This study was designed to examine the in vitro antioxidant activities of the methanol extracts of four Helichrysum species (Helichrysum noeanum Boiss., H. chionophilum Boiss. & Bal., H. plicatum DC. subsp. plicatum, H. arenarium (L.) Moench. subsp. aucheri (Boiss.) Davis & Kuphicha). The extracts were screened for their possible antioxidant activitiy by two complementary test systems, namely DPPH free radical-scavenging and β-carotene/linoleic acid systems. In the first case, non-polar subfractions of the methanol extracts of Helichrysum species studied did not show any antioxidant activity, while the most active one was H. chionophilum (IC50 =40.5 μg/ml) among the polar subfractions. In the β-carotene/linoleic acid test system, inhibition rates of the oxidation of linoleic acid of H. noeanum and H. arenarium were very close to each other. The inhibition rate of the synthetic antioxidant BHT was determined to be 96%. Since the polar extracts of Helichrysum species dealt with here exhibited interesting antioxidant activities when compared to BHT, we believe that it would be useful to take the results into consideration as an alternative for food processing industries.  相似文献

The ethyl acetate and methanol extracts from 16 Salvia L. species were screened for their inhibitory activity against acetylcholinesterase, butyrylcholinesterase, lipoxygenase, and tyrosinase; the enzymes linked to neurodegeneration. Their antioxidant activity was also tested using DPPH radical scavenging, metal-chelation, and ferric-reducing antioxidant power (FRAP) assays. Total flavonoid content of the extracts was determined by AlCl3 reagent, while HPLC technique was applied for analysis of various phenolic acids in the extracts. The extracts exerted weak cholinesterase and tyrosinase inhibition, and remarkable inhibition against lipoxygenase (13.07 ± 2.73-74.21 ± 5.61%) at 100 μg ml−1. The methanol extracts showed higher antioxidant activity in DPPH radical scavenging and FRAP assays. The extracts were analyzed for their gallic, protocateuchic, p-hydroxy-benzoic, vanillic, caffeic, chlorogenic, syringic, o- and p-coumaric, ferulic, rosmarinic, and tr-cinnamic acid contents and the methanol extract of Salvia ekimiana (153.50 mg 100 g−1) was revealed to be the richest in terms of rosmarinic acid.  相似文献

This study was designed to examine the in vitro antioxidant activities and rosmarinic acid levels of the methanol extracts of Salvia verticillata subsp. verticillata and S. verticillata subsp. amasiaca. The extracts were screened for their possible antioxidant activity by two complementary test systems, namely DPPH free radical-scavenging and β-carotene/linoleic acid systems. In the first case, S. verticillata subsp. verticillata was superior to the subsp. amasiaca with an IC50 value of 14.5 ± 1.21 μg mg−1. In the β-carotene/linoleic acid test system, inhibition capacity of S. verticillata subsp. verticillata was 74.4 ± 1.29%. Antioxidant activities of BHT, ascorbic acid, curcumin and α-tocopherol were determined in parallel experiments. Activity of rosmarinic acid was also screened for better establishing the relationship between rosmarinic acid level and antioxidant activity for the plant extracts. S. verticillata subsp. verticillata had the highest rosmarinic acid level with a value of 28.7 ± 0.89 μg mg−1. There is a strong correlation between the rosmarinic acid level and antioxidant activity potential. Our results showed that rosmarinic acid and its derivatives are more likely to be responsible for most of the observed antioxidant activities of Salvia species.  相似文献

