Chicks were fed isocaloric and isonitrogenous diets containing 6% (w/w) added fat consisting of various proportions of animal tallow and flaxseed oil (FSO). No differences among treatments were seen in growth rate, muscular deposition of protein and lipids nor in the muscle phospholipid (PL) and triglyceride (TG) contents. Prostaglandin (PG)E2 synthesis in isolated skeletal muscle was depressed significantly by feeding FSO or by treatment with naproxen (6-methoxy-α-methyl-2-napthaleneacetic acid), an inhibitor of PG synthesis. The changes associated with diet may be related to differences in the fatty acid composition of muscle lipids. Levels of saturated fatty acids in muscle PL and TG were relatively insensitive to dietary treatments. Monounsaturated fatty acid levels were significantly lower in the FSO-fed groups. FSO diets caused significant depression in muscle PL 20∶4ω6 and almost completely inhibited 22∶5ω6 incorporation. FSO diets decreased ratios of ω6/ω3 fatty acids and increased the unsaturation index of muscle PL. Muscles of chicks fed FSO showed increased levels of 18∶3ω3, and of its derivatives 20∶4ω3 and 22∶5ω3. These results suggest that FSO inhibits PG synthesis and modifies the fatty acids of PL and TG of chick muscle. These changes may have implications for PG-dependent and/or membrane-dependent processes in muscle metabolism.  相似文献

Total lipid, phospholipid, and FA composition and distribution of FA between polar lipids (PL) and neutral lipids (NL) were investigated in the boreal soft coral Gersemia rubiformis from the Bering Sea. The total lipids were mostly hydrocarbons and waxes (33.7%) and PL (33.1%). The content of monoalkyldiacylglycerols (9.7%) exceeded the content of TAG (6.7%). PC and PE constituted 31.4% and 25.6% of total phospholipids, respectively. Principal FA were 16∶0, 16∶1n−7, 18∶0, 18∶1n−9, 18∶1n−7, 20∶1n−7, 20∶4n−6, 20∶4n−3, 20∶5n−3 22∶5n−3, 22∶6n−3, 24∶5n−6, and 24∶6n−3. Most n−6 PUFA (52% of total FA) were associated with the PL fraction; this was especially true for arachidonic and tetracosapentaenoic acids. The NL were enriched with mono-, di-, trienoic, and n−3 PUFA. The variation in EPA levels in both NL and PL suggests an origin of this acid from lipids of diatoms consumed by the corals.  相似文献

Diets rich in meat are claimed to contribute to the high tissue arachidonic acid (20∶4ω6) content in people in Westernized societies, but there are very few direct data to substantiate this assertion. Because meat contains a variety of long-chain polyunsaturated fatty acids (PUFA) that are susceptible to oxidation, we initially examined the effect of cooking on the long-chain PUFA content of beef, and then determined the effect of ingestion of lean beef on the concentration of long-chain PUFA in plasma phospholipids (PL). First, we examined the effect of grilling (5–15 min) and frying (10 min) different cuts of fat-trimmed lean beef on the long-chain PUFA content. Second, we investigated the effect of including 500 g lean beef daily (raw weight) for 4 wk on the fatty acid content and composition of plasma PL in 33 healthy volunteers. This study was part of a larger trial investigating the effect of lean beef on plasma cholesterol levels. In the first two weeks, the subjects ate a very low-fat diet (10% energy) followed by an increase in the dietary fat by 10% each week for the next 2 wk. The added fat consisted of beef fat, or olive oil (as the oil or a margarine) or safflower oil (as the oil or a margarine). This quantity of beef provided 60, 230, 125, 140 and 20 mg/d, respectively, of eicosatrienoic acid (20∶3ω6), 20∶4ω6, eicosapentaenoic acid (20∶5ω3), docosapentaenoic acid (22∶5ω3) and docosahexaenoic acid (22∶6ω3). Grilling for 10–15 min, but not frying, of the fat-trimmed lean beef resulted in 20–30% losses of the 20 and 22 carbon PUFA. The consumption of the lean beef during the first two-week period, when there was a very low level of dietary fat, was associated with significant increases in the proportion and concentration of 20∶3ω6, 20∶4ω6, 20∶5ω3 and 22∶5ω3 in the plasma PL and a significant decrease in the proportion and content of 18∶2ω6. The addition of beef fat or olive oil to the diets containing lean beef did not alter the plasma PL fatty acid profile compared with the very low-fat diet, whereas the addition of safflower oil maintained the significant increases in 20∶4ω6 and 22∶5ω3 but led to decreases in 18∶3ω3 and 20∶5ω3 compared with the very lowfat diet. The results showed that diets rich in lean beef increased the 20∶3ω6, 20∶4ω6 and the long-chain ω3 PUFA levels in the plasma PL. A high level of linoleic acid in diets rich in lean beef prevented the rise in the plasma level of 20∶3ω6 and 20∶5ω3, two fatty acids known to antagonize the effects of 20∶4ω6 on platelet aggregation.  相似文献

