The activity of morantel citrate (5.94 mg/kg base) was determined in laboratory tests against field isolates of benzimidazole-resistant nematodes. Its efficacies against adult and seven-day-old worms were 100 per cent and 100 per cent for Cooperia curticei, 95.1 per cent and 69.8 per cent for Haemonchus contortus and 100 per cent and 82 per cent for Ostertagia circumcincta. Morantel citrate was 100 per cent effective against benzimidazole-susceptible Nematodirus battus and Trichostrongylus vitrinus, and it reduced faecal egg counts by 97.9 per cent in sheep infected naturally with benzimidazole-resistant H contortus and O circumcincta.  相似文献

A study on the seasonal variations in the population structure of Haemonchus contortus and Trichostronglyus colubriformis was conducted for a period of 12 months in a typical large scale sheep farm on improved pasture in Peninsular Malaysia which has a wet tropical climate. Successive groups of helminth-free tracer lambs were grazed for 4 weeks together with naturally infected sheep and were necropised for worm counts 2 weeks after their removal from the pasture. The monthly populations of H. contortus fluctuated slightly except in May and August during which more worms were found in the tracer animals. The numbers of T. colubriformis were comparatively high from October to December 1992 and again in March 1993, low during April and June 1992. Small numbers of hypobiotic larvae of H. contortus were detected in the tracer animals. Development and survival of infective larvae of H. contortus and T. colubriformis on pasture were investigated by spreading faeces containing eggs on grass plots in October 1993, February and May 1994. Development of the eggs to the infective larvae occurred within one week and their survival times were 7 weeks in the 3 experiments. The potential for control by rotational grazing is discussed.  相似文献

The efficacy of five daily treatments with 7.5 mg fenbendazole/kg bodyweight against mucosal cyathostome larvae was evaluated in 20 12- and 24-month-old ponies with naturally acquired cyathostome infections. After three weeks communal grazing on infected pasture and six weeks indoors, one group of 10 ponies were treated. Six weeks later, both groups of ponies were humanely destroyed and their burdens of large intestinal cyathostome worms, including luminal parasites and mucosal larvae, were assessed. In the control animals approximately 7 per cent of the total worm burden was present in the gut lumen and 93 per cent was present as larvae in the large intestinal mucosa. The efficacy of fenbendazole against the luminal cyathostomes was 90.7 per cent (P < 0.01). The total numbers of mucosal larvae, recovered after digestion, were reduced by 95.3 per cent (P < 0.0005); mucosal late third and fourth stage larvae were reduced by 99.4 per cent (P < 0.0001), and early inhibited third stage larvae by 91.5 per cent (P < 0.005).  相似文献

One hundred and four post mortem examinations of randomly selected sheep (52) and goats (52) were carried out weekly from March 1990 to February 1991. Gastrointestinal parasites were identified and counted. A large spectrum of 16 helminth species was found in 101 (97%) infected animals. The nematodes infecting small ruminants were, in order of predominance: Trichostrongylus colubriformis (96% of all animals), Oesophagostomum columbianum (82%), Haemonchus contortus (67%), Strongyloides papillosus (55%), Gaigeria pachyscelis (38%), Cooperia spp. (49%) and Trichuris ovis (12%). Forty-five (43%) necropsies revealed an infection with Moniezia benedeni, Avitellina centripunctata or Stilesia globulosa. Infections with Schistosoma bovis and Paramphistomum spp. were rarely seen. Goats carried significantly lower worm burdens than sheep and were less often infected by tapeworms. Nematode worm burdens followed a similar seasonal pattern in both host species. While Trichostrongylus colubriformis was also recovered in high numbers in the mid dry season, other species such as H. contortus, Strongyloides papillosus and Oesophagostomum columbianum reached distinct peaks in the rainy season (July-October). Different survival strategies for the prolonged dry season were observed. Haemonchus contortus outlived the unfavourable climatic conditions as inhibited larvae in the abomasal mucosa, while the other nematode species survived as adults with a reduced fecundity. Egg production per adult worm specimen was found to depend heavily on the season and egg counts in the dry season (November-June) did not correspond to the size of worm burdens. Unexpectedly high worm burdens were recovered from adult and old animals. Investigations of young animals indicated that no reinfection can occur during the dry season. The findings are discussed with regard to their relevance for strategic control of gastrointestinal nematodes in small ruminants.  相似文献

