Most species belonging to Scrophularia genus had been used as antiinflammatory drugs by the folk medicine. The phenylpropanoids are considered to be the active principles of these drugs with antiinflammatory action by different Authors, especially harpagoside and harpagide. In this report, the antiinflammatory effects of Scrophularia frutescens L. (Scrophulariaceae) was studied and the iridoid glucoside harpagoside has been evidenced and isolated for the first time from this plant. Aqueous extract, methanolic extract and harpagoside, isolated from the methanolic extract, were tested for antiinflammatory activity on the rat paw oedema. The results obtained showed that the aqueous extract has a small but significant antiinflammatory effect on carrageenan-induced oedema test, while methanolic extract has a lower antiinflammatory activity and the activity of the isolated harpagoside is remarkably low. Thus, the conclusion may be that S. frutescens L. is a potential antiiflammatory agent but its activity is not due to harpagoside.  相似文献

The effects of Lunularia cruciata (L.) Dum (Bryophyta) acetonic extract was studied in vitro by means of luminol-dependent chemiluminescence (CL) emission from human peripheral whole blood phagocytes and isolated polymorphonuclear leukocytes (PMNs). L. cruciata adult thalli underwent extraction with acetone. CL emission was evaluated in an automated luminometer, measuring the oxygen free-radical production by phagocytes incubated with opsonized zymosan (OZ) or phorbol myristate acetate (PMA), in absence or in presence of various concentrations of L. cruciata extract. The CL results indicated that L. cruciata induced significant changes in light emission from whole blood phagocytes, as well as isolated PMNs. Its inhibitory activity was more evident when resting isolated PMNs were studied. When the cells were activated, the greatest inhibitory effect was observed with PMA. The L. cruciata activity could be caused by several compounds, such as flavonoids and/or sesquiterpenes, present in the acetonic extract.  相似文献

In this work, we have studied the molluscicidal activity of different extracts obtained from Cotula cinerea and Quercus lusitania var. infectoria galls. The hydroalcoholic extract of Cotula cinerea, acetonic extract and gallotanin of Quercus infectoria galls have presented high activity against Bulinus truncatus. The hydroalcoholic extract of Cotula cinerea was fractionated by chromatography on silica gel column. We have isolated two very active fractions at concentrations respectively of 52.5 and 27.5 ppm.  相似文献

A polysaccharide containing 2-acetamido-2-deoxy-D-glucose (GlcNAc), 2-acetamido-2-deoxy-L-fucose (FucNAc), and 2-acetamido-2-deoxy-D-galacturonic acid (GalNAcA) was isolated from an aqueous phenol extract of lipid-free, isolated cell walls of the reference strain for Acinetobacter baumannii serogroup O5, by mild acid hydrolysis of the extract and chromatography of the water-soluble products on Sephadex G-50. By means of NMR studies, methylation analysis, carboxyl reduction and chemical degradations, the repeating unit of the polymer was identified as a branched tetrasaccharide of the structure shown. Bardanae Furctus (Goboshi) extract showed potent in vitro cytotoxicity and in vivo antitumor activity against human hepatoma HepG2 cells and mouse sarcoma 180 cells, respectively. In this study, the cytotoxicities of four fractions and three major components (arctiin, arctigenin, and chlorogenic acid) isolated from Goboshi extract were examined. Arctiin and arctigenin, which were contained in the ethylacetate fraction and n-butanol fraction, respectively, showed strong cytotoxicity against HepG2 cells, but little toxicity against Chang liver cells. Chlorogenic acid isolated from the water fraction did not affect the viability of these cells. The cytotoxicity of arctigenin against Chang liver cells was markedly potentiated by treatment with glutathione (GSH) synthesis inhibitor, L-buthionine-(S,R)-sulfoximine (BSO). On the other hand, in HepG2 cells, the cytotoxicity of arctigenin was hardly changed by BSO. The cytotoxicity of arctigenin against HepG2 cells increased in an exposure-time dependent manner. These characteristics of arctigenin were similar to those of Goboshi extract, as previously observed. We therefore conclude that the principal cytotoxic components of Goboshi extract are arctiin and its aglycone arctigenin.  相似文献