The purpose of this study was to assess the FA composition of both cholesteryl esters (CE) and phospholipids (PL) in maternal plasma during pregnancy and at delivery and in umbilical plasma at birth. A longitudinal study of 32 normal pregnant women was carried out with three cutoff points during pregnancy (first, second, and third trimester) and at delivery. Few significant differences occurred in the FA profile of maternal CE: 18∶1n−9 increased, 18∶2n−6 dropped slightly, and 18∶3n−3 decreased with progressing gestation. In maternal PL, long-chain highly unsaturated FA concentrations dropped and were replaced by saturated FA as gestation progressed. Additionally, changes in saturated FA in PL occurred: Shorter-chain 16∶0 was higher whereas longer-chain 18∶0 was lower at delivery compared to early pregnancy. The FA profile of umbilical venous plasma was strikingly different from that of maternal plasma at delivery. Cord plasma CE contained more saturated and monounsaturated FA than maternal CE. The polyunsaturates 18∶2n−6 and 18∶3n−3 are lower in umbilical CE than in maternal CE whereas 20∶4n−6 and 22∶6n−3 are twice as high in umbilical CE. Cord plasma PL have a higher content of long-chain highly unsaturated FA than maternal plasma PL at delivery. In contrast to maternal plasma PL, 16∶0 was lower and longer-chain saturated FA were higher in cord plasma PL. The FA profile of umbilical plasma at birth shows preferential accumulation of 20∶4n−6 and 22∶6n−3, with low concentrations of 18∶2n−6 and 18∶3n−3 in CE and PL, indicating a preferential supply of the fetus with long-chain highly unsaturated FA needed for fetal development.  相似文献

Several psychiatric disorders, including juvenile Attention Deficit/Hyperactivity Disorder (ADHD), have been associated with abnormalities of certain long-chain PUFA (LCPUFA). Despite this reported association, the FA levels of patients with the adult form of ADHD have not previously been evaluated. In this study we measured the total blood phospholipid FA concentrations in 35 control subjects and 37 adults with ADHD symptoms to determine whether adults with ADHD symptoms would show abnormalities of FA relative to control subjects. In the serum phospholipids, adults with ADHD symptoms had significantly lower levels of total saturated, total polyunsaturated, and total omega-6 (n−6) FA, as well as the omega-3 (n−3) LCPUFA DHA (22∶6n−3), and significantly higher levels of total monounsaturated FA and the n−3 LCPUFA docosapentaenoic acid (22∶5n−3). In the erythrocyte membrane phospholipids, adults with ADHD symptoms had significantly lower levels of total PUFA, total n−3 FA, and DHA, and significantly higher levels of total saturated FA. Neither serum nor erythrocyte membrane phospholipid DHA was related to ADHD symptom severity (as assessed by the Amen questionnaire) in ADHD subjects. Although the exact cause of these variations is unknown, both environmental and genetic factors may be involved.  相似文献

Rat peri-renal and epididymal pre-adipocytes in culture undergoing triglyceride (TG) accumulation were incubated with oleic (18∶1), linoleic (18∶2), α-linolenic (18∶3ω3), arachidonic (20∶4) and 4,7,10,13,16,19-docasahexaenoic (22∶6ω3) acids in the presence of 0.8 μM insulin. The fatty acids were incorporated in cellular TG with relative enrichments over control from 1.4-fold for 18∶1 to greater than 40-fold for 18∶3ω3. Greater than 80% of fatty acids taken up were incorporated into cellular TG. The balance was distributed, in decreasing amounts, into phospholipids, unidentified intracellular constituents, and ketone bodies. The P/S ratio of cellular TG was at least an order of magnitude lower than that of the external milieu for both cell types and for all treatment groups, including controls. Doubling the concentration of treatment fatty acid increased its incorporation into cellular TG. However, it did not affect the accumulation of the other fatty acids in TG. Epididymal cells consistently acquire a higher proportion of treatment fatty acids in cell TG than peri-renal cells. Pre-adipocytes with polyunsaturated fatty acids (PUFA)-enriched TG is a potential model for the study of PUFA metabolism in these types of cells  相似文献

