Paints are composed of an extensive variety of hazardous substances, such as organic solvents and heavy metals. Biomonitoring is an essential tool for assessing the risk to occupational health. Thus, this study analyzed the levels of biomarkers of exposure for toluene, xylene, styrene, ethylbenzene, and lead, as well as the oxidative stress biomarker alterations in painters of an industry. Lipid peroxidation biomarker (MDA), δ-aminolevulinate dehydratase (ALA-D), nonprotein thyol groups, superoxide dismutase and catalase (CAT) were analyzed in exposed and nonexposed subjects. We estimated which of the paint constituents have the greatest influence on the changes in the biomarkers of oxidative stress in this case of co-exposure. The results demonstrated that despite the fact that all the biomarkers of exposure were below the biological exposure limits, the MDA levels and antioxidant enzyme activities were increased, while nonprotein thyol groups and ALA-D levels were decreased in painters when compared with nonexposed subjects. After statistic test, toluene could be suggested as the principal factor responsible for increased lipid peroxidation and inhibition of ALA-D enzyme; however, further studies on the inhibition of ALA-D enzyme by toluene are necessary.  相似文献

In a total of 395 subjects with varying fish consumption habits, mercury levels in whole blood (B-Hg), and selenium levels in plasma (P-Se) were studied. Also, in subcohorts, mercury levels in blood cells (Ery-Hg; n = 79), plasma (P-Hg; n = 158) and urine (U-Hg; n = 125) were analysed. There were statistically significant associations between fish intake on the one hand, and B-Hg, Ery-Hg and P-Hg, on the other, but not so with U-Hg. In subjects who never had fish, the average B-Hg was 1.8 ng/g, in subjects who had at least two fish meals each week, 6.7 ng/g. Ery-Hg, and to a less extent P-Hg, were associated with levels of marine n-3 polyunsaturated fatty acids (PUFA) in serum phosphatidylcholine. P-Hg and U-Hg were associated with numbers of teeth with amalgam fillings. P-Se also correlated with fish intake. In subjects who never had fish, P-Se averaged 80 micrograms/l, in subjects who had at least two fish meals per week, 91 micrograms/l. There was an association between PUFA and P-Se. Further, there were statistically significant associations between P-Se on the one hand, and B-Hg, Ery-Hg and P-Hg on the other. The data clearly demonstrate the importance of fish for the exposure to methylmercury and selenium in the Swedish diet, and the impact of amalgam as a source of exposure to inorganic mercury.  相似文献

The aim of the study was to evaluate the impact of environmental contaminants on oxidative stress biomarkers in hepatocytes of Indian estuarine water grey mullet, Mugil cephalus collected from unpolluted Kovalam and polluted Ennore estuaries. Initially, a comparison was made between the general water chemistry and environmental pollutants like heavy metals to identify the ecotype of the estuaries. Biomarker responses and bioaccumulation of metals were determined along with histological studies of fish hepatocytes to assess contamination impact. Water chemistry data with higher temperature, salinity, pH, biochemical oxygen demand, chemical oxygen demand and low dissolved oxygen and environmental pollutants with higher metal concentration (p < 0.05) were observed at Ennore than Kovalam indicating its polluted environment. A significant increase (p < 0.05) in lipid and protein oxidation markers, decrease (p < 0.05) in thiol status and antioxidant enzyme activities were observed in the Ennore fish hepatocytes compared to Kovalam counterpart. Bioaccumulation of heavy metals was significantly higher (p < 0.05) in fish from Ennore. Hepatocytes from Ennore also featured extensive lipid-type vacuolation, increased size and membrane disruption. All the findings highlighted the value of oxidative stress biomarkers and membrane disruption as the sensitive parameters of environmental pollutant contamination and their importance in biomonitoring of aquatic ecosystems. This is also the first such attempt reported at the cellular level from South India stressing the importance of biomarkers in biomonitoring programmes using fish hepatocytes as the model system.  相似文献

