Heterotrophic plate count (HPC) constitutes a common indicator for monitoring of microbiological water quality in distribution systems (DS). This paper aims to identify factors explaining the spatiotemporal distribution of heterotrophic bacteria and model their occurrence in the distribution system. The case under study is the DS of Quebec City, Canada. The study is based on a robust database resulting from a sampling campaign carried out in about 50 DS locations, monitored bi-weekly over a three-year period. Models for explaining and predicting HPC levels were based on both one-level and multi-level Poisson regression techniques. The latter take into account the nested structure of data, the possible spatiotemporal correlation among HPC observations and the fact that sampling points, months and/or distribution sub-systems may represent clusters. Models show that the best predictors for spatiotemporal occurrence of HPC in the DS are: free residual chlorine that has an inverse relation with the HPC levels, water temperature and water ultraviolet absorbance, both having a positive impact on HPC levels. A sensitivity analysis based on the best performing model (two-level model) allowed for the identification of seasonal-based strategies to reduce HPC levels. 相似文献

Effects of substrate concentrations on the growth of heterotrophic bacteria and algae in secondary facultative ponds 总被引:1,自引:0,他引:1
This paper presents the effect of substrate concentration on the growth of a mixed culture of algae and heterotrophic bacteria in secondary facultative ponds (SFPs) utilizing settled domestic sewage as a sole source of organic carbon. The growth of the mixed culture was studied at the concentrations ranging between 200 and 800 mg COD/l in a series of batch chemostat reactors. From the laboratory data, the specific growth rate (micro) was determined using the modified Gompertz model. The maximum specific growth rate ( micro(max)) and half saturation coefficients (K(s)) were calculated using the Monod kinetic equation. The maximum observed growth rate ( micro(max)) for heterotrophic bacteria was 3.8 day(-1) with K(s) of 200 mg COD/l. The micro(max) for algal biomass based on suspended volatile solids was 2.7 day(-1) with K(s) of 110 mg COD/l. The micro(max) of algae based on the chlorophyll-a was 3.5 day(-1) at K(s) of 50mg COD/l. The observed specific substrate removal by heterotrophic bacteria varied between the concentrations of substrate used and the average value was 0.82 (mg COD/mg biomass). The specific substrate utilization rate in the bioreactors was direct proportional to the specific growth rate. Occurrence of sulfonamide and tetracycline-resistant bacteria and resistance genes in aquaculture environment 总被引:20,自引:0,他引:20
The occurrence of sulfonamide and tetracycline resistance and their pollution profile in the aquaculture environment of Tianjin, northern China, were investigated. The presence of antibiotic-resistant bacteria was identified and the corresponding antibiotic resistance genes (ARGs) were quantified at 6 aquaculture farms in Tianjin. Sulfonamide-resistance genes were prevalent and their concentrations were the highest detected (3.0 × 10−5 to 3.3 × 10−4 for sul1/16S rDNA, 2.0 × 10−4 to 1.8 × 10−3 for sul2/16S rDNA) among the various ARGs, most likely because the use of sulfonamides is more prevalent than tetracyclines in this area. Bacillus was the most dominant bacterial genus in both sulfamethoxazole resistant bacteria (63.27% of the total resistant bacteria) and tetracycline-resistant bacteria (57.14% of the total resistant bacteria). At least two of those genes (tetM, tetO, tetT, tetW, sul1 and sul2) were detected in the isolates of Bacillus cereus, Bacillus subtilis, Bacillus megaterium and Acinetobacter lwofii, and all of the above genes were detected in B. cereus, suggesting the occurrence of multi-resistance in the studied area. The genetic transfer of sul1 between intestinal bacteria (e.g., Enterococcus spp.) and indigenous bacteria (e.g., Bacillus spp.) was implied by phylogenetic analysis. Several strains of resistant opportunistic pathogens (e.g., Acinetobacter spp.) were found in indigenous bacteria, which increase the risk of ARGs to public health. Overall, this is the first study to comprehensively investigate the antibiotic resistance profile by analyzing the species of antibiotic-resistant bacteria and adopting qualitative and quantitative methods to investigate ARGs at a typical aquaculture area in northern China. 相似文献

