Sixty-eight samples of cereals products, including breakfast cereals (n = 48) and infant cereals (n = 20), purchased from supermarkets and pharmacies in Rabat-Salé area from Morocco were analysed for the determination of six emerging mycotoxins: four enniatins ENs (ENA, ENA1, ENB and ENB1), beauvericin (BEA) and fusaproliferin (FUS). Samples were extracted with a mixture of acetonitrile:water (85:15, v/v), using an Ultra-Turrax® homogeniser. Mycotoxins were then identified and quantified by liquid chromatography (LC) with diode array detection (DAD). Positive samples were confirmed by LC–MS/MS.  相似文献

In the present study, the plant, grain and flour characteristics of 48 Amaranthus hypochondriacus and 11 Amaranthus caudatus lines are reported. A. hypochondriacus grains had a higher thousand kernel weight (0.62–0.88 g), a creamish yellow colour (L∗ = 61.38–68.29, b∗ = 5.26–6.8 and a∗ = 19.71–23.84), whereas A. caudatus grains had a lower thousand kernel weight (0.46–0.70 g) and a reddish-brown colour (L∗ = 47.1–51.2, b∗ = 11.82–14.02 and a∗ = 7.72–13.29). The A. caudatus lines had a higher protein content, fat content and tendency for retrogradation, and lower α-amylase activity as compared to A. hypochondriacus lines. The Amaranthus lines with higher a∗-values and lower thousand kernel weight, L∗- and b∗-values, had a higher fat, crude fibre and protein content. The correlation of fat and crude fibre content with the thousand kernel weight, L∗-, a∗- and b∗-values, was significant. The A. hypochondriacus lines showed higher pasting temperature, and lower peak viscosity, breakdown and setback, as compared to the A. caudatus lines. The result reflected that the lines with higher α-amylase activity showed a lower peak viscosity, breakdown, final viscosity and gel hardness, and a higher pasting temperature.  相似文献

Enniatins (ENs) are secondary fungal metabolites with hexadepsipeptidic chemical structure and they possess a number of potent biological activities that can contaminate several kind of food and foodstuffs increasing the exposure risk for consumers. ENs are produced by several Fusariun strains including Fusarium subglutinans, Fusarium proliferatum and Fusarium tricinctum. Production of a mixture of ENs was performed by culturing F. tricinctum ITEM 9496 on white corn as substrate. The solid culture components were dried and subsequently extracted with water/methanol (50/50 v/v, 0.5% NaCl), homogenised, filtered, extracted by ethyl acetate and analysed by liquid chromatography with diode array detection (LC-DAD). The crude extract was first separated by low pressure liquid chromatography (LPLC) and then further purified by liquid chromatography (LC), resulting in six compounds with a purity higher than 95% as calculated by 1H NMR, and with a yield of 30–300 mg per compound. The chemical structures of the ENs were determined by liquid chromatography coupled to mass spectrometry (LC–MS) and nuclear magnetic resonance (NMR). The biological activity of the resulting ENs was determined using a mitochondrial respiration test. We discovered that all the ENs studied induced an increase in the mitochondrial respiration resulting in uncoupling of the oxidative phosphorylation. This effect was most likely due to flux of K+ ions into the mitochondrial matrix. The order of potency of the ENs derivatives was: A1 > B1 > B > A > B4 > J1. These results suggest a correlation between the chemical structures and bioactivity and confirm the severe risks for human associated with consumption of enniatins.  相似文献

This study was carried out to determine the temporal effect of the antioxidants butylated hydroxyanisol (BHA) and propyl paraben (PP) at doses of 500 and 1000 μg/g on the growth of Fusarium verticillioides and F. proliferatum inoculated on natural maize grain in the presence of the competing mycoflora and fumonisin production at 0.98 and 0.95 water activity (aw) over a 28-day storage period. The reduction in the log colony forming units (CFU) of Penicillium, Aspergillus and Fusarium populations was 10-100 fold depending on dose of BHA or PP, aw and time. However, the populations of all three groups were higher at 0.98 aw than 0.95 aw. BHA at 500 μg/g and 0.95 aw reduced the fumonisin content by 82% after 7-14 days incubation, but at the end of the experimental period the reduction was only 32%. A higher reduction in the level of fumonisin produced (77%) was achieved with BHA at 1000 μg/g after 28 days. PP at 500 and 1000 μg/g decreased fumonisin production throughout the incubation period in the drier treatment, but at 0.98 aw control of toxin production was only achieved after 7-14 days. The reduction in the fumonisin levels could be due to the combined effect of antioxidants, and the competing mycoflora, mainly Aspergillus and Penicillium species.  相似文献

