In this study, vascular stents were fabricated from poly (lactide-ɛ-caprolactone)/collagen/nano-hydroxyapatite (PLCL/Col/nHA) by electrospinning, and the surface morphology and breaking strength were observed or measured through scanning electron microscopy and tensile tests. The anti-clotting properties of stents were evaluated for anticoagulation surfaces modified by the electrostatic layer-by-layer self-assembly technique. In addition, nano-composite scaffolds of poly (lactic-co-glycolic acid)/polycaprolactone/nano-hydroxyapatite (PLGA/PCL/nHA) loaded with the vascular stents were prepared by thermoforming-particle leaching and their basic performance and osteogenesis were tested in vitro and in vivo. The results show that the PLCL/Col/nHA stents and PLGA/PCL/nHA nano-composite scaffolds had good surface structures, mechanical properties, biocompatibility and could guide bone regeneration. These may provide a new way to build vascularized-tissue engineered bone to repair large bone defects in bone tissue engineering.  相似文献

The aim of this study was to investigate the feasibility and advantages of the dual delivery of bone morphogenetic protein-2 (BMP-2) and basic fibroblast growth factor (bFGF) from nano-composite scaffolds (PLGA/PCL/nHA) loaded with vascular stents (PLCL/Col/nHA) for large bone defect regeneration in rabbit mandibles. Thirty-six large bone defects were repaired in rabbits using engineering bone composed of allogeneic bone marrow mesenchymal stem cells (BMSCs), bFGF, BMP-2 and scaffolds composed of PLGA/PCL/nHA loaded with PLCL/Col/nHA. The experiments were divided into six groups: BMSCs/bFGF/BMP-2/scaffold, BMSCs/BMP-2/scaffold, BMSCs/bFGF/scaffold, BMSCs/scaffold, scaffold alone and no treatment. Sodium alginate hydrogel was used as the carrier for BMP-2 and bFGF and its features, including gelling, degradation and controlled release properties, was detected by the determination of gelation and degradation time coupled with a controlled release study of bovine serum albumin (BSA). AlamarBlue assay and alkaline phosphatase (ALP) activity were used to evaluate the proliferation and osteogenic differentiation of BMSCs in different groups. X-ray and histological examinations of the samples were performed after 4 and 12 weeks post-implantation to clarify new bone formation in the mandible defects. The results verified that the use of sodium alginate hydrogel as a controlled release carrier has good sustained release ability, and the combined application of bFGF and BMP-2 could significantly promote the proliferation and osteogenic differentiation of BMSCs (p < 0.05 or p < 0.01). In addition, X-ray and histological examinations of the samples exhibited that the dual release group had significantly higher bone formation than the other groups. The above results indicate that the delivery of both growth factors could enhance new bone formation and vascularization compared with delivery of BMP-2 or bFGF alone, and may supply a promising way of repairing large bone defects in bone tissue engineering.  相似文献

We investigated the effect of bioactive nanoparticles on the in-vitro degradation of PLCL and PLCL/nHA composite scaffolds. The concentration of nanohydroxyapatite significantly affected the degradation rate. An increase in the crystallinity of the amorphous portion of the polymer was observed. This increased crystallinity was more pronounced in the pure PLCL samples than in those with more nHA. During the degradation process, we observed the appearance of multiple micropores on the scaffold walls as the hydrolysis process progressed and, by the sixth week, the remains of the degradation products were visible on the pore walls.  相似文献

