醇溶玉米蛋白膜的差热分析结果表明其玻璃化转变温度为171℃,热分解温度为262℃根据热重分析,温度为81℃时,失重率为2.3%,主要是乙醇及水分子的蒸发,262 ̄355℃为产品急剧分解阶段,与差热分析结果相吻合,电镜扫描结果表明高于玻璃化温度的薄膜热处理,能部分修饰表面缺陷,其次还讨论了薄膜形成条件对蛋白膜形态结构的影响。  相似文献

Sixty-seven Spanish honeys from different floral origins, nectars and honeydews, were examined for potential antimicrobial and antioxidant activities. Antioxidant capacities were determined in terms of their antiradical capacity using the stable free radical DPPH?. The bacterial species Micrococcus luteus and Staphylococcus aureus were used as the resistant microorganism in the antibacterial assays. The results showed that honeydew honeys presented higher antioxidant capacities than nectar honeys. On the other hand, honey capacity to inhibit Micrococcus luteus and Staphylococcus aureus was firstly evaluated by an agar diffusion method. Secondly, the active samples were tested with a spectrophotometic method to quantify their “non-peroxide” antimicrobial capacity to inhibit Staphylococcus aureus . We did not observed any relationship between the physicochemical parameters of the samples, and their ability to inhibit these microorganisms. The results obtained by the spectrophotometric method show that this method is easy and useful in the evaluation of the antibacterial capacity of honeys.  相似文献

Stochastic models can be useful to predict the risk of foodborne illness. The presence of Bacillus cereus in liquid egg can pose a serious hazard to the food industry, since a mild heat treatment cannot guarantee its complete inactivation. However, most of the information available in the scientific literature is deterministic, including growth of B. cereus. In this paper, a stochastic approach to evaluate growth of B. cereus cells influenced by different stresses (presence of nisin and lysozyme separately or in combination) was performed, using an individual-based approach of growth through OD measurements. Lag phase duration was derived from the growth curves obtained. From results obtained, histograms of the lag phase were generated and distributions were fitted. Normal and Weibull distributions were ranked as the bestfit distributions in experiments performed at 25 °C. At 16 °C, lag values (obtained in presence of combinations of both antimicrobials) were also fitted by a Gamma distribution. Predictions were compared with growth curves obtained in liquid egg exposed to mild heat, nisin and/or lysozyme to assess their validity.  相似文献

This paper is concerned with transverse cut resistant properties of UHMWPE yarns by the methods of experimental and theoretical analysis. Rheological theory of fabric was applied to evaluate relation of stress and strain of UHMWPE yarn, when the UHMWPE yarn is subjected to external cut force. The dynamic response of yarn and the contact volume of yarn and blade in the cutting have been identified to interpret cut failure mechanism of yarn. The experimental results show that yarn structure, blade edge, and test parameters have great influence on cut resistant property of UHMWPE yarn, and which are consistent with the theoretical analysis. It provides a theoretical basis for quantifying the cutting resistance of fabrics.  相似文献

Seed protein stability and seed deterioration during storage were studied in seeds of two different quinoa (Chenopodium quinoa Willd.) cultivars, i.e., cv. Ollagüe and cv. Baer II. Germination and viability tests proved that cv. Baer II was more longevous than cv. Ollagüe. Protein insolubilisation was detected during storage and correlated with longevity. However, protein solubility was restored by priming in both cultivars, disregarding their germination capability. Extremely high contents of advanced glycation end-products (AGEs), assessed by carboxymethyllysine (CML) quantification, were obtained from stored seeds as viability was reduced; primed seeds showed slightly lower AGE content, indicating a strong association between quinoa seed ageing and Maillard products accumulation. High intensity fluorescence values were observed in seeds stored in the gene bank bearing 100% germination values, which indicate that the detection of fluorescence is not by itself an appropriate indicator of protein damage by Maillard-type product accumulation in quinoa seeds. This work establishes for the first time a direct association between seed viability and AGE accumulation, due to Maillard reaction, in quinoa seeds.  相似文献

