Fermented foods have shown promise in preventing or treating ulcerative colitis (UC) via regulating intestinal flora and correcting metabolic disorders. However, the prevention effect of fermented Wallace melon juice (FMJ) on UC is unclear. In this study, the effects of FMJ on dextran sodium sulfate (DSS)-induced UC were investigated via 16S rRNA sequencing and non-targeted metabolomics. The results showed that FMJ was effective in alleviating the symptoms of UC, reducing histological damage and oxidative stress, decreasing the levels of pro-inflammatory cytokines. After FMJ treatment, the level of propionic acid, butyric acid, and valeric acid increased by 14.1%, 44.4%, and 52.4% compared to DSS-induced UC mice. Meanwhile, the levels of harmful bacteria such as Oscillospira, Bacteroidetes, and Erysipelotrichaceae and Clostridium decreased, while the levels of beneficial bacteria such as Akkermansia, Lactobacillus, and Bifidobacterium increased. Fecal metabolomics analysis identified 31 differential metabolites, which could regulate metabolic disorders in UC mice by controlling the primary bile acid biosynthesis, purine metabolism, and pantothenate and CoA biosynthesis pathway. Additionally, the abundances of butyric acid, bile acids, and pantothenic acid were positively correlated with Allobaculum, Bifidobacterium, and other beneficial bacteria (R2 > 0.80, p < 0.01). The results indicated that FMJ played a role in regulating the structure of intestinal flora, which in turn helped in repairing metabolic disorders and alleviated colitis inflammation. 相似文献

为研究脱脂豆粕中提取的大豆低聚糖(soybean oligosaccharide;SBOS)体外消化和发酵特性;分析了其单糖组成和分子质量的变化;并通过16S rRNA基因测序和液相色谱-质谱联用非靶向代谢组学研究SBOS对微生物群落结构和代谢物的影响。结果表明;SBOS在模拟消化过程中不易被酶解;可通过消化系统到达大肠。体外发酵后SBOS的分子质量显著降低;表明其可被肠道菌群利用;同时短链脂肪酸和乳酸的含量增加;导致发酵液的pH值降低。此外;核心群落包括布劳特氏菌(Blautia)、乳酸杆菌(Lactobacillaceae)和片球菌(Pediococcus)。SBOS上调有益的差异代谢物如肌醇、乳糖和葡萄糖;这些代谢物与半乳糖、氨基糖和核苷酸糖代谢密切相关。本研究为探索SBOS肠道菌群与代谢物之间的关系提供参考;并为SBOS作为功能性产品原料的开发和应用提供了依据。 相似文献

目的 应用超高效液相色谱/四级杆-飞行时间质谱技术分析鉴定了克伦特罗在猪尿中的代谢产物,并推测克伦特罗在猪体内的主要代谢途径。方法 按10 mg/kg bw的剂量,给猪口服灌食克伦特罗,分别采集给药前及给药后的尿液样品。应用超高效液相色谱/四级杆-飞行时间质谱技术对样品进行分析,采用MetaboLynx XS软件进行数据处理,共检测到6 种克伦特罗的代谢产物,并根据碎片离子信息进行了结构鉴定。结果 猪尿中克伦特罗的代谢产物包括4-N-羟基克伦特罗(4-N-OH-CLE)、4-硝基克伦特罗(4-NO2-CLE)、克伦特罗及4-N-OH-CLE的葡萄糖醛酸结合物(GLU-CLE和GLU-OH-CLE)等。结论 根据所检测到的代谢产物,克伦特罗在猪体内的代谢途径包括4-N-氧化和葡萄糖醛酸结合等。 相似文献

Clinical trials investigating the health effects of flavan-3-ols yield heterogeneous results due to interindividual variability in the gut microbiota metabolism. In fact, different groups in the population have similar metabolic profiles following (−)-epicatechin and (+)-catechin gut microbial metabolism and can be regrouped into so-called metabotypes. In this study, the capacity of 34 donors to metabolize polymeric B-type flavan-3-ols from aronia and oligomeric A-type flavan-3-ols from cranberry is investigated by in vitro fecal batch fermentations. Less than 1% of the flavan-3-ols from both sources are converted into microbial metabolites, such as phenyl-γ-valerolactones (PVLs). To further confirm this result, gut microbial metabolites from flavan-3-ols are quantified in urine samples collected from participants, before and after a 4-day supplementation of cranberry extract providing 82.3 mg of flavan-3-ols per day. No significant difference is observed in the urinary excretion of flavan-3-ols microbial metabolites. Hence, it demonstrates by both in vitro and in vivo approaches that flavan-3-ols from aronia and cranberry are poorly degraded by the gut microbiota. The beneficial health impacts of these molecules likely stem from their capacity to affect gut microbiota and their interactions with the gut epithelium, rather than from their breakdown into smaller metabolites. 相似文献

