Exogenous fibrolytic enzymes have been shown to be a promising way to improve feed conversion efficiency (FCE). β-Mannanase is an important enzyme digesting the polysaccharide β-mannan in hemicellulose. Supplementation of diets with β-mannanase to improve FCE has been more extensively studied in nonruminants than in ruminants. The objective of this study was to investigate the effects of β-mannanase supplementation on nutrient digestibility, FCE, and nitrogen utilization in lactating Holstein dairy cows. Twelve post-peak-lactation multiparous Holstein cows producing 45.5 ± 6.6 kg/d of milk at 116 ± 19.0 d in milk were randomly allotted to 1 of 3 treatments in a 3 × 3 Latin square design with 3 periods of 18 d (15 d for adaptation plus 3 d for sample collection). All cows were fed the same basal diet and the 3 treatments differed only by the β-mannanase dose: 0% dry matter (DM; control), 0.1% of DM (low supplement, LS), and 0.2% of DM (high supplement, HS) supplemented to the basal diet. Supplementation of β-mannanase enzyme at the LS dose reduced dry matter intake (DMI) but did not affect milk yield or milk composition. Cows receiving LS produced 90 g more milk per kg of DMI compared with control cows. Somatic cell count (SCC) in milk was lower for cows fed the LS diet compared with cows fed control diets. Cows fed LS diet had lower DM, organic matter and crude protein digestibility compared with cows fed control diets. Starch, neutral detergent fiber, and acid detergent fiber digestibility were not affected by LS. Milk yield, DMI, SCC, and nutrient digestibility did not change for HS. Despite the reduced crude protein digestibility, reduced N intake led to similar fecal N excretions in LS cows and control cows (234 vs. 235 g/cow per day). Urinary N excretions remained similar between enzyme-fed and control cows (~190 g/cow per day), although the percentage of N intake partitioned to urinary N tended to be greater in LS than in control cows (31 vs. 27%). Cows fed LS significantly improved the percentage of apparently absorbed N partitioned to milk protein N (42 vs. 38%). When supplemented at 0.1% of dietary DM, β-mannanase can improve FCE and lower the SCC of dairy cows without affecting milk yield, milk composition, or total manure N excretions of dairy cows. 相似文献

Transformation of Arg to nitric oxide and ornithine (Arg-Orn) constitutes the main route of Arg metabolism in mammals. The primary objective of this work was to determine the effects of inhibiting the Arg-Orn pathway via Nω-hydroxy-nor-l-arginine (nor-NOHA) on health of lactating cows. Furthermore, we also explored the effect of Arg-Orn inhibition on the efficiency of nitrogen utilization to find support for previous research that showed the inhibition of Arg-Orn inhibited milk protein synthesis. Six healthy Chinese Holstein cows of similar body weight (550.0 ± 20 kg), parity (4.0 ± 0), body condition score (3.0 ± 0), milk yield (21.0 ± 1.0 kg), and days in milk (80 ± 2 d) were selected and randomly assigned to 3 treatments in a replicated 3 × 3 Latin square design with 22 d for each period (7 d for infusion and 15 d for washout). The treatments were (1) saline infusion (control); (2) infusion of 125 mg/L of nor-NOHA; and (3) infusion of 125 mg/L of nor-NOHA with 9.42 g/L of Arg. Dry matter intake, apparent digestibility of nutrients, urinary N, N in milk, and blood indices of metabolism and immune function were determined. Compared with the control, the infusion of nor-NOHA had no effect on the concentrations of cholesterol, high-density lipoproteins, IgA, IL-1β, tumor necrosis factor-α, and alanine transaminase. In addition, the dry matter intake, apparent digestibility of N, and the concentration of milk protein N in the Nor-NOHA did not differ from the control; however, the infusion of nor-NOHA and Arg resulted in greater concentrations of high-density lipoprotein, IgA, IL-1β, and tumor necrosis factor-α, and lower concentrations of cholesterol in serum compared with the control. Moreover, the addition of Arg to cows infused with nor-NOHA increased the concentration of nitrate (the indicator of nitric oxide) in serum and was associated with greater milk protein N production due to greater milk yield compared with those infused with nor-NOHA. Overall, the results indicated important roles of Arg in immunity and mammary N utilization, whereas a minor role of the Arg-Orn pathway in these physiologic processes was found. 相似文献

