BACKGROUND: This work aimed to investigate the influence of both ripening stage and cultivation system on the total phenolic compounds (TPC) and total antioxidant activity (TAA) of passion fruit pulp. TPC extraction was optimized using a 23 central composed design. The variables were fruit pulp volume, methanol volume and extraction solution volume. TPC was determined using the Folin–Ciocalteu reaction, and TAA using the ABTS radical reaction. RESULTS: The conditions to extract TPC were 2 mL passion fruit pulp and 9 mL extraction solution containing 40% methanol:water (v/v). TPC values increased in the passion fruit pulp during ripening for both cultivation systems, ranging from 281.8 to 361.9 mg gallic acid L?1 (P?0.05) for the organic pulp and from 291.0 to 338.6 mg gallic acid L?1 (P?0.05) for the conventional pulp. CONCLUSION: TPC values increased during ripening for both organic and conventional passion fruit. The same was true for TAA values for conventional passion fruit. For organic passion fruit, however, TAA values were highest at the initial ripening stages. These results suggest that antioxidant compounds exert strong influence on the initial ripening stages for organic passion fruit, when TPC still did not reach its maximum level. Sour cherry (Prunus cerasus L.) cultivars native to Hungary and some of them grown worldwide, display great phenotypic variability. Eleven sour cherry cultivars were analysed for their main quality attributes including fruit weight, soluble solid content, acidity, pH, specific sugars (glucose and fructose), antioxidant capacity as well as total polyphenolic (TPC), total anthocyanin (TMAC) and vitamin C contents. Results showed wide variation in both fruit quality and antioxidant parameters of Hungarian sour cherries. The anthocyanin contents varied from 11.3 to 93.5 mg/100 g. An amarelle‐type cultivar, ‘Pipacs 1’ showed the highest antioxidant capacity (21.85 mmol AA L?1), TPC (49.04 mg GA L?1) and vitamin C (8.98 mg/100 g) content. Key enzymes in anthocyanin biosynthesis were expressed in both yellow‐coloured flesh and red skin of ‘Pipacs 1’ fruits at all ripening stages. The detected diversity presents a choice that can satisfy different consumer preferences, and meet specific nutritional requirements.  相似文献

Total antioxidant activity (TAA), total phenolic compounds (TPC), and physicochemical characteristics of ready-to-drink orange juice and nectar from the most consumed brands available in Brazil were evaluated. TPC ranged from 18.7 to 54.2 mg of gallic acid/100 mL, and TAA varied from 57.88 to 349.32 μmol TEAC/100 mL ready-to-drink orange juice and nectar. The ascorbic acid content was the only physicochemical parameter that showed strong variation among packages and brands. Correlation of TPC with TAA showed that the higher the level of TPC the higher the TAA. Correlation of ascorbic acid content with TAA is higher for ready-to-drink orange juice than nectar. The same was found for the correlation of ascorbic acid content with TPC. The results confirm the contribution of the TPC to TAA.  相似文献

ABSTRACT

With the diffusion of commercial apple varieties since the second half of the last century, hundreds of different local cultivars rapidly disappeared from orchards in Italy, and the particular quality attributes of these fruits are still at risk of being lost today. The aim of this research was to use sensory, nutritional and genetic techniques in order to define the overall quality of fruits from ancient apple cultivars grown in Northern Italy. Cluster analysis arranged the 10 genotypes into five groups; in most cases, the analyzed cultivars demonstrated higher sensory and nutritional qualities than the control (Golden Delicious cv). The cultivars showed higher values of total polyphenolic compounds and a stronger antioxidant activity. The vitamin C content also differed from the control. The sensory analysis, performed by the Fruit Tasters Italian Organization, was an effective characterization tool and genetic analysis further helped to characterize the identity of these interesting cultivars.

