An attempt was made to enzymatically extract the protein fraction (PF) from sunflower seeds and investigate its dietary effect on the growth, plasma and tissue lipid profiles, plasma protein content, erythrocyte membrane lipid profile and organ weights of rats. Extraction of dehulled sunflower seeds in an aqueous medium in the presence of the enzyme pectinase at 50 ± 1 °C and pH 4.5 yielded a protein fraction with low residual chlorogenic acid and fibre contents. Rats fed the sunflower seed protein fraction showed a similar body weight gain and food efficiency ratio as rats fed casein. However, the different dietary proteins had different effects on plasma total cholesterol and triglyceride contents. The rats fed the sunflower seed protein fraction showed a significant decrease in plasma cholesterol (p < 0.02) and triglyceride (p < 0.02) concentrations compared to the casein‐fed rats. The PF‐fed rats also exhibited a significant difference in erythrocyte membrane phospholipids and phosphatidylcholine/phosphatidylethanolamine ratio. Rats fed the different dietary proteins did not show much variation in plasma protein content, liver and brain lipids and organ weights. These results demonstrate the hypolipidemic action of the sunflower seed protein fraction and that it can be considered a suitable edible protein like casein. © 2000 Society of Chemical Industry

Five castrated male Iberian pigs (100 ± 2 kg body weight) fitted with T‐shaped cannulas at the terminal ileum were used to determine ileal digestibility of legume seed meals. The diets were based on defatted soybean, lupin or chickpea seed meals and contained similar levels of digestible energy (14.2–15.1 kJ g?1) and protein (107 g kg?1). Protein‐free and a hydrolysed casein diets were used to study endogenous ileal amino acid flows. Chromium oxide (10 g kg?1 diet) was added to the diets as an indigestible marker. Ileal flows in pigs fed the hydrolysed casein diet were different (p < 0.05) in amino acid contents and composition from those on the protein‐free diet. Ileal sialic acid flows in pigs fed lupin‐ or chickpea‐based diets were higher (p < 0.05) than those of animals fed soybean or casein diets. Among essential amino acids, only the apparent ileal digestibilities of phenylalanine and valine in lupin meal were lower (p < 0.05) than those in soybean. Apparent ileal digestibilities of lupin aspartate and proline, together with chickpea aspartate, were also lower (p < 0.05) than those of soybean. True ileal digestibility of nitrogen in pigs fed lupin or chickpea meals, calculated according to values from animals fed the protein‐free diet, was lower (p < 0.05) than that for soybean or casein. Among individual essential amino acids, only the true ileal digestibility of phenylalanine in lupin was lower (p < 0.05) than that in soybean. True ileal digestibility of nitrogen calculated according to values obtained with pigs fed a hydrolysed casein diet was not different among soybean, lupin or chickpea meals. Among essential amino acids, only the true ileal digestibilities of isoleucine and lysine in chickpea were lower (p < 0.05) than those of soybean. It is concluded that true ileal nitrogen and amino acid digestibilities of lupin and chickpea meals are comparable to those of defatted soybean in Iberian pigs. The results with protein‐free diets tended to underestimate endogenous protein secretion in pigs fed on diets containing protein. Copyright © 2005 Society of Chemical Industry

The effect of lupin seed protein on rat cholesterol metabolism has been studied with diets high in fat (20% by weight) and containing cholesterol (1%). The proteins used were characterised by their lysine/arginine ratios (sweet white lupin (SWL) 0·5; sweet yellow lupin (SYL) 0·4; and casein 1·8), because the lysine/arginine ratio is one of the parameters suspected to decrease or increase plasma cholesterol depending on the ratio; the diets were also characterised by their total fibre content (casein diet 5%; SWL 22%; SYL 17%) because chronic fibre intake could improve glucose tolerance and modify glucoregulatory hormone levels. Methionine content of diets was balanced. Differences among the total serum cholesterol levels of rat groups fed these diets for 28 days were not significant. But, compared to the casein diet, both lupin diets increased plasma glucagon levels and faeces. Compared to casein and SYL diets, the SWL diet significantly decreased plasma triglyceride levels and the insulin/glucagon ratio, as well as unesterified liver cholesterol and plasma LDL triglycerides levels. The SYL diet significantly increased plasma glucose and insulin, as well as liver total cholesterol compared to the casein and SWL diets. The total fibre content of the diets could indirectly modulate the effect related to the insulin/glucagon ratio on cholesterol metabolism and explain the differences observed between the SWL and SYL diets. © 1998 SCI.