A GH67 α-glucuronidase gene derived from Bacillus halodurans C-125 was expressed in E. coli to obtain a recombinant enzyme (BhGlcA67). Using the purified enzyme, the enzymatic properties and substrate specificities of the enzyme were investigated. BhGlcA67 showed maximum activity at pH 5.4 and 45 °C. When BhGlcA67 was incubated with birchwood, oat spelts, and cotton seed xylan, the enzyme did not release any glucuronic acid or 4-O-methyl-glucuronic acid from these substrates. BhGlcA67 acted only on 4-O-methyl-α-D-glucuronopyranosyl-(1→2)-β-D-xylopyranosyl-(1→4)-β-D-xylopyranosyl-(1→4)-β-D-xylopyranose (MeGlcA3Xyl3), which has a glucuronic acid side chain with a 4-O-methyl group located at its non-reducing end, but did not on β-D-xylopyranosyl-(1→4)-[4-O-methyl-α-D-glucuronopyranosyl-(l→2)]-β-D-xylopyranosyl-(1→4)-β-D-xylopyranosyl-(1→4)-β-D-xylop- yranose (MeGlcA3Xyl4) and α-D-glucuronopyranosyl-(l→2)-β-D-xylopyranosyl-(1→4)-β-D-xylopyranosyl-(1→4)-β-D-xylopyranose (GlcA3Xyl3). The environment for recognizing the 4-O-methyl group of glucuronic acid was observed in all the crystal structures of reported GH67 glucuronidases, and the amino acids for discriminating the 4-O-methyl group of glucuronic acid were widely conserved in the primary sequences of the GH67 family, suggesting that the 4-O-methyl group is critical for the activities of the GH67 family. 相似文献

Scope
The phytosteroid ecdysterone is present in spinach. In this study, the urinary elimination of ecdysterone and its metabolites in humans is investigated following spinach consumption of two different culinary preparations.Methods and results
Eight participants (four males, four females) ingested 950 (27.1) g sautéed spinach (average [±standard deviation (SD)]) and 912 (70.6) g spinach smoothie as second intervention after washout. Post-administration urines are analyzed by liquid chromatography tandem mass spectrometry (LC-MS/MS). After intake of both preparations, ecdysterone and two metabolites, 14-deoxy-ecdysterone, and 14-deoxy-poststerone, are excreted in urine. The maximum concentration of ecdysterone is ranging from 0.09 to 0.41 µg mL−1 after sautéed spinach and 0.08–0.74 µg mL−1 after smoothie ingestion. The total excreted amount (mean% [±SD]) in the urine as a parent drug plus the metabolites is only 1.4 (1.0) for both sautéed spinach and smoothie. The apparent sex related differences in 14-deoxy-poststerone excretion will need further investigations.Conclusion
Only a small proportion of ecdysterone from spinach is excreted into urine. No significant differences are found in concentration and recovered amount (%) of ecdysterone, 14-deoxy-ecdysterone, and 14-deoxy-poststerone in urine between sautéed spinach and smoothie ingestion. A discrimination between ecdysterone from food or preparations will be challenging based on urinary concentrations only, at least for later post-administration samples. 相似文献9.
目的:采用1H-NMR代谢组学技术研究口服葡萄籽提取物后人体尿液中内源性代谢产物的变化规律。方法:选择16名健康男生随机平均分成两组,即试食组(A)和对照组(B)。试食组每人每天口服葡萄籽提取物软胶囊2粒,连续14d,对照组连续服用安慰剂14d。分别收集试食组及对照组服药前(0d)和服药后(14d)的晨尿,运用1H-NMR代谢组学技术分析尿液中内源性代谢产物的变化及其规律。结果:与对照组相比,服药14d后,试食组人体尿液中乳酸水平下降,肌酐水平上升,且具有显著性差异(p<0.05,0.01);结论:葡萄籽提取物(GSE)对人体尿液中内源性代谢产物的调节作用显著,涉及能量代谢和氨基酸代谢通路,对能量代谢的调节尤为显著,是一种很好的运动营养补充剂。1H-NMR代谢组学技术在运动科学研究中的应用前景广阔。 相似文献