The effects of electron beam irradiation and heat treatments on the variability of inactivation of Bacillus cereus spores (CECT 131/ATCC 10876) and of the lag phase of single surviving cells have been studied. In general, dispersion in the number of survivors increased as the stress became more intense. A polynomial relationship was derived between the coefficient of variation of the survivor number and the inactivation achieved. Heat treatments caused wider distributions than irradiation for the same substrate and for a similar degree of microbial inactivation.Increasing the intensity of the inactivation treatment lengthened the lag phase of survivors and increased its variability. Comparison of lag phases of heated and irradiated spores did not show any clear relationship. Heating did not affect the specific growth rate of surviving cells, whereas irradiation lowered the maximum specific growth rate in proportion to the dose applied. These results suggest that the shelf life of irradiated foods is longer than that of heated foods.  相似文献

Cells of six strains of Cronobacter were subjected to dry stress and stored for 2.5 months at ambient temperature. The individual cell lag time distributions of recovered cells were characterized at 25 °C and 37 °C in non-selective broth. The individual cell lag times were deduced from the times taken by cultures from individual cells to reach an optical density threshold. In parallel, growth curves for each strain at high contamination levels were determined in the same growth conditions. In general, the extreme value type II distribution with a shape parameter fixed to 5 (EVIIb) was the most effective at describing the 12 observed distributions of individual cell lag times. Recently, a model for characterizing individual cell lag time distribution from population growth parameters was developed for other food-borne pathogenic bacteria such as Listeria monocytogenes. We confirmed this model’s applicability to Cronobacter by comparing the mean and the standard deviation of individual cell lag times to populational lag times observed with high initial concentration experiments. We also validated the model in realistic conditions by studying growth in powdered infant formula decimally diluted in Buffered Peptone Water, which represents the first enrichment step of the standard detection method for Cronobacter. Individual lag times and the pooling of samples significantly affect detection performances.  相似文献

Abstract: Plasticization and glass transition of amorphous components in food powders often result in stickiness and caking. The glass transition temperature (Tg) of milk powders was measured by differential scanning calorimetry (DSC) and a viscometer method was used to determine sticky-point temperatures. Water sorption isotherms were established for varying solids compositions. Lactose contents were analyzed by high-performance anion exchange chromatography with pulsed amperometric detection (HPAE-PAD) and proteins were identified using SDS-PAGE gel electrophoresis. Solids composition and water affected both the Tg and stickiness behavior. Stickiness was governed by carbohydrates and water plasticization. At low protein contents, precrystallization of lactose decreased the sticky point temperature, but increasing protein content in all milk powders decreased stickiness at all water activities. The results showed that glass transition can be used to describe time-dependent stickiness and crystallization phenomena, and it can be used as a parameter to control and reduce stickiness of dairy solids with various compositions. Practical Application: Glass transition of component sugars in milk powders with various water contents was responsible for a solid-liquid transformation which resulted in their viscous flow at particle surfaces and stickiness of the powders. Stickiness leads to wall deposition in dehydration and caking of powders in storage when the amorphous carbohydrate-rich components gain liquid characteristics. High protein contents in milk powders decreased stickiness, but precrystallization of lactose prior to spray drying increased stickiness at low protein content. Milk powders in storage gained higher water contents with increasing protein contents, but stickiness was reduced and lactose crystallization was delayed which improved storage stability.  相似文献

This study was carried out to determine the temporal effect of the antioxidants butylated hydroxyanisol (BHA) and propyl paraben (PP) at doses of 500 and 1000 μg/g on the growth of Fusarium verticillioides and F. proliferatum inoculated on natural maize grain in the presence of the competing mycoflora and fumonisin production at 0.98 and 0.95 water activity (aw) over a 28-day storage period. The reduction in the log colony forming units (CFU) of Penicillium, Aspergillus and Fusarium populations was 10-100 fold depending on dose of BHA or PP, aw and time. However, the populations of all three groups were higher at 0.98 aw than 0.95 aw. BHA at 500 μg/g and 0.95 aw reduced the fumonisin content by 82% after 7-14 days incubation, but at the end of the experimental period the reduction was only 32%. A higher reduction in the level of fumonisin produced (77%) was achieved with BHA at 1000 μg/g after 28 days. PP at 500 and 1000 μg/g decreased fumonisin production throughout the incubation period in the drier treatment, but at 0.98 aw control of toxin production was only achieved after 7-14 days. The reduction in the fumonisin levels could be due to the combined effect of antioxidants, and the competing mycoflora, mainly Aspergillus and Penicillium species.  相似文献