Successful construction of small-diameter double-layer vascular scaffolds (SDVSs) whose inner diameters are less than 1.5 mm, especially those with multilayer mimic structures, remains a challenge in vascular tissue engineering. In this study, poly(L-lactide-co-caprolactone) (PLCL)/poly(Ɛ-caprolactone) (PCL)/tussah silk fibroin (TSF) SDVSs with a double-layer structure are prepared by one-step method based on friction twisting core-spun electrospinning technology. The constructed PLCL/PCL SDVSs grafted TSF have an obvious double-layer structure; tube wall thickness 524 ± 28 µm; and inner tube diameter 1390 ± 40 µm. Compared with traditional nanofiber vascular scaffolds (TS), the axial and radial tensile strengths of PLCL/PCL SDVSs grafted TSF increase by 86% and 34%, respectively. They also show good scaffold elastic recovery and burst pressure (BP) (8505 ± 875 mmHg). Compared with the PLCL/PCL SDVSs, the inner and outer layers of PLCL/PCL SDVSs grafted TSF show good hydrophilicity and protein adsorption performance. The in vitro cell viability results indicate that the inner and outer layers of PLCL/PCL SDVSs grafted TSF show enhanced proliferation and adhesion of vein endothelial cells (VECs) and smooth muscle cells (SMCs), respectively. Therefore, the successful preparation of PLCL/PCL SDVSs grafted TSF provides more possibilities for the clinical transplantation of small-diameter vascular scaffolds.  相似文献

For soft tissue engineering applications, 3-D macroporous acetylated chitosan/poly(l-lactideco-ε-caprolactone) (PLCL) scaffolds were prepared by acetylation and particulate leaching using sodium acetate in an acidic water/dioxane solution. Acetylated 5 wt% chitosan/PLCL scaffold of 90% porosity was determined and confirmed through various tests. The physiochemical properties of acetylated chitosan/PLCL hybrid scaffolds were examined by measuring water contact angles, pore morphology and interconnectivity using scanning electron microscopy (SEM), and dye release testing. In addition, mechanical properties such as tensile strength and bending stress recovery for determining the elasticity of scaffolds were measured. The fibroblast cell line NIH-3T3 was used to test relative cell affinities for the acetylated chitosan/PLCL vs. normal chitosan/PLCL films and porous scaffolds. The acetylated chitosan/PLCL films and scaffolds showed a high initial cell adhesion after 4 h of cell culture and increased cell proliferation compared to that of the control. The acetylated chitosan/PLCL scaffolds produced by particulate leaching showed a highly porous structure and improved the biocompatibility and stability of chitosan compared to that of chitosan-coated PLCL scaffolds. Thus, these scaffolds may be very useful for a variety of tissue engineering applications.  相似文献

A new mini‐deposition system (MDS) was developed to fabricate scaffolds with interconnected pore structures and anatomical geometry for bone tissue engineering. Polycaprolactone/hydroxyapatite (PCL/HA) composites with varying hydroxyapatite (HA) content were adopted to manufacture scaffolds by using MDS with a porosity of 54.6%, a pore size of 716 μm in the xy plane, and 116 μm in the z direction. The water uptake ratio and compressive modulus of PCL/HA composite scaffold increase from 8 to 39% and from 26.5 to 49.8 MPa, respectively, as the HA content increases from 0 to 40%. PCL/HA composite scaffolds have better wettability and mechanical properties than pure PCL scaffold. A PCL/HA composite scaffold for mandible bone repair was successfully fabricated with both interconnected pore structures and anatomical shape to demonstrate the versatility of MDS. POLYM. ENG. The objective of this study was to develop novel porous composite scaffolds for bone tissue engineering through surface modification of polycaprolactone–biphasic calcium phosphate‐based composites (PCL–BCP). PCL–BCP composites were first fabricated with salt‐leaching method followed by aminolysis. Layer by layer (LBL) technique was then used to immobilize collagen (Col) and bone morphogenetic protein (BMP‐2) on PCL–BCP scaffolds to develop PCL–BCP–Col–BMP‐2 composite scaffold. The morphology of the composite was examined by scanning electron microscopy (SEM). The efficiency of grafting of Col and BMP‐2 on composite scaffold was measured by X‐ray photoelectron spectroscopy (XPS) and Fourier transform infrared spectroscopy (FTIR). Both XPS and FTIR confirmed that Col and BMP‐2 were successfully immobilized into PCL–BCP composites. MC3TC3‐E1 preosteoblasts cells were cultivated on composites to determine the effect of Col and BMP‐2 immobilization on cell viability and proliferation. PCL–BCP–Col–BMP‐2 showed more cell attachment, cell viability, and proliferation bone factors compared to PCL–BCP‐Col composites. In addition, in vivo bone formation study using rat models showed that PCL–BCP–Col–BMP‐2 composites had better bone formation than PCL–BCP‐Col scaffold in critical size defect with 4 weeks of duration. These results suggest that PCL–BCP–Col–BMP‐2 composites can enhance bone regeneration in critical size defect in a rat model with 4 weeks of duration. © 2017 Wiley Periodicals, Inc. J. Appl. Polym. ABSTRACT

