The objective of this study was to determine the effect of changing viscosity and glass transition temperature in the continuous phase of nanoemulsion systems on subsequent stability. Formulations comprising of β-casein (2.5%, 5%, 7.5%, and 10% w/w), lactose (0% to 20% w/w), and trehalose (0% to 20% w/w) were generated from Design of Experiments (DOE) software and tested for glass transition temperature and onset of ice-melting temperature in maximally freeze-concentrated state (T(g) ' & T(m) '), and viscosity (μ). Increasing β-casein content resulted in significant (P < 0.0001) increases in viscosity and T(m) ' (P= 0.0003), and significant (P < 0.0001) decreases in T(g) '. A mixture design was used to predict the optimum levels of lactose and trehalose required to attain the minimum and maximum T(g) ' and viscosity in solution at fixed protein contents. These mixtures were used to form the continuous phase of β-casein stabilized nanoemulsions (10% w/w sunflower oil) prepared by microfluidization at 70 MPa. Nanoemulsions were analyzed for T(g) ' & T(m) ', as well as viscosity, mean particle size, and stability. Increasing levels of β-casein (2.5% to 10% w/w) resulted in a significant (P < 0.0001) increase in viscosity (5 to 156 mPa.s), significant increase in particle size (P= 0.0115) from 186 to 199 nm, and significant decrease (P= 0.0001) in T(g) ' (-45 to -50 °C). Increasing the protein content resulted in a significant (P < 0.0001) increase in nanoemulsion stability. A mixture DOE was successfully used to predict glass transition and rheological properties for development of a continuous phase for use in nanoemulsions.  相似文献

采用酶法提取小米麸皮水溶性膳食纤维(soluble dietary fiber,SDF),并使SDF在弱碱性条件下与三价铬离子(Cr3+)发生配位反应,生成SDF-Cr(III)配合物。运用扫描电子显微镜、傅里叶变换红外光谱、原子力显微镜对小米麸皮SDF和SDF-Cr(III)配合物进行结构表征;运用常温凝胶渗透色谱及高效液相色谱(high performance liquid chromatography,HPLC)比较改性前后小米麸皮SDF分子质量分布及其单糖组成;通过测定1,1-二苯基-2-三硝基苯肼自由基、2,2’-联氮-双-3-乙基苯并噻唑啉-6-磺酸阳离子自由基、羟自由基清除率和总抗氧化能力,比较改性前后SDF体外抗氧化活性。结果表明:小米麸皮SDF-Cr(III)配合物表面结构疏松,呈现出多孔状;岛屿状突起聚集度有所下降,分子间变得更加分散。两种SDF官能团差异较大,傅里叶变换红外光谱分析结果显示小米麸皮SDF-Cr(III)配合物活性官能团(羟基、酯基、羰基等)伸缩振动减弱,特征吸收峰位置发生蓝移,在530.27 cm-1处有Cr—O伸缩振动特征吸收峰,说明SDF与Cr3+结合形成配合物。凝胶渗透色谱分析结果显示,SDF的数均分子质量(mn)为0.195×104 Da、重均分子质量(mw)为0.940×104 Da、分布宽度指数D为4.81;SDF-Cr(III)的mn为0.742×104 Da、mw为4.708×104 Da、分布宽度指数D为6.35。由HPLC分析结果可知,SDF-Cr(III)单糖相对含量高于SDF。体外抗氧化实验结果表明,SDF-Cr(III)比SDF具有显著的体外抗氧化性。综上,小米麸皮SDF-Cr(III)配合物具有一定的抗氧化能力,具备开发成为具有降糖功效保健品的潜力。  相似文献