The association between oxidative stress and exposure to environmental chemicals was assessed in a group of Japanese preschool children. The concentrations of 8-hydroxy-2′-deoxyguanosine (8-OHdG), 1-hydroxypyrene (1-OHP), inorganic arsenic (iAs) and monomethylarsonic acid (MMA), and cotinine in spot urine samples, collected from 134 children (3-6 yrs) from a kindergarten in Kanagawa, Japan, were measured as biomarkers of oxidative stress or exposure to environmental chemicals. For 76 subjects of the 134, intakes of anti-oxidant nutrients (vitamins A, C, and E, manganese, copper, zinc and selenium (Se)) were estimated from a food consumption survey carried out 2-4 weeks after urine sampling and by urine analysis (Se). The median (min-max) creatinine-corrected concentrations of urinary biomarkers were 4.45 (1.98-12.3), 0.127 (0.04-2.41), 4.78 (1.18-12.7), and 0.62 (< 0.6-19.0) μg/g cre for 8-OHdG, 1-OHP, iAs + MMA, and cotinine, respectively. Multiple regression analysis was carried out using 8-OHdG concentration as a dependent variable and urinary biomarkers of exposure and Se intake, intakes of vitamins and biological attributes of the subjects as independent variables. To explain 8-OHdG concentrations, intake of vitamin A and age were significant variables with negative coefficients, while 1-OHP concentration had a positive coefficient. These results indicated that oxidative stress of children is affected by chemical exposure at environmental levels, by nutrient intake and by physiological factors in a complex manner. On the other hand, unstable statistical results due to sub-grouping of subject, based on the availability of food consumption data, were found: the present results should further be validated by future studies with suitable research design.  相似文献

Certain physiologic disorders, attributed to lead (Pb) exposure are related to the generation of oxidative stress. Organisms rely on a complex antioxidant system, composed of endogenously produced compounds such as glutathione (GSH), superoxide dismutase (SOD) and GSH peroxidase (GPX); and dietary antioxidants such as vitamins A and E. The aim of this work was to study the interactions between both groups of antioxidants and to evaluate their role in fighting Pb-induced oxidative stress in wild ungulates living in a Pb mining area. We studied red deer (n = 54) and wild boar (n = 60) from mining and control sites. Liver Pb, copper (Cu) and selenium (Se) levels were measured in liver and bone. Levels of GSH, oxidized GSH (GSSG), lipid peroxidation (TBARS), α-tocopherol, free retinol and retinyl esters, and the activities of SOD and GPX were measured in liver. Wild boar and red deer from the mining sites had higher bone (geometric means: 7.36 vs. 1.23 μg/g dw for boar; and 0.46 vs. 0.11 μg/g dw for deer) and liver Pb levels (0.81 vs. 0.24 μg/g dw for boar; and 0.35 vs. 0.11 μg/g dw for deer) than the controls. Pb exposure in deer was associated with lower GSH, α-tocopherol and retinyl esters, and higher free retinol and TBARS. A similar effect on vitamin A balance was observed in wild boar, but this was accompanied with an increase in GSH and α-tocopherol. GPX activity and Se levels were higher in wild boar (mean in controls: 670 IU/mg protein and 1.3 μg/g dw, respectively) than in red deer (150 IU/mg protein and 0.3 μg/g). These differences may indicate that red deer is more sensitive to Pb-induced oxidative stress than wild boar. Both endogenous and nutritional antioxidants may be negatively affected by Pb exposure, and their interactions are essential to fight against Pb-mediated oxidative damage.  相似文献

