为了改善秘鲁鱿鱼鱼糜的凝胶特性,本文研究了添加不同种类的蛋白添加剂(卵清蛋白,猪血浆蛋白,乳清蛋白,大豆蛋白)对秘鲁鱿鱼鱼糜凝胶破断力,凹陷距离,凝胶强度,色泽的影响,并通过SDS-PAGE电泳分析其肌原纤维蛋白的降解情况。结果发现:添加卵清蛋白可显著增强鱿鱼鱼糜的破断强度和凝胶强度(p<0.05),添加乳清蛋白和大豆蛋白对秘鲁鱿鱼鱼糜破断强度和凝胶强度的提高没有明显的促进作用,而添加猪血浆蛋白粉则对秘鲁鱿鱼鱼糜的破断强度和凝胶强度有一定的降低作用。添加各种蛋白粉的鱿鱼鱼糜凝胶凹陷度变化不大。而添加几种蛋白粉均会使鱼糜凝胶的白度降低,其中添加猪血浆蛋白粉对鱼糜白度的影响最大。通过SDS-PAGE分析,秘鲁鱿鱼鱼糜在添加蛋白后肌球蛋白的变化不明显。这表明蛋白添加剂的蛋白酶抑制剂作用效果不大。   相似文献

The object of the study was to analyse the Theological behaviour occurring during heat-induced gelation (measured by thermal scanning rigidity monitor) of meat batters containing various proportions of fat (48, 106, 147 and 208 g kg?1) and sardine surimi (0, 100, 200 and 300 g kg?1 fish protein added with respect to total protein present) and subjected to different heat treatments (A, heating from 14 to 76?C at a rate of 1?C min?1; B, gradual temperature increase at a rate of 1?C min?1 up to 40?C, maintenance at this temperature for 30 min, then heating from 40 to 76?C at 1?C min?1; C, samples kept at 4?C for 20 h and then heated in the same way as process A). The results indicate that differences in levels of fat content significantly affect the gelation patterns of pork batters, gels being stiffer the higher the fat content and the lower the water content. A highly significant correlation (r = 0.78, P < 0.01) has been established between fat content and maximum values of G, which occur at the final experimental temperature of 76?C. In contrast, in general, neither addition of surimi nor variation in the heat treatment applied has any major effect on the gelation process in the system as formulated.  相似文献

Protein dispersions with different ratios of α-lactalbumin to β-lactoglobulin were heat-denatured at pH 7.5 and then acidified with glucono-δ-lactone to form gels at room temperature. Heat treatment induced the formation of whey protein polymers with reactive thiol group concentrations ranging from 1 to 50 μmol/g, depending on protein composition. During acidification, the first sign of aggregation occurred when the zeta potential reached −18.2 mV. Increasing the proportion of α-lactalbumin in the polymer dispersions resulted in more turbid gels characterized by an open microstructure. Elastic and viscous moduli were reduced, while the relaxation coefficient and the stress decay rate constants were increased by raising the proportion of α-lactalbumin in the gel. After one week of storage at 5 °C, gel hardness increased by 12%. The effect of protein composition on acid-induced gelation of whey protein is discussed in relation to the availability and reactivity of thiol groups during gel formation and storage.  相似文献

A protein dispersion blend of β-lactoglobulin and α-lactalbumin was heat-denatured at pH 7.5, hydrolyzed by α-chymotrypsin and then acidified with glucono-δ-lactone to form gels at room temperature. Heat treatment induced the formation of whey protein polymers with high concentration of reactive thiol groups (37 μmol/g). The reactive thiol group concentration was reduced by half after 40 min enzymatic hydrolysis. It was further reduced after enzyme thermal deactivation. During acidification, the first sign of aggregation for hydrolyzed polymers occurred earlier than for non hydrolyzed polymers. Increasing the hydrolysis duration up to 30 min resulted in more turbid gels characterized by an open microstructure. Elastic and viscous moduli were both reduced, while the relaxation coefficient and the stress decay rate constants were increased by increasing the hydrolysis duration. After one week storage at 5 °C, the hardness of gels made from hydrolyzed polymers increased by more than 50%. The effect of polymer hydrolysis on acid-induced gelation is discussed in relation to the availability and reactivity of thiol groups during gel formation and storage.  相似文献

