目的检测人初乳、集中饲养的新西兰进口荷斯坦乳牛的初乳、常乳和血液中胰岛素样生长因子-1(IGF-1)和神经生长因子(NGF)的含量,比较人初乳和牛初乳、牛初乳和牛常乳以及牛乳和同期采集的血液中IGF-1、NGF的含量。方法采用放射免疫试剂盒测定人初乳、牛初乳、牛常乳和血中IGF-1、NGF的含量,使用SPSS 13.0进行统计分析。结果本研究中测定的人初乳中IGF-1的含量是26.91μg/L,荷斯坦乳牛的初乳、常乳和血清中IGF-1的含量分别是38.40、20.14和37.35μg/L。人初乳中NGF的含量是300.47 ng/L,荷斯坦乳牛的初乳、常乳和血清中NGF的含量分别是69.82、110.37和9.63 ng/L。经统计学分析发现牛初乳和人初乳中IGF-1的含量差异没有显著性,牛初乳中IGF-1的含量高于牛常乳(P<0.05),牛乳中IGF-1的含量与同期血液中IGF-1的含量差异没有显著性。人初乳中NGF的含量高于牛初乳(P<0.05),牛初乳和牛常乳中NGF的含量差异没有显著性,同期牛血中NGF的含量低于牛乳中NGF的含量(P<0.05)。结论本研究发现牛初乳与人初乳IGF-1含量无明显差别,...  相似文献

Colostrum composition and management were surveyed via sample and data collection from 55 dairy farms in Pennsylvania. Colostrum samples were analyzed for fat, protein, lactose, total solids, ash, Ig, lactoferrin, water- and fat-soluble vitamins, and minerals. Mean percentages of fat, protein, and lactose in colostrum were 6.7, 14.9, and 2.5, respectively. Concentrations of IgG1, IgG2, IgA, IgM, and lactoferrin were 35.0, 6.0, 1.7, 4.3, and 0.8 mg/mL, respectively. Mean concentrations of fat-soluble vitamins, including retinol, tocopherol, and β-carotene, were 4.9, 2.9, and 0.7 μg/g, respectively. Mean concentrations of water-soluble vitamins were 0.34, 0.90, 4.55, 0.60, 0.15, 0.21, and 0.04 μg/mL for niacin, thiamine, riboflavin, vitamin B12, pyridoxal, pyridoxamine, and pyridoxine, respectively. Mean concentrations (mg/kg) of selected minerals in colostrum were also determined (Ca 4,716; P 4,452; Mg 733; Na 1,058; K 2,845; Zn 38; Fe 5.3; Cu 0.3; S 2,595; and Mn 0.1). The findings of this study revealed that the mean concentrations of most nutrients in colostrum have increased when compared with values previously reported. Results also showed that management practices have improved over time, particularly with regard to colostrum storage and feeding. Additionally, we observed that herd size influenced colostrum management and quality. It can be inferred, based on these findings, that although improvements have been made with regard to colostrum management and quality, there is still a need to educate producers on issues related to storage and timely feeding of colostrum to increase passive transfer and decrease the rate of calf morbidity and mortality.  相似文献