Five controlled trials were conducted in Germany or in the United Kingdom, using 74 female sheep of merino or Dorset horn breeds, to evaluate the efficacy of an ivermectin controlled-release capsule against naturally acquired or induced infections of gastrointestinal nematodes, lungworms and nasal bot larvae and against incoming infections with gastrointestinal and pulmonary nematodes. Half of the animals were treated with one ivermectin controlled-release capsule that delivered ivermectin at the rate of 1.6 mg per day for 100 days while the other half remained untreated. Parasites were counted 21, 28, 35 or 56 days after administration of the capsule. The treatment was highly effective (> or = 99 per cent) against established parasites of the following species: Haemonchus contortus (adults and fourth-stage larvae), Ostertagia circumcincta, O pinnata, O trifurcata, Ostertagia species fourth-stage larvae, Trichostrongylus axei, T colubriformis, T vitrinus, Cooperia curticei, Nematodirus battus, N filicollis, Strongyloides papillosus, Chabertia ovina, Oesophagostomum venulosum, Trichuris ovis, Tr skrjabini, Dictyocaulus filaria, Protostrongylus rufescens and Oestrus ovis (larvae). The treatment prevented the establishment of the gastrointestinal nematodes H contortus, O circumcincta, T axei, T colubriformis, C curticei, N battus, N filicollis, Ch ovina, Oe vennulosum and the establishment of the lungworm D filaria by > 99 per cent compared with untreated controls (P < or = 0.01).  相似文献

Forty-five calves with artificial and pasture-acquired nematode infections were medicated with albendazole at dose levels of 0, 2.5, 5.0, or 10 mg/kg of body weight. A dose level of 2.5 mg/kg removed at least 99% of adult Trichostrongylus axei, Trichostrongylus colubriformis, Cooperia oncophora, and Bunostomum phlebotomum. Burdens of Haemonchus contortus, Strongyloides papillosus, and Ostertagia ostertagi were reduced 79, 88, and 97%, respectively. At a dose level of 5.0 mg/kg, at least 95% of all adult nematodes were removed; at 10 mg/kg, at least 97% were removed. At least 99% of 4th-stage larvae of O ostertagi, T axei, C oncophora and T colubriformis and 96% of H contortus were expelled at a dose level of 2.5 mg/kg. At 5.0 and 10 mg/kg, 99 to 100% of all species of larvae were removed. Trichuris spp adults were slightly susceptible at all dose levels; larvae were susceptible (83%) only at 10 mg/kg.  相似文献

Plasma concentrations of febantel and its major metabolites, fenbendazole, oxfendazole and fenbendazole sulphone, were determined after oral administration of 7.5 mg/kg febantel in lambs before and 28 days after infection with 100,000 L3 larvae of a benzimidazole (BZ)-sensitive or BZ-resistant strain of Ostertagia circumcincta or with 75,000 L3 larvae of a BZ-sensitive Trichostrongylus colubriformis strain. The febantel concentrations were always low, and in only a few samples were higher than the limit of detection. A mean decrease in the area under the curve (AUC) for the three metabolites of 10.2%, 16.4% and 4.9% in lambs infected, respectively, with BZ-sensitive O. circumcincta, BZ-resistant O. circumcincta and T. colubriformis was observed. The Cmax for all the metabolites was higher in the BZ-sensitive O. circumcincta group than in the naive sheep, while the Tmax occurred earlier. The Cmax and the Tmax values for all the metabolites were lower in the BZ-resistant O. circumcincta group than in their own naive controls. In the T. colubriformis group the Cmax values of the metabolites were lower and the Tmax occurred much later.  相似文献