This study examined the effect on the plasma lipids and plasma phospholipid and cholesteryl ester fatty acids of changing from a typical western diet to a very low fat (VLF) vegetarian diet containing one egg/day. The effect of the addition of saturated, monounsaturated or polyunsaturated fat (PUFA) to the VLF diet was also examined. Three groups of 10 subjects (6 women, 4 men) were fed the VLF diet (10% energy as fat) for two weeks, and then in the next two weeks the dietary fat in each group was increased by 10% energy/week using butter, olive oil or safflower oil. The fat replaced dietary carbohydrate. The VLF diet reduced both the low density lipoprotein (LDL)-and high density lipoprotein (HDL)-cholesterol levels; addition of the monounsaturated fats and PUFA increased the HDL-cholesterol levels, whereas butter increased the cholesterol levels in both the LDL- and HDL-fractions. The VLF diet led to significant reductions in the proportion of linoleic acid (18∶2ω6) and eicosapentaenoic acid (20∶5ω3) and to increases in palmitoleic (16∶1), eicosatrienoic (20∶3ω6) and arachidonic acids (20∶4ω6) in both phospholipids and cholesteryl esters. Addition of butter reversed the changes seen on the VLF diet, with the exception of 16∶1, which remained elevated. Addition of olive oil resulted in a significant rise in the proportion of 18∶1 and significant decreases in all ω3 PUFA except 22∶6 compared with the usual diet. The addition of safflower oil resulted in significant increases in 18∶2 and 20∶4ω6 and significant decreases in 18∶1, 20∶5ω3 and 22∶5ω3. These results indicate that the reduction of saturated fat content of the diet (<6% dietary energy), either by reducing the total fat content of the diet or by exchanging saturated fat with unsaturated fat, reduced the total plasma cholesterol levels by approximately 12% in normocholesterolemic subjects. Although the VLF vegetarian diet reduced both LDL- and HDL-cholesterol levels, the long-term effects of VLF diets are unlikely to be deteterious since populations which habitually consume these diets have low rates of coronary heart disease. The addition of safflower oil or olive oil to a VLF diet produced favorable changes in the lipoprotein lipid profile compared with the addition of butter. The VLF diets and diets rich in butter, olive oil or safflower oil had different effects on the 20 carbon eicosanoid precursor fatty acids in the plasma. This suggests that advice on plasma lipid lowering should also take into account the effect of the diet on the fatty acid profile of the plasma lipids.  相似文献

The fatty acid (FA) compositions of plasma and oviduct phospholipids (PL) and triglycerides (TG) were studied throughout the natural sexual development of the Japanese quail. In the oviduct, PL concentration increased rapidly during the period of active oviduct cell proliferation and then remained at a constant level during the phase of cellular hypertrophy. Oviduct and plasma TG concentrations were 2- and 10-fold higher, respectively, in fully developed animals than in immature ones. During natural sexual maturation of the quail, the FA compositions of PL and TG were markedly modified both in plasma and in oviduct. These qualitative changes occurred predominantly during the period of intense cellular proliferation of oviduct cells, and also were observed in immature animals injected with physiological doses of estradiol. In oviduct PL, the proportions of 20∶4n−6 and 22∶4n−6 decreased significantly (from 20 to 10% and 3.5 to 0.7%, respectively) whereas those of 18∶2n−6 increased (from 8.5 to 21%). In contrast, the plasma PL proportions of 20∶4n−6, 22∶4n−6 and 18∶2n−6 were decreased significantly and the percentage of 18∶1n−9 doubled, suggesting that the oviduct is able to utilize certain plasma FA to a greater extent than others. Changes in plasma and oviduct lipid composition occurring in the quail during sexual development may be attributed to estradiol, which stimulates hepatic Δ9 desaturase and inhibits the oviduct Δ6 desaturase. The changes in FA composition observed in oviduct phospholipids are discussed in relation to eicosanoid production and cellular proliferation.  相似文献