This study involves the evaluation of the effect of certain stabilizers, that is, citric acid (CT), tartaric acid (TA) and boric acid (BA) on the degradation of ascorbic acid (AH(2) ) in oil-in-water cream formulations exposed to the UV light and stored in the dark. The apparent first-order rate constants (0.34-0.95 × 10(-3) min(-1) in light, 0.38-1.24 × 10(-2) day(-1) in dark) for the degradation reactions in the presence of the stabilizers have been determined. These rate constants have been used to derive the second-order rate constants (0.26-1.45 × 10(-2) M(-1) min(-1) in light, 3.75-8.50 × 10(-3) M(-1) day(-1) in dark) for the interaction of AH(2) and the individual stabilizers. These stabilizers are effective in causing the inhibition of the rate of degradation of AH(2) both in the light and in the dark. The inhibitory effect of the stabilizers is in the order of CT > TA > BA. The rate of degradation of AH(2) in the presence of these stabilizers in the light is about 120 times higher than that in the dark. This could be explained on the basis of the deactivation of AH(2) -excited triplet state by CT and TA and by the inhibition of AH(2) degradation through complex formation with BA. AH(2) leads to the formation of dehydroascorbic acid (A) by chemical and photooxidation in cream formulations.  相似文献

Dry beans are rich sources of dietary fiber and phytochemicals such as flavonoids and phenolics. In the current study, we determined the effects of two processing methods (soaking and toasting) on total phenolics, flavonoids, proanthocyanidin and antioxidant potential by 2,2-diphenyl-1-picrylhydrazyl (DPPH) activity and ferric reducing antioxidant potential (FRAP) in selected dry beans (red kidney beans (K), black-eyed peas (B), pinto beans (P) and soy beans (S)).Total phenolics (mg/g dwb) expressed as gallic acid equivalents (GAE), total flavonoids (mg/g dwb) as catechin equivalents (CE) and proanthocyanidin expressed as leucocyanidin equivalent (mg LE/g) ranged from 3.42 to 7.21, 0.61 to 0.84 and 0.51 to 3.13 in raw beans; 3.58 to 6.94, 0.19 to 0.99 and 0.43 to 3.13 in soaked beans and 4.55 to 9.52, 0.23 to1.00 and 0.20 to 3.25 in toasted beans, respectively.FRAP (μg/g) in raw, soaked and toasted dry beans ranged from 0.00097 to 0.00424 while DPPH(T30) (%) ranged from 43.9 to 62.61.Our results indicate that processing methods (soaking and roasting) influenced total phenolic, flavonoid and antioxidant contents (DPPH, FRAP) in selected dry beans.  相似文献

French dairy herds (n = 534) were enrolled in the National 'Zero Mastitis Objective' Program to highlight management practices characterizing very low somatic cell score (SCS) herds. The herds studied were stratified into 2 groups. The first group (LOW) included herds within the first 5 percentiles and the second group (MED) herds within the 50 to 55 percentiles of herds on the basis of mean SCS for the 36 mo preceding the program. Potential explanatory variables, collected through questionnaire surveys, were analyzed using multistep logistic regression models. Twenty-six variables were significant factors in the final models, in which 18 were considered as primary factors for very low SCS. The probability for a herd belonging to the LOW group was associated with: (1) regular use of teat spraying; (2) herdsman precise in his techniques; (3) less than 1 person-year used at activities other than dairy herd; (4) teat dipping after mammary infusion at dry off; (5) heifers kept in a calving pen around parturition; (6) cows locked in feed-line lockups after milking; (7) dry cows with prepartum Ca restriction; (8) heifers on a nondamp pasture; (9) cows culled when at least one damaged teat; (10) heifers at pasture not drinking water from a river; and (11) disinfecting teat ends with alcohol before intramammary infusion at dry off. The probability for a herd belonging to the MED group was associated with: (1) milking cows housed in a straw yard; (2) checking heifers for mastitis only beginning at 2-wk prepartum; (3) no mastitis treatment when at least one clot was observed in milk at successive milkings; (4) distance of herdsman's house to cowshed >300 m; (5) only dirty teats washed before milking; (6) free access of cows from pasture to cowshed during bad weather; and (7) more than 18% of spring calvings. The variables associated with very low SCS should be applied as part of a thorough mastitis-control program adapted to each herd.  相似文献