Resistance to levamisole and the benzimidazoles appears to be achieved by one or, at most, two mechanisms in the common trichostrongyloid parasites of sheep. For the avermectin/milbemycin anthelmintic class the picture is more complex. In-vitro assays employing the free-living stages of trichostrongyloid nematodes were used to investigate structure-activity relationships for the avermectins/milbemycins. While avermectin/milbemycin-susceptible isolates of Haemonchus contortus, Trichostrongylus colubriformis and Ostertagia circumcincta were found to differ in their intrinsic sensitivities to avermectin/milbemycin inhibition of larval development and L3 motility, structure-activity profiles against all three species were similar. In-vivo avermectin/milbemycin resistance was associated with a reduced sensitivity to avermectin/milbemycin inhibition of larval motility and/or development in some, but not all, isolates. Where a reduced sensitivity to avermectin/milbemycin inhibition of larval development was observed, different groups of resistant isolates displayed different structure-activity profiles. Many avermectin/milbemycin-resistant isolates showed an increased sensitivity to paraherquamide. These in-vitro data have allowed the classification of avermectin/milbemycin-resistant isolates into a number of distinct types. Study of the inheritance of avermectin/milbemycin resistance in two resistance types suggests that the in-vitro differences between resistant isolates reflect important differences in the mechanism of resistance present. The kinetics of expulsion of H. contortus, T. colubriformis and O. circumcincta from sheep following treatment with ivermectin indicate that, in vivo, the critical action of avermectins/milbemycin against O. circumcincta may be different to that which results in H. contortus and T. colubriformis elimination. This observation provides some explanation for the differences between resistant isolates. If, for different species, the critical event(s) leading to expulsion are different, then it follows that the mechanisms of resistance observed may also differ.  相似文献

Four groups of 8 parasite-naive Dorset-crossbred lambs, 3-4 months old, were turned out on infected pasture on 2 May and allocated to 4 separate paddocks. From May to September, 2 groups received Duddingtonia flagrans (10(6) chlamydospores per kg body weight per lamb per day) mixed in 100 g of barley, while the other 2 groups received barley only. All groups remained set-stocked until slaughter for worm counts on 10 October. In late June, all lambs were treated with fenbendazole due to severe parasitic gastroenteritis in all groups. The faecal egg counts were comparable for the 2 treatments throughout the grazing period. Larval development of Ostertagia/Trichostrongylus spp. in faecal cultures was 1-28% in the fungi-fed groups compared with 60-80% in the untreated groups (P < 0.05). In September, pasture larval counts of Ostertagia/Trichostrongylus were 930 and 4400 L3 kg-1 on paddocks of fungi-fed and untreated groups, respectively. Corresponding figures for Nematodirus spp. were 7200 and 11600 L3 kg-1, respectively. At slaughter, the number of immature Ostertagia spp. was 62% lower in the fungi-fed groups compared with the untreated groups (P < 0.05). Four parasite-free lambs were introduced to each paddock during the period 3-23 October and slaughtered for worm counts after 3 weeks of housing. The total worm burden of tracers on paddocks previously grazed by fungi-fed lambs was reduced 86% (P < 0.05; geometric means) compared with control groups, while significant reductions were also seen in abomasal worm counts (68%; P < 0.05), N. spathiger (98%; P < 0.05) and for N. battus (97%; P < 0.01). It is concluded that dosing sheep with D. flagrans while grazing may limit the build up of pasture contamination in the late grazing season and subsequently limit the intake of larvae in sheep.  相似文献

First stage (L1) larvae of Haemonchus contortus, Trichostrongylus colubriformis and Ostertagia circumcincta can be cryopreserved in the presence of DMSO using a two-step freezing protocol involving an initial period at -80 degrees C prior to transfer to liquid nitrogen. Thawed L1 larvae continue development in vitro producing third stage (L3) larvae that are infective to sheep when dosed per os. Establishment rates for L3 larvae grown from thawed L1 larvae were 40 and 80% for H. contortus and T. colubriformis, respectively. There was no difference in survival or infectivity between benzimidazole (BZ)-susceptible and BZ-resistant H. contortus isolates and cryopreservation caused no shift in their BZ-resistance status as indicated in an in vitro larval development assay. Cryopreservation also had no effect on the sensitivity of these isolates to the avermectins or levamisole in vitro. High survival rates (60-70%), good levels of establishment and the stability of anthelmintic resistance status of isolates indicate that little if any selection occurs during the cryopreservation process. L1 larvae of all 3 species have been successfully recovered after 16 months storage in liquid nitrogen, cultured to the L3 stage and established in sheep.  相似文献