The concentrations of total mercury (Hg), methylmercury (MeHg) and total selenium (Se) were determined in muscle, liver and brain tissues of young-of-the-year walleye (Stizosedion vitreum) specimens collected from 8 boreal lakes that are located within 107 km around the Sudbury smelters in Ontario, Canada. Dry weight basis concentrations of Hg were highest in muscle and lowest in brain (p < 0.05), those of MeHg were higher in muscle than in liver and brain but there was no significant difference between liver and brain (p < 0.05). The highest Se concentrations were found in liver and the lowest in brain (p < 0.05). Considering the biomass of the studied tissues, muscle was the part of the body where most of Hg, MeHg and Se were accumulated. In fish muscle, the percentage of MeHg over Hg was the highest and this percentage was the lowest in liver. The concentrations of Hg, MeHg and Se in the studied tissues were closely related to the concentrations of total dissolved Se in lake waters which vary with the distance of the lakes from the smelters. Thresholds of Se concentrations in tissues were revealed (6.2, 12.0 and 3.5 mg kg− 1 dry wt., for muscle, liver and brain, respectively), above which a significant reduction of MeHg concentrations was observed in all studied tissues compared to lower Se levels in the same tissues. Based on the collected information and data analysis, possible mechanisms for the biological processes behind the observed inverse relationships between Se and Hg in fish tissues are discussed.  相似文献

With the purpose of knowing seasonal variations of Cd, Cr, Hg and Pb in a river basin with past and present mining activities, elemental concentrations were measured in six fish species and four crustacean species in Baluarte River, from some of the mining sites to the mouth of the river in the Pacific Ocean between May 2005 and March 2006. In fish, highest levels of Cd (0.06 μg g− 1 dry weight) and Cr (0.01 μg g− 1) were detected during the dry season in Gobiesox fluviatilis and Agonostomus monticola, respectively; the highest levels of Hg (0.56 μg g− 1) were detected during the dry season in Guavina guavina and Mugil curema. In relation to Pb, the highest level (1.65 μg g− 1) was detected in A. monticola during the dry season. In crustaceans, highest levels of Cd (0.05 μg g− 1) occurred in Macrobrachium occidentale during both seasons; highest concentration of Cr (0.09 μg g− 1) was also detected in M. occidentale during the dry season. With respect to Hg, highest level (0.20 μg g− 1) was detected during the rainy season in Macrobrachium americanum; for Pb, the highest concentration (2.4 μg g− 1) corresponded to Macrobrachium digueti collected in the dry season. Considering average concentrations of trace metals in surficial sediments from all sites, Cd (p < 0.025), Cr (p < 0.10) and Hg (p < 0.15) were significantly higher during the rainy season. Biota sediment accumulation factors above unity were detected mostly in the case of Hg in fish during both seasons. On the basis of the metal levels in fish and crustacean and the provisional tolerable weekly intake of studied elements, people can eat up to 13.99, 0.79 and 2.34 kg of fish in relation to Cd, Hg and Pb, respectively; regarding crustaceans, maximum amounts were 11.33, 2.49 and 2.68 kg of prawns relative to levels of Cd, Hg and Pb, respectively.  相似文献

Evidence suggests that dietary-intakes of the essential element selenium have fallen in Scotland in recent years, due to changing sources of bread-making wheat. The Scottish environment is thought to be Se-poor due to the geology and climate. This initial study assessed whether geological parent-materials could be used to predict relatively high and low soil-Se areas in Scotland and whether differences in soil-Se were reflected in foodstuff-Se produced on them. Samples (n = 8 per farm) of wheat, calabrese (broccoli), potato, beef-steak, milk, cattle pasture (grass) and soil were collected from pairs of farms (one in each high/low predicted Se area (PSA)). Potatoes and soils were collected from a further 34 farms in high/low PSAs to assess a greater geographical zone. Total soil-Se ranged from 0.115 to 0.877 mg kg-1 but most samples (90%) could be classed as Se-deficient (< 0.6 mg kg−1), irrespective of PSA. Total soil-Se was significantly higher (p < 0.05) in the high than in the low PSAs as expected; however, the difference between the two was small (mean 0.48 and 0.37 mg kg−1, respectively). Water-soluble soil-Se (6.69 to 26.78 μg kg−1) concentrations were not significantly different between the two PSAs (p = 0.71). Soil loss-on-ignition (indicating organic matter content) correlated significantly with total and water-soluble soil-Se (p < 0.001) and exerted a greater control than parent-material on soil-Se. Significant differences between the PSAs for beef-Se (p < 0.001), wheat-Se (p < 0.001), calabrese-Se (p < 0.01) and beef-farm grass-Se (p < 0.05) indicated partial success of the parent-material soil-Se prediction. However, only wheat-Se (p < 0.001) and potato-Se (p < 0.001) correlated significantly with total soil-Se. The results suggest that soil-Se concentrations in the main agricultural areas of Scotland are generally low. Given the low Se concentrations also reported in the food commodities; further investigations may be warranted to fully characterise the Se-status of Scottish produce and dietary-Se intakes in Scotland.  相似文献