PRACTICAL APPLICATIONS

The characterization and valorization of old apple cultivars are essential to avoid loss of potentially useful germplasm. The reduction of biodiversity of cultivars found in agriculture could lead to problems such as higher susceptibility to widespread outbreaks of plant diseases and pests, and at the same time, the loss of precious source of nutrients. The conservation of apple biodiversity is valuable in order to maintain the gene pool of the species and to introduce superior quality traits into apple‐breeding programs.  相似文献

BACKGROUND: Plums are a rich source of nutritive and bioactive compounds. The objective was to evaluate the impact of fruit tissue (flesh and peel), ripening stage and cultivar on the composition of phytochemicals in plums (Prunus domestica L.) and to analyse the relation between total antioxidant activity (TAA) and the measured variables. RESULTS: The content of total sugars and total organic acids was higher in flesh but total phenolic content (TPC) and TAA was statistically higher in plum peel. The composition of sugars depended on fruit tissue, ripening stage and the cultivar. Ripening and fruit tissue affected the composition and concentrations of organic acids. TPC of ‘Haganta’ peel increased during ripening but there was no significant time trend for ‘Jojo’ peel and flesh of both cultivars. The composition of phenolics in peel depended on the cultivar, but in flesh on the cultivar and ripening. Ripening resulted in increased total anthocyanins and TAA in peel. TAApeel was positively correlated with TPCpeel, total anthocyaninspeel, cyanidin 3‐glucosidepeel and peonidin 3‐rutinosidepeel in the cultivar ‘Haganta’. CONCLUSION: The composition of phytochemicals in P. domestica fruit depends on fruit tissue, ripening stage and the cultivar. The present work investigated the phenolic profiles (including nonanthocyanin and anthocyanin phenolics), antioxidant activities, and neuroprotective potential of mulberry fruit (MF) (Morus atropurpurea Roxb.) grown in China at different ripening stages. High‐performance liquid chromatography‐tandem mass spectrometry method (HPLC‐MS/MS) was used to identify and quantify the phenolic compounds. The antioxidant capacity, total phenolic content (TPC), total flavonoid content (TFC), and total monomeric anthocyanin content (TAC) were determined using spectrophotometric methods. The neuroprotective effects of MFs at different ripening stages were investigated using Aβ25‐35‐treated PC12 cells as the cellular model of Alzheimer's disease. Of the 19 phenolic compounds characterized from the MF extracts, the contents of rutin and anthocyanins increased and that of chlorogenic acid decreased significantly with maturity. At the fully ripened stage, MF extracts showed the highest amounts of TPC (11.23 mg gallic acid equivalents/g fresh weight), TFC (15.1 mg rutin equivalents/g fresh weight), and TAC (1177 mg cyanidin 3‐O‐glucoside equivalents/100 g fresh weight). Meanwhile, antioxidant activity of MF extracts at this stage was highest according to ABTS (an IC50 value of 4.11 μg/mL) and DPPH (an IC50 value of 10.08 μg/mL) assays. Cellular assays revealed increased cell viability in cells treated with the ripe MF extracts; compared with the control groups, the ripening fruits also increased the antioxidant enzyme levels in PC12 cells. Together, these results suggest that the antioxidant activities and neuroprotective properties of ripening MFs are related to the contents and types of phenolic compounds that are present in the fruits.  相似文献

Solvent, impregnation time, sonication repetitions, and ultrasonic power were important factors in the process of ultrasound‐assisted extraction from chicory (Cichorium intybus) root, while there were no studies about optimizing these 4 factors for extract yield, total phenolic content (TPC), antioxidant, antibacterial, and antifungal activity of the extracts using orthogonal matrix design. The present research demonstrated that the solvent composition played a significant role in the improving extract yield, TPC, antioxidant, and antibacterial activities. The other 3 factors had inequable effect on different purposes, ultrasonic power could improve TPC and antioxidant activity, but long time of extraction lowered antioxidant activity. The TPC increased from 22.34 to 27.87 mg GAE (gallic acid equivalents)/100 g (dry extracts) with increasing solvent polarity. The half inhibition concentration (IC50, μg/mL) of the radical scavenging activity of the chicory extracts ranged from 281.00 to 983.33 μg/mL. The content of caffeoylquinic acids of root extract, which was extracted by the optimal combination was 0.104%. Several extracts displayed antibacterial activities against Escherichia coli, Staphylococcus aureus, Bacillus thuringiensis, Bacillus subtilis, and Salmonella typhi, while Penicillium sp. and Aspergillus sp. resisted against all the extracts. Combination of 70% ethanol v/v, 24‐h impregnation time, 3 sonication rounds, and 300‐W ultrasonic input power was found to be the optimal combination for the chicory extract yield, TPC, antioxidant activity, and antibacterial activity.  相似文献