The enhanced removal of carbon tetrachloride (CCl4), tetrachloroethene (C2Cl4), and trichloroethene (C2HCl3) by chloride green rust (GR(Cl)) in the presence of copper ions was investigated. X-ray powder diffraction (XRPD) and X-ray photoelectron spectroscopy (XPS) were used to characterize the crystallization and chemical speciation, respectively, of the secondary mineral phases produced in the GR(Cl)-Cu(II) system. The addition of Cu(II) to GR(Cl) suspensions resulted in enhanced dechlorination of the chlorinated hydrocarbons examined in this study. The degradation reactions followed pseudo-first-order kinetics and the pseudo-first-order rate constant (k(obs)) for CCl4 (20 microM) removal by GR(CI) at pH 7.2 was 0.0808 h(-1). Addition of 0.5 mM Cu(II) completely dechlorinated CCl4 within 35 min, and the k(obs) was 84 times greater than that in the absence of Cu(II). Chloroform (CHCl3), the major chlorinated product in CCl4 dechlorination, accumulated at a concentration up to 13 microM in the GR(Cl) system alone, but was completely dechlorinated within 9 h in the GR(Cl)-Cu(II) suspension. Also, rapid removal of C2Cl4 and C2HCl3 by GR(Cl) was observed when Cu(II) was added. The k(obs) values for the removal of chlorinated ethenes were 4.7-7 times higher than that obtained in the absence of Cu(II). In addition, the k(obs) for PCE removal increased linearly with respect to Cu(II) concentrations in the range from 0.1 to 1.0 mM. Addition of Cu(II) at a concentration higher than 1.0 mM decreased the k(obs) for the removal of both C2Cl4 and C2HCl3 due to the decrease in structural Fe(II) concentration in GR(Cl) and the changes in redox potentials and pH values. Moreover, the highest removal efficiency and rate of C2Cl4 was obtained at near-neutral pH when Cu(II) was added into the GR(Cl) suspension. XPS and XRPD results showed that the Fe(II) in the GR(Cl) suspension could reduce Cu(II) to both Cu(I) and metallic Cu. These findings are relevant to the better understanding of the role of abiotic removal of chlorinated hydrocarbons during remediation and/or natural attenuation in iron-reducing environments.  相似文献

Studies were conducted to evaluate the effect of heat treatment on polyphenol oxidase (PPO) activity of whole-wheat flour (WWF) as well as its chapatti making quality. Whole wheat flour samples were heat treated through various means such as (i) dipping in boiling water (BW) for 20, 30, 40, 50, and 60 min; (ii) inpack heating under pressure (PH) at 0.352 kg/cm2 for 4, 6, 8, 10, and 12 min; (iii) microwave heat treatment (MW) at 900 Watt, 2450 MHz for 20, 40, 60, 80, and 100 s. Studies showed that the heat treatment effectively reduced PPO level in whole wheat flour; although, it had adverse effect on the dough-making quality. Based on textural analysis of dough and chapatti as well as sensory scores of chapatti, the conditions for each of the treatments were optimized, i.e., (i) Dipping in boiling water (BW) for 30 min; (ii) inpack heating under pressure (PH) at 0.352 kg/cm2 for 10 min; and (iii) microwave heating (MW) for 80 s. A maximum reduction (71.2%) in PPO activity of WWF using microwave treatment could be achieved followed by PH (56.9%) and BW (38.3%). The changes in colour of unbaked chapattis (flattened circular dough, diameter 150.0 mm and thickness 2.0 mm) and changes in quality of baked chapattis were measured to assess the effectiveness of the heat treatment. The L-value (lightness) decreased from 65.2 to 55.8, 65.7 to 58.3, 65.9 to 61.4, and 64.8 to 49.1 in case of BW, PH, MW treated, and control samples, respectively during the 72 h of storage under refrigeration temperature (5–6°C).  相似文献