Since Salvia species (Lamiaceae) have been recorded to be used against memory loss in European folk medicine, we herein examined in vitro anticholinesterase and antioxidant activities of 56 extracts prepared with petroleum ether, chloroform, ethyl acetate and methanol obtained from 14 Salvia species (Salvia albimaculata Hedge and Hub, Salvia aucheri Bentham var. canescens Boiss and Heldr, Salvia candidissima Vahl. ssp. occidentalis, Salvia ceratophylla L., Salvia cryptantha Montbret and Bentham, Salvia cyanescens Boiss and Bal., Salvia frigida Boiss, Salvia forskahlei L., Salvia halophilaHedge, Salvia migrostegia Boiss and Bal., Salvia multicaulis Vahl., Salvia sclarea L., Salvia syriaca L., Salvia verticillata L. ssp. amasiaca) growing in Turkey. The antioxidant activities were assessed by both chemical and enzymatic methods against 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical-scavenging and xanthine/xanthine oxidase (XO) system generated superoxide anion radical inhibition. Anticholinesterase effect of the extracts was tested against both acetylcholinesterase (AChE) and butyrylcholinesterase (BChE) at concentrations of 0.2 and 1 mg/ml using a microplate-reader assay based on the Ellman method. Most of the extracts did not show any activity against AChE at 0.2 mg/ml, while the chloroform extracts had noticeable inhibition against BChE between 47.7% and 74.7%. The most active extracts at 1 mg/ml for AChE inhibition were observed to be petroleum ether extract of Salvia albimaculata (89.4%) and chloroform extract of Salvia cyanescens (80.2%), whereas ethyl acetate extracts of Salvia frigida and Salvia migrostegia, chloroform extracts of Salvia candidissima ssp. occidentalis and Salvia ceratophylla, as well as petroleum ether extract of Salvia cyanescens were found to inhibit potently BChE (92.2%, 89.6%, 91.1%, 91.3%, and 91.8%, respectively). Particularly, the ethyl acetate and methanol extracts were observed to be highly active against both DPPH and XO. Our data indicates that nonpolar extracts of Salvia species for anticholinesterase activity and the polar extracts for antioxidant activity are worth further phytochemical evaluation for identifying their active components.  相似文献

In the current work, our target was to screen inhibitory potentials of 55 Turkish Salvia taxa, 28 of which are endemic, against acetylcholinesterase (AChE), which is a chief enzyme in pathogenesis of Alzheimer’s disease (AD). The dichloromethane, ethyl acetate, and methanol extracts prepared from 55 Salvia taxa were tested for their AChE inhibitory activity at 25, 50, and 100 μg/ml using an ELISA microplate reader. The extracts were also screened for their scavenging effect against DPPH radical and iron-chelating capacity. Total phenol and total flavonoid contents of Salvia fruticosa were determined. Among the 165 Salvia extracts screened, only the dichloromethane extract of S. fruticosa showed inhibition towards AChE at 100 μg/ml having 51.07% of inhibition, while only the dichloromethane and ethyl acetate extracts of Salvia cilicica had a notable iron-chelating capacity at 100 μg/ml having 54.71% of chelating capacity. Most of the extracts showed remarkable scavenging effect against DPPH radical.  相似文献

Teucrium polium L. (Lamiaceae) aerial parts are used widely in the daily diet and for medicinal purposes. This plant is used also as a spice and refreshing beverage. Phytochemical and bioactivity studies of this plant have been carried out. Aerial parts of the plant were extracted with petroleum ether, chloroform, methanol and water successively. Fractionation of the methanol extract yielded four major flavonoids. The crude extracts and isolated compounds were screened for their antioxidant and free radical scavenging activities using DPPH radical-scavenging, beta-carotene/linoleic acid and ammonium thiocyanate methods. Methanol extract, rutin and apigenin were found to be the most active fractions as radical-scavengers with IC50 values of 20.1 ± 1.7, 23.7 ± 1.9 and 30.3 ± 2.1 μg/ml, respectively. The samples with the highest inhibition of oxidation of beta-carotene and lipid peroxidation in ammonium thiocyanate methods were also found to be methanol extract, rutin and apigenin. Methoxylated flavonoids exhibited a lesser antioxidant activity.  相似文献

Leaves from the plant species belonging to the genus Ligustrum are widely used as tea or herbal medicine in Europe, China, and Japan. The antioxidant properties of five Ligustrum species from Taiwan were compared using in vitro antioxidant methods such as DPPH radical scavenging, TEAC, and FRAP assays. Cell-based antioxidant methods were used, including Fe2+/ascorbate-induced lipid peroxidation on brain homogenate and AAPH-induced erythrocyte haemolysis. The amounts of major phenolic compounds from the Ligustrum species, including phenylpropanoids, flavonoids, and iridoids, were determined by spectrophotometric methods. The results showed that all Ligustrum species exhibited antioxidant, radical-scavenging, anti-haemolytic, and lipid peroxidation-inhibiting activities at different magnitudes of potency. A significant correlation was found between antioxidant activity and the amount of antioxidant components, in particular, total phenolics and phenylpropanoids. Among all Ligustrum species from Taiwan, Ligustrum morrisonense is presented as potential source of natural antioxidants.  相似文献

