从精浆前列腺特异抗原的分离纯化及鉴定   总被引:1,自引:0,他引:1  
用PEG-SephadexG-150两步法,从精浆中分离纯化前列腺特异抗原(PSA)。精浆标本先经PEG粗提,约去除80%的杂蛋白,再经柱层析,进一步提纯,提纯产物经SDS-PAGE及ELISA法证实为免疫纯。  相似文献

目的 分离提纯肌钙蛋白I(TnI)以便制备心肌梗塞诊断试剂。方法 采用离子交换、分子筛、疏水 层析相结合的方法分离纯化兔骨骼肌肌钙蛋白C(sTnC).制备TnG-Sepharose4B亲和层析柱。结果 所提纯的sT- nC,经SDS-PAGE分析已达到电泳纯,经UV-250扫描显示出特异性吸收曲线,证明其含有较多的苯丙氨酸,而不含 色氨酸,进一步证明其纯度。结论 为提纯TnI创造了条件。  相似文献

浓缩Vero细胞乙脑疫苗经SepharoseCL-6B柱层析可去除大部分杂蛋白,再经SphacrylS-500HR层析可进一步去除比病毒大的杂质及残余小分子量杂蛋白。经两步凝胶过滤层析化后,疫苗总蛋白含量减少约99%,抗的比活怀提高87-165倍,抗原回收率达30-50%,残余牛血清和残余细胞DNA均符合WHO有关规程要求,提纯疫苗的效力达到或超过中国地鼠肾灭惭脑疫苗和日本提纯鼠脑疫苗的参考苗。  相似文献

为了提高特种炸药爆轰合成纳米氧化铈的纯度, 采用酸洗方法提纯, 设计正交实验研究了提纯温度、时间, 硝酸、蒸馏水和聚乙二醇(PEG)加入量对提纯结果的影响。利用X射线衍射仪、X射线荧光光谱仪分析氧化铈提纯前后的晶型及杂质含量, 以氧化铈纯度为判定标准, 确定最佳提纯参数。结果表明:提纯温度过高会导致硝酸浓度降低, 时间过长会使过多氧化铈溶解;利用透射电镜观察颗粒表面, 加入PEG可改善纳米氧化铈颗粒团聚现象。最佳提纯参数为:温度80℃, 时间24h, 硝酸量0.333mL/g, 蒸馏水量6.666mL/g, PEG量16.666mg/g, 利用此参数提纯后的纳米氧化铈纯度为99.59%。分析杂质残留的原因为爆轰高温条件使Fe2O3和Al2O3转化为α-Fe2O3和α-Al2O3, 导致0.12%Fe2O3和0.17% Al2O3杂质残留。  相似文献

对聚乙二醇(PEG)-碱金属盐(MX)高分子固体电解质型抗静电剂在感光材料中的应用进行了研究,并考察了PEG的分子量、不同碱金属盐、明胶溶液的pH值和涂层厚度等对抗静电性能的影响。结果表明,PEG-MX体系具有优良的抗静电性能,尤其是在低湿度和真空环境下。  相似文献

对醇类(甘油和PEG-4000)、脲类及胺类活性剂的透明性、变色情况、加工性能及硫化性能进行了对比试验。结果表明,PEG-4000配料方便,混炼时易分散,硫化胶的物理性能最佳,变色程度居中,是最的白黑活性剂;脲类活性剂最透明,变色最轻,可以减小用量与PEG-4000并用;甘油的活化性能较好,但配料误差大,工艺性能不稳定,胺类活性剂变色最重,活化性能不好,故二者不宜使用。  相似文献

采用离子交换层析纯化了大肠杆菌表达的重组人肿瘤坏死因子(rhTNF-α),考察了PEG在离子交换层析中的作用,包括PEG分子量和浓度等的影响,初步探索了PEG和蛋白质相互作用机理。少量残余的杂蛋白继以疏水相互作用层析除去,获得了比活性为3.21×107u·mg-1的rhTNF-α,纯化倍数为253,总回收率为91.3%。  相似文献

目的 建立简便、适合大规模肾综合征出血热灭活疫苗的纯化方法。方法 采用超滤浓缩、醋酸锌沉淀和柱层析(亲和层析、离子交换层析和凝胶过滤层析)等方法,对出血热原代地鼠肾细胞灭活疫苗进行提纯对比试验。结果 用超滤浓缩和Sepharose 4FF凝胶柱层析或用醋酸锌沉淀和凝胶柱层析,纯化的疫苗均可达到质量标准,而用 cellufine sulfate gel亲和层析和离子交换层析均不理想。结论 建立了经济、适合大规模肾综合征出血热灭活疫苗的纯化方法。  相似文献

应用聚乙二醇(PEG)沉淀结合层析技术纯化乙肝病毒表面抗原(HBsAg),对PEG浓度、pH值、离子强度等影响PEG沉淀的因素进行了正交实验研究. 结果表明,浓度为0.12 g/mL的PEG6000在4℃, pH 9.0条件下沉淀HBsAg纯化效果比较理想,纯化倍数达3.7,回收率96.8%,有效地去除了生物大分子杂质和牛血清白蛋白(BSA),PEG在后续的凝胶过滤中可以除去,整个工艺的回收率可提高到41%.  相似文献

