Antioxidant activity (AA), total phenolic content, and reducing power of the crude extract, fractions, and subfractions derived from a red alga, Polysiphonia urceolata, were evaluated and determined. The antioxidative activity was measured using the α,α-diphenyl-β-picrylhydrazyl (DPPH) radical scavenging assay and the β-carotene–linoleate assay systems, and compared with that of butylated hydroxytoluene (BHT), gallic acid (GA), and ascorbic acid (AscA). The results showed that the crude extract and the ethyl acetate-soluble fraction exhibited higher AA than BHT in the DPPH assay model, at all of four concentration levels tested (from 0.4 to 50 μg/ml), while, in the β-carotene–linoleate assay system, the crude extract and the ethyl acetate-soluble fraction exhibited similar or, in most cases, higher AA than GA and AscA at the same concentrations (from 10 to 200 μg/ml). The ethyl acetate-soluble fraction was further fractionated into seven subfractions F1–F7 by silica gel vacuum liquid chromatography. F1 was found to be the most effective subfraction in both assay systems. The total phenolic content and reducing power were determined using the Folin–Ciocalteu and the potassium ferricyanide reduction methods, respectively. Statistical analysis indicated a significant association between the antioxidant potency and total phenolic content as well as between the antioxidant potency and reducing power.  相似文献

The methanolic fresh leaf extracts of Macaranga gigantea, Macaranga pruinosa, Macaranga tanarius and Macaranga triloba were screened for their antioxidant properties (AOP), tyrosinase inhibition and antibacterial activities. Total phenolic content (TPC), 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging, ferric-ion reducing power (FRAP), ferrous-ion chelating (FIC) and lipid peroxidation inhibition (LPI) activities were used to evaluate the AOP. Modified 3,4-dihydroxy-L-phenylalanine (L-DOPA) method was used to determine tyrosinase inhibition activity, whereas antibacterial activity was determined using the disc-diffusion technique. TPC screening of the same species from different collection sites showed no significant difference between sites. M.triloba showed the highest ascorbic acid equivalent antioxidant activity (AEAC), FRAP and LPI values. M. tanarius, which showed the lowest TPC, AEAC, FRAP and LPI activities, exhibited the best FIC activity. M. pruinosa showed the best tyrosinase inhibition activity, whereas M. triloba showed the best antibacterial activity against Gram-positive bacteria species, with minimal inhibition dosage (MID) values as low as 10 μg/disc.  相似文献

This study was designed to examine the in vitro antioxidant activities of the methanol extracts of six Salvia species [Salvia caespitosa Montbret & Aucher ex Bentham (ENDEMIC), Salvia hypargeia Fisch. & Mey. (ENDEMIC), Salvia euphratica subsp. euphratica Montbret & Aucher ex Bentham (ENDEMIC), Salvia sclarea L., Salvia candidissima subsp. candidissima Montbret & Aucher ex Bentham and Salvia aethiopis L.] from Turkey. The extracts were screened for their possible antioxidant activities by two complementary test systems, namely DPPH free radical-scavenging and β-carotene/linoleic acid systems. Non-polar subfractions of the methanol extracts of Salvia species studied did not show any antioxidant activity in both test systems. In the first case, the most active plant was S. euphratica subsp. euphratica, an endemic species, with an IC50 value of 20.7 ± 1.22 μg/ml, followed by S. sclarea (IC50 = 23.4 ± 0.97 μg/ml) among the polar subfractions. In the β-carotene/linoleic acid test system, polar extract of S. hypargeia was superior to the polar extracts of other Salvia species studied (69.2% ± 1.90%). This activity was followed by S. sclarea with 63.5% ± 4.24% inhibition rate. The inhibition rate of the synthetic antioxidant, buthylated hydroxytoluene (BHT), was also determined to be 96%. Since the polar extracts of Salvia species dealt with here exhibited excellent antioxidant activities when compared to BHT, it seems possible to keep perishable fat-containing food longer by direct addition of an extract of sage.  相似文献

The antioxidant activity of Ruellia tuberosa L. (Acanthaceae) was investigated by the 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical-scavenging assay and the hydrogen peroxide-induced luminol chemiluminescence assay. The methanolic extract (ME) and its four fractions of water (WtF), ethyl acetate (EaF), chloroform (CfF), and n-hexane (HxF) were prepared and then subjected to antioxidant evaluation. The results of both methods revealed that R. tuberosa possesses potent antioxidant activity. The antioxidant activities of the different fractions tested decreased in the order of EaF > CfF > ME > WtF > HxF according to the hydrogen peroxide-induced luminol chemiluminescence assay, and results were the same with the exception of the rank order of HxF and WtF according to the DPPH free radical-scavenging assay. The results provide useful information on the pharmacological activities associated with free radicals of this traditional folk remedy.  相似文献