The objectives of this study were to induce hindgut and metabolic acidosis via abomasal infusion of corn starch and β-hydroxybutyrate (BHB), respectively, and to determine the effects of these physiological states in early-lactation dairy cows. In a 6 × 6 Latin square design, 6 rumen-fistulated Holstein-Friesian dairy cows (66 ± 18 d in milk) were subjected to 5 d of continuous abomasal infusion treatments followed by 2 d of rest. The abomasal infusion treatments followed a 3 × 2 factorial design, with 3 levels of corn starch and 2 levels of BHB. The infusions were water as control, 1.5 kg of corn starch/d, 3.0 kg of corn starch/d, 8.0 mol BHB/d, 1.5 kg of corn starch/d + 8.0 mol BHB/d, or 3.0 kg of corn starch/d + 8.0 mol BHB/d. A total mixed ration consisting of 35.0% grass silage, 37.4% corn silage, and 27.6% concentrate (on a dry matter basis) was fed at 90% of ad libitum intake of individual cows. The experiment was conducted in climate respiration chambers to facilitate determination of energy and N balance. Fecal pH decreased with each level of corn starch infused into the abomasum and was 6.49, 6.00, and 5.15 with 0.0, 1.5, and 3.0 kg of corn starch/d, respectively, suggesting that hindgut acidosis was induced with corn starch infusion. No systemic inflammatory response was observed and the permeability of the intestine or hindgut epithelium was not affected by the more acidic conditions. This induced hindgut acidosis was associated with decreased digestibility of nutrients, except for crude fat and NDF, which were not affected. Induced hindgut acidosis did not affect milk production and composition and energy balance, but increased milk N efficiency. Abomasal infusion of BHB resulted in a compensated metabolic acidosis, which was characterized by a clear disturbance of acid-base status (i.e., decreased blood total CO2, HCO3, and base excess, and a tendency for decreased urinary pH), whereas blood pH remained within a physiologically normal range. Abomasal infusion of BHB resulted in increased concentrations of BHB in milk and plasma, but both remained well below the critical threshold values for subclinical ketosis. Induced compensated metabolic acidosis, as a result of abomasally infused BHB, increased energy retained as body fat, did not affect milk production and composition or inflammatory response, but increased intestinal permeability. 相似文献

Several studies have identified beneficial effects of butyrate on rumen development and intestinal health in preruminants. These encouraging findings led to further investigations related to butyrate supplementation in the mature ruminant. However, the effects of elevated butyrate concentrations on rumen metabolism have not been investigated, and consequently the maximum tolerable dosage rate of butyrate has not been established. Therefore, the first objective of this work was to evaluate the effect of a short-term increase in rumen butyrate concentration on key metabolic indicators. The second objective was to evaluate the source of butyrate, either directly dosed in the rumen or indirectly supplied via lactose fermentation in the rumen. Jugular catheters were inserted into 4 ruminally fistulated Holstein cows in a 4 × 4 Latin square with 3-d periods. On d 1 of each period, 1 h after feeding, cows were ruminally dosed with 1 of 4 treatments: (1) 2 L of water (CON), (2) 3.5 g/kg of body weight (BW) of lactose (LAC), (3) 1 g/kg of BW of butyrate (1GB), or (4) 2 g/kg of BW of butyrate (2GB). Sodium butyrate was the source of butyrate, and NaCl was added to CON (1.34 g/kg of BW), LAC (1.34 g/kg of BW), and 1GB (0.67 g/kg of BW) to provide equal amounts of sodium as the 2GB treatment. Serial plasma and rumen fluid samples were collected during d 1 of each period. Rumen fluid pH was greater in cows given the 1GB and 2GB treatments compared with the cows given the LAC treatment. Cows administered the 1GB and 2GB treatments had greater rumen butyrate concentrations compared with LAC. Those cows also had greater plasma butyrate concentrations compared with cows given the LAC treatment. Plasma β-hydroxybutyrate was greater and insulin tended to be greater for butyrate treatments compared with LAC. No difference in insulin was found between the 1GB and 2GB treatments. Based on plasma and rumen metabolites, singly infusing 3.5 g/kg of BW of lactose into the rumen is not as effective at providing a source of butyrate as compared with singly infusing 1 or 2 g/kg of BW of butyrate into the rumen. Additionally, rumen pH, rumen butyrate, plasma β-hydroxybutyrate, glucose, and plasma butyrate were less affected in cows administered the 1GB treatment than in cows given the 2GB treatment. This finding suggests that singly dosing 1 g/kg of BW of butyrate could serve as the maximum tolerable concentration for future research. 相似文献

The objective of this study was to obtain information on variation between dairy cows in muscle and fat tissue mobilization around parturition and to study the association between protein and fat mobilization and serum β-hydroxybutyrate (BHBA) concentrations (hyperketonemia) in this period. Thirty-four cows kept under similar conditions at a university dairy farm (no experimental treatments) were monitored from 4 wk before until 8 wk after calving. Mobilization of muscle protein was investigated by analysis of plasma 3-methylhistidine concentrations (3-MH, analyzed by a recently developed HPLC tandem mass spectrometry method) and ultrasound measurements of longissimus muscle thickness. Mobilization of fat tissue was monitored by serum nonesterified fatty acid (NEFA) concentrations and ultrasound measurements of backfat thickness. Large variation was observed between cows in onset and duration of periparturient protein and fat mobilization. Plasma 3-MH concentrations and muscle thickness profiles indicated that protein mobilization started, on average, before parturition and continued until approximately wk 4 of lactation. Serum NEFA concentrations and backfat thickness profiles showed that fat mobilization occurred from parturition until the end of the study. Thus, muscle protein mobilization occurred in advance of fat mobilization in most cows from this study. We hypothesized that this might be due to a prepartum amino acid deficiency in the absence of negative energy balance. The incidence of hyperketonemia in this study was 16/34 = 47%. With the exception of 3 cows defined as having severe hyperketonemia, cows with lower 3-MH concentrations had higher serum BHBA concentrations. A possible explanation for this observation might be that higher mobilization of protein around calving might restrict ketone body production due to the higher availability of glucogenic precursors in the period of most severe negative energy balance and highest fat mobilization. The validity of this hypothesis needs to be confirmed, but data from this study indicate that further research on the role of protein mobilization in the etiology of hyperketonemia in dairy cows is needed. 相似文献

