidant activity (AOA) of 50% aqueous methanol extracts of the marine algae, Padina antillarum, Caulerpa racemosa and Kappaphycus alvarezzi were studied. TPC was measured using Folin-Ciocalteu method while 1, 1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging, ferric reducing antioxidant power (FRAP), ferrous ion chelating (FIC) assay and beta carotene bleaching (BCB) assay were used to study their AOA. P. antillarum was found to have the highest TPC, 2430±208 mg gallic acid equivalents (GAE) per 100 g dried sample and ascorbic acid equivalent antioxidant capacity (AEAC), 1140±85 mg AA/100 g. C. racemosa and K. alvarezzi displayed lower TPC and AEAC. C. racemosa had 144±22 mg GAE/100 g dried sample of TPC and 14.3±2.0 mg AA/100 g of AEAC, while K. alvarezzi had 115±35 mg/100 g dried sample of TPC and 37.8±16.8 mg AA/100 g of AEAC. In addition, P. antillarum displayed the highest reducing power, 15.7±2.6 mg GAE/g and highest chelating ability. C. racemosa and K. alvarezzi exhibited lower reducing power, 0.737±0.423 mg GAE/g and 0.561±0.269 mg GAE/g, and lower chelating ability. However, the AOA of these three seaweeds as assessed by BCB assay were equally high.  相似文献

The present study aimed to extract total phenolic compounds (TPC), total flavonoid compounds (TFC), and ascorbic acid (AA) from the fruit of rugosa rose (Rosa rugosa Thunb.) by ultrasound-assisted extraction (UAE), and to evaluate their antioxidant activities. UAE significantly increased the extract yield compared with that obtained using the conventional method. TPC, TFC, and AA were extracted, depending on the extraction conditions (temperature, time, and ethanol concentration), in the range of 50.73–96.69, 15.93–31.88, and 3.06–6.08 mg/g, respectively. TPC and TFC were effectively extracted at a relatively high temperature (50 °C) than AA was (30 °C). The solvent condition used to extract TPC, TFC, and AA was 50% ethanol. The UAE condition for the highest antioxidant activity was obtained 30 °C, 30 min, and 50% ethanol, which were the same condition for the highest AA extraction. Among the extracts, AA showed a strong correlation with antioxidant activity at p-value of 0.001.  相似文献

Antioxidant capacity (AC), total phenolic content (TPC) and total carotenoid content (TCC) in palm oils at various stages of the refining process from two technological modes were determined. The obtained mean FRAP and DPPH values for the methanolic extracts of palm oils from mode 1 (19.5–102.8 μmol TE/100 g and 18.8–103.0 μmol TE/100 g) were lower than for oils from mode 2 (25.6–134.8 μmol TE/100 g and 25.4–135.4 μmol TE/100 g). The total phenolics (4.1–12.4 mg GA/100 g) and carotenoids (0.18–45.8 mg/100 g) in the studied oils were correlated with their antioxidant capacities determined by FRAP and DPPH methods (r 0.6623–0.9878). During the refining process, for both technological modes resulted in a loss of AC by 80%, TPC by 26–55% and TCC by 99%. The bleaching step caused the highest losses of AC as determined by FRAP 41% and 46%, DPPH by 43% and 48%, while TPC loss was 45% and 23% and loss of carotenoids was 49% and 56%, in mode 1 and mode 2, respectively.  相似文献

While other extraction methods have been tempted, a microwave-assisted extraction (MAE) method coupled with the orthogonal array design was investigated for efficient extraction of the phenolic compounds in potato downstream wastes. Four parameters were examined for the MAE of the total phenolic content (TPC) and optimized at 60% ethanol, 80 °C, 2 min, solid-to-solvent ratio 1:40 (g/ml). The MAE was proven more efficient than the conventional solvent extraction by refluxing. The optimized model showed that the downstream wastes, both the supernatant and the residue contained high TPC, particularly the former (11.0 ± 0.26 mg GAE/g DW). The antioxidant activities (DPPH and FRAP) closely correlated with the TPC of the samples (r = 0.92–0.97). Chlorogenic acid and caffeic acid were found to be the predominant phenolic acids. The extracts of the downstream wastes from potato processing can be a promising candidate for functional foods and nutraceutical ingredients.  相似文献