A MeOH extract from Z. africana was examined for topical antiinflammatory activity and proved to be active against arachidonic acid (AA) acute edema, 12-O-tetradecanoylphorbol 13-acetate (TPA)-induced chronic inflammation, and oxazolone delayed-type hypersensitivity in mice. The extract also showed significant inhibitory activity of Naja naja phospholipase A2 when a polarographic method was used. Two oleanane-type triterpene saponins, zanhasaponins A (1) and B (2), and the cyclitol pinitol (4), isolated from the extract, were active as inhibitors of PLA2. A further saponin, zanhasaponin C (3) was inactive in this assay.  相似文献

A fraction stimulating the antibody synthesis to sheep erythrocytes in administration during the neonatal period to mice and partially restoring the antibodygenesis in neonatally thymectomized rats was isolated from a calf thymus extract. Morpho-histochemical studies demonstrated a marked plasmocytic reaction in the spleen of neonatally thymectomized rats under the effect of the thymus extract fraction in response to immunization with sheep erythrocytes; however, there occurred no normalization of morphotopographic peculiarities of the splenic tissue, including the thymus-dependent regions. Administration of the extract was also not accompanied by any weight gain of the lymphoid organs, or increase in lymphocyte count in the peripheral blood and the count of splenic cells transforming into blasts under the effect of phytohemagglutinin.  相似文献

1. Effect of water soluble fraction of alcoholic extract of G. sylvestre leaves on glycogen content by isolated rat hemidiaphragm was studied in normal and glucose fed hyperglycemic rats. 2. The leaf extract by itself failed to alter the hepatic glycogen content in normal rats. 3. In glucose fed rats, the leaf extract lowered the glycogen content of the tissue significantly (P<0.05) and this was further lowered when both exogenous insulin and leaf extract was administered. 4. Two new flavone glucosides, 5,2',6'-trihydroxy-6,7,8-trimethoxyflavone 2'-O-glucoside and 5,2',6'-trihydroxy-6,7-dimethoxyflavone 2'-O-glucoside were isolated from the aqueous methanol extract of the roots of Scutellaria baicalensis. From the extract, seven phenolics, 5,7,2',6'-terahydroxyflavone, 5,7,2',5'-tetrahydroxy-8,6'-dimethoxyflavone, skullcapflavone II, baicalin, baicalin methyl ester, wogonin 7-glucuronide and 3,5,7,2',6'-pentahydroxyflavanone were also isolated.  相似文献

Triclisia dictyophylla Diels (Menispermaceae) is a woody climber indigenous to West Africa which has been used natively as a medicinal in the treatment of several ailments. Chromatography of an extract of the whole plant afforded tridictophylline (3), a new morphinan alkaloid whose structure was established by a consideration of spectral data and confirmed by x-ray crystallographic analysis. The bisbenzylisoquinoline dibenzodioxin alkaloids cocsuline (1) and trigilletimine (2) were also isolated from the same extract.  相似文献

The present study examined the effect of the MeOH extract, partially purified fraction (IV), and pure compounds from Argemone mexicana L. (Papaveraceae) on the morphine withdrawal in guinea pig isolated ileum. The MeOH extract, the partially purified fraction (IV), and the pure compounds isolated from A. mexicana significantly and in a concentration-dependent manner reduced the morphine withdrawal. Since the pure compounds were identified as protopine and allocryptopine, the observed effects could be related to these compounds. The results of the present study suggest that isoquinoline alkaloids may be potential agents in the treatment of drug abuse.  相似文献