The objective of this meta-analysis was to evaluate the effect of blending canola meal (CM) with other protein sources on production responses in lactating dairy cows. To evaluate this effect, a data set was assembled containing 22 studies reporting at least 3 isonitrogenous dietary treatments (total of 74 treatment means). Each study needed to report 1 diet with CM <0.3 kg/d, 1 or more diets consisting of CM blended with another protein source, and 1 diet with CM as the main protein source in the protein supplement (>85%). The crude protein (CP) concentration of CM averaged 37.4 ± 3.09% (dry matter basis), and the predictor of interest was the intake of CP from CM, which averaged 0.46 ± 0.413 kg/d among studies. The maximal CP from CM ranged from 0.47 to 1.55 kg/d among studies. The quadratic relationship between CP from CM and responses in milk true protein concentration was significant, the maximum response (3.19%) being reached at 0.79 kg of CP from CM; the quadratic relationships were not significant for the other dependent variables. Responses in dry matter intake; yields of milk, energy-corrected milk, and milk true protein; and apparent N efficiency were related positively to CP from CM and negatively for responses in milk fat and milk urea N concentrations. Remembering that diets were isonitrogenous within studies, this indicates no nutritional benefit of blending CM with another protein source. Taken together, the results indicate that the whole-body N utilization efficiency by the dairy cow improved and that more dietary protein was used to synthesize milk protein when CM was used as the sole protein source in the protein supplement up to 1.55 kg/d. 相似文献

Whether supplemental dietary β-carotene affects periparturient cows and vitamins A and E in cows when dietary vitamin A is adequate remains uncertain. Our objective was to assess the effect of β-carotene supplementation during the close-up dry period in a herd with adequate status of vitamins A and E but low in β-carotene. The study was conducted on a large commercial dairy farm in Indiana during early summer of 2015. Ninety-four multiparous Holstein cows were assigned to either control (CON; n = 47) or β-carotene (BC; n = 47) treatments. When locked in headgates each morning, each cow received a topdress of β-carotene (Rovimix, 8 g/d; provided 800 mg of β-carotene) or carrier from 21 d before expected calving until calving. Blood samples were collected at 21 ± 1 d (mean ± standard deviation) before expected calving (before treatments began), 7 ± 1 d before calving, immediately following parturition, and 7 ± 1 d postpartum. Blood serum was analyzed for vitamins A and E, β-carotene, cholesterol, and other metabolites and enzymes. Data were analyzed using the MIXED procedure in SAS (SAS Institute Inc.). Cows had low β-carotene concentrations (0.85 μg/mL) in blood serum before treatments began. Compared with CON cows, BC cows had higher overall mean concentrations of β-carotene (2.87 μg/mL vs. 0.73 μg/mL) and retinol (165 vs. 143 ng/mL). Cows fed BC had lower α-tocopherol in serum than cows fed CON (2.26 vs. 2.46 µg/mL). Cows fed BC had lower peak milk than cows fed CON (50.9 vs. 55.3), but total lactation milk yield did not differ significantly. No effects of BC were observed on days to conception (100 d) or times bred (2.4). Treatments did not affect incidences of ketosis, retained placenta, displaced abomasum, off feed, lameness, footrot, mastitis, or metritis. In conclusion, in pregnant cows already receiving adequate vitamin A but with low serum β-carotene concentration, supplementation of β-carotene increased concentrations of β-carotene and vitamin A in blood serum, but did not affect production, reproduction, or health. 相似文献

Two trials were conducted to examine factors potentially influencing the measurement of blood β-hydroxybutyric acid (BHBA) in dairy cows. The objective of the first trial was to study effects of sampling time on BHBA concentration in continuously fed dairy cows. Furthermore, we determined test characteristics of a single BHBA measurement at a random time of the day to diagnose subclinical ketosis considering commonly used cut-points (1.2 and 1.4 mmol/L). Finally, we set out to evaluate if test characteristics could be enhanced by repeating measurements after different time intervals. During 4 herd visits, a total of 128 cows (8 to 28 d in milk) fed 10 times daily were screened at 0900 h and preselected by BHBA concentration. Blood samples were drawn from the tail vessels and BHBA concentrations were measured using an electronic BHBA meter (Precision Xceed, Abbott Diabetes Care Ltd., Witney, UK). Cows with BHBA concentrations ≥0.8 mmol/L at this time were enrolled in the trial (n = 92). Subsequent BHBA measurements took place every 3 h for a total of 8 measurements during 24 h. The effect of sampling time on BHBA concentrations was tested in a repeated-measures ANOVA repeating sampling time. Sampling time did not affect BHBA concentrations in continuously fed dairy cows. Defining the average daily BHBA concentration calculated from the 8 measurements as the gold standard, a single measurement at a random time of the day to diagnose subclinical ketosis had a sensitivity of 0.90 or 0.89 at the 2 BHBA cut-points (1.2 and 1.4 mmol/L). Specificity was 0.88 or 0.90 using the same cut-points. Repeating measurements after different time intervals improved test characteristics only slightly. In the second experiment, we compared BHBA concentrations of samples drawn from 3 different blood sampling locations (tail vessels, jugular vein, and mammary vein) of 116 lactating dairy cows. Concentrations of BHBA differed in samples from the 3 sampling locations. Mean BHBA concentration was 0.3 mmol/L lower when measured in the mammary vein compared with the jugular vein and 0.4 mmol/L lower in the mammary vein compared with the tail vessels. We conclude that to measure BHBA, blood samples of continuously fed dairy cows can be drawn at any time of the day. A single measurement provides very good test characteristics for on-farm conditions. Blood samples for BHBA measurement should be drawn from the jugular vein or tail vessels; the mammary vein should not be used for this purpose. 相似文献

