The effect of dietary C18:1 trans fatty acids on back fat composition in pigs was investigated with special emphasis on conjugated linoleic acids (CLA). A total of 12 × 4 siblings of Large White and Swiss Landrace breed were housed in groups and fattened from 22 to 103 kg live weight. Pigs were fed a control diet (barley, wheat, soybean meal) or experimental diets which consisted of the control diet with a 5% replacement of olein or stearin fractions of pork fat, or partially hydrogenated fat. The hydrogenated fat was rich in C18:1 trans fatty acids but contained only negligible amounts of CLA. In contrast olein and stearin fractions contained far less C18:1 trans fatty acids but some CLA. In the control diet no C18:1 trans fatty acids and only traces of CLA were detected. The partially hydrogenated fat led to the highest CLA content in back fat (0.44%). Intermediate amounts of CLA were measured in pigs fed the fractionated pork fat (0.22/0.23%). In pigs fed the control diet, also small amounts of CLA were detected. The results indicate that CLA may be produced by endogenous Δ9‐desaturation out of dietary trans vaccenic acid in pigs. 相似文献

Seasonal variation of octopus (Octopus vulgaris) lipid composition was investigated in four tissues: arm, mantle, ovary and digestive gland. A non‐homogeneous fat distribution was observed, with the digestive gland exhibiting a higher (p <0.05) lipid content than the other tissues. The ovary showed a higher (p <0.05) fat content than both muscle tissues, reaching its highest (p <0.05) value in winter. Neutral lipids – free fatty acids (FFA), triacylglycerols, and sterols (ST) – exhibited their highest (p <0.05) concentrations in the digestive gland and their lowest (p <0.05) values in muscle tissues. The phospholipid (PL) content of the ovary was the highest (p <0.05) of all tissues analysed, with the PL content also being significantly (p <0.05) higher in the digestive gland than in arm and mantle. The concentrations of most lipid classes (FFA, PL and ST) exhibited a seasonal variation. The fatty acid composition showed a remarkable difference between the digestive gland and all other tissues analysed. Thus, the digestive gland exhibited higher (p <0.05) contents in monounsaturated fatty acids and also lower (p <0.05) contents in both saturated (SFA) and polyunsaturated (PUFA) fatty acids. The highest mean values in SFA and PUFA were observed in ovary and muscle tissues, respectively. A seasonal effect was observed for SFA and PUFA. 相似文献

Palmitoleic acid has been classified as an insulin-sensitizing lipokine, but evidence for this from human studies has been inconsistent. We hypothesized that this is related to either the types of samples or conditions under which samples are collected. We measured plasma palmitoleic acid and total free fatty acids (FFA) using ultra-performance liquid chromatography in blood samples collected from 34 adults under a variety of conditions. We collected duplicate samples of adipose (n = 10), FFA (n = 9), and very low density lipoprotein triacylglycerol (VLDL-TAG) (n = 7) to measure the palmitoleic acid as a percentage of total fatty acids. We tested whether the percentage of palmitoleic acid was correlated with insulin resistance, as measured by homeostatic model of insulin resistance (HOMA-IR). Adipose stearoyl-coenzyme A desaturase 1 (SCD-1) protein was measured by capillary Western blotting. FFA-palmitoleic acid percentage increased as a function of total FFA and was greater (p < 0.005) in females than males. Adipose palmitoleic acid percentage was greater in females than males (p < 0.001), as was adipose SCD-1. Palmitoleic acid was greater in femoral fat than in abdominal fat in both females and males (p < 0.001), and correlated positively with HOMA-IR only in females. The test–retest reliability values for percentage palmitoleic acid were 7 ± 10% for adipose, 24 ± 26% for VLDL, and 53 ± 31% for FFA. Because FFA-palmitoleic acid percentage varies as a function of total FFA, investigators should re-evaluate how palmitoleic acid data is presented. The positive relationship between adipose palmitoleic acid and HOMA-IR in females suggests that it is not a potent insulin-sensitizing lipokine in humans. 相似文献