Adult feeding intensity, oviposition, and larval development of Sitophilus granarius (Coleoptera: Curculionidae) were observed on grain from three Polish wheat varieties (Begra, Korweta, and LGR 896/64a) washed with petroleum ether to remove cuticular lipids. Extraction of lipids did not cause any statistically significant changes in the physicochemical, biochemical and technological (milling, rheological and baking) properties of the wheat grain studied. Wheat grain washed with petroleum ether did not show any visible changes in the surface and morphology of the outer layer of the wheat grain. However, differences were noted in the microstructure of the kernel surface. Grain with a thicker seed coat (LGR 896/64a) was infested at a lower rate than the other varieties. Of the 18 hydrocarbons extracted from the grain surface, three compounds - n-heptacosane (C27), n-nonacosane (C29) and n-hentriacontane (C31) - were found in significant amounts. In general, beetles produced 64-95% less dust and laid 7-16% fewer eggs in kernels from which cuticular lipids had been removed. This implies that these compounds have a major role in food selection and the search for an oviposition site prior to grain infestation.  相似文献

Powder and essential oil obtained from dry ground leaves of Chenopodium ambrosioides were tested under laboratory conditions (25±1°C, 70-75% r.h.) for their ability to protect grains from damage by six insect pests, Callosobruchus chinensis, C. maculatus, Acanthoscelides obtectus, Sitophilus granarius, S. zeamais and Prostephanus truncatus. The insects were reared and tested on whole maize grain for S. zeamais and P. truncatus, whole wheat for S. granarius, green peas for C. chinensis, mung bean for C. maculatus and white bean for A. obtectus. The powder prepared from dry leaves of C. ambrosioides was mixed with grains at different dosages ranging from 0.05-0.80% (wt/wt) for C. chinensis, C. maculatus and A. obtectus and from 0.8-6.4% (wt/wt) for S. granarius, S. zeamais and P. truncatus. The dosage of 0.4% killed more than 60% of all the bruchids 2 days after treatment, while a dosage of 6.4% induced total mortality of S. granarius and S. zeamais within the same exposure time. All levels of the dry ground leaf concentrations inhibited F1 progeny production and adult emergence of the tested insects. The dosage of 0.2 μl/cm2 of the essential oil killed 80-100% of the beetles within 24 h except C. maculatus and S. zeamais, where this dosage induced only 20% and 5% mortality, respectively. These results indicate a scientific rationale for the use of this plant in grain protection by local communities in the western highlands of Cameroon.  相似文献

Out of four Cheyletus species occurring in grain stores in Central Europe, only C. eruditus is employed for the biocontrol of stored-pest arthropods. Unlike laboratory test data, field data on the differential natural performance (measured as frequency, abundance and predator-to-prey density dependence) of various species of Cheyletus spp. are not available. Therefore we investigated the relations between population densities of pest mites (Acari: Acaridida) and Cheyletus spp. predatory mites (Acari: Cheyletidae) in 147 grain stores in the Czech Republic. More than 1,000,000 individual pest mites and 40,000 individual predatory mites were extracted. We found that 29% of samples did not contain mites, 41% contained only pest mites, 4% only predatory mites, and in 26% both groups occurred simultaneously. Most abundant of the predatory mites were C. eruditus (79%) followed by “minor” species; C. aversor (10%), C. trouessarti (9%) and C. malaccensis (2%). There was a significant positive correlation between the occurrence and population density of the predatory and pest mites, except for C. malaccensis. Our results revealed C. eruditus as the mite predator with the highest natural performance in the field, indicating that it was the most pre-adapted species for biocontrol in central European grain stores.  相似文献