Three medicinal plants namely Trigonellafoenum-graecum, Glycinemax and Sesamumindicum were evaluated for invitro acetyl cholinesterase inhibitory activity. These plants have been selected based on their use as memory enhancing as well as their nutrient value. These plants have been consumed as nutritious food and are believed to play an important role in health-promoting. The results were expressed as IC50 and the percent of inhibition of acetylcholinesterase (AchE) activity. Diphenyl picrylhydrazil (DPPH) assay and beta-carotene bleaching method were used for antioxidant studies and brine shrimp lethality test (BSL) was used for cytotoxicity assay. The obtained results showed that the G.max extract has inhibited AChE activity strongly in a dose-dependent manner (IC50 = 4.69 mg/mL). The most inhibition of AChE activity was due to G.max extract (68.4%). This extract was also able to scavenge DPPH radical with IC50 = 454.3 μg/mL. The G.max extract has shown the least cytotoxicity (IC50 value of 1112.6 μg/mL) in BSL assay. T.foenum-graecum and S.indicum also exhibited noticeable AchE inhibition.  相似文献

The total phenols index (TPI) and antioxidant activity of persimmon vinegars produced by different processes were evaluated. A novel extraction method was designed and optimised for this purpose with respect to the type and concentration of solvent and ultrasonication time. The best extraction conditions found were the use of 80% ethanol and 25 min of ultrasonication. Antioxidant capacity was determined by the oxygen-radical absorbance capacity of fluorescein (ORAC-FL) and 2,2′-diphenyl-1-pycrylhydrazyl (DPPH) free-radical assays. The antioxidant activities were the same in the fruit and the vinegar, except in the ORAC assay, which showed a significant decrease during the acetification process. The results showed that using the wild yeast strain native to the persimmon produced vinegars with higher antioxidant activity than that of an inoculated alcoholic fermentation. Finally, a comparison between our vinegars and other commercial examples was made. The TPI and antioxidant activity values of persimmon vinegars were always higher than those obtained from white and red-wine vinegars. The antioxidant activity and total phenols of the final product indicate that persimmon vinegar is a competitive product in the market.  相似文献

The fermented products of Monascus sp. are known for their antihypercholesterolaemic effects, however, their antioxidant activities are different from those of many plant-derived foods. To evaluate the effect of ginger addition into the medium on the antioxidant activity of Monascuspilosus fermented products, we cultured uninoculated PDB medium (PDB), inoculated PDB medium (MP), uninoculated ginger-containing medium (PDBG), and inoculated ginger-containing medium (MPG). The broth and mycelia were collected, freeze-dried, and extracted to evaluate their free radical scavenging activities, inhibition of peroxidation, phenolic content, inhibition of DNA damage, cellular antioxidant activity, and expression of the antioxidant enzymes. The results showed that MPG had significantly higher antioxidant activity than PDB, MP, and PDBG at all fermentation time points. Moreover, the fermentation process significantly increased the antioxidant activities of MPG. After the inherent level of antioxidant capacity was increased, the modified M. pilosus fermented product demonstrated a higher anti-atherosclerotic value than the unmodified product.  相似文献

Four different omega-3 rich autotrophic microalgae, Phaeodactylum tricornutum, Nannochloropsis oculata, Isochrysis galbana and Chlorella fusca, were supplemented to the diet of laying hens in order to increase the level of omega-3 long-chain polyunsaturated fatty acids (n-3 LC-PUFA) in egg yolk. The microalgae were supplemented in two doses: 125 mg and 250 mg extra n-3 PUFA per 100 g feed. Supplementing these microalgae resulted in increased but different n-3 LC-PUFA levels in egg yolk, mainly docosahexaenoic acid enrichment. Only supplementation of Chlorella gave rise to mainly α-linolenic acid enrichment. The highest efficiency of n-3 LC-PUFA enrichment was obtained by supplementation of Phaeodactylum and Isochrysis. Furthermore, yolk colour shifted from yellow to a more intense red colour with supplementation of Phaeodactylum, Nannochloropsis and Isochrysis, due to transfer of carotenoids from microalgae to eggs.  相似文献