The effect of highly purified sardine proteins was compared with that of casein on serum and lipoproteins lipid peroxidation and reverse cholesterol transport. Lecithin:cholesterol acyltransferase (LCAT) activity, high density lipoproteins (HDL2 and HDL3) composition and serum lipid and lipoproteins peroxidation were determined in rats fed a cholesterol-rich diet. Hypercholesterolemic rats were divided into two groups fed diets enriched with cholesterol and containing 20% of highly purified sardine proteins (SPc) or casein (CASc) for 28 days. A control group was fed a standard diet (CAS). Compared with CAS and CASc, the thiobarbituric acid reactive substances (TBARS) concentrations of low density lipoprotein (LDL)–HDL1 in SPc were 3.5- and 1.7-fold higher compared with casein diets. TBARS in HDL2 and HDL3 were, respectively, 2.3- and 1.6-fold lower in SPc compared with CASc. In SPc group, LCAT activity was higher compared to CASc and CAS (P < 0.05). Purified sardine proteins had no beneficial effects on LDL-cholesterol and lipid peroxidation. However, they reduced HDL oxidation and improved reverse cholesterol transport, in the hypercholesterolemic rat.

To investigate the effect of peptides from black soy (rat's eye) bean (Rhynchosia volubilis Lour.) on weight gain and lipid composition, Sprague–Dawley male rats were fed either a black soy peptide (BSP; 2, 6 or 10% of energy)‐containing diet or a casein control diet (20% of energy) enriched with 36% fat and 1% cholesterol for 28 days. Compared with the casein group, the BSP groups showed significant attenuation of body, liver and epididymal adipose tissue weight gains (P < 0.05). Moreover, rats that consumed BSP had a lower total cholesterol concentration and low‐density lipoprotein/high‐density lipoprotein (LDL/HDL) ratio in serum and a lower level of hepatic triglycerides as well as higher excretion of faeces compared with those fed only casein (P < 0.05). Molecular weights of peptides from black soy were mainly below 10 kDa, and Arg/Lys and Gly/Met ratios in BSP were five and three times of those in casein respectively. It is concluded that BSP can be a potent nutraceutical component for anti‐obesity and hypolipidaemic benefits. Copyright © 2007 Society of Chemical Industry

The aim of this work was to evaluate the effects of three dietary fibre‐ and polyphenol‐rich materials obtained from grapes on serum cholesterol, lipoprotein and triglyceride concentrations as well as on body weight, fat and protein digestibility in adult Wistar rats fed on cholesterol‐free and 10 g kg−1 cholesterol‐added diets. Dietary fibre content of materials ranged from 540 to 590 g kg−1. Intake of grape products increased stool weight and the amount of fat and protein excreted in faeces, but did not affect negatively animal growth or protein efficiency of the diet in Wistar rats. Studied grape fractions lower serum total cholesterol and LDL cholesterol concentrations in hypercholesterolemic rats. These results could be taken into account in potential applications of these products as a food ingredient. © 2000 Society of Chemical Industry