High concentrations of polyunsaturated fatty acids (PUFA) in meat have detrimental effects on its technical properties. The present study was carried out to investigate whether PUFA levels in pork can be reduced by increasing the concentrations of oleic acid in pig diets. To this end a bifactorial experiment was carried out with 48 female growing finishing pigs. Six different diets were used with two different concentrations of linoleic acid (12 vs. 24 g/kg) and three different concentrations of oleic acid (12 vs. 18 vs. 24 g/kg). The experiment started at a body weight (BW) of 58 kg and continued until 115 kg BW. The fatty acid composition of total lipids of backfat, perirenal fat and musculus (m.) longissimus dorsi was analysed. Concentrations of linoleic acid and total PUFA in backfat and perirenal fat were affected only by the dietary linoleic acid content but not at all by the dietary oleic acid content. Increasing the dietary concentration of oleic acid raised the level of oleic acid in those tissues at the expense of saturated fatty acids, suggesting competition between monounsaturated fatty acids and saturated fatty acids for incorporation into triglycerides. At the low dietary linoleic acid concentration, the percentages of linoleic acid and total PUFA in total lipids of m. longissimus dorsi were also unaffected by the dietary oleic acid content. In contrast, at the high dietary linoleic acid concentration, percentages of linoleic acid and total PUFA of the m. longissimus dorsi were reduced by increasing the dietary concentration of oleic acid, suggesting that oleic acid and linoleic acid compete for incorporation into muscle lipids. Thus, at high dietary linoleic acid levels the fatty acid composition of the m. longissimus dorsi was favourably affected by high dietary oleic acid concentrations; in backfat and perirenal fat, however, no beneficial effect of high dietary oleic acid levels was seen. 相似文献

Fatty acid analysis is frequently performed in fat and other raw materials to classify them according to their fatty acid
composition, but the need to carry out online determinations has generated a growing interest in more rapid options. This
research was done to evaluate the ability of a polymer-sensor based electronic nose to classify Iberian pig fat samples with
different fatty acid compositions. Significant correlations were found between individual fatty acids and sensor responses,
proving that sensor response data were not fortuitously sorted. Significant correlations also appeared between some sensors
and water activity, which was considered during the sample classification. Two supervised pattern recognition techniques were
attempted to process the sensor responses: 85.5% of the samples were correctly classified by discriminant analysis, but the
percentage increased to 97.8% using a one-hidden layer back-propagation artificial neural network. The electronic nose (specifically,
sensor responses analyzed by a neural network) achieved success similar to that obtained using the more usual fatty acid analysis

Guinea pigs are assumed to practice caecotrophy to a higher degree than rats. Studies from leporids suggest that through the practice of caecotrophy, hindgut fermenting species could build up microbial fatty acids (FA) in body tissues. We hypothesized that microbial FA would be detectable in the body tissue of guinea pigs and rats, and this to a higher degree in guinea pigs. Twenty-four rats and guinea pigs were fed with four different pelleted diets (lucerne-, meat-, meat-bone-, insect-based) in groups of six animals for 8 weeks. Perirenal adipose tissue differed in FA composition between the species in spite of the common diets. FA typically associated with microbial activity (saturated FA (SFA; typically 18:0), monounsaturated FA (MUFA; typically trans-fatty acids TFA), and odd- and branched-chain FA (Iso-FA)), were all detected. Guinea pigs had higher SFA levels than rats except on the lucerne diet. Concentrations of 18:0 were higher for guinea pigs on the meat and bone diet. Iso-FA concentrations in guinea pigs exceeded those of rats on all diets. FA profiles with a microbial fingerprint appear—although in low proportions—in the body tissue of both species, and this seemingly to a higher extent in guinea pigs. With respect to whether consumption of rodent meat rich in microbial FA has particular effects on human health as shown for ruminant products, microbial FA concentrations are probably too low to cause any distinct effects. 相似文献

Skeletal muscle is essential to maintain vital functions such as movement, breathing, and thermogenesis, and it is now recognized as an endocrine organ. Muscles release factors named myokines, which can regulate several physiological processes. Moreover, skeletal muscle is particularly important in maintaining body homeostasis, since it is responsible for more than 75% of all insulin-mediated glucose disposal. Alterations of skeletal muscle differentiation and function, with subsequent dysfunctional expression and secretion of myokines, play a key role in the pathogenesis of obesity, type 2 diabetes, and other metabolic diseases, finally leading to cardiometabolic complications. Hence, a deeper understanding of the molecular mechanisms regulating skeletal muscle function related to energy metabolism is critical for novel strategies to treat and prevent insulin resistance and its cardiometabolic complications. This review will be focused on both cellular and animal models currently available for exploring skeletal muscle metabolism and endocrine function. 相似文献