The objective of this study was to determine benzene and 1,3-butadiene exposure through ambient air and personal air monitoring, as well as through biomarkers of exposure, and to evaluate the potential health risk of exposure through the use of biomarkers of early biological effects in central Bangkok traffic policemen. Ambient air concentrations of benzene and 1,3-butadiene at the roadsides were significantly higher than in police offices used as control sites (p < 0.001). Traffic policemen had a significantly higher exposure to benzene (median 38.62 μg/m3) and 1,3-butadiene (median 3.08 μg/m3) than office policemen (median 6.17 μg/m3 for benzene and 0.37 μg/m3 for 1,3-butadiene) (p < 0.001). Biomarkers of benzene exposure, blood benzene, and urinary metabolite, trans, trans-muconic acid were significantly higher in traffic policemen than office policemen (p < 0.001). No significant difference between traffic and office policemen was found in urinary benzene metabolite, S-phenyl mercapturic acid, or in urinary 1,3-butadiene metabolite, monohydroxy-butenyl mercapturic acid. Biomarkers of early biological effects, 8-hydroxy-2′-deoxyguanosine in leukocytes (8-OHdG), DNA-strand breaks, and DNA-repair capacity, measured as an increase in gamma ray-induced chromosome aberrations were significantly higher in traffic policemen than controls (p < 0.001 for 8-OHdG, p < 0.01 for tail length, p < 0.001 for olive tail moment, p < 0.05 for dicentrics and p < 0.01 for deletions). Multiple regression model including individual exposure, biomarkers of exposure, ages and years of work as independent variables showed that only the levels of individual 1,3-butadiene exposure were significantly associated with 8-OHdG and olive tail moment at p < 0.0001 indicating more influence of 1,3-butadiene on DNA damage. These results indicated that traffic policemen, who are exposed to benzene and 1,3-butadiene at the roadside in central Bangkok, are potentially at a higher risk for development of diseases such as cancer than office policemen.  相似文献

Mercury (Hg) exposure is associated with disease conditions, including cardiovascular problems. Although the mechanisms implicated in these complications have not been precisely defined yet, matrix metalloproteinases (MMPs) may be involved. The gene encoding MMP-2 presents genetic polymorphisms which affect the expression and activity level of this enzyme. A common polymorphism of MMP-2 gene is the C−1306T (rs 243865), which is known to disrupt a Sp1-type promoter site (CCACC box), thus leading to lower promoter activity associated with the T allele. This study aimed at examining how this polymorphism affects the circulating MMP-2 levels and its endogenous inhibitor, the tissue inhibitor of metalloproteinase-2 (TIMP-2) in 210 subjects environmentally exposed to Hg. Total blood and plasma Hg concentrations were determined by inductively coupled plasma-mass spectrometry (ICP-MS). MMP-2 and TIMP-2 concentrations were measured in plasma samples by gelatin zymography and ELISA, respectively. Genotypes for the C−1306T polymorphism were determined by Taqman® Allele Discrimination assay. We found a positive association (p = 0.0057) between plasma Hg concentrations and MMP-2/TIMP-2 (an index of net MMP-2 activity). The C−1306T polymorphism modified MMP-2 concentrations (p = 0.0465) and MMP-2/TIMP-2 ratio (p = 0.0060) in subjects exposed to Hg, with higher MMP-2 levels been found in subjects carrying the C allele. These findings suggest a significant interaction between the C−1306T polymorphism and Hg exposure, possibly increasing the risk of developing diseases in subjects with the C allele.  相似文献

Background

Mercury is a known neurotoxicant; however, the relationship between childhood exposures and neurodevelopmental outcomes is uncertain, and may be modified by nutrition-related susceptibilities. In vitro studies found that mercury inhibited methionine synthase, an enzyme that interacts with vitamin B-12 and folate to regenerate the amino acid methionine from homocysteine, and inhibition of methionine synthase diverted homocysteine to cysteine and glutathione synthesis. The relationships between mercury, homocysteine, B-12, and folate have not been examined in children.