The main components of okara, a by-product from soybean, are dietary fibre and protein. In this study we fed okara based diets with different amounts of fibre, protein and isoflavones to determine their effect on the lipid profile in the plasma, liver and faeces of an animal model. Male golden Syrian hamsters were fed high-fat diets supplemented with okara for 3 weeks. The supplemented diets contained 13% or 20% of okara fibre (OK-13 and OK-20), low-protein okara with 13% of fibre (OK1-13), and isoflavone-free okara with 13% of fibre (OK2-13). Okara supplemented diets did not produce significant differences in the feed intake or body weight gain (p > 0.05). The plasma levels of triglycerides, VLDL- plus LDL cholesterol and total cholesterol in hamsters fed OK-20 decreased significantly (p < 0.05) with respect to the 20% control group (C-20). However, no significant differences (p > 0.05) were found in LDL- and HDL-cholesterol plasma levels in all experiments. Total lipids, triglycerides, total and esterified cholesterol concentrations in liver were reduced by OK-20 diet. Regarding the hamsters fed OK-13 diets, the mean values of the total lipid, triglyceride and cholesterol in the plasma and liver decreased as compared to the control (C-13), but the differences were not statistically significant. All of the okara diets assayed increased the faecal excretion of total lipids, triglycerides, free cholesterol and total nitrogen (p < 0.05) compared to their respective controls. Our results suggest that the main components of okara, dietary fibre and protein, could be related with the total lipids and cholesterol decrease in the plasma and liver, as well as with the faecal output increase in high-fat fed hamsters. Okara might play an interesting role in the prevention of hyperlipidemia and could be used as a natural ingredient or supplement for functional food preparation.

This study was performed to investigate the effects of the peptic undigested fraction derived from soybean protein hydrolysate (UDSP) on lipid metabolism in rats fed a cholesterol‐enriched diet (1%). Eighteen male Wistar rats‐weighing 205–235 g were randomly divided into three groups: the control group (20% casein), U2 (18% casein + 2% UDSP), and U5 group (15% casein + 5% UDSP). After 4 weeks, rats were sacrificed, and the lipid profiles of the plasma, liver, and feces were determined. Body weight gain, daily food intake, and liver weight showed no differences among the groups, but the feeding efficiency ratio in the U5 group was significantly lower than that in the other groups (P < 0.05). There were no changes in plasma cholesterol, LDL‐C, HDL‐C, and liver cholesterol levels in each group. However, the U5 group showed a significantly lower VLDL‐C compared to the control and U2 groups. In addition, the plasma and liver TG content were lower in the U2 and U5 groups than in the control group (P < 0.05). Moreover, the fecal bile acid and total neutral steroid excretions were higher in the U2 and U5 groups (P < 0.05) compared to the control group.

The dietary influence of sesame protein isolate (protein content 91.5%), produced from dehulled, defatted sesame meal, on blood and tissue lipid profile and lipid peroxidation has been assessed in normal and hypercholesterolemic rats. To evaluate their hypocholesterolemic and antioxidative activity in vivo, we fed 18% sesame protein isolate with or without 2% cholesterol in comparison with casein to rats for 28 d. We determined plasma total protein, total cholesterol, high-density lipoprotein (HDL)-cholesterol, low-density lipoprotein (LDL)-cholesterol, triacylglycerol as well as susceptibility of plasma and erythrocyte membrane lipid to oxidation ex vivo. Liver tissue lipid, cholesterol, phospholipids, and lipid peroxidations were also determined. The total cholesterol, LDL-cholesterol and triacylglycerol levels were significantly reduced in the sesame protein isolate and isolate containing cholesterol group than the corresponding control casein groups. HDL-cholesterol level was also increased in sesame protein isolate (41%) and protein isolate containing cholesterol group (38%) than the corresponding control casein and casein containing cholesterol groups. There was 49% and 64% lowering of plasma lipid peroxidation as well as 36% and 56% lowering of lipoprotein oxidation susceptibility (LOS) in the 2 experimental groups (sesame protein isolate and isolate containing cholesterol group) than the corresponding control (casein and casein containing cholesterol) groups. There was significant lowering of erythrocyte membrane lipid peroxidation (68% and 63% lowering in sesame protein isolate and isolate containing cholesterol groups) and liver lipid peroxidation (61% and 76% lowering in the 2 experimental groups than the corresponding control casein groups). Therefore, our results indicate that sesame protein isolate decreases cholesterol concentration in plasma, increases HDL-cholesterol, and also decreases plasma and erythrocyte membrane lipid peroxidation with or without cholesterol fed diet in rats.