The aim of this work was to model the growth of Aspergillus parasiticus and Aspergillus ochraceus, both mycotoxin producers, near to the growth/no growth boundaries and validate those models in sterile maize grain, peanuts and coffee beans. Malt extract agar was adjusted to six different water activities: 0.93, 0.91, 0.89, 0.87, 0.85 and 0.80. Plates were incubated at 10, 15, 20, 25, 30, 37 and 42 °C. For each of the 42 conditions, 10 Petri dishes were inoculated. Both kinetic and probability models were applied to colony growth data. The results of the present study indicate that the developed probability modelling approach could be satisfactorily employed to quantify the combined effect of temperature and water activity on the growth responses of A. ochraceus and A. parasiticus. However, validation of kinetic results led to poor goodness of prediction. In this study, the validation samples were placed near to the expected boundaries of the models in order to test them under the worst situation. Probability of growth prediction under extreme growth conditions was somewhat compromised, but it can be considered acceptable.  相似文献

Seed characteristics and nutrient composition of three morphotypes of big-grained Mucuna urens (L.) Medikus were studied. Results showed that 100-seed weight ranged from 3200.2 to 4700.9 g, cotyledon weight per seed (23.2–26.6 g) and testa weight per seed (9.0–21.2 g). The testa constituted 43.7%, 28.1% and 44.7% of the average seed weight for morphotypes 1, 2 and 3, respectively. The cotyledon also constituted 56.3%, 71.9% and 55.3% of the average seed weight for morphotypes 1, 2 and 3, respectively. Nutrient composition analyses showed that the three morphotypes of M. urens are good sources of crude protein (19.97–20.57 g 100 g−1), carbohydrate (72.39–75.49 g 100 g−1) and fat (1.84–5.05 g 100 g−1). Other nutritional components, including ascorbic acid, calcium and phosphorus are present in the three morphotypes in moderate amounts. The iron content of the M. urens is low. The three morphotypes contain appreciable amounts of essential amino acid. The oxalate content is low. The variations observed in the seed characteristics and nutrient compositions are suspected to be due to genotype. Genetic improvement of this plant is recommended to remove the itching hair trait so as to encourage its cultivation.  相似文献

The individual-based approach of the lag phase is gaining interest, especially for pathogens that initially contaminate food products in low amounts. In this paper, the effect of temperature (30, 10, 7, 4 and 2 °C) and pH (7.4, 6.1, 5.5, 5.0, 4.7 and 4.4) on the individual cell lag phase of Listeria monocytogenes was examined in a factorial design, using OD measurements. Individual lag phases of about 100 individual cells per condition were examined and calculated using a linear extrapolation method. Generation times were calculated out of the slope.The obtained data were analyzed at three different levels: in a first approach, the mean values were calculated for each set of environmental conditions and compared to predictions made by the USDA's Pathogen Modeling Program (PMP) for analogous growth conditions. The PMP predictions of the generation times were in the same order of magnitude as the obtained data, although a persistent underestimation could be observed. The observed individual cell lag data differed from lag phase predictions by PMP. Possible reasons for this discrepancy are discussed.Secondly, histograms of individual lag phase measurements were constructed for the different temperature–pH combinations. In this way, the influence of both factors on the variability of individual lag phases could be estimated. At low stress levels, most individual cells showed a short lag phase resulting in a compression of the histograms at the zero-lag level, while, at high stress levels, the histograms shifted to longer lag phases with a significant increase in variability.Thirdly, 37 different distribution types were fitted to the datasets to reveal the distributions that fitted best the obtained data. The gamma distribution was preferred at moderate stress levels, while the Weibull distribution was chosen for harsher growth conditions.  相似文献

The seed cake is a big by-product after crushing cooking oil from the seeds of Camellia oleifera Abel. Chemical investigation on the seed cake of C. oleifera led to the isolation of two new kaempferol acetylated glycosides (1 and 2). In addition, five kaempferol glycosides (37) and their aglycone, kaempferol (8), were also obtained, in addition to gallic acid (9). Their structures were determined by the detailed spectroscopic analysis and acidic hydrolysis. The new compounds were characterised as kaempferol-3-O-[4′′′′-O-acetyl-α-l-rhamnopyranosyl-(1→6)]-[β-d-glucopyranosyl-(1→2)]-β-d-glucopyranoside (1) and kaempferol-3-O-[4′′′′-O-acetyl-α-l-rhamnopyranosyl-(1→6)]-[β-d-xylopyranosyl-(1→2)]-β-d-gluco-pyranoside (2), respectively. The DPPH radical scavenging activity of all the isolated compounds was described.  相似文献