Use of bottled water in Sri Lanka has increased over the last decade, while new brands of bottled water are often introduced to the market. However, the manufacturers' adherence to bottled water regulations is questionable, raising concerns regarding the quality of bottled water. The objective of the current study was to investigate the microbiological and chemical quality of bottled water in Sri Lanka. Thirty bottled water brands were sampled and their chemical and microbiological parameters were analyzed. Microbiological analysis was carried out within 1 to 3, 3 to 6, 6 to 9, and 9 to 12 mo after the date of manufacture. The results indicated that 63% of brands tested exceeded the levels permitted by the Sri Lanka Standards Institution (SLSI) for presumptive total coliforms (TC) (<10 cfu per 100 mL) whereas 97% brands exceeded the World Health Organization (WHO) permitted level. Thirty percent of brands exceeded the limit for presumptive fecal coliforms (FC) (0 cfu per 100 mL in accordance with WHO permitted levels, SLSI and the Sri Lanka Health Ministry requirement). Eighty percent of brands showed higher heterotrophic plate counts (HPC) which exceeded the WHO guidelines for bottled drinking water. Throughout their shelf life, the counts of TC, FC, and HPC bacteria decreased. Bacteria identified were Klebsiella pneumoniae ssp. pneumoniae, Enterobacter cloacae, Pseudomonas aeruginosa, and Pasteurella haemolytica, the most frequently being P. aeruginosa. The dominant fungi identified were Aspergillus sp. and Penicillium sp. Inorganic chemical parameters were within permitted levels for all brands except for initial content of ammonia. The results of this study show the need for the bottling industry to be monitored closely by relevant authorities, in order to provide safe bottled drinking water to consumers in Sri Lanka. 相似文献

BACKGROUND: A variety of methods have been developed for estimating lignin concentration within plant materials. The objective of this study was to compare the lignin concentrations produced by six methods on a diverse population of forage and biomass materials and to examine the relationship between these concentrations and the portions of these materials that are available for utilisation by livestock or for ethanol conversion. RESULTS: Several methods produced lignin concentrations that were highly correlated with the digestibility of the forages, but there were few relationships between these methods and the available carbohydrate of the biomass materials. The use of Na2SO3 during preparation of residues for hydrolysis resulted in reduced lignin concentrations and decreased correlation with digestibility of forage materials, particularly the warm‐season grasses. CONCLUSION: There were several methods that were well suited for predicting the digestible portion of forage materials, with the acid detergent lignin and Klason lignin method giving the highest correlation across the three types of forage. The continued use of Na2SO3 during preparation of Van Soest fibres needs to be evaluated owing to its ability to reduce lignin concentrations and effectiveness in predicting the utilisation of feedstuffs and feedstocks. Because there was little correlation between the lignin concentration and the biomass materials, there is a need to examine alternative or develop new methods to estimate lignin concentrations that may be used to predict the availability of carbohydrates for ethanol conversion. There has been a growing interest in cofermentation of starter cultures with probiotics in milk. In this study, we analyzed the effects of adding the single probiotics Lactobacillus casei Zhang (Zhang) or Bifidobacterium animalis ssp. lactis Probio-M8 (M8) or a combination of Zhang and M8 to starter cultures on volatile and nonvolatile metabolomic profiles after 14 d of storage at 4°C and compared using a liquid chromatography-tandem mass spectrometry (LC-MS) and GC-MS-based metabolomics approach. Principal component analysis, heatmap plots, and Spearman correlation results showed that Zhang alone had a greater effect on volatile and nonvolatile metabolomic profiles than M8 alone. The combination of Zhang and M8 had additive effects on the production of metabolites. For volatile metabolites, the levels of acetaldehyde, diacetyl, acetoin, and acetic acid were higher for the combination of Zhang and M8 compared with either single probiotic culture. Significantly increased nonvolatile components induced by adding Zhang were identified were enriched in the galactose, amino- and nucleotide sugar, fructose and mannose, purine, phenylalanine metabolism, and arginine biosynthesis pathways. The metabolism and biosynthesis of starch, sucrose, tyrosine, galactose metabolism, and aminoacyl-tRNA biosynthesis were significantly upregulated by adding the combination of Zhang and M8. This work provides a detailed insight into different effects of Zhang and M8 used alone or in combination on the volatile and nonvolatile metabolomic profiles of yogurts. 相似文献