The lipid and FA composition of the total lipids of the pearl oyster Pinctada fucata martensii, in different seasons and in different areas, were analyzed to clarify its lipid physiology and to estimate the possible influence of its prey phytoplankton. TAG and sterols were the major components in the neutral lipids in all conditions, whereas high levels of phospholipids (PE and PC) were found in the polar lipids. The major FA in the TAG in all samples were 14∶0, 16∶0, and 18∶0 as saturated FA (saturates); 16∶1n−7, 18∶1n−9, and 18∶1n−7 as monoenoic FA (monoenes); and 20∶4n−6 (arachidonic acid: AA), 20∶5n−3 (EPA), and 22∶6n−3 (DHA) as PUFA. The major components found in the polar lipids were 16∶0 and 18∶0 as saturates; 22∶2n−9, 15 and 22∶2n−7, 15 as non-methylene-interrupted dienes (NMID), and AA, 22∶3n−6, 9, 15, EPA, and DHA as PUFA. Although it is a marine animal, characteristically high levels of AA were found in both the TAG and phospholipids. This result suggests that lipids of P. fucata may be influenced by those of its phytoplanktonic prey. The increase in levels of NMID from TAG to PE with a decrease in those of monoenes suggests that the tissues of this species are able to biosynthesize only the less unsaturated PUFA, such as NMID. In particular, NMID derivatives are considered to be biosynthesized in the PE; thus, they might play a particular role in the membrane, because NMID were characteristically localized only in the PE.  相似文献

We have investigated the effect of oleic (18∶1) and 4,7,10,13,16,19-docosahexaenoic (22∶6ω3) acids on triglyceride (TG) accumulation, secretion and reuptake in rat hepatocytes in culture. We also calculated the percentage of total TG, TG-esterified 18∶1 and TG-esterified 22∶6ω3 that were secreted relative to the total accumulation (intra+extracellular TG). Both fatty acids were incorporated mainly in the intracellular TG fraction. Treatment with 18∶1 but not with 22∶6ω3 increased the quantity of TG secreted into the culture medium relative to controls. Treatment with 22∶6ω3 caused a greater accumulation of intracellular TG than 18∶1. This arises in part from the preferential retention of 22∶6ω3-enriched TG by the hepatocytes. At 24 hr, there was no longer any difference in the net secretion of TG by 18∶1 and 22∶6ω3-treated cells, which may be a consequence of the reuptake of TG-esterified 18∶1. There was no reuptake of TG-esterified 22∶6ω3. We conclude that inhibition of hepatocyte TG synthesis is not obligatory for 22∶6ω3-induced diminution of TG secretion.  相似文献

The effects of clofibrate on the content and composition of liver and plasma lipids were studied in mice fed for 4 wk on diets enriched in n−6 or n−3 polyunsaturated fatty acids (PUFA) from sunflower oil (SO) or fish oil (FO), respectively; both oils were fed at 9% of the diet (dry weight basis). Only FO was hypolipidemic. Both oil regimes led to slightly increased concentrations of phospholipids (PL) and triacylglycerols (TG) in liver as compared with a standard chow diet containing 2% fat. Clofibrate promoted hypolipidemia only in animals fed SO. Its main effect was to enlarge the liver, such growth increasing the amounts of major glycerophospholipids while depleting the TG. SO and FO consumption changed the proportion of n−6 or n−3 PUFA in liver and plasma lipids in opposite ways. After clofibrate action, the PUFA of liver PL were preserved better than in the absence of oil supplementation. However, most of the drug-induced changes (e.g., increased 18∶1n−9 and 20∶3n−6, decreased 22∶6/20∶5 ratios) occurred inrrespective of lipids being rich in n−6 or n−3 PUFA. The concentration of sphingomyelin (SM), a minor liver lipid that virtually lacks PUFA, increased with the dietary oils, decreased with clofibrate, and changed its fatty acid composition in both situations. Thus. oil-increased SM had more 22∶0 and 24∶0 than clofibrate-decreased SM, which was significantly richer in 22∶1 and 24∶1.  相似文献