Objective

This study aimed to evaluate associations between Hg and homocysteine in male and female children differentiated by higher and lower methylmalonic acid (MMA, an indicator of vitamin B-12 deficiency) and folate status.

Design

Cross-sectional data on total blood mercury (Hg), plasma homocysteine, MMA, and serum folate were obtained from the 1999-2004 National Health and Nutrition Examination Surveys for children aged 3-5 years (n = 1005). We used multiple linear regression to evaluate relationships between homocysteine and Hg quartiles, stratified by sex, MMA ≥ and folate < sample medians, adjusted for demographic, anthropometric, and environmental factors.

Results

In boys with higher MMA and lower folate (n = 135), but not in other children, we observed inverse associations between homocysteine and Hg. Children with Hg > 3.49 μmol/L showed 1.14 μmol/L lower homocysteine (p < 0.001) relative to the lowest quartile (≤ 0.70 μmol/L) {p-value for trend < 0.001}. Compared to other subsamples, this subsample had significantly higher homocysteine levels.

Conclusion

Hg was inversely correlated with plasma homocysteine in young boys, but not girls, with higher MMA and lower folate. Additional studies are merited to evaluate Hg and amino acid metabolism in susceptible children.  相似文献

Determination of total antioxidant capacity, instead of the measurements of limited number of antioxidants, is very important for the understanding of how antioxidants interact with reactive oxygen species (ROS). Several techniques already exist with this propose, although some of them are extremely time-consuming. A new methodology is proposed, based on the detection of ROS by fluorometry (ex/em: 485/520 nm) employing 2′,7′-dichlorofluorescein diacetate (H2DCF-DA) as substrate. Supernatant of homogenized samples from different organs (gill, muscle, liver, and brain) of the teleost fish Jenynsia multidentata (Anaplebidae) were exposed to peroxyl radicals generated by thermal (35 °C) decomposition of 2,2′-azobis (2 methylpropionamidine) dihydrochloride (ABAP, 4 mM). Different protein concentrations (0.5, 1, 2 and 8 mg/ml) were assayed to get the best signal and curve fitting of fluorescence data over time (30 min). Total antioxidant capacity against peroxyl radicals was estimated as the difference in ROS area with and without ABAP, relative to the fluorescence registered without ABAP. For application of this methodology, J. multidentata specimens were exposed for 24 h to microcystins, cyanotoxins known to induce oxidative stress. Almost all organs showed a lower antioxidant capacity (p < 0.05) in samples with 8 mg proteins/ml, when compared to protein content of 1-2 mg/ml. In liver samples, higher (p < 0.05) free iron content was determined in samples with 8 mg proteins/ml. Sensitivity test employing GSH spiked in homogenized samples showed the protocol efficiency in detecting total antioxidant capacity. In the test with microcystins a dose-dependent decrease (p < 0.05) of antioxidant competence in gills and brain and an inverse result with liver samples were observed. The use of antioxidant defenses was efficient in avoiding oxidative damage, as the content of oxidized proteins was not altered. Data obtained show the potential of this new methodology to be used in ecotoxicological studies.  相似文献