目的 探究4种回收水溶性蛋白对罗非鱼鱼糜凝胶特性及质构的影响。方法 分别将等电点沉降法回收蛋白、海藻酸钠单一絮凝剂回收蛋白、壳聚糖单一絮凝剂回收蛋白和海藻酸钠-壳聚糖复合絮凝剂沉降回收蛋白以0%、5%、10%、15%、20%掺入罗非鱼鱼糜制备鱼肠,比较各样品在凝胶强度与全质构分析(texture profile analysis, TPA)参数上的差异,包括硬度、胶着性、咀嚼性、内聚性与弹性。结果 4种回收蛋白替代会对凝胶强度产生不利影响,通过等电点沉降法回收的蛋白对凝胶强度的影响最大,凝胶强度持续下降,最低达1708g·mm;以复合絮凝剂回收蛋白的掺入对原凝胶强度的影响最小,通过替代5%鱼糜,凝胶强度降为3833 g·mm。当用壳聚糖单一絮凝剂回收蛋白掺入5%时,与原鱼糜制品的TPA参数最接近,表现为硬度2505 g,胶着性1788 g,咀嚼性192.3 mJ,且内聚性和弹性也没有表现出明显差异。结论 这4种回收蛋白均以5%的添加量替代罗非鱼鱼糜时,制品凝胶强度在企业的可接受线性范围内,综合凝胶强度与TPA参数,以壳聚糖单一絮凝剂回收的蛋白替代产生的影响最小,可为回收鱼糜蛋白这一资源...  相似文献

The cold-set gelation of soy protein isolate (SPI)-gellan gum was induced by the addition of salts (KCl or CaCl2) using two different procedures: the direct addition of salts (fast gelation) or the diffusion of salts through a membrane (slow gelation). The mechanical properties, syneresis and microstructure of the mixed gels were evaluated, as well as for gellan and SPI gels. The mixed gels induced by calcium diffusion were stronger and more deformable than gels induced by the direct addition of calcium, while the opposite occurred for potassium-induced gels. All the mixed gels were macroscopically homogeneous, but at the microscopic level two independent networks could be observed. These two separate networks were more evident for the calcium-induced gels, and the structural characteristics depended strongly on the concentration of the protein and the polysaccharide. However an organized microstructure with the formation of microtubes surrounded by other network was only observed for the mixed gels induced by calcium diffusion at the higher protein/polysaccharide (10:1) ratio. Thus besides the composition and concentration of the biopolymers, the results showed that the type of salt and its velocity of incorporation led to gels with different structures and consequently different mechanical properties.  相似文献

为探究蛋清蛋白对豌豆淀粉凝胶化及凝胶特性的影响。分别以0%、3%、6%、9%和12%的蛋清蛋白替代豌豆淀粉,研究蛋清蛋白对豌豆淀粉糊化特性、热特性、静态流变、动态流变、凝胶质构及水分子状态的影响。结果表明:蛋清蛋白以浓度依赖的方式影响豌豆淀粉的糊化特性,峰值黏度、谷值黏度、崩解值、最终黏度、回生值均随蛋清蛋白添加量增加而降低,而糊化温度则升高。蛋清蛋白对豌豆淀粉凝胶化温度影响不显著,但降低了淀粉的凝胶化焓值。添加不同量蛋清蛋白的豌豆淀粉糊均表现为假塑性流体特征,Herchel-Bulkley模型能够很好地拟合其静态流变行为,稠度指数和屈服应力均随蛋清蛋白添加量增加而降低。添加蛋清蛋白降低了豌豆淀粉糊体系的黏弹性及凝胶的硬度、强度和可塑性。蛋清蛋白对豌豆淀粉凝胶中自由水含量影响不显著,但使结合水含量增加而不可流动束缚水含量降低。  相似文献