The lipid, FA, and sterol composition of the New Zealand green lipped mussel (NZGLM, Perna canaliculus) and of the Tasmanian blue mussel (TBM, Mytilus edulis) were compared using TLC-FID and GC-MS. The respective mussel species were obtained from three different sites in both New Zealand (NZ) and Tasmania. Lipid class distribution of both mussel species was characterized by a high proportion of phospholipid (PL, 57–79%) and TG (10–25%), FFA (7–12%), and sterols (ST, 12–18%). The NZGLM had higher proportions of TG, FFA, and ST (P<0.01), whereas the TBM had a higher proportion of PL (P<0.01). There were higher proportions of total PUFA, saturated FA, n−3 FA, and hydroxy and nonmethyleneinterrupted FA (P<0.05) in the TBM compared with the NZGLM. The major FA in the NZGLM were 16∶0 (15–17%), 20∶5n-3 (14–20%), and 22∶6n-3 (11–17%). The same FA dominated lipids in the TBM, although there were significantly higher proportions of 16∶0 (P=0.000) and 22∶6 n−3 (P=0.003) and lower proportions of 20∶5n-3 (P=0.0072) in the TBM. A novel PUFA, 28∶8n-3, was detected in both mussels with higher amounts in the TBM, which probably reflects a greater dietary contribution of dinoflagellates for this species. Cholesterol was the dominant sterol in both mussels. Other major sterols included brassicasterol, 22-methylcholesterol, trans-22-dehydrocholesterol, and desmosterol. There were significant differences (P<0.05) between the NZGLM and TBM for 12 of the 20 sterols measured. Six sterols showed significant site differences for the NZGLM, and 10 for the TBM. The differences in the FA and sterol composition between the two species may be due to the diet of the NZGLM being more diatom-derived and the diet of the TBM having a greater dinoflagellate component.  相似文献

The activity of the overt form of rat liver mitochondrial carnitine palmitoyltrasferase or CPT0 (EC 2.3.1.21) towards different fatty acid substrates was studied. The following non-esterified fatty acids (NEFA) and their CoA esters in the presence of 1% bovine serum albumin (BSA) were tested: 16∶0, 18∶0, 18∶1, 18∶2, 18∶3ω3, 20∶4, 20∶5ω3 and 22∶6ω3. The data fit a square hyperbolic model for enzyme catalysis (p<0.001, non-linear regression). Asymptotic Vmax and K0.5, substrate concentration at one-half Vmax, were calculated using total concentrations of acyl-CoA, or unbound concentrations of NEFA. BSA was found to act as a true substrate reservoir for NEFA in that the dissociation of the NEFA-BSA complex was 10–330 times faster than the CPT0 reaction. Regardless of form (NEFA or CoA ester), 18∶3ω3 gave the highest, while 22∶6ω3 and 18∶0 gave the lowest rates of acylcarnitine synthesis. Except for 18∶3ω3 and 18∶2, Vmax for NEFA was generally lower than for acyl-CoA, with the greates differences observed for 20∶4, 20∶5ω3 and 22∶6ω3, suggesting that acyl-CoA synthesis may also be important in the control of the entry of these fatty acids into the mitochondria. The data provide an enzymatic rationale for the relatively low content of 18∶3ω3 in esterified lipid.  相似文献

Hyperphagia was achieved by continuous intracerebroventricular infusion of a melanocortin receptor antagonist (HS024; Neosystem, Strasbourg, France) in rats. The effects of hyperphagia on FA composition and concentration of plasma phospholipids (PL), plasma FFA, and adipose tissue TAG were studied in rats for 8 d [short-term hyperphagia (STH); n=8], or 28 d [longterm hyperphagia (LTH); n=9]. The control rats were treated with artificial cerebrospinal fluid for 8 d (n=8) or 28 d (n=10). The rats were fed the same regular diet. In STH rats the plasma PL and fasting plasma FFA contained higher concentrations of saturated FA (SFA) and monounsaturated FA (MUFA), and plasma FFA contained lower n−6 PUFA than in the control rats. In LTH rats the plasma PL contained higher concentrations of SFA, MUFA, and n−3 PUFA and higher proportions of 16∶1n−7 and 18∶1n−9 at the expense of 18∶2n−6 than in the control rats. In LTH rats the abundant dietary intake of 18∶2n−6 did not enrich 18∶2n−6 of the plasma PL or adipose tissue TAG. In LTH rats the fasting plasma FFA contained more than twofold higher concentrations of SFA and MUFA, and higher proportions of 16∶1n−7 and 18∶1n−9 at the expense of 18∶2n−6 than in the control rats. This animal obesity model shows that LTH affects the FA composition and concentration of plasma PL, plasma FFA, and adipose tissue TAG, a result consistent with changes associated with increased risk of various diseases in humans. These results also demonstrate that LTH alters the FA composition of plasma PL and adipose tissue TAG in a way that does not reflect the FA composition of dietary fat.  相似文献