The objective of this study was to extend a stable isotope-based assessment of AA absorption from rumen-degradable protein (RDP) sources to include determination of essential AA (EAA) availability from microbial protein (MCP). To demonstrate the technique, a study using a 2 × 2 factorial arrangement of treatments applied in a repeated 4 × 4 Latin square design was undertaken. Factors were high and low rumen-degradable protein and high and low starch. Twelve lactating cows were blocked into 3 groups according to days in milk and randomly assigned to the 4 treatment sequences. Each period was 14 d in length with 10 d of adaption followed by 4 d of ruminal infusions of 15N-labeled ammonium sulfate. On the last day of each period, a 13C-labeled AA mixture was infused into the jugular vein over a 6-h period to assess total AA entry. Rumen, blood, urine, and milk samples were collected during the infusions. Ruminal bacteria and blood samples were assessed for AA enrichment. Total plasma AA absorption rates were derived for 6 EAA from plasma 13C AA enrichment. Absorption of 6 EAA from MCP was calculated from total AA absorption based on 15N enrichment in blood and rumen bacteria. Essential AA absorption rates from total protein, MCP, and rumen-undegradable protein were derived with standard errors of the mean of 6, 14, and 14%, respectively. An average of 45% of absorbed EAA were from MCP, which varied among 6 EAA and was interactively affected by starch and RDP in diets. Microbial AA availability measured by isotope dilution method increased with the high RDP diets and was unaffected by starch level, except for Met, which decreased with high starch. Microbial protein outflow, estimated from urinary purine derivatives, increased with RDP and was not significantly affected by starch. This was consistent with measurements from the isotope dilution method. Total AA absorption rates measured from isotope dilution were similar to estimates from CNCPS (v. 6.55), but a lower proportion of absorbed AA was derived from MCP for the former method. Compared with the isotope and CNCPS estimates, the Fleming model underestimated microbial EAA and total EAA availability. An average of 58% of the absorbed EAA was converted into milk, which varied among individual AA and was interactively affected by starch and RDP in diets. The isotope dilution approach is advantageous because it provides estimates of EAA availability for individual EAA from rumen-undegradable protein and MCP directly with fewer errors of measurement than can be achieved with intestinal disappearance methods. 相似文献

Although there is evidence that ketosis negatively affects fertility, the effect of late and early ketosis on the reproductive performance of lactating cows has not been systematically investigated. The aim of this study was to evaluate the association between time and amplitude of elevated milk BHB (EMB) occurring within 42 d in milk (DIM) and subsequent reproductive performance of lactating Holstein cows. The dairy herd information data of 30,413 cows with 2 test-day milk BHB recordings during early lactation periods 1 and 2 (5–14 and 15–42 DIM, respectively) assessed as negative (<0.15 mmol/L), suspect (0.15–0.19 mmol/L), or positive (≥0.2 mmol/L) for EMB were used in this study. Based on the time and amplitude of milk BHB, cows were grouped into 7 groups: (1) healthy cows negative in both periods 1 and 2 were classified as NEG; (2) suspect in period 1 and negative in period 2: EARLY_SUSP; (3) suspect in period 1 and suspect/positive in period 2: EARLY_SUSP_Pro; (4) positive in period 1 and negative in period 2: EARLY_POS; (5) positive in period 1 and suspect/positive in period 2: EARLY_POS_Pro; (6) negative in period 1 and suspect in period 2: LATE_SUSP; and (7) negative in period 1 and positive in period 2: LATE_POS. The overall prevalence of EMB within 42 DIM was 27.4%, with the highest prevalence being EARLY_SUSP (10.49%). Cows in EARLY_POS and EARLY_POS_Pro, but not other EMB categories, had a longer interval from calving to first service compared with NEG cows. For the reproductive parameters, first service to conception interval, days open and calving interval, cows in all EMB groups except EARLY_SUSP had longer intervals compared with NEG cows. These data indicate that there is a negative association between EMB within 42 d and reproductive performance after the voluntary waiting period. The intriguing findings of this study are the unaltered reproductive performance of EARLY_SUSP cows, and the negative association between late EMB and reproductive performance. Hence, monitoring and prevention of ketosis during the first 6 wk of lactation is necessary to optimize reproductive performance of lactating dairy cows. 相似文献