We have recently synthesized a series of novel disulfonylmethane compounds that have shown anthelmintic and insecticidal (endectocidal) activity. Several analogues have shown activity against the internal nematode Haemonchus contortus. In sheep studies, these analogues have shown 100% control of this internal parasite at a 10 mg/kg rate. In vitro activity against the biting flies, Stomoxys calcitrans and Haematobia irritans, has been observed at rates as low as 25 and 2.3 ppm, respectively. Only marginal activity against the liver fluke Fasciola hepatica and Trichostrongylus colubriformis was seen. Respiratory control index values on rat liver mitochondria for this series suggested uncoupling of oxidative phosphorylation as a mechanism of action. Compound 1 is considered to be a promising agent for treatment of parasitized sheep.  相似文献

The complement of beta-tubulin alleles in Trichostrongylus colubriformis populations was examined and found to undergo changes similar to those previously reported for Haemonchus contortus following selection for benzimidazole (BZ) resistance. Genomic DNA from BZ-resistant and -susceptible strains was probed with a series of overlapping fragments derived from a T. colubriformis beta-tubulin gene. A susceptible population showed a high level of polymorphism (detected as RFLPs with several enzymes and directly by sequence analysis) at a locus, tcb-1, which appears to be the homologue of the gru-1 locus in H. contortus. This polymorphism disappeared following selection for BZ resistance, leaving a single tcb-1 allele in the resistant population. The same single allele was present in 2 additional, unrelated resistant populations. These data support the hypotheses that tcb-1 and gru-1 are major determinants of BZ susceptibility and hence a major target of BZ-resistance selection.  相似文献

The new drug albendazole (methyl [5-(propylthio)-1H-benzimidazol-2-yl]carbamate) was tested in 90 sheep experimentally infected with Fasciola hepatica. Dose rates of 10, 15, and 20 mg/kg of body weight were active against adult flukes, with efficacies of 98, 95, and 100% respectively. The drug was less active against immature F hepatica at 3 weeks after sheep were inoculated. The drug given at dose rates of 20, 30, 40, and 50 mg/kg had efficacies 24, 47, 63, and 76%. Flukes were not found at posttreatment hour 48 in bile ducts of sheep given a dose of 20 mg/kg. Evidence of drug toxicity was not seen.  相似文献

A total of 46 weaned kids of three genotypes aged about 4-5 months were used to evaluate the effects of trickle infection with a sheep strain of Haemonchus contortus. A completely randomized 3 x 2 factorial design was used. Factors were genotype (Thai native (TN), 75% TN x 25% Anglo-Nubian (AN) and 50% TN x 50% AN) and parasite (control and infected). The animals were infected with 750 infective larvae (L3) of H. contortus three times a week for 3 weeks, with a total of 6750 larvae. The experiment lasted 9 weeks. Each week animals were weighed, faecal egg counts done and blood examined for haematological and biochemical variables. Twenty-seven kids were slaughtered at the end of experiment for worm recovery. Weight gain of infected animals was lower than those of uninfected controls (P < 0.05). The genotype 50% TN x 50% AN had higher growth rate than TN and 75% TN x 25% AN genotypes (P < 0.05). Eggs per gram of faeces (EPG) were significantly higher in 50% TN x 50% AN kids than in TN (P < 0.0005) and 75% TN x 25% AN (P < 0.0001) kids. There was a large variation in the EPG of individual animals within a genotype. The percent establishment of L3 was 8.2% in TN, 16.97% in 50% TN x 50% AN and 17.91% in 75% TN x 25% AN kids. TN kids had worm counts lower than 50% TN x 50% AN (P < 0.05) and 75% TN x 25% AN (P = 0.07) kids. Infection had a significant effect on packed cell volume (PCV), haemoglobin, total protein and albumin. The decrease in the level of these blood parameters was less in TN kids than in 50% TN x 50% AN and 75% TN x 25% AN kids. There was no significant difference between genotypes in the values of total and differential leucocyte counts and mean corpuscular volume (MCV). It can be concluded that TN goats are more resistant to H. contortus than 50% TN x 50% AN goats. However, under the present experimental conditions, the liveweight gain of 50% TN x 50% AN was higher than the TN goats.  相似文献