A facile method was used for fabricating in-situ-polymerized shape memory polyurethane (PU) hydroxyapatite (HA) nanocomposite scaffolds through electrospinning. The chemical structure and hydrogen bonding index (HBI) content were investigated using Fourier transform infrared spectroscopy. The crystalline morphology was analyzed by X-ray diffraction patterns. Differential scanning calorimetry was conducted to determine the glass transition temperature (Tg ) and degree of crystallinity. The results revealed that HA nanoparticles provide significant reinforcement of PU. Furthermore, incorporation of HA nanoparticles improved the shape memory properties of nanocomposites. The effect of HA nanoparticles on PU/HA electrospun fiber diameter, porosity, mechanical, and shape memory properties were examined.  相似文献

Bone defect and osteochondral injury frequently occur due to diseases or traumatism and bring a crucial challenge in orthopedics. The hybrid scaffold has shown promise as a potential strategy for the treatment of such defects. In this study, a novel biomimetic porous collagen (Col)/hydroxyapatite (HA) scaffold was fabricated through assembling layers of Col containing gradual amount of HA under the assistance of “iterative layering” freeze‐drying process. The scaffold presents a double gradient of highly interconnected porosity and HA content from top to bottom, mimicking the inherent physiological structure of bone. Owing to the biomimetic structure and component, significant increase of cell proliferation, alkaline phosphatase activity, and osteogenic differentiation in vitro was observed, illustrating potential application of the excellent Col/HA scaffold as a promising strategy for bone tissue engineering. © 2017 Wiley Periodicals, Inc. A dome-shaped elastic poly(l-lactide-co-caprolactone) (PLCL) scaffold with a channel and pore structure was fabricated by a combinative method of 3D printing technology and the gel pressing method (13 mm in diameter and 6.5 mm in thickness) for patient-specific regeneration. The PLCL scaffold was combined with adipose decellularized extracellular matrix (adECM) and heart decellularized extracellular matrix (hdECM) hydrogels and human adipose-derived stem cells (hADSCs) to promote adipogenesis and angiogenesis. These scaffolds had mechanical properties similar to those of native adipose tissue for improved tissue regeneration. The results of the in vitro real-time PCR showed that the dECM hydrogel mixture induces adipogenesis. In addition, the in vivo study at 12 weeks demonstrated that the tissue-engineered PLCL scaffolds containing the hydrogel mixture (hdECM/adECM (80:20)) and hADSCs promoted angiogenesis and adipose tissue formation, and suppressed apoptosis. Therefore, we expect that our constructs will be clinically applicable as material for the regeneration of patient-specific large-sized adipose tissue.  相似文献