The hypolipidemic effect of cold extracted virgin coconut oil (CEVCO) and hot extracted virgin coconut oil (HEVCO) compared to commercially available coconut oil (CCO) was studied using male wistar rats. Groups were designated as control, hypercholesterimic control (HC Control), HC+10% (CEVCO), HC+15% (CEVCO), HC+10% (HEVCO), HC+15% (HEVCO) and HC+15% (CCO). Control rats were fed a basal diet while other groups were initially fed a hypercholesterimic diet (2% cholesterol and 0.25% bile salts) for 15 days, then fed with CEVCO or HEVCO (both at 10% and 15% levels), and commercial coconut oil (15%) for 30 days. Total cholesterol and LDL cholesterol levels were significantly (p<0.05) decreased by approximately 25 and 40%, respectively, in the blood plasma, while HDL cholesterol increased significantly (p<0.05) by approximately 21% in HEVCO fed rats. Tissue cholesterol and triacylycerol (TAG) levels in both liver and heart tissues decreased significantly (p<0.05) in CEVCO and HEVCO fed animals, compared with CCO fed rats.

This study was designed to evaluate the efficacy of an anthocyanin pigmented rice (e.g. black rice) to mitigate the onset of hypercholesterolemia in rats-fed atherogenic diets. Male Wistar (n = 10/group) rats were fed with atherogenic diets containing 0.5% cholesterol in the presence and in the absence of bile salt (e.g. 0.05% cholic acid) along with a standardized black rice extract (BRE) (e.g. 3%, w/w). All animals were individually housed in stainless steel cages and fed with the experimental diets during a 12-h period for 10 weeks. Body weights of rats were measured every week of the experiment. After 10 weeks fed on experimental diets, rats were sacrificed and plasma total cholesterol, HDL and LDL cholesterol and triacylglycerols were measured immediately. The total cholesterol (TC) content in the liver, heart and aorta, and the concentration of triacylglycerol (TAG) were measured after lipid extraction using Folch method. Rats fed with 0.5% cholesterol containing diets which also included bile salt exhibited a considerably more severe hypercholesterolemia than counterparts fed diets containing only 0.5% cholesterol. The inclusion of the BRE in diets significantly (p < 0.05) decreased the level of TC, LDL–TC and TAG in plasma of rats-fed control diets that either contained or were absent in bile salt (p < 0.05). There were no differences in HDL-level. Liver crude lipids and total cholesterol levels were also significantly (p < 0.05) decreased in experimental groups relative to the control group in both experiments. Thus, supplementation of atherogenic experimental diets with BRE effectively decreased lipid levels in hypercholesterolemic rats. In lieu of the mixture of bioactive components present in BRE, it is possible that more than one mechanism underlying this reduction in lipids is involved.

The aim of this experiment was to assess, using sheep, the nutritive value of lupin seed transgenically modified to contain sunflower seed albumin. Eighty Merino wether sheep of mean live weight 32.3 kg were divided into two groups and fed 796 g dry matter (DM) day−1 of a cereal hay‐based diet containing 350 g kg−1 of either the transgenic or parent (unmodified) lupin seed for 6 weeks. Measurements were made of wool growth and live weight gain. After 6 weeks, half the sheep in each group were selected for a urine and faeces balance study in which organic matter (OM), nitrogen (N) and urinary purine metabolites were measured. Blood samples were taken from all sheep at the beginning and end of treatment and analysed for amino acids and plasma metabolites. A comprehensive chemical analysis of the grains showed that there was little difference between them in terms of most nutritional components, but the transgenic lupin seed contained a 2.3‐fold higher methionine concentration and 1.3‐fold higher cysteine than did the parent. There were no significant differences between grains in OM digestibility, rumen microbial protein synthesis or in sacco degradability of dry matter. Sheep fed the transgenic lupin grain had an 8% higher rate of wool growth (P < 0.01) and 7% higher live weight gain (P < 0.05) than sheep fed the parent grain. The sulphur (S) concentration of wool and the cysteine concentration of plasma were also higher in the sheep fed the transgenic lupin by 2.7% and 11.5% respectively (P < 0.05). Plasma methionine was increased by 10%, but the differences were not statistically significant (P > 0.1). Plasma urea N was lower in the sheep fed the transgenic grain than those fed the parent grain (6.5 vs 6.8 mmol l−1, P < 0.05). The results show that genetic modification of a feed grain can improve its nutritive value for ruminants. The size and nature of the responses were consistent with the transgenic lupins providing more methionine to the tissues, a first‐limiting amino acid for sheep. © 2000 Society of Chemical Industry

