HPLC法测定北虫草中的虫草素含量 总被引:1,自引:0,他引:1
建立了快速测定北虫草及其制品中虫草素的含量测定方法。采用ApolloC18色谱柱(250mm×4.6mm);流动相甲醇(10%~40%)—水(90%~60%),30min内梯度洗脱;流速1.0ml/min;检测波长260nm。在此液相条件下,虫草素在浓度0.05~0.25mg/ml范围内有良好的线性关系(r=0.9999);仪器精密度考察RSD为1.65%;稳定性考察RSD为0.51%;重现性考察RSD为0.70%;加样回收率试验RSD为1.25%。结果表明,该法测定北虫草及其制品中虫草素的含量,快速准确,重现性好。 相似文献

研究了不同变温方式对绿芦笋贮藏品质及相关酶类活性的影响。实验以阿波罗品种绿芦笋为试材,通过3种不同的升温方式,研究了变温方式对绿芦笋货架品质的影响。实验结果表明:逐渐升温的方式可以有效地延长绿芦笋的货架期,其中A-3处理效果最为明显,该处理可以使绿芦笋保持较好的感官品质和更高的可溶性固形物含量(TSS)。货架期为8d,A-3感官评分为17分,TSS为3.91%。而A-1感官评分为10分,TSS为3.42%;且A-3处理绿芦笋的硬度变化较小;同时,A-3能减缓绿芦笋电导率的上升,货架期为8d,其电导率为18.27%,而A-1处理为其1.2倍;A-3处理绿芦笋的PPO、POD和CAT活性总体高于其余两种处理和对照。 相似文献

Prototypes of the automatic-dipping system Apollo were tested with the IQ milking cluster (GEA Farm Technologies GmbH, Bönen, Germany) to determine the teat-dip residues in the milk and the dipping performance (number of dipped teats) of the system compared with manual (hand) dipping. A laboratory trial and a field trial at a dairy farm were performed to determine the iodine level in the milk when an iodine-based teat dip was used. In the laboratory trial, the mean difference between the 53 paired samples (sampling upstream and downstream of the cluster) was 18.9 ± 3.18 μg of iodine/kg. A field trial at a 300-cow commercial dairy farm consisted of taking 2 sets of individual cow milk samples 6 wk apart. Three weeks before the second test day, the iodine-based teat dip was replaced by an iodine-free teat dip. The mean difference between the 2 sets of 55 samples was 25.1 ± 5.22 μg/kg. Compared with manually applying an iodine-based teat dip, the increase in the iodine content resulting from the use of the tested cluster with automatic dipping was very low and would not be an issue of food safety. The dipping performance tests were completed on the same 300-cow commercial dairy farm as the field iodine level trial was performed. Zusammenfassung Die Proteine der Dinkelsorten Steiners roter Tiroler (SRT), Oberkulmer Rotkorn (OKR) und Rouquin (ROU) wurden mit einem modifizierten Osborneverfahren extrahiert. Die Proteinverteilung auf die Osbornefraktionen und die Aminosaurezusammensetzung der Proteinfraktionen ist sehrähnlich und entspricht den von Weichweizen bekannten Werten. Die RP-HPLC der Gliadine liefert für SRT und OKR sehr ahnliche Muster, während ROU insbesondere im ersten Teil der-Gliadinfraktion abweicht. REK hat ein anderes Gliadinmuster. SRT und OKR habenauch ein sehr dhnliches Gluteninmuster. ROU weicht ab und entspricht insbesondere im Bereich der HMW-Untereinheiten guten Weichweizensorten wie z. B. REK. Der LMW-Teil der Chromatogramme ist bei allen untersuchten Dinkelsorten von REK verschieden. Die Untersuchung der Glutenine mit Hilfe der SDS-PAGE bestatigt die mit der RPHPLC erhaltenen Ergebnisse. Das Bandenmuster von SRT und OKR korrespondiert im Bereich der HMW Untereinheiten mit dem schwacher Weichweizen. ROU zeigt im HMW-Bereich acht Banden, von denen drei bei Weichweizen mit guten Backeigenschaften korreliert sind. Drei weitere Banden wurden bei Weichweizen von uns bisher nicht beobachtet.

Summary The proteins of the varietiesSteiners roter Tiroler (SRT),Oberkulmer Rotkorn (OKR) andRouquin (ROU) of the hexaploid wheat spelt (Triticum spelta) were extracted by a modified Osborne procedure. The amounts of protein in the Osborne fractions and the amino acid compositions of the total proteins, the albumins/globulins, the gliadins and the glutenins are in the range of values for the corresponding protein fractions from soft wheat varieties (T.aestivum)Rektor (REK) and Apollo (APO).Reversed phase HPLC of the gliadins (cf. Z Lebensm Unters Forsch (1987) 185:371) produced very similar patterns for SRT and OKR, while ROU is somewhat different, expecially in the first part of the -gliadin fraction. The pattern of REK is clearly different. The glutenin patterns of SRT and OKR obtained by RP-HPLC (cf. Z Lebensm Unters Forsch (1987) 185:487) are also similar. ROU exhibits a different pattern and corresponds expecially in the range of the high-molecular-mass subunits to REK and to other strong varieties of soft wheat. The low-molecular-mass part of the chromatograms of all threeT. spelta varieties differs from REK. The results of reversed-phase HPLC were confirmed by sodium dodecyl sulfate/polyacrylamide gel electrophoresis (SDS-PAGE). The high-molecular-mass pattern of SRT and OKR is similar to that of poor soft wheat varieties. ROU contains eight bands in this range, three of which are known to be correlated with good baking properties. Three other bands have not so far been observed by us in soft wheat varieties (cf. Aerobically grown pitching yeast is very rich in unsaturated fatty acids and sterol esters compared to traditional, anaerobic yeast. The principal fatty acids in aerobic yeast cells are unsaturated palmitoleic and oleic acids, whereas in anaerobic cells saturated palmitic acid predominates. The difference in fatty acid distribution between aerobic and anaerobic cells is most marked in the sterol esters. The fatty acids of phospho-lipids are more stable, although remarkable differences are observed. The sterols of aerobic cells are almost entirely in esterified form and zymosterol is the principal sterol. During the first hours of fermentation a rapid synthesis of palmitoleic acid is observed when anaerobic yeast is used for pitching and the wort is aerated. The synthesis of oleic acid requires more oxygen and time than is available under normal brewing conditions. When aerobic pitching yeast is used no more unsaturated fatty acids are synthesised and the lipid stores of pitching yeast are distributed among the daughter cells. The decrease in acetate ester production by aerobic pitching yeast is concluded to be due to a decrease in acetyl CoA synthesis, which may be caused by the high proportion of unsaturated fatty acids in membrane lipids. 相似文献