The lipid and fatty acid composition of muscle of 10 species of freshwater fish obtained from a market of Shanghai City was examined. Total lipids (TL) ranged over 0.9–4.7% of muscle for all samples. The content of triacylglycerol (TG) in muscle ranged over 0.2–3.4% and that of polar lipids (PL) was 0.5–1.3%. Differences of TL content were dependent on TG contents. The predominant important fatty acids (>10% of the total fatty acids in TL) were 16∶0 and 18∶1n−9 with some 16∶1n−7, 18∶2n−6, and 22∶6n−3. The polyunsaturated fatty acids (PUFA) content was 10.2–43.4%, and especially Chinese sea bass contained above 20% of 22∶6n−3 in the total fatty acids. There were higher levels of PUFA such as 20∶5n−3 and 22∶6n−3 in PL than in neutral lipids. Muscle of the silver carp was stored at 20°C, and changes of lipid classes during storage were examined. Free fatty acids increased, and PL decreased during storage. This phenomenon was inhibited by heating the muscle, suggesting that lipid hydrolysis by phospholipase occurred in silver carp muscle.  相似文献

The influence of age and cholesterol on polyunsaturated fatty acids (PUFa) levels was studied in young and old male Sprague-Dawley rats. Animals were fed a fat-free diet supplemented with 10% (by wt) safflower oil with or without 1% cholesterol for 8 wk. As a result of cholesterol feeding, proportions of linoleic acid (18∶2n−6) and dihomo-γ-linolenic acid (30∶3n−6) were increased and and that of arachidonic acid (20∶4n−6) was decreased in the liver and platelet phospholipids in 64-wk-old rats, suggesting inhibitory effects of cholesterol on 20∶4n−6 synthesis from 18∶2n−6. The prominent age-dependent effect on the levels of PUFA was a retention of C−22 n−3 PUFA, accompanied by decreased C−22 n−6 PUFA and increased 20∶3n−6 in the liver and platelet phospholipids. Ratio of 20∶3n−6/20∶4n−6 increased in 64-wk-old rats regardless of dietary cholesterol, suggesting depressed Δ5-desaturase with age. In aorta phospholipids, 20∶3n−6 content and 20∶3n−6/20∶4n−6 ratio increased with cholesterol supplementation, but not with age. These results suggest that changes of PUFA composition of platelet phospholipids with age are closely linked with changes in liver phospholipids. The 20∶4n−6 content in both platelet and aorta phospholipids is kept constant, despite other n−6 and n−3 PUFA being affected by age.  相似文献

We have previously shown that a continuous infusion of tumor necrosis factor-α (TNF-α) in rats results in an increase in plasma triglyceride (TG), liver protein and DNA, and at the same time a reduction in muscle protein. However, there is no information on the associated effects of TNF-α on liver and muscle lipids. The present study, therefore, determined the effect of TNF-α on the TG and phospholipid (PL) content and their fatty acid composition, in the liver and carcass of rats and correlated with the plasma levels of insulin, corticosterone, and catecholamaines. Total parenteral nutrition that met the daily nutrient requirements was continuously infused for 10 d through a central vein catheter in two groups of rats, one receiving infused TNF-α (100 μg/kg/d) and a control group. Hepatic TG and PL, expressed either as mg/g dry weight or total organ content, were significantly increased in the TNF-α-infused group compared with controls. Livers of TNF-α infused rats contained significantly less saturated and monounsaturated fatty acids and significantly more polyunsaturated fatty acids (PUFA) of the ω3 and ω6 series than controls. The carcass, on the other hand, contained significantly more monounsaturated and significantly less polyunsaturated fatty acids of the ω6 series than controls. There were no changes in corticosterone level. Although plasma glucose levels were comparable between control and TNF-α infused rats, the latter had a marked increase in insulin levels, demonstrating insulin resistance. In addition TNF-α infused rats had raised norepinephrine levels. The fall in carcass and rise in liver TG and PL are consistent with the presence of insulin resistance and raised norepinephrine levels. However, the increase in PUFA content of the liver, especially the increase in 20∶4ω6 levels in liver PL and TG, is likely to be the effect of TNF-α and suggests increased activity of Δ5-desaturase.  相似文献