The hexane extract from leaves of Vernonia brasiliana (L.) Druce (Compositae) was active in vitro against Plasmodium falciparum and in vivo in mice infected with Plasmodium berghei. This extract was subjected to a bioassay-guided fractionation protocol based on the in vitro model. Lupeol was identified as a compound responsible for the activity, inhibiting the P.falciparum growth by 45% when tested at 25 micrograms/ml. However, this triterpene was inactive in vivo when 15 mg/kg were administered per os during four consecutive days to mice infected with P.berghei. beta-Amyrin and germanicol, isolated from the same fraction that yielded lupeol, were inactive in the in vitro assay.  相似文献

Their structures were elucidated by chemical and spectral methods as 1-O-beta-D-glucopyranosyl chrysophanol and 1-O-beta-D-glucopyranosyl emodin. The methanol extract from Machilus thunbergii showed a suppressive effect on umu gene expression of the SOS response in Salmonella typhimurium TA1535/pSK1002 against the mutagen, 3-amino-1,4-dimethyl-5H-pyrido[4,3-b]indole (Trp-P-1), which requires liver metabolizing enzymes. The methanol extract from M. thunbergii was successively re-extracted with chloroform, butanol and water. A suppressive compound in the chloroform extract fraction was isolated by SiO2 column chromatography and identified as meso-dihydroguaiaretic acid by GC-MS, and 1H- and 13C-NMR spectroscopy. Meso-dihydroguaiaretic acid inhibited of the SOS-inducing activity of Trp-P-1 in the umu test. Gene expression was suppressed by 62% at less than 0.18 mumol/ml, the ID50 value being 0.08 mumol/ml. Compound 1 was also assayed with aflatoxin B1 (AfB1) and showed a suppressive effect.  相似文献

The in vitro activity of root-tuber-peel extract of Flemingia vestita, an indigenous plant consumed by the natives in Northeast India, was tested against helminth parasites. Live parasites (nematode: Ascaris suum from pigs, A. lumbricoides from humans, Ascaridia galli and Heterakis gallinarum from domestic fowl; cestode: Raillietina echinobothrida from domestic fowl; trematode: Paramphistomum sp. from cattle) were collected in 0.9 % physiological buffered saline (PBS) and maintained at 37 +/- 1 degrees C. In vitro treatment of the parasites with the crude extract (50 mg/ml) in PBS revealed complete immobilization of the trematode and cestode in about 43 and 20 min, respectively. However, the cuticle-covered nematodes did not show any change in physical activity and remained viable even after a long period of exposure to the extract. Exposure of R. echinobothrida to genistein (0.5 mg/ml), an active principle isolated from the root-tuber peel, caused spontaneous loss of movement (paralysis) in 4.5 h, which was slower than the time required for praziquantel, the reference flukicide and cestodicide. The treated parasites showed structural alteration in their tegumental architecture. This study suggests the vermifugal activity of this plant extract against trematodes and cestodes.  相似文献

Following bronchial provocation tests with inhalent allergens, late asthmatic reactions (LAR) frequently follow immediate asthma reactions (IAR). This study of atopic asthmatic children demonstrated that patients could show IARs to one allergen, ryegrass extract, but isolated LARs without any preceding immediate response when challenged with an unrelated allergen extract of D. pteronyssinus. The patients' degree of skin sensitivity to the concentration of extract inhaled appeared one factor which determined whether an isolated LAR or IAR followed allergen inhalation.  相似文献

A new furanoid diterpene, 15,16-epoxy-12-oxo-8(17), 13(16), 14-labdatrien-20,19-olide (1) was isolated from an ethanolic extract of Potamogeton nodosus. Its structure was elucidated by the usual spectroscopic methods, including 2D NMR techniques. Incubation of ampicillin with whole cells of Streptomyces sp. DRS-1 resulted in accumulation of four compounds different from ampicillin. One of them was isolated, purified and partially characterized. On the basis of spectroscopic characteristics, RF value and antibacterial activity the compound was identified as cephalexin.