Recent studies in mid- and late-lactation dairy cows showed that β-hydroxybutyrate (BHB) infusion had a considerable effect on glucose metabolism and immune response during intramammary lipopolysaccharide challenge. The objective of the present study was to infuse BHB during the dry period and after parturition to investigate the effects of elevated plasma BHB concentrations on metabolism and endocrine changes in transition dairy cows. The hypothesis tested was that regulation of glucose metabolism would change at different physiological stages and an additional elevation of BHB concentration would alter glucose concentration. Multiparous Holstein cows in wk ?2 (antepartum, a.p.; n = 6) and wk +2 (postpartum, p.p.; n = 8) relative to calving were infused (4 h from 0800 to 1200 h) with a BHB solution to increase plasma BHB concentration to 1.5 to 2.0 mmol/L (HyperB). The same period the next day without any infusion was considered the control period (CON). Blood samples were taken 1 h before the start of infusion as reference samples and every 30 min during the following 6 h (4 h of infusion and 2 h after infusion) in the HyperB and CON periods, and analyzed for glucose, BHB, insulin, and glucagon concentrations. During the steady state period (the latter 2 h of the 4-h infusion), plasma BHB concentration reached 1.87 ± 0.05 mmol/L (a.p.) and 1.93 ± 0.05 mmol/L (p.p.) in HyperB compared with 0.55 ± 0.06 mmol/L (a.p.) and 0.64 ± 0.04 mmol/L (p.p.) in CON, respectively. The 4-h average BHB infusion rate was 12.4 ± 1.0 and 13.3 ± 0.9 μmol/kg of BW per minute in wk ?2 and +2, respectively. Infusion of BHB caused a decrease of plasma glucose concentrations relative to preinfusion levels both before and after parturition, although basal glucose concentrations were different before and after calving. Infusion of BHB increased plasma insulin concentrations a.p. but not p.p., despite a higher basal insulin concentration before than after parturition. These findings show that effects of hyperketonemia on plasma glucose concentrations are similar before and after calving but that endocrine adaptation to hyperketonemia differs before and after parturition. We assume that BHB is a metabolic key regulator in early lactating dairy cows and may affect glucose concentration by further pathways such as gluconeogenesis and altered lipolysis. 相似文献

High energy intake in the dry period has reportedly had adverse effects on mobilization of body reserves, dry matter intake, and productivity of dairy cows. We investigated whether grass silage (GS) fed ad libitum (high energy intake, HEI; 141% of daily metabolizable energy requirements) in an 8-wk dry period affects metabolic adaptation—specifically, peripheral insulin resistance—compared with a total mixed ration consisting of GS, wheat straw, and rapeseed meal (55/40/5%; controlled energy intake, CEI; 108% of metabolizable energy/d) fed ad libitum. Multiparous Ayrshire dairy cows (n = 16) were used in a randomized complete block design until 8 wk after parturition. Commercial concentrates were fed 1 and 2 kg/d during the last 10 to 6 and 5 to 0 d before the expected calving date, respectively. Postpartum, a similar lactation diet with ad libitum access to GS and increasing concentrate allowance (maximum of 16 kg/d) was offered to all. The HEI group gained more body weight and had higher plasma insulin, glucose, and β-hydroxybutyrate concentrations than the CEI group prepartum. Postpartal plasma glucose tended to be higher and milk yield was greater from wk 5 onward for HEI compared with CEI cows. An intravenous glucose tolerance test (IVGTT) was performed at ?13 ± 5 d and 9 ± 1 d relative to calving. The HEI cows had greater insulin response to glucose load and smaller area under the response curve for glucose than CEI cows in prepartal IVGTT. Thus, compensatory insulin secretion adapted to changes in insulin sensitivity of the peripheral tissues, preserving glucose tolerance of HEI cows. Higher insulin levels were needed in HEI cows than in CEI cows to elicit a similar decrement of nonesterified fatty acid concentration in prepartal IVGTT, suggesting reduced inhibition of lipolysis by insulin in HEI cows before parturition. In conclusion, high energy intake of moderately digestible GS with low concentrate feeding in the close-up dry period did not have adverse effects on metabolic adaptation, insulin sensitivity, and body mobilization after parturition. Vitamin D exists in 2 forms that are important regarding vitamin D status and supply in cattle: vitamin D2 (D2) and vitamin D3 (D3). To become physiologically active, both D2 and D3 must undergo 25-hydroxylation in the liver. The resulting 25-hydroxyvitamin D2 [25(OH)D2] and 25-hydroxyvitamin D3 [25(OH)D3] are measured as indicators of the physiological vitamin D status of cattle. The study used 14 Danish Holstein cows housed without access to sunlight. The cows were orally administered 250 mg (1.0 × 107 IU) of D2 and D3 in a cross-over design with 2 treatment groups and 2 study periods, rendering 4 treatments when carryover effects were taken into account: D2 given first, D2 given last after D3, D3 given first, and D3 given last after D2. Two weeks elapsed between the treatment in the first study period and the treatment in the second study period. Blood samples were collected 0, 3, 6, 14, 17, 20, 23, 26, 40, 48, 70, 94, 166, and 214 h after providing the oral bolus of vitamin to the cows. Comparisons between plasma levels of the metabolites D2, D3, 25(OH)D2, and 25(OH)D3 over time were made by comparing areas under the plasma concentration curves. Oral administration of D3 increased plasma D3 (182.6 ± 17.1 ng/mL; mean ± SEM) and 25(OH)D3 (103.5 ± 10.0 ng/mL) more efficiently than oral administration of D2 increased plasma D2 (49.1 ± 32.6 ng/mL) and 25(OH)D2 (27.9 ± 2.1 ng/mL). The D3 given after an oral dose of D2 was less efficient for increasing plasma concentrations of 25(OH)D3 (61.2 ± 12.0 ng/mL) compared with D3 given without previous D2 administration (103.5 ± 10.0 ng/mL), whereas the plasma concentrations of D3 itself were the same when given first (182.6 ± 17.1 ng/mL) as when given after D2 (200.0 ± 123.9 ng/mL). The same occurred for plasma concentrations of D2 metabolites both if D2 was given first (49.1 ± 32.6 ng/mL) and after D3 (54.7 ± 7.7 ng/mL). In conclusion, D3 given after D2 is less efficient at increasing the plasma status of 25(OH)D3 than D3 given without previous D2 administration. 相似文献