The bruchid Callosobruchus maculatus causes major losses during the storage of the seeds of Vigna unguiculata in West Africa. An endemic parasitoid, the pteromalid Dinarmus basalis is present in the stores and can reduce the increase in bruchid populations. African farmers often place in the stores the leaves of a shrub, Boscia senegalensis Lam.(Capparaceae), which release methylisothiocyanate (MITC). This compound is toxic to adult bruchids and could reduce seed losses but its influence on the bruchid natural enemy D. basalis was hitherto unknown. The susceptibility of C. maculatus and D. basalis to MITC was assessed under laboratory conditions. The susceptibility of adults of both species towards MITC were similar. After a 24 h exposure, the LC50 was respectively 0.48 and 0.54 mg/l for C. maculatus and D. basalis. The ovicidal activity of this compound was high; the LC50 of C. maculatus eggs was 0.04 mg/l after a 24 h exposure to MITC. High MITC concentrations only slightly affected the survival of C. maculatus during its post-embryonic development in the seeds of V. unguiculata. Gas chromatographic analysis demonstrated that 25-35% of the MITC present in the atmosphere of the experimental jars was absorbed by the seeds but concentrations inside the cotyledons were too low to influence the survival of the C. maculatus larvae. The D. basalis larvae developing at the expense of their host inside the larval galleries, were more affected by the treatment. The introduction of B. senegalensis releasing MITC in the storage systems could reduce the density of the parasitoid population and so increase the seed losses by permitting the development of the bruchid population.  相似文献

This study was designed to examine the chemical composition of essential oil and in vitro antioxidant and antibacterial activity of the essential oil and methanol extracts of Eucalyptus oleosa F. Muell. The chemical composition of the hydrodistilled essential oil of the leaves of E. oleosa was analyzed by gas chromatography and gas chromatography/mass spectrometry. The main constituents of the oil were found to be 1,8-cineole (45.1%), α-pinene (14.52%), and α-terpineol (4.35%). The essential oil showed strong antibacterial activity against the test microorganisms studied, while polar subfraction of methanol extract had moderate antibacterial activity and the nonpolar subfraction of methanol extract did not show any antibacterial activity. In contrast, the extract showed much better activity than the essential oil in antioxidant activity assays employed, e.g., in DPPH systems, the highest radical-scavenging activity was shown by the polar subfraction (15.1 ± 0.7 μg/ml). In the second case, the inhibition capacity (%) of the nonpolar subfraction (98.2% ± 1.5) was found to be the stronger one. In addition, the amounts of total phenol components in the polar subfraction (186.3 ± 2.1 μg/mg) and the nonpolar subfraction (79.6 ± 1.4) were determined.  相似文献

Enterobacter sakazakii infections in preterm neonates and infants have been epidemiologically associated with consumption of reconstituted powdered infant formula. The bacterium has been isolated from grain, infant cereals, and cereal factory environments. A study was done to determine the survival characteristics of E. sakazakii initially at populations of 0.31 and 5.03 logCFU/g of infant rice cereal (a(w) 0.30, 0.45-0.46, and 0.68-0.69). Cereal was stored at 4, 21, and 30 degrees C and populations were monitored for up to 12 months. Survival of the pathogen in infant rice, barley, oatmeal, and mixed grain cereals (a(w) 0.63-0.66, 0.76, or 0.82-0.83) initially containing a population of 4.93-5.64 logCFU/g and held at 4, 21, and 30 degrees C up to 24 weeks was determined. Populations decreased significantly (p < or = 0.05) in all cereals stored at 21 and 30 degrees C regardless of a(w). Increases in a(w) or storage temperature accelerated the rate of death of E. sakazakii in dry infant cereals. However, at an initial population of 0.31 logCFU/g, E. sakazakii survived in rice cereal (a(w) 0.30-0.69) for up to 12 months at all storage temperatures. Survival of E. sakazakii was not affected by the composition of dry infant rice, barley, mixed grain, and oatmeal cereals (initial a(w) 0.63-0.83) stored for up to 24 weeks at 4, 21, or 30 degrees C. This study demonstrated that E. sakazakii can survive for up to 12 months in infant cereals having a wide range of a(w) when storage is at temperatures simulating those to which they may be exposed during distribution, at retail, and in the home.  相似文献