Five castrated male Iberian pigs (100 ± 2 kg mean live body weight) fitted with T‐shaped ileal cannulas were used to determine ileal digestibility of legume seed meals. The diets were based on defatted soybean (256 g kg?1), lupin (451 g kg?1) or chickpea (731 g kg?1) seed meals and contained similar amounts of digestible energy (14.2–15.1 kJ g?1) and protein (107 g kg?1). Only myristic acid (C14:0) ileal apparent digestibility was higher (p < 0.05) in lupin than in chickpea meal. Ileal apparent digestibility of unsaturated fatty acids (FA) (710–920 g kg?1) was higher (p < 0.05) than that of saturated (480–710 g kg?1) FA for both seed meals. Ileal digestibility of chickpea starch was 850 g kg?1. Ileal digestibility of oligosaccharides (sucrose, raffinose, stachyose, verbascose and ciceritol) in lupin (820 g kg?1) and chickpea (690 g kg?1) was higher (p < 0.05) than that of defatted soybean (510 g kg?1). However, total amounts of oligosaccharides digested were higher (p < 0.05) for lupin and chickpea meals (29.6 and 24.6 g kg?1 feed respectively) than for soybean (15.2 g kg?1 feed). Among individual non‐starch polysaccharide (NSP) sugars, ileal digestibility values for rhamnose and xylose in lupins were higher (p < 0.05) than those for soybean or chickpea. Ileal digestibility of lupin meal NSP (650 g kg?1) was higher (p < 0.05) than those of soybean (560 g kg?1) or chickpea (430 g kg?1). Up to the terminal ileum, higher (p < 0.05) amounts of lupin total NSP (94.1 g kg?1) than for soybean (24.3 g kg?1) or chickpea (27.1 g kg?1) were digested. Lactate, acetate and isobutyrate concentrations were higher (p < 0.05) in ileal contents of pigs fed lupin and chickpea seed meals compared with soybean‐fed animals. It is concluded that higher lactate and short chain fatty acids concentrations in ileal contents of pigs fed lupin or chickpea seed meals compared with soybean are likely to be the result of higher total amounts of lupin NSP and/or lupin and chickpea oligosaccharides digested up to the terminal ileum. This might have a protective effect on Iberian pig's intestinal physiology. Both lupin and chickpea meals are likely to constitute a valuable alternative to defatted soybean in Iberian pig feeding. The effects on organ relative weights (g per 100 g bw) and plasma amino acid concentrations of diets based in legume (faba bean (Vicia faba), lupin (Lupinus angustifolius), chickpea (Cicer arietinum) and soybean (Glycine max)) seed meals as the only source of protein were studied with growing rats using lactalbumin or casein as controls. Also, legume seed meal extracted globulins were included in control diets replacing lactalbumin to produce legume globulin‐based diets, and legume residue fractions, containing most of the starch and/or insoluble fibre (NSP+lignin) from the seed meals, were included in control diets to reach the same amounts present in whole legume seed meal diets. All experimental diets were formulated to contain the same amounts of protein (100 g kg−1) and energy (15.5 kJ g−1), and were supplemented with essential amino acids. Compared with lactalbumin, higher relative weights of gastrointestinal sections were determined in rats fed legume seed meals or their corresponding residue fractions. On the contrary, spleen relative weight was lower in rats fed diets containing lupin, chickpea or soybean meals or extracted globulins, while residue fractions had no effect on it. Thymus relative weight was also lower in rats fed whole chickpea seed meal or any of the extracted legume globulins. Except for chickpea meal, animals fed legume‐ or legume protein‐based diets had lower liver relative weights than controls. Lower proportions (mg g−1 tissue) of glycogen, and lower total protein and RNA, were also determined in the livers of rats fed lupin seed meal. Free plasma concentrations (mM litre −1) of glycine, histidine and arginine were higher, and threonine, leucine and lysine were lower, in rats fed diets based in all legume seed meals of their respective globulin proteins. The possible reasons and implications of these results are discussed. © 1999 Society of Chemical Industry