There has been substantial recent interest in shortening dry periods; however, the effects of this management change on reproduction have not been adequately evaluated. Holstein cows (n = 58) were assigned in a randomized block design to 1 of 3 treatments: 1) traditional (T) dry period (∼56 d) in which cows were fed a low energy diet from 56 to 29 d prepartum followed by a moderate energy diet for 28 d; 2) shortened (S) dry period (∼28 d) in which cows were fed continuously a high energy diet; or 3) no planned (N) dry period in which cows were fed continuously a high energy diet. All cows received a high energy lactation diet after calving. Ovaries were evaluated by ultrasound and blood samples collected 3 times weekly beginning at d 6 or 7 postpartum until 7 d after second ovulation. Average days from calving until first detection of a 10-mm follicle were fewer in N (8.0 d) and S (8.9 d) than in T (10.5 d) cows. Time from calving to first ovulation was earlier in N (13.2 d) than in S (23.8 d) and T (31.9 d) cows. A greater percentage of follicles of the first follicular wave ovulated in N (89%; 16/18) than in T (42%; 8/19), with S (62%; 13/21) cows being intermediate. Double ovulation rate at the first ovulation was greater in T (61%) than N (16%), with S (35%) intermediate. No difference was detected in double ovulation rate at second ovulation (13/56). Number of cows with persistent corpus luteum (>30 d; 15/56) was not different among groups; however, short luteal phases were greater in N (28%; 5/18) than S (0%; 0/20) cows. Days to first artificial insemination were fewer in N (69.4 d) and S (68.0 d) than in T (75.0 d). First-service conception rate was greater in N (55%; 11/20) than in T (20%; 4/20), with S (26%; 6/23) cows being intermediate. Days open in pregnant cows were fewer in N (93.8 d) than in T (145.4 d), with S (121.2 d) cows being intermediate. Thus, shortening or eliminating the dry period leads to earlier postpartum ovulation and the results highlight the need for future large field studies to accurately evaluate the effect of dry period length on reproductive performance of lactating dairy cows.  相似文献

An anaerobic, gram positive, spore-forming bacterium Clostridium sp., common in soils and wastes, capable of reduction of Fe(III) to Fe(II), Mn(IV) to Mn(II), Tc(VII) to Tc(IV), and U(VI) to U(IV), reduced Pu(IV) to Pu(III). Addition of 242Pu (IV)-nitrate to the bacterial growth medium at pH 6.4 resulted in the precipitation of Pu as amorphous Pu(OH)4 due to hydrolysis and polymerization reactions. The Pu (1 x 10(-5) M) had no effect upon growth of the bacterium as evidenced by glucose consumption; carbon dioxide and hydrogen production; a decrease in pH of the medium from 6.4 to 3.0 due to production of acetic and butyric acids from glucose fermentation; and a change in the Eh of the culture medium from +50 to -180 mV. Commensurate with bacterial growth, Pu was rapidly solubilized as evidenced by an increase in Pu concentration in solution which passed through a 0.03 microm filtration. Selective solvent extraction of the culture by thenoyltrifluoroacetone (TTA) indicated the presence of a reduced Pu species in the soluble fraction. X-ray absorption near edge spectroscopic (XANES) analysis of Pu in the culture sample at the Pu LIII absorption edge (18.054 keV) showed a shift of -3 eV compared to a Pu(IV) standard indicating reduction of Pu(IV) to Pu(III). These results suggestthat, although Pu generally exists as insoluble Pu(IV) in the environment, under appropriate conditions, anaerobic microbial activity could affect the long-term stability and mobility of Pu by its reductive dissolution.  相似文献

Effects of phosphate on uranium(VI) adsorption to goethite-coated sand   总被引:2,自引:0,他引:2  
U(VI)-phosphate interactions are important in governing the subsurface mobility of U(VI) in both natural and contaminated environments. We studied U(VI) adsorption on goethite-coated sand (to mimic natural Fe-coated subsurface materials) as a function of pH in systems closed to the atmosphere, in both the presence and the absence of phosphate. Our results indicate that phosphate strongly affects U(VI) adsorption. The effect of phosphate on U(VI) adsorption was dependent on solution pH. At low pH, the adsorption of U(VI) increased in the presence of phosphate, and higher phosphate concentration caused a larger extent of increase in U(VI) adsorption. Phosphate was strongly bound by the goethite surface in the low pH range, and the increased adsorption of U(VI) at low pH was attributed to the formation of ternary surface complexes involving both U(VI) and phosphate. In the high pH range, the adsorption of U(VI) decreased in the presence of phosphate at low total Fe concentration, and higher phosphate concentration caused a larger extent of decrease in U(VI) adsorption. This decrease in U(VI) adsorption was attributed to the formation of soluble uranium-phosphate complexes. A surface complexation model (SCM) was proposed to describe the effect of phosphate on U(VI) adsorption to goethite. This proposed model was based on previous models that predict U(VI) adsorption to iron oxides in the absence of phosphate and previous models developed to predict phosphate adsorption on goethite. A postulated ternary surface complex of the form of (>FePO4UO2) was included in our model to account for the interactions between U(VI) and phosphate. The model we established can successfully predict U(VI) adsorption in the presence of phosphate under a range of conditions (i.e., pH, total phosphate concentration, and total Fe concentration).  相似文献