对肉类食品来说,辐照不仅能有效杀灭其中的微生物,而且能作用于蛋白质等生物大分子,影响蛋白间的相互作用力,从而改变蛋白质构象,导致蛋白变性、聚集或凝胶化,因此利用电子束辐照处理,有望成为改善肌肉蛋白凝胶品质的一种有效手段。本文列举了常用于表征蛋白质结构的光学仪器如拉曼光谱、红外光谱、圆二色谱等在肌肉蛋白构象研究中的应用,综述了辐照引起肌肉蛋白生化特性、化学作用力和构象的改变,以及这种改变对肌肉凝胶网络结构形成的作用,在分子水平上探讨了辐照对肌肉凝胶特性的影响机理,为利用辐照技术改善肌肉凝胶性能提供理论指导。   相似文献

为了探究无机盐对大豆蛋白酶促胶凝过程热动力学特性及凝胶强度的影响规律,以大豆分离蛋白分散液(Dispersion of soybean protein isolate,DSPI,4%,m/m)为材料,以木瓜蛋白酶为凝固剂进行酶促胶凝试验,测定不同无机盐种类及浓度下的胶凝时间,并通过Arrhenius方程求出反应活化能;以凝胶强度为指标,利用Box-Behnken中心组合设计试验建立以Ca2+浓度、温度及酶添加量为参数的凝胶强度模型。结果表明添加无机盐(Na Cl、KCl、Ca Cl2、Mg Cl2和Mg SO4)可降低酶促DSPI凝胶的活化能,但Arrhenius曲线仍呈线性关系,说明无机盐可加快反应速度,但胶凝反应仍遵循同一机理;显著性检验及响应面分析表明所建凝胶强度模型拟合度较高,木瓜蛋白酶添加量对凝胶强度影响显著(P0.05),Ca Cl2添加量与温度的交互作用对凝胶强度的影响极显著(P0.01),Ca Cl2和温度同时较高或较低时,均不利于提高凝胶强度。  相似文献

The role of soluble and insoluble aggregates induced by soy protein isolate (SPI) processing in the gelling properties of myofibrillar protein (MP) was studied. Incorporating soluble SPI aggregate could greatly improve (< 0.05) the elastic modulus (G’) and water‐holding capacity (WHC) of MP gel, but had no notable effect on MP gel strength. In contrast, incorporating the insoluble SPI aggregate significantly enhanced the G’, strength and WHC of MP gel, although the improvement in WHC was smaller than that produced by the soluble aggregate. The results of environmental scanning electron microscopy showed that the soluble SPI aggregate induced a less randomly composite gel structure, which may explain its notable enhancement of WHC. However, the insoluble SPI aggregate appeared to be granules embedded in the continuous MP gel matrix, which may be related to the reinforcement of gel strength. Hence, the results of this study suggest further means of processing commercial SPI for use in meat products.  相似文献

研究不同盐浓度下肌原纤维蛋白的氧化对后继的微生物谷氨酰胺转移酶(MTG)交联反应及凝胶能力的影响。分散在三种盐浓度(0.15、0.45和0.6mol/L NaCl)下的肌原纤维蛋白在4℃分别氧化2h和24h,然后在0.6mol/L NaCl条件下加入MTG于30℃交联2h。通过电泳和溶解度研究蛋白的氧化对MTG交联反应的影响,通过凝胶强度研究蛋白的氧化对MTG交联反应后凝胶性的影响。研究表明,三种盐浓度下的氧化都会促进MTG交联反应,特别是高盐条件下的氧化(提高量达47.8%)。所有样品经MTG交联后凝胶强度均显著增加(p<0.05),高盐条件下氧化的蛋白其凝胶强度明显高于低盐条件,然而在所有盐浓度下,氧化蛋白的凝胶强度增幅均小于未氧化的。结果表明过多的交联反应反而不利于蛋白成胶性。   相似文献

Rheological properties of heat-induced pea protein isolate (PPI) gels with added microbial transglutaminase (MTGase) were studied under various reaction conditions. A positive linear relationship was observed between level of MTGase used (0 to 0.7% w/w) and shear stress and shear strain of heat-set commercial pea protein isolate (PPIc) gels at 92 °C following incubation at 50 °C. Use of MTGase allowed for preparation of PPIc gels of similar strength and elasticity as commercial soy protein isolate gels and commercial meat bologna. MTGase treatment did not alter thermal properties of PPI gels. The shear stress and strain of PPIc gels were also improved following low temperature (4 °C) incubation of PPI with MTGase. Enhancement of shear strain or gel elasticity of heat-induced PPI gels with MTGase has not been reported before and provides opportunities for extending the properties of pea proteins when developing new food products.  相似文献