This study compared the Quanti-Disc most probable number (MPN) test for heterotrophic bacteria from drinking water with the widely used yeast extract agar (YEA) pour plate method. The Quanti-Disc test module contains 50 reaction wells in which a medium has been pre-deposited. The medium contains a suite of three fluorogenic enzyme substrates selected for the detection of enzymes expressed widely by heterotrophic bacteria. The MPN of heterotrophic bacteria is calculated from the number of fluorescing reaction wells after incubation of a sample. Quanti-Disc and the YEA pour plate method were compared according to guidance on comparing methods given in United Kingdom national guidance and ISO 17994:2004. The two methods were also challenged with reference strains and isolates of heterotrophic bacteria from drinking water. This indicated that heterotrophic bacteria commonly encountered in drinking water are detected by both the YEA pour plate method and Quanti-Disc. Analysis of data from split water samples (723 for 37 degrees C tests and 872 for 22 degrees C tests) from nine geographically diverse laboratories in England and Wales demonstrated that the Quanti-Disc method is equivalent to the YEA pour plate method for the analysis of heterotrophic bacteria from drinking and similar waters at 37 degrees C, and superior to YEA for the analysis at 22 degrees C. The Quanti-Disc method is a simple and efficient alternative method for the enumeration of heterotrophic bacteria from drinking water. 相似文献

Little is known about the microbial selectivity of UV treatment for antibiotic resistant bacteria, and the results of limited studies are conflicting. To understand the effect of UV disinfection on antibiotic resistant bacteria, both total heterotrophic bacteria and antibiotic resistant bacteria (including cephalexin-, ciprofloxacin-, erythromycin-, gentamicin-, vancomycin-, sulfadiazine-, rifampicin-, tetracycline- and chloramphenicol-resistant bacteria) were examined in secondary effluent samples from a municipal wastewater treatment plant. This study examined the impacts of bacteria on the algal biomass, lipid content and efficiency of wastewater treatment during the heterotrophic cultivation of Chlorella pyrenoidosa. Our results showed that soybean-processing wastewater can enhance the accumulation of lipids in algal cells and thus raise the lipid yield in the pure culture. The bacteria coexisting with algae improved the degradation of total nitrogen (TN), total phosphorus (TP), glucose and chemical oxygen demand (COD). Although the biomass productivity of algae was not significantly affected, the total algal lipid content and lipid production rate were slightly reduced when bacteria coexisted with algae. The difference in the compositions of the medium is presumed to be the main contributing factor for the variation in total lipid content in presence and absence of bacteria. The TN, TP, and COD decreased during the assimilatory process undertaken by C. pyrenoidosa, and the removal efficiency of TN by bacteria depended on the type of nitrogen species in the medium. Additionally, the apparent interaction between the bacterial and algal cultures varied with the changes in experimental conditions. Algae could compete with bacteria for the carbon and energy sources, and inhibit the growth of the bacteria in the presence of high organic matter concentration in the medium. 相似文献

本文利用中型动三轴仪对不同降解度的城市固体废弃物(MSW)的残余变形进行试验研究,分别采用应力水平相关的改进沈珠江模型和固结应力比相关的改进沈珠江模型对MSW的残余变形模型参数进行拟合,得出不同降解度MSW的残余变形参数值。研究表明:降解度为0%和50%的MSW的残余变形规律,均可以采用沈珠江改进模型进行模拟,应力水平指数为0.5或固结应力比指数为0.8时,参数拟合性均较好;在不同破坏标准下,随着降解度的增加,MSW的残余变形参数均逐渐减小。本文的研究成果对垃圾填埋场的抗震设计和垃圾填埋场震后变形的预估有重要的参考价值。 相似文献

Analysis by HPLC identified 14 individual chlorophenols, nitrophenols and phenoxyalkanoic acids in a small stream in West Yorkshire; concentrations fluctuated markedly over 12 months. Abundance of bacteria (as direct counts and colony-forming units) and heterotrophic activity (as fraction of glucose substrate mineralized) were measured in water and artificial gravel substratum at two sites in this stream. Mean bacterial abundance, and at one site activity, were at least similar to those found in an adjacent unpolluted stream. Sometimes, however, colony-forming units and activity were depressed. Multiple-regression analysis demonstrated relationships between bacteria and pollution-related variables; low pH and nitrophenols (particularly picric acid), were identified as possible causes of bacterial inhibition. Taxonomic investigation of isolates from sediment demonstrated that bacteria in the polluted stream were dominated by Pseudomonas, whereas a wide range of mainly Gram-negative bacteria was present in the unpolluted stream. 相似文献