Several factors have been reported to influence colostrum quality (immunoglobulin concentration). To date, knowledge of the influence of climatic factors in association with other potential influencing factors on colostrum quality is scarce. Associated influential factors are parity, body condition score, length of dry period, ration fed ante partum (AP), β-hydroxybutyrate postpartum (PP), milk yield, milk fat and protein, as well as somatic cell counts from previous and current lactation. The objective of the present study was to examine the effect of barn climate and the aforementioned factors on colostrum quality. Data were collected from 1,381 multiparous Holstein Friesian cows kept on one dairy farm over a period of one year (August 2014 to August 2015). Colostrum was harvested on farm within 1 h PP. The quantity and quality of first colostrum (estimated by Brix refractometry) were recorded for each cow. Additional data recorded were parity, body condition score at drying off, length of dry period, ration fed AP, milk yield data from previous and current lactation, milk somatic cell counts, and β-hydroxybutyrate PP. During the study period, temperature and humidity were recorded in the barn every hour, and temperature-humidity-index (THI) was calculated. Linear regression was performed with colostrum quality as the dependent variable. In the final model, colostrum quantity (L), length of dry period, parity, and climatic factors (specifically, median humidity in the 3rd week AP and hours with THI ≥72 in the last 14 and 21 d AP, respectively) were significant. Colostrum quality improved with parity and length of dry period and decreased with colostrum quantity, humidity, and hours with a THI ≥72. A classification and regression tree analysis revealed that colostrum quantity was the most important factor in this model [normalized importance (NI) 100%]. Parity (NI 42.7%), length of dry period (NI 37.1%), and climatic factors (NI 0.4 to 1.9%) followed with decreasing importance. These results indicate that the most important factors for colostrum quality (i.e., colostrum quantity and parity) may not be influenced by management. The 2 factors that can be influenced by management [i.e., length of dry period and THI (e.g., by cooling)], were quantitatively of minor importance compared with the other 2 factors. Further studies are necessary to determine whether changing these factors can improve colostrum quality significantly.  相似文献

目的:研究牛初乳中胰岛素生长因子-I(IGF-I)对大鼠受损肠黏膜的修复作用。方法:应用氨甲喋呤制备大鼠小肠炎模型,实验设正常对照组,MTX模型对照组(20μg/kg),牛初乳IGF-I提取物修复组(75μg/kg),IGF-I修复组(75μg/kg)。后2组每天灌胃给药1次,给药时间从造模后第2d开始至实验结束,共5d。第6d检测大鼠血浆二氨氧化酶(DAO)活性、血浆D-乳酸水平,光镜下观察小肠组织病理形态学改变。结果:与对照组相比,模型组大鼠血浆DAO活性和血浆D-乳酸水平明显升高(P<0.05),小肠损伤程度明显高于对照组(P<0.05),牛初乳中IGF-I提取物组和IGF-I组可降低血浆DAO活性及D-乳酸水平(P<0.05),减轻小肠损伤程度(P<0.05)。结论:牛初乳中IGF-I提取物能够明显减轻大鼠肠黏膜的损伤,同时可减轻炎细胞的浸润,对实验性大鼠的小肠炎具有修复作用。   相似文献

To determine the effects of the multifunctional iron-binding glycoprotein, lactoferrin (LF) and related compounds on the growth of leukemic cells, human myeloid leukemic cells (HL-60) were exposed to bovine lactoferrin (bLF) and proteolytic hydrolysates of bLF. Pepsin hydrolysates of bLF showed a greater growth suppressive effect than tryptic hydrolysates or mature bLF. Four peptides with proliferation inhibition activity were purified from pepsin hydrolysates by ion-exchange chromatography, reverse-phase HPLC, and gel-filtration. All peptides were from the N-terminal end, in a region where lactoferricin B (Lfcin B), an antibacterial peptide, is located. Among the four peptides, peptide 1 (pep1) was found to exhibit highest activity and corresponded to residues 17 to 38 of bLF, with a molecular weight of 2753.88. The IC50 value of this peptide was 6.3 micrograms/ml. Three other peptides were less active and corresponded to sequences 1 to 16 and 45 to 48, linked by disulfide-bridge (pep2, molecular mass of 2430.13), 1 to 15 and 45 to 46 linked by disulfide bridge (pep3, molecular mass of 2017,92) and from residues 1 to 13 (pep4, molecular mass of 1558.73). Cell proliferation inhibition activity of the peptides was thought to be due to induction of apoptosis, which was evaluated by DNA ladder formation, DNA fragmentation, enhanced expression of phosphatidyl serine, and morphological changes. The IC50 values of the three peptides were confirmed using synthetic peptides and were consistent with those of purified peptides.  相似文献