Leaves from the plant species belonging to the genus Ligustrum are widely used as tea or herbal medicine in Europe, China, and Japan. The antioxidant properties of five Ligustrum species from Taiwan were compared using in vitro antioxidant methods such as DPPH radical scavenging, TEAC, and FRAP assays. Cell-based antioxidant methods were used, including Fe2+/ascorbate-induced lipid peroxidation on brain homogenate and AAPH-induced erythrocyte haemolysis. The amounts of major phenolic compounds from the Ligustrum species, including phenylpropanoids, flavonoids, and iridoids, were determined by spectrophotometric methods. The results showed that all Ligustrum species exhibited antioxidant, radical-scavenging, anti-haemolytic, and lipid peroxidation-inhibiting activities at different magnitudes of potency. A significant correlation was found between antioxidant activity and the amount of antioxidant components, in particular, total phenolics and phenylpropanoids. Among all Ligustrum species from Taiwan, Ligustrum morrisonense is presented as potential source of natural antioxidants.  相似文献

Antioxidant activity of seed phenolics was studied in the following Lathyrus species: Lathyrus hirsutus, Lathyrus filiformis, Lathyrus sativus, Lathyrus cicera, Lathyrus angulatus, Lathyrus sphaericus, Lathyrus annuus, Lathyrus clymenum, Lathyrus pratensis, Lathyrus ochrus, Lathyrus aphaca, Lathyrus latifolius, Lathyrus setifolius, Lathyrus tingitanus and Lathyrus amphicarpos. Phenolic contents ranged from 3.8 mg/g meal in L. setifolius to 29.2 mg/g meal in L. sphaericus. In general, non-cultivated Lathyrus species contained higher phenolic contents than cultivated ones. A negative correlation between seed size and phenolic contents was observed and was related to the higher proportion of hulls in the smaller seeds. L. annuus possessed phenolics with highest specific antioxidant activity. These phenolics were more than two times more antioxidant than equivalent amounts of phenolics extracted from commercial chickpea, lupin or soy. On the other hand, L. aphaca possessed the highest antioxidant activity per mg of flour extract. This antioxidant activity was twice that observed in same amounts of extracted flours from commercial chickpea, lupin or soy. Results show that studied Lathyrus species are rich in phenolic compounds with higher antioxidant activity than phenolics of widely consumed legumes such as soy, chickpea or lupin. In conclusion, Lathyrus may represent an interesting source of phenolic compounds with high antioxidant activity that may be useful as natural antioxidants and contribute to revalorize the cultivation of these legumes.  相似文献

Flavonols, betacyanins content and antioxidant activity of red-purple cactus Opuntia macrorhiza fruits, a promising cactus pear species, were evaluated in comparison to the well known cactus pear fruits of Opuntia ficus-indica. Flavonol profile was evaluated by HPLC-DAD prior to and after enzymatic hydrolysis (glycosides vs. aglycons). In addition betacyanins, responsible for the purple to red color of cactus pear fruits were also estimated and compared to Beta vulgaris ssp. (roots) and red O. ficus-indica fruits. In all cases, cactus fruit pulps showed no flavonols at all. While different derivatives of isorhamnetin glycosides were found in O. ficus-indica fruit's peel, isorhamnetin-3-O-rutinoside was the only compound in O. macrorhiza fruit. Correspondingly, antioxidant activity assays showed a high antioxidant activity of both, O. macrorhiza fruit's peel and pulp. With regard to betacyanins, O. macrorhiza fruit's peel and pulp provide a deep red-purple color, whose average impact is higher compared to red beet (B. vulgaris spp.) and about 8-fold higher than red fruits from O. ficus-indica. Supporting these results, estimation of color attributes (L*, a*, b*, C and H°) showed also a high color impact of both O. macrorhiza fruit's peel and pulp.  相似文献

This study was carried out to investigate changes in the free amino acid contents and antioxidant activity of Panax ginseng induced by steaming at different temperatures. For this purpose, white ginseng (WG), red ginseng (RG, ginseng steamed at 100 °C) and ginseng steamed at 120 °C (SG) were prepared using an autoclave. Most free amino acids were decreased significantly by steam treatment, with the greatest reduction observed in SG. Total content of free amino acids, 17.9 mg/g in WG was reduced to 12.2 mg/g in RG and 2.79 mg/g in SG. As for Arg which is the most predominant amino acid in ginseng, the content, 10.4 mg/g in WG, decreased significantly to 1.38 mg/g in SG. In particular, β-N-oxalyl-L-α,β-diaminopropionic acid (β-ODAP), a well-known neurotoxin, was reduced by 92.9% in SG. In contrast, the level of Maillard reaction products (MRPs) increased with steam treatment, which indicates that the reduction of most amino acids is attributed to the extent of the Maillard reaction. Based on MRPs being useful antioxidants, we assayed the scavenging activity against free radicals produced by 2,2′-azobis-(2-amidinopropane) dihydrochloride (AAPH). The radical scavenging activity of a ginseng extract increased with steam treatment, with the most potent activity in SG. Further, MRPs-rich fraction in SG showed powerful antioxidant activity, which indicates MRPs are major contributors to antioxidant activity enhanced by steam treatment.  相似文献