Nitroaromatic pesticides (NAPs) are hydrophobic contaminants that can accumulate in sediments by the deposition of suspended solids from surface waters. Fe(II) and dissolved organic matter (DOM), present in suboxic and anoxic zones of freshwater sediments, can transform NAPs in natural systems. We studied the reduction of pentachloronitrobenzene (PCNB) to pentachloroaniline (PCA) in controlled studies using Fe(II) and surface water DOM isolates from Pony Lake, Antarctica, and Suwannee River, GA, in unfiltered and 0.45 microm filtered solutions. We observed rapid reduction of PCNB to PCA in the presence of Fe(II) and DOM (t(1/2) approximately = 30 min to 4 h) and very limited reduction in DOM-only systems. DOM in unfiltered systems inhibited iron colloid formation and potentially limited the formation of reactive Fe(ll)-iron colloid surface complexes, causing reductive transformation in Fe(II)-DOM media to be slower in some cases relative to Fe(ll)-only controls. Conversely, in 0.45 microm filtered solutions, PCNB reduction in Fe(III)-DOM media was faster than the Fe(II)-only controls, suggesting that DOM enhances the reductive capacity of Fe(ll) in the absence of iron colloids. This work shows that DOM may significantly affect the reactivity of Fe(ll) toward NAPs under suboxic and anoxic conditions in natural wetland sediments.  相似文献

ABSTRACT:  Lactobacillus acidophilus 4461, L. acidophilus 4962, L. casei 290, and L. casei 2607 were used to hydrolyze isoflavone glycosides (IG) to biologically active forms—isoflavone aglycones (IA)—in soymilk (SM) prepared from soy protein isolate (SPI) and soymilk supplemented with 0.5% (w/v) of lactulose (SML). L. acidophilus 4461 utilized the highest level of lactulose (3.01 mg/mL) and L. acidophilus 4962 utilized the least (0.86 mg/mL) at 24 h of incubation. The pH values decreased to 4.00 to 5.00 in SML, while they remained relatively high (6.15 to 6.36) in SM. Supplementation with lactulose significantly ( P < 0.05) enhanced the viable counts of all the 4 Lactobacillus strains. At the end of incubation, the viable counts of Lactobacillus ranged from 8.08 to 8.25 log CFU/mL in SML compared to 6.99 to 7.11 log CFU/mL in SM. Supplementation with lactulose increased the biotransformation of IG to IA after 6 h of incubation. The presence of lactulose in the medium enhanced the biotransformation level of IG to IA by Lactobacillus up to 21.9%. The hydrolysis level of malonyl genistin and acetyl genistin in SML was much higher than in SM by all the 4 probiotic organisms. The biotransformation of IG to IA occurred rapidly during the 1st 12 h of incubation in both SML and SM. Among the 4 Lactobacillus strains, L. acidophilus 4461 biotransformed the highest level (88.8%) of IG to IA in SML compared to 68.2% in SM after 24 h of incubation.  相似文献

The objective of this study was to determine the impact of different slaughter procedures on animal welfare and meat quality. Before slaughter in a dip-lift, one-gondola system, 460 pigs were exposed to an atmosphere containing either 80% or 90% CO(2) for 70 or 100s, and at the longer exposure time with stun-to-stick intervals of either 25-35 or 40-50s. Clinical parameters (reflexes, catecholamines and lactate) showed deficiencies in animal welfare after stunning with 80% CO(2) for 70 and 100s, with an interval of 40-50s, and with 90% CO(2) for 70s. Stunning with 80% CO(2) for 70 or 100s always induced stress, as indicated by higher lactate levels, and reduced meat quality as indicated by low pH(24) values (5.4 in Musculus longissimus and 5.5 in Musculus semimembranosus) and low impedance (Py(24)) values, especially in combination with the longer stun-to-stick interval (40-50s). Stunning with 80% or 90% CO(2) in a dip-lift system was found to be acceptable for animal welfare (percentage of clinical reflexes) only in combination with the longer exposure time of 100s and the shorter stun-to-stick times of 25-35s. When 90% CO(2) was used, the longer stun-to-stick interval of 40-50s also gave results acceptable with regard to animal welfare. Generally, in comparison to 80% CO(2) stunning meat quality was superior (higher pH(24) and Py(24) values) after stunning with 90% CO(2).  相似文献

