综述了肠道病毒的种类、结构、生物学特性、致病性及肠道病毒的检测方法.通过对各方法优缺点的比较,认为免疫学检测方法在方便快速检测肠道病毒中的独特优势,尤其是免疫胶体金标记法和酶联免疫吸附试验备受青睐.提出确定肠道病毒的共同抗原和研制胶体金多元化检测试纸条可作为今后肠道病毒的通用检测方法的研究重点. 相似文献

猪圆环病毒2型(PCV2)是引起断奶猪多系统衰竭综合症(PMWS)的主要致病原,在全世界范围内对养猪业造成了巨大的经济损失。猪圆环病毒2型Cap蛋白(PCV2-Cap)是制备猪蓝耳病疫苗的主要靶蛋白。针对目前PCV2疫苗生产工艺复杂、成本较高的问题,本研究利用毕赤酵母表达系统研究PCV2-Cap蛋白的重组表达及疫苗活性,将PCV2-Cap基因序列去除核定位信号序列(NLS)后,经密码子偏好性优化,构建重组表达载体,并转化至毕赤酵母宿主菌X-33,经Zeocin抗性筛选获得重组菌株。SDS-PAGE和Western blot结果显示Cap蛋白在酵母表达系统中成功表达,蛋白质大小为26 ku。5 L罐高密度发酵小试,得到PCV2-Cap蛋白在发酵上清液中的浓度约为0.53 mg/m L,占总蛋白的23.5%。动物免疫试验显示重组Cap蛋白能刺激机体产生特异性抗体。结果表明,PCV2-Cap基因在毕赤酵母中实现了分泌表达,为PCV2-Cap毕赤酵母亚单位疫苗的研制提供了数据支持。 相似文献

克罗诺杆菌是一种食源性致病菌,能够感染婴幼儿并导致坏死性小肠结肠炎、脑膜炎和菌血症,死亡率最高可达80%。克罗诺杆菌广泛地存在于食品和自然环境当中,并且具有极强的抗干燥能力,因此容易污染乳粉和其原料并在其中长期存在。控制克罗诺杆菌的污染需要增强食品生产质量控制,也需要开发相应的检测技术。本研究主要从生理生化检测、免疫学检测技术、核酸检测技术等对克罗诺杆菌的检测方法进行综述,对上述各种检测方法的原理和优劣势进行了分析和总结,并且对克罗诺杆菌检测方法的未来发展进行展望。需要指出的是,由于克罗诺杆菌在婴幼儿食品中污染量低、婴幼儿食品基质成分复杂且检测要求高的特点,增菌培养是不同检测方法不可缺少的步骤并且是检测的限速步骤,未来还需要对这一环节进行更为深入的研究以提升检测效率。 相似文献

Five yogurts were fermented with each bacteria strain. The viability and pH of each yogurt during fermentation or storage
were evaluated, and then the cytotoxicity and antiviral activity against enterovirus (EV) 71 of cell-free supernatants (CFS)
of the metabolites of each yogurt were compared with those of de Man, Rogosa, and Sharpe (MRS) broth fermented with the same
bacteria. As the results, the number of viable bacteria for each yogurt after fermentation or during storage always remained
higher than 5 log CFU/mL and the pH of those ranged from 4 to 6. The CFS of all yogurts showed antiviral activity over 45%
against EV71, while it didn’t exhibit cytotoxicity in Vero cells. Specially, the CFS of yogurt fermented with Lactobacillus plantarum and Bifidobacterium bifidum exhibited high anti-EV71 activity of 92.74 and 90.44%, respectively. In contrast, the CFS of each MRS broth fermented with

Flavonoids are associated with multiple biological and pharmacological activities, including anti-enterovirus activity. An internal ribosomal entry site (IRES) required for viral protein translation is a potential drug target for enterovirus 71 (EV71). Regulation translation initiation requires the interaction of IRES specific trans-acting host factors with viral IRES element. By evaluation of 12 flavonoids against EV71 infection, we found that (a) 7,8-dihydroxyflavone, kaempferol, quercetin, hesperetin and hesperidin exhibited more than 80% of cell survival and inhibition of EV71 infection; however, no anti-oxidative effects were noted from these flavonoids; (b) among them, only 7,8-dihydroxyflavone, kaempferol and hesperetin showed 40% of viral IRES activity; (c) kaempferol interfered with EV71 virus replication and pseudotyped virus production; and (d) FUBP1, FUBP3, HNRPD, HNRH1 and HNRPF proteins are associated with EV71 5′-UTR as shown using RNA affinity pull-down assay coupled with LC–MS/MS analysis. We firstly found that kaempferol may change the composition of these IRES associated trans-acting factors, and affect IRES function and EV71 virus replication. The flavour and nutritional quality of UHT processed milk is discussed and suggestions are made for ways of minimizing undesirable changes during heat treatment. UHT treatment of milk using an infusion heater is described, and it is concluded that whole milk processed in this way is a typical directly heated UHT milk. 相似文献