Very long chain (VLC) PUFA of the n−6 and n−3 series are known to occur in mammalian testis. The aim of this work was to characterize further two testicular lipid classes with VLCPUFA, cholesterol esters (CE) and total triglycerides (TG) in rat and mouse testis. The VLCPUFA predominating in these lipids were a series of n−6 pentaenes and tetraenes with 24 to 32 carbons, including small amounts of odd-chain PUFA, 28∶5n−6 and 24∶5n−6 prevailing in CE and TG, respectively. Most of the VLCPUFA of TG were concentrated in a small fraction of TG, made up by 1-O-alkyl-2,3-DAG. This TG subclass was absent altogether from the TG of sexually immature testis. The TG and the CE with VLCPUFA only occurred in testis of adult fertile animals. The proportion of VLCPUFA in total TG and CE was higher in rodents than in other mammals. In the n−6 PUFA-rich adult mouse testis, the amounts of testicular triacylglycerols decreased significantly after consumption of fish oil for 2 wk. Whereas 18∶2n−6 was significantly reduced, the amounts of 22∶5n−6 and longer n−6 PUFA were less affected in all mamor testicular lipids including PC and PE, where they were unchanged. The 1-O-alkyl-2,3-DAG and their n−6 VLCPUFA were virtually unaffected by the diet. The VLCPUFA-containing molecular species of CE and TG may represent a form of storage of cholesterol and polyenoic FA required to sustain spermatogenesis. Via chain-shortening, VLCPUFA stored in the neutral lipids may serve as precursors of the major C22 PUFA typical of cell membrane glycerophospholipids, protecting testicular cells against shifts in FA composition induced by dietary changes.  相似文献

Existence of a dietary maximal level or threshold for incorporation of ω3 fatty acids into membrane phospholipids is of interest as it may further define understanding of the dietary requirement for ω3 fatty acids. To test whether feeding increasing levels of dietary ω3 fatty acids continues to increase membrane ω3 fatty acid content, weanling rats were fed a nutritionally adequate semipurified diet which provided increasing amounts of C20 and C22 ω3 fatty acids, such as 20∶5ω3 and 22∶6ω3. Dietary 20∶5ω3 and 22∶6ω3 were provided by substituting a purified shark oil concentrate of high 22∶6ω3 content for safflower oil high in 18∶2ω6. After four weeks of feeding, nuclear envelopes from four animals in each diet group were prepared, lipid was extracted and phospholipids separated. Arachidonic acid content in membrane phosphatidylcholine, phosphatidylethanolamine, phosphatidylinositol and phosphatidylserine was significantly reduced by feeding increased dietary levels of ω3 fatty acids. Decline of 20∶4ω6 level in phospholipid tended to stabilize when the dietary content of total ω3 fatty acids reached 4–5% of total fatty acids. Above this level, dietary ω3 fatty acids did not result in a further decrease in membrane content of 20∶4nω6. Increase in membrane phospholipid content of 20∶5ω3 occurred as the dietary intake of ω3 fatty acids increased from 1.1% to 5% of total fatty acids. A dietary ω3 fatty acid level of 2.2–3% was sufficient to result in maximum incorporation of 22∶6ω3 into membrane phosphatidylcholine and phosphatidylethanolamine, but not into phosphatidylinositol or phosphatidylserine.  相似文献

The fatty acid (FA) composition and distribution in a variety of phospholipids (PL) and neutral lipids (NL) at two discrete stages during the embryonic rat brain development were investigated. Over 96% of the FA were acylated into fetal brain PL at embryonic day 17 after the peak of neuronal proliferation and at embryonic day 20, one day prior to delivery. Phosphatidylcholine constituted approximately 60% of the total PL pool, phosphatidylethanolamine (PE) 30%, phosphatidylserine (PS) 6%, and phosphatidylinositol (PI) 4%. The diacylglycerols and triacylglycerols constituted 1–2% of the fetal brain lipids. α-Linolenic acid (18∶3n−3) and linoleic acid (18∶2n−6) were found in very low amounts in all fetal brain PL and NL. The percentage of the n−6 polyunsaturated FA, consisting of arachidonic acid (AA), 22∶4n−6 and 22∶5n−6, remained unchanged in all the fractions, except in Pl, in which the proportion of AA increased. The concentration of docosahexaenoic acid (DHA) increased with age in all the fractions, with the bulk of accumulation accounted for by its increase in PE and, to a lesser extent, in PS. This finding suggests a “DHA accretion spurt” during the last three days of pregnancy.  相似文献