The methanolic extracts of 16 Helichrysum species were investigated for their in vitro antioxidant, radical scavenging and antimicrobial activities. All the extracts showed strong antioxidant and radical scavenging activity. The highest total antioxidant capacity as ascorbic acid equivalents (AAE) of 194.64 mg/g dry extract was obtained for Helichrysum noeanum in the phosphomolybdenum assay. The highest IC50 value (7.95 μg/ml) was observed for the extract of Helichrysum stoechas subsp. barellieri in the DPPH assay. The total phenolic contents of the extracts ranged from 66.74 to 160.63 mg gallic acid equivalents (GAE)/g dry extract. The major component present in the extracts was identified as chlorogenic acid followed by apigenin-7-glucoside and apigenin by HPLC analysis. All the extracts showed significant antimicrobial activity against microorganisms containing 13 bacteria and two yeasts in the agar diffusion method.  相似文献

The aerial parts of two endemic Pimpinella [Pimpinella anisetum Boiss. & Ball. and Pimpinella flabellifolia (Boiss.) Benth. ex Drude] were hydro-distilled to produce oils in the yields of 2.07% (v/w) and 2.61% (v/w), respectively. The oils were analysed by GC and GC/MS. Twenty-one and nineteen components were identified, representing 99.5% and 99.7% of the oils, respectively. The main compounds of P. anisetum were (E)-anethole (82.8%) and methyl chavicol (14.5%), whereas limonene (47.0%), (E)-anethole (37.9%) and α-pinene (6.0%) were the major constituents of P. flabellifolia. The oils were screened for their possible antioxidant activities by two complementary test systems, namely DPPH free radical-scavenging and β-carotene/linoleic acid systems. In the first case, P. anisetum oil exerted greater antioxidant activity than that of P. flabellifolia oil with an IC50 value of 5.62 ± 1.34 μg/ml. In the β-carotene/linoleic acid test system, the oil of P. anisetum was superior to P. flabellifolia with 70.5% ± 2.86 inhibition rate. Essential oils of the plants studied here were also screened for their antimicrobial activities against six bacteria and two fungi. The oils showed moderate antimicrobial activity against all microorganisms tested.  相似文献

The antioxidant activity of Ruellia tuberosa L. (Acanthaceae) was investigated by the 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical-scavenging assay and the hydrogen peroxide-induced luminol chemiluminescence assay. The methanolic extract (ME) and its four fractions of water (WtF), ethyl acetate (EaF), chloroform (CfF), and n-hexane (HxF) were prepared and then subjected to antioxidant evaluation. The results of both methods revealed that R. tuberosa possesses potent antioxidant activity. The antioxidant activities of the different fractions tested decreased in the order of EaF > CfF > ME > WtF > HxF according to the hydrogen peroxide-induced luminol chemiluminescence assay, and results were the same with the exception of the rank order of HxF and WtF according to the DPPH free radical-scavenging assay. The results provide useful information on the pharmacological activities associated with free radicals of this traditional folk remedy.  相似文献

Total phenolic content, proanthocyanidins, gallotannins, flavonoids, and antioxidant activities of Sclerocarya birrea and Harpephyllum caffrum methanolic extracts were evaluated using in vitro assays. S. birrea young stem extract contained the highest levels of total phenolic content (14.15 ± 0.03 mg GAE/g), flavonoids (1.21 ± 0.01 mg CE/g) and gallotannins (0.24 ± 0.00 mg GAE/g). H. caffrum stem bark extract had the highest content of proanthocyanidins (1.47%). The EC50 values of the extracts in the DPPH free radical scavenging assay ranged from 4.26 to 6.92 μg/ml, compared to 6.86 μg/ml for ascorbic acid. A dose-dependent linear curve was obtained for all extracts in the ferric-reducing power assay. Dichloromethane and methanol extracts exhibited dose-dependent acetylcholinesterase inhibitory activity. Similarly, all extracts exhibited high antioxidant activity comparable to butylated hydroxytoluene based on the rate of β-carotene bleaching (84.1–93.9%). The two Anacardiaceae species provide a source of natural antioxidants and acetylcholinesterase inhibitors, and may be beneficial to the health of consumers.  相似文献