Hydroxyapatite (HA), the bone mineral and Cissus quadrangularis (CQ), a medicinal plant with osteogenic activity, are attaining increasing interest as a potential therapeutic agent for enhanced bone tissue regeneration. In the present study a synergistic effect of these two agents were analyzed by fabricating PCL‐CQ‐HA nanofibrous scaffolds by electrospinning and compared with PCL‐CQ and PCL (control) nanofibrous scaffolds. Morphology, composition, hydrophilicity, and mechanical properties of the electrospun PCL, PCL‐CQ, PCL‐CQ‐HA nanofibrous scaffolds were examined by Field emission scanning electron microscopy (FESEM), Fourier transform infrared spectroscopy (FTIR), Contact angle and Tensile tests, respectively. The response of human foetal osteoblast cells on these scaffolds were evaluated using MTS assay, alkaline phosphatase activity, alizarin red staining, and osteocalcin expression for bone tissue regeneration. While the observed cellular response to both groups of scaffolds was better than for the control PCL scaffold, the PCL‐CQ‐HA nanofibrous scaffolds provided the most favorable substrate for cell proliferation and mineralization. The results showed that PCL‐CQ‐HA nanofibrous scaffolds had appropriate surface roughness for the osteoblast adhesion, proliferation, and mineralization comparing with other scaffolds. The observed investigation of physicochemical and biological properties suggests that the CQ‐HA loaded PCL nanofibrous scaffolds serve as a potential biocomposite material for bone tissue engineering. © 2013 Wiley Periodicals, Inc. To achieve novel polymer/bioceramic composite scaffolds for use in materials for bone tissue engineering, we prepared organic/inorganic hybrid scaffolds composed of biodegradable poly(ε‐caprolactone) (PCL) and hydroxyapatite (HA), which has excellent biocompatibility with hard tissues and high osteoconductivity and bioactivity. To improve the interactions between the scaffolds and osteoblasts, we focused on surface‐engineered, porous HA/PCL scaffolds that had HA molecules on their surfaces and within them because of the biochemical affinity between the biotin and avidin molecules. The surface modification of HA nanocrystals was performed with two different methods. Using Fourier transform infrared, X‐ray diffraction, and thermogravimetric analysis measurements, we found that surface‐modified HA nanocrystals prepared with an ethylene glycol mediated coupling method showed a higher degree of coupling (%) than those prepared via a direct coupling method. HA/PCL hybrid scaffolds with a well‐controlled porous architecture were fabricated with a gas‐blowing/particle‐leaching process. All HA/PCL scaffold samples exhibited approximately 80–85% porosity. As the HA concentration within the HA/PCL scaffolds increased, the porosity of the HA/PCL scaffolds gradually decreased. The homogeneous immobilization of biotin‐conjugated HA nanocrystals on a three‐dimensional, porous scaffold was observed with confocal microscopy. According to an in vitro cytotoxicity study, all scaffold samples exhibited greater than 80% cell viability, regardless of the HA/PCL composition or preparation method. © 2011 Wiley Periodicals, Inc. Bone transplants are used to treat fractures and increase new tissue development in bone tissue engineering. Grafting of massive implantations showing slow curing rate and results in cell death for poor vascularization. The potentials of biocomposite scaffolds to mimic extracellular matrix (ECM) and including new biomaterials could produce a better substitute for new bone tissue formation. A purpose of this study is to analyze polycaprolactone/silk fibroin/hyaluronic acid/minocycline hydrochloride (PCL/SF/HA/MH) nanoparticles initiate human mesenchymal stem cells (MSCs) proliferation and differentiation into osteogenesis. Electrospraying technique was used to develop PCL, PCL/SF, PCL/SF/HA and PCL/SF/HA/MH hybrid biocomposite nanoparticles and characterization was analyzed by field emission scanning electron microscope (FESEM), contact angle and Fourier transform infrared spectroscopy (FT-IR). The obtained results proved that the particle diameter and water contact angle obtained around 0.54 ± 0.12 to 3.2 ± 0.18 µm and 43.93 ± 10.8° to 133.1 ± 12.4° respectively. The cell proliferation and cell-nanoparticle interactions analyzed using (3-(4,5-dimethyl thiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium inner salt) MTS assay (Promega, Madison, WI, USA), FESEM for cell morphology and 5-Chloromethylfluorescein diacetate (CMFDA) dye for imaging live cells. Osteogenic differentiation was proved by expression of osteocalcin, alkaline phosphatase activity (ALP) and mineralization was confirmed by using alizarin red (ARS). The quantity of cells was considerably increased in PCL/SF/HA/MH nanoparticles when compare to all other biocomposite nanoparticles and the cell interaction was observed more on PCL/SF/HA/MH nanoparticles. The electrosprayed PCL/SF/HA/MH biocomposite nanoparticle significantly initiated increased cell proliferation, osteogenic differentiation and mineralization, which provide huge potential for bone tissue engineering.  相似文献