The purpose of the present study was to apply a modelling approach to define the growth rate and growth/no growth interface of Byssochlamys fulva and Byssochlamys nivea on a synthetic medium as a function of temperature and water activity. Both fungal species were grown on malt extract agar at different temperatures (10, 15, 20, 25, 30, 35, 40 and 45 °C) and aw levels (0.88, 0.90, 0.92, 0.94, 0.96 and 0.99) for a period of 30 days. Growth responses were evaluated over time in terms of colony diameter changes. Growth data were fitted to the primary model of Baranyi and the resulting growth rates were further modeled as a function of temperature and water activity using the cardinal model with inflection (CMI) ( Rosso et al., 1993). A logistic regression quadratic polynomial model was also employed to predict the probability of growth over storage time. Estimated parameters for minimum, maximum and optimum temperatures for growth were 9.1 °C, 46.4 °C and 32.1 °C for B. fulva and 10.5 °C, 43.2 °C and 32.1 °C for B. nivea. The respective values for aw were 0.893, 0.993 and 0.985 for B. fulva and 0.892, 0.992 and 0.984 for B. nivea. No growth was observed at 0.88 aw regardless of temperature for both species, whereas B. nivea ascospores could not grow at 10 and 45 °C irrespective of aw. Regarding growth boundaries, the degree of agreement between predictions and observations was >98% concordant for both species. The erroneously predicted growth cases were 1.4–4.2% false positive and 2.1–3.5% false negative for B. nivea and B. fulva, respectively. The developed logistic model was validated with two literature data sets as well as with data from independent experiments carried out on fruit juices. Validation results showed that agreement with literature data for growth was 25 out of 36 (69.4%) cases, whereas validation on fruit juice data failed in only 6 cases (5 false positives and 1 false negative) out of 128 cases.  相似文献

The Burkholderia multivorans strain ATCC 17616 carries three circular chromosomes with sizes of 3.4, 2.5, and 0.9 Mb. To reveal the distribution and organization of the genes for fundamental cell functions on the genome of this bacterium, the dnaA and dnaK gene regions of ATCC 17616 were cloned and characterized. The gene organization of the dnaA region was rnpA-rmpH-dnaA-dnaN-gyrB with a single consensus DnaA-binding box (TTATCCACA) between the rmpH and dnaA genes. This intergenic region, however, did not work as an autonomously replicating sequence in ATCC 17616. On the other hand, the gene organization of the dnaK region was grpE-orf1 (gene for thioredoxin homologue)-dnaK-dnaJ-pabB (gene for p-aminobenzoate synthetase component homologue). A putative heat-shock promoter that showed good homology to the sigma32-dependent promoter consensus sequence in Escherichia coli was found upstream of the grpE gene, suggesting that these five genes constitute an operon. In M9 succinate minimal medium the dnaJ mutant grew more slowly than the wild-type strain, indicating that this operon is functional. Pulsed-field gel electrophoresis and Southern blot analyses indicated that both the dnaA and dnaK gene regions exist as single copies on the 3.4 Mb chromosome.  相似文献

Stochastic models, including the variability in extent and probability of microbial growth, are useful for estimating the risk of foodborne illness (i.e. Nauta, 2000). Risk assessment typically has to embrace all sources of variability. In this paper, a stochastic approach to evaluate growth of heat damaged Listeria monocytogenes cells influenced by different stresses (pH and presence of eugenol) was performed, using an individual-based approach of growth through OD measurements. Both the lag phase duration and the “work to be done” (h0 parameter) were derived from the growth curves obtained. From results obtained histograms of the lag phase were generated and distributions were fitted. Histograms showed a shift to longer lag phases and an increase in variability with high stress levels. Using the distributions fitted, predictions of time to unacceptable growth (102 cfu/g) of L. monocytogenes were established by Monte Carlo simulation and they were compared with results from statistical methods. It was evidenced that both methods (Monte Carlo and regression analysis) gave a good indication of the probability of a certain level of growth other than the average. Tornado plots were obtained to establish a sensitivity analysis of the influence of the conditions tested (heat, pH, eugenol) applied to the microorganism and their combinations.  相似文献