In this study, the antioxidant activity of methanol extracts of five plants from the genus Phyllanthus was evaluated by various antioxidant assays, including total antioxidant, free radical scavenging, superoxide anion radical scavenging, hydrogen peroxide scavenging, nitric oxide scavenging, reducing power and metal ion chelating activities. The various antioxidant activities were compared to standard antioxidants such as butylated hydroxytoluene and ascorbic acid. All the extracts showed strong antioxidant activity in all the tested methods. Among the five plants Phyllanthus debilis has been found to possess the highest activity in all tested models, the activity decreased in the order Phyllanthus debilis>Phyllanthus urinaria>Phyllanthus virgatus>Phyllanthus maderaspatensis>Phyllanthus amarus. In addition to the antioxidant activity of these plants, the total phenolic compounds, flavonoids and flavonols were measured in the extracts. A correlation between the antioxidant activity and total phenolic content was observed.  相似文献

The methanolic fresh leaf extracts of Macaranga gigantea, Macaranga pruinosa, Macaranga tanarius and Macaranga triloba were screened for their antioxidant properties (AOP), tyrosinase inhibition and antibacterial activities. Total phenolic content (TPC), 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging, ferric-ion reducing power (FRAP), ferrous-ion chelating (FIC) and lipid peroxidation inhibition (LPI) activities were used to evaluate the AOP. Modified 3,4-dihydroxy-L-phenylalanine (L-DOPA) method was used to determine tyrosinase inhibition activity, whereas antibacterial activity was determined using the disc-diffusion technique. TPC screening of the same species from different collection sites showed no significant difference between sites. M.triloba showed the highest ascorbic acid equivalent antioxidant activity (AEAC), FRAP and LPI values. M. tanarius, which showed the lowest TPC, AEAC, FRAP and LPI activities, exhibited the best FIC activity. M. pruinosa showed the best tyrosinase inhibition activity, whereas M. triloba showed the best antibacterial activity against Gram-positive bacteria species, with minimal inhibition dosage (MID) values as low as 10 μg/disc.  相似文献

The antioxidant activities of phenolic rich fractions (PRFs) from crude methanolic extract (CME), and its fractions using ethyl acetate (EAF), hexane (HF) and water (WF) of black mahlab (Monechma ciliatum) and white mahlab (Prunus mahaleb) seedcakes were investigated. The total phenolic compounds were found to be higher in white mahlab than black mahlab seedcakes. The antioxidant activity determined by the DPPH method revealed that black mahlab PRFs had the highest antioxidant activity, compared to white mahlab fractions. The presence of antioxidants in the two mahlab PRFs reduced the oxidation of β-carotene by hydroperoxides from these extracts/fractions. The effect of the two mahlab PRFs on the oxidative stability of corn oil at 70 °C was tested in the dark and compared with butylated hydroxyanisole (BHA). The CME performed better antioxidant activity in inhibiting the formation of both primary and secondary oxidation products. The qualitative and quantitative characterisation of phenolic compounds was carried out by HPLC/DAD.  相似文献

The chloroform and aqueous fractions of Carissa opaca fruit, a traditional medicinal fruit in Pakistan possessed a high amount of total phenolic and flavonoid contents as compare to other solvent fractions with potent antioxidant activities in scavenging DPPH, superoxides, hydroxyl, hydrogen peroxide, ABTS radicals, and had strong iron chelating activity. On the other hand, the ethyl acetate fraction showed the highest inhibition of β-carotene linoleic acid peroxidation and phosphomolybdate assay. A high correlation coefficient existed between EC50 values of DPPH, superoxides, hydroxyl, hydrogen peroxide, ABTS radicals, total phenolics and flavonoids, but a non significant correlation was found in the case of iron chelaters, β-carotene and phosphomolybdate assay. This study verified that the chloroform and aqueous fractions have strong antioxidant activities which were correlated with its high level of phenolics and flavonoids. These fractions can be used as a source of potential antioxidant or functional food material.  相似文献