Oestradiol (E(2)) is a key hormone in the regulation of reproductive processes. The aims of this work were a) to examine the distributions of oestrogen receptor α (ERα) and ERβ in the neurons of the superior mesenteric ganglion (SMG) in the oestrus stage by immunohistochemistry, b) to demonstrate whether E(2) in the SMG modifies progesterone (P(4)), androstenedione (A(2)) and nitrite release in the ovarian compartment on oestrus day and c) to demonstrate whether E(2) in the ganglion modifies the activity and gene expression in the ovary of the steroidogenic enzymes 3β-hydroxysteroid dehydrogenase (3β-HSD) and 20α-hydroxysteroid dehydrogenase (20α-HSD). The ex vivo SMG-ovarian nervous plexus-ovary system was used. E(2), tamoxifen (Txf) and E(2) plus Txf were added in the ganglion to measure ovarian P(4) release, while E(2) alone was added to measure ovarian A(2) and nitrites release. Immunohistochemistry revealed cytoplasmic ERα immunoreactivity only in the neural somas in the SMG. E(2) increased ovarian P(4) and A(2) release at 15, 30 and 60?min but decreased nitrites. The activity and gene expression of 3β-HSD increased, while the activity and gene expression of 20α-HSD did not show changes with respect to the control. Txf in the ganglion diminished P(4) release only at 60?min. E(2) plus Txf in the ganglion reverted the effect of E(2) alone and the inhibitory effect of Txf. The results of this study demonstrate that ERα activation in the SMG has an impact on ovarian steroidogenesis in rats, thus providing evidence for the critical role of peripheral system neurons in the control of ovarian functions under normal and pathological conditions.  相似文献

Barrows (n=164) and gilts (n=249) from crosses of a Pietrain homozygous halothane recessive (Pi nn) and two Pietrain homozygous dominant (Pi NN-a and Pi NN-b) sire lines with Landrace×Large White NN sows, were used to study the effect of terminal sire and pre-slaughter treatment on meat quality and animal welfare. The pigs from each of the two farms where they were finished were delivered to the abattoir in two batches differing in the pre-slaughter conditions. A total of 90 pigs (54 NN and 36 Nn) were assigned to a long pre-slaughter treatment (6 h transport and 14.5 h lairage) and 89 (57 NN and 33 Nn) to a short pre-slaughter treatment (4.5 h transport and 2.5 h lairage) in Farm 1, and 118 (65 NN and 53 Nn) to the long (7 h transport and 14 h lairage) and 114 (66 NN and 48 Nn) to the short pre-slaughter treatment (1.5 h transport and 2 h lairage) in Farm 2. In Farm 1, heart rate of 3 NN and 3 Nn gilts was recorded throughout loading and transport and blood samples from 5 NN and 5 Nn were collected before loading and after transport to measure cortisol, creatine phosphokinase (CPK) and lactate dehydrogenase (LDH). Carcasses were classified and commercial cutting was carried out. At 24 h, meat quality was assessed on the Longissimus thoracis muscle by measuring electrical conductivity (PQM), colour (Minolta CR 200 and Japanese scale), pHu and drip losses. Halothane carriers showed a higher mean heart rate and higher increase in CPK levels (P<0.05) after transport in the short pre-slaughter treatment than halothane free pigs. No effect was observed in cortisol or LDH values. Pi NN-a sired pigs had a higher live and carcass weight (P<0.001) and loin depth (P<0.05), but lower killing out percentage (P<0.01) and leg yield (P<0.01) compared with the progeny of the other two terminal sires. Gilts were leaner (P<0.001), had a higher killing out percentage (P<0.001) and higher yields of primal cuts (P<0.001) compared with barrows. Pi nn sired pigs had poorer meat quality (higher PQM values in both farms, P<0.01) than Pi NN-a sired pigs. Long pre-slaughter treatment resulted in darker meat (P<0.01) in both farms and in higher pHu (P<0.001) in Farm 1 than short pre-slaughter treatment. Conversely, pigs subjected to the short pre-slaughter treatment showed higher PQM values (P<0.01) in Farm 1 and higher PSE percentage (P<0.05) in both farms compared to the ones subjected to the long pre-slaughter treatment. These results suggest that Pietrain halothane free sire lines could produce similar results on carcass quality to halothane carriers, without compromising meat quality and welfare.  相似文献