In this study we aimed to estimate the level of mercury in women's hair and its association with fish consumption and amalgam fillings in an Iranian southern port town (named Mahshahr). A total number of 195 hair samples were collected randomly from women aged 25-45 years free from occupational exposures to mercury. Additional information about these women's age, weight, height, number of pregnancies, level of education, number of amalgam fillings and fish consumption were also gathered. The determination of Hg level in hair samples was carried out by the LECO, AMA 254, Advanced Mercury Analyzer, according to ASTM standard No. D-6722. The average mercury level in these women's hair was 2.95 ± 2.33 µg/g (ranged from 0.07 to 24.46 µg/g). In the next stage, the study participants were divided into four groups with respect to their fish consumption and number of amalgam fillings; A) < 3 amalgam fillings, < 3 times fish consumption per month, B) < 3 amalgam fillings, > 7 times fish consumption per month, C) > 6 amalgam fillings, < 3 times fish consumption per month, and D) > 6 amalgam fillings, > 7 times fish consumption per month. Statistical analysis showed that the Hg level was significantly different in these four groups (P < 0.001). Mercury levels in this sample exceeded the USEPA-recommended 1 μg/g in 76.4% of the studied population. Finally, the multiple linear regression results revealed that frequency of fish consumption and amalgam restorations are the most important routes of Hg exposure. However, the main determinant of Hg exposure was the intake of Hg through fish consumption.  相似文献

Traffic related fine particulate emissions, enriched in metal contents, are directly linked to respiratory disorders in human subjects. In view of the growing traffic related emissions in India, the present study was undertaken to estimate the heavy metal exposure among non-occupationally exposed two vehicle riders of Lucknow City and related health effects via application of face masks (FMs) fitted with cellulose nitrate filters and measuring the peak respiratory flow rate (PEFR). Carefully selected 200 volunteers (asymptomatic n = 154 and symptomatic n = 46) were advised to use FMs during their deriving time for 30 days and PEFR test was conducted on each subject at the beginning, i.e. 0 day, and at end of the study period, i.e. 30 days. On completion of the prescribed study period, filters from the used FMs were collected, acid leached and analyzed for Fe, Mn, Cu, Zn, Pb, Ni, Cr and Cd. Asymptomatic and symptomatic subject groups were further divided into two age groups of 15-40 years and 41-68. Pb, Cu and Cd were significantly higher in lower age group (15-40) of symptomatic group and Cr was in asymptomatic group. Negative associations were observed between metals viz. Pb (r = −0.39, p < 0.001), Cd (r = −0.26, p < 0.001), Fe (r = −0.37, p < 0.001), Mn (r = −0.15, p < 0.05) and the lung functioning. 30 days PEFR of all subjects were higher by nearly 10% than 0 day in all 200 samples irrespective of age and symptomatic nature of the subject. The improvement could also be due to metals and other organic species, not analyzed herein. Nevertheless the results indicate that FM usage has a role to play for immediate, if not ultimate, improvement in public health and need further studies.  相似文献

A survey was conducted in the Haicheng River near Liaodong Bay to analyze the characteristics and sources of the in-stream nitrogen pollution throughout the year 2010. The results indicated that the total nitrogen (TN) concentrations in the river water all exceeded the GB3838-2002 standard for Class V guideline of 2.0 mg/l at all sampling sites during the sampling seasons. Ammoniacal nitrogen (NH3-N) dominated TN during the spring season, while nitrate nitrogen (NO3-N) dominated during the summer and autumn seasons. Different forms of nitrogen had significant seasonal variations (p < 0.01 or p < 0.05). Only NH3-N and NO3-N displayed distinct spatial differences at p < 0.05 and p < 0.001, respectively. Most forms of nitrogen were interrelated with physicochemical parameters during different seasons, displaying nitrification and denitrification processes that occurred in the river rather than seasonal biological demand. Based on the nitrogen parameters, the 30 sampling sites were divided into three clusters, by which the pollution sources from chemical nitrogen fertilizers, animal wastes, domestic sewage, and industrial wastewater were identified. Generally, the Haicheng River basin provided about 700 tons of the annual TN flux, contributing to the eutrophication of the Liaodong Bay and Bohai Sea.  相似文献