目的研究钙盐、外源蛋白及其复配物对含微生物产转谷氨酰胺酶(MTG)的鲢鱼糜凝胶品质的影响。方法将碳酸钙、乳酸钙、卵清蛋白、大豆分离蛋白及其复配物添加到含MTG(2 U/gg_(鱼蛋白))的鲢鱼糜中并制成凝胶,测定鱼糜凝胶的持水性、质构特性(破断强度、凹陷深度、硬度、内聚性、咀嚼性)、白度和聚丙烯酰胺凝胶电泳图谱。结果添加碳酸钙、乳酸钙、卵清蛋白、大豆分离蛋白及其复配物均可不同程度地改善含MTG的鱼糜凝胶的凝胶特性,且添加其复配物优于单独添加碳酸钙、乳酸钙、卵清蛋白、大豆分离蛋白。经试验优化,将8μmol/U_(MTG)碳酸钙、4 mg/U_(MTG)卵清蛋白、4 mg/U_(MTG)大豆分离蛋白添加到含有2 U/g_(鱼蛋白)MTG的鲢鱼糜中可获得最佳凝胶品质,鱼糜凝胶的破断力、凹陷深度最高,硬度、咀嚼性、白度均显著高于单独添加MTG(P0.05),也明显高于添加2 U/g_(鱼蛋白)其他品牌商品MTG的(P0.05)。结论 CaCO_3与外源蛋白复配物对含有MTG的鱼糜凝胶特性具有显著促进作用,将碳酸钙、卵清蛋白、大豆分离蛋白与MTG按比例混合可制成鱼糜制品专用的复配酶制剂。  相似文献

The influence of fried fish consumption on dietary iron bioavailability in rats was studied. Sardines (Clupea pilchardus), raw and fried in olive oil, were used whole or defatted to prepare the samples. Five diets containing casein and the different samples as protein source were prepared. After in vitro digestion of the diets a greater proportion of soluble iron was found in the casein diet compared with the sardine diets. Five groups of rats were fed the diets over 28 days. Body weight and food intake were monitored weekly, and during days 21–28 an iron balance study was carried out. No significant differences were observed in food intake, body weight and protein efficiency ratio between groups. Absorbed and retained iron and apparent iron digestibility were higher in rats fed the sardine diets with respect to the control group, except in the whole fried sardine group. Metabolic efficiency and retention efficiency were increased in the four sardine groups. The frying process led to significant changes in the overall digestive and nutritive efficiency of dietary iron, which were different depending on whether the diets contained whole sardine or its protein. It was concluded that the consumption of sardines, whether raw or fried in olive oil, enhances dietary iron availability. © 2002 Society of Chemical Industry  相似文献

ABSTRACT:  The effect of rainbow trout plasma protein (RPP) on the gelation of Alaska pollock surimi was determined to evaluate the possibility of its commercialization as a new protein additive. For modori gel, the breaking force, deformation, whiteness, and water holding capacity increased as the addition amount of RPP (0 to 0.75 mg/g) increased, and decreased at higher concentration of RPP (0.75 to 1.50 mg/g) ( P < 0.05). Protein solubility of modori gel in the mixture of SDS, urea, and β-mercaptoethanol decreased as the addition amount of RPP increased up to 0.75 mg/g, and increased at higher concentration of RPP (0.75 to 1.50 mg/g) ( P < 0.05). The contents of trichloroacetic acid-soluble peptide decreased as the addition amount of RPP (0 to 1.50 mg/g) increased ( P < 0.05). Based on the result of sodium dodecyl sulfate (SDS)-polyacrylamide gel electrophoresis (PAGE), most myosin heavy chain of surimi was not degraded when RPP was added. Thus, RPP was supposed to act as a protease inhibitor in the gelation of Alaska pollock surimi. An RPP of 0.75 mg/g was the optimal concentration to prevent the gel weakening of Alaska pollock surimi. Compounds with molecular weights less than 10 kDa in RPP had no significant effect on the gelation of Alaska pollock surimi based on the result of the dialyzed RPP.  相似文献