Resistance to primary and secondary infections with Haemonchus contortus was studied in 10-month-old Manchego and Merino sheep. No notable interbreed differences were observed after primary infections in the parameters determined (prepatency period, faecal egg output, abomasal worm burden). Previously infected sheep (200 L-3/kg live weight (lw)) from both breeds showed notable protection after challenge (400 L-3/kg lw), evidenced by lower eggs/g faeces (epg) values and worm burdens. A protective response in the Manchego breed was associated with arrested development of fourth stage larvae in the abomasal mucosa, whereas in the Merino breed a more rapid expulsion mechanism seems to be involved. Serum antibody levels (IgG, IgA) were infective dose-dependent and protection from re-infection was not clearly related to the parasite-specific IgG response estimated by enzyme-linked immunosorbent assay (ELISA) and Western blotting.  相似文献

The roles of gamma interferon (IFN-gamma) and interleukin-12 (IL-12) in mediating and/or enhancing the in vivo trypanosomicidal activity of the nitroheterocyclic derivative benznidazole (Bz) were evaluated during early stages of experimental Chagas' disease. Our results show that treatment of Trypanosoma cruzi-infected mice with anti-cytokine monoclonal antibodies (MAbs) had no apparent effect when the optimal dose of Bz (100 mg/kg of body weight) was used. In contrast, treatment with anti-IL-12 or anti-IFN-gamma MAbs enhanced the parasitemia and accelerated the mortality of mice treated with a suboptimal dose of Bz (25 mg/kg). Simultaneous treatment with a suboptimal dose of Bz and recombinant IL-12 (rIL-12) enhanced the efficacy of drug treatment in terms of parasitemia and mouse survival. Interestingly, we found that drug-resistant T. cruzi strains were found to be poor inducers of IL-12 both in vitro and in vivo compared to strains of T. cruzi which are susceptible or partially resistant to Bz treatment. These results suggest that early activation of the cellular compartment of the immune system by IL-12 may favor in vivo Bz activity against T. cruzi. In order to test this hypothesis mice infected with the drug-resistant Colombiana strain of T. cruzi were treated with 100 mg of Bz per kg plus different concentrations of rIL-12. By using the results of PCR and serological and parasitological methods as the criteria of a cure, our results indicate that a higher percentage of mice treated with Bz combined with rIL-12 than mice treated with Bz alone are cured.  相似文献

Six in vitro methods for the detection of anthelmintic resistance were compared using benzimidazole/levamisole-resistant Haemonchus contortus and benzimidazole/levamisole/ivermectin-resistant Ostertagia circumcincta as well as susceptible strains of both parasite species. The degree of resistance to thiabendazole and levamisole was compared by (1) an egg hatch assay, (2) an egg hatch paralysis assay, (3) a larval development assay, (4) a larval paralysis assay (5) a larval paralysis assay with physostigmine and (6) larval micromotility assay. The degree of resistance for each assay was expressed as resistance factor--RF. For the detection of thiabendazole and levamisole resistance, the larval development test was observed as the most sensitive to measure quantitatively a degree of resistance between susceptible and resistant strains. For this test the RF for thiabendazole and levamisole was 14.3 and >32.5, respectively in H. contortus strains and 21.1 and 3.5 in strains of O. circumcincta. Egg hatch assay was also found to be sensitive and accurate in determining of resistance to benzimidazole. For measurement of levamisole resistance the egg hatch paralysis assay and larval paralysis assay were found to be able to distinguish between strains, but some disadvantages of these techniques make it unsuitable for field detection of levamisole resistance. Other in vitro assays as larval paralysis assay with physostigmine and larval micromotility assay were also investigated. Significant differences in paralysis of the larvae were observed using larval paralysis assay.  相似文献