Catalase from Amsonia orientalis was purified by ATPS, and its efficiency was compared against hydrophobic interaction chromatography. Activity recovery and purification fold of purified catalase by ATPS were examined under varying experimental conditions. The effects of various factors such as type of phase-forming salts, (PEG) mass, with their different concentrations, pH and temperature effects on partitioning were investigated. The highest activity recovery (156%) and purification fold (8.67) of catalase were obtained in the ATPS system containing 10% (g/g) PEG4000, 15% (g/g) Na2SO4 at pH 6.0 and room temperature. In hydrophobic interaction chromatography, the enzyme has been purified 12.54-fold with 57.5% recovery. The molecular weight of catalase was determined as 75 kDa by SDS-PAGE.  相似文献

Sequential precipitation with polyethylene glycol (PEG) and KCl followed by preparative anion exchange high-performance liquid chromatography was used to isolated acid phosphatases (APases) from germinating soybean seeds. KCl inhibited the PEG precipitation of APases. Thus individual APases can be isolated directly from crude extracts by controlling PEG and KCl concentrations. Four of the isolated APases were phytases. APases 3 and 5 exhibited optimal activity at pH 4.5 and and 5.8. Phytase activity was at its peak at 40°C for APase 4 and at 60°C for APases 3, 5, and 6. This isolation method can be used to identify endogenous and/or exogenous phytases that are suitable for phytic acid hydrolysis in many food and feed systems, especially during the processing. of oilseeds and cereal-based ingredients.  相似文献

Chlorohydrin water‐swellable rubber—composed of chlorohydrin rubber (CHR), crosslinked polyacrylate (CPA), the amphiphilic compatibilizer poly(vinyl alcohol)‐g‐poly(butyl acrylate) (PVA‐g‐PBA), precipitated silica (PSA), and poly(ethylene glycol) (PEG)—was prepared. The dispersion of PEG in the blend was characterized by wide‐angle X‐ray diffraction (WAXD). The dependence of the water absorbing ratio by weight, the water‐swelling ratio by volume, and the percentage loss by weight on PEG and PSA contents were investigated. The effects of PEG and PSA on the second water‐swelling behaviors and long‐term water‐retention behaviors were also studied. 目的纯化重组人睫状神经营养因子(rhCNTF)。方法破菌液上清经硫酸铵分级沉淀和G-25脱盐后,用QSepharose FF阴离子交换层析初纯,再经Superdex 75 prepgrade凝胶过滤精制,并对纯化产物进行各项检测。结果纯化的rhC-NTF纯度达95%以上,蛋白浓度约为2mg/ml,收率约30%,相对分子质量21600,等电点为6.15,与理论值相符合。Western blot检测证明纯化蛋白能够与特异性抗体结合,并能够明显刺激鸡胚背根神经节突触生长。结论采用盐析、离子交换、凝胶过滤等方法有机组合,成功分离纯化了rhCNTF蛋白。  相似文献

分离纯化了自行筛选的耐有机溶剂的地衣芽孢杆菌Bacillus licheniformis YP1所产的溶剂稳定性蛋白酶. 发酵液经硫酸铵沉淀、疏水层析及强阳离子交换层析纯化,得到电泳纯的蛋白酶;分子量约为28 kDa,纯化蛋白酶的比酶活达到1.18′105 U/mg,纯化倍数为37.2,酶活回收率为20.8%. 纯化的YP1蛋白酶对50%(j)的多种亲水或疏水有机溶剂具有很高的耐受性,其中50%(j)的DMF和DMSO能显著促进YP1蛋白酶活力. YP1蛋白酶为Zn2+蛋白酶,最适反应温度为55℃,最适反应pH为10.0,pH 8.0~13.0范围内具有高活力. 以酪蛋白为底物,YP1蛋白酶的米氏常数Km为0.048 g/L.  相似文献

An extracellular NP-degrading enzyme secreted by Bacillus cereus.Frankland was purified to homogeneity by a combination of ammonium sulfate precipitation,Phenyl-Sepharose hydrophobic-interaction chromatography and DEAE anion-exchange chromatography.On SDS(sodium dodecyl sulfate)-polyacrylamide gel electrophoresis analysis,the purified enzyme showed a relative molecular mass of 58.3 kDa.The depolymerzation of subunits was accompanied with the loss of NP-degrading enzyme activity,and removing denaturing factors by dialysis could restore the dimer structure and enzymatic activity.The enzyme had an isoelectric point of 5.5 and an optimal temperature of 60℃,and was the most active at pH 6.0.The enzymatic activity was stable at pH 4-8 and inhibited by Cu2+.TenN-terminal amino acids were determined to be ASVNSIKIGY,demonstrating that the purified enzyme was a novel one.The hydrolysis pattern of the purified enzyme indicated that the NP-degrading enzyme was an endo NP-degrading enzyme.The extraordinary thermo-stability provided the enzyme with a good prospect to be used as a new tool for clean-production process for textile industry.  相似文献