Stearidonic acid (SA, 18:4n‐3) is a polyunsaturated fatty acid (PUFA) that constitutes the first metabolite of α‐linolenic acid (ALA, 18:3n‐3) in the metabolic pathway leading to C20–22 PUFA, such as eicosapentaenoic acid (EPA, 20:5n‐3), and docosahexaenoic acid (DHA, 22:6n‐3), which recently have received much attention because of their various physiological functions in the human body. Recently, several studies indicated that dietary SA increased EPA more efficiently than ALA. Thus, vegetable oils containing SA may become a dietary source of n‐3 fatty acids that is more effective in increasing tissue n‐3 PUFA concentrations than the current ALA‐containing vegetable oils. Nevertheless, few SA sources occur in nature, although there are still a large number of species untested to date. SA has been detected in variable amounts in several species of algae, fungi and animals tissues, but the seeds of some plant families seem to be better sources of SA, especially Echium (Boraginaceae) species. The influence of the distribution of polyunsaturated fatty acids on the glycerol backbone of dietary triacylglycerols on the fatty acid profile of adipose tissue and muscle phospholipids was investigated in growing‐finishing pigs (48) and broiler chicken (84). The animals were fattened on barley/soybean meal diets supplemented with a blend of soybean oil and beef tallow, either in the ratio 3:1 w/w (high‐PUFA) or 1:3 w/w (low‐ PUFA). Part of the high‐ and low‐PUFA blends was chemically interesterified to randomly distribute all fatty acids over the three positions of the glycerol. Thus, two sets of diets of identical overall fatty acid composition, but differing in the distribution of fatty acids in the triacylglycerols, were fed. Growth performance and carcass composition were neither affected by fatty acid composition nor by randomisation of dietary fats in either animal species. Apparent digestibility of energy was slightly lower in pigs fed the low‐PUFA blends. Fatty acid profile of subcutaneous fat of pigs and broilers as well as of internal body fat (lamina subserosa) and muscle phospholipids of pigs varied according to the dietary fatty acid composition but was not affected by randomisation of dietary fats. These findings are explained in terms of the hydrolysis of TAG during transport of lipids from enterocytes to adipose tissue cells and the continuous lipolysis and re‐esterification of fatty acids that take place in adipose tissue cells. 相似文献

Fatty acids (FAs) have numerous functions in all living organisms, ranging from structural roles and energy production to the biosynthesis of secondary metabolites. Because of the high energy content of exogenous FAs, their acquisition is central of metabolism, and several biological systems are known, although their precise roles are not yet entirely clear. We investigated the roles of FadD (CoA ligase) and FadL (FA transporter) in different bacterial strains by using an improved version of click‐chemistry‐assisted labelling of azido‐FAs. The high sensitivity of this method allows a direct and precise assessment of FA metabolism, and is thus far better suited than growth experiments. Our results show that although FA activation is indeed essential for FA degradation, their transport can be independent of transporters like FadL. 相似文献

A total of 82 dietary fats sold on the Bulgarian market in the period 1995—2000 were analyzed. The samples included 68 table margarines (50 of which were imported), 10 frying fats (6 imported) and 4 salad dressings (all imported). A validated analytical method, thin‐layer chromatography‐AgNO3‐densitometry, was used. It enabled direct determination of all fatty acid groups, differing by degree of unsaturation and double bonds geometry. Low levels of trans fatty acids (TFA) down to 0.1% of the total for mono trans‐trienoic (Tcct) and mono trans‐dienoic (Dct), and down to 0.2% for trans‐monoenoic (Mt) were quantitated, with an error under 3% and a standard deviation of 0.1—1.5. The total content of TFA in table margarines varied from 0 to 26.9% with a mean value of 8.6 ± 7.2% for imported and 1.6 ± 3.4% for Bulgarian samples. Saturated fatty acids (SFA) content varied from 11.5 to 45.7%, with a mean value of 25.4 ± 5.7% for imported and 26.9 ± 5.2% for Bulgarian margarines. A general trend of lower levels of TFA and SFA in imported margarines was observed over the studied period. Additionally, the content of individual saturated fatty acids was determined by gasliquid chromatography in 37 of all studied samples. 相似文献