A total of 120 gastro-intestinal tracts and 960 faecal samples were examined to assess the prevalence and seasonal changes in the gastro-intestinal helminth parasites of Red Sokoto (maradi) goats slaughtered at Ibadan between May 1991 and April 1992. Egg types of strongyles, Strongyloides, Trichuris, Skrjabinema, Dicrocoelium and Moniezia were encountered in 93%, 83%, 44%, 0.9%, 2.3% and 31% of the faecal samples respectively. However, only strongyle, Strongyloides and Trichuris eggs occurred in large numbers and were more common during the rainy season than in the dry season. The parasites recorded and their prevalences were Haemonchus contortus (90.0%), H. ovis (5.0%), Strongyloides papillosus (80.8%), Trichostrongylus colubriformis (78.3%), T. axei (69.2%), Trichuris ovis (72.5%), T. globulosa (38.3%), Oesophagostomum columbianum (67.5%), Cooperia curticei (58.3%) Gaigeria pachyscelis (40.8%), Skrjabinema ovis (5.0%), Nematodirus battus (5.8%), Moniezia expansa (29.2%), M. benedeni (10.0%), Paramphistomum spp. (5.0%) and Cysticercus tenuicollis (33.3%). Haemonchus ovis is reported for the first time in Nigeria. Mixed infections were most prevalent. Young goats were more commonly infected and had higher worm counts than adult goats. Only Haemonchus, Trichostrongylus, Strongyloides and Cooperia spp. occurred in large numbers. Irrespective of the age of the goats, higher worm counts were generally encountered during the rainy season than in the dry season. The results are discussed in relation to the control of helminthiasis in grazing animals in Nigeria.  相似文献

The resistance status of gastro-intestinal nematodes to anthelmintics was evaluated on 881 sheep farms throughout Australia during 1991-92. Resistance was shown to be widespread. Overall, 85% of farms had sheep infected with nematodes resistant to benzimidazole, 65% to levamisole and 34% to combination (benzimidazole+levamisole) products. Resistance to ivermectin was not detected. On only 9% of farms did all anthelmintic groups reduce egg counts by greater than or equal to 95%. The culture of faeces from untreated sheep showed Telodorsagia circumcincta, Trichostrongylus spp, Chabertia ovina and Haemonchus contortus to be the principal species. The nature and prevalence of resistance was not significantly correlated with stocking rate. However, resistance to combination products was almost twice as prevalent on farms in areas with an average annual rainfall of greater than 500 mm.  相似文献

The efficacy of moxidectin was evaluated in young ponies naturally infected with gastrointestinal parasites. Eight animals were treated orally with moxidectin at 0.4 mg/kg bodyweight and eight received only the vehicle. They were all necropsied two weeks later. Faecal samples were examined daily for egg counts and larval cultures. Parasites were recovered from total faecal samples collected daily and from the gastrointestinal tracts at necropsy. Moxidectin reduced the strongyle egg counts by > 99 per cent from three days after treatment but some individuals remained positive for 10 days. The drug had little or no ovicidal activity. As evaluated in the critical-controlled test, moxidectin was 99 to 100 per cent effective (P < 0.05) against luminal stages of parascaris equorum, Strongylus vulgaris, Triodontophorus species, Craterostomum acuticaudatum, 19 cyathostome species and Oxyuris equi. Adult S edentatus were also completely removed. Its efficacy against third stage larvae of Gasterophilus intestinalis was 95 per cent (P < 0.05). Luminal nematode stages were removed within a few days, and bots continued to be eliminated for at least two weeks after treatment. No activity was observed against Anoplocephala perforliata. As evaluated in the controlled test, moxidectin was 100 per cent effective against Habronema muscae (P < 0.05) and had a 76 per cent but not significant efficacy against encysted small strongyle larvae.  相似文献