Methanolic extracts of six cultivars of Portulaca oleracea were analyzed for their total phenol content (TPC) using the Folin–Ciocalteu method. The antioxidant activity was measured using the 1,1-diphenyl-2-picrylhydrazyl, ferric-reducing antioxidant power (FRAP) and β-carotene bleaching (BCB) assays. The iodine titration method was used to determine the ascorbic acid content (AAC). The TPC of the cultivars of P. oleracea ranged from 127 ± 13 to 478 ± 45 mg GAE/100 g of fresh weight of plant. There was good correlation between the TPC value and its AEAC, IC50 and FRAP values (r2 > 0.9) for all the cultivars. The AAC for the cultivars ranged from 38.5 ± 0.6 to 73.0 ± 17.5 mg/100 g. The TPC value of the common variety PO1, was the lowest compared to the ornamental cultivars (PO2–PO6). The BCB assay showed that all cultivars were capable of inhibiting lipid peroxidation and the inhibition power did not correlate with TPC value.  相似文献

Methanolic extracts from fresh leaves of five Etlingera species were screened for total phenolic content (TPC), antioxidant activity (AOA), and antibacterial activity. Analysis of TPC was done using the Folin–Ciocalteu method. Evaluations of AOA included 1,1-diphenyl-2-picrylhydrazyl free radical-scavenging ability, ferric-reducing antioxidant power (FRAP), ferrous-ion chelating (FIC) ability, and β-carotene bleaching (BCB) activity. Antibacterial activity was screened using the disc-diffusion method. Highest TPC, ascorbic acid equivalent antioxidant capacity (AEAC), and FRAP were found in leaves of E. elatior and E. rubrostriata. Leaves of E. maingayi, with the lowest TPC, AEAC, and FRAP, had the highest FIC ability and BCB activity. Ranking of TPC and AOA of different plant parts of E. elatior was in the order: leaves > inflorescences > rhizomes. Leaves of highland populations of Etlingera species displayed higher values of TPC and AEAC than those of lowland counterparts. Leaves of Etlingera species exhibited antibacterial activity against Gram-positive but not Gram-negative bacteria.  相似文献

To investigate the relationship between bioactive compounds and antioxidant property, total phenolic content (TPC), total flavonoid content (TFC), individual and total isoflavone content of soymilk and their correlations with oxygen‐radical‐absorbing capacity (ORAC), ferric reducing antioxidant power (FRAP) and 1,1‐diphenyl‐2‐picrylhydrazyl (DPPH) radical scavenging capacity values were evaluated and compared. Results showed that TPC, TFC, isoflavones (including concentration and profiles) and antioxidation activity significantly varied among ten tested soybean cultivars. Significant correlations were established between DPPH and TFC (= 0.553, < 0.01), DPPH and TPC (= 0.753, < 0.01), FRAP and TFC (= 0.599, < 0.01) and FRAP and TPC (= 0.616, < 0.01). Positive linear correlations were found between subtotal, total isoflavones and ORAC. Unlike other isoflavone monomers, aglycone isoflavones correlated positively with DPPH and FRAP significantly. DPPH well correlated with FRAP, whereas neither DPPH nor FRAP correlated with ORAC. TPC, TFC, individual and total isoflavone values are potentially useful for soymilk antioxidant activity assessment.  相似文献

The antioxidant capacity of twenty nine rapeseed varieties was determined by using ferric reducing antioxidant power (FRAP) and 2,2′-diphenyl-1-picrylhydrazyl (DPPH) methods. Mean FRAP (3190–6326 μmol Trolox/100 g) and DPPH (3194–6346 μmol Trolox/100 g) values for methanolic extracts of rapeseed cultivars did not differ significantly. Moreover, the total content of phenolics (756–1324 mg sinapic acid/100 g), glucosinolates (4.2–87.5 μmol/g, respectively), erucic acid (0.0–56.1%) and colour parameters of the studied rapeseed cultivars were analysed. Antioxidant capacity determined by FRAP and DPPH methods correlated significantly with total phenolic content (TPC) in rapeseed cultivars (r = 0.9332, 0.9339, p < 0.001). Also, significant, inverse correlations were found between antioxidant capacity, total phenolics and luminosity (L) or red colour intensity (a) of rapeseed cultivars. Principal component analysis (PCA) allowed the rapeseed varieties to be differentiated based on their antioxidant capacities, total amounts of phenolics, glucosinolates, erucic acid and colour parameters.  相似文献