Chitosan/hydroxyapatite (CS/HA) bone repair scaffolds crosslinked by glutaraldehyde (GA) were prepared. Characterization of morphology, structure, mechanical property, and porosity of scaffolds were evaluated. The influences of CS viscosity, HA content, and crosslinking degree on properties of scaffolds were discussed. SEM images showed that CS/HA scaffolds were porous with short rod‐like HA particles dispersing evenly in CS substrate. When [η]CS = 5.75 × 10?4, HA content = 65%, and crosslinking degree = 10%, the resulting CS/HA scaffolds had a flexural strength of 20 MPa and porosity of 60%, which could meet the requirements of bone repair materials. The scaffolds were used as drug carriers for icariin, and the impacts of loading time and crosslinking degree of scaffolds on drug‐loading dose were discussed. The suitable loading time was 24 h and it would be better to keep crosslinking degree no more than 10%. The drug release behavior demonstrated that the icariin‐loading CS/HA scaffolds could achieve basic drug sustained release effect. © 2013 Wiley Periodicals, Inc. J. Appl. Polym. An excellent bioactive scaffold material which could induce and promote new bone formation is essential in the bone repair field. In this study, the bioactive material hydroxyapatite (HA) and the bone morphogenetic protein‐2 (BMP‐2) were added to poly‐l‐lactic acid (PLLA) using the electrospinning method. Scanning electron microscopy investigations performed on four different fiber scaffolds, PLLA, PLLA/HA, PLLA/BMP‐2 and PLLA/HA/BMP‐2, revealed that the fibers of all scaffolds are closely interwoven, and the presence of large interconnected voids between the fibers, resulting in a three‐dimensional porous network structure that was similar to the structure of the extracellular matrix of healthy bones. In the MG63 cell culture growth experiments, the composite scaffold material PLLA/HA/BMP‐2 showed a higher bioactivity than the other three scaffold materials. The four scaffold materials were implanted in rabbits’ tibia for 30 and 90 days. The results of the animal experiments indicate that the capability of the PLLA/HA/BMP‐2 composite to induce and promote bone tissue formation was better compared with PLLA/HA or PLLA/BMP‐2, suggesting that PLLA combined with HA/BMP‐2 is a promising material for bone tissue repair. © 2015 Wiley Periodicals, Inc. In composites of hydroxyapatite (HA) nanoparticles with a polymer matrix, the aggregation of nanoparticles would induce structural defects. In order to improve the dispersibility of HA nanoparticles in poly(ethylene glycol) dimethacrylate (PEGDMA) matrix and enhance mechanical properties of the HA/PEGDMA composite as potential bone substitute material, surface‐grafted HA nanoparticles with poly(ethylene glycol) monomethacrylate (PEGMA) were prepared, and crosslinked with PEGDMA under UV light to form a composite. The structure of HA‐g‐PEGMA was characterized by X‐ray diffraction (XRD) and thermal gravimetric analysis (TGA). The dispersibility of HA‐g‐PEGMA nanoparticles in poly(PEGDMA) was evaluated by SEM. The mechanical properties of the composites were investigated by compressive test. The dispersibility of HA‐g‐PEGMA nanoparticles in poly(PEGDMA) matrix was better than the bare HA. The physio-chemical properties of natural bone are significantly determined by the biological environment. Thus, it is desirable to prepare advanced bone tissue engineered scaffold by mimicking nature bone formation process. In this study, we proposed a new simple method (physio-chemical mixing) to prepare a hybrid hydroxyapatite (HA) scaffolds mimicking nature bone. Nano powder phases constitution was studied by SEM, XRD, and TEM. The results founded that the simple method could affect the physio-chemical properties (i.e., compositions and morphologies) of HA scaffolds. Moreover, the scaffolds prepared could have mimicked the nature bone.  相似文献