Eight major mulberry cultivars [Nakhonratchasima 60 (NS 60), Buriram 60 (BR 60), Chumphon (CP), Wavee (WV), Chaingmai (CM), Pikultong (PT), Kamphaengsaen (KS) and Kamnanchul (KJ)] cultivated in Thailand were assessed for their flavonoid and phenolic acid composition using HPLC and tested for antioxidant potential using 2,2‐diphenyl‐1‐picrylhydrazyl hydrate (DPPH) assay. The total phenolic content (TPC) ranged from 104.78 to 213.53 mg GAE/100 g DW, and total flavonoid content (TFC) ranged from 69.58 to 211.01 mg CE/100 g DW. The major flavonoid compounds in mulberry fruit cultivars were (+)‐catechin (309.26–750.01 mg/100 g DW), procyanidin B1 (62.59–224.41 mg/100 g DW), quercetin (5.36–58.42 mg/100 g DW), rutin (18.73–26.90 mg/100 g DW) and (?)‐epicatechin (8.47–29.21 mg/100 g DW). Gallic acid, cinnamic acid and p‐hydroxybenzoic acid were found to be the major phenolic acids in mulberry fruit cultivars. The gallic acid and cinnamic acid contents ranged from 7.33 to 23.90 mg/100 g DW and from 11.64 to 15.05 mg/100 g DW, respectively. p‐Hydroxybenzoic acid content ranged from 1.77 mg/100 g DW (PT) to 7.13 mg/100 g DW (KJ). DPPH‐scavenging ability was excellent for ethanolic extract of NS 60, and EC50 value of NS 60 (241.83 μg mL?1) was significantly lower than those of the others (< 0.05). TPC and TFC of the mulberry fruit were positively correlated with the DPPH‐scavenging ability.  相似文献

In this study, the physicochemical characteristics and antioxidant potentials of 10 Chinese olive cultivars were investigated. Considerable differences were found between cultivars in weight, edible yield, water content, size, shape, total soluble solids, and total titratable acidity. The major sugars and organic acids in all the cultivars were glucose, fructose, and malic acid. Phenolics were the most abundant antioxidants (1174.0 to 1799.6 mg gallic acid equivalents/100 g fresh weight). Twelve phenolic compounds were identified and quantified by UPLC‐MS/MS. Six compounds were identified first in the Chinese olive, with 3‐O‐galloylquinic acid and geraniin isomers as the most abundant compounds. The results also indicated that the phenolic content (TPC) correlated with the antioxidant properties of Chinese olive fruit extracts. A principal component analysis indicated that the Tantou and Tanxiang cultivars were a valuable source, with high TPC and antioxidant activity, whereas Maken22, Changying, and Green changying cultivars may be eaten fresh and had high sugar and low phenolic contents. This information will be useful for selecting suitable cultivars for industry and the market.  相似文献

Total phenolic content (TPC) and antioxidant capacity of lettuce were evaluated using the Folin-Ciocalteu method and DPPH assay, respectively, on 25 cultivars of lettuce, including leaf, romaine, crisphead, and butterhead types, cultivated over two harvest periods. Leaf lettuce possessed the highest TPC and highest DPPH scavenging ability, followed by romaine, butterhead and Batavia (crisphead subtype). Within a lettuce type, red pigmented lettuce cultivars had higher TPC and antioxidant capacity than did green cultivars grown under the same conditions. In addition, lettuce harvested in July possessed higher TPC and antioxidant capacity than did lettuce harvested in September, suggesting that environmental conditions could influence the phenolic content and antioxidant activity of lettuce. These results suggest that Colorado grown lettuce may serve as potential dietary sources of natural phenolic antioxidants.  相似文献