Mercury (Hg) exposure causes health problems including cardiovascular diseases. Although precise mechanisms have not been precisely defined yet, matrix metalloproteinases (MMPs) may be involved. The gene encoding MMP-9 presents genetic polymorphisms which affect the expression and activity level of this enzyme. Two polymorphisms in the promoter region [C− 1562T and (CA)n] are functionally relevant, and are implicated in several diseases. This study aimed at examining how these polymorphisms affect the circulating MMP-9 levels and its endogenous inhibitor, the tissue inhibitor of metalloproteinase-1 (TIMP-1) in 266 subjects environmentally exposed to Hg. Blood and plasma Hg concentrations were determined by inductively coupled plasma-mass spectrometry (ICP-MS). MMP-9 and TIMP-1 concentrations were measured in plasma samples by gelatin zymography and ELISA, respectively. Genotypes for the C− 1562T and the microsatellite (CA)n polymorphisms were determined. We found a positive association (P < 0.05) between plasma Hg concentrations and MMP-9/TIMP-1 ratio (an index of net MMP-9 activity). When the subjects were divided into tertiles with basis on their plasma Hg concentrations, we found that the (CA)n polymorphism modified MMP-9 concentrations and MMP-9/TIMP-1 ratio in subjects with the lowest Hg concentrations (first tertile), with the highest MMP-9 levels being found in subjects with genotypes including alleles with 21 or more CA repeats (H alleles) (P < 0.05). Conversely, this polymorphism had no effects on subjects with intermediate or high plasma Hg levels (second and third tertiles, respectively). Objective

The heavy metals lead (Pb) and mercury (Hg) are ubiquitous environmental pollutants with high neurotoxic potential. We aimed to compare perinatal Pb and Hg concentrations and to explore the potential association between Pb and Hg exposure and newborn anthropometry.

Study design

Pregnant women were recruited in 2005 at the General Hospital Vienna for participation in this longitudinal study. Pb and Hg concentrations were measured in maternal blood and hair, placenta, cord blood, meconium, and breast milk of 53 mother-child pairs by CV-AAS, GF-AAS, and HPLC-CV-ICPMS. We conducted bivariate analyses and categorical regression analysis (CATREG) to evaluate the determinants of Pb and Hg exposure, and of infant anthropometry.

Results

Median Pb and total Hg contents were low, i.e., 25 μg/L (maternal blood-Pb), 13 μg/L (cord blood-Pb), 0.7 μg/L (maternal blood-Hg), and 1.1 μg/L (cord blood-Hg). Hg levels in maternal and fetal tissues were frequently correlated (r > 0.3, P < 0.05, respectively). Regarding Pb, only maternal blood and cord blood concentrations correlated (P = 0.043). Cord blood levels indicated higher Hg exposure but lower Pb exposure relative to maternal blood contents. Adjusted CATREG models indicated the significant predictors of birth length (placenta-Pb, gestational length, meconium-Pb), birth weight (placenta-Pb, gestational length, maternal blood-Pb), and head circumference (maternal education, maternal height). Besides one significant correlation between maternal hair Hg and birth length, the mercury levels were not associated with newborn anthropometry.

Conclusions

Our data implicate that different modes of action may exist for placentar transfer of Pb and Hg as well as that low Pb exposure levels can result in lower birth weight. The findings related to newborn anthropometry need to be confirmed by the examination of larger study groups. Further research is needed to clarify the mechanisms of Pb and Hg transfer via the placenta, and to explore how prenatal Pb exposure is related to intrauterine growth.  相似文献