Bioactive calcium phosphate nanoparticles as reinforcing filler have widely been used to produce polymer nanocomposite scaffolds suitable for application in bone tissue engineering; however, no study has investigated the effect of geometry, size, and surface properties of these nanoparticles on physical and mechanical behavior of scaffolds. This study was therefore devoted to determine how the critical features of the reinforcing nanoparticles could tailor the efficiency of polymeric nanocomposites. For this, we developed fibrous nanocomposite systems in which poly(3-hydroxybutyrate-co-3-hydroxyvalerate), PHBV, was combined with hydroxyapatite (HA) nanoparticles having different sizes, geometries, surface chemical groups, and concentrations. The results showed that critical properties could be controlled by incorporating appropriate amounts of nanoparticles with a specific geometry and surface properties. It was also seen that the tensile modulus of fibrous nanocomposite increased from 77 MPa for neat PHBV to about 161 MPa for the nanocomposites containing 15 wt% surface-modified nanoparticles with elongated morphology.  相似文献

Continuous carbon fibre (CF) reinforced HA (CF/HA) composite scaffolds were prepared using a self-designed and manufactured 3D printer. The optimised design of nozzle structure and the tailored viscoelastic property of HA inks ensured compound extrusion of monofilament and multifilament CF with HA rod. The composite scaffolds designed using the CAD programme and sintered via a suitable process exhibited a hierarchical macro/microporous structure and contained approximately 50% HA and 50% β-TCP. The continuous CF synchronously enhanced the strength and toughness of the scaffolds. The compressive strengths of 1CF/HA and 5CF/HA were 11.4 ± 1.7 MPa and 16.3 ± 2.6 MPa, respectively, which were approximately double and triple compared with that of HA scaffolds. The fracture toughness of 1CF/HA was approximately double that of HA scaffolds and close to that of cortical bone. In vitro and in vivo studies demonstrated that 1CF/HA also had apatite formation capability and adequate bone regeneration capacity.  相似文献

A flexible and bioactive scaffold for adipose tissue engineering was fabricated and evaluated by dual nozzle three-dimensional printing. A highly elastic poly (L-lactide-co-ε-caprolactone) (PLCL) copolymer, which acted as the main scaffolding, and human adipose tissue derived decellularized extracellular matrix (dECM) hydrogels were used as the printing inks to form the scaffolds. To prepare the three-dimensional (3D) scaffolds, the PLCL co-polymer was printed with a hot melting extruder system while retaining its physical character, similar to adipose tissue, which is beneficial for regeneration. Moreover, to promote adipogenic differentiation and angiogenesis, adipose tissue-derived dECM was used. To optimize the printability of the hydrogel inks, a mixture of collagen type I and dECM hydrogels was used. Furthermore, we examined the adipose tissue formation and angiogenesis of the PLCL/dECM complex scaffold. From in vivo experiments, it was observed that the matured adipose-like tissue structures were abundant, and the number of matured capillaries was remarkably higher in the hydrogel–PLCL group than in the PLCL-only group. Moreover, a higher expression of M2 macrophages, which are known to be involved in the remodeling and regeneration of tissues, was detected in the hydrogel–PLCL group by immunofluorescence analysis. Based on these results, we suggest that our PLCL/dECM fabricated by a dual 3D printing system will be useful for the treatment of large volume fat tissue regeneration.  相似文献