The prevalence of Campylobacter and Salmonella was assessed in 3959 raw red meats in the UK during 2003-2005. Meats were more frequently contaminated with Campylobacter (7.2%) than with Salmonella (2.4%). Lamb and other meats (e.g. mutton, rabbit) exhibited the highest contamination from Campylobacter (12.6% and 19.8%, respectively), compared with pork (6.3%) and beef (4.9%). Pork however had the highest contamination from Salmonella (3.9%), followed by lamb (2.0%), other meats (2.0%) and beef (1.3%). Offal samples (36.6%) were more frequently contaminated with Campylobacter or Salmonella than muscle tissue (7.0%). C. jejuni predominated in all meat types. C. coli isolates were more likely to exhibit antimicrobial drug resistance, including quinolones, than C. jejuni. Salmonella typhimurium was the most frequent Salmonella serotype isolated from meats; S. typhimurium DT104/104b isolates exhibited higher rates of multiple drug resistance than other serotypes. The findings reinforce the importance of adequate cooking of meat and good hygiene to avoid cross-contamination.  相似文献

Enniatins represent an emerging food safety issue because of their extensive incidence, documented in recent decades, in various small grain cereals. This study was concerned with the fate of these Fusarium mycotoxins within malting, brewing, milling and baking, when employed for the processing of contaminated barley and wheat. Besides enniatins A, A1, B and B1, also deoxynivalenol and its conjugated form (deoxynivalenol-3-glucoside) were determined in almost all tested cereal-based samples. Significant decline of enniatins occurred within all technologies, with the largest drop in their concentrations observed in the brewing process. While enniatins were not detectable in final beers, they were almost quantitatively transferred to spent grains, probably because of their limited water solubility. Regarding bread baking, levels of enniatins decreased down to 30% of their concentration in the initial flour used for baking. In this case, degradation at higher temperatures might be assumed.  相似文献

The aim of this study was to survey the presence of Staphylococcus aureus and Listeria monocytogenes during the cheese making process in small-scale raw milk cheese production in Norway.The prevalence of S. aureus in bovine and caprine raw milk samples was 47.3% and 98.8%, respectively. An increase in contamination during the first 2-3 h resulted in a 73.6% prevalence of contamination in the bovine curd, and 23 out of 38 S. aureus-negative bovine milk samples gave rise to S. aureus-positive curds. The highest contamination levels of S. aureus were reached in both caprine and bovine cheese after 5-6 h (after the first pressing). There was no contamination of L. monocytogenes in caprine cheeses and only one (1.4%) contaminated bovine cheese.This work has increased our knowledge about S. aureus and L. monocytogenes contamination during the process of raw milk cheese production and gives an account of the hygiene status during the manufacture of Norwegian raw milk cheeses.  相似文献

Data were collected in 1998-2002 from wheat stored in commercial grain elevators in south-central Kansas. Bins at these elevators had concrete walls and were typically 6-9 m in diameter and 30-35 m tall. A vacuum-probe sampler was used to collect grain samples in the top 12 m of the wheat in each bin. The primary insect species found in the wheat samples were: Cryptolestes ferrugineus, Rhyzopertha dominica, and Tribolium castaneum. In the top 3.7 m of grain, R. dominica, C. ferrugineus, T. castaneum and Sitophilus oryzae made up 44, 36, 19 and 1% of the insects found in the samples, respectively. From 3.8 to 12.2 m, R. dominica, C. ferrugineus, T. castaneum and S. oryzae were present at 84, 8, 8, and 1%, respectively. The most prevalent species also changed over time. In June, the start of wheat harvesting and storage in Kansas, insect density was low in the bins. At this time, C. ferrugineus was the most common insect, and it was found mostly in the top grain sample (0-1.2 m). In September through November, C. ferrugineus and R. dominica were at similar densities; however, from February to March, R. dominica was more common.Generally, insect density was greatest at the top and decreased with grain depth. Very few insects were found in samples collected from greater than 12 m (most of the bins contained grain to depths of 24-36 m). Insect density for all species increased rapidly from June through October. During this period less than 20% of the bins had economically significant insect densities (>2 insects/kg). From October until February, the average insect density remained fairly constant but it was greatly reduced in April, May, and June. Bins that had insect densities >2 insects/kg tended to be located adjacent to other heavily infested bins.  相似文献

