用质粒pEF-BOS的增强子、肽链延长因子基因的启动子及其第一内含子替换质粒pED-GCSF的增强子,腺病毒主要晚期启动子及杂合内含子。构建成了更高表达质粒pEF-GCSF,转染CHO-dhfr-细胞,加入并逐渐升高氨甲喋呤的浓度。结果显示:一定范围内,随着MTX浓度增高,重组人粒细胞集落刺激因子表达量随之提高;筛选并建立了一株稳定高表达rhG-CSF cDNA的细胞株,其表达量为5.0 ̄5.6μ  相似文献

采用DSC、WAXD、元素分析、FT-IR、ESCA、GPC、交联度和密度测定,研究了紫外线辐照了HDPE的结晶结构及化学结构变化。结果表明,紫外线辐照后,HDPE的熔融温度降低、熔融热焓升高、结晶度增大、晶胞结构未发生变化;HDPE分子甸主了COOH、C=O、C-O、OOH、OH等含氧官能团,分子量降低,分子量分布加宽,基本无交联发生。  相似文献

采用DSC技术,对增强增韧尼龙66进行了结晶行为的研究。实验结果表明,在合金化改性PA66结晶过程中,所填充的玻纤增强剂成核剂作用,而共混低密度聚乙烯、聚丙烯及其马来酸酐接枝物等增韧剂,使成核作用下降,总的成核作用顺序是:PA/GF〉PA/GF/LDPE-g-MAH〉PA/GF/PP〉PA/GF/PPg-MAH。  相似文献

利用等离子体表面处理和化学接枝的方法对聚四氟乙烯(PTFE)薄膜表面进行了化学改性。利用XPS研究了改性后的PTFE薄膜的表面结构和价键状态,并通过接触角的测定研究了表面改性对薄膜亲水性的影响。研究结果发现PTFE薄膜经等离子体处理后,薄膜表面的C-F键发生了断裂,形成了C-C键、C-H键及C-O键;等离子体处理后的薄膜再经丙烯酸化学处理,强亲水性的丙烯酸基被接枝到PTFE表面,使得PTFE薄膜获  相似文献

Kidney gene therapy using the hepatocyte growth factor (HGF) gene may offer new strategies for the treatment of chronic renal disease such as kidney fibrosis, because HGF has the potential to promote tubular repair and to inhibit tissue fibrosis. As a non-viral vector for gene delivery, polyethylenimine (PEI) exhibits high gene expression due to its buffering capacity with cytotoxicity, although its cytotoxicity depends on its molecular weight. In this study, to minimize the cytotoxicity of PEI with a high transfection efficiency, biodegradable poly(ester amine) (PEA) based on glycerol dimethacrylate (GDM) and low molecular weight PEI (LMW PEI) was synthesized and kidney targeting peptide was conjugated to the PEA (PEP-PEA) to give it kidney cell specificity. The PEP-PEA showed good physicochemical properties as a gene delivery carrier, such as DNA condensation ability, protection of the DNA in the complexes from enzyme degradation, and formation of nanosized complexes with spherical shapes. Higher transfection efficiency in 293T cells was achieved with the PEP-PEA than with the PEA and the PEI 25 kDa with lower cytotoxicity. Also, the HGF gene that was complexed with the PEP-PEA was specifically delivered to the obstructed kidney in the unilateral ureteral obstruction (UUO) model rats. The delivered HGF gene exhibited potency in recovering renal functions, which indicates the potential of the PEP-PEA as a safe and efficient carrier for the treatment of kidney fibrosis.  相似文献