ABSTRACT

The antifungal, aflatoxin inhibitory and antioxidant activity of methanol–aqueous extract (2:1) of 62 medicinal plants was explored. Based on the antifungal results, the extracts of 25 plants showed more than 50% antifungal activity and were further investigated for their aflatoxin inhibition and antioxidant properties. Methanol–aqueous extracts of Phyllanthus emblica and Terminalia chebula fruits caused 100% inhibition of aflatoxin production by the toxigenic strain of Aspergillus flavus in semisynthetic medium at 1 mg/mL. In addition, P. emblica (IC50 = 4.1 µg/mL) and T. chebula (IC50 = 6.9 µg/mL) fruits extracts exhibited strong antioxidant activity during the 2,2‐diphenyl‐1‐picrylhydrazyl radical‐scavenging assay in comparison with butylated hydroxytoluene (IC50 = 8.1 µg/mL) and butylated hydroxyanisole (IC50 = 6 µg/mL).

PRACTICAL APPLICATIONS

Based on the results of the present study, methanol–aqueous extracts of Phyllanthus emblica and Terminalia chebula, being endowed with strong antifungal, aflatoxin inhibitory and antioxidant activity, may be recommended as plant‐based preservatives for the enhancement of shelf life of food items and their protection from the undesirable harmful effects of molds, aflatoxin and free radical‐mediated damages.  相似文献

Seventeen natural population of Seabuckthorn (SBT), which comprised 187 plants from trans-Himalaya, were studied to find out variability and genotypic effect on total phenolic content (TPC), total antioxidant capacity (TAC), ascorbic acid and carotenoids content in fruit pulp. The fruits were found to be rich in TPC ranging from 964 to 10,704 mg gallic acid equivalent/100 g. The free radical-scavenging activity in terms of inhibitory concentration (IC50) ranged from 0.7 to 9.1 mg/ml and ferric reducing antioxidant potential (FRAP) from 180 to 1355 FeSO4·7H2O μg/ml. The ascorbic acid and carotenoids content ranged from 56 to 3909 mg/100 g and 0.1–14.4 mg/100 g, respectively. A variation of 1–11 fold in TPC, 1–14 folds in IC50 by 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay and 1–8 fold in ferric reducing antioxidant potential, 1–70 fold in ascorbic acid content and 1–206 fold in carotenoid content among the examined fruit across 17 populations underlines the important role played by genetic background and the geographical location for determining the health promoting compounds. Significant correlation was observed between TPC, IC50, FRAP, carotenoids, ascorbic acid, fruit lightness (L*) and plant height. Among the 20 morphological traits studied, fruit colour and plant height showed positive correlation with the health promoting compounds.  相似文献

The fruit quality parameters and antioxidant capacity (ferric reducing antioxidant power, FRAP) and total phenolic content (TPC) were determined in 27 apricot cultivars and hybrids of diverse origins. Twenty one- to 35-fold variations were measured among FRAP and TPC values. Besides genotype, harvest year also contributed significantly (P≤0.05) to the variations of TPC presumably due to the climatic differences between years. A subset of genotypes (15) was also analyzed for their antiradical activities (2,2'-diphenyl-1-picrylhydrazyl, DPPH; total radical-scavenging activity, TRSA; water-soluble antioxidant capacity, ACW; and lipid-soluble antioxidant capacity, ACL), and vitamin C contents as well as color indices (CIE H°, L*, and chroma). The hybrid "Preventa" had outstanding FRAP (>10.4 mmol ascorbic acid/L), DPPH (74.45%), TRSA (0.002%), ACW (33587.5 nmol AA/L) and ACL (78.65 nmol Trolox/L), TPC (>2890.0 mg gallic acid/L), and vitamin C (16.17 mg/100 g FW) levels and an average carotenoid content estimated from the hue angle (66.99°). Most antioxidant and antiradical activities correlated significantly except for TRSA; the closest correlation was observed between FRAP and ACW (r=0.952). Only TRSA showed significant correlations with color indices, H° and chroma, suggesting TRSA measures at least a fraction of the antioxidant capacity attributable to apricot carotenoids.  相似文献