Hydro-distilled volatile oils from the aerial parts of three Satureja species were investigated, mainly by a combination of GC and GC/MS. One hundred and thirteen compounds were identified, representing 82.9–92.0% of the total oil. Among the identified components, spathulenol, cis-piperitone oxide, α-bisabolol oxide-B, terpinen-4-ol, linalool, bornyl acetate, β-bourbonene, isomenthone, thymol, neoisomenthol and menthone were found as the main components. Furthermore, the essential oils were investigated for their antimicrobial activity, by the agar dilution technique. The antimicrobial test results showed that the oils had a high antimicrobial activity against two Gram-positive and four Gram-negative bacteria, two oral pathogens and three pathogenic fungi. Gram-positive bacteria were more sensitive to the investigated oils than were Gram-negative bacteria. These results could support the suggestion of Satureja species as a source of antimicrobial ingredients for the food industry.  相似文献

Eleven Lactobacillus plantarum strains isolated from traditional Chinese fermented foods were investigated for their in vitro scavenging activity against hydroxyl and 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radicals, and their resistance to hydrogen peroxide. L. plantarum C88 at a dose of 10(10) CFU/ml showed the highest hydroxyl radical and DPPH scavenging activities, with inhibition rates of 44.31% and 53.05%, respectively. Resistance of intact cells to hydrogen peroxide was also found in all strains. L. plantarum C88 was the most resistant strain against hydrogen peroxide. When L. plantarum C88 was administered to senescent mice suffering oxidative stress induced by d-galactose, the serum superoxide dismutase activity, the glutathione peroxidase activity and the total antioxidant capacity in liver increased significantly, while the level of malondialdehyde in liver decreased significantly. L. plantarum C88 isolated from traditional Chinese fermented dairy tofu could be considered as a potential antioxidant to be applied in functional foods.  相似文献

The number of methods to measure antioxidants in botanicals, foods, nutraceuticals and other dietary supplements are increased considerably in the last 10 years. However most techniques require long experimental times and high costs to determine antioxidant capacity of hydrophilic or lipophilic compound in a food mixture. By means of a photochemiluminescence method, we assessed the Integral Antioxidant Capacity (IAC) which represents the sum of the antioxidant capacity of hydrophilic and lipophilic antioxidants. In this study the IAC of extracts from Adansonia digitatata (i.e. red fiber, fruit pulp and leaves), was assessed in comparison to those deriving from other natural sources of antioxidants (i.e. orange, kiwi, apple and strawberry). When compared, IAC values for the examined product resulted as follows: Adansonia digitata red fibre ? Adansonia digitata fruit pulp ? Adansonia digitata fresh leaves ? Adansonia digitata seeds ? Adansonia digitata radix cuticle ? orange fresh pulp ? strawberry fresh fruit pulp > Adansonia digitata radix > bilberry fresh pulp ? kiwi fruit pulp. Results clearly indicate the interesting antioxidants properties of Adansonia digitata red fibre, in particular the IAC value of baobab red fibre was 66 time higher than that of orange pulp, with value of 1617.3 μmol/g and 24.3 μmol/g, respectively.  相似文献

In this study, the antioxidant activity of methanol extracts of five plants from the genus Phyllanthus was evaluated by various antioxidant assays, including total antioxidant, free radical scavenging, superoxide anion radical scavenging, hydrogen peroxide scavenging, nitric oxide scavenging, reducing power and metal ion chelating activities. The various antioxidant activities were compared to standard antioxidants such as butylated hydroxytoluene and ascorbic acid. All the extracts showed strong antioxidant activity in all the tested methods. Among the five plants Phyllanthus debilis has been found to possess the highest activity in all tested models, the activity decreased in the order Phyllanthus debilis>Phyllanthus urinaria>Phyllanthus virgatus>Phyllanthus maderaspatensis>Phyllanthus amarus. In addition to the antioxidant activity of these plants, the total phenolic compounds, flavonoids and flavonols were measured in the extracts. A correlation between the antioxidant activity and total phenolic content was observed.  相似文献