Synthetic vitamin supplementation is not consistent with organic production, so it is important to investigate whether dairy cows can maintain their health and production without synthetic vitamins being added to their diet. In basic dairy cow diets, provitamin A (β-carotene) and vitamin E are mainly found in pasture and in grass and legume silages, but the concentrations are highly variable. This study compared the vitamin status and health of cows without synthetic vitamin supplementation (NSV group) with control cows (CON group) fed synthetic vitamins according to Swedish recommendations (600 IU of vitamin E and 80,000 IU of vitamin A per cow per day) to investigate whether dairy cows can fulfill their requirements of vitamins A and E without supplementation with synthetic vitamins. Vitamin concentrations in blood plasma and milk, health, fertility, milk yield, and milk composition were measured in Swedish Holstein cows (n = 28) during 2 complete lactations. All cows were fed a 100% organic diet containing grass-legume silage, cold-pressed rapeseed cake, peas, cereal grains, and minerals. Blood samples were collected from each cow 3 wk before expected calving, at calving, and 3 wk, 3 to 5 mo, and 7 to 9 mo after calving. Samples of colostrum were taken and milk samples were collected 4 d after calving and at the same time as the 3 blood samplings after calving. The only difference in vitamin status between groups was found in colostrum in yr 1, when CON cows tended to have a higher concentration of α-tocopherol, and their β-carotene concentration was higher compared with NSV cows. The NSV cows tended to have more cases of mastitis than CON cows in yr 2. Within the NSV group, fewer cows were healthy and more cases of mastitis were observed in yr 2 than in yr 1. The groups did not differ in production parameters. In conclusion, the vitamin status in blood and milk of the studied cows indicated that cows in organic dairy production can fulfill their requirements of vitamins A and E without any supplementation of synthetic vitamins, except at the time around calving, when the requirements are high. However, the impaired health of NSV cows in yr 2 may indicate a long-term negative health effect in cows fed no synthetic vitamins. 相似文献

The accumulation of urine and feces can be responsible for many cow and environmental problems. Despite this, little is known about the factors affecting defecation and urination. In the first experiment, the occurrence of defecation and urination behaviors of 48 lactating Holstein cows was observed [days in milk (DIM) = 144.7 ± 38.0 d, body weight (BW) = 667.1 ± 72.0 kg, parity = 2.8 ± 2.3] in freestalls over 48 h. In the second experiment, defecation and urination by 29 lactating Holstein dairy cows were observed (DIM = 62 ± 22.1 d, BW = 590 ± 70.0 kg, parity = 2 ± 1.3) in another freestall barn over a period of 5 d and related to cow activity and feeding behavior. In both experiments, based on total occurrence of eliminative behaviors, cows mainly defecated (experiment 1: 33.4 ± 2.0%; experiment 2: 42.3 ± 3.1%) and urinated (experiment 1: 28.2 ± 2.5%; experiment 2: 42.7 ± 4.0%) in the feed alley and while occupying a stall (defecation: experiment 1: 28.5 ± 1.0%; experiment 2: 26.2 ± 3.0%; urination: experiment 1: 42.2 ± 1.5%; experiment 2: 39.9 ± 3.8%). Occupying a stall included lying, standing in the stall, or standing with 2 feet in the stall and 2 feet in the alley. In both experiments, differences were found between cows in frequency of defecation (experiment 1: 9.8 ± 4.2/d, range = 3 to 20; experiment 2: 15.4 ± 4.3/d, range = 6 to 36) and in frequency of urination (experiment 1: 7.0 ± 3.1/d, range = 2 to 18; experiment 2: 9.3 ± 2.8/d, range = 3 to 19). Large differences between cows were observed in the frequency of defecation and urination, but these were not correlated with parity, milk production, BW, DIM, or dry matter intake. 相似文献