Methanolic extracts of six cultivars of Portulaca oleracea were analyzed for their total phenol content (TPC) using the Folin–Ciocalteu method. The antioxidant activity was measured using the 1,1-diphenyl-2-picrylhydrazyl, ferric-reducing antioxidant power (FRAP) and β-carotene bleaching (BCB) assays. The iodine titration method was used to determine the ascorbic acid content (AAC). The TPC of the cultivars of P. oleracea ranged from 127 ± 13 to 478 ± 45 mg GAE/100 g of fresh weight of plant. There was good correlation between the TPC value and its AEAC, IC50 and FRAP values (r2 > 0.9) for all the cultivars. The AAC for the cultivars ranged from 38.5 ± 0.6 to 73.0 ± 17.5 mg/100 g. The TPC value of the common variety PO1, was the lowest compared to the ornamental cultivars (PO2–PO6). The BCB assay showed that all cultivars were capable of inhibiting lipid peroxidation and the inhibition power did not correlate with TPC value.  相似文献

ABSTRACT

Seven red raspberry cultivars (Heritage, Canby, Willamette, Hollanda Boduru, Newburgh, Tulameen and Meeker) were evaluated for their total anthocyanin (TACY), total phenolics (TP), trolox equivalent antioxidant capacity (TEAC), fatty acids (FAs) and soluble sugars properties. The TACY, TP and TEAC content ranged from 171.5 to 475.0 µg/g fresh weight (fw), 864.4–1246.9 µg gallic acid equivalent/g fw and 19.4–32.7 µmol TE/g fw, respectively. The highest values of TACY, TP and TEAC content were obtained from the Newburgh cultivar. The FA compositions of raspberry cultivars ranged from 4.97 to 20.31% saturated FAs, 14.65–18.47% monounsaturated FAs (MUFAs) and 62.85–78.68% polyunsaturated FAs (PUFAs). Cultivars Meeker and Newburgh had the highest level of MUFAs, respectively, while the highest level of PUFAs was observed in cultivar Hollanda Boduru. The main soluble sugars of the seven raspberry genotypes were also separated, identified and quantified using high‐performance liquid chromatography with refractive index detection. According to the results, fructose, glucose and sucrose were detected as the main soluble sugars, while xylose and mannose were not detected in the raspberry fruits.

PRACTICAL APPLICATIONS

There have been no previous studies comparing the fruit quality characteristics of raspberry cultivars. Raspberries may be a good alternative crop with a high market value, especially highly profitable due to the long harvesting period, because of the ideal climatic conditions. The objective of this work was to evaluate seven red raspberry cultivars in terms of their total anthocyanin, total phenolic, antioxidant capacity, fatty acids profile and soluble sugar properties.  相似文献

In this study, the antioxidant activity of persimmon fruit of 4 different persimmon cultivars cultured at Korea was evaluated. There were 3 astringent persimmon cultivars [‘Bongok’ (BO), Japanese Hatchiya; ‘Cheongdobansi’ (CB); and ‘Dogeunjosaeng’ (DJ), Japanese Tonewase] a 1 non-astringent persimmon cultivar [‘Seochonjosaeng’ (SJ), Japanese Nishimurawase] fruits. After preparing extracts of fruits according to parts (calyx, pulp, and peel) with 4 different solvents (acetone, ethanol, methanol, and water), total phenolic contents (TPC) and antioxidant activity were determined. The extracts of calyx showed significantly higher TPC and antioxidant properties than those of the other persimmon parts. The highest 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity (IC50=43.36±1.78 μg/mL) and reducing power (IC50=81.93±1.18 μg/mL) were found at acetone extract of SJ calyx, while SJ pulp extracts showed relatively lower antioxidant activities. Astringent persimmons showed relatively higher antioxidant activity than non-astringent persimmons in peel and pulp parts. These results indicated that persimmons have different antioxidant activity depending on cultivars and parts.  相似文献