This review summarises the published information on the behaviour of beetles in grain bulks that could lead to the identification of additional factors with the potential for enhancing existing lures or developing new ones. The review concentrates mainly on the adult stage of the five beetle pest species Oryzaephilus surinamensis (L.), Sitophilus granarius (L.), Cryptolestes ferrugineus (Stephens), Typhaea stercorea (L.) and Ahasverus advena (Waltl). After an analysis of the grain store environment and the origins of storage pests, particular attention is paid to behaviour associated with beetle movement and distribution within grain bulks, and their response to food and other multi-species attractants. Potential sources of new semiochemicals are identified; as well as the grains themselves, these include attractive volatiles associated with hot spots in grain, mouldy grain, and alternative insect pest habitats such as mammals’ and birds’ nests. It might also be worth looking for new semiochemicals produced during the mass upward movements of beetles from grain often seen following disturbance, although these could be repellents rather than attractants. Other sources of repellent semiochemicals with potential as grain protectants could be predatory and parasitic species of insects. Consideration is given to behavioural influences that may conflict with the intended response to attractive lures; for example, the presence of damaged grains, moisture and temperature gradients in the grain bulk may act as rival attractants, while many species prefer to hide away from bright light. As population densities rise, inter- and intra-specific competition becomes increasingly important. Human activities associated with grain handling, drying, cooling and pest control, as well as the presence of natural enemies and pathogens, all have the potential to affect insect response to a lure. Opportunities for the development of more effective lures are highlighted.  相似文献

The aim of this study was to assess infectious foot diseases, including identification and characterization of Dichelobacter nodosus and Treponema spp., in herds having problems with interdigital dermatitis (ID) and heel horn erosion (E) and in control herds expected to have few problems. We also wanted to compare diseased and healthy cows in all herds. The study included 14 dairy herds with a total of 633 cows. Eight herds had a history of ID and E, and 6 were control herds. All cows were scored for lameness, and infectious foot diseases on the hind feet were recorded after trimming. Swabs and biopsies were taken from the skin of 10 cows in each herd for bacterial analyses. In total, samples were taken from 34 cows with ID, 11 with E, 40 with both ID and E, and 8 with digital dermatitis (DD), and from 47 cows with healthy feet. Swabs were analyzed for identification and characterization of D. nodosus by PCR, culture, virulence testing, and serotyping. Biopsies were analyzed by fluorescent in situ hybridization regarding histopathology, identification, and characterization of Treponema spp., and identification of D. nodosus. Interdigital dermatitis was the most frequent foot disease, with a prevalence of 50.4% in problem herds compared with 26.8% in control herds. Heel horn erosion was recorded in 34.8% of the cows in problem herds compared with 22.1% in control herds. Dichelobacter nodosus was detected in 97.1% of the cows with ID, in 36.4% with E, in all cows with both ID and E, in all cows with DD, and in 66.0% of cows with healthy feet. All serogroups of D. nodosus except F and M were detected, and all isolates were defined as benign by the gelatin gel test. Treponema spp. were detected in 50.0% of the cows with ID, in 9.1% with E, in 67.5% with ID and E, in all cows with DD, and in 6.4% of those with healthy feet. In total, 6 previously described phylotypes (PT) of Treponema were detected: PT1, PT3, PT6, PT13, and PT15 in cows with ID, PT1 in a cow with E, and PT1, PT2, PT3, PT6, and PT13 in cows with both ID and E. One new phylotype (PT19) was identified. The epidermal damage score was higher but the difference in inflammatory response of the dermis was minor in cows with ID versus those with healthy feet. Fisher’s exact test revealed an association between ID and D. nodosus, and between ID and Treponema spp. Logistic regression revealed an association between both ID and E and dirty claws (odds ratios = 1.9 and 2.0, respectively). Our study indicates that D. nodosus, Treponema spp., and hygiene are involved in the pathogenesis of ID.  相似文献