Commercially available colostrum replacers (CR) are commonly used when maternal colostrum (MC) is unavailable, for managerial convenience, to ensure quality consistency at first feeding, or in disease control and eradication programs. The objective of this study was to determine the efficacy of feeding First Day Formula (Accelerated Genetics, Baraboo, WI) CR versus pooled MC on immunological status, growth, and health of preweaned dairy calves. A total of 1,220 Jersey and Jersey × Holstein calves born on a California Central Valley dairy farm were assigned after birth to receive either CR or MC following a systematic allocation procedure. Calves assigned to MC were tube fed 2.8 L of MC, and calves assigned to CR were tube fed a total of 500 g of CR (150 g of immunoglobulin G; IgG) mixed into 1.9 L of water at 1 h ± 5 min after the calf was born. A subset of calves was selected for passive transfer (n = 592) and growth (n = 268) analyses. Although both coliform count and total bacteria count were low for MC and CR fed to calves during the study, the predicted probability of calves receiving contaminated liquid feed (coliform count >10,000 cfu/mL) at first feeding was reduced for calves fed CR (1.5%) compared with calves fed MC (6.1%). The mean blood concentration of IgG was lower for calves fed CR than for calves fed MC (19.6 vs. 23.4 mg/mL). However, the apparent efficiency of absorption of IgG did not differ between treatments (34.4 and 35.9% for CR and MC, respectively). Total proteins were lower in calves fed CR compared with MC at 24 h (5.16 vs. 5.84 g/dL, respectively). Calves fed CR were 1.5 kg lighter at weaning and gained 0.03 kg less per day (0.30 vs. 0.33 kg/d, respectively) than calves fed MC before weaning. Height at weaning did not differ between the 2 treatment groups. Calves fed CR tended to have a higher predicted probability of not being treated for diarrhea than calves fed MC (0.142 vs. 0.110, respectively). However, when the disease was present, CR had a higher number of treatment days compared with MC (11.6 vs. 10.8 d, respectively). The hazard ratio of dying did not differ between MC and CR; however, CR calves had a numerically higher risk (hazard ratio = 1.347) of dying compared with calves that received MC. In conclusion, IgG absorption and serum concentration of calves were adequate when calves were fed either CR or MC. The CR-fed calves had a lower probability of receiving contaminated liquid feed and performed similar in terms of health compared with calves receiving high-quality MC, although they were slightly lighter at weaning. Therefore, the CR evaluated in this study is a valid alternative to high-quality (>50 mg of IgG/mL) MC.  相似文献

Mycoplasma bovis is an important cause of mastitis in dairy cattle, and pneumonia, arthritis, and otitis in calves. Milk and colostrum are considered important sources of infection for calves. knowledge on the effect of on-farm freezing (?18°C) and thawing methods on the recovery of M. bovis from colostrum samples is missing. In this study, 2 separate experiments were performed. The first experiment consisted of a longitudinal study examining the survival [as measured by log(10) reduction] of 2 M. bovis strains in frozen colostrum over 14 wk. The second experiment examined the effect of different thawing temperatures (45 and 20°C), thawing frequencies (once or twice), and initial colostrum titer (104 or 106 cfu/mL) on M. bovis survival. A single freeze-thaw cycle led to an approximate 1 log reduction of M. bovis titer, independent of the thawing temperature. Freezing for 14 wk did not significantly further reduce the titer of bacteria compared with freezing for 2 wk. A second freeze-thaw cycle further reduced the M. bovis count by approximately 0.5 log compared with a single freeze-thaw cycle. Thawing temperature and initial bacterial concentration did not significantly affect M. bovis reduction. In conclusion, storage of colostrum samples in the freezer at ?18°C during epidemiological studies, herd monitoring, or test and cull programs will probably have little influence on qualitative bacteriological test results for M. bovis. The epidemiological or clinical relevance of an approximate 1 log reduction of M. bovis in colostrum is currently unclear.  相似文献