Fat is the second most abundant component of the nutrient composition of the mealworm Tenebrio molitor (Coleoptera: Tenebrionidae) that represents also an interesting source of PUFA, especially n-6 and n-3 fatty acids, involved in prevention of cardiovascular diseases. This study investigated the possibility of modifying the fat content and the FA composition of yellow mealworms through feeding and how this would be influenced by developmental stages, pupal sex, and generation with the future aim of applying this coleopteran as a diet supplement for human health. Growth rate and cumulative mortality percentage on the different feeding substrates were also evaluated to select the optimal conditions for a mass-raising of this insect species. Despite the different fat content in the six different breeding substrates used, T. molitor larvae and pupae contained a constant fat percentage (>34% in larvae and >30% in pupae). A similar total fat content was found comparing larvae and male and female pupae of the second generation to those of the first generation. On the contrary, FA composition differed both in larvae and pupae reared on the different feeding substrates. However, the exemplars reared on the diets based on 100% bread and 100% oat flour showed SFA, PUFA percentages, and an n-6/n-3 ratio more suitable for human consumption; the diet based on beer yeast, wheat flour, and oat flour resulted in a contemporary diet that most satisfied the balance between a fat composition of high quality and favorable growth conditions. 相似文献

Globoid cell leukodystrophy or Krabbe disease is an inherited autosomal recessive disorder caused by mutations in the galactosylceramidase gene. The objective of the study was to present information about the fatty acid (FA) composition of the brain and serum of twitcher mice, a mouse model of Krabbe disease, compared to wild type, in order to identify biomarker of disease progression. We defined the FA profiles by identifying the main components present in serum and brain using GC‐EI‐MS analysis. The FA percentage composition was measured and data were analyzed considering the disease and the mouse age as experimental factors. Significant correlations were established, both in brain and in serum, in the fatty acid percentage composition of twitcher compared to wild type mice. The most abundant saturated fatty acid in brain was the palmitic acid (C16:0) with mean values significantly increased in twitcher mouse (p = 0.0142); moreover, three monounsaturated, three polyunsaturated (PUFA) and a plasmalogen were significantly correlated to disease. In the serum highly significant differences were observed between the two groups for three polyunsaturated fatty acids. In fact, the docosahexaenoic acid (C22:6n3c) content was significantly increased (p = 0.0116), while the C20 PUFA (C20:3n6c and C20:5n3c) were significantly decreased in twitcher serum samples. Our study shows a specific FA profile that may help to define a possible pattern that could distinguish between twitcher and wild type; these data are likely to provide insight in the identification of new biomarkers to monitor the disease progression and thereby permit the critical analysis of therapeutic approaches. 相似文献

Three surveys of the content of trans fatty acids (TFA) in foods on the Danish market were carried out before and after the Danish regulation was introduced in January 2004 restricting the use of industrially produced (IP)‐TFA to a maximum of 2 g per 100 g fat in any food product. For this purpose, food samples were collected in 2002–3, 2004–5, and 2006–7. Of these, 60 paired samples (defined as samples included in two of the three investigations and with higher levels of IP‐TFA in the first determination than in the second) were identified. Comparisons of the fatty acid profiles showed that, in 68% of the products (e.g. sweets, cakes and cookies as well as fast food such as pie and tortilla), IP‐TFA were mainly substituted with saturated fatty acids (SFA). In some cases, the SFA source was coconut fat, whereas in other products, palm oil was added instead of partially hydrogenated oils. However, in important cases like frying fats, healthier fat substitutes with monounsaturated fatty acids were used. The surveys showed that the IP‐TFA content has been reduced or removed from most products with originally high IP‐TFA content, like French fries, microwave oven popcorn and various bakery products, so that IP‐TFA are now insignificant for the intake of TFA in Denmark. 相似文献

Even though colorectal cancer (CRC) is one of the most preventable cancers, it is currently one of the deadliest. Worryingly, incidence in people <50 years has increased unexpectedly, and for unknown causes, despite the successful implementation of screening programs in the population aged >50 years. Thus, there is a need to improve early diagnosis detection strategies by identifying more precise biomarkers. In this scenario, the analysis of exosomes is given considerable attention. Previously, we demonstrated the exosome lipidome was able to classify CRC cell lines according to their malignancy. Herein, we investigated the use of the lipidome of plasma extracellular vesicles as a potential source of non-invasive biomarkers for CRC. A plasma exosome-enriched fraction was analyzed from patients undergoing colonoscopic procedure. Patients were divided into a healthy group and four pathological groups (patients with hyperplastic polyps; adenomatous polyps; invasive neoplasia (CRC patients); or hereditary non-polyposis CRC. The results showed a shift from 34:1- to 38:4-containing species in the pathological groups. We demonstrate that the ratio Σ34:1-containing species/Σ38:4-containing species has the potential to discriminate between healthy and pathological patients. Altogether, the results reinforce the utility of plasma exosome lipid fingerprint to provide new non-invasive biomarkers in a clinical context. 相似文献