ABSTRACT: Hypercholesterolemic hamsters were fed for 4 wk on diets rich in saturated fatty acids and cholesterol, differing only in protein source (20 %): casein (control group, HC), whole cowpea seed (HWS), and cowpea protein isolate (HPI). Hamsters fed on HWS and HPI presented significant reductions in plasma total cholesterol and non‐HDL cholesterol. HPI and HC presented similar protein digestibility, which were significantly higher than that of HWS. Animals fed on HWS presented significantly higher levels of bile acids and cholesterol in feces than did the animals fed on casein or HPI diets. Histological analyses of the liver showed that HC diet resulted in steatosis widely distributed throughout the hepatic lobule, while HWS and HPI diets promoted reductions in liver steatosis. The effectiveness of HWS for modulating lipid metabolism was greater than that of HPI, as measured by plasma cholesterol reduction and liver steatosis.

BACKGROUND: Declines due to Papaya ringspot virus (PRSV) are the primary constraint limiting production of papaya (Carica papaya L.) in tropical and subtropical regions. Given the lack of suitable sources of resistance in the genus, transgenic papayas carrying the coat protein gene of a local strain of PRSV were recently developed to manage the disease in Jamaica. For assurance of food safety, a 90 day subchronic whole‐food feeding study in rats was conducted with transgenic and non‐transgenic papayas. RESULTS: Male and female Wistar rats (N = 56) received diets containing transgenic or non‐transgenic papayas at twice the equivalent of the average daily human consumption of fresh papayas. No adverse effects on animal behaviour or differences in body weight and organ weight between the animal groups (P < 0.05) were observed during the study. Necropsy at the end of the study indicated that neither pathological nor histopathological abnormalities were present in the liver and kidneys of rats in the control and treated groups. In addition, the levels of activity of plasma, liver and kidney acid and alkaline phosphatases and alanine and aspartate transaminases were comparable (P < 0.05). Although significant differences (P < 0.05) were found in the cholesterol and triglyceride levels in the plasma and liver among animals of the treated groups, overall the values were comparable to those of the control. CONCLUSION: The results suggest that consumption of transgenic papayas may not have adverse effects in rats as regards the parameters considered in this study. This study describes the hypocholesterolaemic effect of whole lupin and its protein in hamsters. The diets were: casein (control group HC), lupin protein isolate (group HPI) and whole lupin seed (group HWS). Diets from HPI and HWS promoted a significant reduction of total cholesterol and non-HDL cholesterol in the hamsters’ plasma as compared with HC. The true digestibility of HPI and HC groups were similar and differed significantly from the HWS one, which in turn showed a significant difference in total sterol excretion as compared to the former groups. Histological analysis of the liver revealed that animals fed on HPI and HWS diets presented a low level of steatosis (level 1) as compared to the ones fed on HC diet (level 4). Our findings demonstrate that protein isolate from Lupinus albus from Brazil has a metabolic effect on endogenous cholesterol metabolism and a protector effect on development of hepatic steatosis.