β-Carotene, a pro-vitamin A carotenoid, acts as an antioxidant, able to scavenge free radicals to prevent oxidative damage. It has also been shown to increase rumen microbial production in vitro. When supplemented prepartum, β-carotene increased colostral fat and protein concentrations. The objective of this experiment was to evaluate the effects of supplementing 700 mg/d β-carotene (BC) daily for 4 wk prepartum on cow performance, colostrum, and performance in subsequent calves. Eighteen multiparous Holstein cows housed in a tiestall barn were blocked by expected calving date and randomly assigned to treatments at 4 wk prepartum. Blood samples were collected 3 times a week for analysis of nonesterified fatty acids, ketones, β-carotene, and IgG. Urine samples were also collected 3 times a week for analysis of creatinine and purine derivatives. Colostrum was collected within 90 min after parturition. Calves were removed from their dams before suckling, weighed within 30 min of birth, and received 4 L of maternal colostrum. Blood samples were collected from calves before colostrum administration (0 h) and at 24 h via jugular venipuncture for analysis of IgG concentration and apparent efficiency of IgG absorption. The 18 calves born were blocked based on treatments of dams. All calves were fed 449 g/d dry matter of milk replacer (20% crude protein, 20% fat) and an 18% crude protein textured starter and water ad libitum at 2 d of age until weaning at 42 d. There were no differences in any blood parameters of cows during the prepartum period. Supplemental BC increased the solids content of colostrum compared with control (22.89% control; 27.75% BC). Calves born from control fed cows had greater efficiency of IgG absorption than those born from BC supplemented cows (52.16% control; 39.50%, BC). Calves born from BC fed cows had greater feed efficiency (average daily gain/dry matter intake) compared with those born from control supplemented cows (0.33 for control; 0.44 for BC). These data indicate that although supplementing β-carotene to cows in the prepartum period negatively affects apparent efficiency of IgG absorption, it improved feed efficiency in calves. 相似文献

The effectiveness of using serum vitamin concentrations as biomarkers to predict diseases in dairy cows during the periparturient period is not well known. The objective of this study was to evaluate the association between serum β-carotene, retinol, and α-tocopherol concentrations and periparturient cow diseases in commercial dairies. We measured serum concentrations of these vitamin-active compounds at dry-off and during close-up (approximately 3 wk before calving) and early lactation (approximately 7 d post-calving), and we examined their association with clinical diseases in the first 30 d in milk. Diseases were diagnosed by trained personnel and recorded using database software. Blood samples were taken from 353 cows from 5 different farms over a 3-yr period. Blood samples were analyzed for β-carotene, retinol, α-tocopherol, and cholesterol. We built separate mixed logistic regression models for each disease outcome: hyperketonuria, lameness, mastitis, uterine diseases (retained placenta or metritis), and an aggregate outcome. For the aggregate outcome, a cow was considered positive if she had one or more of the following: hyperketonuria, lameness, mastitis, uterine disease, pneumonia, milk fever, or displaced abomasum. Concentrations of all 3 fat-soluble vitamins decreased significantly in early lactation relative to the 2 prepartum sampling times. Serum retinol concentrations at close-up and early lactation were negatively associated with odds of developing postpartum hyperketonuria. At early lactation, cows with uterine disease had lower serum retinol concentrations than cows without uterine disease. Similarly, lower serum retinol concentrations were associated with greater odds of having any one disease in the aggregate outcome. First-test 305-d mature-equivalent milk yield was positively correlated with increased serum α-tocopherol and negatively correlated with β-carotene concentrations. This study demonstrates the potential for serum β-carotene, retinol, and α-tocopherol to serve as biomarkers for disease risk. 相似文献

Luteolysis is a key event in Ovsynch programs of lactating dairy cows. Studies indicate that as many as 20% of cows treated with a Presynch/Ovsynch program have delayed or incomplete luteolysis using dinoprost tromethamine. Cows must have complete luteolysis to have a chance to become pregnant. Dinoprost tromethamine has a short half-life of approximately 7 to 8 min. Cloprostenol sodium is more resistant to endogenous metabolism and is maintained in circulation for a longer time (half-life = 3 h). The objective was to determine if cloprostenol sodium could increase the percentage of cows with complete luteolysis and subsequent pregnancy per artificial insemination (P/AI) in lactating dairy cows compared with dinoprost tromethamine when administered within a presynchronization plus Ovsynch program for first artificial insemination (n = 652) and an Ovsynch resynchronization program for second or later AI (second+; n = 394). Blood samples were collected daily for 5 d beginning at the PGF2α of Ovsynch in a subset of cows (n = 680) for first and second+ AI to measure circulating concentrations of progesterone (P4) and estradiol (E2). Complete luteolysis was defined as cows with functional corpus luteum (CL) at time of treatment and serum concentrations of P4 <0.5 ng/mL at 56, 72, and 96 h after treatment. Percentage of cows with functional CL that had complete luteolysis after treatment was not greater for cloprostenol sodium compared with dinoprost tromethamine in first (79 vs. 80%, respectively) or second+ AI (70 vs. 72%, respectively). In addition, mean serum concentrations of P4 were not less for cows treated with cloprostenol sodium following treatment. Pregnancy per AI of cows treated with cloprostenol sodium tended to be greater than dinoprost tromethamine for first (40 vs. 35%; respectively) but not second+ AI (23 vs. 21%, respectively). Cows with greater serum P4 concentrations at time of PGF2α of Ovsynch had a greater probability of undergoing complete luteolysis after PGF2α of Ovsynch and pregnancy at 39 d after timed AI (i.e., 50% pregnant at 8 vs. 28% pregnant at 4 ng/mL P4). Serum concentrations of E2 at 56 h after PGF2α of Ovsynch were a positive predictor of pregnancy at 39 d after timed AI. In summary, cloprostenol sodium tended to improve P/AI. Cows with greater serum concentrations of P4 at time of PGF2α of Ovsynch had a greater chance of luteolysis and pregnancy. 相似文献