制备具有缓释功能的壳聚糖-明胶-碱性成纤维细胞生长因子(bFGF)-肝细胞生长因子(HGF)三维大孔支架,探讨两种生长因子在体外对骨髓间充质干细胞(BMSCs)增殖的协同作用。方法:采用冷冻干燥法,将不同比例的壳聚糖、明胶、bFGF和HGF依次混合,使其成为具有一定孔径的三维缓释支架。取SD大鼠乳鼠股骨和胫骨骨髓,分离、培养BMSCs。取生长状态良好的第三代BMSCs,将其接种于96孔板后,加入支架混合培养,5d后进行MTT细胞增殖测定。结果:在与含1μg/mL的bFGF支架混合培养,以及与同时含1μg/mL的bFGF、1μg/mL的HGF支架混合培养后,BMSCs增殖明显(P<0.05),但这二组间无统计学差异(P>0.05)。分别与含0.1μg/mL的bFGF支架、0.01μg/mL的bFGF支架、1μg/mL的HGF支架混合培养后,细胞增殖无统计学意义(P>0.05)。结论:壳聚糖-明胶可作为生长因子缓释支架材料;bFGF具有促进BMSCs增殖的作用,促进作用的大小与加入bFGF的量有关;HGF对BMSCs不具有增殖作用;在实验浓度范围内bFGF和HGF体外促进BMSCs增殖上不具有协同性。  相似文献

A remarkable feature of the disposition of polypeptide hormones is the contribution of specific binding sites (receptors) on the cell-surface to their clearance in the body. Receptor-mediated endocytosis (RME) is now well recognized as a polypeptide clearance system. In addition, drug targeting utilizing RME is expected to have promise as a drug delivery system (DDS) to carry some drug specifically into the target cell expressing receptors on its plasma membrane. Therefore, it is important to analyze the RME process kinetically, both to clarify the pharmacokinetics of polypeptide itself and to estimate the efficiency of drug targeting via polypeptide receptors. We have been studying the hepatic handling of epidermal growth factor (EGF) and hepatocyte growth factor (HGF) using perfused rat liver, isolated rat hepatocytes, and primary cultured rat hepatocytes. Kinetic analysis of hepatic clearance of EGF enabled us to determine the kinetic parameters for the construction of a kinetic model for RME. Based on such a kinetic model, we can discuss the contribution of each process in RME on the efficiency of drug targeting into the cell. Not only RME, but also a non-specific uptake mechanism was found to contribute to hepatic HGF disposition. This finding was important piece of information in developing a DDS for HGF, aimed at decreasing such non-specific uptake, with the result of prolonged plasma residence time.  相似文献

A poly(dl-lactic-co-glycolic acid) (PLGA) sandwiched adipose derived stem cell (ADSC)/fibrin tubular construct, fabricated using a step-by-step mold/extraction method, was characterized in this work. The ADSCs were also induced into smooth-muscle-like cells using growth factors such as hepatocyte growth factor (HGF), platelet-derived growth factor BB (PDGF-BB), transforming growth factor β1 (TGFβ1), and basic fibroblast growth factor (b-FGF). Compared with the non-induced cells, the proliferation ability of induced cells was much smaller. The PLGA sandwiched cell/hydrogel construct was shown to be useful for controlling the cellular microenvironment and cellular behaviors such as growth, migration, proliferation and differentiation. This strategy seems promising in tissue engineering and organ manufacturing.  相似文献

Two types of scaffolds composed of microstructured chambers made in a photosensitive biodegradable polymer were microfabricated by softlithography techniques. The fabrication process is fast and flexible. Single- and multi-stepwise microstructures could be fabricated on one polymer layer forming an array of microstructured chambers. The static cell culture of Hep G2 cells and fetal human hepatocyte (FHH) cells were successfully performed on those microstructured layers and show the biocompatibility of this polymer. Morphological observations have shown various tissue-like behaviours during the static cell culture of Hep G2 and FHH cells such as cells aggregations and 3D arrangements. As a first physiological response of the cells to the stepwise structures, the albumin production rate has shown a higher cell activity in the microstructured chambers compared to conventional flat dish cultures.  相似文献