The free radical scavenging activities of three flavonoids (quercetin, rutin and catechin) and four hydroxycinnamic acids (caffeic, ferulic, sinapic, and chlorogenic acids) were evaluated using both oxygen radical absorbance capacity (ORAC) and lipid peroxidation inhibition capacity (LPIC) assays. The cytoprotective effects of these compounds were also measured by the degree of protection against H(2)O(2)-induced damage of human Jurkat cells. All compounds exhibited protection against H(2)O(2)-mediated cytotoxicity in a dose-dependent manner. The concentrations required to result in a 50% reduction in cell death (EC(50) value) were calculated from their dose-response curves. These ranged from 0.15-2.65 microM. Overall, the four hydroxycinnamic acids tested were less effective than the three flavonoids, and of all compounds tested, quercetin offered the strongest protection against H(2)O(2)-induced cell death. A comparison of the results showed that the ability to inhibit peroxidation of lipids in a liposomal system (LPIC) correlated well with the cytoprotective activities (EC(50)), but not with the ability to protect an aqueous fluorescent substrate in the ORAC assays. The results suggest that the behavior of antioxidants in a liposomal membrane is to some extent similar to the mechanism involved in the protection of living cells from oxidative damage.  相似文献

Synopsis The aim of this study was to develop hydrogel patch using crosslinked chitosan-starch as polymeric matrix for controlling the release of the natural alpha-hydroxy acid (AHA) contained in the extract of tamarind's fruit pulp. The chitosan (MW 100 000) was blended with corn, tapioca or rice starch in various ratios and then crosslinked with glutaraldehyde. The physical characteristics, mechanical resistance, bio-adhesion property and surface morphology of the prepared hydrogel patches with and without the extract were investigated. The release patterns of the hydrogel patches containing the extract were investigated by measuring the amount of tartaric acid, a major AHA present in the tamarind's fruit pulp extract, accumulated in the receptor medium of the vertical diffusion cell at various time intervals over a period of 6 h. The results indicated that the formulations of chitosan : corn starch 4.5 : 0.5 with glutaraldehyde 0.02% w/w (C(4.5)C(0.5)G(0.02)) or 0.04% w/w (C(4.5)C(0.5)G(0.04)), chitosan : tapioca starch 4.5 : 0.5 with glutaraldehyde 0.04% w/w (C(4.5)T(0.5)G(0.04)) or 0.05% w/w (C(4.5)T(0.5)G(0.05)), and chitosan : rice starch 4.5 : 0.5 with glutaraldehyde 0.04% w/w (C(4.5)R(0.5)G(0.04)) and chitosan : rice starch 4.0 : 1.0 with glutaraldehyde 0.03% w/w (C(4.0)R(1.0)G(0.03)) provided the flexible and elastic patches with good bio-adhesive property. The tensile strength values ranged from 5 to15 N mm(-2) and the elasticity ranged from 30 to 60%. The addition of the extract in these formulations significantly increased the tensile strength values of the obtained patches. The patch of C(4.0)R(1.0)G(0.03) formulation containing the extract showed relatively highest porosity, corresponding to its highest amount (12.02 +/- 0.33 mg) and rate (0.452 +/- 0.012 mg mm(-2) min(-1/2)) of tartaric acid released. The amounts of tartaric acid released from the developed hydrogel patches were proportional to a square root of time (Higuchi's model), particularly the release from C(4.0)R(1.0)G(0.03) (R(2), 0.9978 +/- 0.0020) and C(4.5)R(0.5)G(0.04) (R(2), 0.9961 +/- 0.0024) patches.  相似文献