Black rice bran contains phenolic compounds of a high antioxidant activity. In this study, the 40% acetone extract of black rice bran was sequentially fractionated to obtain 5 fractions. Out of the 5 fractions, ethyl acetate fraction was subfractionated using the Sephadex LH‐20 chromatography. The antioxidant activity of phenolic compounds in the extracts was investigated by 2,2‐diphenyl‐1‐picrylhydrazyl (DPPH) radical assay, 2,2‐azino‐bis‐(3‐ethylenebenzothiozoline‐6‐sulfonic acid) (ABTS) radical cation assay, reducing power. The subfraction 2 from ethyl acetate fraction had the highest total phenolic contents (TPC) (816.0 μg/mg) and the lowest EC50 values (47.8 μg/mL for DPPH radical assay, 112.8 μg/mL for ABTS radical cation assay, and 49.2 μg/mL for reducing power). These results were 3.1, 1.3, and 2.6 times lower than those of butylated hydroxytoluene (BHT), respectively. At a concentration of 100 μg/mL, the antioxidant activity and TPC of various extracts was closely correlated, with correlation coefficients (R2) higher than 0.86. The major phenolic acid in subfraction 2 was identified as ferulic acid (178.3 μg/mg) by HPLC and LC‐ESI/MS/MS analyses. Our finding identified ferulic acid as a major phenolic compound in black rice bran, and supports the potential use of black rice bran as a natural source of antioxidant.  相似文献

ABSTRACT

Content of individual sugars, organic acids, total phenolics (TPH), some important phenolic compounds (kaempferol, myricetin, quercetin, and ellagic acid) and total antioxidant capacity (TAC) in the fruits of strawberry, raspberry and blackberry were studied. A comparison was made between cultivars and wild relatives of each species (Fragaria vesca L., Rubus idaeus L. and Rubus fruticosus L.). The main sugars found were fructose and glucose both in the fruits of wild species and the studied cultivars. Citric acid was determined to be the major organic acid in most of tested berries with the exception of blackberry cultivars, where malic acid was dominant. The content of individual phenolic compounds varied among the wild species, as well as among the studied cultivars. Ellagic acid content was higher in F. vesca, R. idaeus and R. fruticosus (122.5 µg/g FW, 12.71 µg/g FW and 61.7 µg/g FW, respectively) than that obtained in analyzed cultivars. Overall, TPH expressed higher values in the wild strawberry and blackberry species in comparison to the studied cultivars, and consequently, the highest levels of TAC were recorded in F. vesca (5.78 mg asc/g FW), followed by R. fruticosus (4.95 mg asc/g FW).

PRACTICAL APPLICATIONS

In recent years, both wild and cultivated berries have become very attractive for consumers because of potentially beneficial phytochemicals contained in these fruits. Fruit nutritional quality can be described by standard quality parameters (sugars and organic acids), and the analysis of antioxidant capacity influenced by specific related compounds. The importance of flavonoids and other phenolics have been suggested to play a preventive role in the development of cancer and heart disease. A significant positive correlation observed in this study between total phenolics and total antioxidant capacity indicate the need for the use of wild species in the breeding programs of small fruits, especially strawberry and blackberry, in order to increase their nutritive value and the health benefits.  相似文献

The antioxidant and the tyrosinase inhibitory activities of 4 different solvents (acetone, ethanol, methanol, and water) for preparation of extracts from guava (branch, fruit, leaf, and seed) were evaluated by measuring total phenolic contents (TPC), DPPH radical scavenging activity, ABTS radical scavenging activity, reducing power (RP), and tyrosinase inhibitory activity. The extracts of branch and leaf showed relatively higher antioxidant properties than those of fruit and seed. The highest TPC (141.28 mg/g gallic acid equivalents), DPPH radical scavenging activity (IC50=34.01 μg/mL), ABTS radical scavenging activity (IC50=3.23 μg/mL), and RP (IC50= 75.63 μg/mL) were found in acetone extract of leaf, while water extract of seed had the lowest antioxidant activity. The tyrosinase inhibitory activity of ethanol extract from guava leaf was 69.56%, which was the highest activity among the extracts. These results indicate that useful bioactive substances exist in the guava branch as well as leaf extracts.  相似文献