Commodities may be attacked by a multitude of pest species simultaneously and so to be cost-effective any lure must be attractive to a range of target species. The objective of this study was to test a multi-species lure formulation with different dispensers under conditions as close as possible to their use in practice. The attractant effect of the multi-species lure formulation was tested using two types of dispensers with populations of the three principal grain beetle pests in the UK (Oryzaephilus surinamensis, Sitophilus granarius and Cryptolestes ferrugineus). The trials were performed over a period of six weeks with the lures tested in PC™ Traps: first in a grain bulk and in six grain bins and second in the surrounding area using PC™ Floor Traps. The amounts of volatiles released in and around in situ traps were measured using Solid-Phase Microextraction (SPME).The first lure dispenser tested was attractive to O. surinamensis and C. ferrugineus but it attracted fewer S. granarius than the control traps. It was shown that most of the volatiles were released at the beginning of the trial and therefore lures were not effective over the whole six-week period. The second lure dispenser released the attractant volatiles more consistently over six weeks and exerted significant attraction to O. surinamensis and C. ferrugineus in PC™ Traps in the grain bulk and to O. surinamensis and S. granarius in floor traps. Too few C. ferrugineus were caught in floor traps to make a comparison. The reason for the repellent effect on S. granarius in the grain bulk was unclear and this is discussed along with the importance of measuring volatiles released by the lure.  相似文献

Studies were conducted to observe the effect of grain quantity on the oviposition, distribution of eggs, adult emergence, adult body weight and sex ratio of Sitophilus zeamais, an important pest of maize. Three quantities of maize grains (200, 400 and 800 grains/glass jar of 1 l) were used under laboratory conditions (30°C and 70% r.h.). Twenty five male/female pairs of maize weevil were introduced into each glass jar for 12 d. The greatest and lowest number of eggs was laid on batches of 800 and 200 grain kernels, respectively. The total number of grains attacked followed a similar trend. The aggregation of eggs expressed as a ratio of the variance to mean increased as the amount of maize grains was increased. The aggregation parameter k ranged from 2.35 on 200 grains to 4.49 on 800 grains. On average, a significantly higher proportion of grains were infested (90%) when less grain was present. Emergence of adult weevils was, however, maximal (472 individuals/glass jar) when the grain quantity was highest. The mean weight of the emerged adults was not significantly influenced by grain quantity, but female weevils were heavier than males (mean weight 3.16 vs. 3.05 mg) irrespective of maize quantity and eggs laid. Sex ratio (males/100 females) of emerged adults did not differ among treatments. The importance of grain quantity, and of oviposition, in internally feeding granivores and the possibility that there is an adaptive reproductive strategy are discussed.  相似文献

Fusarium head blight is a fungal disease causing yield losses and mycotoxin contamination in wheat and other cereals. Wheat kernels (cultivar Ritmo) were sampled in 2001, 2002, 2003, and 2006 and Fusarium-damaged kernels were separated from sound grain based on visual assessment. Subsequently, grain lots containing 0, 20, 40, 60, 80, and 100% of damaged kernels were compiled. Each lot was split and the spectrometric reflectance (wavelengths 350-2500 nm) was measured using subgroup one, while the concentration of the mycotoxin deoxynivalenol (DON) was determined by high-performance liquid chromatography in subgroup two. DON concentrations in batches classified as sound were not significantly different from 0. Estimating DON contents from the percentage of Fusarium-damaged kernels was impeded by vast variability, resulting in a coefficient of determination of 0.49. Using spectrometric data subjected to partial least square regression allowed estimating DON contents with higher accuracy, in particular at elevated percentages of damaged kernels. The coefficient of determination was 0.84 for the relationship between DON contents estimated based on spectrometric data and the DON contents measured. The intercept of a regression line fitted through a plot of estimated versus measured DON contents was 0.89 ± 3.61 mg/kg. Since intercept + standard error was larger than the actual legal limit (1.25 mg DON per kg dry grain in the European Union), the spectrometric procedure was still not precise enough to allow a reliable separation of grain samples with DON contents below 1.25 mg/kg from samples with DON contents above the limit. However, spectrometric data also allowed estimating the DON content of the average damaged kernel within a given lot composed of sound and damaged kernels, which is probably the reason for the reduction of the fraction of unexplained variance by 35% compared to the visual approach and illustrates that spectrometric approaches can make a contribution to reducing DON contents of wheat grain.  相似文献