The aim of this study was to evaluate the potency of polyphenolic extract from black radish (Raphanus sativus L. var. niger) root peel waste (PEBRRP) as an antioxidant food additive. PEBRRP was found a powerful antioxidant extract, in which had 172.9 ± 11.5, 462.72 ± 3.05, and 796.51 ± 10.4 mg TE per g dw of total antioxidant capacity (TAC) by CUPRAC, DPPH, and FRAP assays, respectively. The total phenolic content (TPC) and total flavonoid content (TFC) of PEBRRP were found as 305.51 ± 5.2 mg GAE per g dw and 171.58 ± 6.2 mg CAE per g dw, respectively. The ferulic acid (28.02 ± 4.5 mg per g dw) and epicatechin (19.82 ± 3.6 mg per g dw) were identified as the main polyphenols in undigested PEBRRP. The bioaccessibility of total phenolic and total flavonoid compounds of PEBRRP was found approximately 5%, and the antioxidant capacity of bioaccessible PEBRPP fraction was determined 6% in CUPRAC, 36% in DPPH and 47% in FRAP methods.  相似文献

An investigation into the effects of ethanol concentration (0–100%, v/v), extraction time (20–120 min) and extraction temperature (25–65 °C) on the extraction of phenolic antioxidants from mengkudu (Morinda citrifolia) was performed using a single-factor experiment. Total phenolic content (TPC) and total flavonoid content (TFC) assays were used for determination of phenolic compounds. Antioxidant capacity was evaluated by measuring the scavenging effect on 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) and 2,2′-diphenyl-1-picrylhydrazyl (DPPH) radicals. Experimental results showed that extraction conditions had significant effect on extraction of phenolic compounds and antioxidant capacities. The optimised conditions were 40% ethanol for 80 min at 65 °C, with values of 919.95 mg GAE/100 g DW for TPC, 472.73 mg CE/100 g DW for TFC, 791.71 μmol TEAC/100 g DW for ABTS and 1928.5 μmol TEAC/100 g DW for DPPH. TPC was significantly correlated with DPPH under the effects of ethanol concentration (r = 0.932) and extraction time (r = 0.938).  相似文献

Chemical, physicochemical and functional properties of fibrous materials from freeze dried (FDPSP) and oven dried (ODPSP) Parkia speciosa pod (PSP) were evaluated in order to examine the potential applications in food. Preliminary screening of PSP using scanning electron microscope showed that PSP consisted of starches and fibres. Proximate analysis showed that both samples consisted of 4.3–5.2% of moisture, 9.5–10.7% of protein, 0.1–0.2% of fat, 0.01% of ash, 16.5–16.8% of crude fibre and 68.3–68.7% of carbohydrates. However, different functional properties were shown in these samples. FDPSP gave a lighter colour compared to ODPSP. Higher values of solubility (30.8%), water- and oil-holding capacities (3.72 and 1.55 g/g, respectively) were found in FDPSP. Conversely, higher emulsifying activity of 62.7% with high emulsion stability of 99.9% was shown in ODPSP. Higher antioxidative properties were also shown in FDPSP which consist TPC of 110.0 mg gallic acid equivalent/g sample and TFC of 8.5 mg pyrocatechol equivalents/g sample. These extracts (pre-diluted 50×) gave %DPPHsc, %ABTSsc and FRAP values of 65.3%, 77.4% and 1.9 mM FeSO4, respectively. Therefore, it was suggested that PSP has the potential in producing functional fibres that could be imparted into the food system.  相似文献

Aqueous extracts from 20 Malaysian edible plants were screened for total phenolic content (TPC), antioxidant activity and genotoxic effects on freshly isolated human lymphocytes. Ferric reducing antioxidant power (FRAP) and 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging assays were performed to determine the antioxidant activity of the extracts. Single-cell gel electrophoresis (SCGE) or comet assay was carried out to determine the level of DNA damage in human lymphocytes. Of the 20 plant aqueous extracts tested, two exerted more than 50% DNA strand breaks (severe damage), nine exerted 25–50% strand breaks (moderate damage) and nine exerted <25% strand breaks (mild damage). Strong positive correlations between the extent of DNA damage and FRAP level (r = 0.816), DNA damage and TPC (r = 0.830) and DNA damage and DPPH radical scavenging activities (r = 0.859) were observed. It is evident from this study that plants rich in antioxidants have greater genotoxic effect.  相似文献