The objective of this study was to evaluate the ability of milk infrared spectra to predict blood β-hydroxybutyrate (BHB) concentration for use as a management tool for cow metabolic health on pasture-grazed dairy farms and for large-scale phenotyping for genetic evaluation purposes. The study involved 542 cows (Holstein-Friesian and Holstein-Friesian × Jersey crossbreds), from 2 farms located in the Waikato and Taranaki regions of New Zealand that operated under a seasonal-calving, pasture-based dairy system. Milk infrared spectra were collected once a week during the first 5 wk of lactation. A blood “prick” sample was taken from the ventral labial vein of each cow 3 times a week for the first 5 wk of lactation. The content of BHB in blood was measured immediately using a handheld device. After outlier elimination, 1,910 spectra records and corresponding BHB measures were used for prediction model development. Partial least square regression and partial least squares discriminant analysis were used to develop prediction models for quantitative determination of blood BHB content and for identifying cows with hyperketonemia (HYK). Both quantitative and discriminant predictions were developed using the phenotypes and infrared spectra from two-thirds of the cows (randomly assigned to the calibration set) and tested using the remaining one-third (validation set). A moderate accuracy was obtained for prediction of blood BHB. The coefficient of determination (R2) of the prediction model in calibration was 0.56, with a root mean squared error of prediction of 0.28 mmol/L and a ratio of performance to deviation, calculated as the ratio of the standard deviation of the partial least squares model calibration set to the standard error of prediction, of 1.50. In the validation set, the R2 was 0.50, with root mean squared error of prediction values of 0.32 mmol/L, which resulted in a ratio of performance to deviation of 1.39. When the reference test for HYK was defined as blood concentration of BHB ≥1.2 mmol/L, discriminant models indicated that milk infrared spectra correctly classified 76% of the HYK-positive cows and 82% of the HYK-negative cows. The quantitative models were not able to provide accurate estimates, but they could differentiate between high and low BHB concentrations. Furthermore, the discriminant models allowed the classification of cows with reasonable accuracy. This study indicates that the prediction of blood BHB content or occurrence of HYK from milk spectra is possible with moderate accuracy in pasture-grazed cows and could be used during routine milk testing. Applicability of infrared spectroscopy is not likely suited for obtaining accurate BHB measurements at an individual cow level, but discriminant models might be used in the future as herd-level management tools for classification of cows that are at risk of HYK, whereas quantitative models might provide large-scale phenotypes to be used as an indicator trait for breeding cows with improved metabolic health. 相似文献

The objective of this meta-analysis was to assess the efficacy of the treatment of bovine metritis with common antibiotic and nonantibiotic treatment options. Acute puerperal metritis, a systemic and potentially painful illness with rectal temperature >39.5°C and signs of toxemia due to an infection of the uterus, occurs within 21 d after parturition. Because of the infectious nature, antibiotics are considered beneficial for the treatment of acute puerperal metritis. Each use of an antimicrobial drug, however, is associated with selective pressure for eventual emergence of resistant bacteria. The 23 trials evaluated in the course of a previously conducted systematic review were the basis for meta-analytic investigations. Selected trials were screened regarding their eligibility for the following investigations: (1) comparison of different antibiotic treatments with respect to metritis prevalence at time of re-examination, (2) efficacy of ceftiofur treatment with respect to metritis prevalence at time of re-examination, (3) comparison of efficacy of antibiotic versus nonantibiotic drugs with respect to metritis prevalence at time of re-examination, and (4) equivalence assessment of treatment effects on reproductive performance measures. Where at least 3 trials had investigated the same outcome variable and met the inclusion criteria (inclusion of a control or reference group diagnosed with metritis; reporting means and standard deviation in case of continuous data), meta-analytic investigations were carried out. Due to a shortage of comparable studies, we could not conduct investigations (1) and (3). Ceftiofur treatment of 828 metritic cows was associated with a decrease in the prevalence of metritis following treatment in comparison to 804 untreated cows. In conclusion, meta-analytic investigations uncovered a need for more high-quality studies. Furthermore, a positive effect of the most commonly used antibiotic drug, ceftiofur, for the treatment of bovine metritis could be shown. A comparison with other antibiotic or nonantibiotic treatment options could not be made. 相似文献