The root of Scorzonera austriaca has been used in indigenous cuisines as a delicious food and in the Tibetan traditional medicine in northwestern China. Two novel dimeric guaianolides linked by a carbon–carbon bond with a rare carbon skeleton, termed biguaiascorzolides A (1) and B (2), respectively, have been isolated from roots of S. austriaca. Acetylation of 1 gave 1a. The structures of 1, 1a and 2 were characterised by HR-ESI-MS, EI-MS, UV, IR, and 1D- and 2D-NMR techniques (1H and 13C NMR, 1H-1H COSY, HMQC, HMBC, and NOESY experiments). The cytotoxicity of 1a was assayed against selected cancer cell lines, including the human erythroleukaemia adriamycin-resistant subline (K562/ADM) and human stomach carcinoma (MGC-803) cell lines. Compound 1a exhibits a moderate activity against K562/ADM cell lines (IC50 39.8 μm) and is inactive towards MGC-803 cell lines.  相似文献

Maize ear rots reduce grain yield and quality with implication on food security and health. Some of the pathogenic fungi produce mycotoxins in maize grain posing a health risk to humans and livestock. Unfortunately, the levels of ear rot and mycotoxin infection in grain produced by subsistence farmers in sub-Saharan countries are not known. A survey was thus conducted to determine the prevalence of the ear rot problem and levels of mycotoxins in maize grain. A total of 114 farmsteads were randomly sampled from 11 districts in Lusaka and southern provinces in Zambia during 2006. Ten randomly picked cobs were examined per farmstead and the ear rot disease incidence and severity were estimated on site. This was followed by the standard seed health testing procedures for fungal isolation in the laboratory. Results indicated that the dominant ear rots were caused by Fusarium and Stenocarpella. Incidence of Fusarium verticillioides ranged from 2 to 21%, whereas that of Stenocarpella maydis reached 37% on ear rot diseased maize grain. In addition, 2-7% F. verticillioides, and 3-18% Aspergillusflavus, respectively, were recovered from seemingly healthy maize grain. The mean rank of fungal species, from highest to lowest, was F. verticillioides, S. maydis, A.flavus, Fusarium graminearum, Aspergillus niger, Penicillium spp., Botrydiplodia spp., and Cladosporium spp. The direct competitive ELISA-test indicated higher levels of fumonisins than aflatoxins in pre-harvest maize grain samples. The concentration of fumonisins from six districts, and aflatoxin from two districts, was 10-fold higher than 2 ppm and far higher than 2 ppb maximum daily intake recommended by the FAO/WHO. The study therefore suggested that subsistence farmers and consumers in this part of Zambia, and maybe also in similar environments in sub-Saharan Africa, might be exposed to dangerous levels of mycotoxins due to the high levels of ear rot infections in maize grain.  相似文献

Sixty-three strains of the taxonomically related species Lactobacillus plantarum subsp. plantarum, L. plantarum subsp. argentoratensis, L. paraplantarum and L. pentosus isolated from sourdoughs and other food and non-food sources and 14 strains of other members of the genus Lactobacillus were screened for their tolerance of acid, alkaline, heat, oxidative, osmotic, detergent and starvation stresses in order to evaluate the diversity of stress response. Most strains of the L. plantarum group were highly tolerant of acid, alkaline and osmotic stress and highly sensitive to detergent stress, while a larger diversity was found for other stress. Multivariate analysis allowed grouping the strains in clusters with similar response patterns. Stress response patterns in the L. plantarum group were similar to those of species of the L. casei/L. paracasei group but clearly different from those of other mesophilic Lactobacillus. No relationship was found between grouping obtained on the basis of stress response patterns and by genotypic fingerprinting (rep-PCR), nor with the taxonomic position or isolation source of the strains. Further experiments with selected strains showed that exponential phase cells were generally but not always more sensitive than stationary phase cells. The ability to grow under stressful conditions showed a slightly better correlation with the ecological conditions prevailing in the isolation niches of the strains.This study will be the basis for further investigations to identify and exploit the basis of diversity in the stress response of lactic acid bacteria.  相似文献