The purpose of this study was to evaluate the angiogenic effect of topical application of three types of ONO-1301-loaded poly (lactide-co-glycolide) microspheres (ONO-1301 PLGA MS). ONO-1301 PLGA MS were prepared with PLGA 5010, 5020 and 5050 (with molecular weights of 10?K, 20?K and 50?K, respectively), using the solvent evaporation method. The lactide:glycolide ratio was fixed at 50:50; only the molecular weight was varied. The microspheres had an average diameter of almost 25?µm, and a loading efficiency of at least 70%. The sustained-release effect and its dependence on the molecular weight of the polymer used was confirmed in an in vitro drug-release test and by measuring subcutaneous plasma levels after administration of the three types of ONO-1301 PLGA MS to rats for 28 days. In the murine sponge model, the three types of ONO-1301 PLGA MS were administered to mice in a subcutaneously placed sponge and hemoglobin and hepatocyte growth factor (HGF) levels in the sponge were measured at predefined intervals up to 28 days. The hemoglobin and HGF levels obtained were significantly higher than those obtained after daily administration of ONO-1301 powder. Additional in vivo fluorescence imaging showed that PLGA MS remained in the sponge for 28 days. In conclusion, the three types of ONO-1301 PLGA MS prepared with PLGA three different molecular weight suppress the burst release, stimulate angiogenesis on topical application in a murine sponge model. This formulation may therefore be capable of improving the clinical picture in some types of vascular disease.  相似文献

在太阳能热发电站中,选择Al-12.07%Si作为储能材料,20G钢作为换热管材料,并将20G钢沉浸在Al-12.07%Si中进行长达1080h的腐蚀研究。通过SEM、EDAX、XRD方法分别对腐蚀后试样腐蚀层的形貌、成分和腐蚀试样的物相进行分析。研究结果发现,20G钢的腐蚀层形状不平整规则,试样表面均出现了Fe2Al5和FeAl3相。随着腐蚀时间延长,腐蚀试样的失重和腐蚀层厚度逐渐稳定,试样的腐蚀速率下降。  相似文献

Endpoints related to cell cycle perturbations were investigated in human primary fibroblasts irradiated with 1-4 Gy of either protons (7.7 and 28.5 keV.micron-1) or X rays. Regardless of the quality of radiation, 8 h after irradiation cells accumulated in the G2 phase, but such accumulation was maintained at 24 h from treatment only in 28.5 keV.micron-1 protons. A marked G1/S-transition block was observed in proton as well as in X ray-treated cultures up to 24 h, with doses as low as 1 Gy. On the other hand, the number of cells positive to p21 antibody reaction was higher after proton than after X ray treatment, indicating that the 'complexity' of lesions may play a critical role in the induction of p21.  相似文献

The objective of this study is to prepare a novel gene carrier from pullulan, a polysaccharide with an inherent affinity for the liver and evaluate the feasibility in gene transfection in the mice liver. Pullulan with a weight-average molecular weight of 22,800 was cationized by the chemical introduction of spermine (spermine-pullulan). The cationized pullulan derivative was complexed with a plasmid DNA and intravenously injected for in vivo gene transfection. The level of gene expression by the spermine-pullulan in the liver depended on the extent of spermine introduced and the highest level was observed for the spermine-pullulan with an introduction extent of 5.60. When a plasmid DNA coding NK4 of a hepatocyte growth factor (HGF)/scatter factor antagonist complexed with the spermine-pullulan was intravenously injected to mice 1 day before the inoculation of RLmale1 tumor cells, the tumor-bearing mice survived for a longer time period, while the GPT level and the number of tumor cells grown in the liver were low compared with those of free plasmid DNA injection. These findings indicate that the liver targeting of NK4 plasmid DNA by complexation with the spermine-pullulan specifically enhanced the liver expression level, resulting in augmented suppression effect on tumor growth therein.  相似文献

Self-healing is receiving increasing interest worldwide as a technology to autonomously address the effects of damage in composite materials. This paper describes the results of four point bend flexural testing (ASTM-D6272-02) of T300/914 carbon fibre reinforced epoxy with resin filled embedded hollow glass fibres (HGF) which provided a self-healing functionality. The study investigated the effect of the embedded HGF on the host CFRP mechanical properties and also the healing efficiency of the laminates after they were subjected to quasi-static impact. Specimens were tested in the undamaged, damaged and healed conditions using a commercial two-part epoxy healing agent (Cytec Cycom 823). Microscopic characterisation of the embedded HGF was also undertaken to characterise the effect on the host laminate fibre architecture.  相似文献