A study was conducted to identify the optimal temperature and time at which heat treatment of bovine colostrum would least change viscosity and IgG concentrations yet reduce bacterial count. First-milking colostrum with >50 g of immunoglobulins/L (measured by colostrometer) was collected from 30 Holstein cows. Aliquots of colostrum were heated for 0, 30, 60, or 90 min at 57, 60, or 63°C in a water bath. Samples were examined for viscosity, IgG1, and IgG2 concentrations, standard plate count, coagulase-negative staphylococci, environmental streptococci, coliform, gram-negative noncoliform, Streptococcus agalactiae, and Staphylococcus aureus counts. All heat treatments reduced counts of all bacteria groups measured compared with untreated colostrum samples. Heat treatment at ≥60°C denatured IgG1 compared with untreated colostrum; however, colostral IgG2 levels were not reduced when temperature was held at 60°C for <60 min. Viscosity was not affected when temperature was held at 60°C for <60 min. In this study, heat treatment of bovine colostrum at 60°C for 30 or 60 min reduced bacterial count, slightly reduced IgG concentration, and did not affect viscosity.  相似文献

通过添加稳定剂提高牛初乳的热稳定性并将其加工成酸奶。牛初乳中按0.5 g/100g添加Na2HPO4,65℃加热,记录凝固时间及评价杀菌效果;确定葡萄糖和蔗糖的添加比例,将杀菌初乳制成酸奶,进行乳酸菌和杂菌计数。结果表明,添加Na2HPO4的牛初乳加热40 min才开始凝固,而未添加组加热6 min完全凝固。65℃加热15 min,牛初乳菌落总数和大肠菌群均合格;当葡萄糖添加量为3 g/100g,蔗糖添加量为4 g/100g时,牛初乳酸奶口感最佳,乳酸菌数为1.26×106 cfu/mL。因此按0.5 g/100g添加Na2HPO4可以显著提高牛初乳的热稳定性,加工制成的初乳酸奶兼具了牛初乳和酸乳的双重功效。  相似文献

利用牛初乳中的免疫球蛋白活性因子,开发具有独特生理功能的乳制品成为牛初乳利用的热点。本研究利用十二烷基硫酸钠-聚丙烯酰胺电泳法和酶联免疫吸附法对市售的牛初孔制品的质量进行了分析与评价。结果表明,目前牛初乳制品的蛋白含量与组成有很大的差异,并且活性免疫球蛋白含量较低。目前牛初乳制品存在的问题亟待解决,市场亟待规范。  相似文献

The objective of this study was to characterize absorption of colostrum replacer (CR) or supplement (CS) containing fractions of bovine plasma. Immunoglobulin concentrate (IGC) was prepared from bovine blood to a final purity of approximately 90% IgG. Blood was also processed to produce a fraction containing elevated concentrations of IGF-I and TGF-beta (GF). Both IGC and GF were spray-dried and blended with other ingredients to produce CR (30% IgG) or CS (15% IgG) containing 0 or 5% GF. Holstein bull calves were assigned to one of five treatments: 1) Pooled colostrum (MC)--1.9 L of pooled colostrum at 1 and 8 h of age to provide over 150 g of IgG; 2) Low supplement (LC)--1.9 L of CS at 1 and 8 h of age to provide 150 g of IgG; 3) Low supplement + GF (LG)--1.9 L of a CS with GF at 1 and 8 h of age to provide 150 g of IgG; 4) High supplement (HC)--1.9 L of CR at 1 h of age to provide 150 g of IgG and 1.9 L of a commercial milk replacer (MR) at 8 h of age; and 5) High supplement + GF (HG)--1.9 L of a CR with GF at 1 h of age to provide 150 g of IgG and 1.9 L of MR at 8 h of age. Apparent efficiency of IgG absorption was higher for calves fed HC and HG compared with those fed LC and LG and was lower for calves fed LG and HG compared with those fed LC and HC. IgG concentrations at 24 h were highest in calves fed MC compared with other calves and were higher in calves fed HC and HG compared to LC and LG. Calves fed LG and HG had lower IgG concentrations at 24 h of age compared with those fed LC and HC. These results indicate that 150 g of IgG provided in one dose soon after birth is superior to 150 g of IgG fed in two doses 7 h apart. Also, addition of a plasma fraction containing elevated concentrations of IGF-I and TGF-beta decreased IgG absorption in the neonatal calf.  相似文献