The incidence of hepatocellular carcinoma (HCC) is significantly elevated in a Hispanic community in Bexar County, Texas. Chronic exposure to dietary aflatoxins (AFs) is a major risk factor for HCC; increased risk has been linked to polycyclic aromatic hydrocarbon (PAH) co-exposure and hepatitis virus infection. The aims of this study were to assess AF and PAH exposures, investigate dietary factors that may contribute to increased AF exposure, and determine the prevalence of hepatitis virus infection in Bexar Co. Blood and urine samples were collected from 184 volunteers for biomarker analyses and hepatitis screening. Serum AFB1-lysine adduct, urinary AFM1 and 1-hydroxypyrene (1-OHP) levels were measured using high-performance liquid chromatography. The average AFB1-lysine adduct level detected in 20.6% of serums was 3.84 ± 3.11 pg/mg albumin (range 1.01-16.57 pg/mg). AFM1 was detected in 11.7% of urines, averaging 223.85 ± 250.56 pg/mg creatinine (range 1.89-935.49 pg/mg). AFM1 detection was associated with increased consumption of corn tortillas (p = 0.009), nuts (p = 0.033) and rice (p = 0.037). A significant difference was observed between mean 1-OHP values of non-smokers (0.07 ± 0.13) and smokers (0.80 ± 0.68) μmol/mol creatinine (p < 0.01). A high hepatitis C virus positivity rate (7.1%) was observed. Findings suggest that the incidence and level of AF and PAH exposure were less than those observed in a high-risk population; however, participants consuming higher amounts of foods prone to AF contamination may be more vulnerable to exposure and interactions with other environmental/biological factors (i.e., HCV).  相似文献

The risks and benefits of using mercury (Hg) in dental amalgam have long been debated. This study was designed to estimate Hg body burden and its association with dental amalgam fillings in 182 children (ages: 5-15 years) living in Taif City. Hg was measured in urine (UHg), hair (HHg) and toenails (NHg) by the Atomic Absorption Spectrophotometer with Vapor Generator Accessory system. Urinary Hg levels were calculated as both micrograms per gram creatinine (μg/g creatinine) and micrograms per liter (μg/L). We found that children with amalgam fillings (N = 106) had significantly higher UHg-C levels than children without (N = 76), with means of 3.763 μg/g creatinine versus 3.457 μg/g creatinine, respectively (P = 0.019). The results were similar for UHg (P = 0.01). A similar pattern was also seen for HHg, with means of 0.614 μg/g (N = 97) for children with amalgam versus 0.242 μg/g (N = 74) for those without amalgam fillings (P = 0). Although the mean NHg was higher in children without amalgam (0.222 μg/g, N = 61) versus those with (0.163 μg/g, N = 101), the relationship was not significant (P = 0.069). After adjusting for many confounders, the multiple logistic regression model revealed that the levels of UHg-C and HHg were 2.047 and 5.396 times higher, respectively, in children with dental amalgam compared to those without (P < 0.01). In contrast, a significant inverse relationship was seen between NHg levels and dental amalgam fillings (P = 0.003). Despite the controversy surrounding the health impact of dental amalgam, this study showed some evidence that amalgam-associated Hg exposure might be related with symptoms of oral health, such as aphthous ulcer, white patches, and a burning-mouth sensation. Further studies are needed to reproduce these findings. The present study showed that significant numbers of children with or without amalgam had Hg levels exceeding the acceptable reference limits. The detrimental neurobehavioral and/or nephrotoxic effects of such an increased Hg on children should be a cause of concern, and further investigation is warranted. Our results are alarming and indicate an urgent need for biomonitoring and assessment of exposure. Changes in dental practices involving amalgam, especially for children, are highly recommended in order to avoid unnecessary exposure to Hg.  相似文献

Bioaccessibility, the fraction of an element solubilized during gastrointestinal digestion and available for absorption, is a factor that should be considered when evaluating the health risk of contaminants from food. Static and dynamic models that mimic human physiological conditions have been used to evaluate bioaccessibility. This preliminary study compares the bioaccessibility of arsenic (As), cadmium (Cd), lead (Pb) and mercury (Hg) in two food certified reference materials (CRMs) (seaweed: Fucus sp., IAEA-140/TM; Lobster hepatopancreas: TORT-2), using two in vitro gastrointestinal digestion methods: a static method (SM) and a dynamic multicompartment method (TIM-1). There are significant differences (p < 0.05) between the bioaccessible values of As, Cd, Pb and Hg obtained by SM and TIM-1 in the two CRMs. The specific form in which the elements studied are present in the CRM may help to explain the bioaccessibility values obtained.  相似文献