The effects of microbial transglutaminase (TG) and sodium alginate (SA) systems on cold-set gelation of myofibrillar protein (MP) at various salt levels were investigated. The gelation kinetics data showed that both TG and SA had optimal but different salt levels to form an acceptable cold-set MP gel. Although their gelling characteristics were altered with different salt levels, the combination of TG and SA showed a rapid cold-set MP gel formation, regardless of salt levels. The effect of TG and SA on gel strength was reversed by increasing the salt level, while the cold-set MP gel strength of the TG and SA combination was not affected by salt levels. Furthermore, the SA system contributed to improved water-binding ability and cold-set MP gel formation, while the TG system enhanced the gel strength of the MP after cooking at low-salt levels. In addition, TG's effects on gel strength of MP increased at higher salt levels, the SA system prevented the moisture loss induced by the TG reaction. There was no evidence of any interaction between SA and MP in the thermogram and gel electrophoresis data. Results from this study suggested that a system combining TG with SA had the potential advantage of improving the water-binding ability and producing cold-set MP gelation at an even lower salt level than TG alone.  相似文献

ABSTRACT:  Differential scanning calorimetry (DSC) and dynamic oscillatory shear testing were performed to study the influence of inulin (Raftiline® HP-gel and Raftiline® ST-gel) and oligofructose (Raftilose® P95) on the thermal stability and gelation (using glucono-δ-lactone [GDL] as a coagulant) of soy protein isolate (SPI) dispersions. Addition of 10% (w/v) inulin/oligofructose or sucrose increased ( P < 0.05) the peak denaturation temperatures ( Tm ) of 7S and 11S soy proteins in SPI dispersion (5%[w/v], pH 7.0) by an average of 1.9 and 2.3 °C, respectively. GDL induced SPI thermal gelation, and the gel rheology was affected by both the pH decline and the specific temperature of heating. Addition of inulin/oligofructose (8%, w/v) improved the gelling properties of preheated SPI dispersion (8%, w/v) coagulated with GDL, showing 14.4 to 45.6% increase ( P < 0.05) in gel rigidity ( G ' value) at the end of heating (81 °C). Microstructural examination revealed a denser protein cross-linking structure and reduced pore sizes in SPI gels containing inulin/oligofructose. In general, inulin was more capable of improving SPI gelation than oligofructose, suggesting that the degree of fructose polymerization in the fructans was of thermal and rheological importance.  相似文献

Beef plasma protein (BPP) and egg white, at levels of 10, 20 and 30 g kg−1, and sodium ascorbate (SA) and microbial transglutaminase (MTGase), at levels of 1, 2 and 3 g kg−1, were individually added into red tilapia surimi. This gel was set at 40 °C for 90 min followed by heating at 90 °C for 30 min. Gel qualities were analysed and compared with those of non-supplemented red tilapia control gel. Compared with the control, each additive significantly affected gel strength due to an increase in breaking force. Although BPP could improve texture characteristics, higher addition levels of BPP significantly affected gel whiteness. The addition of SA and MTGase were found to increase surimi gel strength and improve whiteness. Amongst all treatments, the addition of 2 g kg−1 MTGase was found to provide the best gel qualities.  相似文献

利用DSC研究了魔芋葡苷聚糖、大豆分离蛋白及两者的混合物凝胶在升、降温过程中的热效应变化。结果表明,在升温过程中,单独的魔芋葡苷聚糖(15%)在水溶液中或加入弱碱或强碱性凝固剂后所形成的凝胶均表现出放热效应;而单独的大豆分离蛋白水溶液(15%)所形成的凝胶没有产生热效应,但加入0.1mol/L的NaoH后却产生了明显的放热效应。对于魔芋葡苷聚糖与大豆分离蛋白所形成的混合凝胶,在碱性条件下,随魔芋葡苷聚糖含量的增加其放热温度升高,但大豆分离蛋白含量较高的凝胶,在高于85℃处有较强的放热效应;而在弱碱(0.1mol/L的Na2CO3)条件下,KGM(≥13%)含量较高的凝胶产生放热效应,其它凝胶基本没有热效应产生。实验还表明,所有的凝胶样品在降温过程中基本没有出现吸热和放热峰,表明所形成的凝胶为热不可逆凝胶。  相似文献