Soluble dietary fibre with low viscosity has the potential to deliver acceptable mouthfeel and texture when included in the diet in a significant amount to show health benefits. Soluble flaxseed gum (SFG) was reported to have low viscosity, thus has potential in applications as a fibre fortifier. In the present work, SFG extracted from flaxseed hulls was fractionated into a neutral fraction gum (NFG) and an acidic fraction gum (AFG) using ion exchange chromatography. The protein content in SFG (11.8%) and AFG (8.1%) were completely removed by protease to obtain two more protein-free fractions, SFGnP and AFGnP; NFG contained no protein. The uronic acid content in NFG (1.8%) was mostly eliminated, whereas in AFC increased to 38.7%. NFG consisted of high molecular weight (MW) (1470 kDa) arabinoxylans and exhibited pseudoplastic flow behaviour; whereas AFG was mainly composed of rhamnogalacturonans with a higher MW fraction (1510 kDa) and a lower MW fraction (341 kDa) and showed Newtonian flow behaviour. The ranking of intrinsic viscosities (mL g−1) in decreasing order was: SFG (446.0) > NFG (377.5) > AFG (332.5). AFG was expected to have higher chain flexibility for its lower value of Huggins constant (0.16) compared to that of NFG (0.54) and SFG (0.48). In comparison with SFGnP, AFGnP and NFG, SFG and AFG showed better surface activities and emulsifying stabilities due to the presence of protein in both fractions.  相似文献

The concentrations of fatty acids were measured in total lipids, triacyglycerol and phospholipid fractions of intramuscular fat (IMF) from the Longissimus dorsi (LD) muscle of 10 lambs reared to approximately 30kg live weight on natural pasture with their dams. Fatty acid composition was also measured in 25 (five of each) Semitendinosus (ST), Semimembranosus (SM), Rectus femoris (RF), Gluteus (GLU) and Tensor fascia latea (TFL) muscles. Intramuscular fat percentages were similar for all muscles. Aspects of the fatty-acid patterns of relevance to human nutrition tended to favor the leg muscles with lower saturated fatty acids (SFA %), n-6/n-3 fatty acid ratios (p<0.01) and higher concentrations of the conjugated linoleic acid (CLA) (p<0.05). The estimated fatty acid concentrations (mg/100g of meat) showed higher contribution of arachidonic (C20:4 n-6), eicosapentanoic (C20:5 n-3), docosapentanoic (C22:5 n-3) and docosahexanoic (C22:6 n-3) acids in leg compared to LD lipids.  相似文献

Metal ion sorption can be significantly impacted by the presence of other solutes or complexing species. In this research, macroscopic sorption studies were conducted to evaluate the effect of strongly sorbing Se(IV) and weakly sorbing Se(VI) oxyanions on cobalt(II) sorption to gamma-Al2O3. Se(IV) was found to significantly alter Co(ll) sorption as a function of Co(II) surface coverage, while Se(VI) was found to have no effect on Co(II) sorption. Under low Co(II) surface loadings (<0.1 micromol/m2), Se(IV) increased Co(II) sorption as a function of the Se(IV) coverage. At low Se(IV) surface coverages, no change in Co(II) sorption was detectable, while at high Se(IV) loadings Co(II) sorption was significantly increased. The increase in Co(ll) sorption in the bisorbate systems can be explained by either an electrostatic enhancement mechanism or byternary complex formation. Se(IV) decreased Co(II) sorption at high Co(ll) surface loadings (>0.5 micromol/m2) where coprecipitation of Co(II) and A(III) in the form of layered double hydroxides (LDH) is expected to be the dominant sorption mechanism for the single-sorbate case. The extent of the Co(ll) sorption reduction in Co(III)/Se(IV) bisorbate systems compared to the corresponding single-sorbate systems increased with increasing Co(II) surface coverage. The rate of Co(II) desorption was reduced in the presence of Se(IV) compared to the single-sorbate case, indicating a direct interaction between Co(II) and Se(IV). A reaction between Co(II) and Se(IV) is further supported by an increase in Se(IV) sorption in the same bisorbate samples where Co(II) sorption is decreased. Thus, the macroscopic data indicates Se(IV) may be altering the mechanism of Co(II) sorption, potentially forming a ternary surface complex or different surface precipitate.  相似文献