Our objective was to determine if immunoglobulins play a role in the gravity separation (rising to the top) of somatic cells (SC) in skim milk. Other researchers have shown that gravity separation of milk fat globules is enhanced by IgM. Our recent research found that bacteria and SC gravity separate in both raw whole and skim milk and that heating milk to >76.9°C for 25 s stopped gravity separation of milk fat, SC, and bacteria. Bovine colostrum is a good natural source of immunoglobulins. An experiment was designed where skim milk was heated at high temperatures (76°C for 7 min) to stop the gravity separation of SC and then colostrum was added back to try to restore the gravity separation of SC in increments to achieve 0, 0.4, 0.8, 2.0, and 4.0 g/L of added immunoglobulins. The milk was allowed to gravity separate for 22 h at 4°C. The heat treatment of skim milk was sufficient to stop the gravity separation of SC. The treatment of 4.0 g/L of added immunoglobulins was successful in restoring the gravity separation of SC as compared with raw skim milk. Preliminary spore data on the third replicate suggested that bacterial spores gravity separate the same way as the SC in heated skim milk and heated skim milk with 4.0 g/L of added immunoglobulins. Strong evidence exists that immunoglobulins are at least one of the factors necessary for the gravity separation of SC and bacterial spores. It is uncertain at this time whether SC are a necessary component for gravity separation of fat, bacteria, and spores to occur. Further research is needed to determine separately the role of immunoglobulins and SC in gravity separation of bacteria and spores. Understanding the mechanism of gravity separation may allow the development of a continuous flow technology to remove SC, bacteria, and spores from milk.  相似文献

The objectives were to describe the effect of on-farm commercial batch pasteurization on immunoglobulin (IgG) concentrations and the fluid and feeding characteristics of colostrum and to compare serum IgG concentrations in calves fed fresh versus pasteurized colostrum. Newborn calves (123) were systematically allocated to dietary treatments of either fresh or pasteurized colostrum at both the first and second colostrum feedings. The IgG concentrations were measured for batches of colostrum fed fresh and in pre and postpasteurized samples for batches of colostrum fed after being pasteurized and in calf serum. Pasteurization reduced colostrum IgG concentration, with the percentage reduction averaging 58.5 and 23.6% for 95-L and 57-L batches, respectively. Pasteurizing high quality colostrum in 57-L (vs. 95-L) batches resulted in higher IgG concentrations in the end product. Pasteurization of 57-L batches produced colostrum of normal or only mildly thickened consistency that could be fed to calves. Serum IgG concentrations were higher for calves fed fresh colostrum and for calves with a shorter time interval (< or = 6 h) between first and second colostrum feedings. After controlling for the time interval between feedings, serum IgG concentrations were significantly higher for 40 calves fed unpasteurized (19.1 mg/ml) vs. 55 calves fed pasteurized colostrum (9.7 mg/ml) for calves fed 2 L at first feeding. By contrast, there was no difference in serum IgG concentrations between 8 calves fed unpasteurized (16.1 mg/ml) and 20 calves fed pasteurized colostrum (13.5 mg/ml) after calves were fed 4 L at the first feeding. While the latter results suggest that pasteurizing colostrum may work for producers with excellent colostrum management, these results are preliminary and should be interpreted with caution, given the fewer number of calves and batches of colostrum involved with this second comparison.  相似文献
